In Vitro Efficacy of Silver Carbene Complexes, SCC1 and SCC22, Against Some Enteric Animal Pathogens

Silver carbene complexes (SCCs), a group of novel silver-based compounds capable of gradually ‎releasing ‎silver ions, have shown significant antimicrobial activity against a wide range of ‎bacterial pathogens mainly ‎isolated from human cases. The antimicrobial activity against ‎animal isolated pathogens has yet been done. ‎The in vitro efficacy of two SCCs with different ‎carrier molecules (SCC1 with a methylated caffeine backbone ‎and SCC22 with a ‎dichloroimidazolium backbone) was investigated against three important animal and ‎human ‎pathogen species. SCC1 and SCC22 exhibited bacteriostatic and bactericidal effects against ‎multidrug ‎resistant Salmonella Typhimurium (poultry isolate), E. coli 843 and E. coli 1568 ‎‎(swine isolates), and the ‎poultry field isolates Salmonella Heidelberg, Salmonella Enteritidis, and ‎Salmonella Montevideo with MICs ‎and MBCs ranged from 16-21 µM (6-8 µg/mL) and 16-32 µM ‎‎(6-12 µg/mL), respectively. Clostridium ‎perfringens type A was sensitive to both SCC1 and ‎SCC22 with the MICs being 11 (4 µg/mL) and 21 µM ‎‎(8 µg/mL), respectively. These values were ‎comparable to the MICs and MBCs for silver acetate. The MBCs ‎against C. ‎perfringens was >85 µM for ‎SCCs and >192 µM for silver acetate (>32 µg/mL for all compounds). Ten hours incubation ‎of C. ‎perfringens with ‎‎40 µg/mL of all three products showed down regulation of virulence genes plc and netB, ‎‎suggesting viable cells and silver can modulate the virulence. Treating the C. ‎perfringens with higher ‎concentration (100 ‎‎µg/mL) of each SCC for 10 hours inhibited more bacteria compared to the ‎untreated bacterial cells, however, no ‎differences in the ultrastructure of lysed bacteria were seen ‎and this concentration might not induce viable ‎but non-culturable (VBNC) state as suggested by ‎transmission electron microscopy findings. SCCs showed a ‎broad antimicrobial activity against ‎all bacterial species tested including multidrug resistant pathogens. Both ‎SCCs demonstrated ‎inhibitory effect against the Gram-positive anaerobic C. perfringens type A ‎which ‎could have a high accumulation capacity for silver ion. These data suggest that SCCs may ‎represent a ‎novel class of broad-spectrum antimicrobial agents, which may be used to reduce the ‎burden of pathogenic ‎bacteria in the gastrointestinal tract of poultry‎‎‎.

Microbiome analyses of poultry feeds: Part II. Comparison of different poultry feeds

Within the realm of poultry feed mill operations, the persistent concern over microbial feed quality necessitates the establishment of a robust baseline for enhancing and sustaining the standards of commercial feeds. This dual-phase investigation, comprising Parts I, was previously published, and the current study presented here as Part II aimed to illuminate this baseline using 16S rRNA gene sequencing. In Part II, nine distinct commercial poultry feeds formulated as starters, growers, starter/growers, or supplements, the selected feeds underwent genomic DNA extraction, amplification with custom dual-indexed primers, and subsequent Illumina MiSeq sequencing. Through data analysis in QIIME2-2021.4 and R Studio, the study unveils alpha (Kruskal–Wallis) and beta (ANOSIM) diversity, taxonomic differences (ANCOM), and core microbiomes (core_members), deeming main and pairwise effects statistically significant at p < 0.05 and Q < 0.05. Notably, the investigation identified 30% common core microbial members across the nine feed types, shedding light on potential foodborne poultry pathogens such as Helicobacter and Campylobacter. Probiotic-associated feeds exhibited distinct microbial communities, emphasizing the need to explore their impact on the early poultry gastrointestinal tract (GIT) further.

Microalgae: A promising strategy for aflatoxin control in poultry feeds

Aflatoxins are toxic compounds produced by certain molds, primarily Aspergillus species, which can contaminate crops such as grains and nuts. These toxins pose a significant health risk to animals and humans. Aflatoxin B1 (AFB1) is the most potent of these compounds and has been well-characterized to lead to diminished growth and feed efficiency by disrupting nutrient absorption and metabolism in poultry. AFB1 can trigger apoptosis and inflammation, leading to a decline in immune function and changes in blood biochemistry in poultry. Recently, there has been growing interest in using microalgae as a natural antioxidant to mitigate the effects of aflatoxins in poultry diets. Microalgae have strong antioxidant, antimicrobial, anti-apoptotic, and anti-inflammatory properties, and adding them to aflatoxin-contaminated poultry diets has been shown to improve growth and overall health. This review investigates the potential of microalgae, such as Spirulina platensis, Chlorella vulgaris, and Enteromorpha prolifera, to mitigate AFB1 contamination in poultry feeds. These microalgae contain substantial amounts of bioactive compounds, including polysaccharides, peptides, vitamins, and pigments, which possess antioxidant, antimicrobial, and detoxifying properties. Microalgae can bind to aflatoxins and prevent their absorption in the gastrointestinal tract of poultry. They can also enhance the immune system of poultry, making them more resilient to the toxic effects of AFB1. Based on the data collected, microalgae have shown promising results in combating AFB1 contamination in poultry feeds. They can bind to aflatoxins, boost the immune system, and improve feed quality. This review emphasizes the harmful effects of AFB1 on poultry and the promising role of microalgae in reducing these effects.

Assessing Metschnikowia pulcherrima as a potential probiotic yeast for animal feed

Abstract In response to escalating antimicrobial resistance fuelled by their extensive use in livestock farming, this study explores alternative strategies in animal health management. We investigated the probiotic potential of Metschnikowia pulcherrima strains 4 × 3, DH5, ICS1 and QRI1 for their suitability in poultry environments. Our research demonstrated that previously discovered antimicrobial M. pulcherrima strains exhibit promising probiotic behaviours, including biofilm production, auto-aggregation, cell surface hydrophobicity, adhesion to Caco-2 cells, antioxidant capacity, and resilience to gastric stresses. Notable findings include DH5 exhibiting the highest biofilm formation, 4 × 3 and DH5 showing rapid auto-aggregation, 4 × 3 and ICS1 displaying high cell surface hydrophobicity, and all strains demonstrating considerable adherence to Caco-2 cells. 4 × 3 also exhibited exceptional bile tolerance, while ICS1 showed robust survival under simulated gastrointestinal conditions. These traits suggest M. pulcherrima’s capacity to colonise the poultry gastrointestinal tract, promote animal health, and support more sustainable livestock practices as a potential alternative to conventional antibiotics.

Investigation of the effect of allithiamine-enriched feed on the poultry gut microbiome composition and resistome

Over the past 20–25 years, the poultry industry has evolved into a specific protein production system. However, the stress resulting from intensive rearing practices has led to numerous negative consequences, making the optimisation of livestock gut microbiome composition crucial for mitigating these effects. Advancements in modern molecular biology methods have brought attention to the impacts of nutrients on gut microbiota. In our study, we extensively investigated the changes induced by feed formulations rich in phytonutrients on the gastrointestinal microbiota of livestock using targeted 16S rRNA amplicon sequencing. Our objective is to examine how the developed feed prototype affects the composition of core microbiomes in raised poultry, community diversity, and the resilience of complex microbial networks. We seek correlations between biological livestock and environmental samples to identify which community constituents, in what proportions and occurrences, may play a role in the development of specific diseases. Based on our measurement results, it can be asserted that allithiamine positively modulated "beneficial" community constituents. Beyond the impact of allithiamine-enriched feed rich in phytonutrients, the composition of the microbial community in the poultry gastrointestinal tract is significantly influenced by the age of the birds. Furthermore, due to the presence of multi-drug-resistant pathogens in environmental samples from livestock facilities, appropriate transmission risk management measures are of paramount importance.

The Bactericidal Activity of a Novel Aneurinibacillus aneurinilyticus Isolate Effectively Controls Foodborne Pathogens Campylobacter jejuni and Listeria monocytogenes

Human zoonotic infection with Campylobacter is a major cause of gastroenteritis in the United States and worldwide. Listeria monocytogenes causes a potentially fatal infection in humans and is often attributed to contaminated food. Genetic typing has demonstrated that Campylobacter infection is often associated with the consumption of contaminated poultry products, with Campylobacter often colonizing the poultry gastrointestinal tract, while listeriosis is commonly associated with the consumption of contaminated ready-to-eat (RTE) foods. In this study, a strain of endospore-forming bacterium (strain NH) that is bactericidal towards the human food pathogens Campylobacter jejuni and Listeria monocytogenes was identified and characterized. Transwell experiments demonstrated that the bactericidal effect on both C. jejuni and L. monocytogenes is due to secretions from the spore former. These foodborne pathogens consistently exhibited 7 log reductions in growth when exposed to the NH cell-free culture filtrate. Sequencing of the 16s rRNA gene V4 variable region and analysis of the full-length 16s rRNA gene sequence from the WGS indicated that strain NH belongs to the species Aneurinibacillus aneurinilyticus. A microplate bioassay demonstrated that a bactericidal substance that is sensitive to protease could be collected from cell-free filtrates by salting out with ammonium sulfate. Gel filtration chromatography indicated a native molecular weight for the bactericidal protein of ca. 50 kDa, consistent with a class III bacteriocin. The active protein bound strongly to a cation-exchange resin and with an isoelectric point of ten, suggesting a positively charged protein. Both cation-exchange chromatography and isoelectric focusing indicated the enrichment of an 11 kDa protein on SDS-PAGE. This protein was identified through mass spectroscopy as the flgM protein, an anti-sigma factor. Analysis of whole genome sequencing (WGS) of the strain NH genome indicated the presence of a number of non-conservative amino acid substitutions in the flgM-gene-derived amino acid sequence of strain NH and A. aneurinilyticus compared to other members of the Aneurinibacillus genus. Further investigation is needed to determine whether these substitutions are correlated with the bactericidal activity. The identified strain may be useful as a feed additive for the pre-harvest control of Campylobacter jejuni in poultry.

Multidrug-Resistant Strain ExPEC Isolat Asal Puyuh (Coturnix coturnix japonica)

AbstractExtraintestinal Escherichia coli (ExPEC) causes colibacillosis in poultry, including quail. ExPEC is a pathogenic E. coli that causes colibacillosis outside the gastrointestinal tract of poultry, in the form of peritonitis, pericarditis, salpingitis, synovitis, osteomyelitis, septicemia, pneumonitis, nephritis, pleurisy, proventriculitis and ventriculitis. In poultry farming antibiotics are used to promote growth and egg production, prevention and treatment of bacterial infections. Antibiotic resistance is a new problem that arises in poultry farming. In this study, 24 isolates of ExPEC strain from quail were used. These isolates did not differentiate sorbitol in the MacConkey (SMAC) sorbitol test assay. Antibiotic resistance was tested based on the inhibition of bacterial growth on Muller Hinnton Agar (MHA) media. 10 types of antibiotics were used to observe their resistance in all isolates. Resistance to Amikacin (87.5%), Ampicillin (87.5%), Ciprofloxacin (100%), Clindamycin (95.83%), Cefoxacin (25.0%), Doxycycline (87.5%), Erythromycin (100%), Gentamicin (95.83%), Penicillin (95.83%) and Tetracycline (83.3%). The isolates had multidrug-resistance between 3 to 10 types of antibiotics. This study showed that ExPEC isolates from quail were resistant to the type of antibiotics tested, so that the use of antibiotics in quail culture should be limited to reduce and prevent the emergence of new antibiotic resistance. Peculiarities of the composition of the digestive microbiota in poultry with fodder contamination with glyphosate

Relevance. The gastrointestinal tract in poultry is vulnerable to different irritators from food and water to medicals. That is why the mortality due to diseases of the digestive system may be as much as 30%. It has been established that about 600–900 bacteria species inhabit the poultry gastrointestinal tract. Microflora of healthy organism is a complex and balanced symbiotic microecosystem with normal metabolic characteristics. In addition to normoflora poultry gastroinrestinal tract is populated by opportunistic pathogenic and pathogenic microflora, which may be hazardous to the host.Methods. The issues regarded to establishing limits of different bacteria had been understudied until 2016. “Biotrof+” Ltd. was first to estimate threshold values for different groups of microorganisms under normal and pathological conditions.Results. Experiments proved that one of factors that may affect the structure of poultry microbiome are pesticide residues in feed. The trial on broilers fed with glyphosate showed that the microflora structure was altered by the pesticide significantly: the amount of staphylococci increased 5 times, enterobacteria increased 1,5 times, the amount of beneficial bacteria decreased. Supplementing the glyphosate contaminated feed with probiotic “Probiocid-Ultra” promoted to significant decrease in the opportunistic pathogenic and pathogenic microflora. Compared to the negative control the broilers average live weight fed with probiotic “ProbiocidUltra” was 1,0% higher. That may be the demonstration of the fact that negative effect of pesticide residues on poultry health and productivity can be minimized by supplementing feed with probiotics.

In silico prediction and expression analysis of vaccine candidate genes of Campylobacter jejuni

Campylobacter jejuni (C. jejuni) is the most common food-borne pathogen that causes human gastroenteritis in the United States. Consumption of contaminated poultry products is considered as the major source of human Campylobacter infection. An effective vaccine would be a promising alternative to antibiotic supplements to curb C. jejuni colonization in poultry gastrointestinal (GI) tract. However, the genetic diversity among the C. jejuni isolates makes vaccine production more challenging. Despite many attempts, an effective Campylobacter vaccine is not yet available. This study aimed to identify suitable candidates to develop a subunit vaccine against C. jejuni, which could reduce colonization in the GI tract of the poultry. In the current study, 4 C. jejuni strains were isolated from retail chicken meat and poultry litter samples and their genomes were sequenced utilizing next-generation sequencing technology. The genomic sequences of C. jejuni strains were screened to identify potential antigens utilizing the reverse vaccinology approach. In silico genome analysis predicted 3 conserved potential vaccine candidates (phospholipase A [PldA], TonB dependent vitamin B12 transporter [BtuB], and cytolethal distending toxin subunit B [CdtB]) suitable for the development of a vaccine. Furthermore, the expression of predicted genes during host-pathogen interaction was analyzed by an infection study using an avian macrophage-like immortalized cell line (HD11). The HD11 was infected with C. jejuni strains, and the RT-qPCR assay was performed to determine the expression of the predicted genes. The expression difference was analyzed using ΔΔCt methods. The results indicate that all 3 predicted genes, PldA, BtuB, and CdtB, were upregulated in 4 tested C. jejuni strains irrespective of their sources of isolation. In conclusion, in silico prediction and gene expression analysis during host-pathogen interactions identified 3 potential vaccine candidates for C. jejuni.

Evaluating the Effects of Pine and Miscanthus Biochar on Escherichia coli, Total Aerobic Bacteria, and Bacterial Communities in Commercial Broiler Litter.

Escherichia coli (E. coli) is a commensal bacteria found in the gastrointestinal tract of poultry; however, some strains are pathogenic and can cause a wide range of diseases. In addition, some strains of pathogenic E. coli can survive in the litter between flocks, making litter management critical for reducing E. coli-associated infections. Biochar (BC) is a porous, carbonaceous material that may be a beneficial litter amendment to reduce moisture and microbial loads. The objectives of this study were to evaluate the effects of pine BC, miscanthus BC, and Poultry Litter Treatment (PLT) on E. coli, total aerobic bacteria populations, and bacterial communities when added to used broiler litter. Pine and miscanthus BC were mixed into poultry litter at inclusion rates of 5%, 10%, 20%, 25%, and 30% w/w. PLT was surface applied at a rate of 0.73 kg/m2. Baseline E. coli and aerobics were measured after a 48-hr litter incubation period and just prior to adding litter treatments. Escherichia coli and aerobics were enumerated 2 and 7 days after adding treatments. Overall, pine BC at 30% had the lowest E. coli and aerobic counts (5.98 and 6.44 log 10 colony-forming units [CFU]/g, respectively); however, they were not significantly different from the control (P ≤ 0.05). At day 2, 30% pine BC inclusion rate treatment resulted in a significant reduction in E. coli and aerobic bacteria counts compared to the control. Miscanthus BC application did not result in significant reductions in E. coli or aerobic bacteria at days 2 or 7. PLT had the highest E. coli (7.07 log 10 CFU/g) and aerobic counts (7.21 log 10 CFU/g) overall. Bacterial community analysis revealed that the alpha and beta diversity between pine BC- and PLT-treated litter were significantly different. However, neither BC type significantly impacted bacterial diversity when compared to the control. Differences in E. coli and aerobic counts between BC types may be attributed to variations in feedstock physiochemical properties.

Effects of DNA extraction methods on the real time PCR quantification of Campylobacter jejuni, Campylobacter coli, and Campylobacter lari in chicken feces and ceca contents

Polymerase chain reaction (PCR) method was coupled with a DNA extraction to enumerate Campylobacter spp. from poultry gastrointestinal tract samples. Three experiments were conducted that included: 1) Development of a DNA standard curve related to bacterial DNA primers; 2) Design of a cell/genomic DNA extraction protocol to isolate Campylobacter spp. DNA from complex samples such as poultry feces; and 3) Comparison of PCR quantification to standard plate count methodology. The standard curve using primers for Campylobacter spp. was created for DNA extracted from environmental isolates with a linear range (R2 > 0.95) and with a high specificity for C. coli and C. jejuni recovered from poultry, swine and laboratory isolates. A 2-step extraction process of bacterial DNA from poultry feces was developed in which the cells were first concentrated using a gradient-centrifugation step followed by comparison of 4 DNA extraction methods. Two commercial DNA extraction methods (Zymo Research Quick DNA, and Invitrogen magnetic separation), a traditional phenol-chloroform DNA extraction method using proteinase K to inactivate DNAses, and an in-house isolation method for DNA extraction based on chaotropic salts were used. The middle gradient layer recovered 89% to 98% of the bacteria cells from the sample, with recovery dependent upon the Campylobacter genus. The 4 DNA extractions methods recovered 112 to 302 ug/nL of DNA. Finally, the qPCR and standard plate methods were highly correlated for enumerating Campylobacter spp. in the 2.0 to 8.0-log CFU range. Analyses of the results from this study demonstrate that the combination of the standard curve for Campylobacter spp. DNA primers, the gradient cell concentration method and DNA extraction techniques with qPCR can be used to enumerate Campylobacter spp. from poultry samples with findings similar those of traditional plate count methodology.

Probiotics as a Friendly Antibiotic Alternative: Assessment of Their Effects on the Health and Productive Performance of Poultry

Antibiotics have been used to maintain the overall health of poultry by increasing production efficiency, promoting growth, and improving intestinal function for more than 50 years. However, they have a number of side effects, such as antibiotic resistance, gut dysbiosis, destruction of beneficial bacteria, and the potential to spread diseases to humans. In order to address the aforementioned issues, a lot of effort is put into the development of antibiotic alternatives. One of them is the use of probiotics that can be added to the feed in order to increase poultry performance and avoid the aforementioned problems. Probiotics are live microorganisms consumed as feed additives or supplements. They function in the poultry gastrointestinal tract to benefit the host. Probiotics improve growth performance, bone health, meat and eggshell quality. The addition of probiotics to the diet also positively affects the immune response, intestinal microflora, and disease resistance. Careful selection of probiotic strains is of utmost importance. This review focuses on the significance of probiotics as a potential antibiotic-free alternative and the way in which they can be used as supplements in poultry feed for boosting production and safeguarding health.

Molecular detection and risk factors associated with multidrug-resistant Campylobacter jejuni from broiler cloacal and meat samples in Bangladesh.

The gastrointestinal tract of poultry is a potential source of Campylobacter jejuni. Here, the prevalence, risk factors, antimicrobial susceptibility profile and genetic relationship of C.jejuni were studied in broilers from farms and meat from live bird markets (LBMs) and super shops (SS). Pooled cloacal samples were obtained from farms in six districts of Bangladesh between June 2019 and March 2020. Pooled meat samples were obtained from LBMs and SS in the Chattogram district. Microbial culture, polymerase chain reaction (PCR), antimicrobial susceptibility tests were used to detect multidrug-resistant C.jejuni. A positive PCR amplicon was validated by mapA partial gene sequencing and subsequent phylogenetic analysis. In total, 12.5% (95% CI: 8.5-17.7%) of farms (N=216) and 27.1% (95% CI: 15.28-41.85%) of LBMs and SS (N=48) tested positive for C.jejuni. Moreover, 98% of the isolates were multidrug-resistant, with 86% resistant to five or more antimicrobial groups. Multivariable logistic regression analysis showed a downtime of <14 days, no separate footwear for shed access, and more than one person entering the sheds were significantly associated with C.jejuni colonization. Phylogenetic analysis revealed a strong relationship between C.jejuni strains obtained in Bangladesh and strains isolated in India, South Africa and Grenada from humans, pigs and bats. This study revealed significant contamination of broiler meat with Campylobacter spp. and C.jejuni. Potential sources of contamination and anthropogenic factors associated with the alarming prevalence of C.jejuni identified in this study would aid in reducing the growing risks of broiler-associated pathogens.

Application of Bacteriophages to Limit Campylobacter in Poultry Production.

Campylobacter is a major foodborne pathogen with over a million United States cases a year and is typically acquired through the consumption of poultry products. The common occurrence of Campylobacter as a member of the poultry gastrointestinal tract microbial community remains a challenge for optimizing intervention strategies. Simultaneously, increasing demand for antibiotic-free products has led to the development of several alternative control measures both at the farm and in processing operations. Bacteriophages administered to reduce foodborne pathogens are one of the alternatives that have received renewed interest. Campylobacter phages have been isolated from both conventionally and organically raised poultry. Isolated and cultivated Campylobacter bacteriophages have been used as an intervention in live birds to target colonized Campylobacter in the gastrointestinal tract. Application of Campylobacter phages to poultry carcasses has also been explored as a strategy to reduce Campylobacter levels during poultry processing. This review will focus on the biology and ecology of Campylobacter bacteriophages in poultry production followed by discussion on current and potential applications as an intervention strategy to reduce Campylobacter occurrence in poultry production.

Biochemical property analysis of native probiotic isolates from selective poultry

Isolation and identification of probiotic bacteria are the prerequisites for their safer use in the food and feed industry. The objectives of the present study were the isolation of probiotic bacteria from the selective gastrointestinal tract of poultry obtained from Khulna and Barisal Divisions, and their identification based on bacterial morphological characterization and biochemical property analysis. Ten potential native probiotics were isolated from the poultry gastrointestinal tract and assayed for their morphological, physiological and biochemical properties. It was observed that, all the isolates were rod-shaped, gram-positive, endospore-negative, catalase-negative, non-motile and were able to ferment particular sugars which are an indicator for typical probiotic bacteria. The sugar fermentation pattern, ability to survive and growth in inhibitory substances like 1-4% NaCl, 0.3% bile salt as well as their ability to grow in different temperatures and pH levels ensured the presumptive identification of the lactic acid bacteria. All the ten isolates exhibited a clear zone of inhibition when they were grown with five enteric pathogens which are indicative of their antimicrobial activity. Ten isolates were assayed for their susceptibility to eight antibiotics using the disc diffusion method. All the isolates were resistant to tetracycline and nalidixic acid. Further research regarding molecular characterization and identification of specific genes using different technologies may open the door to utilize these isolates in different probiotic-based inventions. INFLUENCE OF SPORO-LEX AND ANALCIM-SI ON THE MICROFLORA OF THE BIRD'S GASTROINTESTINAL TRACT

The constant interaction between bacteria in the gastrointestinal tract, intestinal epithelial cells and immune system cells is a continuous interaction that is central to maintaining immune homeostasis.&#x0D; "Analtsim-Si" is a montmorillonite rock of Volodymyretsky field. Probiotic "Sporo-lex" is a mixture of probiotic cultures of Bacillus licheniformis VK-25 and Bacillus subtilis MK-3 on a natural standardized sorbent – montmorillonite rock of Volodymyretsky field. The subjects were laying hens of 130 days of age, Lohmann Brown cross (n = 30). The bird of the first group got compound feed with 2% of "Analtsim-Si", the bird of the second group got compound feed with 2% of the probiotic "Sporo-lex". The third group – was a control group and received the usual food.&#x0D; Prior to the start of the study, as well as on days 7, 14 and 28, the bird was weighed, blood and feces were collected for the study. On the 27th day of the experiment, a pathological examination of birds from all groups was performed to study the patency of the gastrointestinal tract.&#x0D; The effect of experimental veterinary drugs "Sporo-lex" and "Analtsim-Si" on the microflora of the gastrointestinal tract of poultry was established. The drug "Sporo-lex" has a more pronounced effect, which is manifested in the suppression of bacteria of the Escherichia coli group due to the antagonistic action of strains of Bacillus licheniformis VK-25 and Bacillus subtilis MK-3, which are part of the drugs.&#x0D; The average growth of turkeys during the use of the probiotic "Sporo-lex" was studied. As a result of the conducted researches it was established that the bird of the experimental group at the end of the experiment had 470 grams more body weight compared to the control. During the autopsy of birds from both groups, no significant signs of pathology of organs and systems were detected.&#x0D; In the future, it is necessary to study the effect of drugs "Sporo-lex" and "Analcym-Si" on the dynamics of changes in the immunological parameters of birds.

Preparation and application of a functional biological additive for quails based on microbial conversion of plant raw materials

When developing functional feed additives it is of interest to use microorganisms that have the ability to take root in the gastrointestinal tract of poultry and normalize its microflora to have a positive effect on the immune system. One of these bacteria is Propionibacterium shermanii, which has a fortifying, immunostimulating, antimutagenic effect, and can reduce the toxic effect of ultraviolet light and chemical compounds. Azotobacter vinelandii, which has probiotic fortifying, antibiotic properties, is effective as a stimulator of the development of the entire microbiota in a contaminated object, and recycles the formed ammonia in bird droppings, was chosen as another microorganism for creating the additive. The purpose of the research was to develop a functional biological additive based on probiotic microflora, using the by-products of crop processing in the formulations, and to select the norms for its introduction into the diet of quails. As a result of the research the technology of joint cultivation of Propionibacterium shermanii and Azotobacter vinelandii strains was developed, a universal starter culture was created that allows to ferment by-products of processing of plant raw materials, a functional additive with immunostimulating, vitamin and provitamin properties for quails has been obtained. The results of the production test confirmed the effectiveness of the developed additive in the diet of quails. Economic efficiency in the application of the proposed functional biological additive has been shown a reduction in feed consumption per 1 kg of live weight gain by 5,13 %. In addition, the economic effect of using the functional biological additive increases. Thus, the profit in the experimental group was 2738,34 rubles, which was 12,45 % higher than in the control group.

Pharmacokinetics of Colistin in the Gastrointestinal Tract of Poultry Following Dosing via Drinking Water and Its Bactericidal Impact on Enteric Escherichia coli

Colistin, a last-line antibiotic of major importance in veterinary medicine and of critical importance in human medicine, is authorized to treat gastrointestinal (enteric) infections caused by non-invasive Escherichia coli in multiple veterinary species including poultry. Its use in veterinary medicine has been implicated in the widespread prevalence of mobilized colistin resistance. The objectives of this study were to determine the intestinal content reached in broiler chickens during 72-h treatment with colistin, to evaluate the associated impact on intestinal E. coli density, and to select less susceptible E. coli populations. In this study, 94 broiler chickens were administered a dose of 75,000 IU/kg/day via drinking water. Intestinal samples were collected pre-, during-, and post-dosing. Luminal intestinal content was assessed for colistin content by ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS), and E. coli were isolated and enumerated on UriSelect agar™. Minimum inhibitory concentration (MIC, for eight isolates per intestine per animal) was determined, and when higher than the epidemiological cutoff (ECOFF 2 mg/l), isolates were screened for mobilized colistin resistance (mcr)-1 to 5. Colistin content increased during treatment to a maximum of 5.09 mg/kg. During this time, the total population of E. coli showed an almost 1,000-fold reduction. An apparent increase in the relative abundance of E. coli with an MIC ≥ ECOFF, either mcr-negative (6.25–10.94%) or mcr-1-positive (4.16–31.25%) was observed, although this susceptibility shift was not maintained post-treatment. Indeed, following cessation of dosing, colistin was eliminated from the intestine, and content was below the limit of quantification (LOQ, 1.1 mg/kg) within 4 h, and the median MIC of E. coli isolates returned below baseline thereafter. Few isolates with a lower susceptibility (mcr-1-positive or negative) were however observed at the end of the study period, indicating maintained sub-populations in the chicken gut. The results of this study show a limited impact on long-term maintenance of less susceptible E. coli populations as a direct result of colistin treatment in individual birds.

The Use of Probiotics in the Reduction of Campylobacter spp. Prevalence in Poultry.

In 2018, the Member States reported 246,571 cases (30% increase since 2015). For years, poultry meat and poultry products have been considered a main source for human infections. In 2018, the highest occurrence of Campylobacter spp. was detected in chicken (37.5%) and turkey meat (28.2%). Considering this situation, there has been ongoing discussion regarding the potential strategies to minimize the level of Campylobacter spp. colonization in poultry and therefore in humans. In 2018, EFSA Panel on Biological Hazards indicated that use of feed and water additives is the second most likely strategy that can be successful in minimizing Campylobacter spp. colonization rate in broiler chickens. One of these water and feed additives are probiotics—living microorganisms which, when supplemented in the right dose, have a positive effect on microbial ecosystem of the host gut by ensuring a favorable balance between commensal and pathogenic microflora. In this review paper, the authors present current results of the studies concerning the potential use of probiotics as a preventive measure of Campylobacter spp. infection, under laboratory conditions and at a chicken farm level.Campylobacter spp. are widely distributed microorganisms, many of which are commensals of gastrointestinal tract in multiple animal species, including poultry. Most commonly detected are C. jejuni and C. coli. Although infections are usually asymptomatic in poultry, poultry meat and products represent main sources of infection with these bacteria to humans. According to recent EFSA report, campylobacteriosis is the most commonly reported zoonotic disease. In 2018, EFSA Panel on Biological Hazards indicated that use of feed and water additives is the second most likely strategy that can be successful in minimizing Campylobacter spp. colonization rate in broiler chickens. One of those feed and water additives are probiotics. From numerous research papers it can be concluded that probiotics exhibit plenty of mechanisms of anti-Campylobacter activity, which were evaluated under in vitro conditions. These results, to some extent, can explain the efficacy of probiotics in in vivo studies, although different outcome can be observed under these two laboratory conditions. Probiotics are capable of reducing Campylobacter spp. population count in poultry gastrointestinal tract and they can reduce carcass contamination. Potential modes of anti-Campylobacter activity of probiotics, results of in vivo studies and studies performed at a farm level are widely discussed in the paper.
