Survivin Regulates Bad Gene Expression by Binding to Its Promoter and Modulates Cell Cycle and Apoptosis in Esophageal Carcinoma Cell.

Esophageal cancer (EC) is the eighth most prevalent cancer and the sixth leading cause of cancer-related mortality worldwide. As an antiapoptotic and a proapoptotic protein, respectively, survivin and Bad play an important role in carcinogenesis of the most human cancers including EC. However, the regulatory relationships between them remain unclear. We sought to investigate the effects of survivin knockdown and overexpression on the expression of Bad gene, cell cycle progression, and apoptosis of esophageal carcinoma cell. The mRNA expression levels of survivin and Bad were determined in EC tissue samples. The knockdown and overexpression experiments were performed in ECA109 and KYSE450 cells via transfection with survivin overexpression and shRNA plasmids. A Bad overexpression experiment was conducted to confirm the biological effect on knockdown of survivin via modulating Bad expression. RT-qPCR and Western blot analysis were used to detect mRNA and protein expression, respectively. Cell cycle and apoptosis were analyzed by flow cytometry. The chromatin immunoprecipitation (ChIP) was conducted to determine the binding sites of survivin on the promoter of Bad gene. By analyzing the mRNA expression of survivin and Bad in 40 ESCC patient specimens, we found that the positive expression rate of survivin in tumor tissues (88%, 35/40) was remarkably high, compared with the distal nontumor tissues (48%, 19/40, p < 0.01). On the other hand, the positive expression rate of Bad in tumor tissues (70%, 28/40) was remarkably low, compared with the distal nontumor tissues (95%, 38/40, p < 0.01). Overexpression of survivin decreases Bad mRNA and protein expression and promotes transformation of cell cycle to S phase. Conversely, knockdown of survivin increases Bad mRNA and protein expression and induces cell cycle arrest and apoptosis. Bad overexpression inducing apoptosis of esophageal carcinoma cell shows the similar apoptotic effect with survivin knockdown. ChIP assays indicate that survivin directly binds to the Bad promoter region, diminishing the transcriptional activity of Bad. In conclusion, the result suggested that survivin regulates Bad gene expression by binding to its promoter and modulates cell cycle and apoptosis in esophageal carcinoma cell.

Expression and its clinicopathological significance of Survivin, MSH2 and MSH6 in colorectal cancer

Objective To analyze the changes of Survivin, MSH6 and MSH2 expression in colorectal cancer and explore their relationship with clinical and pathological parameters. Methods 197 cases of colorectal adenocarcinoma and 20 cases of inflammatory intestinal mucosa were detected by immunohistochemistry with Survivin, MSH6 and MSH2, and the correlation between Survivin, MSH6 and MSH2 expression was analyzed. Results In the control group of 20 cases with inflammatory non-tumor intestinal mucosa, the positive expression rate of MSH2, MSH6, Survivin were 95%, 95%, 10%, respectively. While the positive expression rate of MSH2, MSH6, Survivin were 88.3%, 74.1% and 84.3% in 197 cases of colorectal cancer. Survivin positive expression rate in colorectal cancer group was significantly higher than that in inflammatory control group (P 0.05). MSH2 expression was correlated with tumor size and lymph node metastasis (P 0.05). MSH6 expression was related to gender and lymph node metastasis (P 0.05). Colorectal cancer tissues showed positive correlation between MSH6 and MSH2 (r=0.326, P<0.01), positive correlation between MSH2 and Survivin positive expression (r=0.277, P<0.01), and positive correlation between MSH6 and Survivin positive expression (r=0.435, P<0.01). Conclusions The positive expression rate of Survivin in colorectal cancer is high. MSH2, MSH6 and Survivin in colorectal cancer play an important role in the development and progression of colorectal cancer, and can provide evidence for the detection of these three factors, including metastasis risk, prognosis assessment and clinical treatment. Expression of Fhit, hWAPL and Survivin in Cervical Squamous Cell Carcinoma and Significance Thereof

Objective To preliminarily discuss the role of fragile histidine triad (FHIT) gene, human wings apart-like (hWAPL) gene, survivin gene in lymphatic metastasis of cervical squamous cell carcinoma. Method Expression of FHIT, hWAPL and survivin in 30 cases of normal squamous epithelium, 30 cases of CIN III and 60 cases of cervical squamous cell carcinoma was detected using the immunohistochemical technique. A statistical analysis was conducted for the relationship between the expression levels of FHIT, hWAPL and survivin and clinical pathologic parameters through expression count LVD of FHIT. Results FHIT value is 1.93±0.57 in the group of normal squamous epithelium, 2.87±0.63 in the group of CIN III and 4.12±0.72 in the group of cervical squamous cell carcinoma. The difference has statistical significance (P<0.05). The positive expression of hWAPL in normal cervical tissues is 0.00% (0/30), the positive expression rate is 36.67% (11/30) in the group of CIN III, and the positive expression rate is 63.33% (38/60) in the group of cervical squamous cell carcinoma. The difference has statistical significance (P<0.01). The positive expression rate of survivin in normal cervical tissues is 0.00% (0/30), the positive expression rate is 30.00% (9/30) in the group of CIN III, the expression rate is 61.67% (37/60) in the group of cervical squamous cell carcinoma. The difference has statistical significance (P<0.01). Conclusion FHIT, hWAPL and survivin promote lymphangiogenesis during occurrence and progression of cervical squamous cell carcinoma and lymphangiogenesis plays a vital role in lymphatic metastasis of cervical squamous cell carcinoma.

Correlations of MRI manifestations with survivin gene expression in primary hepatic carcinoma.

To investigate the correlations of magnetic resonance imaging (MRI) manifestations with survivin gene expression in primary hepatocellular carcinoma (HCC). A total of 84HCC patients receiving partial hepatectomy in the Surgery Department and Oncology Department in our hospital from April 2011 to May 2014 were recruited. At 1week before operation, MRI was used to examine the imaging features of liver, a certain size of area was defined and the signal value of each sequence was recorded. HCC and para-carcinoma tissues were collected after operation, and the expression levels of survivin were detected via immunohistochemistry (IHC). All patients were followed up for 30months after operation, and the Kaplan-Meier survival curve was drawn. The correlations of survivin expression with MRI features and signal parameters of each sequence were analyzed. There was no expression of survivin in normal liver tissues. In HCC and para-carcinoma tissues, the nuclei of positive cells showed brown yellow. The positive expression rate of survivin in HCC tissues was 76.19% (64/84), which was significantly higher than that in para-carcinoma tissues (20.81%, 20/84) (p< 0.05). The overall survival (OS) of patients with high expression of survivin was 12.5months, which was significantly shorter than that of patients with low expression of survivin (17.6months) (p< 0.05). Results of chi-square test showed that the survivin level had no correlations with the MRI scan shape and edge in the tumor area (p> 0.05), but it was significantly correlated with tumor diameter, MRI enhancement features and lymphatic metastasis (p< 0.05). Pearson correlation analyses revealed that the survivin IHC score was not correlated with in-phase T1-weighted gradient-recalled echo (GRE), hepatic arterial-phase T1-weighted GRE, portal venous-phase T1-weighted GRE and equilibrium-phase T1-weighted GRE signals (p> 0.05). Besides, the survivin IHC score was negatively correlated with opposed-phase T1-weighted GRE (r=-0.46, p= 0.038), but positively correlated with T2-weighted fast spin echo signal (r=-0.49, p= 0.025). Survivin is significantly up-regulated in HCC tissues and associated with tumor growth and lymph node metastasis. Clinical detection of survivin level combined with MRI examination might be beneficial for clinical diagnosis and treatment of HCC.

Expressions and significance of the second mitochondria-derived activator of caspase protein and Survivin in tissues of high-grade intraepithelial neoplasia and esophageal squamous cell carcinoma

Objective To detect the expressions of the second mitochondria-derived activator of caspase protein(Smac) and Survivin in tissues of high-grade intraepithelial neoplasia(HGIN)and esophageal squamous cell carcinoma (ESCC), and explore the influence of Smac and Survivin on occurrence and development of ESCC. Methods The retrospective case-control study was conducted. The clinicopathological data of 45 healthy controls with suspected esophagitis (control group), 41 patients with HGIN (HGIN group) and 57 patients with ESCC (ESCC group) who were admitted to the Qinghai University Affiliated Hospital between January 2012 and December 2014 were collected. Observation indicators included: (1) expressions of Smac and Survivin were detected by immunohistochemistry. (2) Expressions of mRNA of Smac and Survivin in tumor tissues and adjacent tissues were detected by real-time polymerase chain reaction (RT-PCR). (3) Correlation between relative expressions of Smac and Survivin in tumor tissues and adjacent tissues. (4) Relationship between expressions of Smac and Survivin in tumor tissues and clinicopathological factors. Measurement data with normal distribution were represented as ±s. The comparisons among groups was evaluated with the one-way ANOVA. The comparison between tumor tissues and adjacent tissues were analyzed by the paired t test, and comparison of rates was done using the chi-square test and Fisher exact probability. The correlation between expressions of Smac and Survivin was done using the Pearson correlation analysis. Results (1) Expressions of Smac and Survivin were detected by immunohistochemistry. The positive expression rate of Smac of esophageal tissues in the control group, HGIN group and ESCC group were respectively 82.2%(37/45), 43.9%(18/41) and 40.4%(23/57), with a statistically significant difference among the 3 groups (χ2=20.408, P 0.05). The positive expression rate of Smac of adjacent tissues in the ESCC group was 84.2%(48/57), with a statistically significant difference between tumor tissues and adjacent tissues (χ2=20.530, P 0.05). The positive expression rate of Survivin of adjacent tissues in the ESCC group was 15.8%(9/57), with a statistically significant difference between tumor tissues and adjacent tissues (χ2=32.386, P 0.05) and in adjacent tissues (r=-0.197, P>0.05). There was a correlation in the relative expression of Smac between tumor tissues and adjacent tissues (r=0.527, P 0.05). (4) The relationship between the expressions of Smac and Survivin in tumor tissues and clinicopathologic factors: the positive expression of Smac in tumor tissues was 51.6% in men, 9.1% in women, 64.7% in tumor diameter<3 cm, 28.0% in tumor diameter≥3 cm, 60.0% in stage Ⅰ-Ⅱ of TNM stage and 22.7% in stage Ⅲ-Ⅳ of TNM stage, respectively, showing statistically significant differences in the positive expression of Smac in tumor tissues of patients with different gender, tumor diameter and TNM stage (χ2=6.093, 5.567, 6.041, P<0.05). The positive expression of Survivin in tumor tissues was 54.5% in high- and moderate-differentiated tumors, 100.0% in low-differentiated tumors, 75.0% in stageⅠ-Ⅱ and 100.0% in stage Ⅲ-Ⅳ, respectively, showing statistically significant differences in the positive expression of Survivin in tumor tissues of patients with different tumor differentiation and TNM stage (P<0.05). Conclusions The expression of Smac in the progression of esophageal epithelial cells towards to high-grade intraepithelial neoplastia and proliferatoin is inhibited, while expression of Survivin is activated. Smac and Survivin participate in the occurrence of ESCC and maybe play an role in determining the biological features of carcinoma tissues. Effects of survivin on FVADT chemotherapy for refractory multiple myeloma.

The present study aimed to investigate the effects of survivin, an apoptosis inhibitor protein, on the efficacy of the fludarabine, vincristine, epirubicin, dexamethasone and thalidomide (FVADT) chemotherapy regime for the treatment of refractory multiple myeloma (MM). A total of 82 patients with MM were selected from the Hematology Inpatient Department at The Second Affiliated Hospital of Zhengzhou University (Zhengzhou, China). The initial treatment group consisted of 40 patients with MM, who received the vincristine, epirubicin and dexamethasone (VAD) chemotherapy regime. The refractory group consisted of 42 patients with refractory MM, who received the FVADT chemotherapy regime. Bone marrow biopsies were collected via marrow aspirations, and the protein expression of survivin was analyzed by immunohistochemistry. In addition, the Kaplan-Meier method was used for survival analyses. Intergroup differences in the protein expression levels of survivin were compared, and the association between survivin expression and the short- and long-term effects of FVADT chemotherapy were analyzed. The positive expression rate of survivin was significantly higher in the refractory group, as compared with the initial treatment group (P<0.05). Furthermore, the complete remission rate and the effective rate were significantly lower in the survivin-positive group, as compared with the survivin-negative group (P<0.05). The overall survival, progression free survival and 1 and 3 year survival rates of the survivin-positive group were significantly higher, as compared with the survivin-negative group (P<0.05). The results of the present study suggested that the protein expression of survivin was upregulated in refractory MM tissues, which was indicative of a poor short- and long-term efficacy for FVADT chemotherapy.

Expression of survivin, livin and enhancer of zeste homolog-2 in bladder cancer tissue

Objective To investigate the expression of survivin, livin and enhancer of zeste homolog-2(EZH-2)in bladder cancer tissues and their correlation with bladder cancer. Methods Eightythree patients pathological diagnosed as bladder cancer were randomly selected as the observation group.Fifty patients with bladder benign lesions served as the control group.The bladder cancer tissues from observation group and the normal bladder mucosa samples from control group were obtained detection of survivin, livin and EZH-2.The relationship between the expression of survivin, livin and EZH-2 with the clinical data was analyzed. All patients were subjected to 5-year outpatient follow-up. Results In the bladder cancer tissues, the positive expression rate of survivin, livin and EZH-2 was 74. 70%, 69. 88% and 83. 13% respectively, significantly higher than in normal bladder tissues correspondingly(P 0. 05), but related to tissue origin and tumor recurrence(P 0. 05), but related with tissue origin, TNM staging, pathological grading and recurrence of the tumor(P< 0. 05). There was a positive correlation between survivin and livin(r= 0. 744, P< 0. 01), or between survivin and EZH-2(r= 0. 717, P< 0. 05), or between livin and EZH-2(r= 0. 683, P< 0. 05). Conclusion The expression of survivin, livin and EZH-2 in bladder cancer tissues was significantly higher than that in normal bladder tissue,and related to the occurrence and development of tumor.The detection of the positive expression rate of survivin, livin and EZH-2 can evaluate the prognosis. Expression of p16 and Survivin in gliomas and their correlation with cell proliferation.

The survival rate of glioma patients is very low, and a lack of effective diagnostic techniques are available at present. The current study aimed to investigate the expression of p16 and Survivin and their association with proliferation and apoptosis in gliomas, as well as patient characteristics and prognosis. In total, 62 glioma specimens were surgically resected and pathologically confirmed at the Zhumadian Central Hospital (Zhumadian, China) between June 2008 and February 2014. Clinical data, including the gender and age of the patients, as well as the location, infiltration degree, size and pathological stage of the glioma, was collected. In order to evaluate the expression of p16 and Survivin in the gliomas, the Ki-67 labeling index was used to evaluate cell proliferation activity. The number of argyrophilic nucleolar organizer regions and the rate of cellular apoptosis was examined using the terminal deoxynucleotidyl transferase dUTP nick end labeling method. The results were analyzed using SPSS 14.0 statistical software. The positive rate of p16 gene expression in the gliomas was 46.77% (29 cases), and p16 gene expression was positively correlated with the differentiation status, tumor size and pre-operative symptoms. The positive rate of Survivin expression in the gliomas was 69.88% (58 cases), and Survivin expression was positively correlated with tumor size, differentiation status and clinical stage. The proliferation activity of the gliomas was enhanced with increasing p16 and Survivin expression, while apoptosis was inhibited. In conclusion, the overexpression of p16 and Survivin was closely associated with uncontrolled cell proliferation and the inhibition of apoptosis in gliomas. The combined analysis of the expression of p16 and Survivin in gliomas may provide guidance with respect to the clinical diagnosis, evaluation, treatment and prognosis of patients with glioma.

Over-expression of survivin and VEGF in small-cell lung cancer may predict the poorer prognosis

The expression of survivin, an inhibitor of apoptosis can be seen in most tumors and is correlated with the angiogenic factor vascular endothelial growth factor (VEGF). But little is known about their contribution in small-cell lung cancer (SCLC). This study was designed to investigate the expression of survivin and VEGF in SCLC, and to explore their correlation with clinical-pathological feature and prognosis. Forty-five patients with pathological histology of SCLC were entered into this study. Forty-five cases of matched adjacent non-tumor samples and 10 samples of operated patients with benign lung tumor were also included as control. The expression of survivin and VEGF was detected by immunohistochemistry (IHC, SP). These two sets of data were processed and tested for correlation with major patients' characteristics, and overall survival. The correlations between survivin and VEGF expressions and the clinical-pathological features were evaluated by chi-square test. The correlation between survivin and VEGF expressions was analyzed by Spearman's rank correlation test; the overall survival was analyzed by the Kaplan-Meier method; and the relationship between clinical and pathological features and overall survival was analyzed by the Cox proportional hazard models. Positive expression rate of survivin and VEGF was significantly higher in SCLC than those of adjacent non-tumor tissues and benign lung tumor tissues (73.3 vs. 15.6 vs. 0 %, P < 0.05) and (75.6 vs. 20 vs. 0 %, P < 0.05), respectively. Survivin and VEGF expressions were significantly associated with lymph node metastasis (P = 0.003, 0.011) and clinical stage (P = 0.006, 0.021). The expression of survivin was significantly coincident with the expression of VEGF (r = 0.644, P = 0.000). The median overall survival in survivin positive group and VEGF positive group was significantly shorter than those in survivin negative and VEGF negative group, respectively (log-rank P = 0.000). Moreover, multivariate analysis showed that survivin expression (HR 0.224; 95 % CI 0.074-0.675; P = 0.008) and VEGF expression (HR 0.172; 95 % CI 0.054-0.559; P = 0.003) were statistically independent predictive factors of poorer prognosis for SCLC patients. Our results indicated that survivin and VEGF were over-expressed in small-cell lung cancer, each of them may be an independent poor prognostic factor.

Correlation between expression of Survivin and cysteinyl aspartate-specific protease and apoptosis in sacral chordoma

Objective To investigate the expression and the clinical significance of Survivin and cysteinyl aspartate-specific protease (Caspase)-3 in human sacral chordoma tissues.Methods The expression of Survivin and Caspase-3 was detected in 30 cases of human sacral chordoma tissues and 10 normal notochord tissues by using streptavidin biotin peroxidase (SP) immunohistochemistry.Results The positive expression rate of Survivin in chordoma was 70.0％,while none of 10 cases in the normal notochord tissues expressed Survivin,with significant difference between two groups (P ＜0.05).The positive rate of Survivin was correlated significantly with tumor invasion of the surrounding muscle and recurrence (P ＜ 0.05).The positive expression rate of Caspase-3 was significantly lower in chordoma (33.3％) than that in normal notochord tissues (90.0％),with significant difference between two groups (P ＜ 0.05).The positive rate of Caspase-3 was correlated significantly with tumor invasion of the surrounding muscle and recurrence (P ＜ 0.05).The expression of Survivin and VEGF was correlated negatively with each other (r =-0.463,P ＜ 0.05).Conclusion The high expression of Survivin in sacral chordoma,possibly through inhibition of Caspase-3 activity,may prevent apoptosis and promote tumor cell proliferation,which may play a significant role in cancer development,progression and prognosis. Expression and clinical significance of Survivin and mutant p53 in bladder urothelial carcinoma

Objective To investigate the expression and significance of Survivin and mutant p53(mp53) in bladder urothelial carcinoma,and the relationship between them.Methods The expression of Survivin and mp53 in 86 bladder urothelial carcinoma specimens surgically resected was detected by using immunohistochemistry SP method.For each specimen,the cancerous tissue,paracancerous tissue and the remote normal mucosa were analyzed and compared.Results The immunohistochemisty revealed that the expression of Survivin and mp53 in bladder urothelial carcinoma was significantly higher than that in paracancerous tissue,and no expression was detectable in normal bladder tissues.Among the pathological grades G1,G2 and G3,the positive expression rate of Survivin and mp53 respectively was 68.00％,85.29％,92.59％,and 72.00％,88.24％,96.30％.The positive expression rate of Survivin and mp53in the clinical stages of Tis-T1 was lower than in the stages of T2-T4 (75.00％ vs.94.12％,P ＜ 0.05 ;80.77％ vs.94.12％,P ＜0.05),and that in the group with lymphatic metastasis was higher than in the group with negative lymphatic metastasis (96.00％ vs.77.05％,P ＜ 0.05 ; 96.00％ vs.81.97％,P ＜0.05).There was significant difference among groups (P ＜ 0.05).Moreover,the expression of Survivin was positively correlated with that of mp53 (r =0.452,P ＜ 0.05).Conclusion The overexpression of Survivin and mp53 was closely correlated with the degree of malignancy,and may play synergetic roles in carcinogenesis and progression of bladder urothelial carcinoma. Correlation of tumor relapse and elevated expression of survivin and vascular endothelial growth factor in superficial bladder transitional cell carcinoma

Survivin and vascular endothelial growth factor (VEGF) are newly discovered tumor markers closely correlated with bladder cancer. We analyzed the expression of survivin and VEGF in paraffin-embedded tumor tissues from 78 patients with bladder transitional cell carcinoma (BTCC) using an immunohistochemistry method. Normal bladder mucosae from 10 non-BTCC cases were also included as a control group. All patients were closely followed up for tumor recurrence after undergoing transurethral resection of bladder tumor procedures. The positive expression rates of survivin and VEGF in superficial BTCC were 66.7% (52/78) and 69.2% (54/78), respectively, which were significantly higher than those in the control group, 0% (0/10). A positive correlation was found between survivin and VEGF expression (r = 0.283, P < 0.01). Thirty-two of 78 patients (41.0%) displayed recurrence during follow-up (median: 47; range: 7-62 months). The tumor recurrence rate in survivin(+) patients was 53.8% (28/52), which was significantly higher than that in survivin(-) patients [15.4% (4/26); P < 0.05]. The recurrence rate in VEGF(+)/ VEGF(-) patients was 50.0% (27/54) and 20.8% (5/24), respectively (P < 0.05). The sensitivity for predicting the relapse of superficial BTCC was 87.5% in the survivin(+) group, 84.4% in the VEGF(+) group, and 78.1% in the survivin(+)/VEGF(+) group, and the specificity was 47.8, 41.3, and 65.2%, respectively. Survivin and VEGF interact and jointly regulate the biological behavior of bladder cancer. Our results suggest that overexpression of survivin and VEGF accompany a higher risk of BTCC recurrence, making survivin and VEGF biomarkers for predicting the relapse of bladder cancer.

Expression and clinical significance of survivin and p63 in diffuse large B-cell lymphoma

Objective To investigate the expression and significance of survivin and p63 in diffuse large B-cell lymphoma (DLBCL). Methods Immunohistochemical staining was used to detect the expression of survivin and p63 in the lymphnode tissues from DLBCL patients and lymph node reactive proliferation.Results The positive expression rate of p63 and survivin proteins in DLBCL patients was 63.5 %(33/52) and 76.9 %(40/52), respectively. p63 was not expressed in lymph node reative proliferation. The positive expression rate of survivin in lymph node reactive proliferation was 20.0 %. The expression rates of survivin and p63 between two groups was significant different. The replase rate of groups with positive expression of survivin and p63 were higher than that of the negative groups. The mean survival in groups with positive expression of survivin and p63 were shorter than that of the negative groups. Conclusion Survivin and p63 are useful in therapeutic and prognostic assessment. Patients with poor prognostic can be screened out in the early period of treatment using expression of survivin and p63, which may help to improve the outcome of diffuse large B-cell lymphoma. Expression and clinical significance of survivin in primary hepatocellular carcinoma

Objective To investgate the expression and the clinical significance of survivin in the primary hepatocellular carcinoma (HCC). Methods S-P immunohistochemical staining was performed to detect the expression of survivin in 50 samples of HCC tissue and 20 of normal liver tissue. Results The positive rate of survivin expression in HCC was 66. 0% (33/50) , which was significantly higher than that in normol liver tissues (negative expression) (P<0. 001). The expression of survivin was correlated with intrahepatic dissemination and multiple masses (P = 0. 019, P=0. 030). The high survivin expression was associated with the survival time less than three years(P = 0. 018). Conclusion The survivin may play certain roles in the oncogenesis and progression of HCC. Subcellular localization of survivin in non-small cell lung cancer

Survivin, a member of inhibitors of apoptosis protein (IAP) family, is expressed in most tumors as well as in different subcelluar units of tumors. This study was to investigate the clinical significance of survivin in different subcellular units in non-small cell lung cancer (NSCLC). The protein expression of survivin was detected by immunohistochemistry (IHC) in the specimens from 51 cases of NSCLC and 21 cases of paracancerous tissues. The relationship between survivin expression and clinical characteristics of patients was analyzed using SPSS 13.0 software. Positive expression of survivin was mainly detected in cytoplasm and / or nucleus of NSCLC tissues, and the positive rates were 49.0%(25/51), 72.5%(37/51), 3.9%(2/51), 27.5%(14/51) and 23.5%(12/51) in cytoplasm only, in cytoplasm, in nucleus only, in nucleus, and both in cytoplasm and nucleus, respectively. The positive expression rate of survivin was significantly higher in NSCLC tissues (76.5%, 39/51) than in paracancerous tissues (19.0%, 4/21) (P=0.000). The expression of survivin in cytoplasm was correlated with differentiation of tumors (P=0.007). Positive staining of survivin in nucleus, both in cytoplasm and nucleus were significantly related to clinical stage and N stage of NSCLC (P<0.05). The positive rate of survivin was higher in III+IV or N1+N2 stage patients than in I+II or N0 stage patients, respectively (P<0.05). The five-year survival rate was lower in patients with positive expression of survivin in nucleus than in those with negative expression in nucleus(P<0.05). The clinical stage and status of recurrence or metastasis were two independent prognostic factors for the survival of NSCLC patients. Expression of survivin might be related to the origin and development of NSCLC. The positive expression of survivin in nucleus might be associated with invasion, progression and poor prognosis of NSCLC.

Expression of Survivin and PTEN in gastric cancer and its clinic significance

Objective To investigate the relationship among Survivin, PTEN and the occurrence, development, soakage and prog-nesis of gastric cancer. Methods The in-situ hybrization technique, immunochemistry and TUNEL method were used to detect the expres-sion of survivin mRNA, PTEN protein and apoptotic cell. Results The positive rate of survivin expression in gastric cancer was obvious higher than that in para-cancer tissue(P <0.01). Moreover, survivin expression rate in advanced clinical stage and low differentiation gastric cancer degree.was higher, compared with para-cancer tissue (P <0.01, P <0.05). The positive rate of PTEN expression in gas-tric cancer was obvious lower than that in pars-cancer tissue (P <0.01). The positive rate of PTEN expression in advanced clinical stage and low differentiation degree of tissue in gastric cancer was lower (P < 0.01). Furthermore, survivin and apoptesis indexes manifested negative relationship in gastric cancer(r =-0.861, P < 0.01), While PTEN and apoptosis indexes manifested positive relationship (r = 0.832, P < 0.01). Conclusions Survivin could be used as a novel marker to detect gastric cancer. PTEN gene lost function, which maybe promote the origin, growth, soakage and metastasis of gastric cancer. Expression of survivin, CDK4, Ki-67 and clinical significance in pediatric acute leukemia

The expression of Survivin, CDK4 and Ki-67 and the clinical significance in pediatric acute leukemia (AL) were investigated. The expression of Survivin, CDK4 and Ki-67 proteins was detected by using immunohistochemical assay in 37 children with AL and 10 children with normal bone marrow as controls. The positive expression rate of Survivin, CDK4 and Ki-67 was 45.9 %, 56.8 %, and 40.5 % respectively in 37 AL children, which was significantly higher than in control group accordingly (P<0.05). The expression of Survivin was positively correlated with CDK4 (P=0.007) and Ki-67 (P=0.008). In conclusion, all Survivin, CDK4 and Ki-67 proteins are over-expressed in pediatric AL and involved in the modulation of apoptosis and proliferation in pediatric AL.

STAT1 and survivin expression in full lymph node examined gastric cancer by using tissue microarray technique

Objective: To characterize the relationship between STAT1 and Survivin expression, and the relationship between them and lymph node metastasis, depth of invasion and prognosis in fully lymph node examined gastric cancer patients of China. Methods: Specimens of curative dissection between 1988 and 2003 were collected from the affiliated hospital of Jianghan University. All 140 patients had complete examination data. All lymph nodes were found by clearing fat method. The interrupted serial 4 μm sections, routine hematoxylin and eosin staining and immunohistochemical methods were used to detect the lymph node metastases. Gastric cancer tissue microarray was formed and the expression of survivin and STAT1 in gastric cancer was detected by immunohistochemical method. All data were processed using Spearman rank correlation analysis, Kaplan-Meyer Log-rank method and Cox multivariate analysis (SPSS 12.0 software). Results: Among 140 gastric cancer tissue microarrays constructed, 110 could be used (utilization rate was 78.6%). 7079 lymph nodes were found in 110 cases (64.4/case). Metastases were found in 89 cases and 1679 lymph nodes. Positive expression rate of survivin and STAT1 was 52.7% (58/110) and 40% (44/110) respectively. There was a significant negative correlation between STAT1 expression and survivin expression (r=−0.19, P=0.04). STAT1 expression had a negative correlation with depth of invasion (r=−0.21, P=0.04). Survivin expression had a negative correlation with UICC N stage (r=−0.24, P=0.01) and histological classification (r=−0.21, P=0.03) by Spearman rank correlation analysis. But survivin and STAT1 expression was not related with prognosis. A significant correlation between lymph node metastasis and prognosis was demonstrated by Cox multivariate analysis (x2=4.85, P=0.028). Conclusion: STAT1 has a negative correlation with survivin expression in gastric cancer. Both of them have no correlation with prognosis in gastric cancer. STAT1 expression can be a molecular marker to predict advanced gastric cancer and survivin a molecular marker of lymph node metastasis in gastric cancer. UICC N stage is the most important prognostic factor in gastric cancer in China.

Expression of TR6 and survivin gene in the tissue of cardiac cancer and its clinical significance

OBJECTIVE:To explore the expression of Survivin gene inhibiting apoptosis in the tissue of cardiac cancer and its correlation with clinicopathological features. METHODS: Two-step immunohistochemistry technology (Envision) was adopted to detect the expression of Survivin gene in 66 cases of cardiac cancer, and the relationship between the expression and clinicopathological features of cardiac cancer was investigated. RESULTS: In cancer tissue and tissues surrounding cancer, the positive expression rates of Survivin were 62.1% (41/66) and 24.2% (16/66) respectively,no Survivin expression was found in normal gastric mucosa, and the positive rates among these three groups had a statistical difference,P0.05. Survivin protein positive expression in low differentiation gastric cardiac carcinoma was 73.8% (31/42), and compared with high and moderate differentiation carcinoma positive rate 41.7% (10/24) there was a statistical difference,χ~2=5.409 4,P0.05. Surrivin protein positive expression rate was 73.3% (33/45) in those patients with lymph node metastasis,while only 38.1% (8/21) in those patients without lymph node metastasis, and a statistical difference was found, χ~2=6.132 4,P0.05. Positive Survivin expression rate was 74.4% in those patients with serosal infiltration while only 44.4% (12/27) in those patients without serosal infiltration,also there was a significant difference between them, χ~2=4.862 8,P0.05. TNM classification stage Ⅲ gastric cardiac carcinoma’s positive Survivin expression rate was 95% (19/20) and it was significantly higher than that of stage Ⅰ and Ⅱ, 47.8% (22/46), χ~2=11.254 4,P0.05. The expression of Survivin gene had a positive correlation with gastric cardiac carcinoma differentiation, clinical staging and lymph node metastasis, but it had no correlation with ages, sex and tumor size of cardiac carcinoma. CONCLUSIONS: Survivina inhibitor of apoptosis may correlate with the carcinogenesis and progression of cardiac carcinoma. The detection of Survivin can be worthwhile to judge the malignance and progression of cardiac cancer.
