Assessment of ferrocyanide intake from food-grade salt in the Korean population

A simple high-performance liquid chromatography-ultraviolet (HPLC-UV) method for estimating the daily intake of ferrocyanide (FeCN) ion from food-grade salts was developed and validated. The levels of FeCN ion in food-grade salts available in Korean markets were analysed. FeCN ion was detected in 8.9% of 801 samples, with concentrations ranging from ND (not detected) to 9.6 mg/kg. The FeCN-ion levels were below the legal limit (10 mg/kg) specified by the Korean Food Code. The FeCN-ion intake of the Korean population was evaluated by combining food-intake data from the 2012–2014 Korea National Health and Nutrition Examination Survey (KNHANES), and the concentration data. The mean estimated daily intakes (EDIs) of the general population and consumers were 0.010 and 0.012 μg/kg bodyweight (bw)/day, respectively, when the mean values from the tested samples were used. The 95th percentile intake levels of both groups were 0.036 and 0.039 μg/kg bw/day, respectively. The EDIs of FeCN ion for all groups were much lower than the acceptable daily intake (ADI) of 0.025 mg/kg bw/day. These results provide useful information about the use of FeCNs as anticaking agents for consumers and manufacturers.

Dietary exposure assessment of cyadox based on tissue depletion of cyadox and its major metabolites in pigs, chickens, and carp.

The tissue kinetics of cyadox, an antibacterial agent used in food animals, and its major metabolites in pigs, chickens, and carp were investigated followed by a complete dietary exposure assessment to evaluate the food safety of cyadox. Cyadox and its major metabolites, bisdeoxycyadox (Cy1), 4-desoxycyadox (Cy2), N-(quinoxaline-2-methyl)-cyanide acetyl hydrazine (Cy4), quinoxaline-2-carboxylic acid (Cy6), and 2-hydromethyl-3-hydroxy-quinoxaline (Cy12), were simultaneously quantitated with a high-performance liquid chromatography-ultraviolet (HPLC-UV) method. Pigs, chickens, and carp were fed with 150mg/kg cyadox in feed for consecutive 60, 40, and 30days, respectively. The residue amount of cyadox and its major metabolites in liver, kidney, muscle, and fat (skin) tissues was determined. Cy2 was below the limit of quantitation even at the withdrawal time of 6hr, cyadox, Cy4, Cy6, and Cy12 could be detected at 6-24hr with low level less than 50μg/kg. By contrast, Cy1 persisted for 3days in the kidney of pigs and chickens, and in the liver of carp. Based on these residue depletion data and previous toxicology results, the global estimated chronic dietary exposure assessment of cyadox for general population was conducted, indicating a zero withdrawal time (WDT) may be appropriate for cyadox in food animals when used in feed for prolonged administration. These results provide analytical techniques and safety standards suitable for residue monitoring of cyadox in food animals.

Determination and comparative analysis of 13 nucleosides and nucleobases in natural fruiting body of Ophiocordyceps sinensis and its substitutes

ABSTRACTNucleosides and nucleobases are one of the most important indicators of quality control. A sensitive and reliable high performance liquid chromatography-ultraviolet method was applied to analyse 13 nucleosides and nucleobases simultaneously in 15 batches of nine Ophiocordyceps species and its allies in China. Principal component analysis (PCA) and cluster analysis were conducted by SPSS 22.0 software (IBM Corp., Armonk, NY, USA). The 15 samples of Cordyceps were differentiated successfully based on their nucleoside and nucleobase content. Total nucleosides content in mycelium was significantly higher than that in the natural fruiting bodies of Ophiocordyceps sinensis (NFOS). Five nucleosides or nucleobases – adenine (A), guanosine (Gu), uracil (U), uridine (Ur) and guanine (G) – were the major components contributed to the total variance according to PCA. The profiles of the 13 tested nucleosides and nucleobases (including adenosine, cytidine, guanosine, inosine, thymidine, uridine, cordycepin, adenine, cytosine, guanine, thymine, uracil and hypoxanthine) can discriminate different samples and can be candidate indicators applied for the quality control of Ophiocordyceps and its allies.

A New Oleanolic Acid Derivative against CCl₄-Induced Hepatic Fibrosis in Rats.

A novel hepatoprotective oleanolic acid derivative, 3-oxours-oleana-9(11), 12-dien-28-oic acid (Oxy-Di-OA), has been reported. In previous studies, we found that Oxy-Di-OA presented the anti-HBV (Hepatitis B Virus) activity (IC50 = 3.13 µg/mL). Remarkably, it is superior to lamivudine in the inhibition of the rebound of the viral replication rate. Furthermore, Oxy-Di-OA showed good performance of anti-HBV activity in vivo. Some studies showed that liver fibrosis may affiliate with HBV gene mutations. In addition, the anti-hepatic fibrosis activity of Oxy-Di-OA has not been studied. Therefore, we evaluated the protective effect of Oxy-Di-OA against carbon tetrachloride (CCl4)-induced liver injury in rats. Daily intraperitoneally administration of Oxy-Di-OA prevented the development of CCl4-induced liver fibrosis, which was evidenced by histological study and immunohistochemical analysis. The entire experimental protocol lasted nine weeks. Oxy-Di-OA significantly suppressed the increases of plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels (p < 0.05). Furthermore, Oxy-Di-OA could prevent expression of transforming growth factor β1 (TGF-β1). It is worth noting that the high-dose group Oxy-Di-OA is superior to bifendate in elevating hepatic function. Compared to the model group, Oxy-Di-OA in the high-dose group and low-dose group can significantly reduce the liver and spleen indices (p < 0.05). The acute toxicity test showed that LD50 and a 95% confidence interval (CIs) value of Oxy-Di-OA were 714.83 mg/kg and 639.73–798.73 mg/kg via intraperitoneal injection in mice, respectively. The LD50 value of Oxy-Di-OA exceeded 2000 mg/kg via gavage in mice. In addition, a simple and rapid high performance liquid chromatography-ultraviolet (HPLC-UV) method was developed and validated to study the pharmacokinetic characteristics of the compound. After single-dose oral administration, time to reach peak concentration of Oxy-Di-OA (Cmax = 8.18 ± 0.66 μg/mL) was 10 ± 2.19 h; the elimination half-life and area under the concentration-time curve from t = 0 to the last time of Oxy-Di-OA was 2.19 h and 90.21 μg·h/mL, respectively.

Comparative pharmacokinetics of diaveridine in pigs and chickens following single intravenous and oral administration.

Comparative pharmacokinetic profiles of diaveridine following single intravenous and oral dose of 10mg/kg body weight in healthy pigs and chickens were investigated, respectively. Concentrations of diaveridine in plasma samples were determined using a validated high-performance liquid chromatography-ultraviolet (HPLC-UV) method. The concentration-time data were subjected to noncompartmental kinetic analysis by WinNonlin program. The corresponding pharmacokinetic parameters in pigs or chickens after single intravenous administration were as follows, respectively: t1/2β (elimination half-life) 0.74±0.28 and 3.44±1.07h; Vd (apparent volume of distribution) 2.70±0.99 and 3.86±0.92L/kg; ClB (body clearance) 2.59±0.62 and 0.80±0.14L/h/kg; and AUC0-∞ (area under the blood concentration vs. time curve) 4.11±1.13 and 12.87±2.60μg∙h/mL. The corresponding pharmacokinetic parameters in pigs or chickens after oral administration were as follows, respectively: t1/2β 1.78±0.41 and 2.91±0.57h; Cmax (maximum concentration) 0.43±0.24 and 1.45±0.57μg/mL; Tmax (time to reach Cmax ) 1.04±0.67 and 3.25±0.71h; and AUC0-∞ 1.33±0.55 and 9.28±2.69μg∙h/mL. The oral bioavailability (F) of diaveridine in pigs or chickens was determined to be 34.6% and 72.2%, respectively. There were significant differences between the pharmacokinetics profiles in these two species.

HPLC-UV Polyphenolic Profiles in the Classification of Olive Oils and Other Vegetable Oils via Principal Component Analysis

High performance liquid chromatography-ultraviolet (HPLC-UV) was applied to the analysis and characterization of olive oils and other vegetable oils. A chromatographic separation on a Zorbax Eclipse XDB-C8 reversed-phase column was proposed under gradient elution, employing 0.1% formic acid aqueous solution and methanol as mobile phase, for the determination of 14 polyphenols and phenolic acids, allowing us to obtain compositional profiles in less than 20 min. Acceptable sensitivity (limit of detection (LOD) values down to 80 µg/L in the best of cases), linearity (r2 higher than 0.986), good run-to-run and day-to-day precisions (relative standard deviation (RSD) values lower than 11.5%), and method trueness (relative errors lower than 6.8%) were obtained. The proposed HPLC-UV method was then applied to the analysis of 72 oil samples (47 olive oils and 27 vegetable oils including sunflower, soy, corn, and mixtures of them). Analytes were recovered using a liquid–liquid extraction method employing ethanol:water 70:30 (v/v) solution and hexane as extracting and defatting solvents, respectively. HPLC-UV polyphenolic profiles using peak areas were then analysed by principal component analysis (PCA) to extract information from the most significant data contributing to the characterization and classification of olive oils against other vegetable oils, as well as among Arbequina and Picual olive oil varieties. PCA results showed a noticeable difference between olive oils and the other classes. In addition, a reasonable discrimination of olive oils as a function of fruit varieties was also encountered.

Evaluation of Meropenem Penetration into Cerebrospinal Fluid in Patients with Meningitis After Neurosurgery

Meropenem is important for management of postneurosurgical meningitis, but the data about its penetration into cerebrospinal fluid (CSF) are inadequate. This prospective, open-label study investigated the pharmacokinetic profile of meropenem in patients with postneurosurgical meningitis, especially its CSF penetration. A total of 82 patients with postneurosurgical meningitis were included to receive meropenem intravenously according to a regimen of 2 g every 8 hours, 1 g every 8 hours, or 1 g every 6 hours. After infusion of 4 doses, blood and CSF samples were collected simultaneously at predefined time points. The high-performance liquid chromatography ultraviolet method was used to determine the concentration of meropenem. The peak meropenem concentration in blood and CSF was 43.2 ± 5.3 and 2.4 ± 0.3 mg/L in the group who received 2 g every 8 hours; 28.9 ± 2.7 and 1.2 ± 0.2 mg/L in the group who received 1g every 8 hours; and31.5 ± 3.4 and 1.6 ± 0.2 mg/L in the group who received 1g every 6 hours. The maximal percent penetration into CSF was 17.6% ± 7.3%, 14.3% ± 1.7%, and 30.9% ± 24.2%, respectively. Dosing regimens of meropenem 1 g every 6 hours and 2 g every 8 hours provided higher CSF penetration than 1 g every 8 hours. A higher dose and shorter dosing interval of meropenem may be more useful for clearance of pathogens.

A simple high-performance liquid chromatography for determining lapatinib and erlotinib in human plasma

Lapatinib and erlotinib are used for cancer treatment, showing large interindividual variability. Therapeutic drug monitoring may be useful for assessing the clinical outcomes and adverse events. A simple high-performance liquid chromatography ultraviolet method was developed for the determination of lapatinib and erlotinib in human plasma. An aliquot of plasma sample spiked with IS was treated with acetonitrile to precipitate the proteins. Lapatinib and erlotinib were separated on an octadecylsilyl silica-gel column using a mobile phase consisting of acetonitrile, methanol, water, and trifluoroacetic acid (26:26:48:0.1) pumped at a flow rate of 1.0 mL/min. The detection wavelength was set at 316 nm. The calibration curves for lapatinib and erlotinib were linear (r = 0.9999) in the range of 0.125-8.00 µg/mL. The extraction recoveries for both lapatinib and erlotinib at the plasma concentration of 0.125-8.00 µg/mL were higher than 89.9% with coefficients of variation (CVs) less than 3.5%. The CVs for intraday and interday assays of lapatinib and erlotinib were less than 5.1% and 6.1%, respectively. The present method can be used for blood concentration monitoring for lapatinib or erlotinib in exactly the same conditions.

Development and Validation of a Chromatographic Ultraviolet Method for the Simultaneous Quantification of Dolutegravir and Rilpivirine in Human Plasma.

We have developed and validated a high-performance liquid chromatographic method for the simultaneous quantification, in human plasma, of dolutegravir, a new human immunodeficiency virus (HIV) integrase inhibitor, and rilpivirine, a novel HIV nonnucleoside reverse transcriptase inhibitor. An internal standard (quinoxaline) was added to plasma aliquots (500 μL), and a simple solid-phase extraction procedure was applied. Chromatographic separation of the drugs and internal standard was achieved with a gradient of acetonitrile and acetate buffer, and with an analytical run time of 25 minutes using an XBridge C18 column. The column eluate was monitored at 260 nm for dolutegravir and the internal standard and at 305 nm for rilpivirine. The method was linear in the range of 20-8000 and 20-2000 ng/mL for dolutegravir and rilpivirine, respectively (mean r ≥ 0.993 on 10 replicates for both analytes). Mean intraday and interday precision and inaccuracy were <15% for both compounds. The mean recovery was 73% and 80% for dolutegravir and rilpivirine, respectively. The high-performance liquid chromatography-ultraviolet method we developed showed a good analytical performance required for therapeutic drug monitoring of antiretrovirals, leading to potential improvements in HIV-infected patient care and laboratory management.

Isolation of Nicotinic Acid (Vitamin B3) and N-Propylamine after Myosmine Peroxidation.

The alkaloid myosmine (3-(1-pyrroline-2-yl)pyridine) is widespread in biological matrixes including foodstuffs and tobacco products. Some in vitro tests in cellular systems showed mutagenic activity for myosmine. Myosmine activation including peroxidation mechanism employs unstable oxazirane intermediates. The formation of minor metabolite 3-hydroxymethyl-pyridine in rat metabolism experiments as well as in in vitro peroxidation assays suggests its further oxidation to nicotinic acid and possible concomitant formation of n-propylamine. A sensitive high-performance liquid chromatography-ultraviolet (HPLC-UV) method was developed for the direct analysis of n-propylamine in the peroxidation assay solution of myosmine employing derivatization with 3,5-dinitrobenzoyl chloride. Additionally, during peroxidation procedures, formation of 3-pyridylmethanol to nicotinic acid, the essential vitamin B3, was observed and characterized using HPLC-UV and gas chromatography/mass spectrometry. This new reaction pathway may present further contribution to our knowledge of myosmine's significance in human food including its activation in human organism, foodstuffs, and biological systems.

A simple HPLC-UV method for the quantification of theophylline in rabbit plasma and its pharmacokinetic application.

A simple, precise and accurate high-performance liquid chromatography-ultraviolet method was developed and validated for the quantification of theophylline in rabbit plasma using hydroxyethyl theophylline as an internal standard. Separation was performed on Waters(®) C18 column (µBondapak™ 5 µm, 150 × 3.9 mm) using a mobile phase consisting of water-acetonitrile (96:4 v/v) at a flow rate of 1 mL/min. Validation of the method was performed in order to demonstrate its selectivity, linearity, precision, accuracy and stability. The calibration curves of theophylline were linear over a concentration range of 0.1-25 µg/mL. The within- and between-day coefficient of variation (CV) were <10%. The extraction recoveries of theophylline at the three levels of quality control samples were 63.1, 69.4 and 69.7%. The method was rapid with retention time of theophylline and the internal standard observed at ∼5.2 and 6.5 min, respectively. The developed method was applied successfully for studying the pharmacokinetics of theophylline in rabbits.

Development and validation of an HPLC-UV method for pazopanib quantification in human plasma and application to patients with cancer in routine clinical practice.

Pazopanib, a new oral angiogenesis inhibitor, has demonstrated clinical activity against multiple solid tumors and was approved for the treatment of patients with advanced renal cell carcinoma. As an exposure-response relationship has been observed for pazopanib, its therapeutic drug monitoring could be a valuable tool in clinical practice. Therefore, the aim of this study was to develop and validate a selective and precise high performance liquid chromatography-ultraviolet method for the measurement of pazopanib in plasma from patients with cancer. After liquid-liquid extraction with diethyl ether, pazopanib and gefitinib (internal standard) were separated using isocratic elution on an Ultrabase C18 column using a mobile phase consisting of a mixture in vol/vol proportion of 47:53 of ammonium acetate (pH, 7; 0.02 mol/L) and acetonitrile/methanol (70:30, vol/vol) pumped at a constant flow rate of 1 mL/min. Quantification was performed at 260 nm. Method validation was undertaken as per the guidelines for Bioanalytical Method Validation published by the Food and Drug Administration and European Medicines Agency. Calibration curves were linear over the range 0.5-100 mcg/mL. Interday and intraday coefficients of variations were less than 4.5%. The limit of detection and the lower limit of quantification were 0.2 and 0.5 mcg/mL, respectively. Recovery of pazopanib from plasma was >80%. This is the first high performance liquid chromatography-ultraviolet method for pazopanib quantification that has been validated within a wide range of plasma concentrations and is a suitable method for therapeutic drug monitoring of pazopanib.

Quantification and localization of hesperidin and rutin in Citrus sinensis grafted on C. limonia after Xylella fastidiosa infection by HPLC-UV and MALDI imaging mass spectrometry

A high performance liquid chromatography-ultraviolet (HPLC-UV) method was developed for quantifying hesperidin and rutin levels in leaves and stems of Citrus limonia, with a good linearity over a range of 1.0–80.0 and 1.0–50.0μgmL−1 respectively, with r2>0.999 for all curves. The limits of detection (LOD) for both flavonoids were 0.6 and 0.5μgmL−1, respectively, with quantification (LOQ) being 2.0 and 1.0μgmL−1, respectively. The quantification method was applied to Citrus sinensis grafted onto C. limonia with and without CVC (citrus variegated chlorosis) symptoms after Xylella fastidiosa infection. The total content of rutin was low and practically constant in all analyses in comparison with hesperidin, which showed a significant increase in its amount in symptomatic leaves. Scanning electron microscopy studies on leaves with CVC symptoms showed vessel occlusion by biofilm, and a crystallized material was noted. Considering the difficulty in isolating these crystals for analysis, tissue sections were analyzed by matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) to confirm the presence of hesperidin at the site of infection. The images constructed from MS/MS data with a specific diagnostic fragment ion (m/z 483) also showed higher ion intensities for it in infected plants than in healthy ones, mainly in the vessel regions. These data suggest that hesperidin plays a role in the plant-pathogen interaction, probably as a phytoanticipin. This method was also applied to C. sinensis and C. limonia seedlings, and comparison with the graft results showed that the rootstock had an increased hesperidin content ∼3.6 fold greater in the graft stem than in the stem of C. sinensis seedlings. Increase in hesperidin content by rootstock can be related to induced internal defense mechanisms.

Development of Validated Bioanalytical HPLC-UV Method for Simultaneous Estimation of Amlodipine and Atorvastatin in Rat Plasma.

A simple, economical and robust analytical high-performance liquid chromatography-ultraviolet method was developed and validated for simultaneous chromatographic elution of two cardiovascular drugs viz. amlodipine and atorvastatin in biological fluid for the first time. Only two liquid chromatography-mass spectrometry/mass spectrometry methods are available in literature for quantitation of selected pair of analytes. The bioanalytical method was developed in rat plasma by using Thermo beta-basic C18 (100×4.6 mm, 5 μm) and mobile phase was composed of dibasic phosphate buffer (pH 3.0):acetonitrile in the ratio of 55:45 at a flow rate of 1 ml/min with ultraviolet detection monitored at 240 nm. The selected chromatographic conditions were found to effectively separate amlodipine (5.1 min) and atorvastatin (12.1 min). The parametric statistics,i.e. correlation coefficient of 0.999, was assessed for both the drugs having linearity over the tested concentration range (0.05 to 10.0 μg/ml) in rat plasma using an unweighted calibration curve. The mean recovery (%) was more than 92.8% for both the drugs using protein precipitation method. The accuracy of samples for six replicate measurements at lower limit of quantitation level was within limit. The method was validated and was successfully applied to the nonclinical pharmacokinetic study of combination tablets containing amlodipine and atorvastatin in six Sprague Dawley rats.

HPLC method for simultaneous quantitative detection of quercetin and curcuminoids in traditional chinese medicines.

Objectives:Quercetin and curcuminoids are important bioactive compounds found in many herbs. Previously reported high performance liquid chromatography ultraviolet (HPLC-UV) methods for the detection of quercetin and curcuminoids have several disadvantages, including unsatisfactory separation times and lack of validation according the standard guidelines of the International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use.Methods:A rapid, specific, reversed phase, HPLC-UV method with an isocratic elution of acetonitrile and 2% v/v acetic acid (40% : 60% v/v) (pH 2.6) at a flow rate of 1.3 mL/minutes, a column temperature of 35°C, and ultraviolet (UV) detection at 370 nm was developed. The method was validated and applied to the quantification of different types of market available Chinese medicine extracts, pills and tablets.Results:The method allowed simultaneous determination of quercetin, bisdemethoxycurcumin, demethoxycurcumin and curcumin in the concentration ranges of 0.00488 ─ 200 μg/mL, 0.625 ─ 320 μg/mL, 0.07813 ─ 320 μg/mL and 0.03906 ─ 320 μg/mL, respectively. The limits of detection and quantification, respectively, were 0.00488 and 0.03906 μg/mL for quercetin, 0.62500 and 2.50000 μg/mL for bisdemethoxycurcumin, 0.07813 and 0.31250 μg/mL for demethoxycurcumin, and 0.03906 and 0.07813 μg/mL for curcumin. The percent relative intra day standard deviation (% RSD) values were 0.432 ─ 0.806 μg/mL, 0.576 ─ 0.723 μg/mL, 0.635 ─ 0.752 μg/mL and 0.655 ─ 0.732 μg/mL for quercetin, bisdemethoxycurcumin, demethoxycurcumin and curcumin, respectively, and those for intra day precision were 0.323 ─ 0.968 μg/mL, 0.805 ─ 0.854 μg/mL, 0.078 ─ 0.844 μg/mL and 0.275 ─ 0.829 μg/mL, respectively. The intra day accuracies were 99.589% ─ 100.821%, 98.588% ─ 101.084%, 9.289% ─ 100.88%, and 98.292% ─ 101.022% for quercetin, bisdemethoxycurcumin, demethoxycurcumin and curcumin, respectively, and the inter day accuracy were 99.665% ─ 103.06%, 97.669% ─ 103.513%, 99.569% ─ 103.617%, and 97.929% ─ 103.606%, respectively.Conclusion:The method was found to be simple, accurate and precise and is recommended for routine quality control analysis of commercial Chinese medicine products containing the flour flavonoids as their principle components in the extracts.

Quality assessment of Fritillariae Thunbergii Bulbus sold in Taiwan markets using a validated HPLC-UV method combined with hierarchical clustering analysis

The present paper describes the development of a high performance liquid chromatography-ultraviolet (HPLC-UV) detection method for quantitative determination of peimine and peiminine in Fritillariae Thunbergii Bulbus (FTB). Separation was achieved using a conventional XBridge™ Shield RP 18 column (250 mm × 4.6 mm, internal diameter 3.5 μm) with photodiode array detection at 190–400 nm for UV spectra and 220 nm for quantification. The mobile phase consisted of (A) 0.03% diethylamine aqueous solution and (B) acetonitrile eluted by an isocratic procedure at 45:55 (A:B) over 25 minutes. The method was validated for linearity, limits of detection (LOD) and quantification (LOQ), inter- and intra-day precisions, repeatability, stability, and recovery. All the validation results were satisfactory. The developed method was then applied to assay the contents of the two chemical markers in all the FTB samples collected. Based on the contents of the two analytes, hierarchical clustering analysis (HCA) was performed to reveal the similarities and differences of the samples.

Our aim was to evaluate DFX plasma concentrations according to single-nucleotide polymorphisms in genes involved in its metabolism (UGT1A1, UGT1A3, CYP1A1, CYP1A2 and CYP2D6) and elimination (MRP2 and BCRP1). Further aim was to define a plasma concentration cutoff value predicting an adequate response to therapy. Plasma concentrations were determined at the end of dosing interval (C trough) using an high-performance liquid chromatography-ultraviolet method. Allelic discrimination was performed by real-time PCR. C trough levels were influenced by UGT1A1C>T rs887829, CYP1A1C>A rs2606345, CYP1A2A>C rs762551, CYP1A2C>T rs2470890 and MRP2G>A rs2273697 polymorphisms. A DFX plasma efficacy cutoff value of 20,000 ng ml(-1) was identified; CYP1A1C>A rs2606345 AA and CYP1A2C>T rs2470890 TT genotypes may predict this value, suggesting a negative predictive role in therapy efficacy. Our data suggest the feasibility of a pharmacogenetic-based DFX dose personalization.

Tissue depletion of quinocetone and its five major metabolites in pigs, broilers, and carp fed quinocetone premix.

A residue depletion study was performed to investigate the tissue kinetics of quinocetone (1) and its major metabolites. Quinocetone and its major metabolites were simultaneously quantitated with a high-performance liquid chromatography-ultraviolet (HPLC-UV) method. A total of 25 pigs, 30 broilers, and 50 carp were fed 100 mg/kg quinocetone for 90, 42, and 60 days, respectively. Liver, kidney, muscle, and fat (skin) tissues were collected at five different withdrawal times for analysis. Results revealed that quinocetone, 1-desoxyquinocetone (2), carbonyl-reduced 4-desoxyquinocetone (4), 3-methylquinoxaline-2-carboxylic acid (5), and carbonyl-reduced dideoxyquinocetone (6) could be depleted quickly in tissues; by contrast, dideoxyquinocetone, 3, persisted for a long time in the liver. Therefore, the liver is possibly the target tissue of quinocetone, and 3 is the residual marker; the recommended withdrawal times (WDTs) are 0 days in pigs and carp and 3 days in broilers. These results provided clear monitoring tools and technical standards to evaluate the food safety of quinocetone.
