Proximate, elemental and anti-nutritional composition of <i>Pachycarpus bisacculatus</i> (bitter butter) roots used locally as anti-snake venom

Snake bite remains a public health problem in many countries including Nigeria; hence search for snake antivenom has intensified. One plant that is potentially useful in this regard is Pachycarpus bisacculatus roots (bitter butter). This study determined; proximate, phytochemical, of the aqueous, n-hexane, methanol and ethyl acetate extracts of Pachycarpus bisacculatus roots. These extracts obtained by maceration were subjected to qualitative and quantitative phytochemical screening. Proximate, elemental, anti-nutritional compositions of this plant were carried out using standard methods. Results obtained were: moisture content (71.56±0.06, 7.53±0.37)%, crude protein (3.19±0.19, 1.92±0.05)%, crude fibre (11.33±0.31, 3.98±0.28)%, crude fat (1.05±0.07, 0.30±0.03)%, Ash (4.68±0.17, 2.32±0.32)%, carbohydrate (72.21±0.24, 19.92±0.48)% for the dried and fresh roots respectively. Elemental content showed that level of Ca was highest while that of Zn was the lowest. Al, Si, V, Pb were not detected. Anti-nutritional composition showed: oxalate (1.68±0.02 mg/100g), tannins (7.10±0.78 mg/100g), phytate (8.47±0.25 mg/100g) and cyanogenic glycosides (0.03±0.01 mg/100g). Extraction yields were 21.068% in aqueous solution, 1.6391% in ethyl acetate. The results indicate that root extracts of Pachycarpus bisacculatus contain bioactive chemicals and micronutrients which may be responsible for the medicinal properties reported for the plant.

Evaluation of antimicrobial activity and bioactive compound analysis of Verbascum thapsus L. A folklore medicinal plant

BackgroundThe prevalence of microbial infectious diseases is a major contributor to morbidity and mortality on a global scale due to the emergence of antibiotic-resistant microbes, despite the fact that many new antibiotics have been introduced for several decades. In present scenario, the potential utilization of natural plant derived extracts for medicinal and therapeutic purposes has increased remarkably due to adverse effects of synthetic drugs and resistance among pathogenic microbes. Aim of the studyVerbascum thapsus L. (Common mullein) has been traditionally used as a medicine for lung, skin and throat and many other disorders since time immemorial, therefore, the current study was aimed to evaluate the antimicrobial activity of leaf and root extracts of Verbascum thapsus against pathogenic bacterial and fungal strains and its phytochemical composition by spectral studies. MethodsSoxhlet extraction method was employed for extraction process by using varying polarity solvents like hexane, ethyl acetate and methanol. Phytochemical analysis of methanol extract was determined by Fourier Transform-Infrared Spectroscopy (FTIR) and Gas Chromatography-Mass Spectrometry (GC–MS). Antimicrobial activity was evaluated by determining the MIC and MBC/MFC of each extract by broth dilution method. ResultsThe antimicrobial competence of each extract sample was evaluated against six pathogenic bacterial strains and three fungal strains. Methanolic extract of leaves and the ethyl acetate extract of roots of Verbascum thapsus exhibited significant zones of inhibition against all the tested microorganisms. The highest inhibition zone was observed against the bacterial strain Bacillus subtilis (24.1±0.18 mm) followed by Streptococcus pyogenes (22.9±0.64 mm) and the fungal strain Aspergillus niger (22.8±0.55 mm) at a concentration of 500 μg/ml. Methanolic leaf extract strongly inhibited the growth of tested microbial strains with MIC ranged from 15.625 to 125 μg/ml. The FTIR and GCMS analysis confirms the presence of bioactive compound, possessing broad range of therapeutic activities. ConclusionThis Presence of various important phytoconstituents and broad-spectrum antimicrobial activity points Verbascum thapsus as a promising source for the isolations of molecules responsible for antimicrobial activities.

Phytochemical analysis of root extracts of Rhynchostylis retusa (L.) Blume from the Eastern Ghats of India

Rhynchostylis retusa (Orchidaceae) is an important ethnomedicinal herb in Indian systems of medicine. Tribal healers commonly employ the roots of this epiphytic orchid to treat various ailments. In the present study, the shade-dried root powder of R. retusa was subjected to cold extraction with four solvents, such as n-hexane, acetone, ethyl acetate, and methanol. The crude root extracts were then taken for qualitative phytochemical screening. Furthermore, GC-MS analysis of ethyl acetate and methanol root extracts was carried out. The methanol root extract of R. retusa showed a positive result for all the phytochemicals tested except for saponins. GC-MS analysis of R. retusa methanol root extract revealed the presence of 16 phytoconstituents. Major phytoconstituents such as 9-Hexadecenoic acid, methyl ester, [Z] Heptadecanoic acid, 16-methyl-, methyl ester, Ethanone,1-[4-(4-morpholyl benzylidene amino)phenyl]-, are recorded in the methanol root extract. In the present study, both methanol and ethyl acetate root extracts showed the presence of9-Hexadecenoic acid, methyl ester, [Z] as the major phytocompound. The phytocompounds identified in methanol and ethyl acetate root extracts exhibit various biological activities, including anti-inflammatory, antibacterial, antifungal, antioxidant, and anticancer properties. Based on the findings of the current study, it can be inferred that the roots of R. retusa contain diverse bioactive compounds with medicinal properties. Further investigation of these R. retusa root extracts holds the potential for the discovery and development of innovative pharmaceuticals.

Phytochemical analysis and high‐performance thin‐layer chromatography guided quantification of Boeravinone B from Boerhaavia diffusa root extracts: Assessment of in vitro safety analysis

AbstractAn Indian traditional medicinal plant, Boerhaavia diffusa (Punarnava), is reported for its diuretic activity and helps in impaired kidney function. The present study indicates that alkaloids, carbohydrates, saponins, phytosterols, flavonoids, and glycosides are absent in the petroleum ether extract and present chiefly in the methanolic extract of roots. The phytosterols were found to be present in the ethyl acetate extract, while other phytochemical constituents, that is, alkaloids, carbohydrates, saponins, flavonoids, and glycosides, were absent. From high‐performance thin‐layer chromatography, the results indicated that out of all the three extracts and two marketed formulations, ethyl acetate root extract (LEA) has the highest amount of Boeravinone B content. Further, different root extracts of B. diffusa and two marketed formulations were evaluated on the human embryonic kidney cell line to understand pharmacological safety. From the results, it was observed that LEA and petroleum ether root extracts showed a higher percentage of cell survival and a better safety profile up to the dose of 1 mg/mL. The overall results indicated that further preclinical evaluation of B. diffusa is needed to understand the mechanism behind nephroprotective potential which can further expedite the drug discovery process of nephroprotective molecules.

Evaluation of the Capparis Herbacea Willd's Chemistry, Antioxidant and Cytotoxic Activity.

The Capparidaceae family includes the medicinal herb Capparis herbacea Willd. The aerial and underground parts of plant C.herbacea were studied for their chemical composition, antioxidant, and cytotoxic properties. Using gas chromatography with mass spectrometric detection (7890A/5975C), 94 chemicals were identified in ethanol extract from leaves, roots, seeds, and stems of C. herbacea. Main components were (leaves) phytol 18.16%, hexanedioic acid, bis(2-ethylhexyl) ester 16.75%, vitamin E 11.95%, (roots) sucrose 13.94%, hexadecanoic acid, ethylester 22.80%, octadecanoic acid, ethylester 37.77%; (seeds) hexadecanoic acid, ethylester 13.96%, ethyl9.cis.,11.trans.-octadecadienoate 48.54%, bis(2-ethylhexyl) phthalate 9.77%; (stems) 1-propene-1,2,3-tricarboxylic acid, tributyl ester 42.69%, and tributylacetylcitrate 19.63%. Nine components were identified in the makeup of the C. herbacea sample's essential oil using the method of chromatography-mass spectrometry. The main components were (in%): T-cadinol (29.56), meta-cymene (16.12), pulegone (14.11), and σ-amorphene (12.26). Chloroform and methanol extracts of Capparis herbacia roots at concentrations of 1 mg/ml showed higher average antioxidant activity, while ethyl acetate root extract at concentrations of 0.75 and 1 mg/ml showed higher average antioxidant activity compared to gallic acid AOA. In addition, plant extracts have cytotoxic activity. Essential oils of leaves and stems, fruit and roots of Capparis herbacia plants exhibited cytotoxicity, all larvae died, and larval mortality was 96%.

In silico and In vivo Studies of Calotropis procera leaf and root extracts on Mitochondrial-Related parameters in Wistar Rats

Drugs targeting Mitochondrial Membrane Permeability Transition (MMPT) pore opening are of great interest for conditions arising from apoptosis dysregulation. This study investigate MMPT pore inducing effect of Calotropis procera leaves and root extracts on mitochondrial-related parameters in the liver of healthy male Wistar rats. Extraction was done by standard methods, using Ethylacetate and Butanol solvent. Fifty–two rats weighing between 130 g - 150 g were divided into thirteen groups (n=4). Group 1 received distilled water, other groups were administered ethylacetate leaf extract (ELE), butanol leaf extract (BLE), ethylacetate root extract (ERE) and butanol root extracts (BRE). Liver Mitochondrial pore opening, mitochondrial ATPase activity and lipid peroxidation were assessed spectrophotometrically, while the interaction between human Cyclophilin D (a pore activating protein with PDB ID: 2BIT) and identified phytochemicals of Calotropis procera leaves and roots extracts were studied In-silico. Varying concentrations of the extracts induced MMPT pore opening by 6.6, 7.4 and 5.9 folds for BLE and 10.2, 7.3 and 6.4 folds for ELE at 40, 50 and 60 mg/100g bw respectively when compared with control group. Root extracts showed significant MMPT pore opening with a fold increase of 12.2, 10.1 and 20.4 for BRE and 11.37, 11.84, for ERE at at 40 , 50 and 60 mg/100g bw respectively. BLE, BRE, ELE and ERE showed increase in ATPase activities with respect to the control group. Malondialdehyde (MDA) levels as an indication of lipid peroxidation increased significantly when compared with control. Preliminary Assessment of Phytochemicals and Free Radical Scavenging Activity of Different Plants Collected from the Western Region of Nepal

Medicinal plants produce vital secondary metabolites in curing various diseases and pathological conditions. There are multiple tribes and communities where people are using plants from the medicinal aspect to cure different diseases. The aim of the present study was qualitative and quantitative phytochemical screening, determination of total phenol and total flavonoid content, and free radical scavenging activity of different parts of ethnomedicinally used plants collected from the western region of Nepal. The phytochemical screening of most of the extract showed the presence of phenols, carbohydrates, flavonoids, and glycosides. The ethanolic flower extract of Callicarpa macrophylla showed a higher phenolic content with a value of 195 ± 7.33 µg GAE/mg. Ethyl acetate extract of Elephantopus scaber root revealed the highest amount of flavonoid content with the value of 1692.31 ± 34.79 µg QE/mg of extract, followed by ethanolic flower extract of Callicarpa macrophylla. Ethanolic flower extract of Callicarpa macrophylla showed potent DPPH free radical scavenging activity with a half inhibitory concentration of 7.28 µg/ml. In contrast, Elephantopus scaber ethyl acetate root extract revealed the maximum free extreme scavenging properties among all the ethyl acetate extracts. From the experiment, the free radical scavenging potential of ethanolic flower extract of the Callicarpa macrophylla was comparable with ascorbic acid. This comparable activity may be attributed to higher phenols and flavonoid content in ethanolic extract. The result signifies that the ethanolic extract of Callicarpa macrophylla acts as an antioxidant and may contain a potent bioactive compound which, with further investigation, may lead to a novel compound.

A New Saponin (Zygo-albuside D) from Zygophyllum album Roots Triggers Apoptosis in Non-Small Cell Lung Carcinoma (A549 Cells) through CDK-2 Inhibition.

Phytochemical study of the ethyl acetate root extract of Zygophyllum album has resulted in the isolation of a new saponin, Zygo-albuside D (1), along with two known compounds; (3-O-[β-D-quinovopyranosyl]-quinovic acid) (2), which is first reported in the root, and catechin (3), first reported in the genus. Their chemical structures were established by NMR and high-resolution mass spectrometry (HRMS). The new saponin (1) exhibited promising cytotoxicity with IC50 values of 3.5 and 5.52 μM on A549 and PC-3 cancer cell lines, respectively, compared to doxorubicin with IC50 values of 9.44 and 11.39 μM on A549 and PC-3 cancer cell lines, respectively. While it had an IC50 value of 46.8 μM against WISH cells. Investigating apoptosis-induction, compound 1 induced total apoptotic cell death in A549 lung cancer cells by 32-fold; 21.53% compared to 0.67% in the untreated control cells. Finally, it upregulated the pro-apoptotic genes and downregulated the antiapoptotic gene using gene expression levels. Compound 1 exhibited remarkable CDK-2 target inhibition by 96.2% with an IC50 value of 117.6 nM compared to Roscovitine. The molecular docking study further confirmed the binding affinity of compound 1 as CDK2 and Bcl2 inhibitors that led to apoptosis induction in A549 cancer cells. Hence, this study highlights the importance of compound 1 in the design of a new anticancer agent with specific mechanisms.

A Comparative Study of Dichloromethane and Ethyl Acetate Root Extracts of <i>Celosia trigyna</i>: Phytochemical and Wound Healing Effect Analyses

Background: <i>Celosia</i> species have been validated to possess antimicrobial, anti-inflammatory, and antioxidant potential. This study was thus designed to comparatively determine the effect of dichloromethane and ethyl acetate root extracts of <i>Celosia trigyna</i> on Wistar rat wounds created by 8 mm biopsy punches.Methods: Phytochemical screening was performed on dichloromethane and ethyl acetate root extracts of <i>C. trigyna</i>. For animal studies, 25 adult female Wistar rats were randomly divided into five groups (group A-E; n=5). Group A served as the negative control, group B as a positive control, group C received dichloromethane extract (once/day), group D ethyl acetate extract (once/day), and group E was the vehicle control. All extracts and medication were administered topically. Wound area, wound contraction, period of epithelialization, macroscopic analysis, and histological examination were studied.Results: Phytochemical screening showed that saponins and terpenoids were particularly high in dichloromethane extracts while ethyl acetate extracts were highly rich in terpenoids. The results of wound areas, wound contraction, periods of epithelialization, and macroscopic and histological analyses showed that dichloromethane extract, ethyl acetate extract, and penicillin-treated groups showed better remediation respectively in order of efficacy compared to the negative and vehicle control groups. The histological examination revealed an evidence of tissue regeneration and collagen deposition in the extract-treated groups compared to the positive, negative, and vehicle control groups.Conclusion: In conclusion, both extracts of <i>C. trigyna</i> have a better healing potential compared to penicillin treatment with dichloromethane extract having a better wound healing potential than the ethyl acetate extract.

Nematicidal Coumarins from Cnidium monnieri Fruits and Angelica dahurica Roots and Their Physiological Effect on Pine Wood Nematode (Bursaphelenchus xylophilus).

Pine wood nematode (PWN), Bursaphelenchus xylophilus, is a major pathogen of pine wilt disease (PWD), which is a devastating disease affecting pine trees. Eco-friendly plant-derived nematicides against PWN have been considered as promising alternatives to control PWD. In this study, the ethyl acetate extracts of Cnidium monnieri fruits and Angelica dahurica roots were confirmed to have significant nematicidal activity against PWN. Through bioassay-guided fractionations, eight nematicidal coumarins against PWN were separately isolated from the ethyl acetate extracts of C. monnieri fruits and A. dahurica roots, and they were identified to be osthol (Compound 1), xanthotoxin (Compound 2), cindimine (Compound 3), isopimpinellin (Compound 4), marmesin (Compound 5), isoimperatorin (Compound 6), imperatorin (Compound 7), and bergapten (Compound 8) by mass and nuclear magnetic resonance (NMR) spectral data analysis. Coumarins 1-8 were all determined to have inhibitory effects on the egg hatching, feeding ability, and reproduction of PWN. Moreover, all eight nematicidal coumarins could inhibit the acetylcholinesterase (AChE) and Ca2+ ATPase of PWN. Cindimine 3 from C. monnieri fruits showed the strongest nematicidal activity against PWN, with an LC50 value of 64 μM at 72 h, and the highest inhibitory effect on PWN vitality. In addition, bioassays on PWN pathogenicity demonstrated that the eight nematicidal coumarins could effectively relieve the wilt symptoms of black pine seedlings infected by PWN. The research identified several potent botanical nematicidal coumarins for use against PWN, which could contribute to the development of greener nematicides for PWD control.

Comparative Evaluation of the Phytochemical Contents, Antioxidant and some Biological Activities of Khaya grandifoliola Methanol and Ethyl Acetate Stem Bark, Root and leaf Extracts

Khaya grandifoliola extracts have been conventionally found to be effective in the treatment of wounds and ulcers. This study assessed the phytochemical content of K. grandifoliola methanol and ethyl acetate extracts and used in vitro bioassays for bioactivity testing to confirm the ethnomedicinal uses. Phytochemical content and bioactivity of K. grandifoliola ethyl acetate and methanol extracts; (stem bark, root, leaf) were assessed using spectrophotometric analysis with standard calibration curves for respective phytochemicals and in vitro bioactivity assays including MTT, free radical scavenging activity, lipopolysaccharide-activated RAW 264.7 macrophages assay for cytotoxicity, anti-inflammatory antioxidant, and antimicrobial activities. The polyphenolic content of methanol extracts (37.49±1.40 -53.57±1.50 mg GAEQ/100 mg) was the highest phytochemical content measured. The methanol root extract had the highest terpenoid concentration (35.78±2.14 mg LIN EQ/100 mg). Methanol stem bark and root extracts exhibited anti-inflammation activity at 200 μg/mL, potent antioxidant activity (IC50; 38.68±5.09 µg/mL and 46.34±9.56μg/mL), and growth inhibition against Staphylococcus aureus (MIC; 1 and 2 mg/mg). Ethyl acetate root and leaf extracts inhibited the growth of Streptococcus pyogenes (MIC; 0.25 mg/mL). In conclusion, the high phenolic and terpenoid content of extracts which correlated positively with their biological activities could be the contributing factor to the biological effects such as wound healing of the extracts.

Exploring the Phytochemical Profile and Biological Activities of Clerodendrum infortunatum.

Clerodendrum infortunatum (C. infortunatum), the hill glory bower, is reputed as the prodigious treasure for Indian folk medicine. The study has focused on exploring the phytochemistry and antitumor potential of the C. infortunatum root extract in vitro and in vivo. The ethyl acetate root extract has demonstrated the highest cytotoxicity in a series of nine human tumor cell lines. Further fractionation of the same has yielded seven compounds. The structures of these compounds were confirmed with spectroscopic techniques. Considering the toxicity observed with the crude extract, cytotoxicity of these compounds was further assessed in two breast carcinoma cell lines (MCF-7[ER/PR-positive HER2-negative] and MDA-MB-231 [ER/PR/HER2-negative]) and in two cervical cancer [human papilloma virus (HPV)-negative C33A and HPV-positive SiHa] cell lines. Betulinic acid (BA) was found as the active principle contributing the cytotoxic activity, and cervical cancer cell lines documented the minimum IC50 value in 24 h. In order to validate the in vitro experimental data, we have established a xenograft model of HPV-positive cervical cancer in female NOD/SCID mice treated with BA using doxorubicin as the positive control. BA treatment gradually reduced the tumor size, maintaining healthy hematological and biochemical parameters, and improved the survival rate of tumor-bearing mice considerably. Thus, our findings suggest that the C. infortunatum root extract has a promising anticancer property against HPV-positive cervical cancer and supports its usage by traditional healers for treating cervical cancer.

GC-MS Analysis and Antioxidant Evaluation of Ativisa Root Extract

Plants provide an unlimited source of bioactive metabolites that have been utilized in the pharmaceutical industry as a source of diet and medication. Free radicals are known to be the major aspect of oxidative stress that creates damage to the cell. The natural sources of antioxidants which preventing and repairing the damage caused by the free radicals to the cells. Aconitum heterophyllum (Ativisa) belongs to family Ranunculaceae is traditionally utilized externally as well as internally for treating various human diseases such as rheumatic fever, painful joints, gastroenteritis, diarrhoea, oedema, Bronchial asthma. In the present study ethanolic and ethyl acetate extract of the roots of Acontium heterophyllum was investigated for its antioxidant activity and phytochemical composition using GC-MS. GC-MS studies exhibited the existence of 24 phytochemical compounds in ethanol root extract and 31 phytochemical compounds in ethyl acetate root extract. The major phytochemicals detected in ethanolic and ethyl acetate roots extracts of Acontium heterophyllum were Alpha-D-Glucopyranoside beta-D-Fructofuranosyl, Benzophenone semicarbazone, Dodecanoic acid ethyl ester, Ethyl iso-allocholate, l-(+)-Ascorbic acid 2,6 dihexadecanoate, Phytol, Methyl ricinoleate, 13-docosenamide(z), 1,2,4-Triazol-3-amine5-(1,3,5-trimethyl-4-pyrazolyl)amino. The antioxidant potency of ethanol and ethyl acetate root extracts of Acontium heterophyllum on DPPH (23.05% and 21.05 %) and NO radicals (48.58% and 46.70%) at the highest concentration (500 μg/ml) compared to standard ascorbic acid. These findings suggest Acontium heterophyllum could be used as an alternative source for the investigation of new antioxidant agents.

In vitro Evaluation of Cytotoxicity and Antibacterial Activities of Ribwort Plantain (Plantago Lanceolata L.) Root Fractions and Phytochemical Analysis by Gas Chromatography-Mass Spectrometry.

Ribwort plantain (Plantago lanceolata L.), which belongs to the Plantaginaceae family, has been widely used as a herbal plant in traditional medicine across the globe. The present study aimed to investigate the biologically active substances of P. lanceolata root fractions, as well as the cytotoxic and antibacterial activities of extracts. The cytotoxic activity of ethyl acetate, dichloromethane, and n-butanol extracts of P. lanceolata root was evaluated by MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay. The P.lanceolata root extracts were also evaluated on gram-positive and negative bacteria by disc diffusion and microtiter broth dilution methods. The phytochemical content was also examined by gas chromatography-mass spectrometry. The P.lanceolata root extracts were cytotoxic; IC50 values against HCT-116 at 72 h were 168.553 μg/mL, 167.458 μg/mL, and 205.004 μg/mL for ethyl acetate, dichloromethane, and n-butanol root extracts, respectively. The dichloromethane extract of P. lanceolata root had the highest inhibitory effect against S. paratyphi (14.00±1.0 mm) at the concentration of 100 mg/mL. The minimum MIC and MBC (5 and 15 mg/mL) were observed for dichloromethane extract of P. lanceolata root against S. paratyphi. The main composition of ethyl acetate extract was 1,2-Benzenedicarboxylic acid and mono(2-ethylhexyl) ester (60.93%). The major compositions in dichloromethane and n-butanol extracts were 1,2-Benzenedicarboxylic acid, mono(2-ethylhexyl) ester (60.64%) and 2-Methyl-1-butanol (.+/-.)- (17.85%). As evidenced by the results of the present research, P. lanceolata extracts are a significant source of bioactive metabolites. Therefore, they can play a prominent role in the production of pharmaceutical materials.

In vitro and Insilico cytotoxicity activity of Aconitum Heterophyllum Phyto niosomes and its ethyl acetate root extract

Niosomes as a novel drug delivery system is a nonionic surfactant-based vesicle, have attracted much attention in pharmaceutical fields due to their excellent behavior in encapsulating both hydrophilic and hydrophobic agents. In the present study a comparative free radical scavenging and anti-proliferative potential of ethyl acetate root extract of Aconitum heterophyllum (EAEAH) and Aconitum heterophyllum roots loaded phyto-niosomes (nEAEAH) were investigated. Initially the EAEAH and nEAEAH were prepared and their antioxidant potential were studied in terms of DPPH, Super oxide, Hydroxyl radical scavenging by employing the In-vitro method. Assessment of anti-proliferative activity of these extracts was done using MCF-7 cancer cell lines (human breast cancer) by employing MTT assay and trypan blue exclusion assay. In-silico docking was performed using Auto Dock Vina software to identify the suitable antagonistic ligand which can inhibit the proteins BRCA1, BRCA2 and HER2 to exert the cytotoxic effect. In the present study a concentration dependent antioxidant, and cytotoxic activity was observed for both the extracts. IC50 values were calculated for MTT and trypan blue assay and the nEAEAH showed 41.71μg/ml, 49.36μg/ml respectively, whereas for EAEAH it was found to be 54.89 and 67.13μg/ml.

Comparative Evaluation of Biomedical Applications of Zinc Nanoparticles Synthesized by Using Withania somnifera Plant Extracts.

Green synthesis of metal nanoparticles is of great importance in the modern health care system. In this study, zinc nanoparticles (ZnONPs) were synthesized using leaf and root extracts of Withania somnifera using four different solvents. ZnONPs were characterized by UV-vis spectrophotometer with a range between 350–400 nm. Scanning electron microscope revealed spherical morphology with an overall size of 70–90 nm and XRD pattern confirmed the crystalline structure. The total flavonoids, phenolic, and alkaloid contents were significantly greater in the crude extracts as compared to ZnONPs. The highest scavenging activity was observed in ZnONPs from n-hexane and ethyl-acetate extracts of roots with IC50 values of 27.36 µg/mL and 39.44 µg/mL, respectively. ZnONPs from methanol and aqueous extracts showed significant antibacterial activity against Escherichia coli, Staphylococcus aureus, and Bacillus subtilis while none of the extracts were found to have significant antifungal activity. Maximum cytotoxic activity was observed in ZnONPs synthesized from aqueous and n-hexane root extracts with LC50 values of 9.36 µg/mL and 18.84 µg/mL, respectively. The highest antidiabetic potential was exhibited by ZnONPs from n-hexane leaf extracts, i.e., 47.67 ± 0.25%. Maximum protein kinase inhibitory potential was observed in ZnONPs of ethyl-acetate extract of roots with a bald zone of 12 mm. These results indicated that Withania somnifera-based ZnONPs showed significant biological activities compared to crude extracts. These findings can further be utilized for in-vivo analysis of nano-directed drug delivery systems.

Antimicrobial Activities of Salacia oblonga Wall Leaf and Root Extracts Against Different Bacterial Strains and Fungal Isolates.

Antibiotic resistance and the hazardous nature of synthetic drugs is threatening issue in the health sector. The alternative for this problem is to focus on plants that attribute to various compounds that exhibit therapeutic properties. Therefore, the study aims to evaluate the antimicrobial efficacy of Salacia oblonga leaf and root extracts against tested human pathogens. The S. oblonga extracts showed a significant zone of inhibition against bacteria and fungi. The leaf and root extracts of S. oblonga are prepared using low polar to high polar solvents in the Soxhlet apparatus and tested on the selected bacterial and fungal strains. Agar well diffusion and broth dilution methods evaluate antibacterial activity, antifungal activity, and Minimum Inhibitory Concentration (MIC) of extracts. Among the extracts tested, the ethyl acetate extract of root showed more antimicrobial activity against the tested bacterial and fungal strains. The most susceptible bacterial and fungal species against ethyl acetate extract are Micrococcus luteus, Mycobacterium tuberculosis, Microsporum canis, Trichophyton interdigitale, and Microsporum gypseum. The MIC for bacteria ranged from 13.0 to > 200µg/ml, whereas for fungi, the MIC ranged from 25.9 to > 200µg/ml. Ethyl acetate extract of root with 100µg/ml concentration showed 29.1mm and 28.7mm zone of inhibition against bacterial strains M. luteus and M. tuberculosis, respectively. The ethyl acetate extract of root with a 100µg/ml concentration showed 15.8, 15.2, and 15.6mm zone of inhibition against fungal isolates M. canis, T. interdigitale, and M. gypseum, respectively. The activity of root and leaf extracts increased in a concentration-dependent manner, and further, the compounds isolated from the crude extracts of leaf and root showed antimicrobial activity. Structural elucidation of isolated compounds Lambertic acid and Ferruginol was done using NMR spectroscopy. Reports indicate that Lambertic acid was isolated previously, but the isolation of hydroxy Ferruginol from S. oblonga leaf extract was reported unprecedented.

IN VITRO AND IN VIVO ANTIDIABETIC EFFECTS OF SALACIA OBLONGA WALL LEAF AND ROOT EXTRACTS

The present study was framed out to evaluate the antidiabetic effects of S. oblonga Wall leaf and root ethyl acetate and toluene extracts in vitro and in vivo. The root ethyl acetate extract found significant inhibition of alpha amylase and alpha glucosidase enzyme functions in vitro. The in vivo studies are carried out according to the guide line of CPCSEA and IAEC. The acute toxicity test of the extracts resulted in the no effect on the animals. The ethyl acetate extract of root showed the increase in body weight of animals at the end of the experiment. The blood glucose levels of diabetic rats are significantly reduced after the treatment of root ethyl acetate extract. The leaf toluene and ethyl acetate extract exhibited average antidiabetic activity. We conclude that root extract exhibited good antidiabetic activity comparing to leaf extract.

Susceptibility of Aedes albopictus Larvae to the Larvicidal Activity of Three Types of Derris elliptica Extract.

<b>Background and Objective:</b> The methanol, ethyl acetate and n-hexane extracts of <i>D. elliptica</i> root have high larvicidal activity against <i>Aedes aegypti</i> larvae, the primary vector of dengue but have not been understood their potential against <i>Ae. albopictus</i> larvae, the secondary vector of dengue that also transmits Chikungunya and Zika viruses. This <i>in vitro</i> study aims to understand the larvicidal activity of the 3 extract types of <i>D. elliptica </i>root against <i>Ae. albopictus</i> larvae. <b>Materials and Methods:</b> The tuba root extract types were obtained from the sequential extraction process with 3 steps of liquid-liquid partition as described in the previous report. Six concentrations were occupied in this experiment ranging of 0.5, 1.0, 2.0, 4.0, 10.0 and 15.0 mg L<sup>1</sup> each concentration was 5 times replicated and placed in 250 mL plastic cups. As many as 20 of 3rd instar larvae of <i>Ae. albopictus</i> were subjected in each treatment cup and larval mortality was observed after 24 and 48 hrs of exposure. <b>Results:</b> Larval mortality rates based on concentration range of 13.75-97.00 and 43,75-100%, 14.00-44.00, 34.00-90.00%, 12.00-47.00 and 28.00-88.00%, with the LC<sub>50</sub> after 24 and 48 hrs of exposure were 2.925 and 0.414, 16.184, 2.900, 15.789 and 4.380 mg L<sup>1</sup>, respectively for methanol, ethyl acetate and n-hexane extracts. <b>Conclusion:</b> The methanol, ethyl acetate and n-hexane extract of tuba root have high larvicidal activity against <i>Ae. albopictus</i> larvae. Further study on prototype formulation of larvicide and elucidation of the specific phytochemical compounds of the extracts were necessarily conducted.
