Optimizing the microwave-assisted hydrothermal extraction of pectin from tangerine by-product and its physicochemical, structural, and functional properties

Microwave-assisted hydrothermal extraction (MAHE) was optimized using a Box-Behnken design (BBD) of the response surface methodology (RSM) for optimal recovery of pectin from tangerine peel (TPP). The effects of three factors (pH, irradiation time and temperature) on extraction yield (EY), galacturonic acid content (GAC) and degree of esterification (DE) of pectin were investigated. The optimal extraction conditions were as follows: pH 1.7, irradiation time 12 min and temperature 109 °C. Under these conditions, the EY, GAC and DE were 30.4, 72.3 and 45.2%, respectively. The low methoxyl content of MHAE (45.2%) compared to CE is confirmed by the 1H NMR and FTIR spectra, and the emulsifying activity is 57.65% and 50.56% for CE and MHAE, respectively. The total phenolic content (TPC) of pectin produced using MAHE is 41.2 mg GAE/g, thus indicating higher antioxidant properties compared to pectin produced with CE, which had a TPC of 38.4 mg GAE/g. In addition, the X-ray diffraction (XRD) and surface morphological analysis (SEM) results showed that TPP had a rough surface and crystalline structure. Overall, our findings show that TTP from MAHE can be used as a natural antioxidant ingredient in the functional food and pharmaceutical industries.

Antioxidant components of the three different varieties of Indonesian ginger essential oil: In vitro and computational studies

Differences in antioxidant activity and the components that contribute to this activity in the essential oil (EO) of three varieties of Indonesian ginger were studied. The EOs were isolated at various distillation times and their composition was identified by GCMS. Antioxidant activities were determined by in vitro analysis, while Pearson correlation followed by density functional theory (DFT)-based reactivity descriptors analysis was used to identify their antioxidant components. A total of 29 components were identified from all studied ginger EOs where geranial was main component of red (28.3 %) and gajah ginger (20.4 %), while zingiberene (16.2 %) was main component of emprit ginger EO. In general, the gajah ginger EO was demonstrated the best antioxidant activity. Pearson correlation analysis was revealed 9 components, namely β-myrcene, D-limonene, α-sabinene, geranyl acetate, α-curcumene, α-zingiberene, α-farnesene, β-bisabolene, and β-sesquiphellandrene, may be contribute for antioxidant activity with strong correlation value (r>0.6). Reactivity descriptors of B3LYP optimized geometry and DEF2-SVP basis set confirmed the reactivity of the nine components and revealed zingiberene as the most reactive. This study has revealed the differences in antioxidant activity and antioxidant components of Indonesian ginger EOs, thus providing clearer information regarding the best variety and composition profile as a natural antioxidant ingredient.

Validation study of isoflavone derivatives and simultaneous analysis of Puerariae Flos

Using LC/MS, isoflavones (glycitin, genistin, tectoridin, glycitein, genistein, and tectorigenin) known as Puerariae Flos marker compounds were identified. The simultaneous analysis and validation of six marker compounds were performed using the HPLC-DAD method. To validate the analysis method of six marker compounds 1–6, the linearity was showed in the calibration curve as coefficient of correlation (R2) range of 0.9997–1.000. The limit of detection (LOD) and limit of quantitation (LOQ) of six marker compounds were measured in the ranges of 0.2720–0.7884 µg/mL and 0.8243–2.3890 µg/mL, respectively. Intermediate precision, repeatability, and accuracy evaluations according to the Ministry of Food and Drug Safety (MFDS) guidelines showed that the relative standard deviation (RSD) less than 1% and the stability less than 2%, respectively. Also, the simultaneous validation method is attributed to analysis of the different isoflavones Flos ne compounds in Puerariae extract. It could be potentially used as sources of pharmaceutical and natural antioxidant ingredients.

High hydrostatic pressure assisted by food-grade enzymes as a sustainable approach for the development of an antioxidant ingredient

Apple is one of the most important fruits in the world. The elaboration of apple-based products such as cider or apple juice generates around 3–4.2 million tonnes of apple by-product that are annually discarded. Apple by-product has been described as an interesting source of bioactive compounds, including polyphenols. These valuable components, once extracted, could enable the usage of the apple by-product as a natural antioxidant ingredient. The aim of the present work was to address the effect of high hydrostatic pressure (HHP), and HHP assisted by two alternative food-grade enzymes, Celluclast®, and Ultraflo®. For that purpose, the total phenolic content was measured by Folin-Ciocalteu and Fast-Blue BB assays. Furthermore, antioxidant capacity was determined by three analytical methods (DPPH, ORAC and FRAP) and the characterization of phenolic compounds was carried out by Chromatography and Mass Spectrometry analysis. Results indicated a significant increase of the total phenolic content and the antioxidant capacity after the HHP aided by Celluclast® or Ultraflo® treatment. Conversely, the exclusive application of HHP induced minor changes. Thus, the combination of HHP and the food-grade enzymes Celluclast® or Ultraflo®, has proved to be an effective strategy for the extraction of natural antioxidant compounds from the apple by-product.

Effects of dietary supplementation with selenium yeast and jujube powder on mitochondrial oxidative damage and apoptosis of chicken

The main objective of this study was to explore the effects of dietary selenium yeast and jujube powder on mitochondrial oxidative damage and cell apoptosis of broilers during postmortem aging, chicken breasts of broilers fed diets supplemented with different concentrations of selenium yeast and jujube powder were used as research subjects. With the prolongation of postmortem aging time, the levels of reactive oxygen species (ROS), carbonyl content, mitochondrial permeability transition pore (MPTP) openness, and mitochondrial membrane permeability increased significantly (P < 0.05). The contents of the sulfhydryl, mitochondrial membrane potential, shear force, and cytochrome C (Cyt-c) reduction level decreased significantly (P < 0.05). The activity of Caspase-3 and Caspase-9 increased from 0 to 24 h postmortem but fell from 24 to 72 h postmortem. Compared with the control group, dietary selenium yeast and jujube powder significantly reduced mitochondrial oxidative damage. They greatly increased the shear force, mitochondrial membrane potential, and Cyt-c reduction levels (P < 0.05). Among them, the combination group of high-dose selenium yeast and jujube powder had more significant effects on ROS scavenging, reducing cell membrane permeability, protecting cell membrane integrity, and increasing Cyt-c reduction level (P < 0.05). In conclusion, cell apoptosis intensifies during the chicken breast's aging time, and muscle tenderness continues. Still, different doses of dietary selenium yeast and jujube powder can inhibit mitochondrial oxidation to various degrees. The combined group of selenium yeast and jujube powder with 0.6 mg·kg−1 has the best effect. This study is of great significance for applying natural antioxidant ingredients such as selenium yeast and jujube powder in the development and utilization of poultry feed.

Twelve Antioxidant Peptides From Protein Hydrolysate of Skipjack Tuna (Katsuwonus pelamis) Roe Prepared by Flavourzyme: Purification, Sequence Identification, and Activity Evaluation.

For using aquatic by-products to manufacture high-value products, Skipjack tuna (Katsuwonus pelamis) roes were degreased, pretreated with microwave, and hydrolyzed using five proteases. The protein hydrolysate (TRPH) generated using Flavourzyme displayed the strongest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. Twelve antioxidative peptides were prepared from TRPH by ultrafiltration and chromatography methods and determined to be SGE, VDTR, AEM, QDHKA, TVM, QEAE, YEA, VEP, AEHNH, QEP, QAEP, and YVM with molecular weights of 291.24, 489.50, 349.41, 597.59, 349.44, 475.42, 381.36, 343.37, 606.58, 372.35, 443.42, and 411.49 Da, respectively. AEM, QDHKA, YEA, AEHNH, and YVM presented the strongest scavenging activity on DPPH radical (EC50 values of 0.250±0.035, 0.279±0.017, 0.233±0.012, 0.334±0.011, and 0.288±0.015 mg/ml, respectively), hydroxyl radical (EC50 values of 0.456±0.015, 0.536±0.021, 0.476 ± 0.051, 0.369 ± 0.052, and 0.413 ± 0.019 mg/ml, respectively), and superoxide anion free radical (EC50 values of 0.348 ± 0.018, 0.281 ± 0.013, 0.305 ± 0.022, 0.198 ± 0.011, and 0.425 ± 0.021 mg/ml, respectively). Moreover, AEM, QDHKA, YEA, AEHNH, and YVM presented high lipid peroxidation inhibition ability, Ferric-reducing power, and significant protective function on H2O2-induced Chang liver cells. Therefore, AEM, QDHKA, YEA, AEHNH, and YVM could be natural antioxidant ingredients used in pharmaceutical and functional products.

Antioxidant peptides from protein hydrolysate of skipjack tuna milt: Purification, identification, and cytoprotection on H2O2 damaged human umbilical vein endothelial cells

To make full use of tuna by-products to prepare high-value products, skipjack tuna (Katsuwonus pelamis) milt was degreased, pretreated with microwave and separately hydrolyzed using trypsin, Neutrase, papain, and pepsin. Neutrase hydrolysate with the highest degree of hydrolysis (29.54 ± 1.19 %) and antioxidant activities were separated by ultrafiltration and a series of chromatographic methods, and thirteen antioxidant oligopeptides were prepared and identified as GHHAAA, PHPR, AKHQ, GRVPRV, ADMYW, VDDDD, SVTEV, VKIYI, VRDQY, IRDDY, YREY, AQRPR, and SMDV with molecular weights of 562.6, 505.6, 482.7, 654.8, 685.1, 578.0, 534.2, 634.6, 679.2, 681.2, 630.0, 626.4, and 450.0 Da, respectively. Moreover, GHHAAA, PHPR, SVTEV, VRDQY, and SMDV showed the highest Ferric-reducing power and radical scavenging activity on DPPH (EC50 values of 1.09 ± 0.08, 1.19 ± 0.07, 0.89 ± 0.08, 0.88 ± 0.11, and 1.24 ± 0.09 mg/mL, respectively) and ABTS (EC50 values of 1.12 ± 0.06, 1.14 ± 0.11, 1.02 ± 0.07, 1.04 ± 0.12, and 1.16 ± 0.07 mg/mL, respectively). In addition, GHHAAA, PHPR, SVTEV, VRDQY, and SMDV presented significant protective function on H2O2 oxidative damaged HUVECs by increasing the HUVECs viability and antioxidant enzyme activity, and lower the contents of ROS and MDA. This study suggested that GHHAAA, PHPR, SVTEV, VRDQY, and SMDV could be natural antioxidant ingredients used in pharmaceutical and functional products.

Performance of Strobilanthes crispus Leaves Extract as Potential Antioxidant Compound in Oil-in-water Emulsion

Excess of free radicals accelerates the degradation of oil-in-water (O/W) emulsion due to oxidative damage resulting in low emulsion quality such as in lotion formulation. The blend of antioxidants in O/W emulsion could reduce the damage caused by the radical compounds. The crude extract of Strobilanthes crispus (S. crispus) leaves was screened for assessing bioactive phytochemical constituents. By using various chemical assays, the extract of S. crispus leaves revealed the presence of tannins, steroids, flavonoids, terpenoids and glycosides. Besides that, the antioxidant activity of the leaf extract, spray-dried extract, a lotion containing extract and other local and abroad commercial lotions were determined as references using standard methods such as total phenolic content (TPC), total flavonoid content (TFC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and reducing power assay. In terms of TFC, the highest, 10.591±0.01 mg QE/g was recorded in crude extract along with the highest reducing power and percentage inhibition, 25.40±0.02 mg GAE/g and 88.89 % respectively. The result from TPC value can be a good indicator for a longer shelf life of the O/W emulsion if compared to other results from TPF, reducing power and DPPH value. From this study, it can be concluded that S. crispus has potential as a good natural antioxidant ingredient for the emulsion of O/W formulation.

Valorization of Peel Wastes of Purple Turnip (Brassica rapa L.): Extraction of Polyphenolics Through Ultrasonic-Assisted Extraction and Investigation of Changes in Total Phenolic Content, Total Monomeric Anthocyanin Content and Total Antioxidant Capacity During in vitro Gastro-Intestinal Digestion

Fruits and vegetables include high levels of bioactive compounds with health-promoting effects. Especially, peels, which are one of the generaly inedible fractions of fruit or vegetable, have higher amount of polyphenolic compounds than their pulps or seeds, since they accumulate in their peels. Purple turnip (Brassica rapa L.) is one of the important members of Brassicaceae family due to its nutritional composition. In this context, the aim of the study was to reveal the potency of peels from purple turnip as a natural antioxidant ingredient To reach this aim, the spectrophotometric assays including the Folin-Ciocalteu method for total phenolic content (TPC), the pH differential method for total monomeric anthocyanin content (TMAC) and CUPRAC and DPPH methods for total antioxidant capacity (TAC) before and after in vitro gastro-intestinal digestion were carried out. According to results of the study, TPC and TMAC of the polyphenolic axtract obtained by ultrasound-assisted extraction were 169.29±6.89 mg GAE/g dw and 159.53±10.82 mg CGE/L, respectively. The TAC of the extracts was 44.19±0.10 mg TE/g dw in CUPRAC assay and 38.52±0.06 mg TE/g dw in DPPH assay. The bioaccessibilities of total phenolics and anthocyanins from extracts after in vitro digestion process were approximately 17% and 6% and the antioxidant capacity of the bioaccessible extract fraction was approximately 28% (CUPRAC) and 0% (DPPH).

Development of a Natural Preservative from Chestnut Flowers: Ultrasound-Assisted Extraction Optimization and Functionality Assessment

This study was carried out with the aim of optimizing the ultrasound-assisted extraction (UAE) of phenolic compounds from male chestnut flowers (C. sativa Mill) to develop a bioactive extract with potential to be used as a natural antioxidant preservative ingredient in the food industry. Time (t, 1–39 min), solvent concentration (S, 0–100%), and ultrasonic power (P, 5–500 W) were used as the independent variables for a 5-level experimental circumscribed central composite design (CCCD) coupled with response surface methodology (RSM) to optimize the extraction of phenolic compounds by UAE. Regarding the variables, the three showed a significant effect on the extraction of phenolic compounds. The content of phenolic compounds (including flavonoids and tannins) and the extraction yield (extract weight gravimetrically assessed) were the response criteria for the optimization. Based on the statistically validated predictive polynomial models, it was possible to reach a maximum content of phenolic compounds at the global optimal conditions of 24 ± 3 min, 259 ± 16 W, and 51 ± 7% ethanol. Additionally, pentagalloyl-glucoside and trigalloyl-hexahydroxydiphenoyl-glucoside were the major phenolic compounds identified. The optimized extract was then analyzed for their biological properties. The bioactive potential of the chestnut flower extract obtained under these optimized conditions was evaluated using in vitro assays for antioxidant, anti-inflammatory, and antimicrobial activity, as well as cytotoxicity and hepatotoxicity tests. The results revealed that the enriched extract has antioxidant, antitumoral, and anti-inflammatory activities without toxicity issues. Overall, this study allowed to define the optimal conditions for the extraction of phenolic compounds from chestnuts male flowers by UAE, to obtain an enriched extract with biological properties that could be further used as a natural antioxidant ingredient with applications on functional foods.

Karakteristik Mutu Food Grade Grease Ramah Lingkungan Berbahan Baku Minyak Bekas Penggorengan

Food grade grease is a lubricant based on vegetable oils, in its safe use for health, does not pollute the environment so it is very beneficial for the pharmaceutical, animal feed, cosmetics and food industry industries in particular. This study aims to obtain a basic formulation of natural antioxidant ingredients and optimum long storage periods that can produce good and long lasting food grade grease, according to quality standards including: corrosion resistance, dropping point, lubricant texture, and pH . The purpose is more detailed, namely to know (1) the treatment of the percentage of activated charcoal (b / b) on the weight of the material as bleaching material at a temperature of 1050C for 1 hour; (2) the effect of increasing the concentration of natural antioxidant ingredients on food grade grease quality; (3) the best storage period for maintaining quality food grade grease quality; and (4) the interaction between the addition of the concentration of anti-corosit chemicals with the treatment period of storage for 5 days, 10 days and 15 days against the quality of food grade grease. The results of the study show (1). 5% activated charcoal is the best treatment, because it produces FFA values, low water content values ​​and visual appearance (color) with values ​​of 0.22%, 0.12%, and bright yellow, and (2). The treatment of the concentration of natural antioxidant material as much as 10% (A3) and treatment of 10 days storage period (T2) is the best treatment to produce food grade grease quality including corocyte test value, melting point, lubricant texture, and pH respectively: 7.99 ; 83.99; 2.49; and 6.99.

Combination of light and oxygen accelerates formation of covalent protein-polyphenol bonding during chill storage of meat added 4-methyl catechol

Plant polyphenols applied as natural antioxidant ingredients, are known to bind to cysteine residues on meat proteins. The aim of this study was to examine the effect of light exposure on the formation of cysteine-phenol adduct in meat added 4-methylcatechol (4MC), a model polyphenol, during storage through quantitative LC-MS/MS-based analysis. Cysteine-4-methylcatechol adduct (Cys-4MC) formation in meat added 1500 ppm 4-MC increased significantly (by 50%) when stored under light in oxygen at 4 °C for 7 days as compared to storage in the dark. This was reflected by a significant decrease in thiol concentrations in the same sample. Gel electrophoresis showed loss in myosin heavy chain (MHC), and a resulting increase in cross-linked MHC (CL-MHC) and larger protein polymers in samples added 4MC. Protein blots stained with nitroblue tetrazolium (NBT) showed intensive protein-polyphenol binding in the meat samples added 4MC, but no major differences between storage conditions.

Acerola fruit as a natural antioxidant ingredient for gluten-free bread: An approach to improve bread quality.

In this study, we evaluated the effect of enrichment of bread using acerola fruit powder on the physical, sensorial and antioxidant properties of gluten-free breads. We tested different proportions of acerola fruit powder (0-5% w/w) in rice flour. According to the results, loaf volume increased from 423.33 cm3 to 571.67 cm with increasing amount of acerola fruit powder cm3 with increasing amount of acerola fruit powder (from 0 to 5% w/w). Acerola fruit powder improved the structural parameters of the crumb by increasing the size and area fraction of cells. All tested quantities of acerola fruit powder improved textural parameters by decreasing firmness and chewiness and by increasing springiness. In addition, acerola fruit powder positively affected the antioxidant properties of enriched breads. The total phenolic content and antioxidant activity of extracts was found to be increased with the addition of acerola fruit powder. All antioxidant activities were found to be increased with increasing quantities of acerola fruit powder. The sensory attributes of the bread showed that a partial replacement of the rice flour with up to 3% of acerola fruit powder provided satisfactory results. The optimum level of acerola fruit powder for all parameters tested was found to be 3% w/w.

Characterization of Antioxidant Potential of Seaweed Extracts for Enrichment of Convenience Food

In recent years, there has been a growing interest in natural antioxidants as replacements of synthetic compounds because of increased safety concerns and worldwide trend toward the usage of natural additives in foods. One of the richest sources of natural antioxidants, nowadays largely studied for their potential to decrease the risk of diseases and to improve oxidative stability of food products, are edible brown seaweeds. Nevertheless, their antioxidant mechanisms are slightly evaluated and discussed. The aims of this study were to suggest possible mechanism(s) of Fucus vesiculosus antioxidant action and to assess its bioactivity during the production of enriched rye snacks. Chemical and cell-based assays indicate that the efficient preventive antioxidant action of Fucus vesiculosus extracts is likely due to not only the high polyphenol content, but also their good Fe2+-chelating ability. Moreover, the data collected during the production of Fucus vesiculosus-enriched rye snacks show that this seaweed can increase, in appreciable measure, the antioxidant potential of enriched convenience cereals. This information can be used to design functional foods enriched in natural antioxidant ingredients in order to improve the health of targeted consumers.

Cocoa pod husk has not been used optimally by farmers, most of cocoa pod husk will discarded around the cacao tree or collected in one hole and then piled with soil. Cocoa pod husk waste can be used optimally by extracting polyphenol compounds which are used as natural antioxidant ingredients. This research aimed to determine the effect of particle size and maceration time on cocoa pod husk extract as a source of antioxidant and determine the best particle size and maceration time to produce cocoa pod husk extract as a source of antioxidants. This research used factorial randomized block design with two factors. Factor 1 is a particle size consisting of 40, 60, and 80 mesh. Factor 2 is maceration time consisting of 24, 36, and 48 hours. The data were analyzed by analysis of variance and then it will be proceed with tukey test. The results showed that particle size and maceration time affected yield, total phenolic and antioxidant capacity of cocoa pod husk extract, while the interaction was affected on total phenolic and antioxidant capacity but had no effect on yield of cocoa pod husk extract. The best treatment to produce cocoa pod husk extract as a source of antioxidants 80 mesh particle size and maceration time for 48 hours, with a yield value of 5.22 ± 0.05 percent, a total phenolic of 148.09 ± 0.00 mg GAE / g and antioxidant capacity of 118.71 ± 0.13 mg GAEAC / g.&#x0D; Keywords: cocoa pod husk, extraction, antioxidants, particle size, maceration time

Investigation of antioxidant capacity of peptide fractions from the Tra catfish by-product-derived proteolysate using Flavourzyme® 500 mg

In this study, the antioxidant potential of peptide fractions isolated from the Tra catfish (Pangasius hypophthalmus) by-product-derived hydrolysate using ultrafiltration centrifugal devices with 3 distinct molecular-weight cutoffs (MWCOs) of 5KDa, 10KDa, and 30KDa was investigated. Firstly, the chemical composition of the Tra catfish byproducts was analyzed. The result showed that the Tra catfish by-products contained 58.5% moisture, 33.9% crude protein, 50.1% crude lipid and 15.8% ash (on dry weight basis). Secondly, the effects of enzyme content, hydrolysis time on the antioxidant activity of the hydrolysate were studied using DPPH• (2,2-diphenyl-1-picrylhydrazyl) radical scavenging method (DPPH• SM), and FRAP (ferric reducing antioxidant potential) method. Flavourzyme® 500 MG was used for hydrolysis. The result revealed that the antioxidant activity of the proteolysate reached the peak when the hydrolysis time was 3h, enzyme/substrate (E/S) ratio was 25 U/g protein, hydrolysis temperature was 500C, pH was 7, and the degree of hydrolysis (DH) of the hydrolysate reached 37.4%. Next, the proteolysate was further fractionated using MWCOs of 5KDa, 10KDa, and 30KDa and the peptide fractions were investigated for their antioxidant activity. The result showed that the &lt;5KDa fraction showed strongest antioxidant activity with the 50% DPPH• inhibition concentration (IC50) of 3079.34±75.29 μg/mL and FRAP value of 611.28±4.5 μM Trolox equivalent. The second strongest fraction was 5-10KDa with the IC50 and FRAPS value of 4709.3 ± 222.22 μg/mL and 323.62±2.54 μM Trolox equivalent, respectively. The 10-30KDa fraction showed weakest antioxidant capacity with the IC50 of 5847.73 ± 128.68 μg/mL and FRAP value of 291.4±2.65 μM Trolox equivalent. The antioxidant peptides derived from Tra catfish by-products proteolysate have the potential to be used as natural antioxidant ingredients in pharmaceutical and food industry.

Red seaweed Pyropia columbina as antioxidant supplement in feed for cultured juvenile Pacú (Piaractus mesopotamicus)

The aim of this study was to evaluate the antioxidant effect of red seaweed Pyropia columbina on juvenile Pacu (Piaractus mesopotamicus). Two hundred and ten (n = 210) fish were fed for 90 days with a control extruded feed (CEF) or the same feed added with P. columbina (35 g kg−1) (PcEF). No significant difference in final body weight, specific growth rate, condition factor, and liver somatic index was detected between dietary treatments (p > 0.05). Fish fed with PcEF showed lower lipid peroxidation (28.2 ± 2.9 vs. 39.4 ± 3.7 mmol malondialdehyde mg−1 protein) and superoxide dismutase (23.3 ± 2.3 vs. 37.7 ± 3.0 U mg−1 protein) and reduced SOD/CAT ratio (5.5 ± 0.3 vs. 8.1 ± 0.7) in intestine than those fed with CEF (p 0.05). PcEF showed higher hemoglobin (7.5 ± 0.4 vs. 6.6 ± 0.3 g 100 mL−1) and mean corpuscular hemoglobin concentration (18.4 ± 1.8 vs. 25.7 ± 2.7%) than those fed with CEF diet (p < 0.05). Beneficial effects on lipid metabolism were observed in fish fed with PcEF respect to control diet. Pyropia columbina could be used as a natural antioxidant ingredient in fish feeding contributing a better nutritional status of cultured fish.

Controlling Lipid Oxidation and Volatile Compounds in Frozen Fried Fish Cake Prepared with Rice Bran Hydrolysate

ABSTRACTFried fish cakes formulated with (1) no antioxidant (control), (2) 1% rice bran hydrolysate (RBH), (3) 2% RBH, (4) 0.05% rosemary oil, and (5) 0.02% butylated hydroxyanisole/butylated hydroxytoluene (BHA/BHT) were investigated for their oxidation values following 0–9 freeze-thaw cycles. Both lipid oxidation and protein oxidation were significantly obstructed when RBH or BHA/BHT was used. RBH at 2% was equally effective as 0.02% BHA/BHT. Headspace solid phase microextraction (HS-SPME), which measures volatile compounds that determine lipid oxidation, demonstrated the effectiveness of RBH. The development of rancid volatile compound (i.e., hexanal) levels in fried fish cakes decreased during 0–9 freeze-thaw cycles. This study demonstrates that RBH is as an effective antioxidant comparable to commercial antioxidants (BHA/BHT) in frozen fried fish cakes. Consequently, RBH can be a consumer-friendly and natural antioxidant ingredient.

Glycosaminoglycans from a Sea Snake (Lapemis curtus): Extraction, Structural Characterization and Antioxidant Activity.

Sea snakes have wide application prospects in medicine, health food and other fields. Several novel polysaccharides were successfully obtained from the skin and the meat of a sea snake (Lapemis curtus). The structures of polysaccharides LSP3 and LMP3, which were extracted and purified from Lapemis curtus, were determined to be new and highly heterogenic glycosaminoglycans (GAGs) by means of FT-IR, ESI-MS/MS and NMR. LSP3 is a hybrid dermatan sulfate (DS) and composed of 48% 4-sulfated disaccharides (Di4S), 42% 6-sulfated disaccharides (Di6S) and 5% disulfated disaccharides (Di2,6S), while LMP3 is a hybrid chondroitin sulfate (CS) and composed of 70% Di4S, 20% Di6S, and 8% Di2,6S. More importantly, LSP3 and LMP3 showed a strong scavenging ability of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, iron (Fe2+) chelating activity and total antioxidant capacity in vitro, especially LSP3, with high contents of uronic acid and sulfate, which possessed a higher scavenging ability of DPPH radicals than other fractions. These data suggested that the sea snake polysaccharides could be promising candidates for natural antioxidant ingredients.
