Exploratory study using proton induced X-ray emission analysis and histopathological techniques to determine the toxic burden of environmental pollutants

The aim of this novel research was to determine the toxic burden of increased elements in water resources on the inhabitant wild animals (squirrels, turtles, bats), using particle induced x-ray emission (PIXE) and histopathological approaches. PIXE analysis of skin, muscle, lung, liver and kidney revealed significant increase in Al, Cl, Fe, Mg, Mn, Si and V. Moreover, data clearly reflect a significant (P < 0.001) deposition of toxic elements (Al, Cl, Fe and K) in the lung producing interstitial/proliferative pneumonitis, intra-alveolar hemorrhages, and thickening of alveolar capillary walls. The results obtained from the liver samples emphasized that majority of the animals were intoxicated with Cl, Mg, S, Si and V, which have produced profound deterioration and swelling of the hepatocytes. Likewise, histopathology of the kidney sections spotlighted severe nephritis and degenerative changes, which could be associated with the elevated amount of Al, Cl and Mg. This data undoubtedly provide relevant information on the heavy burden of toxic elements and their pathological outcomes in wild animals and highlight their potential risks for human exposure. Thus, the information provided is critical for developing effective strategies in dealing with health hazards associated with elemental exposures.

Proton induced X-ray emission analysis of soils with various length of fallow: The effect on soil total elemental composition

Fallowing of whatever kind is beneficial to elemental (nutrients) recovery. To what length (years) should a land be left fallowed for optimum elemental recovery is of serious concern to stakeholders owing to the limited availability of land. This work aimed at accessing the effect of fallow duration on soil total elemental composition. Proton induced X-ray emission (PIXE) technique has been used to assess the effect of relative longer length of fallowed land on soil total elemental composition using a 5SDH Tandem Pelletron accelerator available at Centre for Energy Research and Development, Obafemi Awolowo University Ile-Ife. The results at a 2.5MeV energy revealed the presence of elements: Si, P, Ca, Ti, Mn, Fe, Zn, Rb and Zr with the only sharp difference being the fact that elements Zn and Rb were only detected in the soil of continuous cropping probably due to the agrochemical applications prevalent in the study area. The inter-elemental correlation trend revealed a significant positive correlation of P to Ti and Cr (.916 and .937) at P<0.05 level of significance. A similar trend was obtained for Ca to Mn and Zr (.955 and .925). There was a very strong positive significant inter elemental correlation between Ti and Cr (.994) at P<0.01 level of significance. On the whole, leaving a land fallow for a relative longer period of time is beneficial in nutrients replenishment.

Elemental Characterization Of Indoor Breathable Particles at a Portuguese Urban Hospital

Hospitals are a specific indoor environment with highly susceptible individuals for whom indoor air pollution represents additional health risks. Particulate matter (PM) is one of the most health-relevant indoor pollutants due to its association with respiratory and cardiovascular diseases. Particles can also act as a carrier for various harmful organisms present in the air of hospitals, thus leading to airborne transmission of infectious diseases. Thus, the objective of this study was to characterize indoor PM collected in a hospital in consideration of concentration, size distribution, and elemental composition. Emission sources of indoor PM were indentified and risks associated with indoor PM estimated. Sampling was performed at radiology ward of a Portuguese urban hospital where PM10, PM2.5, and PM1 were collected during a period of 4 wk; PM elemental composition was determined by proton-induced x-ray emission (PIXE) analysis. Data showed that indoor PM10 concentrations ranged from 13 to 58.8 μg/m3 and from 10.5 to 41.9 μg/m3 for PM2.5. Fine particles constituted 77% of PM10, indicating that PM2.5 made a significant contribution to indoor air quality at the hospital. PM1 ranged from 9.9 to 35.6 μg/m3, accounting for 93% of PM2.5. PIXE identified 21 elements in PM, including health-hazardous metals (manganese, iron, copper, and vanadium) and carcinogenic elements (nickel, chromium, arsenic, and lead). However, no significant indoor source of PM emissions was identified, while outdoor air was the major contributor of indoor particles. Further, no significant risks existed through PM10 inhalation for population at the respective hospital.

Status of ion sources at National Institute of Radiological Sciences

The National Institute of Radiological Sciences (NIRS) maintains various ion accelerators in order to study the effects of radiation of the human body and medical uses of radiation. Two electrostatic tandem accelerators and three cyclotrons delivered by commercial companies have offered various life science tools; these include proton-induced x-ray emission analysis (PIXE), micro beam irradiation, neutron exposure, and radioisotope tracers and probes. A duoplasmatron, a multicusp ion source, a penning ion source (PIG), and an electron cyclotron resonance ion source (ECRIS) are in operation for these purposes. The Heavy-Ion Medical Accelerator in Chiba (HIMAC) is an accelerator complex for heavy-ion radiotherapy, fully developed by NIRS. HIMAC is utilized not only for daily treatment with the carbon beam but also for fundamental experiments. Several ECRISs and a PIG at HIMAC satisfy various research and clinical requirements.

QUALITATIVE AND QUANTITATIVE STUDY OF TRACE ELEMENTS IN DRUGS (OPIUM, HASHISH, ECSTASY PILL) BY PIXE AND NAA

In investigating drug addiction and its side effects from a social health point of view, the attention is usually focused on the drug itself. Yet trace amounts of other elements may have side effects no less harmful than addiction itself. The knowledge of these elements can be of help in the cure of drug addiction. The purpose of this study is to determine the trace elements in some drugs by methods of nuclear analysis. In this study, the Proton Induced X-ray Emission (PIXE) and Neutron Activation Analysis (NAA) techniques have been applied to measure the elemental composition and concentration of 55 opium, hashish and ecstasy pill samples. PIXE analysis shows the samples contain various elements including Mg, Al, Si, P, S, Cl, K, Ca, Ti, Fe, Cu, Zn, Rb and Sr .

Occupational genotoxicity among copper smelters

Occupational exposure in a copper smelting industry may produce various adverse health effects including cancer. Despite a number of well-documented studies reporting an increased risk of cancer among copper smelter workers, the data on genotoxic effects in this industry are scarce. In view of the above, an assessment of DNA damage in peripheral blood leukocytes by Comet assay from copper smelter workers was undertaken. Additionally, the proton-induced X-ray emission (PIXE) analysis was assessed to determine the metal content of samples. The study was conducted with all workers from a copper smelter (males; n=11), and a control group (n=11) was recruited. The results of our study showed a significant increase (p<0.001; Mann-Whitney test) in DNA damage in peripheral blood lymphocytes of smelter workers, compared to the controls (p<0.001; Mann-Whitney test). No correlation between DNA damage or metal concentration and age mean or time of exposure was found under study. Our findings indicate that copper smelter workers have increased levels of DNA damage in somatic cells, suggesting a potential health risk for the workers. PIXE results show the presence of copper, iron, and other metals.

Structure of melt-quenched AgIn3Te5

Polycrystalline AgIn3Te5 synthesized by melt-quench technique has been analyzed using proton induced X-ray emission (PIXE), X-ray diffraction (XRD), and selected area electron diffraction. PIXE analysis yielded the content of Ag, In, and Te, respectively, to be 9.76%, 31.18%, and 59.05% by weight. Structure refinement was carried out considering those space groups from I- and P-type tetragonal systems which possess 4 symmetry and preserve the anion sublattice arrangement of the chalcopyrite structure (space group: I42d) as well. The results showed that AgIn3Te5 synthesized by melt-quench method crystallizes with P-type tetragonal structure (space group: P42c; unit-cell parameters a = 6.2443(8) and c = 12.5058(4) Å), the presence of which was corroborated by selected area electron diffraction studies.

Characterization of tannery effluent wastewater by proton-induced X-ray emission (PIXE) analysis to investigate their role in water pollution

Over the last few decades, the chromium-based tanning industry has shown rapid growth in Pakistan. However, the rules and regulations promulgated by the government are not strictly followed for processing the effluent discharge from the tanneries. Consequently, tannery effluents have become a great source of water pollution in surrounding areas. In this case study, characterization of tannery effluent wastewater (TW), shallow groundwater (SW), and deep groundwater (DW) samples was carried out to determine the source of water pollution in the district of Kasur, Pakistan. The concentrations of calcium (Ca), chlorine (Cl), chromium (Cr), iron (Fe), potassium (K), Mg, sulfur (S), silicon (Si), and Sr in TW were significantly higher than SW and DW, which also exceeded the international limits. In addition, increased concentrations of major toxic elements (Cl, Cr, Fe, K, Ni, and Si) were also observed in SW, which were higher in comparison to DW. Strikingly, the concentrations of Cr and Si in various DW samples were also beyond World Health Organization (WHO) safe limit, which reinforced the trend that water pollution in the area is directly linked to the distance from the source (TW). The particle-induced X-ray emission (PIXE) indices also suggested that TW is a main contributory source of water-based pollution in the area, which is imposing great threat to local inhabitants due to known hazardous and carcinogenic potential of these elements. Protecting the water resources will be a formidable challenge in the study area, which requires modernization of tannery industry, thereby improving the recovery and recycling of TW. Moreover, PIXE analysis presented here as a successful tool, could serve as landmark for the contemporary research in environmental toxicology.

Rectangular Beads from the Final Gravettian Level of the Abri Pataud: Raw Material Identification and its Archaeological Implications

The Final Gravettian level (level 2) of the abri Pataud (Dordogne, France) yielded a large assemblage of body ornaments that consists essentially of 85 quite standardized rectangular beads. Some uncertainty remained about the raw material in which these small beads were made: mammoth ivory, reindeer antler or bone? Non-invasive methods were employed in order to determine the raw material. First chemical analyses using microbeam Proton Induced X-ray Emission analysis (microPIXE) did not enable us to conclude definitively. Therefore, synchrotron and laboratory X-ray microtomography (microCT) were applied on eight beads and allowed us to identify ivory for all of them except for one, which shows slightly different morphological features.

Direct characterization of airborne particles associated with arsenic-rich mine tailings: Particle size, mineralogy and texture

Windblown and vehicle-raised dust from unvegetated mine tailings can be a human health risk. Airborne particles from As-rich abandoned Au mine tailings from Nova Scotia, Canada have been characterized in terms of particle size, As concentration, As oxidation state, mineral species and texture. Samples were collected in seven aerodynamically fractionated size ranges (0.5-16 {micro}m) using a cascade impactor deployed at three tailings fields. All three sites are used for recreational activities and off-road vehicles were racing on the tailings at two mines during sample collection. Total concentrations of As in the <8 {micro}m fraction varied from 65 to 1040 ng/m{sup 3} of air as measured by proton-induced X-ray emission (PIXE) analysis. The same samples were analysed by synchrotron-based microfocused X-ray absorption near-edge spectroscopy ({micro}XANES) and X-ray diffraction ({micro}XRD) and found to contain multiple As-bearing mineral species, including Fe-As weathering products. The As species present in the dust were similar to those observed in the near-surface tailings. The action of vehicles on the tailings surface may disaggregate material cemented with Fe arsenate and contribute additional fine-grained As-rich particles to airborne dust. Dating and classification of Syrian excavated pottery from Tell Saka Site, by means of thermoluminescence analysis, and multivariate statistical methods, based on PIXE analysis

Thermoluminescence (TL) dating and Proton Induced X-ray Emission (PIXE) techniques have been utilized for the study of archaeological pottery fragment samples from Tell Saka Site, which is located at 25 km south east of Damascus city, Syria. Four samples were chosen randomly from the site, two from third level and two from fourth level for dating using TL technique and the results were in good agreement with the date assigned by archaeologists. Twenty-eight sherds were analyzed using PIXE technique in order to identify and characterize the elemental composition of pottery excavated from third and fourth levels, using 3 MV tandem accelerator in Damascus. The analysis provided almost 20 elements (Na, Mg, Al, Si, P, S, K, Ca, Ti, Mn, Fe, Co, Ni, Cu, Zn, Rb, Sr, Y, Zr, Nb). However, only 14 elements as follows: K, Ca, Ti, Mn, Fe, Co, Ni, Cu, Zn, Rb, Sr, Y, Zr, Nb were chosen for statistical analysis and have been processed using two multivariate statistical methods, Cluster and Factor analysis. The studied pottery were classify into two well defined groups.

Elemental analysis of limestone samples from Obajana and Mfamosing limestone deposits, Nigeria, using nuclear techniques

Six limestone samples were picked from three different points at the Obajana and Mfamosing limestone deposits. The limestone samples were subjected to elemental analysis by Proton-Induced X-Ray Emission (PIXE) analysis. The samples were irradiated by a 4 mm diameter beam of protons with energy of 2.5 MeV and beam current of 0.2 nA for 0.9 ms. The analysis was carried out with the 1.7 MV Tandem accelerator at the Centre for Energy Research and Development (CERD), Ile-Ife, Osun State, Nigeria. The NIST geology standard NIST 278 was analysed for quality assurance. The elemental composition and concentration of 14 elements were determined in the two locations. Ten elements were found at the Obajana deposit while 13 elements were found at the Mfamosing deposits. The elements: Mg, Al, Ca and Mn do not differ much at both deposits while others differ. The major elements (Ca, Fe, Al, Si and K) present in the limestones were also found in airborne particulate matter studied by earlier researchers. These observations suggest that all particulate emissions and wastes from the Limestone deposit should be closely monitored to reduce their cumulative effects on both health and the environment

Fluorine analysis of human dentin surrounding resin composite after fluoride application by μ-PIGE/PIXE analysis

The use of fluoride for the prevention of caries is based on the transformation of hydroxylapatite to fluoroapatite in the presence of fluoride ions, thereby strengthening tooth structure. Adhesion of dentin and resin composite (tooth-colored restoration material) requires a dentin bonding system, since resin composite is not able to adhere to dentin directly. Demineralization of dentin by acid etching is an important step in the dentin bonding system, however, demineralization also introduces weaknesses in tooth structure. If the demineralized dentin could be strengthened by the application of fluoride, then the dentin–resin composite bond strength might also improve. To test this hypothesis, the present study evaluated the influence of fluoride applications on the strength of the dentin–resin composite bond by (1) tensile strength testing analyses, (2) SEM analyses of tooth structure, and (3) detection of calcium (Ca) and fluorine (F) distribution patterns by micro proton-induced X-ray emission (μ-PIXE) and micro proton-induced gamma-ray emission (μ-PIGE) analyses conducted at the Takasaki Ion Accelerators for Advanced Radiation Application (TIARA) at the Takasaki Advanced Radiation Research Institute (TARRI).In this study, the dentin in extracted human molars was exposed by grinding and the dentin was etched with 35% phosphoric acid. Fluoride was applied at two concentrations, 0.022% (100ppm F) and 2.21% (10,000ppm F) NaF solution, for two time periods, 30 and 60s, prior to bonding the resin composite with the treated dentin. Controls were prepared in the same manner, but without the fluoride application. Bond strength was measured with a micro-tensile testing unit, and the fluorine and calcium distributions at the interface between dentin and resin composite were detected by μ-PIGE and μ-PIXE analysis, respectively.Results indicate that the 10,000ppm F applications resulted in higher bond strengths than observed in either the 100ppm F applications or the control group. In addition, PIGE analyses showed high concentrations of fluorine in the hybrid bonding layer of the 10,000ppm F samples, suggesting that the fluorine contributes to the strength of the dentin–resin composite bond. Detection of fluoroapatite within the hybrid bonding layer suggests that bond strength involves remineralization processes.

In situ analyses of gallstone inner layers by external PIXE

External proton induced X-ray emission analyses were carried out to study the elemental profile at different points of two mixed gallstones inner layers. It was observed that ten minor/trace elements namely P, S, Ca, Mn, Fe, Cu, Zn, Br, Rb and Sr were present in the mixed gallstone layers. Among the elements detected, Ca shows the highest concentration whereas Br, Rb and Sr were found only at some points in trace level. Concentrations of Ca and P were observed to be the highest in the core of the gallstones. The presence of high amount of Cu and Zn on the surface of the gallstones, as compared to other layers, may be due to contamination with bile.

PIXE analysis of trace elements in relation to chlorophyll concentration in Plantago ovata Forsk

Plantago ovata Forsk - an economically important medicinal plant - was analyzed for trace elements and chlorophyll in a study of the effects of gamma radiation on physiological responses of the seedlings. Proton-induced X-ray emission (PIXE) technique was used to quantify trace elements in unirradiated and gamma-irradiated plants at the seedling stage. The experiments revealed radiation-induced changes in the trace element and chlorophyll concentrations.

Elastomeric, Half-Facepiece, Air-Purifying Respirator Performance in a Lead Battery Plant

This workplace protection factor (WPF) study of a half facepiece air-purifying respirator with P100 filters was done in a lead battery manufacturing plant. Paired air samples for lead were collected inside and outside respirators worn by workers who were properly trained and quantitatively fit tested. Of the 45 valid sample sets, only four had detectable lead on the inside sample. WPFs were calculated for these sample pairs by dividing the outside sample lead concentration (C o ) by the inside concentration (C i ). For the remaining 41 sample pairs, the detection limit for lead was used to calculate a maximum estimated C i concentration. The C o for each of these sample pairs was divided by the C i estimate to obtain a minimum WPF value. All the WPFs were rounded down to two significant figures, resulting in values ranging from 12 to > 2500. A rank and percentile procedure resulted in a 50th percentile WPF of 270 and a lower 5th percentile estimate > 50. These WPFs exceed the assigned protection factor of 10 for half facepieces published by the Occupational Safety and Health Administration. This study's results support the APF of 10 and indicate the respirator provided appropriate protection as it was used in this study. The comparability of the two analytical methods commonly used together in WPF studies was also evaluated. The samples collected outside the respirators were analyzed for lead by proton-induced X-ray emission analysis (PIXE) followed by inductively coupled plasma spectrometry (ICP). While the two methods were highly correlated (r2 = 0.965), the mean PIXE lead mass was approximately 45% higher than the mean ICP value. This systematic bias was explained by the assumptions used to interpret the PIXE analytical results. When WPF studies use ICP and PIXE for C o and C i samples, respectively, the calculated WPF values are conservative estimates of respirator performance.

PIXE analysis of medieval silver coins

We applied the proton-induced X-ray emission (PIXE) analytical technique to twenty-eight medieval silver coins, selected from the Tunisian treasury. The purpose is to study the fineness evolution from the beginning of the 7th to the 15th centuries AD. Each silver coin was cleaned with a diluted acid solution and then exposed to a 3 MeV proton beam from a 1.7 MV tandem accelerator. To allow the simultaneous detection of light and heavy elements, a funny aluminum filter was positioned in front of the Si(Li) detector entrance which is placed at 135° to the beam direction. The elements Cu, Pb, and Au were observed in the studied coins along with the major component silver. The concentration of Ag, presumably the main constituent of the coins, varies from 55% to 99%. This significant variation in the concentration of the major constituent reveals the economical difficulties encountered by each dynasty. It could be also attributed to differences in the composition of the silver mines used to strike the coins in different locations. That fineness evolution also reflects the poor quality of the control practices during this medieval period. In order to verify the ability of PIXE analytical method to distinguish between apparently similar coins, we applied hierarchical cluster analysis to our results to classify them into different subgroups of similar elemental composition.

μ-PIXE and SAXS studies at the bone–cartilage interface

Micro Proton Induced X-ray Emission (micro-PIXE) analysis has been employed herein in investigating and quantifying the distribution of a number of essential elements in thin human diseased articular cartilage sections affected by osteoarthritis (OA). Various cations Ca, P and Zn have been reported to play an important role both in the normal growth and remodelling of articular cartilage and subchondral bone as well as in the degenerative and inflammatory processes associated with the disease; they act as co-factors of a class of enzymes known as metalloproteinases which are believed to be active during the initiation, progress and remodelling processes associated with osteoarthritis. Other important enzymes such as alkaline phosphatase are associated with cartilage mineralization. Synchrotron radiation X-ray fluorescence (SR-XRF) for mapping of elemental distributions in bone and cartilage has also been employed by the present group and others. In the current investigations using the cSAXS beamline at the Swiss light source, Small-Angle X-ray Scattering (SAXS) was carried out on decalcified human articular cartilage to explore the structural and organizational changes of collagen networks in diseased articular cartilage.

Atherosclerosis: Contrast-enhanced MR Imaging of Vessel Wall in Rabbit Model—Comparison of Gadofosveset and Gadopentetate Dimeglumine

To investigate the potential of gadofosveset for contrast material-enhanced magnetic resonance (MR) imaging of plaque in a rabbit model of atherosclerosis. All experiments were approved by the animal ethics committee. Thirty-one New Zealand White rabbits were included in one of four study groups: animals with atherosclerosis imaged with gadofosveset (n = 10) or gadopentetate dimeglumine (n = 7) and control animals imaged with gadofosveset (n = 7) or gadopentetate dimeglumine (n = 7). Aortic atherosclerosis was induced through endothelial denudation combined with a cholesterol-enriched diet. Control rabbits underwent a sham surgical procedure and received a regular diet. After 8 weeks, pre- and postcontrast T1-weighted MR images of the aortic vessel wall were acquired. Relative signal enhancement was determined with dedicated software. Statistical analysis was performed by using a generalized linear mixed model. Immunohistochemical staining with CD31 and albumin was used to assess microvessel density and the albumin content of the vascular wall. Group differences were analyzed by using a chi(2) test. Gadofosveset spatial distribution and content within the vessel wall were determined with proton-induced x-ray emission (PIXE) analysis. Postcontrast signal enhancement was significantly greater for atherosclerotic than for control animals imaged with gadofosveset (P = .022). Gadopentetate dimeglumine could not enable discrimination between normal and atherosclerotic vessel walls (P = .428). PIXE analysis showed higher amounts of gadopentetate dimeglumine than gadofosveset in both atherosclerotic and normal rabbit aortas. Immunohistochemical staining revealed the presence of albumin and increased microvessel density in the vascular walls of atherosclerotic rabbits.
