Morphological Transitions of Block Copolymer Micelles: Implications for Mesoporous Materials Ordering

AbstractThe design of block‐copolymer‐based functional materials, including mesoporous membranes and nanoparticles, requires a comprehensive understanding of the hierarchical assembly of block copolymers in selective solvents into micelles and subsequent ordered phases. It is hypothesized that micellar ordering and characteristic assembly can be described using a set of phase parameters that account for entropic and enthalpic interactions. Dissipative particle dynamics (DPD) simulations are used to systematically investigate the self‐assembly of semidiluted block copolymers, resembling isoporous membrane preparation conditions. The effect of Flory–Huggins interaction parameters, block lengths, and concentration on the morphology and polydispersity of the micelles is evaluated. The interaction parameters are mapped into Flory–Huggins theory by considering the block's conformation. These results reveal the effect of polymer concentration and solvent affinity on the morphological transition of the aggregates, in agreement with existing experimental evidence. It is identified that monodisperse‐spherical micelles in solution are fundamental to stabilize ordered states. Weak solvent segregation of the largest block, curvature of the core‐corona interface, and stretching of the corona‐forming one are found to be key to stabilize monodisperse assemblies. These conditions can be predicted using spherical‐micelles packing considerations and a global phase parameter from the Flory–Huggins theory. This study provides valuable insights into the self‐assembly of diblock copolymers and offers a potential way to optimize the preparation of mesoporous ordered structures and micelle ordering in semidiluted systems.

Photo-responsive anti-fouling polyzwitterionic brushes: a mesoscopic simulation.

The antifouling effects of a toothbrush-shaped photo-responsive polyzwitterionic membrane were studied via dissipative particle dynamics simulations in this work. The results reveal that the membrane modified by spiropyran methacrylate brushes displays photo-switchable and antifouling capability due to the photo-induced ring-opening reaction. Namely, surface morphology and hydrophilicity change in response to visible or UV light irradiation, which can be observed visually by protein adsorption and desorption. Further study indicates that: (1) brush-modification density can influence the structure and properties of the membrane. With low modification density, systems cannot establish an intact selective layer, which hinders the antifouling ability; as the modification density increases, the intact selective layer can be formed, which is conducive to the expression of photo-responsiveness and antifouling capability. (2) Factors of toothbrush-hair length and grafting ratio can influence the establishment of a light-responsive surface: as the grafting ratio and toothbrush-hair length increase, the light-responsive surface is gradually formed, meanwhile, the antifouling ability can be continuously reinforced under UV light irradiation. (3) As the brushes switch into a zwitterionic merocyanine state under UV exposure, the selective layer swelling becomes stronger than that with a hydrophobic spiropyran state under visible exposure. This is owing to the enhanced interaction between zwitterionic brushes and water, which is the root of the antifouling effect. The present work is expected to provide some guidelines for the design and development of novel antifouling membrane surfaces.

Methane-Water Interfacial Tension in Nanopores: A Dissipative Particle Dynamics Study.

Imbibition of fracturing fluid in deep shale nanopores has a significant effect on shale gas production. One of the key parameters affecting imbibition is the interfacial tension of the methane-water system. However, studies on the methane-water interfacial tension in nanopores are very limited, and obtaining the accurate value of the methane-water interfacial tension at the nanoscale is difficult and time-consuming. In this work, a dissipative particle dynamics simulation model was built to study the methane-water interfacial tension in nanopores. This model provides reliable access to methane-water interfacial tension for deep shales under high-temperature, high-pressure conditions at low computation cost. It can be easily used to compute the methane-water interfacial tension in nanopores or the confined space in wide application scenarios. A sensitivity study of methane-water interfacial tension on a variety of factors was conducted. Results demonstrate that under high-pressure conditions, the increase in pressure leads to the rise of interfacial tension. When pressure increases from 20 to 120 MPa, interfacial tension rises from 0.0275 to 0.12 N/m, which contributes to the severe imbibition of fracturing fluid in deep shales. The confinement effect was observed by investigating the influence of pore size. Interfacial tension almost remains unchanged in pores smaller than 7 nm because most of the confined space is occupied by interface layer molecules in these pores. When pore size increases from 7 to 15 nm, the confinement effect is reduced. The interfacial tension experiences a growth from 0.1155 to 0.27 mN/m. Compared with pressure and pore size, the effect of temperature on interfacial tension can be neglected during deep shale gas production.

Understanding the phase behavior during CO2 flooding by dissipative particle dynamics

Understanding the phase behavior of a CO2-oil–water surfactant system is critical for carbon dioxide flooding engineering. Using dissipative particle dynamics, we investigate how the alkanes in crude oil are extracted after the injection of supercritical carbon dioxide into the formation, and what effect this extraction has on the miscible and immiscible displacement of carbon dioxide and crude oil. The images of the extraction are exhibited at the molecular level, and the effects of temperature and pressure on the extraction are investigated. The phase distribution of carbon dioxide, oil, and water under immiscible phase is simulated using actual reservoir fluid data based on a comprehensive understanding of extraction. Oil tends to be spread at the interface of carbon dioxide and water when it is immiscible. The miscible pictures were generated under conditions of reduced CO2-oil–water miscible pressure by adding surfactants to the CO2-oil–water system, and the fluid distribution characteristics under immiscible and miscible phases were examined. dissipative particle dynamics (DPD) simulation demonstrates that when the molar proportion of carbon dioxide is less than 0.4, the solid phase surface can always create a water film of varied thickness, regardless of whether the surface charge of the solid phase is positive or negative. As the molar fraction of carbon dioxide increases, the water film’s thickness decreases.

Effects of side-chain lengths on the structure and properties of anhydrides modified starch micelles: Experimental and DPD simulation studies

Anhydride-modified starch micelles have great potential in the delivery of hydrophobic guest molecules. This study aimed to experimentally explore the effects of side-chain lengths on the structure and properties of anhydride-modified starch micelles, and to visualize the self-assembly and loading process of these micelles through Dissipative particle dynamics (DPD) simulations. Starch micelles could only form when the carbon chain length exceeded four. The highly hydrophobic C18 starch micelle exhibited the minimum particle size (65 nm) and maximum loading capability (59.10 μg/mg). For each addition carbon atom in the anhydride side chains, the critical micelle concentration (CMC) of starch micelles decreased average of 1.79 %. Thermodynamic results showed that the micellization was an entropy-dominated driven process, and longer carbon chains enhanced the stability of starch micelles. DPD results showed that the starch chains formed the small clusters then spherical aggregates and finally core-shell structure spherical micelle. Curcumin was loaded into micelles by adjoint aggregation-micellization-adsorption mechanism. Overall, this study provides microscopic insight into the micellization and drug-loading mechanisms for anhydrides modified starch micelles.

Adhesion dynamics of Janus nanocarriers to endothelial cells: A dissipative particle dynamics study.

Janus nanocarriers (NCs) provide promising features in interfacial applications such as targeted drug delivery. Herein, we use dissipative particle dynamics simulations to study the adhesion dynamics of NCs with Janus ligand compositions to the endothelial cell as a function of a series of effects, such as the initial orientation, ligand density, shape, and size of Janus NCs. The Janus NCs, with its long axis parallel to the endothelial glycocalyx (EG) layer, has the best penetration depth due to its lower potential energy and the lowest shell entropy loss. Among different shapes of Janus NCs, both the potential energy and the EG entropy loss control the penetration. In fact, at the parallel orientations, Janus shapes with a robust mechanical strength and larger surface area at the EG/water interface can rotate and penetrate more efficiently. An increase in the ligand density of Janus NCs increases entropy losses of both the hydrophilic and the hydrophobic ligands and decreases the potential energy. Thus, for a specific Janus NCs, functionalizing with an appropriate ligand density would help driving forces prevail over barriers of penetration into the EG layer. For a particular ligand density, once the radius of the Janus NCs exceeds the appropriate size, barriers such as hydrophobic ligands and shell entropy losses are also reinforced significantly and surpass driving forces. Our observations reveal that entropy losses for hydrophobic ligands of Janus NCs and for the shell of NCs are decisive for the adhesion and penetration of Janus NCs to endothelial cells.

Evolution Kinetics of Stabilizing Pickering Emulsion by Brush-Modified Janus Particles: DPD Simulation and Experimental Insights.

In the present study, we report the evolution of stabilizing Pickering emulsions using brush-modified Janus particles (JPs), utilizing the dissipative particle dynamics (DPD) simulation technique. Our results are subsequently corroborated with experimental findings. Each JP has one-half of the hydrophobic surface, with the other half embedded with hydrophilic polymer brushes grown via atom transfer radical polymerization (ATRP). Our generic simulation model analyzes the chemical kinetics of polymer brush growth on one-half of the initiator-embedded microparticle (MP) surface, resulting in the formation of JP. This involves evaluating monomer conversion and reaction rates. Our results exhibit a substantial influence of the number of JPs, grafted brush density, and brush length on oil-in-water emulsion stability. We studied the evolution kinetics and stability of emulsion formation by analyzing the growth of average domain size and corresponding scaling functions up to a late time limit. This study aims to clarify the connection between the size, quantity, and functionality of JPs and the stability of Pickering emulsions.

Dissipative particle dynamics simulation of proposed artificial filament contraction mechanism using variable external force inspired by nature

ABSTRACT In this numerical research, the proposed mechanism of contraction of an artificial muscle filament with regard to the performance of a natural filament and the possibility of industrial production is presented using the Dissipative Particle Dynamics (DPD) method. First, an artificial myosin component has been simulated, which includes a micro-nozzle with a thin fixed two-end polymer membrane in the middle. The swelling of the limited permeable membrane from one side results from moving the DPD fluid and, consequently, the fluid force will be transferred. Also, by changing the direction of the movement of the fluid, the swelling of the polymer membrane is reversed. It has been shown that the amount of swelling is directly dependent on the amount and direction of external force implementation ( F e = ± 0.001, ± 0.003, ± 0.005 and ± 0.007) and there is a delay (almost one DPD unit) in the amount of displacement when the direction of the flow is changed due to the inertia force effect. The proposed mechanism includes six double artificial myosins, whose myosin movement causes that actin to take contraction in a wave motion considering both forward and backward movements of the limited permeable thin membrane. Also, more artificial myosins may provide smoother contraction.

Mitigating strategy and mechanism of cement slurry contamination caused by oil-based mud: Macro-micro experiments, molecular dynamics simulation and dissipative particle dynamics simulation

During the cementing process, the oil-well cement (OWC) inevitably comes into contact with oil-based mud (OBM), which contaminates the OWC due to the incompatibility between OBM and the OWC. As a solution, hydrophilic surfactants can reduce interfacial tension and enhance the wetting compatibility of oil and water. Therefore, adding surfactants to the OWC is expected to alleviate contamination caused by OBM. In this study, a new strategy was proposed to combine non-ionic surfactants and anionic surfactants to form a composite surfactant (OP-S2), which overcomes the defect that excessive use of anionic surfactants deteriorates the performance of the OWC—showing great potential in adjusting the compatibility between OBM and the OWC and reducing cement contamination. The mechanism of contamination alleviation during the cementing process was elucidated by comparing the anticontamination properties of a single non-ionic surfactant (OP-S1) and OP-S2. In addition, molecular dynamics (MD) and dissipative particle dynamics (DPD) simulations were used to verify the mechanism of different types of surfactants in removing contamination further, providing theoretical guidance for future research on anti-contamination agents.

Hybrid Microneedle-Mediated Transdermal Delivery of Atorvastatin Calcium-Loaded Polymeric Micelles for Hyperlipidemia Therapy.

Hyperlipidemia has been a huge challenge to global health, leading to the cardiovascular disease, hypertension, and diabetes. Atorvastatin calcium (AC), a widely prescribed drug for hyperlipidemia, faces huge challenges with oral administration due to poor water solubility and hepatic first-pass effects, resulting in low therapeutic efficacy. In this work, we designed and developed a hybrid microneedle (MN) patch system constructed with soluble poly(vinyl alcohol) (PVA) and AC-loaded polymeric micelles (AC@PMs) for transdermal delivery of AC to enhance the hyperlipidemia therapy. We first prepared various AC@PM formulations self-assembled from mPEG-PLA and mPEG-PLA-PEG block copolymers using a dialysis method and evaluated the physicochemical properties in combination with experiment skills and dissipative particle dynamics (DPD) simulations. Then, we encapsulated the AC@PMs into the PVA MN patch using a micromold filling method, followed by characterizing the performances, especially the structural stability, mechanical performance, and biosafety. After conducting in vivo experiments using a hyperlipidemic rat model, our findings revealed that the hybrid microneedle-mediated administration exhibited superior therapeutic efficacy when compared to oral delivery methods. In summary, we have successfully developed a hybrid microneedle (MN) patch system that holds promising potential for the efficient transdermal delivery of hydrophobic drugs.

New strategy for structure rearrangement of thin-film composite (TFC) polyamide (PA) membranes and offering potential use in both aqueous and organic solvents

The purity of sodium hexafluorophosphate (NaPF6) is crucial for the preparation of electrolytes in manufacturing. We propose a simple process in order to induce membrane structure rearrangement process (SRP) by activating thin-film composite (TFC) membranes with polyamide (PA) structure and explore their potential green recycling application in concentrating sodium-ion batteries (SIBs) electrolyte. The method utilizes a solution containing polar aprotic solvent as the soaking and activating solution to swell and disintegrate the loosely cross-linked PA layer, and a non-solvent solidification solution complete the SRP, enhancing the permeability of the PA layer while maintaining high rejection rates for divalent ions. Subsequently, during the further treatment of the membrane surface in structure rearrangement, tert-butanol (TBA) is introduced into the aqueous solution to reconstruct the surface PA layer, further improving the permeability. The optimal D8010T10 achieved a permeation flux of 24.2 L m−2 h−1·bar−1, with rejection rates for sodium sulfate (Na2SO4), magnesium chloride (MgCl2), and magnesium sulfate (MgSO4) reaching 94.7 %, 98.4 %, and 99.1 % respectively. And the mechanism of SRP with different solvent is analyzed and explained through dissipative particle dynamics (DPD) simulations. This study provides a commercially viable approach for manufacturing high-performance membranes. The D8010T10 membrane achieves more than 90 % separation efficiency for sodium hexafluorophosphate (NaPF6) dissolved in different solvents, providing a potential solution for the concentration and recovery of NaPF6 in lab-scale, verified its potential use in the organic solvent nanofiltration (OSN) and a path way for waste membrane recycling and degrading use in the industry.

Excitation signal optimization for minimizing fluctuations in knock out slow extraction

The synchrotron is a circular particle accelerator used for high energy physics experiments, material and life science, as well as hadron cancer therapy. After acceleration to the desired energies, particle beams are commonly extracted from the synchrotron using the method of resonant slow extraction. The goal is to deliver a steady particle flux—referred to as spill—to experiments and treatment facilities over the course of seconds while slowly emptying the storage ring. Any uncontrolled intensity fluctuations in the spill are detrimental to the efficiency of beam usage, as they lead to detector pileups or detector interlocks, hindering experiments and cancer treatment. Among the most widely used extraction scheme in medical facilities is the Radio Frequency Knock Out (RF-KO) driven resonant slow extraction, where the stored beam is transversely excited with a radio frequency (RF) field and the spill intensity is controlled by the excitation signal strength. This article presents particle dynamics simulations of the RF-KO system with the focus on finding effective mechanism for minimizing the intensity fluctuations while maintaining a good extraction efficiency and other advantages of KO extraction. An improved beam excitation signal which optimizes these main objectives is found, and is rigorously compared experimentally with other commonly applied techniques.

An individualised 3D computational flow and particle model to predict the deposition of inhaled medicines — A case study using a nebuliser

Background and objective: Drug inhalation is generally accepted as the preferred administration method for treating respiratory diseases. To achieve effective inhaled drug delivery for an individual, it is necessary to use an interdisciplinary approach that can cope with inter-individual differences. The paper aims to present an individualised pulmonary drug deposition model based on Computational Fluid and Particle Dynamics simulations within a time frame acceptable for clinical use. Methods:We propose a model that can analyse the inhaled drug delivery efficiency based on the patient’s airway geometry as well as breathing pattern, which has the potential to also serve as a tool for a sub-regional diagnosis of respiratory diseases. The particle properties and size distribution are taken for the case of drug inhalation by using nebulisers, as they are independent of the patient’s breathing pattern. Finally, the inhaled drug doses that reach the deep airways of different lobe regions of the patient are studied. Results:The numerical accuracy of the proposed model is verified by comparison with experimental results. The difference in total drug deposition fractions between the simulation and experimental results is smaller than 4.44% and 1.43% for flow rates of 60 l/min and 15 l/min, respectively. A case study involving a COVID-19 patient is conducted to illustrate the potential clinical use of the model. The study analyses the drug deposition fractions in relation to the breathing pattern, aerosol size distribution, and different lobe regions. Conclusions:The entire process of the proposed model can be completed within 48 h, allowing an evaluation of the deposition of the inhaled drug in an individual patient’s lung within a time frame acceptable for clinical use. Achieving a 48-hour time window for a single evaluation of patient-specific drug delivery enables the physician to monitor the patient’s changing conditions and potentially adjust the drug administration accordingly. Furthermore, we show that the proposed methodology also offers a possibility to be extended to a detection approach for some respiratory diseases.

Investigation of morphology and structure of drug-loaded PLA-b-PEO-b-PLA polymeric micelle: A dissipative particle dynamics simulations study.

The dissipative particle dynamics (DPD) simulation was used to study the morphologies and structures of the paclitaxel-loaded PLA-b-PEO-b-PLA polymeric micelle. We focused on the influences of PLA block length, PLA-b-PEO-b-PLA copolymer concentration, paclitaxel drug content on morphologies and structures of the micelle. Our simulations show that: (i) with the PLA block length increase, the self-assemble structure of PLA-b-PEO-b-PLA copolymers with paclitaxel vary between onion-like structure (core-middle layer-shell) to spherical core-shell structure. The PEO shell thins and the size of the PLA core increases. The onionlike structures are comprised of the PEO hydrophilic core, the PLA hydrophobic middle layer, and the PEO hydrophilic shell, the distribution of the paclitaxel drug predominantly occurs within the hydrophobic intermediate layer; (ii) The system forms a spherical core-shell structure when a small amount of the drug is added, and within a certain range, the size of the spherical structure increases as the drug amount increases. When the drug contents (volume fraction) cdrug = 10%, it can be observed that the PLA4-b-PEO19-b-PLA4 spherical structures connect to form rod-shaped structures. With the length of PLA block NPLA = 8, as the paclitaxel drug concentrations cdrug = 4%, PEO has been insufficient to completely encapsulate the PLA and paclitaxel drug beads. To enhance drug loading capacity while maintaining stability of the system in aqueous solution, the optimal composition for loading paclitaxel is PLA4-b-PEO19-b-PLA4; the drug content is not higher than 4%; (iii) The paclitaxel-loaded PLA4-b-PEO19-b-PLA4 micelle undergo the transition from onionlike (core-middle layer-shell) to spherical (core-shell) to rod-shaped and lamellar structure as the PLA4-b-PEO19-b-PLA4 copolymer concentration increases from ccp = 10% to 40%.

DPD simulation to reproduce lipid membrane microdomains based on fragment molecular orbital calculations

Lipid domains play a critical role in signal transduction and transport across cell membranes. The formation of domains in “HLC” ternary lipid bilayers composed of high transition temperature (high-Tm) lipids, low-Tm lipids, and cholesterol (Chol) has been extensively studied as a raft-like system. Recently, experiments were performed to control the formation of submicron domains in LLC lipid bilayers containing low-Tm phosphatidylethanolamine (PE), low-Tm phosphatidylcholine (PC), and Chol by manipulating the presence or absence of Chol. The formation of microdomains in this LLC mixture was replicated by dissipative particle dynamics simulation. The results show that domain formation can be replicated.

Channel width-dependent viscosity and slip length in nanoslits and effect of surface wettability

The channel width-dependent behaviors of viscosity (μ) and slip length (ls) in nanoslits are investigated using many-body dissipative particle dynamics simulation in both Poiseuille and Couette flow systems. In both systems, the viscosity and slip length increase as the channel width (w) grows in smaller channels, while they reach bulk values in larger channels. Moreover, as the surface wettability decreases, the slip length is found to increase, while the viscosity remains the same. The channel width-dependent behavior in nanoslits can be explained by the unique structure of the confined fluid. As the channel width narrows, the uniform density profile in the central region diminishes, and an oscillation pattern appears throughout the system. The change in the microstructure with the channel width alters friction between layers of fluid in laminar flow and fluid-solid friction, leading to a w-dependent μ and ls. Nonetheless, the alteration of surface wettability influences only fluid–solid interactions but not the friction between layers of fluid.

Simulating Brownian motion in thermally fluctuating viscoelastic fluids by using the smoothed profile method

To investigate the Brownian motion of individual particles suspended in viscoelastic fluids, the stochastic smoothed profile method (SPm) for direct numerical simulation is developed by extending deterministic SPm for suspensions in viscoelastic media. To simulate viscoelastic flow driven by thermal fluctuations in the suspending medium, the random stress in the fluid momentum equation as well as the random driving force for the conformation tensor in the Oldroyd-B model are incorporated according to the fluctuating hydrodynamics and fluctuating viscoelasticity formalisms. The thermal equilibrium and dynamical properties calculated by using numerical simulations successfully reproduce the analytic predictions, validating the direct simulation for the coupled fluctuating Navier–Stokes and Oldroyd-B equations and for the coupling between the stochastic viscoelastic medium and individual particles. As an application of the stochastic SPm, we investigate finite system-size effects under periodic boundary conditions (PBCs) on the passive microrheological relationship between the mean-square displacement (MSD) of a Brownian particle and the medium's dynamic modulus. Comparing the modulus that was microrheologically calculated from the MSD with the input modulus reveals that the effect of periodic image cell interaction appears not only in the long-time diffusive regime but also in the short-time region. A frequency-dependent finite system-size correction is implemented by phenomenologically extending the long-time diffusive regime correction, allowing passive microrheology analysis under PBCs. This result can be directly applied to other mesoscale numerical simulations including coarse-grained molecular dynamics and dissipative particle dynamics simulations.

Characterization of partial wetting by CMAS droplets using multiphase many-body dissipative particle dynamics and data-driven discovery based on PINNs

The molten sand that is a mixture of calcia, magnesia, alumina and silicate, known as CMAS, is characterized by its high viscosity, density and surface tension. The unique properties of CMAS make it a challenging material to deal with in high-temperature applications, requiring innovative solutions and materials to prevent its buildup and damage to critical equipment. Here, we use multiphase many-body dissipative particle dynamics simulations to study the wetting dynamics of highly viscous molten CMAS droplets. The simulations are performed in three dimensions, with varying initial droplet sizes and equilibrium contact angles. We propose a parametric ordinary differential equation (ODE) that captures the spreading radius behaviour of the CMAS droplets. The ODE parameters are then identified based on the physics-informed neural network (PINN) framework. Subsequently, the closed-form dependency of parameter values found by the PINN on the initial radii and contact angles are given using symbolic regression. Finally, we employ Bayesian PINNs (B-PINNs) to assess and quantify the uncertainty associated with the discovered parameters. In brief, this study provides insight into spreading dynamics of CMAS droplets by fusing simple parametric ODE modelling and state-of-the-art machine-learning techniques.

A Structure Independent Molecular Fragment Interfuse Model for Mesoscale Dissipative Particle Dynamics Simulation of Peptides.

There is a need to develop robust computational models for mesoscale simulation of the structure of peptides over large length scales toward the discovery of novel peptides for medical applications to address the issues of peptide aggregation, enzymatic degradation, and short half-life. The primary objective was to predict the structure and conformation of peptides whose native structures are not known. This work presents a new model for computation of interaction parameters between the beads in coarse-grained dissipative particle dynamics (DPD) simulation that is properly calibrated for amino acids, supports compressibility requirement of water molecules, and accounts for subtle differences in the structure of amino acids and the charge in the side chain of charged amino acids. This new model is referred to as Structure Independent Molecular Fragment Interfuse Model, abbreviated as SIMFIM, because it accounts for specific interactions between different beads, which represent molecular fragments of the amino acids, in calculating nonbonded interaction parameters in the absence of knowing the actual peptide structure. The electrostatic interactions are incorporated in this model by using a normal distribution of charges around the center of the beads to prevent the collapse of oppositely charged soft beads. The uniquely parameterized DPD force field in the SIMFIM model is optimized for a given peptide with respect to the degree of coarse-grained graining for simulating the peptide over long times and length scales. The SIMFIM model was tested in this work using four peptides, namely, TrpZip2, Rubrivinodin, Lihuanodin, and IC3-CB1/Gai peptides, whose structures were sourced from the Protein Data Bank. The SIMFIM model predicted radius of gyration (Rg) values for the peptides closer to the actual structures as compared to the conventional model, and there was less deviation between the predicted and actual structures of the peptides.

Assessing the structural stability and drug encapsulation efficiency of poly(ethylene glycol)-poly(L-lactic acid) nanoparticles loaded with atorvastatin calcium: Based on dissipative particle dynamics

Block polymer micelles have been proven highly biocompatible and effective in improving drug utilization for delivering atorvastatin calcium. Therefore, it is of great significance to measure the stability of drug-loading nano micelles from the perspective of block polymer molecular sequence design, which would provide theoretical guidance for subsequent clinical applications. This study aims to investigate the structural stability of drug-loading micelles formed by two diblock/triblock polymers with various block sequences through coarse-grained dissipative particle dynamics (DPD) simulations. From the perspectives of the binding strength of poly(L-lactic acid) (PLLA) and polyethylene glycol (PEG) in nanoparticles, hydrophilic bead surface coverage, and the morphological alteration of nanoparticles induced by shear force, the ratio of hydrophilic/hydrophobic sequence length has been observed to affect the stability of nanoparticles. We have found that for diblock polymers, PEG3kda-PLLA2kda has the best stability (corresponding hydrophilic coverage ratio is 0.832), while PEG4kda-PLLA5kda has the worst (coverage ratio 0.578). For triblock polymers, PEG4kda-PLLA2kda-PEG4kda has the best stability (0.838), while PEG4kda-PLLA5kda-PEG4kda possesses the worst performance (0.731), and the average performance on stability is better than nanoparticles composed of diblock polymers.
