Allylated ketosulphides of benzothiazole as intermediates for stereoselective synthesis of allyl ketones, allyl thiiranes and dienes

α-Ketosulphides of benzothiazole 1 react with allylic carbonates in the presence of palladium acetate in dichloromethane under mild conditions affording α- and α,α-diallylated ketosulphides 2 in high yields. Reductive desulphurization of 2a–d with tributyltin hydride gives diallylated ketones 3a–d, whereas reduction of mono-allylated 2e–k with sodium borohydride in isopropanol affords allyl episulphides 4e–k prevalently as (Z)-isomers which can be transformed stereoselectively into dienes.

Palladium-catalysed synthesis of α-diallylated ketosulphides of benzothiazole and their transformation into diallyl thiiranes and trienes

α-Ketosulphides of benzothiazole 1 react with allylic carbonates in the presence of palladium acetate in dichloromethane under mild conditions affording α,α-diallylated ketosulphides 2 in high yields. Reduction of 2 with sodium borohydride in isopropanol gives episulphides which in turn can be transformed almost quantitatively into trienes.

The Heck Reaction of N-Protected Vinylglycines with 4-Iodoanisole.

Among N-protected and unprotected vinylglycines tested, N-(benzyloxycarbonyl)vinylglycine (1c) provided the highest yield of the coupling product 3c in the reaction with 1-iodo-4-methoxybenzene (2) in N, N-dimethylformamide in the presence of palladium acetate, sodium bicarbonate, and tetrabutylammonium chloride, whereas none of the desired product was obtained in the reaction with 4-methoxyphenyl trifluoromethanesulfonate (7). The stereoselectivity of the reaction was reversed by employing triethylamine instead of sodium bicarbonate to furnish (Z)-3c predominantly. In the presene of sodium bicarbonate, replacement of the solvent by water improved not only the chemical yield and stereoselectivity but also the optical purity : geometrically pure (E)-3c of 96% ee was formed in a good yield.

Palladium-catalyzed β-arylation of modified vinyl ethers with aryl triflates.

The reaction between modified vinyl ethers and aryl triflates in the presence of palladium acetate is described. High β-regioselectivity and high chemical yields are obtained, provided that the aryl triflates are not ortho- or pera-substituted with electron-donating groups.

Synthesis of Casein-Related Peptides and Phosphopeptides. XIII. Solid Phase Synthesis of GLu-Ser(P)-Leu Through Global Phosphorylation of a Resin-Bound Seryl-Containing Peptide

The solid phase synthesis of CF3CO2H.H-Glu-Ser(P)-Leu-OH by the 'global' 'phosphite-triester' phosphorylation of a polystyrene resin-bound seryl -containing peptide by using either dibenzyl or di -t-butyl N,N- diethylphosphoramidite is described. The seryl -containing peptide resin was prepared by successive dicyclohexylcarbodiimide/1H-benzotriazol-1-ol couplings of Fmoc -Ser(But)-OH and Z- Glu ( OBzl )-OH to Boc-Leu-resin (polystyrene) followed by cleavage of the seryl t-butyl ether group with 90% CF3CO2H/CH2Cl2. The seryl hydroxy group was phosphorylated by the use of either dibenzyl or di -t-butyl N,N-diethylphosphoramidite/1H- tetrazole followed by m-chloroperoxybenzoic acid oxidation of the resultant phosphite triester. With benzyl phosphate protection, the protected Ser(PO3Bzl2)-containing peptide resin was deprotected by high-pressure hydrogenation with palladium acetate. In the case of t-butyl phosphate protection, the t-butyl groups were first cleaved by treatment with 10% CF3CO2H/CH2Cl2 and the Ser(PO3H2) tripeptide was then cleaved from the resin support by high-pressure hydrogenation in the presence of palladium acetate.

Selective preparation of non-symmetrically substituted divinylbenzenes by palladium-catalysed arylations of alkenes with bromobenzoic acid derivatives

Palladium-catalysed arylation of alkenes with the three bromobenzoic acids or their acyl chlorides provides an efficient and selective method for the preparation of non-symmetrically substituted divinylbenzene derivatives. In the presence of palladium acetate and a phosphorus ligand the free acids react as aryl bromides, with the exception of 2-bromobenzoic acid. If palladium acetate is used alone as catalyst, all three bromobenzoyl chlorides react only as aroyl chlorides. Using two different alkenes a given non-symmetrically substituted divinylbenzene can be prepared by four different routes, allowing for an optimum choice of synthesis path. Substituent effects in the aromatic derivatives and the reactivity of the alkenes in arylation are the principal features to be taken into account. The reaction pathway can generally be chosen to give excellent yields in short reaction times at low palladium concentrations.
