Abstract

AbstractThe nucleic-acid material in gelatin has been dctermined by hydrolysis and measurcment of thc liberated nucleic-acid bases, adenine, cytosine and thymine, using ion-exchange chromatography. The ratios of the three bases are not the same for all gelatins; neither are they as in native nucleic acid. This show that, in addition to molecular weight and conformation changes, the nucleic acid i altered chemically during gelatin manufacture. De-amination of the amino-bearing bases, especially cytosine, occurs in all gelatins. In addition, for gelalins derived from demineralized ossein, extensive de-purination occurs, as shown by the very low adenine/thymine ratios. The nucleic-acid material in inert ossein gelalins thus differs chemically from that in the active hide gelalins. As judged by lhymine determinations, all gelatins, including “empty” gelatins, contain DNA-derived material.
