In this research, a high-efficiency ratiometric fluorescent probe TZTF for sensitive detecting the fluctuations of acidic pH was synthesized. The fluorescent color changed from bright blue to orange in the pH values range from 8.0 to 1.0 with the pKa value of 5.41. Meanwhile, the probe demonstrated excellent selectivity, good anti-interference ability, prominent reversibility, outstanding photostability, and low cytotoxicity. The detection mechanism of TZTF towards acidic pH was verified through 1H NMR analysis and DFT calculation. Additionally, probe TZTF was also applied to evaluate the freshness of meat samples, as the pH value of meat samples may decrease if they are not stored properly, and the subsequent acid-catalyzed denaturation of proteins may pose a threat to human health through the food chain. More importantly, fluorescence imaging experiments confirmed that TZTF is appropriate for real-time tracking of pH variations in living cells and living zebrafish.
