Actin's role in endocytosis has been challenging to elucidate due to the small scale and transient nature of endocytic structures. Inhibiting actin polymerization in yeast completely halts endocytosis. However, until this landmark paper, the corollary process in mammalian cells was less clear, because ablating actin polymerization only partially stalled trafficking. Yarar et al. (2005) used a mixture of electron and fluorescence microscopy and biochemical methods to establish a clear role for actin polymerization during late stages of clathrin-mediated endocytosis. A clever combination of TIRF and wide-field fluorescence imaging allowed precise vesicle tracking.
