Evaluating the antidiabetes and antioxidant activities of halogenated Schiff bases derived from 4-(diethylamino)salicylaldehyde: in vitro antidiabetes, antioxidant and computational investigation

Six Schiff bases with general name 5-(diethylamino)-2-(((halophenyl)imino)methyl)phenol (where halo = 4-fluoro (H1), 2-fluoro (H2), 2-bromo (H3), 4-bromo (H4), 4-chloro (H5) and 3-chloro-4-fluoro (H6)) were prepared by the condensation reaction between 4-(diethylamino)salicylaldehyde and suitable halogenated aromatic amines. The six halogenated Schiff bases were elucidated using different spectroscopic techniques and the structure of H3 and H6 were confirmed using single-crystal X-ray crystallography. The bond lengths of C7–N1, C7–C8 and C8–C9 obtained from structural analysis for both compounds depicted their enol-tautomeric characteristic form. The Hirshfeld analysis revealed that H‧‧‧H intermolecular contacts contributed most towards the Hirshfeld surfaces of both H3 (47.6%) and H6 (39.9%). Quantum chemical calculation studies showed that H1 and H2 have the highest and lowest energy band gap (∆E = 3.80 eV for H1 and ∆E = 3.73 eV for H2), depicting H2 and H1 as the most and least chemically reactive respectively among all the compounds. α-Amylase and α-glucosidase assay were used to evaluate the antidiabetes prowess of the synthesized compounds. All the compounds exhibited lower IC50 values than acarbose (reference drug) in α-amylase assay experiments and H5 with lowest IC50 value of 63.54 μM could be suggested to have the highest α-amylase inhibitory potential among the test samples. For α-glucosidase assay, H1–H6 displayed good antidiabetic potential. However, none of the compounds outshined acarbose with H6 having highest α-glucosidase inhibitory potential when compared to others i.e., IC50 of H6 = 60.89 μM and IC50 of acarbose = 51.42 μM. We measured the antioxidant potential of H1–H6 exploring 2,2-diphenyl-1-picrylhydrazyl (DPPH), nitric oxide (NO) and ferric reducing ability power (FRAP) assays. The DPPH as well as NO radical scavenging assay showed that all the compounds exhibited excellent antioxidant results with some of the compounds surpassing catechin (reference drug). Compound H5 with IC50 values of 30.32 mM and 31.73 mM outshined catechin with IC50 values of 31.17 mM and 140.62 mM for DPPH and NO assays respectively. All the compounds fell within the threshold of Lipinski’s Ro5 projecting them as orally bioavailable and less toxic future therapeutics.

Exploring the antioxidant and protective effects of Marsdenia thyrsiflora Hook.f. leaf extract against carbon tetrachloride-induced hepatic damage in rat models.

Medicinal plants are vital to healthcare, yet many remain unexplored. Marsdenia thyrsiflora Hook.f., from Bangladesh's Bhawal Forest, lacks research on its medicinal properties, especially its antioxidant capacities and protection against CCl4-induced liver toxicity. This study aims to evaluate the antioxidant properties of M. thyrsiflora leaf extract to determine its protective effects on rodents against CCl4-induced liver injury. After extraction, the total phenol, flavonoid content, and antioxidant capacity of the leaf extract were measured using established protocols. Free radical scavenging abilities were evaluated with 2,2'-diphenyl-1-picrylhydrazyl (DPPH), nitric oxide (NO) assays. Additionally, reducing power was assessed through cupric-reducing and ferric-reducing assays. Based on the OECD 420 recommendation, acute toxicity was tested on Swiss albino mice to establish an effective and safe dosage. For the hepatoprotective study, Sprague-Dawley rats were pre-treated with M. thyrsiflora leaf methanolic extract (MTLM) at 250 and 500mg/kg body weight, and CCl4 was administered to induce liver damage. Serum hepatic enzyme levels (alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT)), lipid profile (total cholesterol, triglycerides), total bilirubin, and markers of lipid peroxidation (Malondialdehyde (MDA)) were measured. The activities of antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH) were also evaluated to assess oxidative stress. The results demonstrated that MTLM, rich in phenolic and flavonoid content, exhibits significant antioxidant activities in DPPH and NO radical scavenging assays, as well as in reducing power assays. The acute toxicity study confirmed the safety of MTLM, with no adverse effects observed even at high doses. For the hepatoprotective study, rats were administered CCl4 to induce liver damage, followed by treatment with MTLM. Results showed that MTLM significantly reduces liver damage markers such as elevated serum hepatic enzyme levels, lipid profile, total bilirubin, and lipid peroxidation and improves the activities of GSH and key antioxidant enzymes such as SOD and CAT. Histopathological analysis corroborated these findings, displaying reduced necrosis, inflammation, and edema in liver tissues treated with MTLM. MTLM extract exhibits potent antioxidant and hepatoprotective properties. Its ability to attenuate oxidative stress, enhance antioxidant enzyme activities, and facilitate histopathological changes in the liver highlights its potential as a natural therapeutic agent for liver damage. However, further investigation is required to understand its molecular processes, safety profiles, and active component characterization.

Myco-nanotechnological approach to synthesize gold nanoparticles using a fungal endophyte, Penicillium oxalicum, and unravelling its antibacterial activity and anti-breast cancer role via metabolic reprogramming

The present study has been designed to fabricate fungal endophyte-assisted gold nanoparticles (AuNPs) and elucidate their anti-breast cancer potential. The aqueous extract of fungal endophytePenicillium oxalicum(PO), associated with the medicinal plantAmoora rohituka, was used for the fabrication of AuNPs (POAuNPs). Physico-chemical characterization using Ultraviolet-visible spectroscopy, Fourier transform infrared, X-ray diffraction, Dynamic light scattering, Zeta potential, Transmission electron microscopy and Field emission scanning electron microscopy analysis revealed stable, uniform distribution, spherical shape and crystalline nature of POAuNPs with a size range of 3-46 nm. Furthermore, the POAuNPs potentially inhibited the growth of pathogenic bacterial strainsEscherichia coliandStaphylococcus aureus. The synthesized POAuNPs have shown potential antioxidant effects against 2,2-diphenyl-1-picrylhydrazyl (DPPH), superoxide and nitric oxide (NO) radical scavenging assays with an EC50value of 8.875 ± 0.082, 52.593 ± 2.506 and 43.717 ± 1.449 µg mL-1, respectively. Moreover, the value of EC50for the total antioxidant capacity of POAuNPs was found to be 23.667 ± 1.361 µg mL-1. The cell viability of human breast cancer cells, MDA-MB-231 and MCF-7, was found to be reduced after treatment with POAuNPs, and IC50values were found to be 19.753 ± 0.640 and 35.035 ± 0.439 µg mL-1, respectively. Further,in vitrobiochemical assays revealed that POAuNPs induces metabolic reprogramming in terms of reduced glucose uptake, increased lactate dehydrogenase (LDH) release and, disruption of oxidative balance through depletion of glutathione levels, increased nitric oxide (NO) and lipid peroxidation levels as a possible pathway to suppress human breast cancer cell proliferation. Apoptosis-specific nuclear modulations induced by POAuNPs in human breast cancer cells were validated through 4',6-diamidino-2-phenylindole (DAPI) nuclear staining. The present investigation thus attempts to show the first ever fabrication of AuNPs using an aqueous extract ofP. oxalicumassociated withA. rohituka. The results revealed unique physico-chemical characteristics of mycogenic AuNPs, and screening their effect against breast cancer via metabolic reprogramming and induction of apoptosis thus adds great significance for cancer therapeutics, suggesting further exploration to develop nanotherapeutic drugs.

Investigating the Therapeutic Property of Galium verum L. (GV) for MSG induced Audiogenic Epilepsy (AEs) and Neuroprotection through In-Silico and In-Vitro Analysis.

Audiogenic Epilepsy (AEs) is a subtype of epileptic seizure that is generally caused by high-intensity sounds. A large number of traditional medicines has been explored in this lieu where our study chased Galium verum L. (Rubiaceae), an herbal plant which is commonly known as Lady's Bedstraw, that contains a highly rich chemical composition including flavonoids (Hispidulin, Quercetin, and Kaempferol), and phenolic acids (chlorogenic acid, caftaric acid, and gallic acid). G verum is well known for its antioxidant, neuroprotective, and anti-inflammatory properties. Recently, the unique role of Adhesion G Protein- Coupled Receptor V1 (ADGRV1) protein in the progression of audiogenic epilepsy has been explored. This study aimed to examine the potent phytoconstituents of the hydroalcoholic extract of G. verum L. (HEGV) using analytical techniques. Additionally, our study sought to evaluate the antioxidant, neuroprotective, anti-inflammatory properties, and antiepileptic potency of HEGV by targeting ADGRV1 via in silico and in vitro analyses using SHSY5Y cells. HPLC and LC-MS techniques were employed to identify the flavonoids, iridoids, and phenolic acid derivatives present in HEGV. DPPH (2,2-diphenyl-1-picrylhydrazyl), nitric oxide (NO), and hydroxyl (OH) radical scavenging assays were performed to confirm the antioxidant potential of the extract. Additionally, in silico molecular docking and molecular dynamic studies were performed using AutoDock Vina software to analyze the possible interactions between crucial phytoconstituents of HEGV and ADGRV1, followed by cell line analysis. In the in vitro analysis, antioxidant, neuroprotective, and anti-inflammatory properties were assessed via cell viability assay, IL, GABA, and glutamate estimation. LC-MS and HPLC analyses revealed high concentrations of hispidulin, a major flavonoid found in HEGV. HEGV exhibited moderate-to-high free radical-scavenging activities comparable to those of ascorbic acid. Docking analysis demonstrated that hispidulin has a stronger binding affinity with ADGRV1 (Vina score = -8.6 kcal/mol) than other compounds. Furthermore, cell line analysis revealed that the MSG exacerbates the neurodegeneration and neuroinflammation, whereas, HEGV and Hispidulin both possess neuroprotective, antioxidant, and antiepileptic activities. HEGV and Hispidulin proved to be promising candidates for treating audiogenic epilepsy by modulating ADGRV1.

Eco-friendly Strategy for Producing Bio-based Silver Nanoparticles (AgNPs) Employing Sepioteuthis lessoniana ink, in Addition to Biological and Degradation of Dye Applications.

The squid, Sepioteuthis lessoniana, is a remarkable fishery product which is exported by many nations for use in industrial production or human consumption. This study focused on the synthesis of silver nanoparticles (AgNPs) from squid ink (SI) and its wide range of applications. The formation of the nanoparticles was confirmed through UV-Visible spectroscopy, FTIR, XRD, SEM with EDX, DLS, and zeta potential analysis. The results showed a strong absorbance peak at 407nm, the presence of various functional groups, a nanocrystalline structure with a crystalline size of 17.56nm, spherical-shaped particles with an average size of 76nm, and the presence of the highest % mass of Ag and uniformly dispersed particles, respectively. The bioactivity of the synthesized squid ink silver nanoparticles was analyzed through antibacterial, antioxidant, anticancer, and toxicity studies. The dye degradation assay was also analyzed as a means of wastewater treatment for different industrial dyes. The antibacterial activity showed the highest zone of inhibition of 24mm at a concentration of 100μg/ml against Escherichia coli, followed by other tested strains. The nitric oxide radical scavenging assay showed the highest antioxidant activity (92%) at a concentration of 100μg/ml. The cytotoxic ability of SI-AgNPs against the MDA-MB-231 breast cancer cell line revealed an IC50 value of 4.52μg/ml. The toxicity study revealed a dose and time-dependent activity with the LC50 value of 5.090 and 3.303mg/ml for 24 and 48h, respectively. The successful degradation of dyes by SI-AgNPs is attributed to the cooperative action of the electron relay system with Ag as a catalyst and SI as a catalytic support. These findings indicate that SI-AgNPs are a novel potential product that should be further studied to improve its pharmacological, biomedical, and environmental applications.

Turmeric-Black Cumin Essential Oils and Their Capacity to Attenuate Free Radicals, Protein Denaturation, and Cancer Proliferation.

Turmeric rhizomes (Curcuma longa) and black cumin seeds (Nigella sativa) are polyherbal ingredients used for the management of cancer and other chronic inflammatory diseases in Nigerian ethnomedicine. Previous studies have shown the antioxidant, anti-inflammatory, and anticancer activities of the individual plant extracts. However, the two spices have not been biologically potentiated in their combined form. Therefore, this study obtained essential oils (EOs) from the combined spices and evaluated their inhibitory effects on free radicals, protein denaturation, and cancer proliferation. The EOs were extracted by hydro-distillation (HD) and characterized by gas chromatography-mass spectrometry (GC-MS). In vitro antioxidant assessment was conducted based on DPPH, hydrogen peroxide (H2O2), nitric oxide (NO), and ferric ion (Fe3+) radical scavenging assays. The cytotoxicity of the oil against non-tumorigenic (HEK293) and cancerous (HepG2 and HeLa) cell lines was determined following the MTT cell viability assay. An in silico molecular docking analysis of the oil constituents was also performed. Six batches of EOs I-VI were afforded, comprising twenty-two major constituents, with aromatic Ar-turmerone being the most prominent compound. There was a marked improvement in the bioactivity of the oils upon repeated HD and as a combination. The batch VI oil exhibited the best activity, with a cytotoxicity (CC50) of 10.16 ± 1.69 µg/100 µL against the HepG2 cell line, which was comparable to 5-fluorouracil (standard, CC50 = 8.59 ± 1.33 µg/100 µL). In silico molecular docking suggested δ-curcumene, Ar-curcumene, Ar-turmerol, and Ar-turmerone among the promising compounds based on their high binding energy scores with NOX2, NF-κB, and mdm2 proteins. In conclusion, the oils from the turmeric-black cumin combined possess a considerable inhibition ability against free radicals, protein denaturation, and cancer proliferation. This study's findings further underscore the effectiveness of turmeric-black cumin as a polyherbal medicinal ingredient.

The antioxidant, anti-obesogenic, and anti-diabetic potential of selected South African marine algal species

South African marine algae are a prevalent traditional medicine utilized by indigenous populations. There has been recent attention on the health advantages of South African marine algae and its bioactive compounds, which have been associated with favourable health outcomes such as diabetes and obesity. This study investigated the antioxidant, anti-obesogenic and anti-diabetic potentials of three South African marine algal species or seaweeds, namely Codium fragile, Radicilingua thysanorhizans and Mazzaella capensis using a variety of in vitro and in silico experiments. The crude extracts, obtained using different solvents (aqueous and ethanol), were examined to assess their total phenolic and total flavonoid content as well as their antioxidant properties by means of 2,2-diphenyl-β-picrylhydrazyl (DPPH), non-site-specific hydroxyl radical (OH•), and nitric oxide (NO) radical scavenging assays. Thereafter, the extracts were subjected to carbohydrate digestive enzyme inhibitory assays, including α-glucosidase and α-amylase, and a lipid digestive enzyme inhibitory assay, such as pancreatic lipase. The glucose uptake by yeast cells was also evaluated in vitro. The subjection of the marine algal extracts to LC-MS analysis revealed several beneficial compounds. These compounds were further docked against α-glucosidase and α-amylase, and a lipid digestive enzyme, pancreatic lipase. The results of this study indicate that the selected South African marine algae possess potent antioxidant capacity while also exhibiting anti-obesogenic and anti-diabetic activities by inhibiting carbohydrate and lipid digesting enzyme activity as well as by increasing glucose uptake in yeast cells. Evaluated ADME profiles revealed that the compounds of these plants have excellent bioavailability and are considered safe for consumption. Overall, the results of this study suggest that all studied seaweeds have potential antioxidant, anti-diabetic, and anti-obesogenic potentials with Codium fragile found to be the most promising among them. The current work serves as a validation of the data currently available regarding the antioxidant, anti-obesogenic and anti-diabetic potential of C. fragile. The study will also serve as a first-report of the aforementioned activities for R. thysanorhizans and M. capensis.

Compositional, structural, and functional characterization of fucoidan extracted from Sargassum polycystum collected from Saint Martin's Island, Bangladesh

Fucoidan is a sulphated polysaccharide with diversified biological activities commonly found in brown seaweeds. In our present study, fucoidan was extracted from Sargassum polycystum collected from Saint Martin's Island in three different extraction methods. The composition of the fucoidans was determined by measuring the total carbohydrate, sulfate, uronic acid, fucose, and protein contents. Fourier-transform infrared spectroscopy (FT-IR) and Proton nuclear magnetic resonance (1H NMR) were studied to determine the structural groups of extracted fucoidans. Functional activities were analyzed by antioxidant activity and anti-diabetic activity assay. Fucoidan yield was obtained highest in the acid extraction method (0.061 ± 0.007 g/g), followed by the water extraction method (0.058 ± 0.005 g/g) and salt extraction method (0.041 ± 0.005 g/g). Water extracted fucoidan showed the highest carbohydrate content (62.97 %), uronic acid content (6.46 %), and protein content (2.62 %). Sulphate content (28.56 %) was highest in salt extracted fucoidan, whereas fucose content (24.0 %) was highest in water extracted fucoidan. FT-IR and 1H NMR analysis confirm several characteristic picks for the extracted fucoidans. Extracted fucoidans presented comparable antioxidant activity, but salt-extracted fucoidan presented the highest DPPH radical scavenging assay (86.85 ± 0.74 %, IC50 = 0.03 mg/mL), ABTS radical scavenging assay (65.13 ± 0.92 %, IC50 = 1.94 mg/mL), Phosphomolybdenum assay (4.356 ± 0.11) and Nitric Oxide (NO) radical scavenging assay (57.08 ± 0.52 %, IC50 = 3.73 mg/mL) in dose-dependency manner. Further anti-diabetic activity assay resulted in the highest α-amylase inhibition activity (62.82 ± 1.36 %, IC50 = 2.30 mg/mL) and α-glucosidase inhibition activity (55.73 ± 0.84 %, IC50 = 3.71 mg/mL) by salt-extracted fucoidan. These functional potentials of seaweed may add significant value to the development of functional food and pharmaceutical materials.

<i>In Vitro</i> Anti-oxidant and Antidiabetic Activity of the Leaves of <i>Stereospermum sauveolens</i> (Roxb.) DC

Background: Diabetes mellitus is a metabolic condition characterised by a persistent rise in blood glucose levels. Diabetes is just one of the many disorders that can be treated using natural remedies made from plants. Stereospermum suaveolens (Roxb.) DC, belongs to the family Bignoniaceae, commonly known as Padri. It is employed as a folk remedy in southern India to cure diabetes. Aim: The aim of this study is to assess the potential anti-diabetic and antioxidant properties of the hydroalcoholic extract of S. suaveolens (Roxb.) DC leaves through phytochemical screening and in vitro assays. Methods: Leaves of S. suaveolens (Roxb.) DC were collected and processed to obtain a hydroalcoholic extract. The extract was subjected to qualitative analysis to identify the presence of various phytochemical constituents. The free radical scavenging activity of the extract was evaluated using Nitric oxide radical scavenging assay and DPPH racial scavenging assay. In vitro anti-diabetic studies were performed through α-amylase inhibition assay and glucose uptake assay. Results: The hydroalcoholic extract of S. suaveolens (Roxb.) DC leaves exhibited the presence of bioactive compounds. The extract demonstrated dose-dependent free radical scavenging activity in the in vitro antioxidant assays. The extract showed remarkable inhibitory activity against α-amylase, suggesting its potential in controlling carbohydrate digestion. Also the extract significantly enhanced glucose uptake by yeast cells, indicating its potential to regulate blood glucose levels. Conclusion: The findings of this study indicate that the hydroalcoholic extract of S. suaveolens (Roxb.) DC leaves possesses both antioxidant and anti-diabetic properties. These results suggest its potential as a natural medicinal agent for the management and treatment of type 2 diabetes mellitus.

Organic wastes of Citrus sinensis Peels- a source of eco-friendly and sustainable bioactive compounds for promoting health

To determine the phytochemicals, radical scavenging and antioxidant potential of orange peel extract. Citrus sinensis were subjected to extraction with ethanol. Gas chromatography (GC) was utilized to determine the phytochemical composition of orange peel extract. Hydrogen peroxide, superoxide, nitric oxide, and hydroxyl radical scavenging assays were conducted to assess radical scavenging potential of the extract. Antioxidant activities of the peel extracts were determined via the 2,2-diphenylpicrylhydrazyl (DPPH) free radical scavenging activity, ferric reducing antioxidant power (FRAP) assay, ABTS scavenging, and total antioxidant capacity (TAC) assay. The GC-FID analysis revealed the presence of alkaloids, flavonoids, polyphenols, tannins, sapogenin, and steroids in the orange peel extract. The results of radical scavenging assays demonstrated the extract’s ability to scavenge hydrogen peroxide, superoxide, nitric oxide and hydroxyl radicals. The scavenging capacity of the extract was observed to be concentration-dependent, with comparisons made to standard antioxidants ascorbic acid and BHT. Peels from citrus sinensis represent a valuable source of phytochemicals, demonstrating significant antioxidant and radical scavenging activities.

Aminoguanidine Hydrazone Derivatives: The Antioxidant, Antineoplastic Profile, and Interaction with ctDNA Studies

Herein, we report the synthesis and evaluation of four aminoguanidine hydrazone derivatives with different aromatic moieties. This class of compounds presents a series of biological applications. Derivative AGH-3 with an indole nucleus offered the highest antioxidant capacity with results comparable to Trolox in 2,2-diphenyl-2-picrylhydrazyl radical (DPPH• ), 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS•+), FeIII reduction assay (FRAP), and nitric oxide (•NO) radical scavenging assays. Furthermore, AGH-3 showed the highest antiproliferative activity against human kidney cancer cells (786-0) with concentration necessary to inhibit 50% cell growth (GI50) = 6.3 µM; additionally, in biophysical studies, AGH-3 interacted with ctDNA (biological target model) forming a fluorescent supramolecular complex with a binding constant (Kb) of 2.89 × 103 M-1 with preferentially an intercalator mechanism. The tested compounds revealed the potential of aminoguanidine hydrazones as a strategic class of compounds with multitarget biological activity.

Antioxidant Effect of Levetiracetam and Brivaracetam Acting on Presynaptic SV2A Receptors: An In-vitro Study

Introduction: Epilepsy is a serious psychological condition associated with social stigma, psychiatric co-morbidity, and a high economic burden. It is pertinent to develop novel treatment methods given the ever-evolving nature of the disease. Recently, various newer Antiepileptic Drugs (AEDs) have been under study, among which a newer targeted modality consists of SV2A receptors. Aim: To compare the antioxidant properties or free radical scavenging activity of the newer heterogeneous AEDs acting on the SV2A receptors, namely Levetiracetam (LEV) and Brivaracetam (BRV). Materials and Methods: This in-vitro study was conducted at Department of Pharmacology, Sri Ramachandra Institute of Higher Education and Research, Chennai, Tamil Nadu, India from July 2021 to August 2021. The study evaluated the antioxidant properties of LEV and BRV using various assays such as the Ferric Reducing Antioxidant Power (FRAP) test, DPPH (1,1-diphenyl-2-picrylhydrazyl) assay, Nitric Oxide (NO) radical scavenging assay, Superoxide Dismutase activity (SOD), and catalase activity. The obtained results were expressed using specific formulas related to the absorbance of the respective chemicals, along with the percentage of inhibition, and have been tabulated in Microsoft excel Sheet Version 16.16.27. Halfmaximal Inhibitory Concentration (IC50) values were calculated using scatter plot graphs. Results: The FRAP values for LEV were reported to be significantly higher compared to BRV at concentrations of 20, 40, 60, 80 μg/mL, while at 100 μg/mL, BRV showed better ferric reducing activity. Although both drugs exhibited antioxidant activity, the results clearly identified LEV as a better NO radical scavenger, with a percentage of inhibition reaching up to 80.52%. During the SOD assay, the percentage inhibition of superoxide generation by LEV was found to be 99.02%, while that of BRV was 97.07% at a concentration of 100 μg/mL. LEV (0.573) also showed a higher degradation of H2O2 per minute than BRV (0.065) at a concentration of 100 μg/mL. Conclusion: The results showed that both drugs, BRV and LEV, exhibited significant antioxidant capacity. However, LEV demonstrated increased antioxidant potency and efficacy compared to BRV.

Antioxidant and Antibacterial potential of Methanol root Extract of Kigelia africana (Sausage tree)

Kigelia africana popularly known as the cucumber or sausage tree that possesses medicinal and traditional uses like anticancer, antiulcer, anti-aging, antioxidant, and anti-malarial activity.This study aimed to investigate GCMS, FTIR, Antioxidant and Antibacterial properties of methanolic root extract of Kigeliaafricana.The antioxidant activity of root extracts of Kigelia was evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical assay, Reducing power (PR) assay, Ferric Reducing Power assay and Nitric oxide (NO) radical scavenging assay. Ascorbic acid was used as a standard antioxidant. GCMS analysis reveals the presence of 9-Octadecanoic acid (z)-methyl ester while theFTIR spectral analysis identify some functional group such as hydroxyl group, Carboxyl group, methyl group, carbonyl group, amino group, and phosphate group. In vitro antibacterialactivityof the extracts wastestedagainst4bacterialstrains viz; Staphylococccus aureus, Escherichia coli, Pseudomonas aeruginosa, and Salmonella spp.The highest antibacterial activity was found against Staphylococcus aureus then followed by Pseudomonas aeruginosa, Salmonella spp and Escherichia coli. The extract showed much higher scavenging activity against DPPH and with most reducing power against ferric cyanide. This study reveals the potency of the root of K. africana to be used as a good natural source for antioxidant and antibacterial activity.

Evaluation of Toxicity and Antioxidant Activity of Hydro-Ethanol Leave Extract of Mentha piperita

Mentha piperita has numerous pharmacological, cosmetic and alimental applications due to its ability to produce terpene and terpenoid compounds. Antioxidants are considered as important bioactive compound on account of many health benefits along with their vital role in delaying oxidative rancidity of numerous food. The study was to investigate acute toxicity evaluation and in vitro antioxidant activity of hydro-ethanol leave extracts of M. piperita. The acute toxicity was evaluate in rat according to (OECD). Animals were daily observed for 24 hours after treatment for mortality. The antioxidant activity of the extract was evaluated based on the method such as DPPH radical scavenging activity and reducing power method. The results showed that mortality was not observed in phase one but was observed in phase two. The LD50 by oral route in rat was less than 5000 mg/kg body weight. The percentage of antioxidant activity of leaf extracts of M. piperita was assessed by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical assay, reducing power (RP) assay, ferric reducing/antioxidant power assay, and nitric oxide (NO) radical scavenging assay. The analysis revealed that NO scavenging activity is higher than DPPH with extract. NO and DPPH exhibited remarkable antioxidant activity. This indicates that the hydro-ethanol leave extract of M. piperita has a good antioxidant activity.

Evaluation of Antioxidant and Anti-diabetic Activity of Stem Bark of <i>Ziziphus jujuba</i> (L.) Gaertn

Diabetes Mellitus (DM), a significant metabolic disorder that increases mortality and morbidity, affects people all over the world. Recently, many individuals are exploring CAM treatment that may have fewer side effects due to the prominent side effects of allopathic medications. Ziziphus jujuba (L.) Gaertn. (Rhamnaceae) stem bark has been a vital part of the conventional system of medicine for the management of diabetes, inflammation, wounds, fever, asthma and liver disorders. In spite of its pharmacological significance, Ziziphus jujuba stem bark lacks scientific evidence of its antidiabetic and antioxidant potential. Hence, an attempt has been made to evaluate the anti-diabetic effect of the stem bark of Ziziphus jujuba using alpha-amylase inhibition assay. In this research work, we further investigated the antioxidant activity of the Ziziphus jujuba using DPPH and Nitric oxide radical scavenging assay. The Chloroform (CEZJ), ethyl acetate (EAZJ) and Ethanol (EEZJ) extracts of Ziziphus jujuba stem bark were used in the study. In the DPPH radical scavenging assay, EEZJ had an IC50 value of 117.1 µg/ml, however, the value for the nitric oxide scavenging assay was 64.65 µg/ml. The Ziziphus jujuba was additionally examined for its in-vitro antidiabetic potential using an alpha-amylase inhibition assay, and it was discovered to possess a considerable percentage of alpha-amylase inhibition. The IC50 of EEZJ was found to be 34.68 µg/ml. These results imply that the Ziziphus jujuba possesses considerable antioxidant and anti-diabetic properties. To thoroughly establish the anti-diabetic potential of Ziziphus jujuba, additional in vitro experiments utilizing cell lines and other enzymes may be conducted.

Antioxidant, Antiproliferative, Pro-apoptotic and cell cycle arrest properties of crude extract and biofractions of Hybanthus enneaspermus Linn. to combat breast cancer

Objective: According to the World Health Organisation, breast cancer is presently the most common cancer diagnosed in women globally. Polyphenolic compounds act as antioxidants, improve health, and reduce risk and proliferation of various types of cancers. Hybanthus enneaspermus Linn. is a beneficial medicinal plant, reported to possess antimicrobial, antioxidant, anti-inflammatory, neuroprotective, cardioprotective, and nephroprotective properties etc. Methods: The current study involved the evaluation the antioxidant, antiproliferative, apoptotic and cell cycle arrest potential of the ethanolic leaf extract of Hybathus enneaspermus Linn. (EEHE), its toluene soluble, toluene insoluble, ethyl acetate and methanol soluble biofractions viz. TFHE, ITHE, EFHE, and MFHE to combat breast cancer. In vitro antioxidant activities were evaluated using DPPH, Hydrogen peroxide, Nitric oxide and ABTS free radical scavenging assays. In vitro antiproliferative activity against MCF-7 cells was assayed using the Sulforhodamine method, while apoptosis and cell cycle assays were analysed by flow cytometry. Results: MFHE exhibited significant in vitro antioxidant activity with IC50 values of 21.10±0.39 μg/mL and 25.99±4.66μg/mL, when compared against standard ascorbic acid with IC50 values of 11.19±1.09 μg/mL and 9.30±0.26μg/mL in DPPH and nitric oxide assays respectively. EFHE displayed substantial antioxidant potential in ABTS and hydrogen peroxide assays with IC50 values of 40.38±0.88μg/mL and 99.11± 13.59μg/mL, while ITHE showed considerable activity with IC50 < 100μg/mL in DPPH, nitric oxide and ABTS assays. TFHE demonstrated significant antiproliferative activity by sulforhodamine assay, with GI50 value of 10.22 6.72µg/mL, while EEHE and ITHE showed substantial activity with GI50 values of 41.42±3.74µg/mL and 64.37±7.07µg/mL respectively, as against the standard drug Adriamycin (GI50 < 10µg/mL) used. In the apoptosis assay, ITHE showed 11.31±0.82% cells in late apoptosis and 34.48±1.57 % cells in necrosis as compared to standard Adriamycin indicated 13.67±1.02 % cells in late apoptosis and 8.58±0.65 % cells in necrosis. In cell cycle analysis, ITHE displayed significant apoptotic activity with 20.15±1.37 % cells in SubG1 phase and 13.99±1.65 % cells arrested in G2-M phase as compared to the control. Conclusion: The study thus revealed that MFHE, EFHE and ITHE biofractions showed significant antioxidant activities, while EEHE, TFHE and ITHE exhibited substantial antiproliferative activity against mammary cancer cells. Additionally, ITHE induced remarkable apoptotic activity and cell cycle arrest in the MCF-7 cells. The therapeutic benefits may be credited to the bioactive constituents present in the ITHE fraction viz. polyphenolics, flavonoids etc.; however, the molecular mechanisms may need to be evaluated further.

Biochemical Screening, Fabrication of Green Nanoparticles and Its Antimicrobial, and Antioxidant Studies of Endophytic Fungus Phlebia Species.

Endophytes are organism dwelling totally dynamic and novel biotopes this makes them able to produce novel biochemicals that may become assets to the future. This study aims at understanding the biochemical components of the endophytic fungus Phlebia sp. synthesis of gold and silver nanoparticles from it, and the antimicrobial as well as antioxidant ability of these green synthesised nanoparticles. Aqueous fungal extract was subjected for HRLCMS analysis which revealed 34 biochemicals within the extract. Silver and gold nanoparticles were also produced from the fungal extract. UV-vis analysis revealed a peak at 450nm for silver nanoparticle and 550nm for gold nanoparticles. FESEM analysis confirmed the presence of these nanoparticles with its spherical shape. Both of these nanoparticles were able to produce a conspicuous zone of inhibition in the antimicrobial tests against Escherichia coli, Salmonella paratyphi. For both of the organisms under study, a concentration-dependent expansion of the zone of inhibition was discovered in the nanoparticles. However, with silver nanoparticles, a relatively high zone of inhibition and vulnerability of the organism was discovered. Four in vitro free radical scavenging assays, including the DPPH, Hydroxyl, Superoxide, and Nitric oxide radical scavenging assays, were used for antioxidant analysis. The results of every test demonstrated that green synthesisedsilver nanoparticles had higher activity than gold nanoparticles. All of the tests showed that silver nanoparticles were more active than gold nanoparticles with the maximum value of 86.254 ± 0.296% being discovered at the greatest concentration of superoxide radical scavenging assay.

In- vitro Antioxidant and Anti-aging activity of a Traditional Ayurvedic Formulation

Aging is an inevitable process for all living organisms. When the skin ages, Reactive Oxygen Species (ROS) are generated, which results in an increase in Hyaluronidase, Collagenase and Elastase, further contributing to skin aging. Amalaki Rasayana (AR) which is a traditional Ayurvedic preparation was investigated to assess its anti-oxidant and anti-aging properties. In order to assess radical scavenging activity, plant extracts/formulations were tested for their ability to scavenge DPPH and nitric oxide radicals. The alcoholic extract was prepared with Amalaki Rasayana (AR) and used for phyto-chemical, anti- oxidant and anti- aging studies. The phyto-chemical screening revealed the presence of tannins, alkaloids and flavonoids. The in- vitro anti- oxidant activity of Amlaki Rasayana (AR) was carried out by DPPH and Nitric oxide radical scavenging assay method. The DPPH assay revealed the concentration dependent scavenging activity with the extract and the results are comparable with standard quercetin. The percentage inhibition was more at 1000 μg/ml of the ethanol extract in Nitric oxide scavenging assay. The in- vitro anti- aging potential was evaluated by anti- collagenase assay method using FALGPA as substrate. The ethanol extract exhibited significant inhibition on collagenase enzyme which was comparable to standard EDTA. The ethanol extract of Amlaki Rasayana (AR) showed significant anti- oxidant and anti- aging activity which may be attributed to phyto- constituents such as flavonoids, alkaloids and tannins. This study offers scientific validation of Amlaki Rasayana (AR) for anti- aging activity and can be recommended to be taken internally for aging problems.

A Study on Phytochemical and Antioxidant properties of the Leaves of Ficus hispida Linn.

Ficus hispida Linn. is a small but well-distributed species of the tropical fig tree. Although used in traditional medicine to treat many ailments, there is a lack of system reports about the phytochemical and antioxidant properties of the leaves of that plant. This study was conducted to investigate the phytochemical and antioxidant properties of F. hispida leaves using four different solvents (methanol, ethanol, chloroform, and petroleum ether). Qualitative and quantitative analysis are performed to determine the presence and content of phytochemicals. Four in vitro assay models were used to investigate the antioxidant activity. Phytochemical experiments confirmed the presence of alkaloids, flavonoids, flavonols, resins, saponins, proanthocyanidins, glycosides, steroids, and tannins. Qualitative analysis showed the total content of polyphenols such as phenolic, flavonoids, flavonols, and proanthocyanidin in those extracts. Among all the Extracts, ethanol extracted from the leaves of F. hispida has shown superior activity in the testing of the total antioxidant capacity assay. In DPPH and ABTS scavenging, ethanol extracted from F. hispida leaves shows the highest scavenging activity among all extractions with IC50 values of 50.33±2.59μg/ml and 83.35±1.98μg/ml respectively. Although in Superoxide radical scavenging assay and Nitric Oxide radical scavenging assay, methanol extraction of F. hispida leaves exhibit with an IC50 value of 104.17±3.49μg/ml and 108.98±4.66μg/ml.

In Vitro Antioxidant and Fibroblast Migration Activities of Fractions Eluded from Dichloromethane Leaf Extract of Marantodes pumilum.

(1) The complexity of diabetes and diabetic wound healing remains a therapeutic challenge because proper and systematic wound care and management are essential to prevent chronic microbial infection and mechanical damage to the skin. Marantodes pumilum, locally known as 'Kacip Fatimah', is an herb that has been previously reported to possess anti-inflammatory, analgesic, antinociceptive and antipyretic properties. The current study aims to assess the antioxidant and fibroblast cell migration activities of the fractions eluded from the dichloromethane extract of M. pumilum leaves. (2) The total antioxidant capacity of M. pumilum was assessed using the total proanthocyanidins and phosphomolybdenum assays, while DPPH, nitric oxide, hydrogen peroxide and superoxide free radical scavenging assays were tested to determine the antioxidant potential of M. pumilum. An in vitro scratch wound assay was performed to measure the fibroblast cell migration rate using normal and insulin-resistant human dermal fibroblast cells. (3) All M. pumilum fractions exhibited good antioxidant and fibroblast cell migration activity, among which fractions A and E displayed the greatest effect. (4) M. pumilum's fibroblast migration activity could be attributed to its strong antioxidant properties along with its previously reported properties.
