A New Way for Beta Cell Neogenesis: Transdifferentiation from Alpha Cells Induced by Glucagon-Like Peptide 1.

Recent studies showed that alpha cells, especially immature cells and proalpha cells, might be the precursors of beta cells. Exposure to glucagon-like peptide 1 (GLP1) can ameliorate hyperglycemia in diabetic mice and restore the beta cell mass. In the present study, we adopted single high-dose (60 mg/kg, i.p.) streptozotocin (STZ) to model diabetes mellitus (DM) and randomly assigned short-tail (SD) rats to a normal group, a diabetic group, GLP1 groups (50 μg/kg, 100 μg/kg, and 200 μg/kg), a GLP1 (200 μg/kg) with exendin (9-39) group, and a GLP1 with LY294002 group. We found that the pancreatic insulin-glucagon-positive cell populations increased according to the increase in GLP1 exposure. By contrast, no insulin-amylase-positive cell populations or insulin/pan-cytokeratin cells were observed in the pancreatic sections. The GLP1 receptor antagonist exendin (9-39) and the phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K) family inhibitor LY294002 not only suppressed protein kinase B (Akt), pancreatic and duodenal homeobox 1 (Pdx1), forkhead box O 1 (FoxO1), and mast cell function-associated antigen A (MafA) mRNA expression but also increased MAFB expression. We concluded that treatment with GLP1 might result in beta cell neogenesis by promoting the transdifferentiation of alpha cells but not by pancreatic acinar cells, ductal cells, or the self-replication of beta cells. The regulation on the GLP1 receptor and its downstream transcription factor PI3K/AKT/FOXO1 pathway, which causes increased pancreatic and duodenal homeobox 1 (Pdx1) and MafA mRNA expression but causes decreased MAFB expression, may be the mechanism involved in this process.

Mast cell inhibitory receptors

Mast cells play an important role in diverse physiological mechanisms as well as taking part in antimicrobial defense. What is more, these cells are important regulators of a number of pathophysiological processes, involving allergic reactions. Therefore, it seems to be very important to know and understand the factors and receptors influencing mast cell activity. Nowadays it is well established that activating signals are counterbalanced by negative or inhibition signals transmitted by inhibitory receptors containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs). Inhibitory receptor engagement leads to ITIM tyrosine phosphorylation, the recruitment and activation of protein tyrosine phosphatases such as SHP-1, SHP-2 and/or SHIP, and the dephosphorylation of activating receptor associated proteins. There is growing evidence that a number of inhibitory receptors have been identified on mast cells. The scope of this paper is to present the current knowledge on mast cell-associated inhibitory receptors, such as FcγRIIB, paired immunoglobulin-like receptor B (PIR-B), CD300, CD172a, gp49B1, CD200R, sialic acid-binding immunoglobulin-like lectin (Siglec) molecules, CD305, allergin-1, mast cell function-associated antigen (MAFA), and CD72. The role of these inhibitory receptors in regulation mast cell activity is also discussed.

Compartmentalization of the Type I Fcε receptor and MAFA on mast cell membranes

The Mast cell Function-associated Antigen (MAFA) is a membrane glycoprotein on rat mast cells (RBL-2H3) expressed at a ratio of ∼ 1:30 with respect to the Type I Fcε receptor (FcεRI). Despite this stoichiometry, clustering MAFA by its specific mAb G63 substantially inhibits secretion of both granular and de novo synthesized mediators induced upon FcεRI aggregation. Since the FcεRIs apparently signal from within raft micro-environments, we investigated possible co-localization of MAFA within these membrane compartments containing aggregated FcεRI. We used cholera toxin B subunit (CTB) to cluster the raft component ganglioside GM1 and studied the effects of this perturbation on rotation of FcεRI and MAFA by time-resolved phosphorescence anisotropy of erythrosin-conjugated probes. CTB treatment would be expected to substantially inhibit rotation of raft-associated molecules. Experimentally, CTB has no effect on rotational parameters such as the long-time anisotropy ( r ∞) of unperturbed FcεRI or MAFA. However, on cells where FcεRI-IgE has previously been clustered by antigen (DNP 14-BSA), CTB treatment increases the FcεRI-IgE's r ∞ by 0.010 and MAFA's by 0.014. Similarly, CTB treatment of cells where MAFA had been clustered by mAb G63 increases MAFA's r ∞ by 0.010 but leaves FcεRI's unaffected. Evaluation of raft localization of FcεRI and MAFA using sucrose gradient ultracentrifugation of Triton X-100 treated membrane fragments demonstrates that a significant fraction of MAFA molecules sediments with rafts when FcεRI is clustered by antigen or when MAFA itself is clustered by mAb G63. The large excess of FcεRI over MAFA explains why clustering MAFA does not substantively affect FcεRI dynamics. Moreover, in single-particle tracking studies of individual FcεRI-IgE or MAFA molecules, these proteins, upon clustering by antigen, move into small membrane compartments of reduced, but similar, dimensions. This provides additional indication of constitutive interactions between FcεRI and MAFA. Taken together, these results of distinct methodologies suggest that MAFA functions within raft microdomains of the RBL-2H3 cell membrane and thus in close proximity to the FcεRI which themselves signal from within the raft environment.

Selective inhibition of the FcɛRI-induced de novo synthesis of mediators by an inhibitory receptor

Aggregation of the type 1 Fc-epsilon receptors (Fc-epsilon-RI) on mast cells initiates a network of biochemical processes culminating in secretion of both granule-stored and de novo-synthesized inflammatory mediators. A strict control of this response is obviously a necessity; nevertheless, this regulation is hardly characterized. Here we report that a prototype inhibitory receptor, the mast cell function-associated antigen (MAFA), selectively regulates the Fc-epsilon-RI stimulus-response coupling network and the subsequent de novo production and secretion of inflammatory mediators. Specifically, MAFA suppresses the PLC-gamma2-[Ca2+]i, Raf-1-Erk1/2, and PKC-p38 coupling pathways, while the Fyn-Gab2-mediated activation of PKB and Jnk is essentially unaffected. Hence, the activities of several transcription/nuclear factors for inflammatory mediators (NF-kappaB, NFAT) are markedly reduced, while those of others (Jun, Fos, Fra, p90rsk) are unaltered. This results in a selective inhibition of gene transcription of cytokines including IL-1beta, IL-4, IL-8, and IL-10, while that of TNF-alpha, MCP-1, IL-3, IL-5, or IL-13 remains unaffected. Taken together, these results illustrate the capacity of an immunoreceptor tyrosine-based inhibitory motif-containing receptor to cause tight and specific control of the production and secretion of inflammatory mediators by mast cells.

Co-clustering activating and inhibitory receptors: Impact at varying expression levels of the latter

Clustering the mast cell function-associated antigen (MAFA) has earlier been shown to inhibit mast cells’ secretory response to the type 1 Fcɛ receptor (FcɛRI) stimulus. MAFA is a type II membrane glycoprotein first identified on rat mast cells and contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) in its cytosolic domain. This inhibition is induced already upon clustering MAFA alone. Still, the inhibitory capacity of MAFA–FcɛRI co-clustering has recently been characterized and co-clustered MAFA molecules were found to exhibit a markedly higher inhibition capacity than MAFA-clusters alone. We have now compared the inhibitory capacity of FcɛRI co-clustered MAFA on the secretory response of rat mucosal-type mast cells (RBL-2H3 line) expressing different levels of this inhibitory protein. Reacting these cells carrying an IgE class, 2,4 dinitrophenyl (DNP)-specific monoclonal antibody with DNP-conjugated F(ab′) 2 fragments of non-specific polyclonal mouse IgG causes clustering of the FcɛRI–IgE. Reaction of these cells with DNP-conjugated F(ab′) 2 fragments of the MAFA-specific, monoclonal antibody G63 co-aggregates MAFA together with the FcɛRI–IgE thereby producing FcɛRI–IgE–MAFA co-clusters. Results of measurements of the secretory responses of RBL-2H3 cells expressing higher or lower MAFA levels than those of unmodified cells provided further support to the notion that co-clustered MAFA molecules exhibit a markedly higher inhibition capacity than MAFA-clusters alone. The molecular basis for this enhanced inhibition is most probably the increased concentration of the inhibitory cell components in the immediate proximity of the co-clustered FcɛRI–MAFA.

Regulation of mast cells’ secretory response by co‐clustering the Type 1 Fcϵ receptor with the mast cell function‐associated antigen

The mast cell function-associated antigen (MAFA) is a type II membrane glycoprotein first identified on rat mast cells and basophils. Clustering MAFA inhibits these cells' secretory response to the type 1 Fcepsilon receptor (FcepsilonRI) stimulus. To quantitatively characterize this inhibition and its dependence on MAFA-FcepsilonRI co-clustering, we investigated the secretory response of rat mucosal-type mast cells of the RBL 2H3 line carrying an IgE class, 2,4 dinitrophenyl (DNP) specific monoclonal antibody to DNP-conjugated Fab and F(ab')(2) fragments of (1) mouse IgG, and (2) of the MAFA-specific, monoclonal antibody G63. The first reagent clusters FcepsilonRI-IgE complexes into oligomers by reacting with the DNP residues. The DNP conjugated G63 Fab and F(ab')(2) fragments, additionally aggregate MAFA and form FcepsilonRI-IgE-MAFA co-clusters. All experiments using these ligands were performed in the absence or presence of an excess of intact mAb G63, which clusters MAFA molecules. Empirical Hill functions were used to relate the secretory response of mast cells to the equilibrium concentrations of FcepsilonRI-IgE or MAFA clusters and co-clusters calculated as function of the employed ligands concentrations. This analysis of the experimental results indicates that co-clustered MAFA molecules have a markedly higher inhibitory capacity than MAFA-clusters alone. The molecular basis of the enhanced inhibition observed upon co-clustering MAFA with the FcepsilonRI is most probably the increased concentration of the inhibitory cell components in the immediate proximity of the activation coupling elements.

243 REPROGRAMMING OF FIBROBLASTS TO EXPRESS MALE GERM CELL OR MAST CELL FUNCTIONS USING TESTIS OR MAST CELL-DERIVED CELL EXTRACTS

The concept of epigenetic reprogramming of a somatic nucleus was supported by the birth of cloned animals and the derivation of embryonic stem cells after nuclear transplantation into oocytes. Moreover, recent studies reported that the direct transformation of one differentiated somatic cell type into another is possible and would be advantageous for producing isogenic replacement cells. In this study, we were able to modulate the cell fate of fibroblasts by introducing cell extracts derived from a mast cell line, RBL-2H3. NIH-3T3 cells were treated with streptolysin O (SLO; 230 ng/mL), which reversibly permeablizes plasma membrane, and incubated for 30 min with the mast cell- derived cell extracts (4 mg/mL). After resealing the membrane of the cells, we incubated the cells for 3 weeks and then analyzed them for the expression of mast cell-specific genes such as MAFA (mast cell function-associated antigen) and FceRI (high-affinity IgE receptor). Our results showed that the cell extracts can activate the expression of mast cell-specific genes, implying that cell extracts can provide regulatory components required for reprogramming the cell fate to initiate a transcriptional program specific for the cell type. Moreover, mast cell-specific degranulation and cell morphology changes were observed in cultured mouse fibroblasts. We could detect mast cell-specific functions even after 15 days of incubation. Next, to induce porcine fibroblasts to take on testis sertoli cell-specific properties, we reversibly permeablized porcine primary fibroblasts with SLO (230 ng/mL) and incubated the cells with porcine testis extracts (4 mg/mL). As expected, in the reprogrammed primary porcine fibroblasts, the porcine testis extracts activated the expression of porcine testis sertoli cell-specific genes including protamine 1 and 2, SOX9, MIS (mullerian inhibitory substance), preproacrosine (ACR), phosphoglycerate kinase-2 (PGK-2), protein C, and c-kit ligand. The male germ cell functions were sustained for more than 10 days after the reprogramming process. Taken together, our data suggest that testis- or mast cell-derived cell extracts can reprogram fibroblasts to express male germ cell or mast cell functions, respectively, supporting the concept that cell extracts can reprogram the genome activity to activate cell-specific gene expression. Stable knockdown of MAFA expression in RBL-2H3 cells by siRNA retrovirus-delivery system

The mast cell function-associated antigen (MAFA) is a type II membranal glycoprotein first discovered on the surface of the rat mucosal-type mast cells of the RBL-2H3 line. A C-type lectin domain and an immunoreceptor tyrosine-based inhibitory motif (ITIM) are located in the extracellular and intracellular domains of MAFA, respectively. MAFA clustering by its specific monoclonal antibody G63, has been previously shown to cause a dose-dependent inhibition of the secretory response of these cells to the FcεRI stimulus. More recently, human and mouse homologues of MAFA have also been discovered. However, they are expressed also or only by NK- and T cells, suggesting that MAFA may play additional functional role(s). To further pursue MAFA’s functional significance in mast cells, we have employed the recently developed siRNA (small inhibitory RNA)-dependent gene-specific silencing protocol. We have first incorporated the key elements of the well-defined and commercially available siRNA vector (pSilencer1.0-U6) together with anti-MAFA hairpin into a retroviral vector (pLXSNneo). This enabled generating retroviruses that induced a stable and efficient downregulation (knockdown) of MAFA’s expression. RBL-2H3 cells with either normal or knocked-down MAFA expression levels are currently examined for their biological responses e.g. FcεRI mediated secretion, cell proliferation and cell adhesion. So far however, no significant changes were resolved in the above biological functions.

Interactions of the mast cell function-associated antigen with the type I Fcε receptor

Clustering the mast cell function-associated antigen (MAFA), a membrane glycoprotein expressed on 2H3 cells, by its specific monoclonal antibody G63 substantially inhibits secretion normally triggered by aggregating these cells’ Type I Fcε receptor (FcεRI). To explore possible MAFA-FcεRI interactions giving rise to this inhibition, we have studied by time-resolved phosphorescence anisotropy the rotational behavior of both MAFA and FcεRI as ligated by various reagents involved in FcεRI-induced degranulation and MAFA-mediated inhibition thereof. From 4 to 37 °C the rotational correlation times (mean±S.D.) of FcεRI-bound, erythrosin-conjugated IgE resemble those observed for MAFA-bound erythrosin-conjugated G63 Fab, 82±17 μs and 79±31 μs at 4 °C, respectively. Clustering the FcεRI-IgE complex by antigen or by anti-IgE increases the phosphorescence anisotropy of G63 Fab and slows its rotational relaxation. Lateral diffusion of G63 Fab is also slowed by antigen clustering of the receptor. Taken together, these results suggest that unperturbed MAFA associates with clustered FcεRI. They are also consistent with its interaction with the isolated receptor, a situation also suggested by FRET measurements on the system.

Clustering the mast cell function-associated antigen (MAFA) leads to tyrosine phosphorylation of p62 Dok and SHIP and affects RBL-2H3 cell cycle

The mast cell function-associated antigen (MAFA) is a type II membranal glycoprotein expressed by rat mast cells and basophils. MAFA clustering by its specific monoclonal antibody, (mAb) G63, efficiently inhibits the FcεRI induced secretory response of mucosal-type mast cells of the RBL-2H3 line, as well as bone marrow-derived mast cells. Here we present results which suggest that MAFA has also a capacity of modulating the cell cycle of the RBL-2H3 line. We found that MAFA clustering, by mAb G63 or by its F(ab′) 2 fragments, reduces the cell proliferation rate. Cell cycle analysis by flow cytometry revealed that the number of cells in sub-G phase is considerably higher for cells on which MAFA was clustered. Results of biochemical experiments established that MAFA clustering leads to a marked increase in the transient tyrosine phosphorylation of the adaptor protein p62 Dok and the inositol phosphatase SHIP. Concomitantly, their respective binding to RasGAP and Shc was increased. Furthermore, the GTP binding protein Sos1 was found to dissociate from Shc upon MAFA clustering, suggesting that SHIP and Sos1 compete for Shc binding. We therefore suggest that MAFA has also a role in regulating RBL-2H3 cell proliferation rate by inhibiting RasGTP formation in the Ras signaling pathway.

The mast cell function-associated antigen and its interactions with the type I Fcepsilon receptor.

The nature of MAFA-FcepsilonRI interactions giving rise to this inhibition remains unclear. Rotational diffusion of a membrane protein is a sensitive probe of its involvement in intermolecular interactions. We have therefore studied by time-resolved phosphorescence anisotropy the rotational behavior of both MAFA and FcepsilonRI as ligated by various reagents involved in FcepsilonRI-induced degranulation and MAFA-mediated inhibition thereof. The protein tyrosine kinase syk activity is reduced by clustering the mast cell function-associated antigen.

The mast cell function-associated antigen (MAFA) is a glycoprotein first identified on the membrane of rat mucosal-type mast cells (RBL-2H3 line). MAFA clustering causes a dose-dependent inhibition of these cells' secretory response to the type I Fcepsilon receptor (FcepsilonRI) stimulus. The inhibition has earlier been shown to take place upstream to the production step of inositol phosphates in the FcepsilonRI coupling cascade. To resolve further the mechanism of action of MAFA, we have investigated the events prior to the activation of phospholipase C. Activities of the non-receptor protein tyrosine kinases Lyn and Syk in untreated cells were compared with those where the FcepsilonRI, MAFA or both were clustered. Syk tyrosine phosphorylation and activation, as well as LAT (linker for activation of T cells) tyrosine phosphorylation, both induced by FcepsilonRI clustering, were found to be reduced upon MAFA clustering. In contrast, the activity of the Src homology domain 2 (SH2)-containing protein tyrosine phosphatase (SHP-2) increased. MAFA clustering also enhanced the co-isolation of SHP-2 and Syk with tyrosine-phosphorylated MAFA in both untreated and FcepsilonRI-stimulated cells. SHP-2 caused a decline in the FcepsilonRI-induced tyrosine phosphorylation of Syk, at least under in vitro conditions. Taken together, these results suggest that one possible mechanism by which MAFA affects the FcepsilonRI stimulation cascade is suppression of Syk activity, i.e. MAFA clustering leads SHP-2 to act on Syk, thereby reducing its tyrosine phosphorylation and its activity.

Genomic structure, alternative splicing, and physical mapping of the killer cell lectin-like receptor G1 gene (KLRG1), the mouse homologue of MAFA.

The mouse killer cell lectin-like receptor G1 (KLRG1), the mouse homologue of the mast cell function-associated antigen (MAFA), is an inhibitory C-type lectin expressed on natural killer (NK) cells and activated CD8 T cells. Here we report the complete nucleotide sequence, alternatively spliced variants, and the physical mapping of the KLRG1 gene in the mouse. The gene spans about 13 kb and consists of five exons. Short interspersed repeats of the B1 and B2 family, a LINE-1-like element, and a (CTT)170 triplet repeat were found in intron sequences. In contrast to human KLRG1 and to the murine KLR family members, mouse KLRG1 locates outside the NK complex on Chromosome 6 between the genes encoding CD9 and CD4.

An immunoreceptor tyrosine-based inhibitory motif, with serine at site Y-2, binds SH2-domain-containing phosphatases.

Clustering of the mast cell function-associated antigen by its specific monoclonal antibody (G63) inhibits the FcepsilonRI-mediated secretory response. The cytosolic tail of the mast cell function-associated antigen contains a SIYSTL stretch, a potential immunoreceptor tyrosine-based inhibition motif. To investigate the possible functional role of this sequence, as well as identify potential intracellular proteins that interact with it, peptides corresponding to residues 4-12 of the mast cell function-associated antigen's N-terminal cytoplasmic domain, containing the above motif, were synthesized and used in affinity chromatography of mast cell lysates. Both tyrosyl phosphorylated and thiophosphorylated mast cell function-associated antigen peptides bound the src homology domain 2 (SH2)-containing tyrosine phosphatases-1 (SHP-1), -2 (SHP-2) and inositol 5'-phosphatase (SHIP), though with different efficiencies. Neither the nonphosphorylated peptide nor its tyrosyl phosphorylated reversed sequence peptide bound any of these phosphatases. Point mutation analysis of mast cell function-associated antigen pITIM binding requirements demonstrated that for SHP-2 association the amino acid residue at position Y-2 is not restricted to the hydrophobic isoleucine or valine. Glycine and other amino acids with hydrophilic residues, such as serine and threonine, at this position also maintain this binding capacity, whereas alanine and acidic residues abolish it. In contrast, SHP-1 binding was maintained only when serine was substituted by valine, suggesting that the Y-2 position provides selectivity for peptide binding to SH2 domains of SHP-1 and SHP-2. These results were corroborated by surface plasmon resonance measurements of the interaction between tyrosyl phosphorylated mast cell function-associated antigen peptide and recombinant soluble SH2 domains of SHP-1, SHP-2 and SHIP, suggesting that the associations observed in the cell lysates may be direct. Taken together these results clearly indicate that the SIYSTL motif present in mast cell function-associated antigen's cytosolic tail exhibits characteristic features of an immunoreceptor tyrosine-based inhibition motif, suggesting it is a new member of the growing diverse family of immunoreceptor tyrosine-based inhibition motif-containing receptors.

Mast cell stimulation by co-clustering the type I Fcε-receptors with mast cell function-associated antigens

The secretory response of rat mucosal-type mast cells (line RBL 2H3) to stimuli produced by clustering or co-clustering two of its membranal components; the type I Fcε receptor and the mast cell function associated antigen (MAFA) was investigated. The primary reagents employed for this purpose were Fab fragments of the monoclonal antibodies J17 and G63 specific to the above respective proteins. The Fabs were then aggregated by F(ab′) 2 fragments of mouse IgG specific goat antibodies. This reaction was assumed to yield predominantly three different bivalent clustering reagents. Namely, dimers of the FcεRI specific (J17-Fab) 2; dimers of the MAFA specific, (G63-Fab) 2 and bispecific (J17-Fab-G63-Fab) dimers. The observed cellular secretory response was analyzed by employing a model which accounts for the clustering and co-clustering of FcεRIs and MAFAs by the above protocols. Results of this analysis provided evidence that at least some of the MAFA molecules are physically associated with the FcεRI. As a consequence, clustering of MAFA and FcεRI by bispecific J17-Fab-G63-Fab dimers induces secretion at comparatively low concentrations of these reagents, though with a significantly lower maximal response than that caused by the respective monospecific reagent (J17-Fab) 2. This result most likely reflects the inhibitory capacity of MAFA-FcεRI interaction.

The mast cell function-associated antigen, a new member of the ITIM family.

A decade has already passed since we discovered, essentially by serendipity, the membranal glycoprotein later named MAst cell Function-associated Antigen (MAFA). While trying to identify the still elusive Ca2+ channel responding to the type 1 Fcɛ receptor (FcɛRI) stimulus, a series of monoclonal antibodies (mAb) were raised to the rat mucosal-type mast cells of the RBL-2H3 line. Inhibitory lectin-like receptors expressed on the surface of hematopoietic cells are critically involved in regulation of their effector functions. Here we report that a novel mAb specific for mouse NK cells, 2F1, recognizes the mouse homolog of the mast cell function-associated antigen (MAFA), an inhibitory lectin-like transmembrane receptor expressed on rat mast cells. The 2F1 antigen (2F1-Ag) and rat MAFA are structurally highly conserved and contain a cytoplasmic motif similar to the immunoreceptor tyrosine-based inhibitory motif that is presumably utilized for inhibitory signaling. We also identified a human homolog that is closely related to the rodent MAFA/2F1-Ag proteins. Like rat MAFA, 2F1-Ag is probably encoded by a single gene, which exhibits relatively little polymorphism. Strikingly, while rat MAFA is considered a mast cell antigen, we have been unable to detect cell surface expression of 2F1-Ag by mouse mast cell lines, bone marrow-derived mast cells, or peritoneal mast cells. Furthermore, mouse bone marrow-derived mast cells were devoid of 2F1-Ag mRNA. Instead, we find that approximately 40% of mouse NK cells express 2F1-Ag. Thus, MAFA/2F1-Ag may modulate immunological responses on at least two different cell types bridging the specific and innate immune system.

MAFA-L, an ITIM-containing receptor encoded by the human NK cell gene complex and expressed by basophils and NK cells.

The natural killer cell gene complex on human chromosome 12p12-13 encodes several C-type lectin receptor genes expressed by NK cells and other hematopoietic cells. We have identified a novel receptor gene in this region encoding a putative type II transmembrane glycoprotein. The product is 54% identical to the rat mast cell function-associated antigen (MAFA), which inhibits mast cell activation by IgE. The human MAFA-like receptor (MAFA-L) and the rat MAFA protein are expressed by basophils and both have an immunoreceptor tyrosine-based inhibitory motif in the cytoplasmic tail, consistent with an inhibitory role in basophil activation. Unlike rat MAFA, expression of the MAFA-L gene is not limited to mast cells and basophils. In common with other genes in the NK cell gene complex MAFA-L is also expressed by natural killer cells as well as the monocyte-like cell-line U937. Expression in NK cells is restricted to peripheral blood NK cells, decidual NK cells do not express MAFA-L. While MAFA-L and rat MAFA might have a similar role in basophils, the expression of MAFA-L in other cell types implies additional functions for this molecule. The presence of the MAFA-L gene in the human NK cell complex indicates that this locus encodes C-type lectin receptors expressed by a variety of cells important in host defense.

Human MAFA has alternatively spliced variants

Human mast cell function-associated antigen (MAFA) cDNA has been cloned. This molecule is similar to the rat form having an intracellular domain containing a putative immunoreceptor tyrosine inhibition motif and an extracellular C type lectin-like domain. However, in contrast to rat MAFA, the amino acid sequence suggests the presence of two additional extracellular N-linked glycosylation sites. In addition, alternative mRNA transcripts are observed that differ substantially from those found in the rat.

The mast cell function-associated antigen exhibits saccharide binding capacity.

The mast cell function-associated antigen (MAFA) is a membrane glycoprotein first identified on rat mucosal type mast cells (line RBL-2H3) and known to inhibit the Fc epsilon RI-mediated secretory response. In its extracellular domain, an amino acid stretch homologous to the carbohydrate binding domain of calcium-dependent animal lectins has been found. To investigate its carbohydrate binding capacity, the MAFA has been expressed in the Spodoptera frugiperda insect cell line (Sf9) using the baculovirus expression system. Analysis by flow cytometry and surface labeling with 125I showed that the recombinant MAFA (rMAFA) was expressed as a monomeric and disulfide-linked homodimeric glycoprotein in the membrane of the insect cells, and both forms exhibited the same epitopes as the protein isolated from RBL-2H3 cells. Immunoaffinity-purified rMAFA was then employed for studies of its saccharide binding capacity by using different neoglycans and glycoproteins. The rMAFA was found to bind specifically terminal mannose residues in a Ca(2+)-dependent manner. These results support the notion that the extracellular domain of the MAFA is indeed able to bind ligands, which may be modulatory for the mast cell response.
