Towards Protection of Nucleic Acids from Herbicide Attack: Self-Assembly of Betaines Based on Pillar[5]arene with Glyphosate and DNA.

Herbicides are one of the main parts of pesticides used today. Due to the high efficiency and widespread use of glyphosate-based herbicides, the search for substances reducing their genotoxicity is an important interdisciplinary task. One possible approach for solving the problem of herbicide toxicity is to use compounds that can protect DNA from damage by glyphosate derivatives. For the first time, a method for developing DNA-protecting measures against glyphosate isopropylamine salt (GIS) damage was presented and realized, based on low-toxicity water-soluble pillar[5]arene derivatives. Two- and three-component systems based on pillar[5]arene derivatives, GIS, and model DNA from salmon sperm, as well as their cytotoxicity, were studied. The synthesized pillar[5]arene derivatives do not interact with GIS, while GIS is able to bind DNA from salmon sperm with lgKa = 4.92. The pillar[5]arene betaine derivative containing fragments of L-phenylalanine and the ester derivative with diglycine fragments bind DNA with lgKa = 5.24 and lgKa = 4.88, respectively. The study of the associates (pillar[5]arene-DNA) with GIS showed that the interaction of GIS with DNA is inhibited only by the betaine pillar[5]arene containing fragments of L-Phe (lgKa = 3.60). This study has shown a possible application of betaine pillar[5]arene derivatives for nucleic acid protection according to its competitive binding with biomacromolecules.

Performance de produtos à base de glyphosate aplicados isolados e em associações à herbicidas na dessecação

ABSTRACT The no-tillage system is a conservation system that helps sustainability and agricultural production. The effectiveness of glyphosate control, applied alone or in combination with other herbicides, can be altered depending on the product’s formulation. The objective of the present study was to evaluate the effectiveness of glyphosate in formulations containing different salts and concentrations, applied alone and in combination with other herbicides, in controlling weeds in advanced stages in the pre-sowing burndown operation. The experiment was carried out in the field in an area with a history of high weed infestation. The experiment was conducted in a randomized block design, evaluating eleven treatments and four replicates. The treatments consisted of the application of three glyphosate-based formulations alone and in combination with clethodim and 2,4-D amine herbicides, in addition to a control without herbicide application. The evaluated variables were percentage of weed control and percentage of desiccation. A comparison of means by contrasts was performed to analyze the percentage of weed control. In general, treatments containing products based on glyphosate potassium salt in the composition have slightly better control performance compared to those consisting of glyphosate isopropylamine salt.

Response of black swallowwort (Vincetoxicum nigrum) to herbicides plus mowing

AbstractThe invasive vine black swallowwort [Vincetoxicum nigrum (L.) Moench = Cynanchum louiseae Kartesz & Gandhi, Apocynaceae] is difficult to control, and herbicide studies are lacking. This long-lived perennial species is primarily found in high-light environments in natural areas and perennial cropping systems in northeastern North America. We conducted a 3-yr herbicide efficacy study, with or without mowing, in an old-field site infested with V. nigrum in Dutchess County, NY, USA. Experimental plots were either herbicide treated in early July or mowed in early July and subsequently herbicide treated in late August for 2 yr with the potassium salt of glyphosate (2.02 kg ae ha−1), the isopropylamine salt of glyphosate (1.35 kg ae ha−1), or the butoxyethyl ester of triclopyr (1.79 kg ae ha−1). Both glyphosate formulations were effective in reducing V. nigrum aboveground biomass, although they were somewhat less effective in reducing cover or stem densities of V. nigrum plants >10-cm tall after 2 yr compared with untreated plots. Mowing did not always enhance the efficacy of foliar glyphosate applications. Triclopyr, with or without mowing, was generally not effective against V. nigrum in our study. The only significant effect of triclopyr was to increase the cover of grasses in the plots. While annual applications of glyphosate can be useful for management of V. nigrum infestations, higher rates and more frequent applications of triclopyr need to be investigated to determine its usefulness for V. nigrum control.

ECOLOGICAL AND TOXICOLOGICAL ASSESSMENT OF THE HERBICIDE EXPOSURE DIRECTOR, W.S. (A.I. ISOPROPYLAMINE SOLT GLYPHOSATE, 410 G/DM3) ON BRANCHED CRUSTACEANS CERIODAPHNIA AFFINIS LILLJEBORG

Analysis of the chronic experiments results showed that the herbicide Director (active ingredient - isopropylamine salt of glyphosate, 41%) is a substance toxic to Ceriodaphnia affinis. This herbicide is characterized by embryotoxicity. Herbicide Director has the greatest toxic effect on C. affinis at a concentration of 50 mg/dm3. All major bioparameters of crustacean activity are suppressed. There is a high mortality of first-generation crustaceans (78%) planted in solutions with the herbicide, low life expectancy of females. The offspring born to females in a number of generations are unviable; reproduction of offspring is suspended mainly at the level of the second generation. Only one female from the second generation offspring on the 8th day gave 4 juveniles of the third generation of the 1st brood, which died a day later. At 50 mg/dm3 offspring reproduction is suspended at the level of the second generation. The productivity of females at the herbicide concentration of 25 mg/dm3 is reduced by 66.5% compared with the control. The productivity decrease at lower concentrations was in the range of 30.4 ÷ 17.6%. The smallest decrease in female productivity (by 9.2%) was observed at a concentration of 0.001 mg/dm3. At the herbicide concentration of 25 mg/dm3, the puberty of young C. affinis was delayed and was longer than in the control and at lower concentrations by 1.42 days. Females gave only 5 generations, while in the control and in the concentration range of 0.001–10 mg/dm3 for the same period there were 6 generations. The total number of offspring on average for 5 generations was 33.58% of control, with a reliability of> 99.9%. To a lesser extent, inhibition of crustacean activity was observed at lower herbicide concentrations. At 10 mg/dm3, the sexual maturity of crustaceans was delayed only in the first generation, and the total number of offspring on average for 6 generations was 72.1% of control. At the same level, a decrease in total productivity was observed at concentrations of 1.0 and 0.1 mg/dm3.

Pamuk ekim alanlarında Ipomoea triloba L. (pembe çiçekli akşam sefası)’nın mücadele olanaklarının belirlenmesi

Cotton (Gossypium hirsutum L.) is used as the raw material in more than fifty industries and is also the main source of raw materials used in the textile, the oil, the animal feed, and the paper industries. Additionally, it is a crop of great economic importance for its producer countries with the added value and employment opportunities. Cotton is among the crops sensitive to weed competition and its yield decreases with the weeds. For this reason, weed control treatments should be employed effectively to obtain high yields in cotton fields. In the recent years, Ipomoea triloba (IPOTR), which has increased in its importance in the agricultural areas of the Mediterranean Region, prevents the development of the cotton because of its invasive features and causes the harvest difficulties during by hand or machine harvest. The aim of this study is to create an effective control program against IPOTR in cotton. Field experiments were carried out in Ceyhan, Adana, Turkey in 2018 and 2019. For this purpose, impact of the treatments including Pyroxasulfone 85% (PYRS) Trifloxysulfuron sodium 75% (TRFS), Pyrithiobac-sodium 383 g/l (PYBS), Glyphosate isopropylamine salt 480 g/l (GLYI), S-metolachlor 915 g/l + Benoxacor 45 g/l + hand hoe (SMEC), inter-row rotary hoe + intra-row hand hoe (FÇEÇ) were investigated 28 day-after treatment and at the harvest during two years. It has been determined that TRFS and PYBS have an efficacy over 90%, while FÇEÇ, SMEÇ, TRFS and PYBS applications have an efficacy over 60%. Applications against IPOTR have increased the cotton lint and yield. However, crop injury was observed after GLYI application (20%).

Glyphosate residues in grain after desiccation of crops in the Ob Region

The residual amounts of glyphosate in wheat grain after desiccation of crops were determined. Glyphosate is the most widely used herbicide in the world. The level of its ecotoxicity is widely discussed in the scientific literature after the compound was classified as “a likely carcinogenic” pesticide. It is assumed that glyphosate residues can be highest when the crops are desiccated before harvesting. The studies were carried out in 2018 (central forest-steppe of the Novosibirsk Ob region (54°53'13.5"N, 82°59'36.7"E). In the experiment, desiccation of wheat was carried out with the herbicide Zero Super (the content of isopropylamine salt of glyphosate was 750 g/kg) at the recommended dose of 1.5- 2.0 kg/ha. Glyphosate residues in plant biomass were determined using the test system, based on the principle of enzyme immunoassay. Pre-preparation of the samples for the analysis was carried out according to the recommendation of Stylab laboratory, which validated the method for the determination of glyphosate in grain. Glyphosate residues were found in all of the 37 samples tested. Regardless of the sampling period after desiccation, herbicide residues in the grain did not exceed 4.4 mg/kg. In 14 days after desiccation, the minimum pesticide level was lower (0.5 mg/kg) compared to the data obtained in 7 days (2.6 mg/kg). The content of the herbicide residues in wheat straw was higher than in grain. The highest values of glyphosate residues were found in the dry biomass of weed vegetation (up to 9 mg/kg). In the grain stored for 1 year, the content of GR has not decreased. The data obtained were compared with the MRL for the residual amount of glyphosate in wheat grain accepted in the world.

Mixtures between glyphosate formulations and ACCase-inhibiting herbicides in the control of Chloris elata

Chloris elata is an important weed for grain and sugarcane crops. In addition to its aggressiveness, it may show resistance to glyphosate herbicide. In this context, the aim of this study was to assess the effectiveness of glyphosate formulations, isolated or in association with ACCase inhibitors, in controlling C. elata (putatively resistant to glyphosate) at different developmental stages. Four experiments were conducted in a completely randomized design. Treatments consisted of glyphosate application under different formulations with ACCase inhibitors, isolated or in mixtures. Applications were carried out at the stages of four fully expanded leaves in Experiment I, four tillers in Experiment II, and at regrowth of the four-tiller plants in Experiments III and IV. Applications of glyphosate isopropylamine salt associated with sethoxydim or clethodim showed to be among the best treatments in Experiments I, II, and III, presenting control scores equal to or greater than 90%. However, not even these treatments could provide successful control in Experiment IV. Moreover, and regardless of the formulation, isolated glyphosate, showed 85% (Experiment III) and 50% (Experiment IV) maximum controls. Associations between glyphosate and ACCase-inhibiting herbicides showed to be effective in controlling C. elata, especially at early developmental stages. In general, isolated herbicides provided lower percentages of control, as well as higher values of dry matter. Sole herbicide applications were not effective in controlling C. elata (putatively resistant to glyphosate), regardless of the developmental stage. Highlights: Mixtures between glyphosate and ACCase-inhibiting have been shown to be effective in C. elata control, especially in the early development stages. Isolated herbicide applications were not effective in controlling C. elata, regardless of the development stage. Mixtures between glyphosate and ACCase-inhibiting is more important in the control than the glyphosate formulation or ACCase-inhibiting used.

Oxidative carbonilation of liver tissue proteins under the influence of pesticide based on glyphosate in a subchronic experiment

Introduction. Currently, the pathogenetic mechanisms of the action on the body of widely used glyphosate-based herbicides and the search for the markers of the health status that changes under the influence of these herbicides remain open for study and discussion. Carbonyl derivatives of proteins can be used as the markers of oxidative stress caused by the herbicidal composition of the isopropylamine salt of glyphosate. The purpose of this study was to assess the severity of oxidative carbonylation of liver tissue proteins under the influence of a glyphosate-based pesticide in a subchronic experiment. Material and methods. The material for the study was subcellular fractions of liver tissue homogenates obtained from 90 Wistar rats, which were administered solutions of glyphosate isopropylamine salt at doses of 280 mg/kg and 100 mg/kg per os for 3 months. The severity of oxidative carbonylation of proteins was determined by the method of R.L. Levine modified by E.E. Dubinina. Results. Statistically significant changes in the content of products of oxidative damage to proteins at various stages of the experiment in the study groups were obtained compared with the control group due to a significant increase in the products of oxidative damage to proteins in subcellular fractions of liver tissue. Conclusion. The changes in carbonyl stress indices and the decrease in the reserve-adaptive potential indicate the depletion of antioxidant protection in the liver cells. A statistically significant increase in secondary markers of carbonyl stress after 1 month from the start of seeding of animals compared with the control with a slight increase in this index after 3 months may indicate the launch of adaptation mechanisms, including the induction of proteolytic utilization of oxidized proteins or additional protein synthesis.

Polymer additives regulate the deposition behavior of pesticide droplets on target plants

Pesticide formulation is usually combined with spraying additives to enhance the spraying effect in the actual field spraying. Polymer additives are new polymer materials developed in recent years and widely used in agriculture field. In this article, four different degrees of polymerization polymer additives (G101, G103, G100A and G200A) regulate the spray droplets of glyphosate herbicide on the target crop foliar deposition performance was studied. Through the study of equilibrium and dynamic surface tension, the dynamic contact angle of the droplet on the hydrophilic and hydrophobic leaves and the droplet impacting the target leaf surface at a high speed in a simulated field spraying process, the results showed that although glyphosate isopropylamine salt (Gly-IPA) with polymer additives can wet the target leaf surface, but only Gly-IPA adding G200A can deposited on the leaf surface during dynamic impact. This work opens up new prospects and provides a series of good evaluation methods for the study of polymer additives in regulating the deposition behavior of pesticide droplets on the target interface.

Pesticide selectivity to the parasitoid Trichogramma pretiosum: A pattern 10-year database and its implications for Integrated Pest Management

Trichogramma pretiosum is one of the main egg parasitoids used in the control of lepidopteran pests in Brazil. This natural enemy can be negatively affected by the use of insecticides, herbicides, and fungicides. The present work used a systematic review and meta-analysis to group information from multiple studies on the selectivity of pesticides (279 commercial products) in rice, corn, soybean, apple and peach crops for immature stages (egg-larva, pre-pupa, and pupa) and adult parasitoids. The selected studies used the International Organization for Biological and Integrated Control (IOBC) methodology with the same adaptations for T. pretiosum. The meta-analysis found that corn crops had the highest frequency of tests (2 0 7). The most frequently tested active ingredients (a.i.) were glyphosate, glyphosate isopropylamine salt, and sulfur at frequencies of 41, 32 and 24 tests, respectively. The pesticides registered for rice crops showed the greatest sublethal effects on T. pretiosum, with an approximately 47% reduction in parasitism (RP) or emergence (RE). The adult stage of the parasitoid showed greater sensitivity to the tested pesticides (65% RP), in comparison to the immature stages. In general, insecticides showed superior toxicity for all development stages of T. pretiosum, compared to herbicides and fungicides, regardless of the recommended dosage for the crop. The present study aggregates information related to selectivity for the four life stages of T. pretiosum, contributing significantly to the integration of biological control and chemical control in rice, corn, soybean, apple and peach crops in Brazil.

Long-Term Control of Hedge Bindweed (Calystegia sepium L.) with Single, Tank Mixture, and Sequential Applications of Glyphosate, 2,4-D, and Dicamba

Hedge bindweed (Calystegia sepium L.) is a widespread troublesome perennial weed species that has strong rhizome regenerative capacity. Four pot trials with randomised, complete block designs were conducted in 2015 to evaluate long-term control of hedge bindweed using individual, tank mixture, and sequential applications of selected herbicides. Two different formulations of N-(phosphonomethyl) glycine (glyphosate; isopropylamine, trimesium salts) were applied at 2000 g active ingredient (a.i.) ha−1. Additionally, two synthetic auxins were applied as 3,6-dichloro-2-methoxybenzoic acid (dicamba) at 500 g a.i. ha−1 and the dimethylamine salt of (2,4 dichlorophenoxy)acetic acid (2,4-D) at 1000 g a.i. ha−1. Tank mixtures and sequential applications (12/24 h separation) of these different herbicides were also included. Long-term control of hedge bindweed, Calystegia sepium L., growth was evaluated 8 months after treatments, as comparisons of shoot and rhizome growth (biomass) between untreated and treated plants. There were no differences between the two formulations of glyphosate alone, with shoot and rhizome biomass reductions of 83% and 42%, respectively. Dicamba alone inhibited shoot and rhizome biomass by 86% and 67%, respectively. By itself, 2,4-D provided the greatest reductions in shoot and rhizome biomasses, 93% and 79%, respectively. Antagonism was seen in the tank mixtures of glyphosate and dicamba or 2,4-D. Tank mixtures were generally comparable to treatments of glyphosate alone, and were less effective compared to dicamba or 2,4-D alone. The greatest reduction of bindweed rhizome biomass was for sequential glyphosate trimesium salt followed by 2,4-D 12 h later, thus showing significantly greater efficacy over glyphosate isopropylamine salt (94% vs. 84%; p ≤ 0.05). These data for reductions of the growth of the rhizome biomass show that the sequential application of glyphosate followed by 2,4-D significantly improves long-term control of hedge bindweed.

Adjuvant contributes Roundup's unexpected effects on A549 cells

Roundup® (RDP) is one of the most representative glyphosate-based herbicides (GBHs), which extensive use increases pressure on environmental safety and potential human health risk. The aim of this study was to investigate whether the adjuvant polyethoxylated tallow amine (POEA) or the herbicidal active ingredient glyphosate isopropylamine salt (GP) in formulation confers RDP cytotoxicity. We demonstrated that RDP and POEA could inhibit the proliferation of human lung A549 cells. Intracellular biochemical assay indicated that collapse of mitochondrial membrane, release of cytochrome c into cytosol, activation of caspase-9/-3, cleavage of poly (ADP-ribose) polymerase (PARP), oxidative DNA damage, DNA single-strand breaks and double-strand breaks are occurred in RDP and POEA treated A549 cells, not occurred in GP treated A549 cells. We conclude that the RDP's effect of apoptosis and DNA damage on human A549 cells is related to the presence of adjuvant POEA in formulation, independent of the herbicidal active ingredient GP. This study would enrich the theoretical basis of the RDP toxicity effects and attract attention on potential human health and environmental safety threat caused by adjuvant.

Determination of acute oral toxicity of glyphosate isopropylamine salt in rats.

In this study, we aim to determine the median lethal dose (LD50) of glyphosate isopropylamine salt (GI), which is commonly used in the world and especially in Turkey against to weeds, in male and female rats by using the probit or logit analysis method. A total of 140 Wistar rats were used, including 70 females and 70 males. To determine LD50, the male and female rats were randomized into 7 groups made up of 10 animals in each group. At doses of 6000, 6500, 7000, 7500, 8000, 8500, and 9000mg / kg, GI was administered to the male and female rats by oral gavage. After dosing, the animals were periodically monitored for 14days. No deaths were observed after 48h of herbicide application. In this study, only logit analysis was used for the LD50 value to be calculated in the male rats within 24h, while other analyses were carried out with the probit method. In the female and male rats, the LD50 levels of GI between 24 and 48h were determined as 7444.26-7878.50mg/kg and 7203.58-7397.25mg/kg, respectively. According to these results, it was concluded that female rats are more sensitive to GI than male rats. We believe that the findings that were obtained will guide researchers, clinicians, and toxicologists through preventive and curative studies against acute poisoning that may occur with GI.

Drift Distance in Aircraft Glyphosate Application Using Rice Plants as Indicators

ABSTRACT: This study aimed to evaluate the potential for glyphosate drift during aerial application using rice plants as sentinels, aiming to determine the effect of drift on irrigated rice crops. For this purpose, a field experiment was performed using an entirely randomized design with four replicates, evaluating different distances from the site of application [control (no application), 0, 12.5, 25, 50, 75, 100, 150, 200, 300, and 400 m]. The experiment was carried out at the Granjas 4 Irmãos farm, located in the Rio Grande city, Rio Grande do Sul State, Brazil. The glyphosate dose application was 1,920 g e.a. ha-1 (Roundup Transorb®, 480 g e.a. L-1 glyphosate isopropylamine salt). A dose-response curve was developed to estimate the drift rate in sentinel plants, by applying increasing glyphosate doses in rice plants and assessing the injury level. The drift rates estimated by the injury level in sentinel plants were 14% (150 m), 13% (200 m), and 5% (400 m). Death of the experimental units was observed for distances between 0 and 50 m, while in distances between 75 and 150 m, 25 to 50% of the plants survived, reducing productivity. In the distances between 200 and 400 m, there was no reduction in productivity when compared to the control, even when the injury levels reached 52 to 82%. Thus, we concluded that a 5% glyphosate drift reached up to 400 m from the application range. Considering the recommendation of zero drift, distances greater than 400 m should be adopted to avoid symptoms in rice plants. We suggest using distances of more than 400 m in future studies.

Synergistic effects of adjuvant A-134 on the herbicidal effects of glyphosate.

Undesirable side effects on ecosystems and strong selection for weed resistance demand an increase in the efficacy and a reduction in the dosage of glyphosate herbicide used. The synergistic effect of tank-mixed adjuvant KAO® A-134 (A-134) on the post-emergence activity of the commercial glyphosate formulation Roundup® (RDP) against crabgrass (Digitaria sanguinalis) was detected. Field study also showed that A-134 can increase the herbicidal effect of RDP. Meanwhile, A-134 concentration-dependently decreased the surface tension and increased the spreading area of RDP, causing faster penetration and improved uptake of glyphosate into crabgrass. Moreover, the tank mix with A-134 also increased the adhesion of spray droplets of glyphosate isopropylamine salt (GP) to the leaf surface after rainfall treatment, thus maintaining its herbicidal effect. Data suggested the necessity of using these synergistic properties of A-134 to reduce environmental exposure and glyphosate resistance selection.

THE SEVERITY OF ENDOGENOUS INTOXICATION AND OXIDATIVE STRESS IN THE BLOOD OF WORKERS IN CONTACT WITH GLYCINE DERIVATIVES

Introduction. Currently, the search for markers of the health status of workers in agro-industrial complexes in contact with pesticides continues. The aim of this study was the assessment of the effect of glyphosate isopropylamine salt on the health of workers in agroindustrial complexes of the Ryazan region according to the severity of endogenous intoxication and oxidative stress in the body. Materials and Methods. Plasma and erythrocytes of peripheral blood of workers were the clinical material for the study. Blood was taken in 87 workers contacted with glyphosate-containing pesticides. As a control group, 20 clinically healthy individuals of the same age category who did not have contact with the pesticides were selected to be studied. The severity of endogenous intoxication was determined by the level of substances of low and medium molecular weight in the plasma and red blood cells. The severity of oxidative stress was determined by the content of the products of oxidative modification of proteins by the method of R.L. Levine modified by E.E. Dubinina. Results. Statistically significant changes in the severity of endogenous intoxication in the blood plasma and erythrocytes of the study groups and the control group of individuals among machine operators and workers who come into contact with pesticides to a lesser extent were obtained. A statistically significant increase in the level of carbonyl derivatives compared with the control among machine operators who have greater contact with glyphosate-containing pesticides according to the time sheets has been revealed. The level of secondary carbonyl derivatives in machine operators was higher than that of warehouse workers and was statistically significant. Conclusion. A statistically significant increase in the level of secondary carbonyl derivatives among machine operators as compared with other workers is a late marker of oxidative stress, which indicates to a depletion of the reserve forces of the body. This may be due to longer contact with pesticides based on glyphosate machine operators as compared to other workers in the agro-industrial complexes.

MODEL STUDY TO INVESTIGATE THE TOXIC INTERACTION BETWEEN GLYPHOSATE AND COPPER SULPHATE ON CHICKEN EMBRYOS

The toxic effects of the Taifun Forte herbicide (360 g/l glyphosate isopropylamine salt) applied alone or in combination with copper sulphate were studied on chicken embryos in the early phase of embryonic development. The test materials were injected in 0.1 ml volume into the air chamber of eggs on the first day of incubation. Subsequently, on the third day of incubation permanent preparations were made from the embryo in order to study the early developmental stage. Embryos fixed on slides and stained with osmium tetroxide solution were studied under light microscope. The embryonic mortality and the developmental anomalies was analysed statistically by Fisher test. According to the result of the statistical evaluation, the embryonic mortality was not influenced by the single treatment of copper sulphate. However, Taifun Forte and its combination with heavy metal significantly increased the early embryonic mortality. Developmental abnormalities were sporadically observed due to the single administration of copper sulphate. The incidence of it was increased due to the treatment with herbicide alone and in combination with copper sulphate. Based on the results, additive toxic interaction may occur between the copper sulphate and glyphosate that can highly reduce the viability of the embryos or can lead to extinction of wild birds in serious cases.

Roundup-Induced AMPK/mTOR-Mediated Autophagy in Human A549 Cells.

The extensive use of pesticide caused an amount of pressure on the environment and increased the potential human health risk. Glyphosate-based herbicide (GBH) is one of the most widely used pesticides based on a 5-enolpyruvylshikimate-3-phosphate synthase target, which does not exist in vertebrates. Here, we study autophagic effects of the most famous commercial GBH Roundup (RDP) on human A549 cells in vitro. Intracellular biochemical assay indicated opening of mitochondrial permeability transition pore, LC3-II conversion, up-regulation of beclin-1, down-regulation of p62, and the changes in the phosphorylation of AMPK and mTOR induced by RDP in A549 cells. Further experimental results indicated that all the effects induced by RDP were related to its adjuvant polyethoxylated tallow amine, not its herbicidal active ingredient glyphosate isopropylamine salt. All these results showed that RDP has the ability to induce AMPK/mTOR-mediated cell autophagy in human A549 cells. This study would provide a theoretical basis for understanding RDP's autophagic effects on human A549 cells and attract attention on the potential human health risks induced by the adjuvant.

Evaluation of the cytotoxic effects of glyphosate herbicides in human liver, lung, and nerve

Glyphosate-based herbicides are broad-spectrum pesticides widely used in the world, which is considered a highly safe pesticide due to their target specificity, but recently, there has been an ongoing controversy regarding their carcinogenicity and possible side effects of glyphosate on human health. Commercial glyphosate-based herbicides (GBHs) consist of declared active ingredient (glyphosate salts) and a number of formulants such as ethoxylated formulants (4130®, 3780®, and A-178®). The aim of our study is to investigate whether the toxicity of GBHs is related to formulants. The effects of GBHs on human health were studied at the cellular level based on their toxicity to liver, lungs and nerve tissue. The inhibitory toxicity to cell viability by GBHs was examined with cell-based systems using three human cell lines: HepG2, A549, and SH-SY5Y. Data obtained showed that all tested ethoxylated formulants and their mixtures with declared active ingredient glyphosate isopropylamine salt (GP) have significant inhibitory effect on cell proliferation, while the declared active ingredient has no significant toxicity. Our study demonstrates that the toxic effect of GBH is primarily due to the use of formulants. This result suggests that GP is relatively safe and a new approach for the assessment of toxicity should be made.

Effective dose of ionic liquids with glyphosate

Summary The effective dose of six herbicidal ionic liquids containing glyphosate [N-(phosphonomethyl)glycine] was investigated. Varied biological activity of the tested compounds was observed depending on the type of cation and targeted plant species. In the case of common lambsquarters, the lowest effective dose was obtained for compounds containing didecyldimethylammonium and di(hydrogenated tallow)dimethylammonium cations. In the case of white mustard, the lowest ED50 and ED90 values were obtained for the reference compound, which contained glyphosate isopropylamine salt. These parameters were determined using dose efficiency curves based on log-logistic models with three or four parameters. The study indicates that ionic liquids with glyphosate may be used as a new form of this herbicide in the future.
