Physiological and Proteomic Responses of Contrasting Alfalfa (Medicago sativa L.) Varieties to High Temperature Stress.

High temperature (HT) is an important factor for limiting global plant distribution and agricultural production. As the global temperature continues to rise, it is essential to clarify the physiological and molecular mechanisms of alfalfa responding the high temperature, which will contribute to the improvement of heat resistance in leguminous crops. In this study, the physiological and proteomic responses of two alfalfa (Medicago sativa L.) varieties contrasting in heat tolerance, MS30 (heat-tolerant) and MS37 (heat-sensitive), were comparatively analyzed under the treatments of continuously rising temperatures for 42 days. The results showed that under the HT stress, the chlorophyll content and the chlorophyll fluorescence parameter (Fv/Fm) of alfalfa were significant reduced and some key photosynthesis-related proteins showed a down-regulated trend. Moreover, the content of Malondialdehyde (MDA) and the electrolyte leakage (EL) of alfalfa showed an upward trend, which indicates both alfalfa varieties were damaged under HT stress. However, because the antioxidation-reduction and osmotic adjustment ability of MS30 were significantly stronger than MS37, the damage degree of the photosynthetic system and membrane system of MS30 is significantly lower than that of MS37. On this basis, the global proteomics analysis was undertaken by tandem mass tags (TMT) technique, a total of 6,704 proteins were identified and quantified. Gene Ontology (GO) analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that a series of key pathways including photosynthesis, metabolism, adjustment and repair were affected by HT stress. Through analyzing Venn diagrams of two alfalfa varieties, 160 and 213 differentially expressed proteins (DEPs) that had dynamic changes under HT stress were identified from MS30 and MS37, respectively. Among these DEPs, we screened out some key DEPs, such as ATP-dependent zinc metalloprotease FTSH protein, vitamin K epoxide reductase family protein, ClpB3, etc., which plays important functions in response to HT stress. In conclusion, the stronger heat-tolerance of MS30 was attributed to its higher adjustment and repair ability, which could cause the metabolic process of MS30 is more conducive to maintaining its survival and growth than MS37, especially at the later period of HT stress. This study provides a useful catalog of the Medicago sativa L. proteomes with the insight into its future genetic improvement of heat-resistance.

Photosynthetic Light-Harvesting (Antenna) Complexes-Structures and Functions.

Chlorophylls and bacteriochlorophylls, together with carotenoids, serve, noncovalently bound to specific apoproteins, as principal light-harvesting and energy-transforming pigments in photosynthetic organisms. In recent years, enormous progress has been achieved in the elucidation of structures and functions of light-harvesting (antenna) complexes, photosynthetic reaction centers and even entire photosystems. It is becoming increasingly clear that light-harvesting complexes not only serve to enlarge the absorption cross sections of the respective reaction centers but are vitally important in short- and long-term adaptation of the photosynthetic apparatus and regulation of the energy-transforming processes in response to external and internal conditions. Thus, the wide variety of structural diversity in photosynthetic antenna “designs” becomes conceivable. It is, however, common for LHCs to form trimeric (or multiples thereof) structures. We propose a simple, tentative explanation of the trimer issue, based on the 2D world created by photosynthetic membrane systems.

Small-Angle X-Ray and Neutron Scattering on Photosynthetic Membranes.

Ultrastructural membrane arrangements in living cells and their dynamic remodeling in response to environmental changes remain an area of active research but are also subject to large uncertainty. The use of noninvasive methods such as X-ray and neutron scattering provides an attractive complimentary source of information to direct imaging because in vivo systems can be probed in near-natural conditions. However, without solid underlying structural modeling to properly interpret the indirect information extracted, scattering provides at best qualitative information and at worst direct misinterpretations. Here we review the current state of small-angle scattering applied to photosynthetic membrane systems with particular focus on data interpretation and modeling.

Ultrastructural modeling of small angle scattering from photosynthetic membranes

The last decade has seen a range of studies using non-invasive neutron and X-ray techniques to probe the ultrastructure of a variety of photosynthetic membrane systems. A common denominator in this work is the lack of an explicitly formulated underlying structural model, ultimately leading to ambiguity in the data interpretation. Here we formulate and implement a full mathematical model of the scattering from a stacked double bilayer membrane system taking instrumental resolution and polydispersity into account. We validate our model by direct simulation of scattering patterns from 3D structural models. Most importantly, we demonstrate that the full scattering curves from three structurally typical cyanobacterial thylakoid membrane systems measured in vivo can all be described within this framework. The model provides realistic estimates of key structural parameters in the thylakoid membrane, in particular the overall stacking distance and how this is divided between membranes, lumen and cytoplasmic liquid. Finally, from fitted scattering length densities it becomes clear that the protein content in the inner lumen has to be lower than in the outer cytoplasmic liquid and we extract the first quantitative measure of the luminal protein content in a living cyanobacteria.

LIPID COMPOSITION AND FUNCTIONAL STATE OF MEMBRANE SYSTEMS IN STELLARIA HUMIFUSA

The object of the study is Stellaria humifusa ( Caryophyllaceae ) – a polycarpic herb with a long taproot; an almost Arctic hygrophyte with circumpolar distribution. Plants form mats and grow on rock outcrops near coastal areas. The plant community consists of only one species, and is covered with a layer of dead leaves from the previous year. The study of lipids showed that during active growth S. humifusa has lipid fractions in the functionally active state. The fraction of neutral lipids (a structural component of the cell and its emergency stores) dominates in terms of the percentage of saturated fatty acids (FA). The fraction of phospholipids (the main membrane system of the cell) has the greatest content of FA with high quantities of diene, triene and saturated FA. This fraction contains the maximum amount of linoleic acid. The fraction of glycolipids (lipids of chloroplast membranes) accounts for around 32 % of the plant’s total FA. It indicates a tense lipid metabolism associated with photosynthetic activity. This fraction contains less of diene and saturated FA, but significantly more of triene FA, including linolenic acid, comparing to the other groups The study of AOS activity in S. humifusa leaves revealed relatively low peroxidase activity (2.2 and 1 μmol TG/mg protein at pH 5 and 7.8, respectively) simultaneously with high catalase activity (366 μmol H 2 O 2 /mg protein) as compared to other herbaceous plants. This fact possibly indicates the species is adapted to the environment. The high level of unsaturation of the lipid structure of the photosynthetic apparatus membrane system contributes a lot to the maintenance of its high functional activity in S. humifusa .

Effects of heat stress on photosynthetic characteristics and chloroplast ultrastructure of a heat-sensitive and heat-tolerant cultivar of wucai (Brassica campestris L.)

The hot climate of southern China from late summer to early fall is one of the major factors limiting the yield and quality of wucai (Brassica campestris L.). To study the effects of high temperature on the growth and development of wucai, two cultivars (heat-sensitive ‘WS-6’ and heat-tolerant ‘WS-1’) were subjected to three heat stress treatments for 5 days. The two cultivars were evaluated for plant growth, photosynthetic characteristics, oxidative damage, and activities of antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). Heat stress treatments higher than 30 °C resulted in significant changes for both cultivars in plant growth (plant height, stem diameter, and single plant weight) and photosynthetic performance [net photosynthetic rate (PN), stomatal conductance (gs), intercellular CO2 concentration (Ci), and leaf transpiration rate (E)]. The chlorophyll (Chl) content was markedly reduced by heat stress, inhibiting photochemical activity. However, larger decreases in growth and photosynthetic parameters occurred under heat stress in WS-6, compared with WS-1. Furthermore, microscopy also revealed greater severity of damage to the photosynthetic apparatus and membrane system of WS-6 under heat stress, compared with WS-1. These effects were probably due to a greater accumulation of reactive oxygen species (ROS) and malondialdehyde (MDA) in WS-6, relative to WS-1. Moreover, higher photochemical efficiency of photosystem II (PSII) and greater activities of antioxidant enzymes were present in WS-1, compared with WS-6. Our results indicated that the extent of heat stress damage in wucai was cultivar dependent. The heat-tolerant cultivar WS-1 had a greater capacity for self-protection and self-regulation under heat stress.

Acute Effects of a Spinach Extract Rich in Thylakoids on Satiety: A Randomized Controlled Crossover Trial

Objective: By retarding fat digestion, thylakoids, the internal photosynthetic membrane system of green plants, promote the release of satiety hormones. This study examined the effect of consuming a single dose of concentrated extract of thylakoids from spinach on satiety, food intake, lipids, and glucose compared to a placebo.Design: Sixty overweight and obese individuals enrolled in a double-blind randomized crossover study consumed the spinach extract or placebo in random order at least a week apart. Blood was drawn for assessments of lipids and glucose before a standard breakfast meal, followed 4 hours later by a 5 g dose of the extract and a standard lunch. Visual analog scales were administered before lunch and at intervals until an ad libitum pizza dinner served 4 hours later. Two hours after lunch a second blood draw was conducted. Mixed models were used to analyze response changes.Results: Compared to placebo, consuming the spinach extract reduced hunger (p < 0.01) and longing for food over 2 hours (p < 0.01) and increased postprandial plasma glucose concentrations (p < 0.01). There were no differences in plasma lipids and energy intake at dinner, but males showed a trend toward decreased energy intake (p = 0.08).Conclusions: At this dose, the spinach extract containing thylakoids increases satiety over a 2-hour period compared to a placebo. Thylakoid consumption may influence gender-specific food cravings.

Nowadays,with the increase of ozone precursor caused by photochemical reactions,the extent of global plants suffering from ozone stress in the troposphere is getting more and more serious. Ozone pollution has been considered to be the primary cause of forest decline and tree dieback in Eastern and Western Europe and throughout the United States. Ozone stress could have strong impacts on the utilization of light and the production of photosynthesis products in plants,and lead to the decline of photosynthetic rate through stomatal limitation and non-stomatal limitation. The effects of ozone on plants are closely related to the accumulation of metabolic substance in plants. Various defensive protection mechanisms are initiated by ozone stress,such as antioxidant system and membrane system. Ozone stress has changed the absorption of carbon and mineral nutrients in plants and caused their redistribution. And it could also induce deep changes in gene expression. In order to adapt to the environment with ozone stress,plants adjust their physiological and biochemical mechanism to ensure their vital movement. For example,the content of permeable mass would be regulated by cell to maintain osmotic potential balance,meanwhile the metabolic pathway would be changed to keep energy reserves and reduce metabolic rate in plants. And all kinds of antioxidant enzymes' activity in the cell would increase to clear up free radicals and mitigate or even avoid cell damages. It' s clear that the ecological environment has important effects on biological evolution. And it's confirmed by the plants' physiological effects from ozone stress,which is also another kind of evidence of biological evolutionism. In this paper,the impacts that ozone does to plants' photosynthetic physiology,respiratory metabolism,antioxidant system,membrane system,nutrients absorption and allocation,and molecular physiology were summarized to get a more comprehensive understanding of the physiological and biochemical mechanism of plants adapting to ozone stress. Hotspots for future ozone researches should be focused on the enhancement of effects of ozone stress on plant physiology in individual and community level,the gene regulation and related mechanism of signal transmission network system of plants under ozone stress,plants breeding to adapt to the environment with ozone stress by methods such as molecular marker,gene mapping,genomics approach and transgenic technology,research condition that should be close to natural environment as much as possible,the effects of ozone stress on major physiological functions of subtropical and tropical forests,and the models to assess the impacts of ozone on plants.

Effects of 24-epibrassinolide on the photosynthetic characteristics, antioxidant system, and chloroplast ultrastructure in Cucumis sativus L. under Ca(NO3)2 stress

The effects of 0.1 μM 24-epibrassinolide (EBL) on plant growth (plant height, leaf area, fresh weight, and dry weight), chlorophyll content, photosynthetic characteristics, antioxidant enzymes, and chloroplast ultrastructure were investigated using cucumber seedlings (Cucumis sativus L. cv. Jinyou No. 4) with 80 mM Ca(NO(3))(2) to induce stress. The presence of Ca(NO(3))(2) caused significant reductions in net photosynthetic rate (P(N)), stomatal conductance (Gs), intercellular CO(2) concentration (Ci), and transpiration rate (Tr) of leaves. In addition, Ca(NO(3))(2) markedly reduced the chlorophyll content and inhibited photochemical activity, including the actual photochemical efficiency (ΦPSII). In contrast, EBL increased the chlorophyll content, especially chlorophyll b, and minimized the harmful effects on photosynthesis caused by the Ca(NO(3))(2). The application of EBL to the plants subjected to Ca(NO(3))(2)-enhanced photochemical activity. EBL protected the photosynthetic membrane system from oxidative damage due to up-regulating the capacity of the antioxidant systems. Microscopic analyses revealed that Ca(NO(3))(2) affected the structure of the photosynthetic apparatus and membrane system and induced damage of granal thylakoid layers, while EBL recovered the typical shape of chloroplasts and promoted the formation of grana. Taken together, EBL compensated for damage/losses by Ca(NO(3))(2) due to the regulation of photosynthetic characteristics and the antioxidant system.

Amino acid-assimilating phototrophic heliobacteria from soda lake environments: Heliorestis acidaminivorans sp. nov. and ‘Candidatus Heliomonas lunata’

Two novel taxa of heliobacteria, Heliorestis acidaminivorans sp. nov. strain HR10B(T) and 'Candidatus Heliomonas lunata' strain SLH, were cultured from shoreline sediments/soil of Lake El Hamra (Egypt) and lake water/benthic sediments of Soap Lake (USA), respectively; both are highly alkaline soda lakes. Cells of strain HR10B were straight rods, while cells of strain SLH were curved rods. Both organisms were obligate anaerobes, produced bacteriochlorophyll g, and lacked intracytoplasmic photosynthetic membrane systems. Although the absorption spectrum of strain HR10B was typical of other heliobacteria, that of strain SLH showed unusually strong absorbance of the OH-chlorophyll a component. Major carotenoids of both organisms were OH-diaponeurosporene glucosyl esters, as in other alkaliphilic heliobacteria, and both displayed an alkaliphilic and mesophilic phenotype. Strain HR10B was remarkable among heliobacteria in its capacity to photoassimilate a number of carbon sources, including several amino acids. Nitrogenase activity was observed in strain HR10B, but not in strain SLH. The 16S ribosomal RNA gene tree placed strain HR10B within the genus Heliorestis, but distinct from other described species. By contrast, strain SLH was phylogenetically more closely related to neutrophilic heliobacteria and is the first alkaliphilic heliobacterium known outside of the genus Heliorestis.

Chloroplast lipid synthesis and lipid trafficking through ER–plastid membrane contact sites

Plant chloroplasts contain an intricate photosynthetic membrane system, the thylakoids, and are surrounded by two envelope membranes at which thylakoid lipids are assembled. The glycoglycerolipids mono- and digalactosyldiacylglycerol, and sulfoquinovosyldiacylglycerol as well as phosphatidylglycerol, are present in thylakoid membranes, giving them a unique composition. Fatty acids are synthesized in the chloroplast and are either directly assembled into thylakoid lipids at the envelope membranes or exported to the ER (endoplasmic reticulum) for extraplastidic lipid assembly. A fraction of lipid precursors is reimported into the chloroplast for the synthesis of thylakoid lipids. Thus polar lipid assembly in plants requires tight co-ordination between the chloroplast and the ER and necessitates inter-organelle lipid trafficking. In the present paper, we discuss the current knowledge of the export of fatty acids from the chloroplast and the import of chloroplast lipid precursors assembled at the ER. Direct membrane contact sites between the ER and the chloroplast outer envelopes are discussed as possible conduits for lipid transfer.

Genetic analysis of Arabidopsis mutants impaired in plastid lipid import reveals a role of membrane lipids in chloroplast division

The biogenesis of photosynthetic membranes in plants relies largely on lipid import from the endoplasmic reticulum (ER) and this lipid transport process is mediated by TGD proteins in Arabidopsis. Such a dependency of chloroplast biogenesis on ER-to-plastid lipid transport was recently exemplified by analyzing double mutants between tgd1-1 or tgd4-3 and fad6 mutants. The fad6 mutants are defective in the desaturation of membrane lipids in chloroplasts and therefore dependent on import of polyunsaturated lipid precursors from the ER for constructing a competent thylakoid membrane system. In support of a critical role of TGD proteins in ER-to-plastid lipid trafficking, we showed that the introduction of the tgd mutations into fad6 mutant backgrounds led to drastic reductions in relative amounts of thylakoid lipids. Moreover, the tgd1-1 fad6 and tgd4-3 fad6 double mutants were deficient in polyunsaturated fatty acids in chloroplast membrane lipids, and severely compromised in the biogenesis of photosynthetic membrane systems. Here we report that these double mutants are severely impaired in chloroplast division. The possible role of membrane lipids in chloroplast division is discussed.

Lipid Transport Mediated by Arabidopsis TGD Proteins is Unidirectional from the Endoplasmic Reticulum to the Plastid

The transfer of lipids between the endoplasmic reticulum (ER) and the plastid in Arabidopsis involves the TRIGALACTOSYLDIACYLGLYCEROL (TGD) proteins. Lipid exchange is thought to be bidirectional based on the presence of specific lipid molecular species in Arabidopsis mutants impaired in the desaturation of fatty acids of membrane lipids in the ER and plastid. However, it was unclear whether TGD proteins were required for lipid trafficking in both directions. This question was addressed through the analysis of double mutants of tgd1-1 or tgd4-3 in genetic mutant backgrounds leading to a defect in lipid fatty acid desaturation either in the ER (fad2) or the plastid (fad6). The fad6 tgd1-1 and fad6 tgd4-3 double mutants showed drastic reductions in the relative levels of polyunsaturated fatty acids and of galactolipids. The growth of these plants and the development of photosynthetic membrane systems were severely compromised, suggesting a disruption in the import of polyunsaturated fatty acid-containing lipid species from the ER. Furthermore, a forward-genetic screen in the tgd1-2 dgd1 mutant background led to the isolation of a new fad6-2 allele with a marked reduction in the amount of digalactosyldiacylglycerol. In contrast, the introduction of fad2, affecting fatty acid desaturation of lipids in the ER, into the two tgd mutant backgrounds did not further decrease the level of fatty acid desaturation in lipids of extraplastidic membranes. These results suggest that the role of TGD proteins is limited to plastid lipid import, but does not extend to lipid export from the plastid to extraplastidic membranes.

Inelastic neutron scattering study of light-induced dynamics of a photosynthetic membrane system

Inelastic neutron scattering was employed to study photoeffects on the molecular dynamics of membranes of the photosynthetic bacterium Rhodopseudomonas viridis. The main photoactive parts of this biomolecular system are the chlorophyll molecules whose dynamics were found to be affected under illumination by visible light in a twofold manner. First, vibrational modes are excited at energies of 12(2) and 88(21) cm(-1). Second, a partial "freezing" of rotational modes is observed at energies of 1.2(3) and 2.9(5) cm(-1). These results are attributed to a possible coupling between molecular motions and particular mechanisms in the photosynthetic process.

Form Follows Function

MICROBIOLOGY Strains of Prochlorococcus cyanobacteria contribute nearly half of the photosynthesis in the open ocean. Different ecotypes have distinct morphologies. The tiny and spherical MED4 strain holds a 1.66-Mb genome and appears to prefer surface waters. In contrast, the larger (2.41-Mb genome) and ovoid MIT9313 ecotype is abundant in deeper (>50 m) subtropical and tropical waters. By rapid freezing of hydrated cultured cells and cryoelectron microscopy, Ting et al. were able to document a substantially divergent cellular organization and structure in these bacterial strains. The MED4 strain possesses a thinner cell wall and a less extensive intracytoplasmic (photosynthetic) membrane system in comparison to MIT9313. The authors also find differences in key genes required for the biosynthesis of the cell wall peptidoglycan, where the greater similarity of these genes in MIT9313 and Synechococcus WH8102 correlates with their much thicker peptidoglycan layers. Overall, it seems that MIT9313 cells are better adapted for photosynthetic growth at low irradiance levels in deeper waters, although nutrient transport could also influence cell size and shape. — CA J. Cryo-Electron Tomography Reveals the Comparative Three-Dimensional Architecture ofProchlorococcus, a Globally Important Marine Cyanobacterium

In an age of comparative microbial genomics, knowledge of the near-native architecture of microorganisms is essential for achieving an integrative understanding of physiology and function. We characterized and compared the three-dimensional architecture of the ecologically important cyanobacterium Prochlorococcus in a near-native state using cryo-electron tomography and found that closely related strains have diverged substantially in cellular organization and structure. By visualizing native, hydrated structures within cells, we discovered that the MED4 strain, which possesses one of the smallest genomes (1.66 Mbp) of any known photosynthetic organism, has evolved a comparatively streamlined cellular architecture. This strain possesses a smaller cell volume, an attenuated cell wall, and less extensive intracytoplasmic (photosynthetic) membrane system compared to the more deeply branched MIT9313 strain. Comparative genomic analyses indicate that differences have evolved in key structural genes, including those encoding enzymes involved in cell wall peptidoglycan biosynthesis. Although both strains possess carboxysomes that are polygonal and cluster in the central cytoplasm, the carboxysomes of MED4 are smaller. A streamlined cellular structure could be advantageous to microorganisms thriving in the low-nutrient conditions characteristic of large regions of the open ocean and thus have consequences for ecological niche differentiation. Through cryo-electron tomography we visualized, for the first time, the three-dimensional structure of the extensive network of photosynthetic lamellae within Prochlorococcus and the potential pathways for intracellular and intermembrane movement of molecules. Comparative information on the near-native structure of microorganisms is an important and necessary component of exploring microbial diversity and understanding its consequences for function and ecology.

Morphological and ultrastructural characterization of an unusual purple sulfur bacterium from a marine microbial-mat community

An unusual purple sulfur bacterium present in the pink layer of the Ebro Delta microbial mats has been identified through the study of its ultrastructural features. As pure cultures of this bacterium have not been obtained, due to its inability to grow in axenic conditions, only enrichment cultures where it reached at least 90% of total biomass have been considered. In enrichment cultures, the cells are rods that are 5.4 ± 0.6 μm wide and 11.0 ± 2.1 μm long. The color of the cell suspensions is pink to pinkish-red. Cells are motile by means of a polar tuft of flagella and multiply by binary fission. This bacterium possesses an extensive internal photosynthetic membrane system consisting of stacks of lamellae, contains bacteriochlorophyll a and carotenoids of normal spirilloxanthin series and deposits sulfur intracellularly. In natural samples, the most abundant phototrophic purple sulfur bacterium developing in this ecosystem has the same kind of intracytoplasmic membrane system, but the cells differ slightly in size and arrangement, in that they are ovoid, 4.2 ± 0.5 μm wide and 6.0 ± 1.4 μm long, and can be seen forming irregular non-motile clumps which are embedded in slime. Differences observed between field samples and enrichment cultures suggest that environmental conditions may bring about changes in the phenotypic characteristics of the cells. The morphological characteristics of the described bacterium resemble those of large Chromatia. However, this bacterium differs from known species in this group, particularly in terms of its photosynthetic membrane system and in its light absorption properties.

Chloroplast ultrastructural development in vascular bundle sheath cells of two different maize (Zea mays L.) genotypes

The leaves of maize have two photosynthesizing tissues with two types of chloroplasts, mesophyll cells (MC) and vascular bundle sheaths cells (BSC). The development of chloroplasts in BSC was followed by transmission electron microscopy and point counting method in the middle part of the third leaf of maize plants. From young (Y) to mature (M) leaves, volume density of photosynthetic membrane system (thylakoids) increased, to senescing (S) leaves it did not significantly change. During the whole leaf ontogeny, small thylakoid appression regions (grana) were present in BSC chloroplasts, currently assumed to be agranal. From M to S leaves, volume density of starch inclusions strongly decreased and that of plastoglobuli strongly increased.

Thiocapsa marina sp. nov., a novel, okenone-containing, purple sulfur bacterium isolated from brackish coastal and marine environments.

Four marine, phototrophic, purple sulfur bacteria (strains 5811T, 5812, BM-3 and BS-1) were isolated in pure culture from different brackish to marine sediments in the Mediterranean Sea, the White Sea and the Black Sea. Single cells of these strains were coccus-shaped, non-motile and did not contain gas vesicles. The colour of cell suspensions that were grown in the light was purple-red. Bacteriochlorophyll a and carotenoids of the okenone series were present as photosynthetic pigments. Photosynthetic membrane systems were of the vesicular type. Hydrogen sulfide, thiosulfate, elemental sulfur and molecular hydrogen were used as electron donors during photolithotrophic growth under anoxic conditions; carbon dioxide was utilized as the carbon source. During growth on sulfide, elemental sulfur globules were stored inside the cells. In the presence of hydrogen sulfide, several organic substances could be photoassimilated. Comparative 16S rDNA sequence analysis revealed an affiliation of these four strains to the genus Thiocapsa. Both phylogenetic analysis and the results of DNA-DNA hybridization studies revealed that these strains formed a separate cluster within the genus Thiocapsa. Thus, according to phenotypic characteristics and mainly the carotenoid composition, 16S rDNA sequence analysis and DNA-DNA hybridization data, it is proposed that these strains should be classified as a novel species, Thiocapsa marina sp. nov., with strain 5811T (=DSM 5653T=ATCC 43172T) as the type strain.

Photosynthesis genes and their expression in Rhodobacter sphaeroides 2.4.1: a tribute to my students and associates.

This minireview traces the photosynthesis genes, their structure, function and expression in Rhodobacter sphaeroides 2.4.1, as applied to our understanding of the inducible photosynthetic intracytoplasmic membrane system or ICM. This focus has represented the research interests of this laboratory from the late 1960s to the present. This opportunity has been used to highlight the contributions of students and postdoctorals to this research effort. The work described here took place in a much greater and much broader context than what can be conveyed here. The 'timeline' begins with a clear acknowledgment of the work of June Lascelles and William Sistrom, whose foresight intuitively recognized the necessity of a 'genetic' approach to the study of photosynthesis in R. sphaeroides. The 'timeline' concludes with the completed genome sequence of R. sphaeroides 2.4.1. However, it is hoped the reader will recognize this event as not just a new beginning, but also as another hallmark describing this continuum.
