Hydrogels comprising oxidized carboxymethyl cellulose and water-soluble chitosan at varied oxidation levels: Synthesis, characterization, and adsorptive toward methylene blue

Chitosan, as a biomaterial, has increasingly garnered attention. However, its limited solubility in water—only dissolving in certain dilute acidic solutions—substantially restricts its broader application. In this investigation, chitosan underwent a solubilization modification to acquire water solubility, facilitating its dissolution in neutral aqueous mediums. Subsequently, this water-soluble chitosan (WSC) was interlinked with oxidized carboxymethyl cellulose (OCMC), characterized by varied oxidation extents, to synthesize hydrogels. Structural characterization verified the formation of imine bonds resulting from crosslinking interactions between the amino groups of water-soluble chitosan and the aldehyde groups of oxidized carboxymethyl cellulose. Employing performance characterization analysis, it was discerned that an increase in the oxidation level of the oxidized carboxymethyl cellulose corresponded to a denser hydrogel network architecture and the hardness increased from 3.01 N to 6.16 N. Moreover, the capacity of these hydrogels to adsorb methylene blue was meticulously examined. Notably, the hydrogel denoted as WSC/66%OCMC manifested an adsorption capability of 28.08 mg/g for methylene blue. Analytical findings from adsorption kinetics and isotherm studies indicate that the adsorption mechanism of the WSC/66%OCMC hydrogel follows the pseudo-second-order kinetic model and corresponds to the Freundlich isotherm model.

Designing and synthesis of injectable hydrogel based on carboxymethyl cellulose/carboxymethyl chitosan containing QK peptide for femoral head osteonecrosis healing

Femoral head necrosis is a debilitating disorder that typically caused by impaired blood supply to the hip joint. In this study, a novel injectable hydrogel based on Oxidized Carboxymethyl Cellulose (OCMC)-Carboxymethyl Chitosan (CMCS) polymers containing an angiogenesis stimulator peptide (QK) with a non-toxic crosslinking interaction (Schiff based reaction) was synthesized to enhance angiogenesis following femoral head necrosis in an animal model. The physicochemical features of fabricated injectable hydrogel were analyzed by FTIR, swelling and degradation rate, rheometry, and peptide release. Also, the safety and efficacy were evaluated following an in vitro hydrogel injection study and an avascular necrosis (AVN) animal model. According to the results, the hydrogel exhibited an appropriate swelling ratio and water uptake (>90 %, 24 h) as well as a suitable degradation rate over 21 days accompanied by a continuous peptide release. Also, data showed that hydrogels containing QK peptide boosted the proliferation, differentiation, angiogenesis, and osteogenic potential of both Bone Marrow mesenchymal Stem Cells (BM-MSCs) and human umbilical vein endothelial cells (HUVECs) (****p < 0.0001 and ***p < 0.001, respectively). Furthermore, molecular and histological evaluations significantly demonstrated the overexpression of Runx2, Osteocalcin, Collagen I, VEGF and CD34 genes (**p < 0.01 and ***p < 0.001, respectively), and also femoral head necrosis was effectively prohibited, and more blood vessels were detected in defect area by OCMC-CMCS hydrogel containing QK peptide (bone trabeculae >9000, ***p < 0.001). In conclusion, the findings demonstrate that OCMC-CMCS-QK injectable hydrogel could be considered as an impressive therapeutic construct for femoral head AVN healing.

Tuning a green carboxymethyl cellulose-based pre-tanning agent via peroxide oxidation for high chrome exhaustion in leather industry

The low chrome uptake by collagen in the conventional tanning process leads to the pollution of the wastewater. Due to environmental concerns, leather scientists are already searching for innovative ways to produce pre-tanning agents as a high exhaustion chrome tanning auxiliary. Herein, a novel kind of pre-tanning agent is engineered by converting carboxymethyl cellulose (CMC) to oxidized carboxymethyl cellulose (OCMC) via the hydrogen peroxide process. FT-IR and carboxyl content analysis demonstrated the increase in carboxyl content after oxidation. After that, the obtained OCMC was utilized as a pre-tanning agent, resulting in a high exhaustion of chrome (92.76 %) which is 27.76 % more than conventional chrome tanning (65 %), and the amount of chrome in wastewater reduced to 7.24 %. The hydrothermal stability of wet-blue increased by increasing the uptake of chrome (Ts = 118 °C). The obtained crust leather represented excellent mechanical properties (Tensile strength: 305.68 kg/cm2; tear strength: 50 kg/cm) and desirable organoleptic properties. The environmental analysis signifies a significant step towards a cleaner and sustainable tanning process (COD = 1600, BOD5 = 560 mg/L) compared to the conventional chrome tanning process. Consequently, the obtained results offer a green pre-tanning agent to meet the requirements of the sustainable development of the leather industry.

Fabrication of carboxymethyl cellulose-based thermo-sensitive hydrogels and inhibition of corneal neovascularization

Corneal neovascularization (CNV) is a common multifactorial sequela of anterior corneal segment inflammation, which could lead to visual impairment and even blindness. The main treatments available are surgical sutures and invasive drug injections, which could cause serious ocular complications. To solve this problem, a thermo-sensitive drug-loaded hydrogel with high transparency was prepared in this study, which could achieve the sustained-release of drugs without affecting normal vision. In briefly, the thermo-sensitive hydrogel (PFNOCMC) was prepared from oxidized carboxymethyl cellulose (OCMC) and aminated poloxamer 407 (PF127-NH2). The results proved the PFNOCMC hydrogels possess high transparency, suitable gel temperature and time. In the CNV model, the PFNOCMC hydrogel loading bone morphogenetic protein 4 (BMP4) showed significant inhibition of CNV, this is due to the hydrogel allowed the drug to stay longer in the target area. The animal experiments on the ocular surface were carried out, which proved the hydrogel had excellent biocompatibility, and could realize the sustained-release of loaded drugs, and had a significant inhibitory effect on the neovascularization after ocular surface surgery. In conclusion, PFNOCMC hydrogels have great potential as sustained-release drug carriers in the biomedical field and provide a new minimally invasive option for the treatment of neovascular ocular diseases.

Oxidized carboxymethyl cellulose/polyaniline-based hybrid nanocomposite for sensitive detection of environmentally hazardous nitrophenol in real samples

Present work aims at fabricating chemically active, reproducible, and reliable electrode material by Ag nanoparticles anchored oxidized-carboxymethyl cellulose (O-CMC) embedded Pani (Ag@Pani/O-CMC) based nanocomposite for the trace detection of 4-nitrophenol. The Ag@Pani/O-CMC was physiochemically characterized in detail using FTIR, SEM, TEM, UV–vis, XPS, and XRD to analyze functional groups, structural morphology, optical properties, and chemical compositions. Differential pulse voltammetry and cyclic voltammetry were adopted to explore the electrochemical performance of Ag@Pani/O-CMC modified glassy carbon electrode (Ag@Pani/O-CMC/GCE). The observed reduction peak current was proportional to the 4-NP concentrations with a high sensitivity (2.48 µA nM−1 cm−2) and displayed a detection limit of 0.58 nM and a good linear range of 10–100 nM. In addition, the sensor exhibited comparable sensing capability in real biological samples, including milk, mixed fruit juice, and tap water, with excellent recovery from 97.20 % to 101 %. It also depicted high repeatability as the current response remained at 84.52 % after 28 days of operation for 4-NP detection. This approach can be projected to result in the electrochemical sensor for safe industrial processing and environmental monitoring, which is expected to have good stability, excellent selectivity, and sensitivity for detecting traces of 4-NP in actual samples.

Immobilization of Thermomyces lanuginosus lipase in a novel polysaccharide-based hydrogel by a two-step crosslinking method and its use in the lauroylation of α-arbutin

The Thermomyces lanuginosus lipase (TLLs) was successfully immobilized within a novel hydrogel matrix through a two-step crosslinking method. TLLs were initially crosslinked through the Schiff base reaction by oxidized carboxymethyl cellulose (OCMC). The water-soluble OCMC@TLLs complex was subsequently crosslinked by carboxymethyl chitosan (CMCSH) in a microfluidic apparatus to form the CMCHS/OCMC@TLLs microspheres. The CD (Circular Dichroism, CD) and FT-IR (Fourier Transform infrared spectroscopy, FT-IR) spectra demonstrated that the crosslinking of TLLs with OCMC resulted in a less significant impact on their structure compared to that with glutaraldehyde. CMCHS/OCMC@TLLs showed decreased catalytic performance due to the mass transfer resistance, while its thermal stability was greatly improved. The CMCHS/OCMC@TLLs were used to catalyze the lauroylation of arbutin in tetrahydrofuran. After 12 h of reaction under optimal conditions, the yield of 6′-O-lauryl arbutin reached an impressive 92.12%. The prepared 6′-O-lauryl arbutin has high lipophilicity and exhibits similar tyrosinase inhibitory activity and higher antioxidant activity compared to its parent compound.Graphical

Laponite nanoparticle-crosslinked carboxymethyl cellulose-based injectable hydrogels with efficient underwater-specific adhesion for rapid hemostasis

Tissue adhesives have attracted intense and increasing interest due to their multiple biomedical applications. Despite the rapid development of adhesive hydrogels, huge challenges remain for materials that can ensure strong adhesion and seal hemostasis in aqueous and blood environments. To address this issue, we have developed an innovative design of PAA-based coacervate hydrogel with strong wet adhesion capability through a simple mixture of PAA copolymers with oxidized-carboxymethylcellulose (OCMC), and tannic acid (TA) as the main components, and structurally enhanced with natural clays (Laponite XLG). The absorbed TA provides solid adhesion to dry and wet substrates via multiple interactions, which endows the XLG-enhanced coacervate with the desired underwater adhesive strength. More importantly, the dielectric constant is introduced to evaluate the polarity of the tested samples, which may be used as guidance for the design of mussel-inspired adhesives with even better underwater adhesive properties. In vivo hemorrhage experiments further confirmed that the hydrogel adhesive dramatically shortened the hemostatic time to tens of seconds. Overall, the persistent adhesion and acceptable cytocompatibility of the hydrogel nanocomposite make it a promising alternative suture-free approach for rapid hemostasis at different length scales and is expected to be extended to clinical application for other organ injuries.

Synthesis of oxidized carboxymethyl cellulose/chitosan hydrogels doped with graphene oxide for pH- and NIR-responsive drug delivery

Biological macromolecules hydrogels are synthesized via the Schiff base reaction between oxidized carboxymethyl cellulose (oxCMC) and chitosan (CS), and graphene oxide (GO) and 5-fluorouracil (5-FU) are co-doped in the oxCMC/CS hydrogels during the cross-linking process. The acylhydrazone bonds (–HC = N–) between oxCMC and CS can be easily hydrolyzed in acidic solutions, leading to the swelling of the hydrogels and facilitated delivery of 5-FU. On the other hand, the doped GO is a promising photothermal agent, and the delivery of 5-FU can also be facilitated by the generated hyperthermia upon near infrared (NIR) irradiation. Therefore, pH- and NIR-responsive delivery of 5-FU from the hydrogels can be achieved. The kinetics of drug delivery indicate that the delivery of 5-FU from the hydrogels is controlled by first-order model. The drug-free carrier (oxCMC/CS/GO) has excellent biocompatibility while the developed drug delivery system (oxCMC/CS/GO/5-FU) exhibits high cytotoxicity against human hepatoma SMMC-7721 cells.

Synthesis of Dialdehyde Carboxymethyl Cellulose-Crosslinked-Chitosan for Preconcentration of Cu(II) Using Batch Method

In a batch system, The Cu(II) preconcentration parameters were optimized using chitosan-crosslinked dialdehyde carboxymethyl cellulose (CS-DCMC). The experiment was started with the oxidation of carboxymethyl cellulose by periodate to form DCMC. The CS-DCMC adsorbent was then prepared with a mass ratio of CS-DCMC=1:1. The adsorbent is characterized by FTIR for functional group analysis and SEM-EDX for morphological analysis and the percentage of elements in CS-DCMC adsorbent. In the optimization of preconcentration parameters, the optimal adsorption, desorption, and enrichment factors were obtained at pH 4, adsorbent mass of 90 mg, adsorption time of 95 min, Cu(II) concentrations of 7 mg/L, and Na2EDTA 1 M. The application of CS-DCMC on wastewater containing Cu(II) under optimal conditions gave good results. Wastewater containing Cu(II) 0.1 mg/L can be concentrated to 0.53 mg/L with an enrichment factor of 5.26 times, 0.2 mg/L Cu(II) can be concentrated to 1.08 mg/L with an enrichment factor of 5.40 times, and 0.3 mg/L Cu(II) can be concentrated to 1.68 mg/L with an enrichment factor of 5.59 times. The enrichment factor obtained is almost close to the theoretical enrichment factor of 6 times.

Mussel-inspired self-healing hydrogel form pectin and cellulose for hemostasis and diabetic wound repairing

Diabetic wound is considered as a kind of chronic wound prone to infection and difficult to repair due to high glucose level in the blood of patients. In this research, a biodegradable self-healing hydrogel with mussel inspired bioadhesion and anti-oxidation properties is fabricated based on Schiff-base cross-linking. The hydrogel was designed from dopamine coupled pectin hydrazide (Pec-DH) and oxidized carboxymethyl cellulose (DCMC) for mEGF loading as a diabetic wound repair dressing. The Pectin and CMC as natural feedstock endowed the hydrogel with biodegradability to avoid possible side effects, while the coupled catechol structure could enhance the tissue adhesion of the hydrogel for hemostasis. The results showed the Pec-DH/DCMC hydrogel formed fast and can cover irregular wounds with good sealing effect. The catechol structure also improved the reactive oxygen species (ROS) scavenging ability of the hydrogel, which can eliminate the negative effect of ROS during wound healing. The in vivo diabetic wound healing experiment revealed the hydrogel as mEGF loading vehicle greatly enhanced the diabetic wound repairing rate in mice model. As a result, the Pec-DH/DCMC hydrogel could show advantages as EGF carrier in wound healing applications.

Biodegradable hydrogel from pectin and carboxymethyl cellulose with Silibinin loading for lung tumor therapy

Serious side effects of chemotherapy drugs greatly limited the anticancer performance, while targeted drug delivery could improve the therapeutic effect and reduce side effects. In this work, biodegradable hydrogel was fabricated from pectin hydrazide (pec-H) and oxidized carboxymethyl cellulose (DCMC) for localized Silibinin delivery in lung adenocarcinoma treatment. The self-healing pec-H/DCMC hydrogel showed blood compatibility and cell compatibility both in vitro and in vivo, and could be degraded by enzymes. The hydrogel also formed fast fit for injectable applications and showed sustained drug release characteristic sensitive to pH based on acylhydrzone bond cross-linked networks. The Silibinin, as a specific lung cancer inhibiting drug targets TMEM16A ion channel, was loaded into the pec-H/DCMC hydrogel to treat the lung cancer in mice model. The results showed that the hydrogel loaded Silibinin significantly enhanced the anti-tumor efficiency in vivo and greatly reduced the toxicity of the Silibinin. Based on the dual effect of improving efficacy and reducing side effects, the pec-H/DCMC hydrogel with Silibinin loading have broad application prospects to inhibit lung tumor growth in clinic.

Oxidized carboxymethyl cellulose/gelatin in situ gelling hydrogel for accelerated diabetic wound healing: Synthesis, characterization, and in vivo investigations

Diabetic wounds are chronic wounds that are currently affecting many patient's quality of life. These wounds are challenging because of the impaired healing cycle and harsh environment. In this study in situ gelling hydrogels based on oxidized carboxymethyl cellulose (OCMC) and gelatin (Gel) were used to hasten the healing rate due to their ease of application. The suggested system in this work is synthesized from entirely natural renewable biomaterials to not only achieve the best biocompatibility and biodegradability but also to develop a sustainable product. The rheological studies showed that the hydrogel is turned into a gel after about 30 s of the mixing process. Moreover, the hydrogel can absorb about ten times its weight, keeping the wound hydrated. In vitro biological investigations indicated optimal biocompatibility, antibacterial, and antioxidant activity for faster tissue regeneration. This product was tested in vivo on normal rats and diabetic mice models to treat full-thickness incisional wounds. Results showed that the OCMC-Gel hydrogel is able to hasten the healing rate in both non-diabetic and diabetic wounds. Pathological examinations of the regenerated skin tissue revealed that the OCMC-Gel treated groups developed much more than the control group.

Fabrication of carboxymethyl chitosan/oxidized carboxymethyl cellulose composite film and its assessment for coating preservation of strawberry

In the current study, a novel chitosan-based composite, carboxymethyl chitosan (CMCHS)/oxidized carboxymethyl cellulose (OCMC) was fabricated and characterized. The composite film (CMCHS 1.5%w/v+OCMC 0.8%w/v) was more uniform and had better tensile properties, UV blocking, water vapor permeability, and antifungal properties than pure CMCHS film. Preservation experiments showed that the CMCHS/OCMC film was more efficient for retaining the quality decrease of strawberry during storage. By the end of 7 days' storage, the hardness, contents of organic acid, soluble solids, and reducing sugar of coated strawberries were increased by 35.1%, 38.5%, 14.1%, and 3.5%, respectively, compared to the control group; and the decay rate of strawberries in CMCHS/OCMC group also dropped to 36%, about 42% decrease than that in control, suggesting the promising application of CMCHS/OCMC composite in coating preservation.

Preparation of functional and reactive nanosilver nanogels using oxidized carboxymethyl cellulose

The designing of functional and reactive nanosilver has been carried out by in-situ reduction of silver nitrate using oxidized carboxymethyl cellulose (OCMC). The reduction process is also accompanied by the stabilization of nanoparticles using the OCMC polymer chain, leading to the formation of a structure where nanosilver is entrapped within OCMC gel. The silver nanogels characterized using transmission electron microscopy (TEM) are found to be ∼22 nm. By virtue of the presence of dialdehyde functionality around the silver nanogels, they have the ability to react with a polymer having a complementary functional group. The nanogels have exhibited prominent antimicrobial activity against both gram-negative and gram-positive bacteria. It has been observed that a 0.3 mM concentration of silver nanogel is active in inhibiting bacterial growth. The antibacterial activity of the synthesized Ag nanogels was dose-dependent, with 99.9 % of E. coli and S. aureus destroyed within 5 h at a concentration of 0.4 mM Ag nanogels. The nanogels disrupted the bacterial cell wall and generated reactive oxygen species inside the cell, which resulted in cell death. This investigation provides a very interesting application as a coating for biomedical implants and devices.

In-situ forming hydrogel based on thiolated chitosan/carboxymethyl cellulose (CMC) containing borate bioactive glass for wound healing

Suitable wound dressings for accelerating wound healing are actively being designed and synthesised. In this study, thiolated chitosan (tCh)/oxidized carboxymethyl cellulose (OCMC) hydrogel containing Cu-doped borate bioglass (BG) was developed as a wound dressing to improve wound healing in a full-thickness skin defect of mouse animal model. Thiolation was used to incorporate thiol groups into chitosan (Ch) to enhance its water solubility and mucoadhesion characteristics. Here, the in situ forming hydrogel was successfully developed using the Schiff-based reaction, and its physio-chemical and antibacterial characteristics were examined. Borate BG was also incorporated in the generated hydrogel to promote angiogenesis and tissue regeneration at the wound site. Investigations of in vitro cytotoxicity assays demonstrated that the synthesised hydrogels showed good biocompatibility and promoted cell growth. These results inspired us to investigate the effectiveness of skin wound healing in a mouse model. On the backs of animals, two full-thickness wounds were created and treated utilising two different treatment conditions: (1) OCMC/tCh hydrogel, (2) OCMC/tCh/borate BG, and (3) control defect. The wound closure ratio, collagen deposition, and angiogenesis activity were measured after 14 days to determine the healing efficacy of the in situ hydrogels used as wound dressings. Overall, the hydrogel containing borate BG was maintained in the defect site, healing efficiency was replicable, and wound healing was apparent. In conclusion, we found consistent angiogenesis, remodelling, and accelerated wound healing, which we propose may have beneficial effects on the repair of skin defects.

Synthesis of keto-biomacromolecule derivatives by oxidation of carboxymethyl cellulose using alkaline hexacyanoferrate (III) for alternative applications in biotechnology

The oxidation kinetics of polysaccharides involving carboxymethyl cellulose (CMC) by several oxidizing agent have been investigated in more detail with shedding some light on the synthesis of their respective keto-derivatives as oxidation precursor products. However, the literature survey indicated that no reports have appeared on the oxidation kinetics of CMC or the synthesis of keto-CMC using hexacyanoferrate (III) ion. Therefore, a lack of information on the transfer of electron process in the rate-determining stage as well as the nature of such keto-derivatives obtained as a result of polysaccharides oxidation using various oxidants in aqueous solutions.Accordingly, the present study presents synthesis of diketo CMC as a biomacromolecule derivative using potassium ferricyanide in alkaline media. The experimental results revealed the formation of either monoketo or diketo-derivatives of CMC based on initial molar ratio between the two reactants. The formation of such keto derivatives was confirmed by the reaction of the oxidation products with both 2,4-dinitrophenyl hydrazine and hydroxyl amine as well as FTIR spectra. The reaction kinetics of oxidation showed unity order in [Fe(CN)63−] and fractional first-orders with respect to both [CMC] and [OH−].The obedience of the reaction behavior to the Michael-Menten kinetics was indicative to the formation of 1:1 intermediate complex prior to the rate-determining step as a pathway route for oxidation.

Cellulose-based solid and gel polymer electrolytes with super high ionic conductivity and charge capacity for high performance lithium ion batteries

Polymer electrolytes are one of the most effective alternatives to liquid electrolytes to extend lithium ion batteries (LIBs) safety. However, they suffer from low conductivity and charge capacity. In this work, to overcome these tremendous disadvantages, cellulose acetate (CA) and oxidized carboxymethyl cellulose (OCMC) polymer electrolytes obtained from sugarcane and cotton, respectively, are used as polymer electrolytes in LIBs. Then, the effect of short (citric acid) and long (polyvinyl alcohol (PVA)) chain crosslinker to create polymer networks and solid and gel state of electrolytes on the electrochemical properties are investigated. Gel CA-PVA and solid OCMC-PVA electrolytes indicated the best results. Additionally, each CA and OCMC, in both gel and solid state exhibited ionic conductivity in order of 10−2 S cm−1, charge capacity over 230 mAh g−1, capacity retention of 90% after 100 cycles, excellent electrochemical stability upper than 4.5 V, supreme lithium transfer number (∼ 0.8), and good thermal stability. The electrolytes prepared here bring safe and highly efficient LIBs and are a very serious competitor to liquid electrolytes.

Cellulose-based thermo-responsive hydrogel with NIR photothermal enhanced DOX released property for anti-tumor chemotherapy

Thermo-sensitive hydrogels change their properties through phase transition, which could be used to regulate various behaviors by changing the temperature. In this study, degradable hydrogels with thermo-response were designed by reaction of oxidized carboxymethyl cellulose (CMC-CHO) with functional P(NIPAM-co-AH). The hydrogels showed biocompatibility and thermo-response with lower critical solution temperature (LCST) regulated by weight ratio of P(NIPAM-co-AH)/CMC-CHO. The photo-thermal property of gold nanorod was triggered by the near-infrared (NIR) to enhance the DOX release for in vivo anti-tumor therapy of model mice. The results showed good biocompatibility and biodegradability of the hydrogel both in vitro and in vivo, and the DOX with hydrogel loading reduced the toxicity through sustained release behavior. The anti-tumor performance further enhanced with NIR triggered drug release regulated by photo-thermal property. In conclusion, the injectable P(NIPAM-co-AH)/CMC-CHO based self-healing hydrogels could act as promising drug delivery vehicle for potential localized anti-tumor therapy.

Fluorescent hydrogels based on oxidized carboxymethyl cellulose with excellent adsorption and sensing abilities for Ag+

Heavy metal contamination in water and soil are harmful and destructive to the environment, it has always been regarded as a big problem. Herein, we developed a self-healing fluorescent hydrogel based on oxidized carboxymethyl cellulose with excellent sensing and adsorption abilities for Ag+. The detection and adsorption effects of hydrogels on heavy metal ions were studied. It turned out that the fluorescent hydrogel has sensitive detection and high adsorption capacity for Ag+, the detection limit was 3.798 μM, and the maximum adsorption capacity was 407 mg/g. The adsorption isotherm fitted the Langmuir model well, and the pseudo-secondary model for adsorption kinetics fitted well. The hydrogel could heal itself without external stimulus, it could be easily regenerated 7 times without loss of adsorption performance. In short, the prepared hydrogel has capability of self-healing, detecting and adsorbing heavy metal ions at the same time, good mechanical strength, these all made it a promising long-life adsorbent and provided a new way for wastewater treatment.
