Processing time affects sequential memory performance beginning at the level of visual encoding.

Electrophysiological studies have demonstrated that theta-band activity is useful for investigating neural mechanisms of memory. However, mechanisms specifically driving memory performance remain poorly understood. In sequential memory, performance can be artificially attenuated by shortening the inter-stimulus interval (ISI) between memory item presentations. Therefore, we sought to clarify the mechanisms of sequential memory performance by analyzing theta-band (4–8 Hz) activity recorded via magnetoencephalogram in 33 participants during performance of a sequential memory task where memory items were presented at either slow or fast rates in accordance with longer or shorter ISIs, respectively. Particularly in the slow task, theta activity clearly modulated in accordance with the presentation of memory items. Common cortical target regions in the occipital and frontal cortex were identified in both tasks and related to visual encoding and memory maintenance, respectively. Compared to the slow task, occipital-theta activity was significantly lower in the fast task from the midterm until the ending of encoding, in correspondence with significantly lower recall for memory items in this same period. Meanwhile, despite a loss of clarity in responsiveness to individual memory items in the fast task, frontal-theta activity was not different between tasks and exhibited particularly strong responses in both tasks during the holding period prior to recall. Our results indicate that shorter processing time erodes sequential memory performance beginning at the level of visual encoding.

Interrupted processing of paired somatosensory stimuli in short interstimulus intervals: role of thalamo-cortical oscillations.

Discrimination of consecutive sensory stimuli is imperative for proper sensory perception and behavioral response. We aimed to investigate the emergence of paired somatosensory responses in relation to the interstimulus interval (ISI) change. Paired stimulus with 35 ms, 50 ms, 80 ms, 140 ms, and 500 ms ISI was applied to the median nerve and evoked responses were recorded from the primary somatosensory cortex in rats. Early and late components of both responses were analyzed in different frequency bands. The amplitudes were comparable for the 1st responses (S1), while the amplitude of the 2nd responses (S2), and S2/S1 sensory gating ratio were significantly lower at 35 and 50 ms ISI values. S2/S1 ratio was close to 1 at 500 ms ISI. The duration and latency of the 2nd response was also different at 35 ms ISI. In the 2nd responses, area of early high-frequency oscillations (150-400 Hz) was significantly lower at 35 ms ISI values. The shaping of 2nd somatosensory response is dependent on ISIs. Early high-frequency oscillations changes without accompanying late high-frequency oscillations alterations, may indicate that reduced thalamo-cortical drive to the cortex take a part in determining the 2nd response at short ISI. Further research is required by using neuropsychiatric disorder models where somatosensory perception is impaired.

Fast acquisition of resting motor threshold with a stimulus–response curve – Possibility or hazard for transcranial magnetic stimulation applications?

ObjectivePrevious research has suggested that transcranial magnetic stimulation (TMS) related cortical excitability measures could be estimated quickly using stimulus–response curves with short interstimulus intervals (ISIs). Here we evaluated the resting motor threshold (rMT) estimated with these curves. MethodsStimulus-response curves were measured with three ISIs: 1.2–2 s, 2–3 s, and 3–4 s. Each curve was formed with 108 stimuli using stimulation intensities ranging from 0.75 to 1.25 times the rMTguess, which was estimated based on motor evoked potential (MEP) amplitudes of three scout responses. ResultsThe ISI did not affect the rMT estimated from the curves (F = 0.235, p = 0.683) or single-trial MEP amplitudes at the group level (F = 0.90, p = 0.405), but a significant subject by ISI interaction (F = 3.64; p < 0.001) was detected in MEP amplitudes. No trend was observed which ISI was most excitable, as it varied between subjects. ConclusionsAt the group level, the stimulus–response curves are unaffected by the short ISI. At the individual level, these curves are highly affected by the ISI. SignificanceEstimating rMT using stimulus–response curves with short ISIs impacts the rMT estimate and should be avoided in clinical and research TMS applications.

Emergence of Frequency-Dependent Motor Variability Within Supra-Second Auditory Cueing.

Motor variability (MV) is an essential feature of the sensory motor system, and it plays an important role in sensory-motor learning. MV facilitates motor adaptation during auditory-motor synchronization (AMS). In AMS, individuals receive a series of similar auditory stimuli that come in a periodic manner at a fixed interval called an inter-stimulus interval (ISI). Peri-second ISI (1 second range) and supra-second ISI (>1 second) are differently processed, since these intervals involve different amount of cognitive resources. Supra-second ISI involves more top-down attention as compared to peri-second ISI. In this study we examined the effect of tone frequency (perceptual property of auditory stimuli) on predictive tapping and MV under peri-second and supra-second ranges. We examined the effect of tone frequency (a perceptual property of auditory stimuli) on predictive tapping and motor variability (MV) under short (peri-second) and long (supra-second) inter-stimulus intervals. Among 30 healthy participants (aged 18-35 years, M = 24.6 years), we randomly assigned equal numbers of these two inter-stimulus conditions to isochronous sound sequences. In their attempt to synchronize their motor responses with the tone, participants reproduced the ISI in their inter-tap intervals (ITIs). We analyzed their predictive tapping in terms of negative asynchrony (in which the tap occurs before the tone) and small positive asynchrony (0-100 ms), whereas we analyzed MV using the coefficient of variation (CV) of the ITI. We found that participants showed predictive tapping under short ISI, irrespective of the tone frequency. Moreover, their MV was unaffected by tone frequency. These findings imply that participants expressed MV in a predictive rather than reactive manner under short, but not long, ISI. Under long ISI, tone frequency had a significant effect on MV such that there was higher MV with the low-frequency than with the high-frequency tone. Thus, low-frequency tones are most suitable for auditory-motor learning in the supra-second range.

Blink reflex recovery cycle distinguishes patients with idiopathic normal pressure hydrocephalus from elderly subjects.

The R2 component of blink reflex recovery cycle (R2BRrc) is a simple neurophysiological tool to detect the brainstem hyperexcitability commonly occurring in several neurological diseases such as Parkinson's disease and atypical parkinsonisms. In our study, we investigated for the first time the usefulness of R2BRrc to assess brainstem excitability in patients with idiopathic Normal Pressure Hydrocephalus (iNPH) in comparison with healthy subjects. Eighteen iNPH patients and 25 age-matched control subjects were enrolled. R2BRrc was bilaterally evaluated at interstimulus intervals (ISIs) of 100, 150, 200, 300, 400, 500 and 750ms in all participants. We investigated the diagnostic performance of R2BRrc in differentiating iNPH patients from control subjects using ROC analysis. Midbrain area and Magnetic Resonance Hydrocephalic Index (MRHI), an MRI biomarker for the diagnosis of iNPH, were measured on T1-weighted MR images, and correlations between R2BRrc values and MRI measurements were investigated. Fourteen (78%) of 18 iNPH patients showed an enhanced R2BRrc at ISIs 100-150-200ms, while no control subjects had abnormal R2BRrc. The mean amplitude of bilateral R2BRrc at the shortest ISIs (100-150-200ms) showed high accuracy in differentiating iNPH patients from controls (AUC = 0.89). R2BRrc values significantly correlated with midbrain area and MRHI values. This study represents the first evidence of brainstem hyperexcitability in iNPH patients. Given its low cost and wide availability, R2BRrc could be a useful tool for selecting elderly subjects with mild gait and urinary dysfunction who should undergo an extensive diagnostic workup for the diagnosis of NPH.

Is a paired-stimuli configuration necessary to obtain typical evoked response differences in studies of psychosis? An MEG study.

Paired-stimuli (S1-S2) procedures have long been used to assess auditory processing in psychosis. Such studies have shown aberrant evoked responses (ERPs) following long (S1 response) and/or short (S2 response) inter-stimulus intervals. The historical tendency from paired stimuli outcomes in the schizophrenia (SZ) literature is for (i) response to the first stimulus (S1) to be smaller among SZ, and (ii) response to the second stimulus (S2) to be larger among SZ in relation to the size of their S1. An interpretation of these two findings is that SZ have poor auditory response suppression to redundant stimuli ("poor gating"). The present study sought to determine if the reported S1 and S2 effects in SZ (smaller S1 and larger S2 in relation to S1 magnitude) require the paired-stimuli presentation format. Participants (18 schizophrenia and 17 healthy persons) were administered the equivalent of S1 (after a 4.5-sec ISI - "long ISI") and S2 (after a 500-ms ISI - "short ISI") stimuli under four conditions (traditional paired long and short, randomly interleaved long and short, block of long, block of short). Neural activity differences were consistent between-groups independent of condition: (i) schizophrenia cases had greater activity in the pre-stimulus to very early post-stimulus period, (ii) healthy persons had greater M100 activity to long ISI stimuli, and (iii) healthy persons had greater activity after the M50/M100 evoked fields (recovery phase) following short ISI stimuli. Simple early auditory processing in psychosis may be largely independent of stimulus presentation condition, an outcome that may help re-frame future translational studies. Traditional paired-stimuli auditory neural response effects may not require the paired-stimuli format.

Short interstimulus intervals (ISI<5ms) between a subthreshold and a suprathreshold pulse cause short-term intracortical inhibition (SICI), whereas longer ISI causes intracortical facilitation (ICF). Task-specific engagement of object-based and space-based attention with spatiotemporally defined objects.

We used a form of ambiguous apparent motion known as Ternus motion to isolate the effects of object-based and space-based attention, and to explore functional differences between them. Two frames of horizontally aligned disks that were shifted by one position between frames were temporally separated by either a short or a long inter-stimulus interval (ISI). Short ISI displays were perceived as element motion where one disk appeared to jump across the other two. Long ISI displays were perceived as group motion where all three disks appeared to move together. Because element and group motion imply mutually exclusive object structures, adding stimuli (e.g., a small gap) to one disk in each frame created conditions of orthogonal object and location status (same or different), depending on ISI. We used two tasks with different functional demands, an identification task (Experiments 1 and 3a) in which observers responded to a single attribute of the final stimulus, and a comparison task (Experiments 2 and 3b) in which observers compared two attributes across two stimuli. Reliable object-specific effects occurred only with the comparison task, whereas location-specific effects occurred with both tasks. These results confirm that attention can be directed to objects separately from spatial locations and vice versa, and, moreover, that object-based and space-based attention are engaged differently depending on the processing demands of the task.

Modeling the temporal dynamics of neural responses in human visual cortex

Cortical responses to visual stimuli exhibit complex temporal dynamics, including sub-additive temporal summation, response reduction with repeated or sustained stimuli (adaptation), and slower dynamics at low contrast. Multiple computational models have been proposed to account for these dynamics in several measurement domains, including single-cell recordings, psychophysics, and fMRI. It is challenging to compare these models because there are differences in model form, test stimuli, and instrument. Here we present a new dataset that is well-suited to compare models of neural temporal dynamics. The dataset is from electrocorticographic (ECoG) recordings of human visual cortex, which measures cortical neural population responses with high spatial and temporal precision. The stimuli were large, static contrast patterns and varied systematically in contrast, duration, and inter-stimulus interval (ISI). Time-varying broadband responses were computed using the power envelope of the band-pass filtered voltage time course (50-170 Hz) recorded from a total of 126 electrodes in ten epilepsy patients, covering earlier (V1-V4) and higher-order (LO, TO, IPS) retinotopic maps. In all visual regions, the ECoG broadband responses show several non-linear features: peak response amplitude saturates with high contrast and long stimulus durations; response latency decreases with increasing contrast; and the response to a second stimulus is suppressed for short ISIs and recovers for longer ISIs. These features were well predicted by a computational model (Zhou, Benson, Kay and Winawer, 2019) comprised of a small set of canonical neuronal operations: linear filtering, rectification, exponentiation, and a delayed divisive gain control. These results demonstrate that a simple computational model comprised of canonical neuronal computations captures a wide range of temporal and contrast-dependent neuronal dynamics at millisecond resolution. Finally, we present a software repository that implements models of temporal dynamics in a modular fashion, enabling the comparison of many models fit to the same data and analyzed with the same methods.

Do shorter inter-stimulus intervals in the go/no-go task enable better assessment of response inhibition?

Young, Sutherland, and McCoy indicated that a Go/No‐Go Task (GNG) becomes more difficult as the inter‐stimulus intervals (ISIs) becomes shorter. However, is the number of commission errors under extremely short ISIs a useful metric for assessing response inhibition? This study challenges the assumption that a shorter ISI in the GNG enables better assessment of response inhibition. University students (N = 213) completed the GNG, the Conners Continuous Performance Test 3rd Edition (CCPT), and the Modified Stroop Task. The GNG comprised four blocks of 400, 600, 800, and 1000 ms ISIs, whereas the stimulus presentation was fixed at 250 ms. Consistent with Young et al., shorter ISIs in the GNG resulted in more commission errors. In the block with the shortest ISI, participants also failed more frequently in responses in go trials than in the other blocks, which appears to increase in error variance of commission errors. Consistent with this interpretation, the association between the number of commission errors in the block with 400 ms ISI and CCPT performance was weaker than those between the number of commission errors in other blocks and CCPT performance. It is concluded that using the number of commission errors in the condition with extremely short ISIs in the GNG might be inappropriate for assessing response inhibition.

Specialization of Neurons with Different Response Patterns in the Mouse Mus musculus Auditory Midbrain and Primary Auditory Cortex during Communication Call Processing

Wriggling call produced by mouse pups when struggling in the nest is one of the mouse early ontogenesis communicative vocalization. Processing of these signals by auditory midbrain and primary auditory cortex neurons with different response patterns was the subject of this study. Responses of single neurons evoked by wriggling call models and a series of models consisting of 4 stimuli with different interstimulus intervals (0–1000 ms) were recorded extracellularly. A third of central nucleus neurons in the inferior colliculus showed a spectral facilitation in responses to various two- and three-tone combinations of the wriggling call frequency components. About 80% of such combination-sensitive neurons had tonic characteristics of responses, i.e. tonic, phasic-tonic, pauser and long-latency discharges. A study of responses of central nucleus neurons to a series of wriggling call models showed that at short interstimulus intervals (0–50 ms) responses to the 2nd–4th signals in a series were completely suppressed or significantly decreased in two-thirds of the recorded neurons, i.e. there arose adaptation to a series of sounds. Such neurons had mainly phasic discharges. In a third of neurons, adaptation did not arise; these neurons were tonically discharging. All neurons in the primary auditory cortex (anterior and primary auditory fields) demonstrated adaptation in responses to a series of wriggling calls. Discharges of neurons were phasic.

Post-activation depression of primary afferents reevaluated in humans

Amplitude variation of Hoffmann Reflex (H-reflex) was used as a tool to investigate many neuronal networks. However, H-reflex itself is a subject to intrinsic changes including post-activation depression (P-AD). We aimed to investigate P-AD and its implication on motor control in humans. Upon tibial nerve stimulation in 23 healthy participants, peak-to-peak amplitude change of H-reflex was investigated using surface electromyography (SEMG) of soleus muscle. Variety of stimulus intensities, interstimulus intervals (ISIs), voluntary contraction levels/types and force recording were used to investigate the nature of P-AD. We have shown that P-AD was significantly stronger in the shorter ISIs. The only exception was the ISI of 200 msecs which had a weaker P-AD than some of the longer ISIs. Sudden muscle relaxation, on the other hand, further increased the effectiveness of the ongoing P-AD. Moreover, P-AD displayed its full effect with the first stimulus when there was no muscle contraction and was efficient to reduce the muscle force output by about 30%. These findings provide insight about the variations and mechanism of P-AD and could lead to improvements in diagnostic tools in neurological diseases.

Low-intensity repetitive paired associative stimulation targeting the motor hand area at theta frequency causes a lasting reduction in corticospinal excitability

ObjectiveSub-motor threshold 5 Hz repetitive paired associative stimulation (5 Hz-rPAS25ms) produces a long-lasting increase in corticospinal excitability. Assuming a spike-timing dependent plasticity-like (STDP-like) mechanism, we hypothesized that 5 Hz-rPAS at a shorter inter-stimulus interval (ISI) of 15 ms (5 Hz-rPAS15ms) would exert a lasting inhibitory effect on corticospinal excitability. Methods20 healthy volunteers received two minutes of 5 Hz-rPAS15ms. Transcranial magnetic stimulation (TMS) was applied over the motor hotspot of the right abductor pollicis brevis muscle at 90% active motor threshold. Sub-motor threshold peripheral electrical stimulation was given to the left median nerve 15 ms before each TMS pulse. We assessed changes in mean amplitude of the unconditioned motor evoked potential (MEP), short-latency intracortical inhibition (SICI), intracortical facilitation (ICF), short-latency afferent inhibition (SAI), long-latency afferent inhibition (LAI), and cortical silent period (CSP) before and for 60 minutes after 5-Hz rPAS15ms. ResultsSubthreshold 5-Hz rPAS15ms produced a 20–40% decrease in mean MEP amplitude along with an attenuation in SAI, lasting at least 60 minutes. A follow-up experiment revealed that MEP facilitation was spatially restricted to the target muscle. ConclusionsSubthreshold 5-Hz rPAS15ms effectively suppresses corticospinal excitability. Together with the facilitatory effects of subthreshold 5-Hz rPAS25ms (Quartarone et al., J Physiol 2006;575:657–670), the results show that sub-motor threshold 5-Hz rPAS induces STDP-like bidirectional plasticity in the motor cortex. SignificanceThe results of the present study provide a new short-time paradigm of long term depression (LTD) induction in human sensory-motor cortex.

A novel rat robot controlled by electrical stimulation of the nigrostriatal pathway.

Artificial manipulation of animal movement could offer interesting advantages and potential applications using the animal's inherited superior sensation and mobility. Although several behavior control models have been introduced, they generally epitomize virtual reward-based training models. In this model, rats are trained multiple times so they can recall the relationship between cues and rewards. It is well known that activation of one side of the nigrostriatal pathway (NSP) in the rat induces immediate turning toward the contralateral side. However, this NSP stimulation-induced directional movement has not been used for the purpose of animal-robot navigation. In this study, the authors aimed to electrically stimulate the NSP of conscious rats to build a command-prompt rat robot. Repetitive NSP stimulation at 1-second intervals was applied via implanted electrodes to induce immediate contraversive turning movements in 7 rats in open field tests in the absence of any sensory cues or rewards. The rats were manipulated to navigate from the start arm to a target zone in either the left or right arm of a T-maze. A leftward trial was followed by a rightward trial, and each rat completed a total of 10 trials. In the control group, 7 rats were tested in the same way without NSP stimulation. The time taken to navigate the maze was compared between experimental and control groups. All rats in the experimental group successfully reached the target area for all 70 trials in a short period of time with a short interstimulus interval (< 0.7 seconds), but only 41% of rats in the control group reached the target area and required a longer period of time to do so. The experimental group made correct directional turning movements at the intersection zone of the T-maze, taking significantly less time than the control group. No significant difference in navigation duration for the forward movements on the start and goal arms was observed between the two groups. However, the experimental group showed quick and accurate movement at the intersection zone, which made the difference in the success rate and elapsed time of tasks. The results of this study clearly indicate that a rat-robot model based on NSP stimulation can be a practical alternative to previously reported models controlled by virtual sensory cues and rewards.

Dissociated modulations of multivoxel activation patterns in the ventral and dorsal visual pathways by the temporal dynamics of stimuli.

IntroductionPrevious studies suggested temporal limitations of visual object identification in the ventral pathway. Moreover, multivoxel pattern analyses (MVPA) of fMRI activation have shown reliable encoding of various object categories including faces and tools in the ventral pathway. By contrast, the dorsal pathway is involved in reaching a target and grasping a tool, and quicker in processing the temporal dynamics of stimulus change. However, little is known about how activation patterns in both pathways may change according to the temporal dynamics of stimulus change.MethodsHere, we measured fMRI responses of two consecutive stimuli with varying interstimulus intervals (ISIs), and we compared how the two visual pathways respond to the dynamics of stimuli by using MVPA and information‐based searchlight mapping.ResultsWe found that the temporal dynamics of stimuli modulate responses of the two visual pathways in opposite directions. Specifically, slower temporal dynamics (longer ISIs) led to greater activity and better MVPA results in the ventral pathway. However, faster temporal dynamics (shorter ISIs) led to greater activity and better MVPA results in the dorsal pathway.ConclusionsThese results are the first to show how temporal dynamics of stimulus change modulated multivoxel fMRI activation pattern change. And such temporal dynamic response function in different ROIs along the two visual pathways may shed lights on understanding functional relationship and organization of these ROIs.

Effect of conditioning and test stimulus intensity on cortical excitability using triad-conditioning transcranial magnetic stimulation

Cortical facilitation assessed with triad conditioning transcranial magnetic stimulation has been termed triad-conditioned facilitation (TCF). TCF has been supposed to reflect increased intracortical facilitation (ICF) at short interstimulus intervals (ISI) around 10 ms and an intrinsic rhythm of the motor cortex at longer ISI around 25 ms. To gain further insight into the pathophysiological mechanism of TCF, we systematically studied the effect of suprathreshold conditioning stimulus (CS) and test stimulus (TS) intensity on TCF. Various CS intensities and TS intensities were used in a triad-conditioning paradigm that was applied to 11 healthy subjects. ISI between pulses were studied between 5 and 200 ms. TCF at 10 ms ISI enhanced with increasing CS intensity but decreased with increasing TS intensity. The duration of facilitation was longer with higher CS intensity. However, TCF at 25 ms ISI could not be elicited with none of the CS and TS intensities addressed here. Our results are consistent with the notion of TCF at short ISI reflecting ICF. The enhanced and prolonged facilitation with increase of CS without additional isolated facilitation at longer ISI suggest a prolongation of ICF.

Retinal ON and OFF pathways contribute to initial optokinetic responses with different temporal characteristics.

Visual information in the retina is processed via two pathways: ON and OFF pathways that originate from ON and OFF bipolar cells. The differences in the receptors that mediate signal transmission from photoreceptors imply that the response speed to light signals differs between ON and OFF pathways. We studied the initial optokinetic responses (OKRs) of mice using two-frame motion stimuli presented with interstimulus intervals (ISIs) to understand functional difference of these pathways. When two successive image frames were presented with an ISI, observers often perceived motion in the opposite direction of the actual shift. This directional reversal results from the biphasic nature of the temporal filters in visual systems whose characteristics can be estimated from the dependence on ISIs. We examined the dependence on ISIs in the OKRs of TRPM1-/- mice, whose ON bipolar cells are dysfunctional, as well as in those of wild-type control mice. Wild type and TRPM1-/- mice showed comparable OKRs in the veridical direction when no ISI was present. Both types of mice showed OKRs that decreased and eventually reversed as the ISI increased, but with a directional reversal at a shorter ISI in TRPM1-/- than wild-type mice. In addition, the temporal filters of TRPM1-/- mice estimated from dependence on ISIs were tuned for higher frequencies, suggesting that compared with wild-type mice, the visual system of TRPM1-/- mice responds to light signals with faster dynamics. We conclude that the ON and OFF pathways contribute to initial OKRs by providing visual signals processed with different temporal resolutions.

Touch anticipation mediates cross-modal Hebbian plasticity in the primary somatosensory cortex

Paired associative stimulation (PAS) protocols can be used to induce Hebbian plasticity in the human brain. A modified, cross-modal version, of the PAS (cross-modal PAS, cm-PAS) has been recently developed. The cm-PAS consists in the repetitive pairings of a transcranial magnetic stimulation (TMS) pulse over the primary somatosensory cortex (S1) and a visual stimulus depicting a hand being touched; a 20 ms of inter-stimulus interval (ISI) is required to affect S1 plasticity, in turn modulating tactile acuity and somatosensory evoked potentials. The present study explores the role of anticipatory simulation in the cm-PAS efficacy, which could be responsible for such a short ISI. To this aim, we compared the effect of the original, fixed-frequency, cm-PAS to that of a jittered version, in which the time interval between trials was not steady but jittered, hence avoiding the anticipation of the upcoming visual-touch stimulus. Moreover, in the jittered PAS, the ISI between the paired stimulations was varied: it could match the early, somatosensory-driven, activation of S1 (20 ms), or the mirror recruitment of S1 by touch observation (150 ms). Results showed that tactile acuity is enhanced by the fixed-frequency cm-PAS, with an ISI of 20 ms between paired stimulation (visual-touch stimulus and TMS pulse over S1), and also by the jittered cm-PAS but only if the ISI is of 150 ms. These findings suggest that the cm-PAS with a jittered frequency, by preventing an anticipatory pre-activation of S1, delays the timing of the interaction between the visual-touch stimulus and the cortical pulse. On a broader perspective, our study highlights the possible involvement of sensory anticipation, likely through mirror-like simulation mechanisms, in tactile mirroring, as well as its influence of the optimal interval between the afferent and the magnetic pulse during PAS protocols.

Modulatory effects of the supplementary motor area on primary motor cortex outputs.

Premotor areas of primates are specialized cortical regions that can contribute to hand movements by modulating the outputs of the primary motor cortex (M1). The goal of the present work was to study how the supplementary motor area (SMA) located within the same hemisphere [i.e., ipsilateral SMA (iSMA)] or the opposite hemisphere [i.e., contralateral (cSMA)] modulate the outputs of M1. We used paired-pulse protocols with intracortical stimulations in sedated capuchin monkeys. A conditioning stimulus in iSMA or cSMA was delivered simultaneously or before a test stimulus in M1 with different interstimulus intervals (ISIs) while electromyographic activity was recorded in hand and forearm muscles. The pattern of modulation from iSMA and cSMA shared some clear similarities. In particular, both areas predominantly induced facilitatory effects on M1 outputs with shorter ISIs and inhibitory effects with longer ISIs. However, the incidence and strength of facilitatory effects were greater for iSMA than cSMA. We then compared the pattern of modulatory effects from SMA to the ones from the dorsal and ventral premotor cortexes (PMd and PMv) collected in the same series of experiments. Among premotor areas, the impact of SMA on M1 outputs was always weaker than the one of either PMd or PMv, and this was regardless of the hemisphere, or the ISI, tested. These results show that SMA exerts a unique set of modulations on M1 outputs, which could support its specific function for the production of hand movements.NEW & NOTEWORTHY We unequivocally isolated stimulation to either the ipsilateral or contralateral supplementary motor area (SMA) using invasive techniques and compared their modulatory effects on the outputs of primary motor cortex (M1). Modulations from both SMAs shared many similarities. However, facilitatory effects evoked from ipsilateral SMA were more common and more powerful. This pattern differs from the ones of other premotor areas, which suggests that each premotor area makes unique contributions to the production of motor outputs.
