Sleep problems and circadian rhythm functioning in autistic children, autism with co-occurring attention deficit hyperactivity disorder, and typically developing children: A comparative study.

Sleep problems are common in autism spectrum disorder (ASD) and different factors can contribute to its occurrence in this population. Misalignment of the biological clock (our circadian system) has been described as one possible explanation. While there is a body of research on sleep problems, relatively less is known about circadian functioning and the specific population of autistic children with co-occurring attention deficit hyperactivity disorder (ADHD). Using an ambulatory circadian monitoring (ACM) system, which resembles a common watch, we gathered sleep parameters and the different rhythms obtained from measuring motor activity, light exposure and distal temperature in 87 autistic children and adolescents, 27 of whom were diagnosed with co-occurring ADHD, and 30 neurotypical children and adolescents as a comparison group. Autistic children and, especially, those with co-occurring ADHD showed greater motor activity during sleep which would be worth studying in future projects which could better define this restless sleep. Of note, we observed an atypical pattern of wrist temperature, with higher values in neurotypical children, followed by autistic children and, ultimately, those with co-occurring ADHD. Temperature is one of the most valuable factors evaluated here as it is closely connected to sleep-wakefulness and the hormone melatonin. Its special pattern during day and nighttime would support the hypothesis of an atypical secretion of melatonin in autistic individuals which would also link with the higher presence of sleep problems in this neurodevelopmental condition.

Direct interaction in digital interactive media and stock performance: Evidence from Panorama.

Media information plays an essential role in the stock market. Recent financial research has verified that media information could shock stock price by influencing investors' expectation. Now, a new type of interactive media, called Digital Interactive Media (DIM), is popular in Chinese stock market and becomes the main channel for investors to understand listed companies. Unlike general news media or investor forums, DIM enables direct interaction between listed companies and investors. In the modern society where digital economy is booming, media information would largely affect investors' decisions. Therefore, it is urgent to use natural language processing (NLP) technology to deconstruct the massive questions and answers (Q&A) interactive information in DIM and extract valuable factors that affect stock prices and stock performances to explore the influence mechanism of digital interactive information on stock performances. This paper firstly uses web crawling technology to obtain approximately 110000 Q&A text information from the digital interactive platform ('Panoramic Network') from 2015 to 2021. Then we use big data text analysis technology and emotional quantification technology to extract valuable influencing factors from the massive text. A Multiple Linear Regression (MLR) model was created to explore specific influence mechanism of digital interactive information on stock price performance. The empirical results show that the emotions implicit in investors' questions do not significantly impact stock performance. However, the emotions and attitudes of the answers by listed companies can significantly affect corresponding stock prices, which indirectly confirms the Proximate Cause Effect of behavioral finance. This effect is particularly evident in the stock prices on the current trading day and the next trading day. In the Robustness Test, this paper replaces dependent variable and adds relevant control variables, and the conclusion remains valid. In the Endogeneity Test, this paper selects sample data before the launch of Panorama Network in 2014 as a comparison, and uses a Difference-in-Difference (DID) model to prove the significant impact of the launch of Panorama Network on Chinese stock market. In the Heterogeneity Test, the paper classifies the market value, region, and industry of listed companies and regressed the sub samples, once again confirming the reliability of the empirical conclusions. The results of Robustness Test, Endogeneity Test, and Heterogeneity Test conducted in this paper all support empirical conclusions.

Atherogenic index of plasma as a predictor of coronary artery disease: a cohort study in south of Iran

BackgroundCoronary artery disease (CAD) is an atherosclerotic disease of an inflammatory nature. Previous studies examining the relationship between triglycerides and high-density lipoprotein cholesterol have highlighted the importance of plasma atherogenic index (AIP) as an important predictor of coronary heart disease. However, due to the lack of adequate information on this topic, this study aimed to investigate the relationship between AIP and coronary heart disease risk.ResultsThis study included 2,226 women and 1,690 men aged 35–70 years who participated in the Bandar Kong Cohort study and met the eligibility criteria. The data was collected using a checklist and questionnaires, which were designed by experienced individuals. After participants completed a registration form and gave informed consent, face-to-face interviews were conducted by trained experts. The validity and reliability of the questionnaire had been verified by the national cohort team prior to its use. The Ethics Committee of Hormozgan University of Medical Sciences (IR.HUMS.REC.1400.171) approved the study. Data from the initial cohort survey using SPSS software version 25, were analyzed to include several factors, including age, sex, smoking status, body mass index (BMI), physical activity level, socioeconomic status, AIP, systolic blood pressure, and diastolic blood pressure. The prevalence of coronary heart disease was found to be 7.5% higher in people with a BMI of 25 or higher. Also, Individuals with low physical activity had a higher prevalence. Individuals with CAD had significantly higher mean values for the AIP, age, systolic blood pressure, and diastolic blood pressure (0.46, 57.50, 128.43, and 81.10, respectively) compared to those without CAD. Furthermore, patients with CAD had lower years of education (2649.45 and 3.59) than individuals without CAD (P < 0.05). Importantly, our findings showed that AIP increased the odds ratio of coronary heart disease by 1.86 as an independent risk factor.ConclusionsBased on our investigation, the AIP is a valuable and independent predictive risk factor for coronary artery disease. This index can be utilized effectively due to its accessibility and affordability, making it a promising tool for risk assessment in clinical settings.

Selecting Pedal Load for Lower-Limb Rehabilitation Based on the Combination of Muscle Synergy and Fourier Series

This paper introduces a new lower-limb rehabilitation machine that meets the rehabilitation needs of hemiplegic patients. First, a left–right independent rotary pedal mechanism was selected to facilitate rehabilitation and adapt to the user’s physical condition. Then, a half model of the lower-limb rehabilitation machine is designed and manufactured with ergonomics in mind. As analytical tools, we combine non-negative matrix factorization and non-negative double singular value decomposition to calculate muscle synergy of the walking muscle surface electromyography (sEMG) signal, and use cosine similarity to evaluate the similarity between walking and pedaling activities. By comparing the results of the walking and pedaling experiments, the effectiveness of pedaling in gait rehabilitation is revealed. To further improve the similarity between walking and pedaling, double integration of the sEMG signal is introduced, and the relationship between load input and rotation angle is described for the first time using Fourier series. The results of the experiment confirmed that more than half of the 10 subjects performed pedaling exercises similar to walking using Fourier series loading compared to pedaling exercises with normal constant loading. This loading parameter may have the potential to improve rehabilitation efficiency for many subjects compared to the usual exercise.

Quantitative analysis of social influence and digital piracy contagion with differential equations on networks

Illegal file sharing of copyrighted contents through popular file sharing networks poses an enormous threat to providers of digital contents, such as, games, softwares, music and movies. Though empirical studies of network effects on piracy is a well-studied domain, the dynamics of peer effect in the context of evolving social contagion has not been enough explored using dynamical models. In this research, we methodically study the trends of online piracy with a continuous ODE approach and differential equations on graphs to have a clear comparative view. We first formulate a compartmental model to study bifurcations and thresholds mathematically. We later move on with a network-based analysis to illustrate the proliferation of online piracy dynamics with an epidemiological approach over a social network. We figure out a solution for this online piracy problem by developing awareness among individuals and introducing media campaigns, which could be a valuable factor in eradicating and controlling online piracy. Next, using degree-block approximation, network analysis has been performed to investigate the phenomena from a heterogeneous approach and to derive the threshold condition for the persistence of piracy in the population in a steady state. Considering the dual control of positive peer influence and media-driven awareness, we examine the system through realistic parameter selection to better understand the complexity of the dynamics and suggest policy implications.

Analysis of rock grinding results in centrifugal-vibrational disintegrating module

This scientific study is devoted to analysis of the results of grinding materials such as marble, sandstone and granite in a disintegrating module. This module combines grinding of particles in the countercurrent centrifugal mill with simultaneous vibrational screening of the finished product. The essence of the study is that two-factor experiments have been carried out according to the full factor scheme. Here, the achieved degree of crushing is used as the response function. The valuable factors have the initial size and the ultimate strength of the material. As a result of two-factor analysis of variance, it has been found that the compressive strength and the initial size of the material, regardless of the sieve vibration amplitude, have a significant and comparable scale effect on the material crushing ratio. At the same time, the interaction of factors has no statistical significance. Also, approximation of experimental data by the first- and the second-order regression equations has been performed. For simplified calculations, it is recommended to use the linear regression equation, while for more accurate ones, the second-order equation is more suitable.

Time to Positivity in Blood Culture Bottles Inoculated with Sonication Fluid from Fracture-Related Infections.

The timely and accurate identification of causative agents is crucial for effectively managing fracture-related infections (FRIs). Among various diagnostic methods, the "time to positivity" (TTP) of cultures has emerged as a valuable predictive factor in infectious diseases. While sonication of implants and inoculation of blood culture bottles with sonication fluid have enhanced sensitivity, data on the TTP of this microbiological technique remain limited. Therefore, patients with ICM criteria for confirmed FRI treated at our institution between March 2019 and March 2023 were retrospectively identified and their microbiological records were analyzed. The primary outcome parameter was TTP for different microorganism species cultured in a liquid culture collected from patients with confirmed FRI. A total of 155 sonication fluid samples from 126 patients (average age 57.0 ± 17.4 years, 68.3% males) was analyzed. Positive bacterial detection was observed in 78.7% (122/155) of the liquid culture pairs infused with sonication fluid. Staphylococcus aureus was the most prevalent organism (42.6%). Streptococcus species exhibited the fastest TTP (median 11.9 h), followed by Staphylococcus aureus (median 12.1 h) and Gram-negative bacteria (median 12.5 h), all of which had a 100% detection rate within 48 h after inoculation. Since all Gram-negative pathogens yielded positive culture results within 24 h, it could be discussed if empirical antibiotic therapy could be de-escalated early and limited towards the Gram-positive germ spectrum if no Gram-negative pathogens are detected up to this time point in the context of antibiotic stewardship.

On the importance of asset pricing factors in the relative valuation

In this study, we examine the implications of multi-factor asset pricing models in corporate valuation, focusing on the relative valuation approach. Our investigation centers around an eight-factor model, which we compare against traditional valuation methods that rely on industry or risk grouping and cross-sectional averages. The purpose of this comparison is to assess the efficacy of incorporating pricing factor variables into the valuation process and to understand their impact on valuation accuracy. The findings of our analysis suggest that the use of the eight-factor model leads to a reduction in estimation errors of valuation multiples, indicating an important improvement in precision compared to conventional valuation methods. Additionally, our research offers insights into the practicality of applying multi-factor models. It contributes to the intersection between empirical asset pricing and corporate finance by providing a detailed evaluation of advanced valuation models and their relevance in improving the accuracy of relative valuation.

A hypoxia–glycolysis–lactate-related gene signature for prognosis prediction in hepatocellular carcinoma

BackgroundLiver cancer ranks sixth in incidence and third in mortality globally and hepatocellular carcinoma (HCC) accounts for 90% of it. Hypoxia, glycolysis, and lactate metabolism have been found to regulate the progression of HCC separately. However, there is a lack of studies linking the above three to predict the prognosis of HCC. The present study aimed to identify a hypoxia–glycolysis–lactate-related gene signature for assessing the prognosis of HCC.MethodsThis study collected 510 hypoxia-glycolysis-lactate genes from Molecular Signatures Database (MSigDB) and then classified HCC patients from TCGA-LIHC by analyzing their hypoxia-glycolysis-lactate genes expression. Differentially expressed genes (DEGs) were screened out to construct a gene signature by LASSO-Cox analysis. Univariate and multivariate regression analyses were used to evaluate the independent prognostic value of the gene signature. Analyses of immune infiltration, somatic cell mutations, and correlation heatmap were conducted by “GSVA” R package. Single-cell analysis conducted by “SingleR”, “celldex”, “Seurat”, and “CellCha” R packages revealed how signature genes participated in hypoxia/glycolysis/lactate metabolism and PPI network identified hub genes.ResultsWe classified HCC patients from TCGA-LIHC into two clusters and screened out DEGs. An 18-genes prognostic signature including CDCA8, CBX2, PDE6A, MED8, DYNC1LI1, PSMD1, EIF5B, GNL2, SEPHS1, CCNJL, SOCS2, LDHA, G6PD, YBX1, RTN3, ADAMTS5, CLEC3B, and UCK2 was built to stratify the risk of HCC. The risk score of the hypoxia-glycolysis-lactate gene signature was further identified as a valuable independent factor for estimating the prognosis of HCC. Then we found that the features of clinical characteristics, immune infiltration, somatic cell mutations, and correlation analysis differed between the high-risk and low-risk groups. Furthermore, single-cell analysis indicated that the signature genes could interact with the ligand-receptors of hepatocytes/fibroblasts/plasma cells to participate in hypoxia/glycolysis/lactate metabolism and PPI network identified potential hub genes in this process: CDCA8, LDHA, YBX1.ConclusionThe hypoxia–glycolysis–lactate-related gene signature we built could provide prognostic value for HCC and suggest several hub genes for future HCC studies.

A Psychometric Validation of the Sociomathematical Norm Scale for Senior High School Students in Mathematics Learning

&lt;p style="text-align: justify;"&gt;Students in mathematics classes do not understand the importance of sociomathematical norms in learning mathematics. This causes sociomathematical norms not to be teachers' focus when learning mathematics. Besides, there is no standardized instrument for assessing this norm, so developing this instrument is necessary to measure socio-mathematical norms in learning mathematics. This study aims to create and verify the psychometric validity of the sociomathematical norm scale. This research used a survey method with 505 senior high school students from Jakarta and West Java as respondents. The results showed that 25 items had convergent validity, with a loading factor value of &gt; 0.700, meaning they could be declared valid. Concurrent validity indicates that each sociomathematical norms indicator is valid as a whole. Discriminant validity shows that the average variance extracted value on the diagonal is higher than the other values, so each item is declared valid. It was concluded that each item of the sociomathematical norms instrument has accuracy in its measurement function. The reliability test shows that each sociomathematical norms item is declared reliable. The reliability value of the sociomathematical norm item is .99, and the person's reliability is .86. Thus, the instruments developed can measure sociomathematical norms in learning mathematics.&lt;/p&gt;

Biological functions and clinic significance of SAF‑A (Review).

As one member of the heterogeneous ribonucleoprotein (hnRNP) family, scaffold attachment factor A (SAF-A) or hnRNP U, is an abundant nuclear protein. With RNA and DNA binding activities, SAF-A has multiple functions. The present review focused on the biological structure and different roles of SAF-A and SAF-A-related diseases. It was found that SAF-A maintains the higher-order chromatin organization via RNA and DNA, and regulates transcription at the initiation and elongation stages. In addition to regulating pre-mRNA splicing, mRNA transportation and stabilization, SAF-A participates in double-strand breaks and mitosis repair. Therefore, the aberrant expression and mutation of SAF-A results in tumors and impaired neurodevelopment. Moreover, SAF-A may play a role in the anti-virus system. In conclusion, due to its essential biological functions, SAF-A may be a valuable clinical prediction factor or therapeutic target. Since the role of SAF-A in tumors and viral infections may be controversial, more animal experiments and clinical assays are needed.

Cholinergic hyperintensity pathways are associated with cognitive performance in patients with asymptomatic carotid artery stenosis

ObjectivesWe aimed to determine whether asymptomatic carotid artery stenosis (ACS) induced cognitive impairments were related to the cholinergic hyperintensity pathway. MethodsThis cross-sectional study included patients with moderate-to-severe ACS, who were categorized into mild cognitive impairment (MCI) and normal cognition groups on the basis of Montreal Cognitive Assessment (MoCA) scores. The cholinergic pathway hyperintensity scale (CHIPS), Fazekas, and medial temporal atrophy (MTA) scores were assessed. SPSS software was used for statistical analyses. ResultsA total of 117 ACS patients (70.89 ± 8.81 years) and 105 controls (67.87 ± 9.49 years) were evaluated (t = 2.46, p = 0.015). The ACS group showed a worse median Mini-Mental Status Examination (MMSE) score (z = −2.41, p = 0.016) and MoCA score (z = −3.51, p < 0.001), and a significantly higher median total CHIPS score (z = 4.88, p < 0.001) and mean Fazekas score (t = 2.39, p = 0.018). In the correlation analysis, the MoCA score showed a significant negative correlation with the CHIPS score (ρ = −0.41, p < 0.001) and Fazekas score (ρ = −0.31, p < 0.001) in ACS group. Logistic regression analyses suggested that CHIPS scores were risk factors for MCI in patients with ACS (odds ratio [OR] = 1.07, 95% Confidence Interval [CI]1.01–1.13 and controls (OR = 1.09, 95%CI 1.01–1.17), while the MTA and Fazekas scores showed no predictive power. The receiver operating characteristic curve showed that the area under the curve of the CHIPS score for predicting MCI was 0.71 in ACS group, but was only 0.57 in controls. ConclusionsPatients with ACS showed poorer cognitive performance and higher CHIPS and Fazekas scores. CHIPS, but not Fazekas, scores were risk factors for cognitive impairment and were a valuable factor to predict MCI in patients with ACS.

Implementation of Learning Management Systems (LMS) in higher education systems through bipolar complex hesitant fuzzy Aczel-Alsina power aggregation operators: A case review for China.

A learning management system (LMS) is a web-based application or software platform computed to facilitate the development, tracking, management, reporting, and delivery of education and training programs. Many valuable and dominant factors are working behind the Learning Management System (LMS), but no one can find which factor is most important and valuable for LMS during COVID-19 among the following five alternatives, called Improved Accessibility, Blended Learning, Collaboration and Communications, Assessment and Evaluation, and Administrative Efficiency. For this, first, we derive the techniques of bipolar complex hesitant fuzzy (BCHF) sets, and then we evaluate some flexible operational laws, called Algebraic operational laws and Aczel-Alsina operational laws. Secondly, using the above techniques, we elaborate the technique of BCHF Aczel-Alsina power averaging (BCHFAAPA), BCHF Aczel-Alsina power weighted averaging (BCHFAAPWA), BCHF Aczel-Alsina power geometric (BCHFAAPG), and BCHF Aczel-Alsina power weighted geometric (BCHFAAPWG) operators. Some basic properties are also investigated for each proposed operator. Further, to evaluate the problem concerning LMS, we compute the multi-attribute decision-making (MADM) techniques for invented operators. Finally, we select some prevailing operators and try to compare their ranking results with our proposed results to enhance the worth and capability of the invented theory.

According to the analysis of the research results of foreign researchers, it was confirmed that ESG management activities had a direct and indirect effect on the financial performance of a company. ESG occupies the most important sector of a company's valuation factors. In particular, in the process of transitioning from shareholder capitalism to stakeholder capitalism, corporate social responsibility (CSR) became an essential for sustainable development, not an option. Research on ESG has increased rapidly through the COVID-19 pandemic, and research results also speak about the positive effects of ESG management in various areas and aspects. This paper examins ESG and management performance, especially focusing on recently published papers among overseas research. Most studies have confirmed a positive relationship between the two. In addition to simple financial performance, ESG was analyzed to have a significant effect in various aspects such as the relationship between ESG practices and debt costs, the relationship between corporate risk taking, the effect of suppressing misconduct by managers, the relationship with corporate default risk, the relationship with the bond credit spread, the relationship with trade credit finance, and the moderating effect of corporate controversy and ESG practices. While the existing ESG research trend analysis studies were external analysis using text mining or keyword network analysis, this study is meaningful in that the trend was analyzed based on the content. However, I think a separate study is needed for the overall trend of the research method and domestic research trend.

Prediction of bleeding in placenta accrete spectrum with lacunar surface: a novel aspect.

Diagnosing the placenta accreta spectrum is crucial to prevent morbidities and mortalities among women with the suspicion of this pathology. We aim to evaluate novel ultrasonography markers for these patients in diagnosing and predicting prognosis. This cross-sectional study was performed in a referral academic hospital. The population was composed of 51 pregnant women with a suspect of placenta accreta spectrum who had scheduled C-sections. Their primary information and past medical histories were documented. Then the ultrasonography markers, including the most bulging volume behind the bladder (area, perimeter, and volume), the Lacune (diameter, length, number, and surface of the largest lacuna obtained by multiplying the length by the width), the most considerable thickness of placenta on the cervix in patients with placenta previa, the most considerable thickness of the placenta behind the bladder, the Jellyfish sign, and sponge cervix were evaluated. Their comparison to the severity of the bleeding, the rate of the hysterectomy, and the following pathology of the placenta accreta spectrum were analyzed. The results showed that 17 (33.3%) of patients had severe bleeding (more than 2500 cc). The diameter, length, and surface of the largest lacunae limited to women with severe bleeding were 13.50 (5.5-21) mm, 20.50 (11-56) mm, 273.00 (60-1176) mm2, and they were 11.00 (5-24) mm, 16.25 (10-39) mm, and 176.25 (50-744) mm2 for women without severe bleeding (P value = 0.039, 0.027, 0.021). 13 (76.5%) women with severe bleeding had Jellyfish signs,16 (94.2%) had bulging on the cervix, and 10(58.8%) had a sponge cervix (P value = 0.046, 0.036, 0.006). Also, 34 (66.66%) patients needed hysterectomy. The diameter, length, and surface of the largest lacunae limited to women with hysterectomy were 12.00 (5-24) mm, 18.00 (11-56) mm, 231.00 (60-1176) mm2, and they were 9.00 (5-18) mm, 15.00 (10-28) mm, and 136.00(50-504) mm2 for women without hysterectomy (P value = 0.012, 0.070, 0.021). 24(70.6%) women with hysterectomy had Jellyfish signs, 29 (85.3%) of them had bulging on the cervix, and 15 (44.1%) had sponge cervix (P value = 0.05, 0.036, 0.028). The cut-off associated with the Lacunar surface was 163.5 mm2. Its sensitivity was 80%, and its specificity was 48% (P value = 0.021). The presence of single large lacunae could be a suitable predictive factor for bleeding in the placenta accreta spectrum; Moreover, there are some other US criteria, including the presence of a sponge cervix or the Jellyfish sign that are valuable predictive factors for negative outcomes for this spectrum, including hysterectomy.

Stock Price Reaction to Environmental, Social, and Governance News: Evidence from Brazil and Financial Materiality

This study explores the relationship between Environmental, Social, and Governance (ESG) practices and the market value of companies, with a focus on Brazil’s largest corporations. Recognizing the limitations of existing research tools for analyzing the impact of ESG factors, we introduce an innovative, open-source Dictionary of ESG Terms. This tool is designed to classify news content into the detailed categories established by the Sustainability Accounting Standards Board (SASB), thereby facilitating a nuanced analysis of ESG-related news and its subsequent effects on stock prices. Our analysis reveals that stock prices exhibit significant positive reactions to favorable ESG news and negative reactions to adverse ESG developments. Crucially, our findings underscore the discernment of investors, who appear to prioritize financially material ESG information over news bearing solely reputational or non-pecuniary significance. This distinction highlights the critical role of financial materiality in shaping market responses to ESG news. By providing empirical evidence from the Brazilian market, this study contributes to the broader discourse on ESG factors in corporate valuation. It offers practical tools and insights for investors, companies, and regulators aiming to better understand the complexities of ESG investment strategies. Through the application of our comprehensive ESG Dictionary, we shed light on the diverse dimensions of ESG impact, suggesting an approach to evaluate how ESG practices influence corporate market value in emerging economies.

Comparison of bioactive substances in novel-developed freeze-dried platelet-rich plasma (PRP) and activated normal PRP, and investigation of bioactive substance levels after long-term storage

IntroductionIn recent years, biotherapy in orthopedics has become widespread, and platelet-rich plasma (PRP) has been readily used to treat sports injuries and osteoarthritis. Production of freeze-dried PRP (PRP-FD) results in PRP that is in powder form, allowing it to be stored for long periods at room temperature. Using this technology, we have developed Valuable Platelet-Derived Factor Concentrate Freeze Dry (VFD). However, whether VFD contains sufficient levels of bioactive substances (BS) remains unknown and retains the same levels of BS during long-term storage. In this study, we examined whether VFD contains sufficient amounts of BS and whether they retain these BS levels during long-term storage. MethodsPeripheral blood was collected from 10 healthy men (mean ± SD: 46.5 ± 15 years old) and various BS, including transforming growth factor β (TGF-β), basic fibroblast growth factor (bFGF), epidermal growth factor (EGF), tissue inhibitors of metalloproteinases-1 (TIMP-1), interleukin-1 receptor antagonist (IL-1ra), matrix metallopeptidase-9 (MMP-9), and interleukin-6 (IL-6), were compared between VFD and normal PRP samples, including both leukocyte-rich PRP (LR-PRP) and leukocyte-poor PRP (LP-PRP). VFD was prepared using two rounds of centrifugation. LP-PRP and LR-PRP were activated by freezing and thawing before measurement. To evaluate the effects of long-term storage, the BS of VFD purified from five professional football players was compared between baseline and 1 year after storage. ResultsIn terms of the growth factors, the TGF-β and EGF levels were higher in LR-PRP than in VFD and LP-PRP (p < 0.05), while the bFGF levels were higher in VFD than in the LR-PRP and LP-PRP groups (p < 0.01). In terms of anti-inflammatory cytokines, the TIMP-1 level was lower in VFD than that in the other groups (p < 0.01), whereas the IL-1ra levels were higher in VFD than those in LP-PRP (p < 0.05) and lower than those in LR-PRP (p < 0.01). In terms of inflammatory enzymes and cytokines, the IL-1ra level was higher in VFD than that in LP-PRP (p < 0.05) and lower than that in LR-PRP (p < 0.01), whereas the IL-6 levels did not differ among the groups. Furthermore, the TGF-β, bFGF, TIMP-1, and IL-1ra levels were 5.61 → 3.38 (x103 pg/μL), 61.0 → 63.0 (pg/μL), 3.4 → 2.7 (x105 pg/μL), and 14.9 → 14.5 (x103 pg/μL) at baseline and 1 year later, respectively. No significant differences in the BS levels were observed between baseline and 1 year after storage. ConclusionsThe VFD samples prepared in this study exhibited higher levels of anti-inflammatory cytokines than LP-PRP and contained growth factor levels similar to LP-PRP and LR-PRP. In addition, the BS levels in VFD samples were maintained after one year of storage. These results suggest that VFD can be prepared and stored and may serve as a novel treatment strategy for sports injuries in high-risk groups, such as athletes.

Phytosulfokine alpha enhances regeneration of transformed and untransformed protoplasts of Brassica oleracea.

Phytosulfokine-α (PSK-α) is a disulfated pentapeptide (YIYTQ) acting as an intercellular signal peptide and growth factor. It was originally isolated from conditioned medium of asparagus mesophyll cell cultures in 1996 and later characterized as a hormone-like signal molecule with important roles in numerous processes of in vivo plant growth and development. It is currently becoming a valuable mitogenic factor in plant breeding and biotechnology due to its stimulatory effect on in vitro cell elongation, proliferation and differentiation. The focus of our work was to review current knowledge about the roles of PSK-α in plant biotechnology and to evaluate its influence on the regeneration of protoplasts of four Brassica oleracea cultivars (two cauliflower and two cabbage) cultured under two distinctive protocols and with different protoplast densities. Protoplast regeneration was studied due to its high value for plant genome editing, which is generally limited by the inefficient regeneration of treated protoplasts of numerous important plant genotypes. Our hypothesis was that the stress related to PEG-mediated protoplast transformation and the following decrease in viable protoplast density in culture could be alleviated by the addition of PSK-α to the culture medium. We therefore tested whether PSK-α could increase cell division at the early stages of culture (5 and 15 days after protoplast isolation) and stimulate the formation of microcallus colonies up to the 30st day of culture and to evaluate its influence on callus organogenesis leading to shoot regeneration. The PSK-α showed a strong stimulatory effect on untransformed protoplast regeneration already during the first days of culture, accelerating cell division up to 5.3-fold and the formation of multicellular microcallus colonies up to 37.0-fold. The beneficial influence was retained at later stages of regeneration, when PSK improved shoot organogenesis even if it was present only during the first 10 days of culture. The highest numbers of shoots, however, were regenerated when PSK was present during the first days of culture and later in solid shoot regeneration medium. Finally, the addition of PSK-α to PEG-transformed protoplasts significantly enhanced their division rate and the formation of microcallus colonies in selection media, up to 44.0-fold.
