Double Hydrophilic Hyperbranched Copolymer-Based Lipomer Nanoparticles: Copolymer Synthesis and Co-Assembly Studies

Double hydrophilic, random, hyperbranched copolymers were synthesized via reversible addition–fragmentation chain transfer (RAFT) polymerization of oligo(ethylene glycol) methyl ether methacrylate (OEGMA) and 2-(dimethylamino)ethyl methacrylate (DMAEMA) utilizing ethylene glycol dimethacrylate (EGDMA) as the branching agent. The resulting copolymers were characterized in terms of their molecular weight and dispersity using size exclusion chromatography (SEC), and their chemical structure was confirmed using FT-IR and 1H-NMR spectroscopy techniques. The choice of the two hydrophilic blocks and the design of the macromolecular structure allowed the formation of self-assembled nanoparticles, partially due to the pH-responsive character of the DMAEMA segments and their interaction with -COOH end groups remaining from the chain transfer agent. The copolymers showed pH-responsive properties, mainly due to the protonation–deprotonation equilibria of the DMAEMA segments. Subsequently, a nanoscopic polymer–lipid (lipomer) mixed system was formulated by complexing the synthesized copolymers with cosmetic amphiphilic emulsifiers, specifically glyceryl stearate (GS) and glyceryl stearate citrate (GSC). This study aims to show that developing lipid–polymer hybrid nanoparticles can effectively address the limitations of both liposomes and polymeric nanoparticles. The effects of varying the ionic strength and pH on stimuli-sensitive polymeric and mixed polymer–lipid nanostructures were thoroughly investigated. To achieve this, the structural properties of the hybrid nanoparticles were comprehensively characterized using physicochemical techniques providing insights into their size distribution and stability.

Influence of pH on the Self‐Assembly Properties of Heterofunctional POEGMA‐Based Block Copolymers During RAFT‐PISA

ABSTRACTPOEGMA‐based block copolymers self‐assemblies with surface‐functionalized carboxylic acid or propargyl groups were synthesized by successive reversible addition‐fragmentation chain transfer (RAFT) polymerization of oligo(ethylene glycol) methyl ether methacrylate (OEGMA) and RAFT‐mediated polymerization‐induced self‐assembly (RAFT‐PISA) in dispersion of 2‐(methacryloyloxy)‐N,N,N‐trimethylethanaminium hexafluorophosphate (METAPF6). The temperature responsive properties of the carboxylic acid–terminated POEGMA (POEGMACDP) and propargyl‐terminated POEGMA (POEGMACDPy) were studied in aqueous buffer solutions at pH 2.3 and 9.2. At pH 2.3, POEGMACDP aqueous solution exhibits a cloud point while no cloud point was observed at pH 9.2. POEGMACDPy shows a cloud point at both pH levels. The RAFT‐PISA in dispersion of METAPF6 at 75°C in either pH 2.3 or 9.2 buffer solution using POEGMACDP or POEGMACDPy as macro‐RAFT agents led to block copolymers, as confirmed by DOSY analysis. For POEGMACDP (DPn = 9), DPn,NMR,PMETAPF6 was 47 at pH 2.3 and 27 at pH 9.2 and for POEGMACDPy (DPn = 10), DPn,NMR,PMETAPF6 was 36 at pH 2.3 and 22 at pH 9.2, as shown by 1H NMR spectroscopy. Both in situ POEGMACDP‐b‐PMETAPF6 and POEGMACDPy‐b‐PMETAPF6 self‐assemblies in aqueous solutions exhibited an increase in Dh and PDI when the pH increased from 2.3 to 9.2, as measured by DLS at 20°C. TEM analyses revealed almost spherical self‐assemblies, unaffected by pH or chain‐end functionality.

Micelles of poly[oligo(ethylene glycol) methacrylate] as delivery vehicles for zinc phthalocyanine photosensitizers

Drug-loaded polymeric micelles have proven to be highly effective carrier systems for the efficient delivery of hydrophobic photosensitizers (PSs) in photodynamic therapy (PDT). This study introduces the micellization potential of poly(oligoethylene glycol methyl ether methacrylate) (pOEGMA) as a novel approach, utilizing the hydrophobic methacrylate segments of pOEGMA to interact with highly hydrophobic zinc phthalocyanine (ZnPc), thereby forming a potential micellar drug carrier system. The ZnPc molecule was synthesized from phthalonitrile derivatives and its fluorescence, photodegradation, and singlet oxygen quantum yields were determined in various solvents. In solvents such as tetrahydrofuran, dimethyl sulfoxide, and N,N-dimethylformamide, the ZnPc compound exhibited the requisite photophysical and photochemical properties for PDT applications. The pOEGMA homopolymer was synthesized via reversible addition-fragmentation chain-transfer polymerization, while ZnPc-loaded pOEGMA micelles were prepared using the nanoprecipitation method. Characterization of the pOEGMA, ZnPc, and micelles was conducted using FTIR,1H-NMR, dynamic light scattering, matrix-assisted laser desorption/ionization time-of-flight mass spectrometries, gel permeation chromatography, and transmission electron microscopy. The critical micelle concentration was determined to be 0.027 mg ml-1using fluorescence spectrometry. The drug loading and encapsulation efficiencies of the ZnPc-loaded micelles were calculated to be 0.67% and 0.47%, respectively. Additionally, the release performance of ZnPc from pOEGMA micelles was monitored over a period of nearly 10 d, while the lyophilized micelles exhibited stability for 3 months. Lastly, the ZnPc-loaded micelles were more biocompatible than ZnPc on L929 cell line. The results suggest that the pOEGMA homopolymer possesses the capability to micellize through its methacrylate segments when interacting with highly hydrophobic molecules, presenting a promising avenue for enhancing the delivery efficiency of hydrophobic PSs in PDT. Moreover, it was also deciphered that obtained formulations were highly biocompatible according to cytotoxicity results and could be safely employed as drug delivery systems in further applications.

Preparation and performance evaluation of intelligent plugging agent for thermo‐sensitive water‐based drilling fluid

AbstractIn the process of unconventional oil and gas exploitation, the invasion of a large amount of drilling fluid into the formation is one of the most critical reasons leading to wellbore instability. A temperature‐sensitive polymer (AOS) was synthesized from acrylamide, oligo (ethylene glycol) methyl ether methacrylate (OEGMA), and sodium p‐styrene sulfonate by emulsion polymerization. The material is a solid‐free fluid with good fluidity at room temperature, after injection into the formation, the fluid is gradually transformed into hard gel above the lower critical solution temperature (LCST) (90°C), thus blocking micro‐fractures and improving wellbore stability. Infrared and NMR characterization confirmed the successful preparation of the target product. The transmittance of AOS with ambient temperature was measured by UV–vis spectrophotometer, and the LCST behavior of AOS was verified by rheological and viscoelastic measurements. The interaction mechanism between AOS and sodium bentonite was evaluated according to the American Petroleum Institute standard. The results show that after aging at 180°C, the filtration loss is reduced by 93%, and the medium permeability (100, 200 mD) formation is effectively plugged, with a plugging efficiency is as high as 74.7%.

Four-Component Statistical Copolymers by RAFT Polymerization.

This manuscript serves as the starting point for in-depth research of multicomponent, statistical, methacrylate-based copolymers that potentially mimic the behavior of proteins in aqueous solutions. These synthetic macromolecules are composed of specially chosen comonomers: methacrylic acid (MAA), oligoethylene glycol methyl ether methacrylate (OEGMA475), 2-(dimethylamino)ethyl methacrylate (DMAEMA) and benzyl methacrylate (BzMA). Monomer choice was based on factors such as the chemical nature of pendant functional groups, the polyelectrolyte/polyampholyte and amphiphilic character and the overall hydrophobic-hydrophilic balance (HLB) of the obtained quaterpolymers. Their synthesis was achieved via a one-pot reversible addition fragmentation chain transfer (RAFT) polymerization in two distinct compositions and molecular architectures, linear and hyperbranched, respectively, in order to explore the effects of macromolecular topology. The resulting statistical quaterpolymers were characterized via 1H-NMR and ATR-FTIR spectroscopies. Their behavior in aqueous solutions was studied by dynamic (DLS) and electrophoretic light scattering (ELS) and fluorescence spectroscopy (FS), producing vital information concerning their self-assembly and the structure of the formed aggregates. The physicochemical studies were extended by tuning parameters such as the solution pH and ionic strength. Finally, the quaterpolymer behavior in FBS/PBS solutions was investigated to test their colloid stability and biocompatibility in an in vivo-mimicking, biological fluid environment.

Oligo(ethylene glycol) methyl ether methacrylate based hydrogel with intrinsic antibacterial activity against Pseudomonas aeruginosa as a model of a major wound infecting human pathogen

Wound dressings represent the first important defense line against potentially life-threatening infections. Successful materials should unify efficiency, simplicity of production with the ease of application. Cationic polymers based on quaternary ammonium salt moieties along the polymer chain promise high efficacy against growth of relevant pathogens including drug-resistant strains. We present a novel and easy-to-handle wound dressing material based on the well-defined copolymers of oligo(ethylene glycol) methyl ether methacrylate (OEGMA) and 2-(dimethylamino)ethyl methacrylate (DMAEMA), using quaternization with 1,6-dibromohexane for hydrogel formation. The material obtained from this robust approach was perfectly biocompatible with human dermal fibroblasts and revealed an auspicious degree of intrinsic antibacterial activity against P. aeruginosa as a model pathogen for hospital-acquired infections. Formation of bacterial biofilms was prevented in early- and late-stages and even planktonic cells were killed upon gel contact. The rheological properties of the hydrogel materials were comprehensively characterized in both dry and swollen states. Excellent biological performance and appropriate viscoelastic properties qualify the hydrogels from our production approach as truly promising novel wound dressing materials with long-lasting and contact-active bactericidal activities. We believe that this charmingly simple procedure and the mechanical properties of the hydrogel may open new avenues towards cost-effective next-generation first-line wound-care materials.

Fabricating hybrid DSPC:DOPC:P(OEGMA-co-LMA) structures: Self-assembly as the milestone of their performance

Lipid/polymer nanoparticles are among the most promising hybrid nanoplatforms for pharmaceutical applications, and their many advantages are well established. Despite the efforts, the design parameters that influence their performance have not been deciphered in total yet. In this study, thermodynamic evaluation of lipid and lipid/copolymer bilayers was conducted via differential scanning calorimetry (DSC). Based on their biophysical behavior and by modulating different design parameters for each system, selected lipid/copolymer nanostructures were prepared by the thin film hydration method. The biomaterials of choice were 1,2-dioctadecanoyl-sn-glycero-3-phosphocholine (DSPC), 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), and a linear statistical (random) copolymer comprised of oligo (ethylene glycol) methyl ether methacrylate (OEGMA) and lauryl methacrylate (LMA). Their features were examined by light scattering techniques, fluorescence spectroscopy, and MTS assay. For the first time DSPC:DOPC:P(OEGMA-co-LMA) hybrid systems were successfully formulated and the most prominent factors influencing their thermodynamic, physicochemical, and toxicological properties were investigated. Our results indicate that self-assembly is pivotal for hybrid systems performance and is mediated via random topology of copolymer, lipid composition, hydrophilic to hydrophobic balance, and lipid to polymer ratio. Having in mind fast clinical translation, we hope this work to contribute to a design road map for efficient lipid/copolymer nanostructure development.

Thermoresponsive Smart Copolymer Coatings Based on P(NIPAM-co-HEMA) and P(OEGMA-co-HEMA) Brushes for Regenerative Medicine.

The fabrication of multifunctional, thermoresponsive platforms for regenerative medicine based on polymers that can be easily functionalized is one of the most important challenges in modern biomaterials science. In this study, we utilized atom transfer radical polymerization (ATRP) to produce two series of novel smart copolymer brush coatings. These coatings were based on copolymerizing 2-hydroxyethyl methacrylate (HEMA) with either oligo(ethylene glycol) methyl ether methacrylate (OEGMA) or N-isopropylacrylamide (NIPAM). The chemical compositions of the resulting brush coatings, namely, poly(oligo(ethylene glycol) methyl ether methacrylate-co-2-hydroxyethyl methacrylate) (P(OEGMA-co-HEMA)) and poly(N-isopropylacrylamide-co-2-hydroxyethyl methacrylate) (P(NIPAM-co-HEMA)), were predicted using reactive ratios of the monomers. These predictions were then verified using time-of-flight-secondary ion mass spectrometry (ToF-SIMS) and X-ray photoelectron spectroscopy (XPS). The thermoresponsiveness of the coatings was examined through water contact angle (CA) measurements at different temperatures, revealing a transition driven by lower critical solution temperature (LCST) or upper critical solution temperature (UCST) or a vanishing transition. The type of transition observed depended on the chemical composition of the coatings. Furthermore, it was demonstrated that the transition temperature of the coatings could be easily adjusted by modifying their composition. The topography of the coatings was characterized using atomic force microscopy (AFM). To assess the biocompatibility of the coatings, dermal fibroblast cultures were employed, and the results indicated that none of the coatings exhibited cytotoxicity. However, the shape and arrangement of the cells were significantly influenced by the chemical structure of the coating. Additionally, the viability of the cells was correlated with the wettability and roughness of the coatings, which determined the initial adhesion of the cells. Lastly, the temperature-induced changes in the properties of the fabricated copolymer coatings effectively controlled cell morphology, adhesion, and spontaneous detachment in a noninvasive, enzyme-free manner that was confirmed using optical microscopy.

Stereo-Complex and Click-Chemical Bicrosslinked Amphiphilic Network Gels with Temperature/pH Response.

Stimulus-responsive hydrogels have been widely used in the field of drug delivery because of their three-dimensional pore size and the ability to change the drug release rate with the change in external environment. In this paper, the temperature-sensitive monomer 2-methyl-2-acrylate-2-(2-methoxyethoxy-ethyl) ethyl ester (MEO2MA) and oligoethylene glycol methyl ether methacrylate (OEGMA) as well as the pH-sensitive monomer N,N-Diethylaminoethyl methacrylate (DEAEMA) were used to make the gel with temperature and pH response. Four kinds of physicochemical double-crosslinked amphiphilic co-network gels with different polymerization degrees were prepared by the one-pot method using the stereocomplex between polylactic acid as physical crosslinking and click chemistry as chemical crosslinking. By testing morphology, swelling, thermal stability and mechanical properties, the properties of the four hydrogels were compared. Finally, the drug release rate of the four gels was tested by UV-Vis spectrophotometer. It was found that the synthetic hydrogels had a good drug release rate and targeting, and had great application prospect in drug delivery.

Polymer-Grafted Cellulose Nanofibrils with Enhanced Interfacial Compatibility for Stronger Poly(lactic acid) Composites

Cellulose nanofibrils (CNFs) are a promising reinforcement for biodegradable composite matrices such as poly(lactic acid) (PLA), but they require commercially scalable drying methods that preserve their fibrillar morphology along with improved interfacial interactions with polymer matrices. In this work, a water-based grafting-through polymerization scheme to modify CNFs improved spray drying behavior and reinforcement capacity in PLA composites. All polymer modifications yielded CNFs with a more fibrillar morphology after spray drying, increasing specific surface area by up to 490% compared to unmodified CNFs, values similar to conventional freeze drying. Polymer-grafted CNFs in PLA composites improved the tensile strength by 16% at 20 wt % loading and stiffness by 22% at a 10 wt % loading with two different graft-polymer chemistries compared to unmodified CNF composites. Surface energy heterogeneity measurements of the reinforcements and PLA matrix were employed to understand the improvements in composite properties. Polymer modifications lowered the total surface energy of the CNFs, and calculated ratios of work of adhesion to work of cohesion suggested improved interfacial compatibility for four of the modified CNFs with PLA. Rheological oscillatory shear studies of the composites correlated solid-like melt behavior, as demonstrated by storage moduli dominance, with higher tensile strength. Thermal analysis of the composites revealed that excessive plasticization by the poly(oligoethylene glycol methyl ether methacrylate)-grafted sample potentially offset mechanical property improvements imparted by the more fibrillar morphology. This work provides an opportunity for large-scale manufacturing of CNF/PLA composites via an entirely aqueous modification scheme and industrially relevant spray drying process.

Development of Hybrid DSPC:DOPC:P(OEGMA950-DIPAEMA) Nanostructures: The Random Architecture of Polymeric Guest as a Key Design Parameter.

Hybrid nanoparticles have gained a lot of attention due to their advantageous properties and versatility in pharmaceutical applications. In this perspective, the formation of novel systems and the exploration of their characteristics not only from a physicochemical but also from a biophysical perspective could promote the development of new nanoplatforms with well-defined features. In the current work, lipid/copolymer bilayers were formed in different lipid to copolymer ratios and examined via differential scanning calorimetry as a preformulation study to decipher the interactions between the biomaterials, followed by nanostructure preparation by the thin-film hydration method. Physicochemical and toxicological evaluations were conducted utilizing light scattering techniques, fluorescence spectroscopy, and MTS assay. 1,2-dioctadecanoyl-sn-glycero-3-phosphocholine (DSPC) and 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) in different weight ratios were the chosen lipids, while a linear random copolymer with pH- and thermoresponsive properties comprised of oligo (ethylene glycol) methyl ether methacrylate (OEGMA) and 2-(diisopropylamino) ethyl methacrylate (DIPAEMA) in different ratios was used. According to our results, non-toxic hybrid nanosystems with stimuli-responsive properties were successfully formulated, and the main parameters influencing their overall performance were the hydrophilic/hydrophobic balance, lipid to polymer ratio, and more importantly the random copolymer topology. Hopefully, this investigation can promote a better understanding of the factors affecting the behavior of hybrid systems.

Synthesis and characterization of chitosan-based graft copolymers with temperature-switchable and self-adaptive plugging performance for drilling in microporous formation

The increasing oil–gas explorations in microporous formations and the intensified environmental requirements necessitate the development of high-performance, environmentally friendly and sustainable plugging materials to maintain wellbore stability in drilling engineering. In this work, a series of graft copolymers of carboxymethyl chitosan-g-poly(oligo(ethylene glycol) methyl ether methacrylate-co-acrylic acid) (GCOA) is synthesized through free-radical graft-from polymerization. The oligo(ethylene glycol) methyl ether methacrylate (OEGMA) and acrylic acid (AA) can be well copolymerized in the grafts as confirmed by structural characterizations, and the incorporation of AA improves the grafting ratio and grafting efficiency. All obtained GCOAs possess the temperature-responsive aggregation behavior, which causes the formation of aggregated meso-assemblies from original nano-micelles when heating above their lower critical solution temperature (LCST), accompanied by the phase separation. Meanwhile, the presence of AA segments increases the LCST due to the resistance of enhanced polymer–polymer electrostatic repulsion to the hydrophobic attraction. By the delicate control of AA molar ratio from 0 to 50 mol%, the LCST values of GCOAs can be adjusted from 90 °C to 110 °C. Furthermore, the GCOAs are formulated as smart plugging fluids and the intelligent plugging evaluation method is established based on core flow test with continuous temperature control. The results indicate that the GCOAs can form effective plugging in both low- and median-permeability cores when temperature reaches above a switching point and the formed plugging can be partially removed after cooling, showing the temperature-switchable and self-adaptive plugging capacity as a consequence of reversible retention of GCOA aggregates and thermo-thickening behavior in pore-throat spaces. These features enable the implementation and removal of in-situ plugging in the specific temperature sections during drilling, and provide a new insight into the design of novel smart plugging materials with sustainable and recycling values.

Hydrogels of Acrylic Acid with Oligoethyleneglycol Methacrylate, Carboxymethyl Cellulose and Graphite Oxide

Changing the content of acrylate-based hydrogels allows adjusting their absorption of liquids. In this study, the hydrogels formed by combining carboxymethyl cellulose and graphite oxide with acrylate and oligoethyleneglycol methyl ether methacrylate monomers were investigated by FTIR spectroscopy and SEM and their water absorption capabilities were examined. Swelling tests were performed to investigate the effect of the materials added to the hydrogel structure. The absorption value of hydrogels decreased at the highest and lowest carboxymethyl cellulose concentrations, while the medium carboxymethyl cellulose concentration allowed to obtain the highest absorption. The effect of graphite oxide concentration on water absorption was not detected in the studied concentration range. Due to the biocompatible groups of cellulose and oligoethyleneglycol and biodegradability, the hydrogel has the potential to be used in agricultural and bioengineering applications.

The Preparation of Novel P(OEGMA-co-MEO2MA) Microgels-Based Thermosensitive Hydrogel and Its Application in Three-Dimensional Cell Scaffold.

Thermosensitive hydrogel scaffolds have attracted particular attention in three-dimensional (3D) cell culture. It is very necessary to develop a type of thermosensitive hydrogel material with low shrinkage, and excellent biocompatibility and biodegradability. Here, five types of thermosensitive microgels with different volume phase transition temperature (VPTT) or particle sizes were first synthesized using 2-methyl-2-propenoic acid-2-(2-methoxyethoxy) ethyl ester (MEO2MA) and oligoethylene glycol methyl ether methacrylate (OEGMA) as thermosensitive monomers by free radical polymerization. Their VPTT and particle sizes were investigated by a nanometer particle size meter and an ultraviolet spectrophotometer. The feasibility of using these P(OEGMA-co-MEO2MA) microgels to construct thermosensitive hydrogel by means of the thermal induction method is discussed for the first time. The prepared thermosensitive hydrogel with the optimum performance was screened for in situ embedding and three-dimensional (3D) culture of MCF-7 breast cancer cells. The experimental results of AO/EB and MTT methods indicate that the pioneering scaffold material has prominent biocompatibility, and cells grow rapidly in the 3D scaffold and maintain high proliferative capacity. At the same time, there is also a tendency to aggregate to form multicellular spheres. Therefore, this original P(OEGMA-co-MEO2MA) thermosensitive hydrogel can serve as a highly biocompatible and easily functionalized 3D cell culture platform with great potential in the biomedical area.

Polymerization-induced self-assembly and disassembly during the synthesis of thermoresponsive ABC triblock copolymer nano-objects in aqueous solution.

Polymerization-induced self-assembly (PISA) has been widely utilized as a powerful methodology for the preparation of various self-assembled AB diblock copolymer nano-objects in aqueous media. Moreover, it is well-documented that chain extension of AB diblock copolymer vesicles using a range of hydrophobic monomers via seeded RAFT aqueous emulsion polymerization produces framboidal ABC triblock copolymer vesicles with adjustable surface roughness owing to microphase separation between the two enthalpically incompatible hydrophobic blocks located within their membranes. However, the utilization of hydrophilic monomers for the chain extension of linear diblock copolymer vesicles has yet to be thoroughly explored; this omission is addressed for aqueous PISA formulations in the present study. Herein poly(glycerol monomethacrylate)-poly(2-hydroxypropyl methacrylate) (G-H) vesicles were used as seeds for the RAFT aqueous dispersion polymerization of oligo(ethylene glycol) methyl ether methacrylate (OEGMA). Interestingly, this led to polymerization-induced disassembly (PIDA), with the initial precursor vesicles being converted into lower-order worms or spheres depending on the target mean degree of polymerization (DP) for the corona-forming POEGMA block. Moreover, construction of a pseudo-phase diagram revealed an unexpected copolymer concentration dependence for this PIDA formulation. Previously, we reported that PHPMA-based diblock copolymer nano-objects only exhibit thermoresponsive behavior over a relatively narrow range of compositions and DPs (see Warren et al., Macromolecules, 2018, 51, 8357–8371). However, introduction of the POEGMA coronal block produced thermoresponsive ABC triblock nano-objects even when the precursor G-H diblock copolymer vesicles proved to be thermally unresponsive. Thus, this new approach is expected to enable the rational design of new nano-objects with tunable composition, copolymer architectures and stimulus-responsive behavior.

Synthesis and Self-Assembly of Thermoresponsive Biohybrid Graft Copolymers Based on a Combination of Passerini Multicomponent Reaction and Molecular Recognition.

Amphiphilic graft copolymers exhibit fascinating self-assembly behaviors. Their molecular architectures significantly affect the morphology and functionality of the self-assemblies. Considering the potential application of amphiphilic graft copolymers in the fabrication of nanocarriers, it is essential to synthesize well-defined graft copolymers with desired functional groups. Herein, the Passerini reaction and molecular recognition are introduced to the synthesis of functional thermoresponsive graft copolymers. A bifunctional monomer 2-((adamantan-1-yl)amino)-1-(4-((2-bromo-2-methylpropanoyl)oxy)phenyl)-2-oxoethyl methacrylate (ABMA) with a bromo group for atom transfer radical polymerization (ATRP) and an adamantyl group for molecular recognition is synthesized through the Passerini reaction. The graft copolymers are prepared by reversible addition-fragmentation transfer (RAFT) copolymerization of ABMA and oligo(ethylene glycol) methyl ether methacrylate (OEGMA) followed by RAFT end group removal and ATRP of di(ethylene glycol)methyl ether methacrylate (DEGMA) initiated by the ABMA units. The graft copolymer P(OEGMA-co-ABMA)-g-PDEGMA can be functionalized with β-cyclodextrin modified peptides, affording a thermoresponsive biohybrid graft copolymer. At a temperature above its lower critical solution temperature, the biohybrid graft copolymer self-assembles into peptide-modified polymersomes.

Hybrid thermoresponsive nanoparticles containing drug nanocrystals for NIR-triggered remote release

The on-demand administration of anaesthetic drugs can be a promising alternative for chronic pain management. To further improve the efficacy of drug delivery vectors, high drug loadings combined with a spatiotemporal control on the release can not only relief the pain according to patient’s needs, but also improve the drawbacks of conventional burst release delivery systems. In this study, a hybrid nanomaterial was developed by loading bupivacaine nanocrystals (BNCs) into oligo(ethylene glycol) methyl ether methacrylate (OEGMA)-based thermoresponsive nanogels and coupling them to NIR-absorbing biodegradable copper sulphide nanoparticles (CuS NPs). Those CuS NPs were surface modified with polyelectrolytes using layer-by-layer techniques to be efficiently attached to the surface of nanogels by means of supramolecular interactions. The encapsulation of bupivacaine in the form of nanocrystals allowed to achieve CuS@BNC-nanogels having drug loadings as high as 65.5 wt%. The nanocrystals acted as long-lasting drug reservoirs, leading to an elevated localized drug content, which was useful for their application in prolonged pain relief. The CuS@BNC-nanogels exhibited favorable photothermal transducing properties upon NIR-light irradiation. The photothermal effect granted by the CuS NPs triggered the nano-crystallized drug release to be boosted by the collapse of the thermoresponsive nanogels upon heating. Remote control was achieved for on-demand release at a specific time and place, indicating their potential use as an externally activated triggerable drug-delivery system. Furthermore, cell viability tests and flow cytometry analysis were performed showing satisfactory cytocompatibility in the dose-ranging study having a subcytotoxic concentration of 0.05 mg/mL for CuS@BNC-nanogels. This remotely activated nanoplatform is a promising strategy for long-lasting controlled analgesia and a potential alternative for clinical pain management.

Application of Redox-Responsive Hydrogels Based on 2,2,6,6-Tetramethyl-1-Piperidinyloxy Methacrylate and Oligo(Ethyleneglycol) Methacrylate in Controlled Release and Catalysis.

Hydrogels have reached momentum due to their potential application in a variety of fields including their ability to deliver active molecules upon application of a specific chemical or physical stimulus and to act as easily recyclable catalysts in a green chemistry approach. In this paper, we demonstrate that the same redox-responsive hydrogels based on polymer networks containing 2,2,6,6-tetramethyl-1-piperidinyloxy (TEMPO) stable nitroxide radicals and oligoethylene glycol methyl ether methacrylate (OEGMA) can be successfully used either for the electrochemically triggered release of aspirin or as catalysts for the oxidation of primary alcohols into aldehydes. For the first application, we take the opportunity of the positive charges present on the oxoammonium groups of oxidized TEMPO to encapsulate negatively charged aspirin molecules. The further electrochemical reduction of oxoammonium groups into nitroxide radicals triggers the release of aspirin molecules. For the second application, our hydrogels are swelled with benzylic alcohol and tert-butyl nitrite as co-catalyst and the temperature is raised to 50 °C to start the oxidation reaction. Interestingly enough, benzaldehyde is not miscible with our hydrogels and phase-separate on top of them allowing the easy recovery of the reaction product and the recyclability of the hydrogel catalyst.

Synthesis of bio‐based poly(methacrylates) using SG1‐containing amphiphilic macroinitiators by nitroxide mediated miniemulsion polymerization

AbstractSG1‐based amphiphilic macroinitiators were synthesized from oligoethylene glycol methyl ether methacrylate and 10 mol% acrylonitrile or styrene (as the controlling comonomer) to conduct the nitroxide mediated polymerization of bio‐based methacrylic monomers (isobornyl methacrylate (IBOMA) and C13 alkyl methacrylate (C13MA)) in miniemulsion. The effect of the addition of surfactant (DOWFAX 8390), co‐stabilizer (n‐hexadecane) and different reaction temperatures (80, 90 and 100°C) on polymerization kinetics was studied. We found that the NMP of IBOMA/C13MA using amphiphilic macroalkoxyamines were most effective during miniemulsion polymerization (linear trend of Mn versus conversion and high latex stability) in presence of 2 wt% surfactant and 0.8 wt% co‐stabilizer (relative to monomer) at 90°C. The effect of surfactant, co‐stabilizer and temperature on particle size during the polymerization was studied and suggested a decrease in initial particle size with the addition of surfactant and co‐stabilizer. Finally, the thermal properties of IBOMA/C13MA polymers, prepared by amphiphilic macroinitiators, were examined thoroughly, indicating a Tg in the range of −44°C < Tg < 109°C.

Inclusion Complexation between α-Cyclodextrin and Oligo(ethylene glycol) Methyl Ether Methacrylate.

The preparation of inclusion complexes based on α-cyclodextrin (α-CD) and oligo(ethylene glycol) methyl ether methacrylate (OEGMA) was investigated aiming to reveal complexation particularities and thermodynamic and kinetic aspects as a function of the oligomer architecture. Small-angle X-ray scattering and isothermal titration calorimetry measurements revealed that oligomer molecular weight controls both the kinetics and thermodynamics of inclusion. Unlike linear ethylene glycol polymers, OEGMA groups possess a methacrylate group, which seems to act as a stopper, affecting their mode of complexation. Nuclear magnetic resonance spectra and relaxation measurements support the fact that methacrylate groups lie outside the α-CD ring and that a full sequential complexation of the oligomer ethylene oxide groups is not observed. These results allied to the temperature sensitivity of these oligomers and enable possible routes for chemical modifications and design of new stimuli-responsive materials.
