Effects of abdominal, vaginal, and laparoscopic hysterectomies on total oxidant/antioxidant levels: A prospective observational study.

This prospective observational study aimed to compare abdominal hysterectomy (AH), vaginal hysterectomy (VH), and total laparoscopic hysterectomy (TLH) in terms of oxidative stress (OS) by measuring serum levels of total antioxidant status (TAS), total oxidant status (TOS), and oxidative stress index (OSI). Of the 3 groups, namely, AH, VH, and TLH, 22 patients were enrolled in each to investigate the aim of the study mentioned above. Patient demographics, clinical and surgical characteristics, and preoperative and postoperative (0th and 24th hours) serum TAS, TOS, and OSI levels were investigated. The groups differed significantly based on surgical indications (P < .001). While abnormal uterine bleeding was more prevalent in the AH (95.5%) and TLH (77.3%) groups, pelvic organ prolapse (POP) was more prevalent in the VH group (90.1%). The mean age and menopausal patient rates were significantly higher in the VH group than in the AH and TLH groups (P < .001). The groups did not differ significantly in terms of simultaneous bilateral salpingo-oophorectomy rates (P = .10). However, additional POP surgery was performed in 14 patients in the VH group. The mean operative time was significantly shorter in the AH group than in the VH and TLH groups (P = .001). The groups did not differ significantly based on either the preoperative or postoperative serum TAS levels (P > .05). Furthermore, the change in serum TAS levels over time was not significant in any of the groups (P > .05). In contrast, preoperative serum TOS and OSI levels were significantly higher in the VH group than in the AH and TLH groups (P < .05). The groups did not differ significantly in terms of serum TOS and OSI levels at 0th and 24th hours postoperatively (P > .05). Serum TOS and OSI levels increased dramatically over time in the AH and TLH groups (P < .05), but not in the VH group (P > .05). post hoc analysis showed that the changes in serum TOS and OSI values in the AH and TLH groups were between the preoperative and postoperative 0th hour levels (P < .001). Our results indicated that the total antioxidant capacity was preserved in all 3 techniques. However, VH causes less OS than the other hysterectomy techniques.

Relationship between oxidative stress and endometrial polyps in pre-and postmenopausal women.

This study aimed to investigate the relationship between oxidative stress (OS) and endometrial polyps (EP) in pre- versus postmenopausal women with abnormal uterine bleeding. This prospective case control study was conducted in the Gynecology Department of Ankara Bilkent City Hospital between January and December 2019. In this study, the EP and control groups included 45 participants each (30 pre- and 15 postmenopausal women). Demographics (age, gravidity, parity, and body mass index), serum complete blood count parameters, serum total antioxidant status, total oxidant status (TOS), oxidative stress index (OSI), and thiol/disulfide balance were compared between groups. Subsequently, all women were stratified based on menopausal status, and the investigated parameters were investigated separately in the pre- and postmenopausal participants between groups. There were no significant intergroup differences in the investigated parameters among all participants (P>0.05). However, unlike in the control group, the age distribution in the EP group was bimodal, peaking in the early 40s and late 50s. Stratification based on menopausal status revealed no significant intergroup differences in the investigated parameters among the premenopausal participants. However, among the postmenopausal participants, serum TOS and OSI levels were significantly higher in the EP versus control group (9.0 vs. 7.2 μmol H2O2 Equiv/L and 0.8 vs. 0.6 AU; P=0.01). Serum OS may play a role in the development of EP, particularly among postmenopausal women. Further investigations are required in this area.

Effects of Cerium Oxide on Kidney and Liver Tissue Damage in an Experimental Myocardial Ischemia-Reperfusion Model of Distant Organ Damage.

Background and Objectives: Ischemia-reperfusion (I/R) injury is a process in which impaired perfusion is restored by restoring blood flow and tissue recirculation. Nanomedicine uses cutting-edge technologies that emerge from interdisciplinary influences. In the literature, there are very few in vivo and in vitro studies on how cerium oxide (CeO2) affects systemic anti-inflammatory response and inflammation. Therefore, in our study, we aimed to investigate whether CeO2 administration has a protective effect against myocardial I/R injury in the liver and kidneys. Materials and Methods: Twenty-four rats were randomly divided into four groups after obtaining approval from an ethics committee. A control (group C), cerium oxide (group CO), IR (group IR), and Cerium oxide-IR (CO-IR group) groups were formed. Intraperitoneal CeO2 was administered at a dose of 0.5 mg/kg 30 min before left thoracotomy and left main coronary (LAD) ligation, and myocardial muscle ischemia was induced for 30 min. After LAD ligation was removed, reperfusion was performed for 120 min. All rats were euthanized using ketamine, and blood was collected. Liver and kidney tissue samples were evaluated histopathologically. Serum AST (aspartate aminotransferase), ALT (alanine aminotransaminase), GGT (gamma-glutamyl transferase), glucose, TOS (Total Oxidant Status), and TAS (Total Antioxidant Status) levels were also measured. Results: Necrotic cell and mononuclear cell infiltration in the liver parenchyma of rats in the IR group was observed to be significantly increased compared to the other groups. Hepatocyte degeneration was greater in the IR group compared to groups C and CO. Vascular vacuolization and hypertrophy, tubular degeneration, and necrosis were increased in the kidney tissue of the IR group compared to the other groups. Tubular dilatation was significantly higher in the IR group than in the C and CO groups. TOS was significantly higher in all groups than in the IR group (p < 0.0001, p < 0.0001, and p = 0.006, respectively). However, TAS level was lower in the IR group than in the other groups (p = 0.002, p = 0.020, and p = 0.031, respectively). Renal and liver histopathological findings decreased significantly in the CO-IR group compared to the IR group. A decrease in the TOS level and an increase in the TAS level were found compared to the IR group. The AST, ALT, GGT, and Glucose levels are shown. Conclusions: CeO2 administered before ischemia-reperfusion reduced oxidative stress and ameliorated IR-induced damage in distant organs. We suggest that CeO2 exerts protective effects in the myocardial IR model.

Increased oxidative stress biomarkers in central serous chorioretinopathy

Current data suggest that oxidative stress may play an important role in the occurrence of acute central serous chorioretinopathy (CSC), as chorioretinal integrity may be affected by disruption of the patient’s metabolic redox balance, indicating the need for biomarkers. In addition to oxidative stress, high-density lipoprotein (HDL) dysfunction due to dyslipidemia can also lead to many types of physical discomfort. However, little is known about the pathophysiology of the disease resulting from oxidative stress and HDL dysfunction in CSC. The aim of this study was to investigate whether serum oxidative stress and HDL functionality markers have an impact on CSC disease. The case series of this study included 33 consecutive patients with treatment-naïve acute CSC. Thirty-five healthy volunteers of similar age were included in this study as non-CSC controls. Serum samples of the participants were taken and routine lipid values, serum Total Antioxidant Status (TAS), Total Oxidant Status (TOS), Oxidized Low Density Lipoprotein (ox-LDL), and Paraoxonase (PON1) levels were measured quantitatively. Serum oxidative stress index (OSI) was then calculated. Serum Ox-LDL, TOS and OSI levels in the acute CSC group, consisting of patients who had never been treated before and had no other disease, were statistically significantly higher than the control group. Conversely, serum PON1 and TAS levels were lower in CSC than in the control group. The relationship between CSC and deterioration in serum redox balance and decrease in PON1 activity, an important marker of HDL functionality, was demonstrated for the first time through this study. According to the literature, serum levels of these biomarkers, which identify acute/chronic inflammation and oxidative stress, have not been measured before in CSC disease. Finally, it is conceivable that redox balance and HDL functionality may be important in the diagnosis and treatment of the acute phase of CSC.

Does metabolic control of the disease related with bone turnover markers in children with type 1 diabetes mellitus in Turkey?

BackgroundThe aim was to evaluate the effect of metabolic control on bone biomarkers in children with type I diabetes.Materials and methodsThe children were divided into two groups according to their glycated hemoglobin (HbA1c) (%) levels: a group with HbA1c levels < 8% (n = 16) and: a group with HbA1c levels > 8% (n = 18). The serum total oxidative status (TOS) (µmol/L), total antioxidant status (TAS) (mmol/L), alkaline phosphatase (ALP) (IU/L), osteocalcin (OC) (ng/ml), procollagen type-1-N-terminal peptide (P1NP) (ng/ml), and vitamin D (IU) levels and food consumption frequencies were determined.ResultsWhen patients were classified according to HbA1c (%) levels, those with HbA1c levels < 8% were found to have lower TOS (µmol/L) values (8.7 ± 6.16, 9.5 ± 5.60) and higher serum OC (ng/mL) (24.2 ± 16.92, 22.0 ± 6.21) levels than those with HbA1c levels > 8% (p < 0.05). Regardless of the level of metabolic control, there was a statistically significant association between serum TOS (µmol/L) and P1NP (ng/ml) (p < 0.05) levels, with no group-specific relationship (HbA1c levels <%8 or HbA1c levels >%8).ConclusionHbA1c and serum TOS levels had an effect on bone turnover biomarkers in individuals with type I diabetes.

Serum pentraxin 3 levels in cats with feline parvovirus infection

Feline parvovirus (FPV) infection continues to be a serious problem in cats and therefore studies are ongoing to investigate all aspects of the disease. This study was designed to determine the levels of PTX-3 in cats with feline panleukopenia (FPL). Blood samples were taken from 12 cats of different breeds and gender with complaints of weakness, listlessness, anorexia, diarrhoea, vomiting and FPV positive on examination and from 7 cats found healthy on physical and laboratory examination. Whole blood, biochemical parameters, total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSi), serum amyloid A (SAA), C-reactive protein (CRP) and pentraxin-3 (PTX-3) concentrations were determined in both sick and healthy cats. The results showed that there was marked panleukopenia and serum PTX-3 (58.69 pg/mL), SAA (59.91 µg/mL), TOS (14.35 μmol H2O2 Eq/L) and OSi (1.17 arbitrary unit) levels were significantly higher in cats diagnosed with FPL compared to healthy subjects. In conclusion, serum PTX- levels were measured for the first time in cats naturally infected with FPV and found to be elevated. Further clinical studies with large numbers of infected cats are needed to clarify these findings and to use PTX-3 as a reliable biomarker in FPV-infected cats.

Assessment of the Serum Paraoxonase1 Level and Its Relation to Total Oxidant Status and Disease Activity in Vitiligo Patients

Abstract Background Vitiligo is characterized by white macules and patches, whose size increases over time, due to the loss of melanocytes.It can appear at any time, and it significantly impairs the patients’ quality-of-life. Vitiligo is a multifactorial disorder due to genetic and environmental factors. One of the important hypotheses in the pathogenesis of vitiligo is the oxidative stress hypothesis, which is based on the reality of the formation of some toxic metabolites throughout pigment biosynthesis. Human paraoxonase-1 (PON1) is a Ca2+ dependent esterase synthesized in the liver. Reduced serum PON1 activity has been reported to be associated with some diseases under oxidative stress and inflammation conditions. Aim of the Work The aim of the work was to evaluate the serum level of paraoxonase1 in patients with vitiligo and its relation to total oxidant status and disease activity in vitiligous patients. Patients and Methods This is a case control study which included 48 vitiligo patients and 48 ageand sex-matched controls. The patients recruited from the Outpatient Dermatology Clinic at Ain Shams University Hospitals during the period from August 2020 till March 2021. Serum level of paraoxonase1 (PON1) and total oxidant status (TOS) where assessed by ELISA technique. Results We observed a statistically significant higher oxidative stress reflected by the high serum TOS levels among the vitiligo compared to controls. This finding supports the aforementioned hypothesis and it also came in accordance with the observations of high oxidative stress state in vitiligo patients which was repeatedly reported in many studies. Furthermore, we also reported the significant inverse relation between PON1 levels and the oxidative stress reflected by the serum TOS levels in vitiligo patients. This reflected the reduced protective antioxidant mechanisms in vitiligo patient making them more vulnerable to oxidative stress Conclusion Oxidative stress may play an important role in the pathogenesis of vitiligo. The finding of a PON1 decrease in vitiligo patients emphasises the underlying hypothesis in the progression of the disease, and it can highlight the effect of free radicals and leading oxidative damage in vitiligo disease. However, further, larger studies are necessary to confirm our results.

The usability of MCP-1, fetuin-A, TAS, and TOS levels in the diagnosis of acute myocardial infarction.

Our aims were to determine whether the levels of plasma monocyte chemotactic protein-1 (MCP-1), fetuin-A, serum total antioxidant status (TAS), and serum total oxidant status (TOS) are cardiac biomarkers and to clarify their relationship with each other in acute myocardial infarction (AMI). The study included 90 participants: 60 patients with AMI [30 with and 30 without ST-segment elevation myocardial infarction (STEMI)] and 30 cardiac patients without AMI. The diagnostic values of serum Hs-cTnT, MCP-1, fetuin-A, TAS, and TOS levels in predicting AMI were evaluated statistically. Median levels of MCP-1 [120.10 ng/L (interquartile range: 76.94-230.54 ng/L)] and TOS [2.89 U/MI (IQR: 2.31-3.94 U/Ml)] were statistically higher, and median levels of fetuin-A [433.52 mg/L (IQR: 387.89-584.49 mg/L)] and TAS (3.10 ± 0.86 U/mL) were lower in patients with AMI than in controls. The parameter with the area under the curve (0.815), sensitivity (73.3%), and specificity (66.7%) closest to those of Hs-cTnT was fetuin-A, followed by MCP-1, TOS, and TAS, respectively. A one-unit increase in MCP-1 levels increased the probability of AMI by 1.023 times (p = 0.002). A one-unit increase in fetuin-A levels decreased the probability of AMI by 0.995 times (p = 0.003). A one-unit increase in serum TOS levels was 1.29 times more characteristic of STEMI than of NSTEMI (p = 0.044). MCP-1, oxidative stress parameters, and fetuin-A might support Hs-cTnT levels in the early diagnosis of AMI. Fetuin-A and MCP-1 levels may be independent risk factors for AMI, whereas TOS could be used to distinguish STEMI from NSTEMI.

Determination of Serum Trace Elements and Oxidative Stress in Bitches with Transmissible Venereal Tumor

Background: This study was conducted to compare the changes in oxidative stress levels with some trace elements in blood serum in healthy bitches and bitches with transmissible venereal tumor (TVT). Methods: A total of 40 bitches were used in the study. According to the genital organ examinations, healthy bitches constituted the control group (n=20) and those with venereal tumors constituted the TVT group (n=20). Total oxidant status (TOS), total antioxidant status (TAS) and oxidative stress index (OSI) were determined spectrophotometrically. In addition, some serum trace element levels such as magnesium (Mg), manganese (Mn), iron (Fe), copper (Cu), zinc (Zn) and selenium (Se) were measured using an ICP-MS analysis device. Result: The serum Se, Mg and Zn levels were lower in the TVT group compared to the control group (p less than 0.01) and serum Fe and Cu levels were higher in the TVT group than in the control group (p less than 0.01). Serum TOS and OSI levels were higher (p less than 0.001) and serum TAS levels were lower (p less than 0.001) in the TVT group compared to the control group. The study results showed that oxidative stress is high in bitches with TVT, and in parallel with this, changes occur in the amount of certain trace elements that are associated with oxidative stress.

Oxidative stress and antioxidant status in threatened preterm labor.

The purpose of this study was the evaluation of total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI) and superoxide dismutase (SOD) levels in women with threatened preterm labor (TPL) and also to compare the levels of these oxidative stress biomarkers of TPL pregnancies that had preterm and term deliveries. This case-control study was conducted on 46 patients diagnosed with TPL and 47 healthy pregnant women matched for gestational age. Patients with threatened preterm labor were divided into two groups: true preterm birth (TPB) group (n = 16) and false preterm birth (FPB) group (n = 30) groups. Maternal serum SOD, TOS and TAS levels were measured by a spectrophotometric method using a commertial kit. OSI level for each patient was calculated by using the formula: (TOS (μmol·H2O2·equiv/L) × 100)/(TAS (μmol·Trolox·equiv/L)). The mean TAS levels of the TPB and FPB groups were significantly lower than those of the control group (0.96 ± 0.3 vs 1.36 ± 0.34, p1 < 0.001; 0.97 ± 0.22 vs 1.36 ± 0.34, p2 < 0.001, respectively). The mean SOD, TOS and OSI levels of the TPB and FPB groups were significantly higher than those of the control group (p < 0.001). There was no significant difference between the TPB and FPB groups for any oxidative stress biomarkers. The maternal serum oxidative stress biomarkers are increased in pregnancies with TPL. However, these are not effective in predicting preterm birth in pregnancies with TPL.

Evaluation of Oxidative Stress Levels and Dynamic Thiol-disulfide Balance in Patients with Retinopathy of Prematurity

Purpose The aim of this study is to evaluate both dynamic thiol-disulfide homeostasis and oxidative stress (OS) levels in patients with retinopathy of prematurity (ROP). Methods A total of 129 infants of <34 weeks gestational age were enrolled in the present study. The thiol-disulfide homeostasis was determined by using the new, cost-effective and fully automated colorimetric method. Total antioxidant status (TAS), Total oxidant status (TOS) and Oxidative stress index (OSI) levels were evaluated. Results We found serum TAS levels were lower while serum TOS and OSI levels were significantly higher in patients with ROP compare to the without ROP group (p < .05). However, native, total and disulfide values were not statistically significant between the groups (p > .05). In addition, we also evaluated the native, total and disulfide levels in patients with ROP according to grades and no statistically significant results were found (p > .05). Low birth weight (p = .001), gestational age (p = .001) and 5-min Apgar score were significantly lower in the ROP group. Conclusion This study revealed that dynamic thiol-disulfide homeostasis was changed in patients with ROP. Increased TOS and decreased TAS levels may be associated with functional reduction of the antioxidant system due to increased OS. This indicate that ROP patients are highly sensitive to OS. The dynamic thiol-disulfide homeostasis may conduce to the pathophysiological mechanism and disease follow-up in patients with ROP. The results of this study show that ROP patients are highly sensitive to oxidative stress.

Effectiveness of coenzyme Q10 on learning and memory and synaptic plasticity impairment in an aged Aβ-induced rat model of Alzheimer's disease: a behavioral, biochemical, and electrophysiological study.

Aging is the major risk factor for Alzheimer's disease (AD), and cognitive and memory impairments are common among the elderly. Interestingly, coenzyme Q10 (Q10) levels decline in the brain of aging animals. Q10 is a substantial antioxidant substance, which has an important role in the mitochondria. We assessed the possible effects of Q10 on learning and memory and synaptic plasticity in aged β-amyloid (Aβ)-induced AD rats. In this study, 40 Wistar rats (24-36 months old; 360-450 g) were randomly assigned to four groups (n = 10 rats/group)-group I: control, group II: Aβ, group III: Q10; 50 mg/kg, and group IV: Q10+Aβ. Q10 was administered orally by gavage daily for 4 weeks before the Aβ injection. The cognitive function and learning and memory of the rats were measured by the novel object recognition (NOR), Morris water maze (MWM), and passive avoidance learning (PAL) tests. Finally, malondialdehyde (MDA), total antioxidant capacity (TAC), total thiol group (TTG), and total oxidant status (TOS) were measured. Q10 improved the Aβ-related decrease in the discrimination index in the NOR test, spatial learning and memory in the MWM test, passive avoidance learning and memory in the PAL test, and long-term potentiation (LTP) impairment in the hippocampal PP-DG pathway in aged rats. In addition, Aβ injection significantly increased serum MDA and TOS levels. Q10, however, significantly reversed these parameters and also increased TAC and TTG levels in the Aβ+Q10 group. Our experimental findings suggest that Q10 supplementation can suppress the progression of neurodegeneration that otherwise impairs learning and memory and reduces synaptic plasticity in our experimental animals. Therefore, similar supplemental Q10 treatment given to humans with AD could possibly provide them a better quality of life.

Tobacco smoke induces oxidative stress and alters pro-inflammatory cytokines and some trace elements in healthy indoor cats.

This study was aimed to assess oxidative stress, pro-inflammatory cytokines and some trace elements in healthy pet cats exposed to environmental tobacco smoke. Forty healthy cats were included in this study. Cats were divided in two groups: Exposed to tobacco smoke (ETS; n = 20) and non-exposed to tobacco smoke (NETS; n = 20). Blood levels of cotinine, total oxidant status (TOS), oxidative stress index (OSI), lipid hydroperoxide (LOOH), protein carbonyl (PCO), advanced oxidative protein products (AOPP), total antioxidant status (TAS), copper, zinc-superoxide dismutase (Cu, Zn-SOD), catalase (CAT), total thiol (T-SH), interferon gamma (INF-γ), tumor necrosis factor (TNF-α), interleukin β (IL-1β), interleukin 6 (IL-6), interleukin-8 (IL-8), inter-leukin 2 (IL-2) and iron (Fe), zinc (Zn), copper (Cu), selenium (Se) levels were measured. Hematological and biochemical parameters were also measured. Serum cotinine, TOS, OSI, PCO, AOPP and LOOH levels were higher, whereas TAS and Cu, Zn-SOD levels were lower in ETS group. In ETS group INF-γ, IL-1β, IL-2, and IL-6 levels were higher. The Cu level was higher in ETS group. Blood reticulocyte number, serum creatinine and glucose were higher in ETS group. It could be concluded that exposure to tobacco smoke in cats impaired the oxidant/antioxidant balance and potentially triggered the release of pro-inflammatory cytokines.

Toxoplasma gondii'li koyunlarda Oksidatif Stresin Glutatyon, Malondialdehit Düzeyi, Oksidatif Stres İndeksi, Toplam Oksidan Durumu ve Toplam Antioksidan Kapasitesi Kullanılarak Değerlendirilmesi

This study was conducted to evaluate oxidative stress using markers such as malondialdehyde (MDA), glu- tathione (GSH), total oxidant status (TOS), total antioxidant capacity (TAC) and oxidative stress index (OSI) in sheep naturally infected with Toxoplasma gondii (T. gondii). A total of 30 Morkaraman breed sheep were used in this study. Blood samples were obtained from 10 healthy control sheep and 20 sheep naturally infected with T. gondii. malondial- dehyde, GSH, TOS and TAC levels were determined in serum samples. The ratio of serum TOS to TAC levels was assessed as OSI. While the serum MDA level increased significantly (P&amp;lt;0.01), the serum TAC and TOS levels de- creased significantly (P&amp;lt;0.01) in naturally infected with T. gondii sheep. There were no different for serum GSH and OSI in between the groups (P˃0.05). Negative correlation (P&amp;lt;0.01) between TAC and OSI and a positive correlation (P&amp;lt;0.01) between TOS and MDA were determined in naturally infected with T. gondii sheep. In conclusion, the results of study suggested that OSI, TAC and MDA could be used as indicator of oxidative stress for sheep naturally infected with T. gondii.

İdiyopatik Konjenital Kataraktı Olan Pediatrik Hastalarda Sistemik ve Oküler Oksidatif Stres Arasındaki İlişkinin İncelenmesi: Kesitsel Çalışma

Objective: To investigate the systemic and ocular oxidant-antioxidant status of pediatric patients with idiopathic congenital cataracts. Material and Methods: This study investigated the serum and aqueous humor samples of 18 cases with idiopathic congenital cataract and the serum samples of 20 age-and sex-matched healthy cases. Total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), total thiol (T-SH), and thiobarbituric acid reactive substances (TBARS) levels in serum and aqueous humor samples were evaluated using spectrophotometric and spectrofluorometric methods. The weight, height, and body mass index parameters were recorded. Results: Study groups had statistically similar serum TAS, TOS, OSI, T-SH, and TBARS levels (p˃0.05). Statistically significant correlations between oxidant-antioxidant status in serum and aqueous humor were not detected in the patient group (p˃0.05). There was a statistically significant positive correlation was observed between the axial length and aqueous humor T-SH levels (r=0.651, p=0.042). Additionally, statistically significant positive correlation was observed between the age and serum TBARS levels in the patient group (r=0.422, p=0.008). Conclusion: In terms of oxidant-antioxidant status, this study did not find a correlation between serum and aqueous humor samples in pediatric patients with idiopathic congenital cataract and no difference in serum samples between study groups. This study suggests that oxidative stress may not be a part of idiopathic cataract development in the pediatric population.

SERUM LEVELS OF OXIDATIVE STRESS MARKERS IN SUBCLINICAL AND OVERT HYPOTHYROIDISM VERSUS CONTROL GROUP IN POPULATION OF KUTAHYA CITY, TURKEY

Background: Overproduction of oxygen-related free radicals and inadequate antioxidant defense are critical in hypothyroidism, as they might lead to future life-threatening diseases. The objectives of this study were to compare serum levels of oxidative stress markers; total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI), paraoxonase-1 (PON-1), and arylesterase (ARYL) in patients with subclinical hypothyroidism (SH) and overt hypothyroidism (OH) versus healthy controls in population of Kutahya city, Turkey.Materials Methods: This cross-sectional study was conducted at Department of Biochemistry, Faculty of Medicine, Kutahya University of Health Sciences, Kutahya, Turkey, from July to December, 2016. Three study groups of 45 each were formed as healthy controls, patients with SH and OH. Serum levels of TOS, TAS, OSI, PON-1, and ARYL were five research variables with ratio scale, but skewed, so described by median and IQR with 95% Cl. Kruskal-Wallis and multiple comparison tests were used for hypotheses testing.Results: Serum levels of TOS and OSI were higher in SH group (p-values 0.032; .0001; respectively) and OH group (p-values .0001; .0001, respectively) than healthy controls, while serum levels of TAS, PON-1 and ARYL were lower in SH group (p-values .0001; .0001; .0001, respectively) and OH group (p-values .0001; .0001; .0001, respectively) than control group.Conclusion: Our study showed that serum levels of TOS and OSI were higher in subclinical and overt hypothyroidism than in healthy controls; while serum levels of TAS, PON-1, and ARYL were lower in subclinical and overt hypothyroidism than in healthy controls.

Evaluation of The Relationship Between Apelin 36 and Oxidative Stress In Patients With General Anxiety Disorder

Objective: The aim of this study was to evaluate the relationship between apelin-36 and oxidative parameters and Generalized Anxiety Disorder (GAD). Apelin which prevents hippocampal neuronal death is an endogenous ligand for G protein bound APJ receptors in the central nervous system. Oxidative Stress were occurred by free radicals which caused apoptosis in the hypothlamus, hippocampus and amygdala regions of the Central Nervous System (CNS).Methods: In this study, 61 patients diagnosed generalized anxiety disorder at psychiatry polyclinic and 55 control subjects were enrolled. The Structured Clinical Interview for DSM-IV Axis I Disorders (SCID-I) and the Hamilton Anxiety Rating Scale (HARS) were used in this study and they were filled by the patients and healthy individuals. Serum apelin-36 level was measured by using an ELISA kit. Total antioxidant status (TAS) and Total Oxidant Status (TOS) in the serum was measured by an automated system. The data were analyzed by using the SPSS.Results: It was found that serum apelin-36, TOS and Oxidative stress index (OSI) levels were significantly lower in patients’ group than controls. In addition, negative correlation was detected between apelin 36 levels and HARS scores, TOS and OSI values in patients’ group. TAS values were found as similar between the patient and control group. There was no correlation between TAS and Apelin 36 in the patient group.Conclusion: In this study it was found that serum apelin-36, TOS and OSI levels were low in GAD. According to the results, Apelin-36 and oxidative stress may play a role in the etiopathogenesis of Generalized Anxiety Disorder.

Oxidative Stress Level in Patients with Subarachnoid Hemorrhage.

One of the antioxidant mechanisms is the dynamic balance between thiol and disulfide, which, in subarachnoid hemorrhage and other chronic diseases, is disrupted in favor of the latter. The two most commonly used oxidative stress (OS) biochemical markers are the oxidative stress index (OSI) value, which indicates the total oxidant status (TOS) and total antioxidant status (TAS) balance, and the thiol-disulfide (TDS) value, which indicates the total thiol (TT) and native thiol (NT) balance. High OS levels require further investigations. We aimed to investigate the OS level in aneurysmal SAH (aSAH) patients. In this clinical prospective study, blood samples were collected from 50 consecutively treated patients with aSAH and 50 volunteers. Serum TOS, TAS, TT, and NT levels were measured using Erel's method via a spectrophotometer. The Glasgow Coma Scale (GCS) scores, Fisher grades, length of hospital stay (LOS), and the Glasgow Outcome Scale (GOS) scores were recorded. Consequently, the OSI and TDS values were calculated in all participants. A statistically significant difference was observed in the TAS, TOS, OSI, and TDS values between the aSAH patients and the controls. The TT and NT values were significantly lower in aSAH patients than in the controls. A correlation was identified between the OSI values and the GCS scores. Although a correlation was observed between the TDS values and the LOS, no correlation was found between the OSI and the TDS values. The OSI and TDS, which are OS indicators, might serve as the additional objective nominal data to evaluate the treatment efficacy and follow-up for SAH patients. Moreover, decreasing the OSI values and increasing the TT values can be used as improvement indicators in the treated aSAH patients. If we can reduce the OS at the early stage of SAH, it could improve the prognosis by reducing both the morbidity and mortality rates. Further randomized investigations are required to prove the findings in this prospective study.

Determination of oxidative, genotoxic, and histopathologic effects of metal pollution on the fish fauna inhabiting Karasu River, Turkey

The water quality of rivers located in areas close to major cities is deteriorating significantly due to increased pollution. The Karasu River, which passes through Erzurum city Turkey, is getting polluted day by day due to domestic and industrial wastewater. In the study, various assessments were made in three different fish species to determine the impact of possible contamination here. Capoeta capoeta, Alburnus mossulensis and Squalius cephalus were caught from four stations of the river. Micronucleus test was applied to find out genotoxic alterations, and plasma total oxidant status (TOS) and total antioxidant status (TAS) were measured to determine oxidative effects. Histological changes in fish brain were also assessed by histopathological alterations index (HAI). The highest serum TOS levels and the lowest TAS levels were detected in fish species inhabiting polluted stations. A high erythrocytic nuclear abnormality frequency and HAI values were observed in all species from the Aşkale stations. The abnormalities observed in the peripheral blood cells were micronucleus, binucleus, kidney-shaped nucleus, notched nucleus, lobed nucleus and bud nucleus. Histological damages like dilatation, congestion, vacuolization in Purkinje layer, infiltration, hyperemia, degenerations, and axonopathy were noticed in all fish brains. It was determined that histopathological changes were more common in fish species inhabiting the polluted stations. S. cephalus showed to be more sensitive to the pollution. The most important result obtained from this study is that the pollution in the river adversely affects the fish species found in the natural fauna of the river. Histopathological abnormalities, genotoxicity and increased oxidative stress detected in fish are the most important indicators of this situation.

The effects of iloprost and beta3 receptor agonist on TRPA1 and TRPC1 immunreactivity in an experimental lower extremty ischemia-reperfusion injury model.

Background/aim In this study, we aimed to investigate the effects of antioxidant iloprost (ILO) and ß3 adrenergic receptor agonist (BRL) on transient receptor potential ankyrin 1 (TRPA1) and transient receptor potential canonical 1 (TRPC1) ion channels on an experimental ischemia and reperfusion injury model in 30 male Wistar albino rats aged 8–10 weeks.Materials and methodsWistar Albino rats aged were divided into 5 equal groups. Group I Sham operation, Group II IR (ischemia-reperfusion) procedure, Group III IR + intravenous ILO administration, Group IV IR + intraperitoneal BRL administration, and Group V IR + intravenous ILO + intraperitoneal BRL administration group. Two ng/kg/min ILO intravenous infusion was applied to the ILO group. A single dose of 5 mcg/kg BRL intraperitoneal was applied to BRL group. TOS (total oxidant status), TRPA1, and TRPC1 levels were measured with ELISA (enzyme linked immunosorbent assay) in serum, immunohistochemical staining in musculus quadriceps femoris tissue. ResultsCompared with the sham group, the IR group had a statistically significant increase in serum levels of TOS (p = 0.004), TRPA1 (p = 0.002), and TRPC1 (p = 0.008) along with TRPA1- and TRPC1-immunoreactivity (p = 0.005, each) in the tissue. When compared with the IR group in terms of serum levels of TRPA1 and tissue TRPA1-immunoreactivity, although there was no statistically significant difference in the IR+Ilo (p = 0.257 and p = 0.429, respectively), IR+Brl (p = 0.024 and p = 0.177, respectively), and IR+Ilo+Brl (p = 0.024 and p = 0.329, respectively) groups, serum levels of TOS and TRPC1 along with tissue TRPC1-immunoreactivity were statistically significantly reduced in the IR+Ilo (p = 0.002, p = 0.008, and p = 0.004, respectively), IR+Brl (p = 0.004, p = 0.008, and p = 0.004, respectively), and IR+Ilo+Brl groups (p = 0.002, p = 0.008, and p = 0.004, respectively). ConclusionIn IR group serum TOS, TRPA1 and TRPC1 levels ,and tissue TRPA1 and TRPC1 immunoreactivity were statistically significant increase when compared to the sham group. In IR+ILO, IR+BRL and IR+ILO+BRL groups serum TRPA1 and tissue TRPA1 immunoreactivity did not change when compared to IR group. Serum TOS and TRPC1 levels, tissue TRPC1 immunoreactivty were statistically significant decreased when compared to IR group. More detailed and expanded population studies are needed to discuss our results.
