A method for the spectrophotometric determination of a three-component system is presented. The method is applied to the simultaneous determination of carboxyhemoglobin and methemoglobin in human blood. The three-component system comprising oxyhemoglobin, carboxyhemoglobin and methemoglobin is determined by measuring the optical density at 4 wavelengths, using a layer thickness of 0.013 cm. Except for a drop of saponin solution to bring about hemolysis, no chemicals are added to the blood. A suitable method for calculating the results is described.
