Most of the significant changes in the mean weight and water content of 18 organs of albino rats at 18 h after the rectal administration of an LD50 of acetylsalicylic acid were found to be dose-dependent. Variations in the dose of drug and in the interval after drug administration selected for measurements were factors which affected the relation between mean changes and close–response correlations. Most mean changes in organ weights and water contents in an acute drug toxicity study will probably be dose–dependent. A dose–response correlation of changes in organ weights and water contents is not necessarily indicated in routine acute toxicity studies on new drugs. Gastric ulcers were found in albino rats 18 h after the rectal administration of 0.8 g of acetylsalicylic acid per kg; and over. It is concluded that measurement of organ weights and water contents in representative groups of animals at death and at intervals in survivors of specified doses of the drug under study will provide probably as much information as the more expensive dose–response studies.

Studies on Experimental Hypertension and Vacular Lesion in Albino Rats, Produced by the Adenosinetriphosphates Fraction (2) : EXPERIMENTAL ARTERIOSCLEROSIS IN ALBINO RATS ELICITED BY THE KIDNEY OR MYOCARDIUM ATP-ase FRACTION

The author attempted to produce the arteriosclerosis in various organs in albino rats. Cholesterol, methyl-thiouracil or butter were additionally administered in the albino rats previously loaded with the ATP-ase fraction of the house-rat's kidney and DCA. Besides the vascular lesions in renal arteries as shown in the previous paper, arteriosclerotic changes were produced in the coronary, mesenteric or cerebral arteries, and, in some animals, myocardial fibrosis or cerebral demyelinating lesion were observed. Myocardium ATP-ase fraction was administered in normotensive albino rats, in combination with the loading of cholesterol, methyl-thiouracil or butter. Coronary sclerosis was produced in normotensive albino rats, but no marked vascular lesions were observed in kidney or other organs.

IN contrast to other malignant animal tumours, such as the Ehrlich adenocarcinoma of mice, the Croker mice sarcoma or the Brown–Pierce rabbit epithelioma, which could be transplanted to the embryos of white rats in serial passages1, the tissue of the Rous chick sarcoma could only exceptionally be grown under such conditions. However, in many rats grown from embryos inoculated with minced tissue of the Rous sarcoma we have detected2 multiple cysts. The observations recorded here reveal that white rats are highly susceptible to the Rous sarcoma virus administered both during embryonic life and the first two days of postnatal life.

Autoradiography of tracks from beta-particle emitters in tissues.

Mounted paraffin sections, 2-4μ thick, ˙were stained, dehydrated, allowed to air dry, and given a thin coating of 1 % Plexi-glas solution in chloroform. The chloroform was allowed to evaporate completely in a dry atmosphere. An emulsion whose dried thickness was 100-150μ, was prepared from Ilford G5 type in gel form and glued to the section by means of a 15% solution of shellac in absolute alcohol. The surface of the emulsion was then cleaned with absolute ethyl alcohol, to remove the impermeable shellac layer. The exposure for radiation reaction was made at about 2°C and required, in the conditions of our experiment, about 24 hrs. The emulsions were processed by the “temperature-development method.” With the described procedure, autoradiographs have been obtained of various organs of albino rats, labeled with P32, S35 and other radioisotopes, and very precise localizations of the origin of electron tracks was attempted.
