Rapid Multiplication of Geographical Indication (GI) Tagged Mysore Mallige (Jasminum azoricum L.) Using Single Node through In vitro Culture

Aims: The present study was undertaken to rapid multiplication of (GI) tagged Mysore Mallige (Jasminum azoricum L.) using single node through in vitro culture. Study Design: The present study follow Completely Randomized Design (CRD). Place and Duration of Study: The study was conducted at the Plant Tissue Culture Laboratory, Department of Horticulture, University of Agricultural Sciences, Gandhi Krishi Vignana Kendra, Bengaluru, from 2020 to 21. Methodology: Single node cutting from the young shoots of field growing plants were used as explants to conduct the experiment. The explants were sterilized and placed on MS medium supplemented with different concentrations and combinations of growth regulators, namely BAP and Kinetin. Results: Among the different combinations of growth regulators, the combination of BAP 1.5 mg L-1 and Kin 1.5 mg L-1 produced maximum number of shoots from single node. Very low intensity of callus formation was observed from the cut ends of single node explants that were cultured on half strength MS medium consisting of 1.5 mg L-1 BAP + 0.0 mg L-1 Kin and 1.5 mg L-1 BAP + 1.0 mg L-1 Kin. Conclusion: The use of growth regulators such as BAP and KIN is reliable for shoot regeneration even when the field explants are used.

The role of silver nanoparticles biosynthesized from Citrus aurantium Leaves in some Vegetative and physiological characteristics of fenugreek plant (Trigonella foenum- graecum L.)

The research was crried out in the plant tissue culture laboratory and the green house of Department of plant Biotechnology in College of Biotechnology/AL-Nahrain University. The aim of it, is the Green-synthesis of silver nanoparticles by using an aqueous extract of Citrus aurantium leaves and characterizied the silver nanoparticles, using visible method depend on colour changing of the solution(mixture of aqueous extract of C. aurantium leaves and 1mMol of AgNO3 under 60C ̊), UV-Visible spectroscopy method and Atomic Force Microscopy (AFM), then study the effects of foliar spray of of AgNPs on some vegetative and physiological characteristics of Fenugreek Plant (Trigonella foenum- graecum L.), such as fresh and dry weight, length of shoot, carbohydrates, proteins and proline. The result showed the positive effect of using AgNPs on these characteristics.

Direct Plant Regeneration from Nodal Explants of Grapefruit (Citrus paradisi Macfad.)

Aims: The present study was undertaken to develop a protocol for in vitro regeneration and multiplication of disease-free quality planting material of Grapefruit (Citrus paradisi Macfad.). Study Design: Completely Randomized Design (CRD) Place and Duration of Study: The study was conducted at the Plant Tissue Culture Laboratory, Department of Horticulture, University of Agricultural Sciences, Gandhi Krishi Vignana Kendra, Bengaluru, from 2020 to 21. Methodology: Nodal segments from the young shoots of field growing plants were used as explants to conduct the experiment. The explants were sterilized and placed on MS medium supplemented with different concentrations and combinations of growth regulators, namely BAP, Kinetin and Gibberellic acid. Results: Among the different combinations of growth regulators, the combination of BAP 1.0 mgL-1 and GA3 3.0 mgL-1 produced maximum number of shoots (7) from nodal segments Conclusion: The use of growth regulators such as BAP and GA3 is reliable for shoot regeneration even when the field explants are used.

Effect of different Potting Mixtures on Acclimatization of In vitro Developed Plants on Survival Percentage under Glass House Condition

The experiment was conducted during the years 2020-22, all of the experiments for the current research were carried out in the Plant Tissue Culture Laboratory, Department of Plant Molecular Biology & Biotechnology, College of Agriculture, Gwalior, Rajmata Vijayaraje Scindia Krishi Vishwa Vidyalaya, Gwalior (M. P.). Pomegranate cv. Bhagwa plantlets were placed in polythene bags containing various potting mixtures, such as soil, FYM, and Vermicompost, alone and in various combinations. The hardened plantlets were then exposed to direct sunshine to help them acclimate to their new surroundings. The percentage of tissue culture plantlets that survived was calculated. Experiments were conducted in a Completely Randomized Design (CRD) with four replications of each treatment and each replication contain three explants. Data were analyzed using Duncan’s multiple range test whereas the survival percentages were analysed by arc-sine transformation. On the basis of present study it is concluded that cocopeat soil combination had the highest survival rate (95.00%), followed by cocopeat (85%). In the case of pomegranate, sand, perlite, vermiculite, vermiculite soil and perlite sand alone were not shown to be effective hardening materials because all of the plants withered within a week. After two months of hardening under polyhouse circumstances, cocopeat soil showed the greatest growth with a notable increase in plant height (37.23 cm), followed by cocopeat (33.03 cm). Therefore, we may conclude that cocopeat is the ideal material for pomegranate plants that have been produced in vitro.

Growth response of sweet kaffir lime (Citrus hystrix) seeds due to kinetin and coconut water application using tissue culture technique

Sweet kaffir lime is one of Aceh’s local fruit crops, which is rarely found anymore. The problem of sweet kaffir lime cultivation is the difficulty of generative and vegetative plant propagation. The general objective of this research is to preserve the Acehnese sweet kaffir lime from extinction. One way that can be done is through plant propagation in tissue culture. The experiment was conducted in the Plant Tissue Culture laboratory of the Faculty of Agriculture, Universitas Malikussaleh. This research used a two-factor, completely randomized design, and ten replications. Two factors were tried: the concentration of kinetin (0, 1, and 2 mg/L) and the concentration of young coconut water (0, 15, 30 %). The results showed that increasing kinetin concentration could reduce shoot length, number of leaves, root length and slow the root emergence time of sweet kaffir lime in vitro. The best treatment was the 0 mg/L kinetin (without kinetin). Increasing coconut water concentration could increase shoot length, number of leaves, and root length of sweet kaffir lime in vitro. The best treatment was 30% coconut water concentration.

Use of Temporary Immersion Systems in the Establishment of Biofactories.

Companies dedicated to the large-scale propagation of plant species are known as biofactories or agricultural biotechnology companies. Globally, there are a large number of biofactories (large-scale production) or plant tissue culture laboratories (small-scale production) in charge of supplying commercial propagules of plants of economic interest. Each biofactory implements technological developments such as temporary immersion (TIS) systems that allow them to reduce costs. This chapter analyzes some of the biofactories established globally, the main plant species propagated, and whether or not they implement the use of TIS.

The Effect of BAP Concentration on In-Vitro Mutant Taro Regeneration

Plant regeneration through invitro culture is strongly influenced by the use of growth regulators. BAP is a class of cytokinins that affect cell division which is very well used to stimulate plant growth. The aim of the study was to determine the effect of BAP concentration on the regeneration of various in vitro mutant taro. The study was carried out at the Plant Tissue Culture Laboratory, Faculty of Agriculture, Hasanuddin University, Makassar. The method used was factorial completely randomized design with 2 factors as the environmental design. The first factor was the BAP concentrations of 0.0 mg/L, 1.0 mg/L, 1.5 mg/L, 2.0 mg/L and 2.5 mg/L. The second factor was mutant plants consisting of control plants (diploid), mutant 1 ( Putative-Tetraploid), mutant 2 (Mixoploid), mutant 3 (Mixoploid), and mutant 4 ( Putative-Tetraploid ). The results showed that the best regeneration was obtained by BAP concentration of 0 mg/L with control plants on the parameters of root length (2.41 cm), number of roots (11.66 pieces) and plantlet formation rate (13 days). The BAP concentration of 1 mg/L was shown by mutant plant 1 on the parameters of the number of shoots (26 pieces) and number of leaves (16 pieces). BAP concentration of 0 mg/L in mutant 4 had a shoot height of 1.83 cm. The results of ploidy flow cytometry analysis of mutant plants with Putative-Tetraploid parental traits after regeneration resulted in detectable diploid chromosomes that were different from the parent types in all BAP concentration treatments

Effect of the source of the plant part and plant growth regulators on the establishment and multiplication of vegetative branches of Rosmarinus officinalis L. in vitro

This search was carried out in the plant tissue culture laboratory at the College of Agriculture at the University of Kerbala from 2021 to 2022. The study included the use of in vitro technology in the use of plant parts and different combinations in the emergence and multiplication of farms vegetative branches of rosemary, the study was carried out in two stages after performing the sterilization process: the first included the emergence of vegetative farms by planting the growing tops and side nodes on the MS nutrient medium with different concentrations (0 , 1, 2, 3 mg.L-1) BA, and the second stage was carried out by cultivating the growths resulting from the nodes grown in the previous stage on the (MS) media, that was prepared with various concentrations (0, 1, 2, 3 mg.L-1) BA and ( 0, 0.1, 0.2, 0.4mg.L-1) NAA, The results of the study showed the superiority of the apical, as it achieved a response rate of 57.5% compared to the lateral shoots, which recorded a response rate of 37.5%, and the con. of 1 mg.l-1 BA was superior and achieved a response rate of 80% comparison with the neutral treat that earn a response rate It reached 15%, and the results showed that the same concentration of benzyl adenine at a concentration of 2 mg.l-1 achieved of higher average in the number and length of branches (4.74 branches.plantlet-1, 4.12 cm), while the concentration achieved 3 mg .l- 1 of it had the higher rate of leaves number and the Fresh and Dry weight of the branches (15.24 leaves.plantlet-1, 2986 mg and 1823 mg), respectively, comparison with the neutral treat that achieved the lower average, and the concentration 0.2 mg.L-1 NAA exceeded in achieving The highest average number of branches, leaves, fresh &dry weight of branches was (4.16 branches.plantlet-1 and 14.21 leaves.plantlet-1, 2606 mg and 1594 mg) respectively, while the concentration exceeded 0.4 mg.L-1 NAA in achieving the highest rate The length of the branches reached 3.74 cm. Role of Plant Growth Regulators on Multiplication of Stevia rebaudiana Inside Manufactured Temporary Immersion Bioreactor and Induction of Callus from Explant

The experiment was conducted at Plant Tissue Culture Laboratory - Department of Horticulture and landscape gardening, from October 2021 to January 2023. micropropagation experiments were conducted using five levels of Kinetin 0.0, 1.0, 2.0, 3.0, and 4.0 mg L. Callogenesis experiments were performed using four levels of Naphthalenacetic Acid 0.0, 1.0, 2.0, and 3.0 mg L. A temporary immersion bioreactor was used with a 10-minute immersion period every 4 hours, and two levels of Kinetin and two levels of benzyl adenine were added to the Murashige and Skoog medium as a comparative treatment. The results showed that the concentration of 1.0 mg L had a significant effect on the number of shoot and leaves, and the comparative treatment showed a significant effect on shoots height. The results of multiplication in the manufactured bioreactor showed a significant effect on shoots height, reaching 4.8 cm, and the concentration of 1.0 mg L benzyl adenine showed a significant effect on the number of shoots, with 3.2 shoot per plant, and the number of leaves for the same treatment was 17.1 leaves per shoot.

Influence of Water Stress on Expression of the X95950 Gene and Several Cellular Callus Components in Violet-Treated Sunflower Varieties

The research was conducted between 2017 and 2019 at the College of Agricultural Engineering Sciences and Laboratory of Plant Tissue Culture for Postgraduate Studies at the University of Baghdad. One experiment used a totally random design. The experiment examined the effects of PEG (Polyethylene glycol) at concentrations of 0, 2, 4, 6, and 8% on the development of three sunflower types (Ishaqi-1, Aqmar, and AL-Haja) exposed to UV-C rays for 40 minutes as a result of the growing of the juvenile peduncle outside the live body. The aim of the study was to better comprehend the physiological and biochemical changes caused by water stress on the callus of several sunflower varieties. The X95950 gene was amplified using qPCR technology to investigate drought tolerance gene expression and callus cell content of glutathione, ascorbic acid, and APX enzyme as indicators to determine the effect of PEG on callus tissue cells in the nutrient medium. The expression of the X95950 gene was influenced by water stress. It was particularly expressed in Ishaqi-1 at 4 and 8% PEG concentrations, with values of 1.64 and 1.01, respectively. The glutathione content and activity of the enzyme ascorbate peroxidase, which were 56.49 mol-1 and 0.149 mg-1 protein absorption units, differed significantly among the varieties. PEG concentrations significantly influenced glutathione content, with 8% achieving the highest average of 50.07 mol g-1 and 4% achieving the highest averages of ascorbic acid and ascorbate peroxidase enzyme activity of 2.462 mg 100 g-1 and 0.138 mg-1 protein absorption units, respectively. The interaction was considerable, with the maximum average glutathione callus content in the Aqmar variety reaching 69.60 mol g-1 at 6% PEG concentration. At 4% PEG concentration, the Aqmar variety outperformed in terms of APX enzyme efficacy, reaching 0.238 mg-1 absorption units of protein.

Effect of the Interaction of Kinetin with Indole Butyric Acid on the Multiplication of Steva Plant (Spanty) In Vitro

This study was conducted in the laboratory of plant tissue culture in the College of Agriculture, University of Diyala for the period 2022-2023 to find an integrated program for propagating and multiplying Stevia outside the living body using some auxins and cytokinins and their interaction in the components of the nutrient medium. Tissue propagation experiments included a sterilization experiment, as a concentration of 15 mg.L-1 of sodium hypochlorite which gave the best sterilization percentage of 100%, and the best time period of 15 minutes gave 76.66%. Regarding the interaction between Kin and IBA, the control was superior in plant height, number of leaves, fresh weight and dry weight. Whereas, the number of branches was superior to the concentration treatment of 2.0 mg.L-1, as it gave the best number of branches, reaching 3.46 branch.plant-1. As for the effect of NAA and IBA on the rooting, NAA was given at a concentration of 0.4 mg.L-1, the best root length was 1.94 cm, and the best number of roots was 6.6 root.plant-1. As for the acclimatization experiment, there was no significant differences between the media used. This may be attributed to the genetic ability of this variety to better tolerate environmental conditions. As for the medium used in all its combinations, it proved successful in the process of acclimatizing tissue culture plants. In general, the method used did not differ from the resulting method in acclimatizing most tissue culture plants.

2,4-Dichlorophenoxyacetic Acid and 6-Benzyladenine Stimulate of Indication and Biomass Accumulation in a Phaseolus Vulgaris Callus Culture

Obtaining callus tissue is one of the essential aims of plant tissue culture technology due to its great importance in subsequent experiments. The present study aimed to determine the interactive effects of plant growth regulators in stimulating calli cultures from the hypocotyl of Phaseolus vulgaris. The study was conducted in the Plant Tissue Culture Laboratory, Desert Studies Center, University of Anbar. Different concentrations of 2,4-D were used, including 1.0, 2.0, 3.0, and 4.0 mg L−1, with concentrations of BA including 0, 0.5, 1.0, 1.5, and 2.0 mg L−1. Sodium hypochlorite (NaOCl) was used to obtain sterile seedlings. The seeds were sterilized with four concentrations of 0, 2, 4 and 6% for 5, 10 and 15 min. We use the complete random design (RCD) for coefficient distribution. The results showed the mixture of NaOCl 4.0% for 10 min recorded the highest percentage of sterile seedling production (SSP) at 90%. As for the induction of callus culture, the hypocotyl extracted from sterilized seedlings was grown in MS medium supplemented with 2,4-D and BA concentrations for 21 days. The results showed that the combination 2.0+1.5 mg L−1 caused the highest callus formation response (CFR) by 100%, the lowest complete callus formation (CCF) by 5.80 days, and the highest fresh weight (FW) by 4.318 g. At the same time, the combination of 2.0+2.0 mg L−1 reduced the start of callus formation (SCF) by 3.40 days.

Induction of Callus of Two Cultivars from Rice Seed Embryos by the Effect of Abiotic Factors for Drought Tolerance ex Vivo

The experiment was conducted at the Plant Tissue Culture Laboratory, College of Agricultural Engineering Sciences, University of Baghdad, during the period 2021-2023, with the aim of inducing Callus from embryos of two rice cultivars: black rice and Amber 33, to enhance drought tolerance in vitro. The Completely Randomized Design (CRD) was applied for experimental arrangements. Different concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) (0, 1.5, 3, 4.5 mg L-1) were used to stimulate Callus, along with UV radiation of type UV-C with a wavelength (<280 nanometers), It does not affect the environment because it is absorbed by the atmosphere’s oxygen and ozone, for various durations of 15, 30, 45, and 60 minutes. A control treatment was also used for comparison. The experimental stress conditions were induced using different concentrations (0, 1.5, 3, 4.5, and 6%) of polyethylene glycol (PEG). The fresh and dry weights of Callus were measured for all treatments, in addition to assessing the drought tolerance index. The results showed significant differences for most of the studied traits. The highest fresh and dry weight of Callus was achieved at a concentration of 3 mg L-1 of 2,4-D, at 258.9 mg and 26.06 mg, respectively. The control treatment with UV-C radiation outperformed with the highest fresh and dry weight of Callus, reaching 356.8 mg and 37.81 mg, respectively, compared to other irradiation doses. The control treatment with other PEG concentrations yielded the highest fresh and dry weight of Callus for the cultivars treated with UV-C radiation, reaching 356.4 mg and 38.81 mg, respectively. The drought tolerance index in terms of dry weight, the treatment with 1.5% PEG yielded the highest rate at 0.73 mg, while the treatment with 6% PEG resulted in the lowest rate at 0.21 mg. additionally; the black rice cultivar treated with UV-C radiation outperformed the Amber 33 rice cultivar in terms of both fresh and dry weights of Callus.

Effect of 2,4-D and NAA in Callus Induction and Differentiation from Different Explants of Moringa Oleifera Lam

This study was carry out in the Laboratory of Plant Cells and Tissue Culture / Department of Forestry Sciences in the College of Agriculture and Forestry / University of Mosul during the year 2021-2022 to study the effect of adding different concentrations of 2,4-Dichloro-Phenoxyacetic acid (2,4-D) or Naphthalene Acetic Acid (NAA) to the MS medium in the induction of callus from different parts of the Moringa oleifera Lam. and then study the effect of different concentrations of 6-Benzyladenine (BA) and Kinetin (Kin) on the differentiation of callus into buds and embryogenesis. Concerning callus induction, the results showed that 2,4-D or NAA in the nutrient medium was necessary after three weeks of cultivation 2,4-D was superior to NAA in the percentage of callus induction, as callus induction occurred by 100% of the explants cultured in the media supplemented with 0.1 mg.l−1 2,4-D. On the other hand, the parts (apical buds, single node, parts of the leaf blade, epicotyl, cotyledon, hypocotyl) differed significantly among themselves in the percentage of their ability on callus induction, as the cotyledons gave the highest rate of induction, amounting to 80%. The shoots resulting from the callus rooted after four weeks of cultivation in media supplemented with different concentrations of IBA at a rate of 100%, regardless of the concentration of IBA. The study proved the existence of a genetic match between the mother plants and the plants produced from the callus induction from them.

In Vitro Propagation of Pomegranate (Punica granatum L.)

This study was conducted in the plant cell and tissue culture laboratory of the Department of Horticulture and Landscaping / College of Agriculture, University of Kirkuk – Iraq. To study in vitro propagation of pomegranate by adding (0.0, 0.5, 1.0, 1.5, and 2.0) mg .L−16-benzyladenine (BA) and (0.0, 2.0, 4.0, 6.0, and 8.0) mg .L−1Kinetin (Kin) on Initiation state. The results showed that the highest rate number of branches is 1.20 branches. Vegetative part −1 for the medium supplemented with 0.5 mg .L−1 BA, the highest average for the longest length branch was 2.00 cm, the highest average for the number of leaves was 6.40. Vegetative part −1 when cultivated in the medium supplemented with 1.5 mg.L−1 BA. Whereas, when treated with Kin, the highest rate of the number of branches was recorded, 0.70 branches, vegetative part−1. The length of the most extended branch was 1.00 cm, at a concentration of 4 mg .L−1. The highest average number of leaves was 3.40 leaves, vegetative part-1, when cultured with the medium supplied with 8 mg. L −1.

Role of Sucrose and Benzyladenine in Stevioside Content of Stevia Rebaudiana Leaves in Temporary Immersion Bioreactor

The experiment was carried out in the plant tissue culture laboratory of the Department of Horticulture and landscape gardening from October 2021 to January 2023. micropropagation experiments were conducted using concentrations of BA 0.0, 0.5, 1.0, 1.5 and 2.0 mg L, and an experiment with the addition of three levels of sugar (30, 60 and 90 g L) with two levels of BA (0.0 and 1.0 mg L). The results of the micropropagation showed that the concentration 1.0 mg L giving the highest average number of shoots of 3.5 shoots per plant, and giving the highest average number of leaves of 16.8 leaves of the shoot and the superiority of the comparison treatment in giving the highest average height of shoots amounting to 4.5 cm. The results showed that concentration 60 interacted with BA giving the highest average height of the shoots amounting to 4.3 plant shoots. and the concentration 60 interacted with the comparison treatment in giving the highest average shoot height of 4.4 cm and the concentration 90 interacted with BA in giving the highest average number of leaves amounting to 14.1 leaf shoot. The results indicated that the concentration 90 sucrose interacted with 1.0 mg L BA in giving the highest content of stevioside amounting 171.02

Isolasi dan Identifikasi Mikroba Penyebab Kontaminasi dari Udara di Laboratorium Kultur Jaringan Tumbuhan UIN-SU Medan

Plant tissue culture laboratory is a technique for growing cells, tissues or slices of plant organs which are often called explants in the laboratory in an artificial media containing aseptic (sterile) nutrients to become a whole plant. This research aims to know the isolation and identification of contaminating microbes in the plant tissue culture laboratory and to know the types of contaminating microbes in the plant tissue culture laboratory. This research was conducted from december 2022 to january 2023 at the Wahdatul Ulum plant tissue culture laboratory, uinsu medan with 5 main sampling points.the sampling point consists of an incubation room, planting room and practicum space which is divided into 3 collection points. Staining tests and biochemical tests are conducted at the USU faculty of mathematics and natural sciences microbiology laboratory. The results of the study found 23 bacterial isolates that contamined the wahdatul ulum plant tissue culture laboratory at UINSU Medan consisting of the genera bacillus, lactobacillus, streptobasil, aeromonas, pseudomonas, serratia, bacteroides and acinetobacter while the contaminating fungi were Mucor sp. Contamination occurs due to mobility and activity in the laboratory room.

Tissue Propagation of Marjoram Plant (in vitro)

The experiment was conducted in the plant tissue culture laboratory of the Department of Horticulture and Landscape Engineering, College of Agriculture, University of Kirkuk for the season 2022. to study the propagation of the marjoram plant tissue from tips of branches outside the body. plant parts were sterilized for a period of 10-15 min, water was used to remove dust, and they were sterilized inside the cultivation table in Naocl sodium hypochlorite solution (commercial minor solution) at a concentration of 6% for 8 minutes in order to remove suspended microscopic parts. plant parts were washed 3 times each for 5 minutes. plant parts were cut, and tips of branches were taken, with (1) cm. with kinetin at, 2, 4, 6, 8) mg. L-1. plants were vaccinated at a temperature of 25 + 2 m 0 with an intensity of illumination was 3000 lux, with a light sequence of 16 hours, followed by 8 hours of darkness, for 4 weeks of cultivation. data were taken for the experiment (length of the longest branch) (number of branches) (number of leaves). after replanting the tips of the branches for another 4 weeks of the multiplication process, data were taken at (length of the longest branch cm) number of leaves/plant part). tips of branches. Regarding the root experience, data were taken (percentage of rooting %) (average number of roots/vegetative part) (average root length/cm) (branch length/cm) (number of leaves/vegetative part) full and half concentration of salt from the same soil were added to cultivated on a medium with different concentrations of IBA.

Conventional propagation of patchouli plants has not met the demand for superior seeds. Propagation of superior seeds in a relatively short time can be done using in vitro culture techniques. This research aims to determine the induction of patchouli shoots of Tapaktuan, Lhokseumawe and Sidikalang varieties with Benzyl Amino Purine (BAP) concentrations in vitro. This research was carried out at the Plant Tissue Culture Laboratory, Faculty of Agriculture, Syiah Kuala University. This research used a Randomized Block Design with a 3x4 factorial pattern consisting of 2 factors. The first factor was patchouli variety which consisted of 3 levels, namely: Tapaktuan, Lhokseumawe, and Sidikalang. The second factor was BAP concentration which consisted of 4 levels, namely: 0 (control), 0.5, 1, and 1.5 mg L-1. The results showed that the percentage of live patchouli explants was 100% in all treatments. The time and percentage of shoot emergence tended to be faster in the Lhokseumawe + BAP 0.5 mgL-1, namely 17.5 days after planting (DAP) and 100%. The number of patchouli explant shoots tended to be high in the Sidikalang + BAP 0.5 mgL-1, namely 12 shoots. The number of patchouli explant leaves tended to be high in the Lhokseumawe + BAP 1 mgL-1, namely 13 sheets. Patchouli explants tended to be taller in the Sidikalang + control, namely 8 cm. The percentage of callus formation tends to be high in the varieties Tapaktuan + BAP 1 mgL-1, Sidikalang + BAP 0.5 mgL-1, Sidikalang + BAP 1 mgL-1, Tapaktuan + BAP 1.5 mgL-1, Sidikalang + BAP 1.5 mgL-1 and Sidikalang + control were above 50% at 4 WAP. Respon Pertumbuhan Tanaman Porang (Amorphophallus muelleri Blume.) Dengan Penambahan NAA (Naphthalene Acetic Acid) dan BAP (6-Benzyl Amino Purine) Pada Perbanyakan Secara In Vitro

Porang is one of the export crops that are much hunted in several countries. The increasing number of needs has resulted in limited porang seeds so that there is a need for propagation of porang plants with good quality, one of which is by propagation through tissue culture or in vitro. In order to propagate porang plants in vitro, this research tries to discover the ideal concentration of NAA and BAP. From August to December 2022, this study was conducted in the Plant Tissue Culture Laboratory of Jember State Polytechnic. The experiment employed a parametric statistical design with 2 treatments: 0.5 mg/L NAA + 1.5 mg/L BAP (P1) and 0.5 mg/L + 2.5 mg/L BAP (P2), with each treatment being repeated 10 times. According to the findings, the parameters of callus emergence time (46 HST) and callus diameter (1.51 cm) were best achieved by administering NAA and BAP at a dosage of 0.5 mg/L NAA + 2.5 mg/L BAP. According to the study aims, the optimal concentration for explant growth and development is believed to be 0.5 mg/l NAA + 2.5 mg/l BAP.
