Antioxidants Characterization of the Fruit, Juice, and Pomace of Sweet Rowanberry (Sorbus aucuparia L.) Cultivated in Estonia.

This study aimed to identify promising candidates of rowanberry cultivars for a wider cultivation and utilization. Antioxidant properties and phenolic content were evaluated for fruit, juice, and pomace samples of 16 different sweet rowanberry cultivars (cvs) and wild rowanberry (S. aucuparia L.), while the antioxidant potential was assessed using three different methods, based on the capacity to scavenge ABTS●+ and DPPH● and measure the oxygen radical absorbance capacity (ORAC). In general, the radical scavenging capacity was higher for hybrid cultivars, e.g., for cvs Likernaja, Burka, Granatnaja, and Rubinovaja in all assays. The highest value in the ABTS●+ assay was determined for the fruit sample Likernaja, and in DPPH● assay in the pomace sample of cv. Likernaja, at 527.55 and 1068.28 µM TE/g dw, respectively. The highest ORAC value was found in the fruit sample of Burka (456.53 µM TE/g dw). Among the Nevezhino rowans, the highest radical scavenging values of all fractions were determined in cv. Solnechnaja. Regarding the total phenolic content (TPC), higher values were obtained in the whole fruits than in separated fractions, juice, and pomace. The tested hybrids had higher TPC values, either in fruit and pomace or in juice extracts, than those in the other analyzed S. aucuparia L. cultivars. While the fruit and juice samples showed higher anthocyanin (ACY) values, the pomace samples had higher hydroxycinnamic acid (HCA) contents on average. The results revealed that the different fractions of selected rowanberry cultivars can be a promising source of antioxidants and polyphenols for further potential applications. It is envisaged that the results of this study will serve in valorizing sweet rowanberry cultivars as value-added functional ingredients for food and non-food applications.

The effect of chocolate drink to hepatosomatic index of diabetes mellitus induced rat

Chocolate (Theobroma cacao) is one of the foods that contain a high antioxidant level. The effect of polyphenol compounds in cocoa beans and the chocolate products is the potential to be developed into healthy chocolate drink products. The cocoa powder has a high ORAC (Oxygen Radical Absorbance Capacity) value (80,933). This indicates that the cocoa powder has the ability to prevent the free radicals harm, including contributing to the heart organ repairing. The study was conducted using an experimental research method with a randomized design. Twenty-five male white rats (Rattus norvegicus) Sprangue Dawley strain were divided into five treatment groups. Negative control (P-) consists of normal rats without chocolate drink treatment. Positive control (P+) consists of diabetes mellitus rats without chocolate drink treatment. Treatment 1 (P1) consists of diabetes mellitus rats treated with 2% of chocolate drink. Treatment 2 (P2) consist of diabetic rats treated with 4 % chocolate drink. Treatment 3 (P3) consists of diabetes mellitus rats treated with 6% chocolate drink. The intervention was carried for 14 days then a wet weight of liver organ was measured. The hepatosomatic index (HSI) level then analyze to show the effect. The Hepatosomatic index value are: P- = 3,79 ± 0,40; P+ = 4,66 ± 0,21; P1 = 4,41 ± 0,30 P2 = 4,45 ± 0,35 and P3 = 4,28 ± 0,58. The ANOVA test showed that there was no significant difference in the hepatosomatic index value in each treatment group (p = 0.15).

Antioxidant Potential of Mung Bean (Vigna radiata) Albumin Peptides Produced by Enzymatic Hydrolysis Analyzed by Biochemical and In Silico Methods.

The objective of this study was to investigate the biochemical antioxidant potential of peptides derived from enzymatically hydrolyzed mung bean (Vigna radiata) albumins using an 2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assay, a ferrous ion chelating assay and an oxygen radical absorbance capacity (ORAC) assay. Peeled raw mung bean was ground into flour and mixed with buffer (pH 8.3, 1:20 w/v ratio) before being stirred, then filtered using 3 kDa and 30 kDa molecular weight cut-off (MWCO) centrifugal filters to obtain albumin fraction. The albumin fraction then underwent enzymatic hydrolysis using either gastrointestinal enzymes (pepsin and pancreatin) or thermolysin. Peptides in the hydrolysates were sequenced. The peptides showed low ABTS radical-scavenging activity (90–100 μg ascorbic acid equivalent/mL) but high ferrous ion chelating activity (1400–1500 μg EDTA equivalent/mL) and ORAC values (>120 μM Trolox equivalent). The ferrous ion chelating activity was enzyme- and hydrolysis time-dependent. For thermolysin hydrolysis, there was a drastic increase in ferrous ion chelating activity from t = 0 (886.9 μg EDTA equivalent/mL) to t = 5 min (1559.1 μg EDTA equivalent/mL) before plateauing. For pepsin–pancreatin hydrolysis, there was a drastic decrease from t = 0 (878.3 μg EDTA equivalent/mL) to t = 15 (138.0 μg EDTA equivalent/mL) after pepsin was added, but this increased from t = 0 (131.1 μg EDTA equivalent/mL) to t = 15 (1439.2 μg EDTA equivalent/mL) after pancreatin was added. There was no significant change in ABTS radical scavenging activity or ORAC values throughout different hydrolysis times for either the thermolysin or pepsin–pancreatin hydrolysis. Overall, mung bean hydrolysates produced peptides with high potential antioxidant capacity, being particularly effective ferrous ion chelators. Other antioxidant assays that use cellular lines should be performed to measure antioxidant capacity before animal and human studies.

α-Glucosidase and non-enzymatic glycation inhibitory potential of Eugenia dysenterica fruit pulp extracts

Considering the nutritional benefits of fruits on human health, this study aimed to screen 11 fruits found in the Brazilian Savanna for glycoside hydrolases inhibitors with antioxidant and anti-glycation properties. Aqueous and ethanol extracts of pulps were used in enzymatic assays to evaluate the inhibitory potential against α-amylase and α-glucosidase, as well as antioxidant (2,2-diphenyl-1-picrylhydrazyl (DPPH) and oxygen radical absorbance capacity (ORAC) assays) and anti-glycation (BSA-fructose model) capacities. Ethanol extracts of Campomanesia xanthocarpa, Byrsonima verbascifolia and Eugenia dysenterica had the lowest DPPH IC50 values (<0.25 mg/ml), while ethanol extracts of Genipa americana and B. verbascifolia had the highest ORAC values (>0.60 mmol trolox equivalents/g). The extracts of E. dysenterica showed potential to inhibit glycation (IC50 < 0.40 mg/ml) and α-glucosidase (IC50 < 20 μg/ml). Liquid-liquid fractionation of the extracts of E. dysenterica fruit pulp resulted in the ethyl acetate fraction showing higher antioxidant (DPPH IC50 10 μg/ml and ORAC 1.3 mmol trolox equivalents/g) and anti-glycation (IC50 0.10 mg/ml) capacities, and α-glucosidase inhibition activity (IC50 11 μg/ml). HPLC-ESI-MS/MS analysis suggested the presence of antioxidant molecules in E. dysenterica fruit pulp, such as ferulic and gallic acids, myricetin, quercetin and kaempferol-pentosides. Non-conventional fruits found in the Brazilian Savanna, a global biodiversity hotspot, with antioxidant and anti-glycation properties, could be potential sources of nutraceuticals to combat oxidative stress, which is present in many pathologies. In addition, the capacity of E. dysenterica to inhibit α-glucosidase and non-enzymatic glycation makes it especially useful for in vivo studies related to diabetes mellitus.

Evaluations of the Peroxidative Susceptibilities of Cod Liver Oils by a 1H NMR Analysis Strategy: Peroxidative Resistivity of a Natural Collagenous and Biogenic Amine-Rich Fermented Product.

High-resolution 1H nuclear magnetic resonance (NMR) analysis was employed to molecularly screen the lipid, lipid oxidation product (LOP), and antioxidant compositions of four natural (unrefined) cod liver oil (CLO) products. Products 1–3 were non-fermented CLOs, whilst Product 4 was isolated from pre-fermented cod livers. Supporting analytical data that were acquired included biogenic amine, flavanone, tannin, phenolic antioxidant, α-tocopherol, and oxygen radical absorbance capacity (ORAC) determinations by recommended HPLC, LC/MS/MS, or spectrophotometric methods. SDS-PAGE, HPLC, and 1H NMR analyses investigated and determined collagenous antioxidants and their molecular mass ranges. 1H NMR analysis of aldehydic LOPs was employed to explore the susceptibilities/resistivities of each CLO product to peroxidation that is induced by thermal stressing episodes (TSEs) at 180°C, or following prolonged (42 day) storage episodes at 4 and 23 °C. Product 4 displayed extremely high ORAC values, which were much greater than those of Products 1–3, and that were predominantly ascribable to significant levels of peroxidation-blocking and/or aldehyde-consuming collagenous polypeptides/peptides and ammoniacal agents therein. Significantly lower levels of toxic aldehydes were generated in the pre-fermented Product 4 during exposure to TSEs, or the above long-term storage episodes. These results confirmed the enhanced peroxidative resistivity of a fermented, antioxidant-fortified natural CLO product over those of non-fermented unrefined products. Water Extract of Cryphaea heteromalla (Hedw.) D. Mohr Bryophyte as a Natural Powerful Source of Biologically Active Compounds.

Bryophytes comprise of the mosses, liverworts, and hornworts. Cryphaea heteromalla, (Hedw.) D. Mohr, is a non-vascular lower plant belonging to mosses group. To the date, the most chemically characterized species belong to the liverworts, while only 3.2% and 8.8% of the species belonging to the mosses and hornworts, respectively, have been investigated. In this work, we present Folin–Ciocalteu and oxygen radical absorbance capacity (ORAC) data related to crude extracts of C. heteromalla obtained by three different extraction solvents: pure water (WT), methanol:water (80:20 v/v) (MET), and ethanol:water (80:20 v/v) (ETH). The water extract proved to be the best solvent showing the highest content of biophenols and the highest ORAC value. The C. heteromalla-WT extract was investigated by HPLC-TOF/MS (High Performance Liquid Chromatography-Time of Flight/Mass Spectrometry) allowing for the detection of 14 compounds, five of which were phenolic compounds, derivatives of benzoic, caffeic, and coumaric acids. Moreover, the C. heteromalla WT extract showed a protective effect against reactive oxygen species (ROS) generation induced by tert-butyl hydroperoxide (TBH) on the murine NIH-3T3 fibroblast cell line.

Anti-aging effects of black raspberry extract on cataract, alopecia, skin whitening, and weight loss

Background: To maintain good health, it is important to eliminate extra reactive oxygen generated in the body. Furthermore, ingesting foods containing antioxidants is beneficial. The oxygen radical absorbance capacity (ORAC) values for black raspberry extract (BRE), blueberry extract (BBE), and raspberry extract (RBE) are 62, 66, and 49 µM Trolox equivalents (TE)/g respectively. These values are higher than those for typical antioxidant foods that have been discovered so far (3–30 µM TE/g). Our aim was to find new functionality from the food with the high ORAC value. Therefore, we have prepared these four berry extracts and examined whether they have anti-aging effects and if those effects correlate with their antioxidant activities.Methods: We studied the following effects of 4 berry extracts: 1) lens cell protective effects; 2) effects against alopecia; 3) induction of uncoupling protein-1 (UCP1), a regulator of fat and energy consumption in adipocytes, and stimulation of irisin secretion from skeletal muscle cells; and 4) inhibitory effects on melanocyte tyrosinase activity. The evaluation method was based on below; 1) a-crystallin, type 17 collagen, heat shock protein 47 (HSP47), UCP1 and Irisin - mRNA by qRT-PCR, 2) the amount of the UCP1 and Irisin protein by ELISA. 3) Inhibition of tyrosinase activity was measured by dopachrome production using L-tyrosine.Results: In lens cells, a-crystallin mRNA expression was induced 1 hour after treatment of the cell with Blabina (a powdered formulation containing BRE) and BRE. The extracts of all four berry species promoted the growth of follicle dermal papilla cells by 3-20% in a concentration-dependent manner. These berry extracts were also discovered to markedly induce the expression of mRNAs of type 17 collagen and HSP47 in the hair follicle stem cell and elevate the expression levels of UCP1 mRNA and its protein in adipocytes in a concentration-dependent manner. BRE and Blabina inhibited 5a-reductase in follicle dermal papilla cells and tyrosinase activity in melanocytes at the concentrations which inhibited dopachrome production by at least 50%. Finally, Blabina was discovered to stimulate the irisin secretion from skeletal muscles.Conclusion: These results suggest that berry extracts, particularly BRE, have anti-aging effects through their higher antioxidant activities.Keywords: Anti-aging; antioxidant; alopecia; black raspberry; weight loss; oxygen radical, absorbance capacity; skin whitening

Antioxidant Activity of Oat Proteins Derived Peptides in Stressed Hepatic HepG2 Cells.

The purpose of this study was to determine, for the first time, antioxidant activities of seven peptides (P1–P7) derived from hydrolysis of oat proteins in a cellular model. In the oxygen radical absorbance capacity (ORAC) assay, it was found that P2 had the highest radical scavenging activity (0.67 ± 0.02 µM Trolox equivalent (TE)/µM peptide) followed by P5, P3, P6, P4, P1, and P7 whose activities were between 0.14–0.61 µM TE/µM). In the hepatic HepG2 cells, none of the peptides was cytotoxic at 20–300 µM. In addition to having the highest ORAC value, P2 was also the most protective (29% increase in cell viability) against 2,2′-azobis(2-methylpropionamidine) dihydrochloride -induced oxidative stress. P1, P6, and P7 protected at a lesser extent, with an 8%–21% increase viability of cells. The protection of cells was attributed to several factors including reduced production of intracellular reactive oxygen species, increased cellular glutathione, and increased activities of three main endogenous antioxidant enzymes.

Comparison of polyphenol, anthocyanin and antioxidant capacity in four varieties of Lonicera caerulea berry extracts

Four varieties of Lonicera caerulea berries—‘Wild’, ‘Beilei’, ‘No. 1’, and ‘No. 2’—were compared with respect to extraction yield, fruit weight, total soluble solids, polyphenol and anthocyanin contents, oxygen radical absorbance capacity (ORAC), and anthocyanin composition. Sixteen individual anthocyanins were identified in the selected varieties. Acylated anthocyanins, cyanidin 3-acetylhexoside and peonidin 3-acetylhexoside, were identified in L. caerulea berries for the first time. Cyanidin-3-glucoside was the most prominent anthocyanin in all four tested varieties. Wild type of L. caerulea fruit (‘Wild’), with the highest polyphenol content, contained 14 anthocyanins and the highest ORAC value. Eleven anthocyanins were found in ‘Beilei’ berries, which had a higher ORAC value than ‘No. 1’ and ‘No. 2’. The highest total soluble solid content and extraction yield were found in ‘No. 2’ and ‘Wild’ berries, respectively.

Free radical scavenging ability of Aspalathus linearis in two in vitro models of diabetes and cancer

The free radical scavenging activity of Aspalathus linearis (Rooibos tea) and its effect on reactive oxygen species (ROS), catalase (CAT), and superoxide dismutase (SOD) were investigated in two in vitro disease models of cancer and diabetes. Although the antioxidant activity of this tea has been reported in several studies, its effects in disease models of ROS-induced oxidative stress have not been systematically evaluated to date. The oxygen radical absorbance capacity (ORAC) assay was used in this study to quantify the antioxidant capacity of the extract, whereas the ROS scavenging ability in hyperglycemia-induced human umbilical vein endothelial cells (HUVECs) and HeLa cells were investigated. The CAT and SOD assays were also carried out in the two disease models in order to evaluate the effect of the extract in the stimulation of these two enzyme activities. The extract was observed to have reduced ROS in a dose-dependent manner in both HUVECs and HeLa cells. The stimulation of the CAT and SOD enzyme activities were observed to be dose-dependent as well. The high ORAC value of the extract indicated the presence of antioxidant compounds which could directly quench ROS, whereby this mechanism of action could be hypothesized to have been further complemented through the stimulation of CAT and SOD. Overall, the Aspalathus linearis extract was observed to have increased the CAT and SOD activities in two in vitro disease models of cancer and hyperglycemia. Given the correlation between the ORAC values, the increases in CAT and SOD activities and the reduction in ROS in a dose-dependent manner, it could be hypothesized that the extract had a significant therapeutic potential for either the prevention of the onset of the two diseases or their progression because ROS has been identified as their root causes.

동결건조 진주담치 추출물의 항산화 및 DNA 손상 보호 활성

동결건조에 의한 진주담치의 생리활성의 변화를 살펴보고자 DPPH 라디칼 소거능, ORAC, CAC 등의 항산화 활성과 comet assay를 이용한 DNA 손상 보호능을 측정하였다. 생 진주담치 및 동결건조 진주담치에서 물 추출물을 제외한 에탄올과 메탄올 추출물에서 DPPH 라디칼 소거능을 확인하였고, 메탄올 추출물의 경우 동결건조에 의해 DPPH 라디칼 소거능이 증가하는 것으로 나타났다. 생 진주담치의 ORAC 수치는 물 추출물에서 가장 높게 나타난 반면, 동결건조 진주담치의 경우 메탄올 추출물에서 가장 높게 나타났다. 동결건조 후 ORAC 수치는 물 추출물에서만 유의적으로 감소된 반면 HepG2 세포의 라디칼 소거능(CAC)의 경우 물 추출물에서 유의적으로 증가하여 소거 대상 라디칼에 따라 항산화활성의 차이가 있는 것으로 나타났다. 생 또는 동결건조 진주담치는 모든 추출물에서 산화적 스트레스에 의한 DNA 손상을 억제하는 보호 효과가 있음이 밝혀졌고 동결건조에 의한 보호 효과는 유의적인 변화가 없는 것으로 나타났다. 결론적으로 진주담치는 물 추출물의 DPPH 라디칼 소거능을 제외하고는 모든 추출물에서 항산화 활성과 더불어 DNA 손상의 보호 효과가 관찰되었고 동결건조 가공처리에 의해서 그 활성이 크게 영향을 받지 않거나 추출물에 따라 오히려 활성이 증가함을 알 수 있었다. 이러한 결과를 바탕으로 우리나라에서 생산되는 진주담치의 식품 첨가물이나 기능성 식품 개발을 위한 생리활성 소재로의 가능성을 확인할 수 있었다.

Phytochemical profiles and antioxidant activities in six species of ramie leaves.

Increased consumption of vegetables or plant food has been associated with decreased risk of developing major chronic diseases, such as cancers, diabetes, cardiovascular diseases, and age-related functional decline. Ramie leaves are rich in phenolics and flavonoids, which have been suggested for human health benefits. Phenolic contents, flavonoid contents, phenolic compounds, and anti-cancer properties in six species of ramie leaves were analyzed by Folin-reagent method, sodium borohydride/chloranil-based assay (SBC), HPLC method and antiproliferation, cytoxicity, respectively. Antioxidant activities were measured through peroxyl radical scavenging capacity (PSC) method, oxygen radical absorbance capacity (ORAC) method, and cellular antioxidant activity (CAA). Research indicated that Boehmeria penduliflora contained the highest total phenolic content (2313.7±27.28 mg GAE/100 g FW), and flavonoid content (1682.4±27.70 mg CAE/100 g FW). Boehmeria tricuspis showed the highest PSC value (9574.8±117.63 µM vit. C equiv./100 g FW), while Boehmeria penduliflora indicated the highest ORAC value (330.44±16.88 µmol Trolox equiv./g FW). The antioxidant activities were correlated with phenolic contents and flavonoid contents. Boehmeria tricuspis had the highest antiproliferative capacity with the lowest EC50 (4.11±0.19 mg/mL). The results for the analyzed ramie for CAA were significantly different from each other (p<0.05), Boehmeria tricuspis had the highest CAA value (133.63±7.10 µmol QE/100 g). Benzoic acid, 4-coumaric acid, caffeic acid, and ferulic acid were the dominant phenolic ingredients in the ramie leaves according to HPLC analysis. Our research is the first report to study the phytochemical profiles and antioxidant activities in different species of ramie leaves for their health benefit.

Effect of methanol extract of Dicranopteris linearis against carbon tetrachloride- induced acute liver injury in rats

BackgroundDicranopteris linearis (family Gleicheniaceae) has been reported to possess anti-inflammatory and antioxidant activities but no attempt has been made to study its hepatoprotective potential. The aim of the present study was to determine the hepatoprotective effect of methanol extracts of D. linearis (MEDL) against carbon tetrachloride (CCl4)-induced acute liver injury in rats.Methods6 groups (n = 6) of rats received oral test solutions: 10% dimethyl sulfoxide (DMSO), 200 mg/kg silymarin, or MEDL (50, 250, and 500 mg/kg), once daily for 7 consecutive days, followed by hepatotoxicity induction with CCl4. Blood and liver were collected for biochemical and microscopic analysis. The extract was also subjected to antioxidant studies (e.g. 2, 2-diphenyl-1-picrylhydrazyl (DPPH)- and superoxide anion-radical scavenging assays, oxygen radical absorbance capacity (ORAC) test and total phenolic content (TPC) determination), phytochemical screening and HPLC analysis.ResultsPretreatment with MEDL and silymarin significantly (P < 0.05) reduced the serum levels of AST, ALT and ALP, which were increased significantly (P < 0.05) in DMSO-pretreated group following treatment with CCl4. Histological analysis of liver tissues in groups pretreated with MEDL and silymarin showed mild necrosis and inflammation of the hepatocytes compared to the DMSO-pretreated group (negative control group). The MEDL showed higher DPPH- and superoxide anion-radical scavenging activity as well as high TPC and ORAC values indicating high antioxidant activity.ConclusionsMEDL exerts hepatoprotective activity that could be partly contributed by its antioxidant activity and high phenolic content, and hence demands further investigation.

Composition and Antioxidant Activity of Aronia Melanocarpa Fruit Juice

Aronia melanocarpa is a woody shrub of the Rosaceae family. Its fruits are used for consumption either fresh, or as juice, jam and wine. The aim of the present study was to measure the concentration of some biologically active substances and the antioxidant activity of two samples of Aronia melanocarpa fruit juice (AMFJ). The juice was prepared from fresh fruits. One of the samples was produced using a juice centrifuge and was preserved with potassium sorbate (1.0 g/l). The other sample was produced by crushing and squeezing of fruits after which the juice was filtered and sterilized at 100 °C for 10 min. The contents of biologically active substances in the juice samples were measured by spectrophotometric and high-performance liquid chromatography methods. The antioxidant activity of the two juice samples was measured by the ORAC (oxygen radical absorbance capacity) and HORAC (hydroxyl radical аverting capacity) methods. The results showed that both samples were extremely rich in polyphenolic substances, amongst which the highest was the concentration of procyanidins followed by phenolic acids (chlorogenic and neochlorogenic) and anthocyanins. Sample 1 had higher concentration of total phenolics and contained higher quantities of most polyphenolic substances with the exception of anthocyanins. It also contained micro-quantities of cyanogens. The variations in the composition of the two samples could be attributed to the different methods of juice production and conservation. Both juice samples showed very high ORAC and HORAC values which were higher for sample 1 (the sample with the higher concentration of polyphenolic substances). The high antioxidant activity of the two AMFJ samples could be attributed to their polyphenolic ingredients.

Evaluation of the Antioxidant Abilities of Water-Soluble Biosubstances Using the ORAC-ESR Assay in Combination with the Hydroxyl and Superoxide Radical-Eliminating Abilities

The oxygen radical absorbance capacity (ORAC) values of various water-soluble biosubstances using the ORAC-electron spin resonance (ESR) assay were examined, and an advanced database of water-soluble biosubstances in combination with the hydroxyl and the superoxide radical-eliminating abilities was prepared. The ORAC-ESR assay is based on the competitive reaction between the spin trapping of the 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH)-derived alkoxyl radical and its elimination by substrate. The alkoxyl radical-eliminating ability of substrate was calculated from the decrease in the ESR signal of the 5-(2,2-dimethyl-1,3-propoxy cyclophosphoryl)-5-methyl-1-pyrroline N-oxide (CYPMPO) spin adduct of the alkoxyl radical. The determination of ORAC values was performed by determining the specificity of a spin trap relative to a standard radical eliminator, 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox). The ORAC values [μmol Trolox equivalent (TE)/g] of water-soluble biosubstances were classified into three groups, the high value group (over 10,000 μmol TE/g), the intermediate value group (400–10,000 μmol TE/g), and the low value group (<100 μmol TE/g). The two-dimensional diagrams for the ORAC values (μmol TE/g) and the hydroxyl and superoxide radical-eliminating abilities (1/[\( {\text{id}}^{\text{h}}_{ 5 0} \)(mg/ml)] and 1/[\( {\text{id}}^{\text{s}}_{50} \)(mg/ml)], respectively) were depicted. Biosubstances having higher ORAC values had higher superoxide radical-eliminating abilities (the correlation factor 0.73). Little correlation between the ORAC values and the 1/[\( {\text{id}}^{\text{h}}_{ 5 0} \)(mg/ml)] values (the correlation factor 0.32) was observed. The obtained diagrams should provide a database for the study of the antioxidant abilities of foods and pharmaceutics.

A Comparative Study on the Antioxidant Activity of Commonly Used South Asian Herbs

The antioxidant activities of curry leaves, fenugreek seeds, Indian malabar leaves, red silk cotton tree leaves, cowitch leaves, holyfruit tree leaves, and black mustard seeds were compared. Their effects on reactive oxygen species (ROS) and superoxide dismutase (SOD) activity were investigated. The Oxygen Radical Absorbance Capacity (ORAC) assay determined the antioxidant potential of the extracts, while the ROS scavenging ability was explored in hyperglycemia-induced human umbilical vein endothelial cells (HUVECs). The SOD assay determined if the extracts stimulated the enzyme activity in the HUVECs. Curry leaf and fenugreek extracts had high ORAC values and superior free radical scavenging abilities compared with the rest of the extracts. The curry leaf extract had also increased the SOD activity. Fenugreek extract had not increased the SOD activity of the HUVECs. Thus, the two herbs displayed two distinct pathways of action for scavenging of ROS.

음료 농축액 및 퓨레의 항산화 활성

본 연구에서는 음료 농축액 및 퓨레로 사용되는 석류 농축액, 블루베리 농축액, 양파추출 농축액, 도라지추출 농축액, 흑마늘 농축액, 홍삼 농축액, 아사이베리 퓨레, 새싹유채 퓨레의 이화학적 특성, 총 페놀 함량, 항산화 효능(DPPH radical scavenging, ORAC) 및 아질산염 소거능을 분석하였다. pH는 석류 농축액이 2.81로 가장 낮았으며 양파추출 농축액이 5.12로 가장 높은 수치를 나타냈다. Brix는 홍삼 농축액이 70.30으로 가장 높았고 새싹유채 퓨레가 1.70으로 가장 낮았다. 총 페놀 함량은 아사이베리 퓨레에서 182.71 mg GAE/mL로 다른 시료에 비하여 높은 수치를 나타냈고 양파추출 농축액이 8.80 mg GAE/mL로 가장 낮았다. DPPH 라디칼 소거능 측정은 총 페놀 함량과 마찬가지로 아사이베리 퓨레에서 69.88%로 가장 높은 활성을 나타내었다. ORAC 지수는 블루베리 농축액에서 27,514 μMTE/mL로 가장 높게 나타났으며 아질산염 소거능도 아사이베리 퓨레에서 28.19%로 유의적으로 높은 활성을 보였다. 이상의 연구 결과로부터 천연물 유래 농축액 및 퓨레는 다양한 항산화 평가모델에서 항산화 활성을 나타내었으며 이들 농축액 및 퓨레를 이용한 항산화 활성을 갖는 건강음료의 원료로서 활용이 가능할 것으로 사료된다.

Antioxidant changes of leek (Allium ampeloprasum var. porrum) during spontaneous fermentation of the white shaft and green leaves

Leek is grown for its thickened cylindrical white shaft made up of long leaf bases. Despite the potentially valuable nutritional profile of the green leaves, a large portion remains unused owing its restricted culinary applications. This large quantity of this plant biomass could be valorized given an adequate stabilization method. In this study, we examined leek fermentation with regard to antioxidant changes. The oxygen radical absorbance capacity (ORAC) increased by 62% when the green leaves were fermented for 21 days, while 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity did not increase significantly. Fermentation resulted in an increase of endogenous polyphenolic compounds such as ferulic acid, astragalin, luteolin and naringenin. Moreover, fermentation stimulated the production of a series of polyphenolic compounds that were not present in the fresh leek. The flavour precursors in leek, i.e. methiin and isoalliin, were reduced by 91-93% and 100%, respectively, when spontaneous fermentation was allowed to occur on the white shaft and green leaves. Our results demonstrated that application of fermentation resulted in a higher ORAC value and polyphenol content of the leek plant, especially in the green leaves. These results indicate the nutritional relevance of fermentation, which hold promise as a stabilization technique.

Oxygen Radical Absorbance Capacity of Different Varieties of Strawberry and the Antioxidant Stability in Storage

Total antioxidant capacity of different varieties of strawberry (Ningfeng, Ningyu, Zijin 4, Toyonoka, Benihope, Sweet Charlie) in different developmental stages (including green unripe stages, half red stages, and red ripe stages) was investigated by oxygen radical absorbance capacity (ORAC) assay. In addition, effects of the antioxidant properties of strawberry stored at 4 °C or −18 °C for a period of five months were studied. The results showed that antioxidant capacity of strawberry changed based on tested part, developmental stage, variety, and time of collection. Calyces had significantly higher ORAC values compared with fruits. Strawberry fruits had higher ORAC values during the green unripe stages than the half red stages and red ripe stages. Strawberries got higher ORAC values during short-time storage, and then decreased during long-time storage. Samples stored at −18 °C exhibited higher antioxidant capacity than those stored at 4 °C, while vacuum treatment could further increase ORAC values. The results indicated the potential market role of strawberries as a functional food and could provide great value in preventing oxidation reaction in food processing and storage for the dietary industry.

Antioxidant Capacity and Polyphenol Content of Extracts from Crops Cultivated in Japan, and the Effect of Cultivation Environment

Oxygen radical absorbance capacity (ORAC) value and polyphenol contents of extracts from 71 crops cultivated in Japan were determined. Hydrophilic ORAC values (H-ORAC) ranged from 194 − 29,830 μmol TE/100 g fresh weight (FW), whereas lipophilic ORAC values (L-ORAC) ranged from 6 − 2,076 μmol TE/100 g FW. High total ORAC value was indicated in the following vegetables and fruits: mulukhiya (Corchorus olitorius), perilla (Perilla frutescens), water spinach (Ipomoea aquatica), edible burdock (Arctium lappa), blueberries (Vaccinium corymbosum) and hardy kiwifruit (Actinidia arguta) (6,562 − 31,802 μmol TE/100 g FW). These extracts showed a strong positive correlation between ORAC value and polyphenol content (r = 0.98). In addition, to gain a better understanding of ORAC variability in Japanese crops, we also discussed the influence of environmental factors on the ORAC value of stem mustard (Brassica juncea) “Umino” grown in three different regions. Regional variations of ORAC in stem mustard likely result from different levels of ultraviolet radiation and fertilizer application regimens specific to those regions.
