In Vitro Probiotic Characterization of Lactiplantibacillus plantarum Strains Isolated from Traditional Fermented Dockounou Paste

This study aimed to evaluate the probiotic properties of 10 lactic acid bacteria (LAB) isolated from artisanal fermented plantain dockounou paste. A preliminary characterization of the LAB isolates was performed based on phenotypic and several biochemical properties, which was subsequently confirmed through 16S rRNA gene sequencing analysis, indicating that these isolates belonged to the species Lactiplantibacillus plantarum. With regard to safety criteria, the strains exhibited no alpha or beta hemolysis activity. Nevertheless, the majority of LAB strains demonstrated high sensitivity to the antibiotics tested. The results demonstrated that the majority of the strains exhibited remarkably high survival rates under simulated gastrointestinal conditions, such as pH = 1.5 (81.18–98.15%), 0.3% bile salts (68.62–100.89%), 0.4% phenol (40.59–128.24%), as well as 0.1% pepsin and pH = 2.5 (88.54–99.78%). The LAB strains demonstrated elevated levels of cell surface properties, indicative of the presence of a considerable defensive mechanism against pathogens. Intact LAB cells exhibited significant antioxidant abilities (48.18–83.58%). They also demonstrated a pronounced inhibitory effect on the growth of foodborne pathogens. Enzyme pattern analysis revealed that the LAB isolates produced both proteases and cellulases, as well as pectinase and/or amylase activity. The potential of the L. plantarum strains FS43, FS44, and FS48, as indicated by the results obtained from the standard in vitro assays, makes them suitable for further study as potential probiotics.

Fermentation of barley wort with Saccharomyces boulardii to generate a beverage with probiotic potential

Developing non-dairy functional beverages based on probiotics is a potential alternative for vegan consumers. It also becomes a big challenge since microorganisms can easily change chemical composition, acidity, color, and consumers’ acceptability. In this work, the probiotic yeast Saccharomyces boulardii was used to elaborate a fermented barley wort beverage. For the beverage formulations, the use of two flavors strawberry (SPB) and nutty (NPB) and two different inoculum concentrations (1 × 103 (SPB or NPB103) and 1 × 104 CFU/mL (SPB or NPB 104)) were considered. The fermentation was carried out at 37 °C under anaerobic conditions. The effect of the inoculum concentration was evaluated over reducing sugar consumption, the concentration of soluble solids, and pH. The best formulation was selected through a sensory evaluation. The profile of oligosaccharides and the resistance of the yeast to gastrointestinal conditions were determined only in the nutty flavor formulation (NPB 104). During the fermentation process, the content of oligosaccharides was increased compared to the unfermented wort, which improved the nutritional value. The analysis of resistance to simulated gastrointestinal conditions showed a decrease from 8.0 × 107 to 1.5 × 107 CFU/mL in the NPB 104 beverage. Barley wort is a potential medium to produce probiotic beverages with the yeast S. boulardii.

The effect of synbiotic coating of flaxseed mucilage‐defatted rice bran carbohydrate on quality of dried mango, viability of Bifidobacterium animalis subsp. LactisBB12 on storage and simulating gastrointestinal condition

AbstractIn the present study, a synbiotic coating of flaxseed mucilage, defatted rice bran carbohydrate, and Bifidobacterium animalis subsp. Lactis BB12 was fabricated for coating dried mango slices (M‐P‐C). The control samples contained only probiotic bacteria without coating (M‐P). Several quality parameters (moisture, weight loss, shrinkage percentage, pH, firmness, and color) were assessed on specific storage circumstances (25°C, relative humidity (RH) = 22%.). In addition, the survival of Bifidobacterium animalis subsp. Lactis BB12 was evaluated on storage and under simulated gastrointestinal (GI) conditions. According to the results, the log number of Bifidobacterium animalis subsp. Lactis BB12 reached 8.1 and 6.2 for coated and uncoated samples, respectively, during the 45 days storage at 25°C (>6 log CFU (log colony‐forming units)/g) and at finished stage of in vitro gastrointestinal circumstances, the log number of probiotic bacterial count reached 6.8 and 4 for coated and uncoated samples, respectively. The coating resulted in significantly less weight loss, moisture loss, and shrinkage of the mango slices than uncoated ones (p < .05). The growth of yeasts and molds was undetectable in both samples. The results of acceptance experiments for M‐P and M‐P‐C dried mango samples showedthat there were no significant differences between M‐P and M‐P‐C samples (p >.05), indeed in the case of purchase intention and overall acceptability. After reading the text highlighting, there was no significant difference in all attributes of M‐P‐C samples pre and post of reading text highlighting. It could be concluded that the synbiotic coating of mango slices improved the quality characteristics of the dried mango as well as viability of the probiotic bacteria at storage time and under simulated gastrointestinal conditions.

Effect of microencapsulation on the bio-preservative and probiotic properties of Enterococcus durans F21

This study aimed to assess the probiotic potential of Enterococcus durans F21 and its microencapsulation. Two microencapsulation methods, spray-drying (SD) and freeze-drying (FD), were employed using sodium caseinate (Cas) as a cell protectant at concentrations of 0.035% and 1% and at two pHs, 3 and 7. Maltodextrins (MD) served as wall material (10%). Microcapsules were analysed for cell viability and membrane damage after drying, survival under simulated gastro-intestinal conditions, antimicrobial activity, stability during storage, and physicochemical characterization. Results showed that E. durans F21 exhibited promising probiotic properties, including moderate auto-aggregation, high co-aggregation with pathogens, moderate biofilm formation, and resistance to simulated gastrointestinal conditions. The encapsulation pH showed to be a crucial factor affecting the viability of microencapsulated cells. Microencapsulation at pH 3 adversely affected cell viability during drying. However, microencapsulation at pH 7 using Cas (at 0.035 and 1%) was found to be most effective in maintaining higher cell viability under similar conditions. No significant difference was detected between both Cas formulations, suggesting that 0.035% Cas concentration might be sufficient for the microencapsulation process at pH 7. Moreover, FD proved to be the most effective method to produce E. durans F21 microcapsules with high viability (cell viability of 93%) and stability during storage (cell viability of 99%). Conclusively, microencapsulation of E. durans F21 using Cas, at pH 7, and employing FD method could be a promising strategy for producing highly viable microcapsules containing E. durans F21 cells with high probiotic and bio-preservative potentials in foods.

Development of a functional dulce de leche (milk jam) with prebiotic carbohydrates and Lacticaseibacillus rhamnosus GG

A functional dulce de leche (DL) was developed by incorporating prebiotic carbohydrates (inulin and maltodextrin) and probiotic Lacticaseibacillus rhamnosus ATCC 53103 (LGG) (either as free cells, or encapsulated in pectin beads with or without prebiotics). Encapsulation improved the viability of LGG cells in DL during refrigerated storage, and its survival under simulated gastrointestinal conditions. Final cell counts in all DL samples after 6 months were >106 CFU g−1. Incorporation of carbohydrates in DL and probiotic encapsulation state had no significant effect on colour (L∗a∗b∗), rheological and mechanical properties. Although encapsulation state caused a significant effect on acceptability of DL, all samples scored above the minimum acceptability level, with more than 80% of consumers acceptance. Storage time had a significant effect (only after 2–3 months) on DL properties (except b∗ and adhesiveness), causing an increase in rheological properties, a decrease in texture parameters, and a decrease in L∗ and a∗.

Artichoke extract and human bifidobacteria to attain symbiotic sheep milk cheeses

There is a growing interest for healthy non-bovine functional dairy products. Therefore, in the present study, an aqueous polyphenol-rich artichoke extract (AE) and two Bifidobacterium strains of human origin (B. pseudolongum INIA P2 and B. longum INIA P132) were added, alone or in combination with AE, for the manufacture of six sheep milk cheeses. In general, addition of 1.25 g of lyophilized AE per 100 g of curd did not affect cheese pH, dry matter, lactoccci starter counts or organic acid levels during ripening. Bifidobacterial strains showed good viability in cheeses with AE during ripening and after simulated major gastrointestinal conditions. Cheeses with AE showed improved functional properties, with significantly (P < 0.01) higher total polyphenol content and, especially, antioxidant activity. A symbiotic cheese with AE andB. pseudolongumINIA P2, a strain known for increasing polyphenol bioavailability, may ensure that consumers could fully benefit from polyphenol health-promoting properties. The cheese with AE and strain INIA P2 fulfil the definition of a symbiotic cheese. It contains prebiotic compounds and Bifidobacterium at adequate levels to sustain a therapeutic effect, which are able to survive major gastrointestinal conditions. This cheese could confer beneficial effects on consumers’ health by contributing to the host antioxidant capacity and increasing polyphenol bioavailability.

Multistimuli responsive microcapsules produced by the prilling/vibration technique for targeted colonic delivery of probiotics

This study aimed to microencapsulate the probiotic strain Lactiplantibacillus plantarum 4S6R (basonym Lactobacillus plantarum) in both microcapsules and microspheres by prilling/vibration technique. A specific polymeric mixture, selected for its responsiveness to parallel colonic stimuli, was individuated as a carrier of microparticles. Although the microspheres were consistent with some critical quality parameters, they showed a low encapsulation efficiency and were discarded. The microcapsules produced demonstrated high yields (97.52%) and encapsulation efficiencies (90.06%), with dimensional analysis and SEM studies confirming the desired size morphology and structure. The results of thermal stress tests indicate the ability of the microcapsules to protect the probiotic. Stability studies showed a significant advantage of the microcapsules over non-encapsulated probiotics, with greater stability over time. The release study under simulated gastrointestinal conditions demonstrated the ability of the microcapsules to protect the probiotics from gastric acid and bile salts, ensuring their viability. Examination in a simulated faecal medium revealed the ability of the microcapsules to release the bacteria into the colon, enhancing their beneficial impact on gut health. This research suggests that the selected mixture of reactive polymers holds promise for improving the survival and efficacy of probiotics in the gastrointestinal tract, paving the way for the development of advanced probiotic products.

Construction of Fu brick tea polysaccharide-cold plasma modified alginate microgels for probiotic delivery: Enhancing viability and colonization

Emerging food processing technologies provide broader avenues for enhancing probiotic delivery systems. In this study, the new Fu brick tea polysaccharide (FBTP) was extracted and combined with cold plasma-modified alginate nano-montmorillonite (AMT) to prepare microgels by ionic gelation to improve the viability of encapsulated Lactobacillus kefiranofaciens JKSP109. Results showed that cold plasma treatment for 3 min changed the surface charge of AMT biopolymer solution, and FBTP addition reduced the particle size to the lowest of 223 ± 5.50 nm. Morphological analysis showed that the AMT treated with cold plasma for 3 min and FBTP (C3AMT + FBTP) formed a dense microgel through electrostatic interaction, and the probiotics were randomly distributed in their internal polysaccharide network, as well as the interlayer and surrounding of nanoparticles. The probiotics immobilized in C3AMT + FBTP microgel exhibited the highest viability (8.48 ± 0.03 log CFU/g) and colonic colonization after exposure to simulated gastrointestinal conditions. In addition, the good antioxidant activity of FBTP reduced the loss of probiotic viability during storage, with only 2.58 log CFU/g decreased after 4 weeks. Therefore, such probiotic products enriched with natural bioactive ingredients can be developed as a potential functional food additive.

Core-shell nanofibers based on microalgae proteins/alginate complexes for enhancing survivability of probiotics

In this study, a novel one-step coaxial electrospinning process is employed to fabricate shell-core structure fibers choosing Chlorella pyrenoidosa proteins (CP) as the core material. These nanofibers, serving as the wall material for probiotic encapsulation, aimed to enhance the stability and antioxidant activity of probiotics in food processing, storage, and gastrointestinal environments under sensitive conditions. Morphological analysis was used to explore the beads-on-a-string morphology and core-shell structure of the electrospun fibers. Probiotics were successfully encapsulated within the fibers (7.97 log CFU/g), exhibiting a well-oriented structure along the distributed fibers. Compared to free probiotics and uniaxial fibers loaded with probiotics, encapsulation within microalgae proteins/alginate core-shell structure nanofibers significantly enhanced the probiotic cells' tolerance to simulated gastrointestinal conditions (p < 0.05). Thermal analysis indicated that microalgae proteins/alginate core-shell structure nanofibers displayed superior thermal stability compared to uniaxial fibers. The introduction of CP resulted in a 50 % increase in the antioxidant capacity of probiotics-loaded microalgae proteins/alginate nanofibers compared to uniaxial alginate nanofibers, with minimal loss of viability (0.8 log CFU/g) after 28 days of storage at 4 °C. In summary, this dual-layer carrier holds immense potential in probiotic encapsulation and enhancing their resistance to harsh conditions.

Optimization of oxalic acid treatment for ulvan extraction from Ulva linza biomass and its potential application as Fe(III) chelator

This study investigated the use of oxalic acid as an eco-friendly compound for extracting ulvan from Ulva linza biomass. The concentration of oxalic acid, temperature, and period of extraction were optimized using Box-Behnken design in response of ulvan yield, molecular weight (MW), sulphate content, uronic acid content, purity ratio (total sugars/total phenolics and proteins), and Fe(III) chelation properties. Under the optimized conditions (1.7 % w/v oxalic acid, 64 °C, and 2.63 h), ulvan yield, MW, sulphate content, uronic acid content, and purity ratio were 29.90 % (w/w), 32.22 kDa, 11.01 % (w/w), 8.12 % (w/w), and 12.84, respectively. The optimized ulvan exhibited good Fe(III) chelation of 18.58 % (w/w). The synthesized Fe-ulvan complex released approximately 73 % of the chelated Fe(III) under in vitro simulated gastro-intestinal conditions. Furthermore, both ulvan and Fe-ulvan complex exhibited potent antioxidant properties. FT-IR analysis confirmed the fundamental role played by hydroxyl, carboxyl and sulphate groups in the coordination of Fe(III). Furthermore, MW, MW/sulphate ratio, and MW/uronic acid ratio should be low to enhance the Fe(III) chelation properties of ulvan. The results of the present study shed light on the use of oxalic acid as a simple and environmentally-benign treatment for ulvan extraction, especially for the recovery of low MW ulvan with high yield and good Fe(III)-binding properties.

Nanostructured Lipid Carriers for oral treatment of leishmaniasis: Design and preclinical evaluation

Pentavalent antimonials are the primary treatment for leishmaniasis due to their proven efficacy and cost-effectiveness. However, they present poor oral absorption, parenteral administration, and serious adverse effects due to prolonged use. Nanostructured Lipid Carriers (NLC) offer a promising solution, providing prolonged release, enhanced intestinal permeability, and oral bioavailability. This study aimed to address these issues by developing Nanostructured Lipid Carrier loaded with meglumine antimoniate (NLC-MA) for oral leishmaniasis treatment. We used design of experiments (DoE) for optimization. The chosen NLC-MA was characterized assessing size, polydispersity index (PdI), zeta potential (ZP), entrapment efficiency (EE), pH, and by analytical techniques such as DSC, FTIR, XRD, and TEM. Stability under simulated gastrointestinal conditions, in vitro release kinetics, cytotoxicity in RAW 264.7 macrophages, and leishmanicidal activity were assessed. NLC-MA presented 41.13 nm, PdI 0.227, ZP −27.9 mV, EE of 62.43%, and pH 5.23. Characterization techniques confirmed drug incorporation and nanoparticles with reduced crystallinity. TEM images showed spherical morphology. NLC remained stable in gastrointestinal pH and under refrigeration storage, while lyophilized formulations retained their initial properties. MA release from NLC followed the Peppas-Sahlin model, an anomalous transport mechanism. Importantly, MA showed no cytotoxicity in macrophages, but the NLCs exhibited cytotoxicity beyond 50 µg/mL. NLC-MA displayed improved efficacy against Leishmania infantum. Overall, these results highlight the potential of utilizing a lipid nanocarrier incorporating meglumine antimoniate as an innovative drug delivery platform for oral treatment against leishmaniasis.

Assessing Metschnikowia pulcherrima as a potential probiotic yeast for animal feed

Abstract In response to escalating antimicrobial resistance fuelled by their extensive use in livestock farming, this study explores alternative strategies in animal health management. We investigated the probiotic potential of Metschnikowia pulcherrima strains 4 × 3, DH5, ICS1 and QRI1 for their suitability in poultry environments. Our research demonstrated that previously discovered antimicrobial M. pulcherrima strains exhibit promising probiotic behaviours, including biofilm production, auto-aggregation, cell surface hydrophobicity, adhesion to Caco-2 cells, antioxidant capacity, and resilience to gastric stresses. Notable findings include DH5 exhibiting the highest biofilm formation, 4 × 3 and DH5 showing rapid auto-aggregation, 4 × 3 and ICS1 displaying high cell surface hydrophobicity, and all strains demonstrating considerable adherence to Caco-2 cells. 4 × 3 also exhibited exceptional bile tolerance, while ICS1 showed robust survival under simulated gastrointestinal conditions. These traits suggest M. pulcherrima’s capacity to colonise the poultry gastrointestinal tract, promote animal health, and support more sustainable livestock practices as a potential alternative to conventional antibiotics.

Co‐encapsulated of potential probiotic yeast Kluyveromyces marxianus and peanut skin polyphenolic extract as a functional ingredient

SummaryThe aim of this study was to develop and evaluate the bioactive properties and storage stability of microcapsules (MCs) obtained by co‐encapsulation of the potential probiotic yeast Kluyveromyces marxianus VM004 and peanut skin polyphenolic extract (PSE) by spray drying, using whey protein concentrate (WPC) and water‐soluble chitosan (WSCh) as wall materials. The results showed that the selected wall materials provided protection to yeast during spray drying, storage, and simulated gastrointestinal conditions, obtaining better results for WPC and higher concentrations of PSE. Moreover, all formulations demonstrated cytoprotective effects against menadione‐induced oxidative stress in normal rat ileum epithelial cells (IEC‐18) at all concentrations and storage temperatures evaluated. These results suggest that the obtained MCs could be potential functional food ingredients, considering their antioxidant, cytoprotective, and potential probiotic properties.

Mangaba pulp fermented with Lacticaseibacillus casei 01 has improved chemical, technological, and sensory properties and positively impacts the colonic microbiota of vegan adults

This study aimed to evaluate the functional, technological, and sensory aspects of mangaba (Hancornia speciosa Gomes) fruit pulp fermented with the probiotic Lacticaseibacillus casei 01 (LC1) during refrigerated storage (7 °C, 28 days). The effects of the fermented mangaba pulp on the modulation of the intestinal microbiota of healthy vegan adults were also assessed. Mangaba pulp allowed high viability of LC1 during storage and after simulated gastrointestinal conditions (≥7 log CFU/g). The fermented mangaba pulp showed lower pH and total soluble solids, and higher titratable acidity, and concentrations of lactic, acetic, citric, and propionic acids during storage compared to non-fermented pulp. Also, it presented a higher concentration of bioaccessible phenolics and volatiles, and improved sensory properties (yellow color, brightness, fresh appearance, and typical aroma and flavor). Fermented mangaba pulp added to in vitro cultured colonic microbiota of vegan adults decreased the pH values and concentrations of maltose, glucose, and citric acid while increasing rhamnose and phenolic contents. Fermented mangaba pulp promoted increases in the abundance of Dorea, Romboutsia, Faecalibacterium, Lachnospira, and Lachnospiraceae ND3007 genera and positively impacted the microbial diversity. Findings indicate that mangaba pulp fermented with LC1 has improved chemical composition and functionality, inducing changes in the colonic microbiota of vegan adults associated with potential benefits for human health.

The Ability of Lacticaseibacillus paracasei MSMC 36-9 Strain with Probiotic Potential to Ferment Coconut Milk and Produce a Yogurt-Type Beverage

The efficacy of the Lacticaseibacillus paracasei MSMC 36-9 strain with probiotic potential to ferment coconut milk and produce coconut milk yogurt-type beverages was examined. Tapioca starch was used as a stabilizer at concentrations of 0, 1.0, and 2.0% (w/w). The samples were stored at 4 °C for 21 days and analyzed for viability and resistance to in vitro gastrointestinal conditions of L. paracasei MSMC 36-9, pH changes, radical scavenging activity using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, and apparent viscosity. The viability of the strain with probiotic potential in the samples remained stable during storage and ranged between 12 and 13 log CFU/g by the end of the storage period. The strain L. paracasei MSMC 36-9 from all samples survived under simulated gastrointestinal conditions. The pH levels of all samples decreased during storage due to post-acidification. The radical scavenging activity of the products fermented with L. paracasei MSMC 36-9 was higher than that of the sample fermented with the commercial yogurt starter culture. The addition of tapioca starch to fermented coconut milk increased the viscosity of the samples. The results suggested that L. paracasei MSMC 36-9 can be used as a starter culture in the production of coconut milk yogurt-type beverages with antioxidant potential.

Characterization of a symbiotic beverage based on water-soluble soybean extract fermented by Lactiplantibacillus plantarum ATCC 8014.

The health benefits of functional foods are associated with consumer interest and have supported the growth of the market for these types of foods, with emphasis on the development of new formulations based on plant extracts. Therefore, the present study aimed to characterize a symbiotic preparation based on water-soluble soy extract, supplemented with inulin and xylitol and fermented by Lactiplantibacillus plantarum ATCC 8014. Regarding nutritional issues, the symbiotic formulation can be considered a source of fiber (2g/100mL) and proteins (2.6g/100mL), and it also has a low-fat content and low caloric value. This formulation, in terms of microbiological aspects, remained adequate to legal standards after storage for 60days under refrigeration and also presented an adequate quantity of the aforementioned probiotic strain, corresponding to 9.11 Log CFU.mL-1. These viable L. plantarum cells proved to be resistant to simulated human gastrointestinal tract conditions, reaching the intestine at high cell concentrations of 7.95 Log CFU.mL-1 after 60days of refrigeration. Regarding sensory evaluation, the formulation showed good acceptance, presenting an average overall impression score of 6.98, 5.98, and 5.16, for control samples stored for 30 and 60days under refrigeration, respectively. These results demonstrate that water-soluble soy extract is a suitable matrix for fermentation involving L. plantarum ATCC 8014, supporting and providing data on the first steps towards the development of a symbiotic functional food, targeting consumers who have restrictions regarding the consumption of products of animal origin, diabetics, and individuals under calorie restrictions.

Effect of Amine, Carboxyl, or Thiol Functionalization of Mesoporous Silica Particles on Their Efficiency as a Quercetin Delivery System in Simulated Gastrointestinal Conditions.

Quercetin (Q) dietary supplements exhibit poor oral bioavailability because of degradation throughout gastrointestinal digestion (GD), which may be overcome using mesoporous silica particles (MSPs) as an oral delivery system (ODS). This study aimed to elucidate the effect of the functionalization of MSPs with amine-(A-MSP), carboxyl-(C-MSP), or thiol-(T-MSP) groups on their efficiency as a quercetin ODS (QODS). The type and degree of functionalization (DF) were used as factors in an experimental design. The Q-loaded F-MSP (F-MSP/Q) was characterized by gas physisorption analysis, loading capacity (LC), and dynamic light scattering and kinetics of Q release at gastric and intestinal pHs. Antioxidant capacity and Q concentration of media containing F-MSP/Q were evaluated after simulated GD. A-MSP showed the highest LC (19.79 ± 2.42%). C-MSP showed the lowest particle size at pH 1.5 or 7.4 (≈200 nm). T-MSP exhibited the maximum Q release at pH 7.4 (11.43%). High DF of A-MSP increased Q retention, regardless of pH. A-MSP preserved antioxidant capacity of Q-released gastric media (58.95 ± 3.34%). Nonetheless, MSP and F-MSP did not protect antioxidant properties of Q released in intestinal conditions. C-MSP and T-MSP showed essential features for cellular uptake and Q release within cells that need to be assessed.

Supramolecular assembly of dual crosslinked nanocomposite polysaccharides hydrogel: Integration of injectable, self-healing, and pH-responsive platform for sustained delivery of polyphenols

This study introduces an innovative approach in the field of biomaterials through the development of a multifunctional, printable scaffold designed for the advanced delivery of polyphenols. Addressing challenges of burst-release, poor compounds solubility and bioavailability, we engineered a supramolecular-assembled dual crosslinked hydrogel combining fucoidan (F) and chitosan (CS) encapsulating nanoliposomes. The novel integration of dibenzaldehyde-terminated PEG (DB) within the hydrogel, created a synergistic crosslinking through the formation of the covalent Schiff base bonds and physical interactions which enhanced the hydrogel's overall characteristics. Tailoring F content (DBCS-F), allowed the crosslinking density modulation, yielding a robust gel network structure with enhanced mechanical properties, evidenced by a remarkable compressive stress resistance of 1.73 MPa at 60% strain, coupled with high compressive resilience (79.31% stress recovery after 6 cycles), surpassing single crosslinked hydrogel (DBCS). The rheological analysis further confirmed the hydrogel's improved viscoelasticity, stable network structure, and shear-thinning behavior, affirming its excellent injectability and suitability for 3D printing applications. Most notably, the incorporation of catechin/juglone-loaded nanoliposomes contributed to a pH-responsive sustained release of polyphenols in simulated gastrointestinal conditions. The developed dual crosslinked hydrogel, integrating injectability, self-healing, and pH-responsiveness presents a significant advancement in biomaterials and holds a promising approach for the fabrication of personalized 3D printed scaffolds aimed for sustained and targeted polyphenols release.

In vitro and in silico investigation of glycyrrhizic acid encapsulated zein nanoparticles: A synergistic targeted drug delivery approach for breast cancer

This study presents an innovative approach for targeted drug delivery through the development of Glycyrrhizic acid-loaded zein nanoparticles (GA-LNPs) as a proficient carrier system. The juxtaposition of zein, a hydrophobic biological macromolecule as a protein carrier, and Glycyrrhizic acid (GA), a hydrophilic therapeutic compound, exemplifies the adaptability of hydrocolloids within cutting-edge drug delivery systems. The characterization and functional traits of research encompass multifaceted analyses of natural macromolecules, which elucidate the homogeneous and spherical morphology of GA-LNPs with an average size of 170.49 nm. The controlled drug release profile of GA, orchestrated under simulated gastrointestinal conditions, adheres to diffusion-based Higuchi kinetics, reflecting the controlled release of the natural macromolecules. The intermolecular interactions among Zein, GA, and cross-linker EDC, facilitated through molecular dynamics simulations, fortify the structural integrity of the encapsulation matrix. In Vitro studies revealed enhanced cellular uptake of GA-LNPs in MCF-7 breast cancer cells. This cellular internalization was further confirmed through cytotoxicity assessments using MTT and apoptosis assays (fluorescence microscopy), which demonstrated the prominent anticancer effects of GA-LNPs on MCF-7 in time/dose-dependent manner. The successful formulation of GA-LNPs, coupled with their sustained release and potent anticancer properties, makes them a potential platform for advanced targeted therapeutic strategies in biomedical applications.
