Impact of Adherence to the Mediterranean Diet on Antioxidant Status and Metabolic Parameters in NAFLD Patients: A 24-Month Lifestyle Intervention Study.

The Mediterranean Diet (MedDiet) is recognized as a healthy dietary pattern. Non-alcoholic fatty liver disease (NAFLD) is characterized by the excessive accumulation of fat in the liver. To assess the antioxidant status in erythrocytes, plasma, and peripheral blood mononuclear cells (PBMCs) of NAFLD patients following a 24-month lifestyle intervention based on the MedDiet. Adult patients (n = 40; aged 40-60 years) diagnosed with NAFLD by magnetic resonance imaging were divided into two groups based on their adherence to the MedDiet. Consumption was assessed using a validated 143-item semiquantitative Food Frequency Questionnaire. Anthropometrics, biochemistry parameters, intrahepatic fat contents (IFC), antioxidants, and inflammatory biomarkers were measured in plasma and erythrocytes before and after the intervention. After the intervention, body mass index (BMI) and plasma levels of total cholesterol, low-density lipoprotein cholesterol (LDL-chol), triglycerides, malondialdehyde (MDA), and cytokeratin-18 (CK18) decreased, and high-density lipoprotein cholesterol (HDL-chol) increased. Participants with high adherence to MedDiet showed lower IFC, hepatic enzyme (AST, ALT, and GGT), glycemia, oxidase LDL (oxLDL) plasma levels, and erythrocyte MDA levels. Higher antioxidant activity (erythrocyte catalase-CAT, superoxide dismutase-SOD, glutathione peroxidase-GPx, glutathione reductase-GRd, and total glutathione-GSH as well as PBMCs-CAT gene expression) was observed in these patients, along with a reduction of PBMCs reactive oxygen species production and Toll-like receptor 4 (TLR4) expression. Inverse associations were observed between adherence to the MedDiet and BMI, glycemia, AST, IFC, and CK18 plasma levels and oxLDL, CAT, SOD, and GRd activities in erythrocytes. A significant linear regression was observed between adherence to the MedDiet and antioxidant score. Adherence to the MedDiet is associated with improved plasma and PBMC antioxidant and inflammatory biomarker profiles and high antioxidant defences in erythrocytes.

Effect of dendritic cell vaccine on blood redox status in patients with cervical cancer

Purpose of the study. The purpose of the study was to evaluate the dynamics of parameters of malondialdehyde (MDA), reduced glutathione and antioxidant enzyme activity in patients with advanced cervical cancer (CC) receiving anticancer treatment with dendritic cell vaccine (DCV).Patients and methods. Levels of malondialdehyde (MDA), reduced glutathione and antioxidant enzyme activity were studied in 27 patients aged 27–65 years with advanced primary and recurrent CC receiving chemotherapy (CT) with DCV or DCV as monotherapy at a dose of 10 million cells intradermally once a week. The same indices were studied in 20 healthy women from the comparison group.Results. The majority of patients showed the initial increase of MDA: in blood plasma by an average of 66.7 %, in red blood cells – statistically insignificant. After CT, MDA levels were elevated respectively in primary and recurrent patients in erythrocytes by 85.6 % and 96.4 % compared to donors, and by 53.9 % and 33.7 % compared to the initial values; the levels in plasma were elevated by 79.8 % and 57.1 % compared to donors. After 5–7 DCVs in combination with CT, MDA levels in erythrocytes and in blood plasma were similar to the donor values. MDA in erythrocytes of patients receiving DCV without CT decreased by 36 % compared to initial values. Oxidase activity of ceruloplasmin, initially increased by 34.2–57.1 %, normalized after DCV. Superoxide dismutase activity decreased by 16.7–27.3 % after 4–6 DCVs and normalized after 7 DCVs. Catalase activity, initially reduced by more than 40 %, remained 20–38 % lower than the norm at all stages of DCV. In this setting, hydrogen peroxide was probably inactivated by glutathione peroxidase whose activity was increased at all stages of treatment and decreased only when MDA was normalized after 7 DCVs.Conclusion. Administration of 5–7 DCVs in combination with CT or as monotherapy to patients with advanced CC normalizes parameters of free radical oxidation and antioxidant system of the blood.

Effect of Cadmium on Oxidative Stress Indices and Vitamin D Concentrations in Children

Heavy metal poisoning can have serious health consequences, including damage to the brain, kidneys, and other organs. Cadmium is a toxic heavy metal that can accumulate in the body over time and the exposure to this element has been linked to a variety of adverse health effects. Cadmium toxicity can lead to an imbalance in the cellular redox state and be a source of oxidative stress. On the molecular level, cadmium ions negatively affect cellular metabolism, including the disruption of energy production, protein synthesis, and DNA damage. The study has been carried out on a group of 140 school-age children (8 to 14 years old) inhabiting the industrialized areas of Upper Silesia. The study population was divided into two sub-groups based on the median concentration of cadmium in blood (0.27 µg/L): Low-CdB and High-CdB. Measured traits comprised blood cadmium levels (CdB) as well as a blood count and selected oxidative stress markers. This research study aimed to demonstrate a correlation between the impact of exposure to elevated cadmium concentrations in a population of children and certain markers of oxidative stress, and 25-OH vitamin D3 concentration. A negative correlation has been found between cadmium concentration and 25-OH vitamin D3 level, protein sulfhydryl groups content in blood serum, glutathione reductase activity, and lipofuscin and malondialdehyde levels in erythrocytes. The concentration of 25-OH vitamin D3 in the High-CdB group was decreased by 23%. The oxidative stress indices can be considered a valuable indicator of early Cd-toxicity effects to be included in the routinely-applied cadmium exposure monitoring parameters, allowing the evaluation of stress intensity to the cell metabolism.

Lepidium meyenii Walp (red maca) Supplementation Prevents Acrylamide-Induced Oxidative Stress and Liver Toxicity in Rats: Phytochemical Composition by UHPLC-ESI-MS/MS.

Lepidium meyenii Walp (red maca) is a high Andean plant cultivated since the Incas and has innumerable therapeutic properties. The study aims to identify its phytochemical composition using UHPLC-ESI-MS/MS, and evaluate its effects on acrylamide-induced oxidative stress. The lyophilized aqueous extract of red maca (LAqE-RM) was orally administered in doses of 1 and 2g/kg body weight for 4weeks. Malondialdehyde (MDA) levels in erythrocytes, brain, and liver, as well as hepatic levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were determined. Administration of acrylamide for 2 and 4weeks significantly increased (p < 0.001) MDA levels in erythrocytes, brain, and liver. However, LAqE-RM prevented (p < 0.001) an increase in MDA levels in all tissues studied. Likewise, the groups treated with LAqE-RM presented significantly (p < 0.001) lower levels of ALT and AST compared to the control. Treatment with LAqE-RM ameliorated the acrylamide-induced oxidative stress by reducing MDA levels in erythrocytes, brain, and liver and by lowering liver levels of ALT and AST in a dose-dependent manner. Twenty-five secondary metabolites were identified and characterized from LAqE-RM based on UHPLC mass spectrophotometry. These include carbolines, alkamides, fatty acids, and macamides, which are probably involved in their antioxidant protective role.

Promotion of the Glutathione Defense System From Aronia Berry Extract With Acute Aerobic Exercise: A Double-Blind Randomized Controlled Trial in Healthy Adults.

ObjectivesThis study aimed to examine the protective effects of Aronia melanocarna berry extract (ABE) supplementation against the acute aerobic exercise (AAE) – induced oxidative stress in healthy subjects.MethodsA 8-week, randomized, double-blind, parallel, placebo-controlled trial were conducted with 70 healthy middle-aged adults. We assessed a battery of AAE-induced oxidate stress biomarkers at pre-exercise, immediately post-exercise, and 30-min post-exercise. The AAE challenge model, as a treadmill exercise for 30 minutes at 60% VO2max, was applied to load oxidative stress at the end of the study. A linear mixed-effect model was used to test differences of safety and effectiveness between two groups and Pearson's correlation coefficients assessed the statistical association.ResultsThe AAE-induced time course-dependent oxidative stress was well-observed in the placebo group regarding the glutathione peroxidase activity and glutathione availability for antioxidant defense; and erythrocyte malondialdehyde, interleukin-6, and lactate levels for oxidative damage. Meanwhile, the ABE supplementation improved the glutathione defense system by increasing glutathione availability and glutathione peroxidase activity during exercise and post-exercise recovery periods. Age-and sex-related changes in selected hematological parameters, lipid peroxidation and erythrocytes osmotic fragility of Turkish Angora cats

This study was conducted to investigate the changes in selected hematological parameters, lipid peroxidation and osmotic fragility of erythrocytes in Angora cats depending on the age and gender. For this purpose, the blood samples were collected from 9 young and 14 adult cats which also classified as male (n=12) and female (n=11). Following hematological analysis, samples were washed with PBS by centrifugation and 10% hematocrit suspension was prepared from the erythrocytes pellet for osmotic fragility test. The concentration of malondialdehyde (MDA) was also measured from lysed erythrocyte to determine lipid peroxidation level. Red blood cells (RBCs), hemoglobin, and hematocrit were high in adults while mean corpuscular volume, mean corpuscular hemoglobin, and mean corpuscular hemoglobin concentration were high in young cats. Erythrocyte MDA level was also higher in adult cats than in young cats. There was no significance in these parameters between male and female cats. Findings of fragility tests were showed that erythrocytes of young and male cats were more susceptible to hypotonic NaCl solutions than those of adult and female cats, respectively. It was concluded that erythrocytes related parameters in Angora cats changed depending on the age rather than gender except stability of RBCs.

Oxidative Stress and Biochemical Parameters among Recovered COVID-19 Patients: A Case-control Study

Introduction: The Coronavirus Disease-2019 (COVID-19) pandemic has spread rapidly, infecting more than 194 million and killing more than 4 million people worldwide. Algeria has not escaped this scourge; according to World Health Organization (WHO), 162,155 confirmed cases and 4,063 deaths have been recorded from 3rd January 2020 to 26th July 2021. Recent studies have indicated the critical role of an altered immune system, and oxidative stress in the pathological process contributing to several complications during COVID-19 disease. Aim: To determine blood markers, oxidant/antioxidant status and biochemical parameters in patients highly recovered from COVID-19 and compare with those who have never contracted COVID-19; considered as controls. Materials and Methods: The present case-control study was conducted in Tiaret, Algeria, between May 2021 and June 2021. Thirty healthy volunteers who had never contracted COVID-19 and 16 volunteers who recovered from COVID-19 in the last six months were included in the study. Blood samples were taken after 8 to 12 hours of fasting, the blood markers and biochemical parameters were evaluated. The participant with chronic diseases (diabetes, hypertension, cardiovascular diseases, kidney disease) was excluded. Student’s t-test was performed for statistical comparison between the two groups. Statistical analysis was performed using Excel Microsoft 2010 software. Results. The control group consisted of 46.7% male (n=14) and 53.3% females (n=16). While, the case group consisted of 62.5% males (n=10) and 37.5% females (n=6). The plasma levels of Low Density Lipoprotein-Cholesterol (LDL-C), p-value=0.004** and creatinine increased very significantly in the cases compared to the controls. While, total cholesterol, p-value=0.04* and Glutamate Pyruvate Transaminase (GPT), p-value=0.03* increased significantly in the case group on comparision to the control group. On the other hand, erythrocyte Malondialdehyde (MDA) levels, p-value=0.009** increased very significantly in the case group compared to controls. The erythrocyte activity catalase decreased highly significantly in the case group compared to the controls. But erythrocyte Reduced glutathione (GSH) decreased very significantly in group cases compared to controls. Conclusion: The findings in the present study confirmed the persistence of metabolic alterations and oxidative stress in COVID-19 patients after recovery. Antioxidant supplementation is recommended to improve redox status and reduce oxidative stress after recovery

Parameters of Oxidative Stress, Vitamin D, Osteopontin, and Melatonin in Patients with Lip, Oral Cavity, and Pharyngeal Cancer.

Lip, oral cavity, and pharyngeal cancers (LOCP) constitute a group of rare neoplasms with unfavorable prognosis. So far, not much is known about the role of vitamin D and oxidative stress in the pathogenesis of LOCP in the European population. The aim of the study was to determine the concentrations of vitamin D, osteopontin, melatonin, and malondialdehyde (MDA) as markers of oxidative stress and/or inflammation, as well as the activities of antioxidant enzymes in the course of LOCP. The vitamin D, melatonin, and osteopontin concentrations in blood serum, the MDA levels in erythrocytes and blood plasma, and the activities of superoxide dismutase (SOD-1), catalase (CAT), and glutathione peroxidase (GPx) in erythrocytes were measured in blood samples taken from 25 LOCP patients of middle age (YCG), 20 LOCP elderly patients (OCG), and 25 healthy middle-aged volunteers. In both cancer groups, decreases in vitamin D and CAT, as well as increases in osteopontin and blood plasma MDA, were observed. An increase in GPx activity in YCG and a decrease in melatonin level in OCG were found. The results indicate the vitamin D deficiency and disturbed oxidant-antioxidant homeostasis in LOCP patients. Osteopontin seems to be associated with LOCP carcinogenesis and requires further research.

Serum levels of anti-carbonic anhydrase antibodies and erythrocyte oxidative stress markers in endometriosis

Aim: To evaluate the serum levels of anti-carbonic anhydrase I-II antibodies and erythrocyte oxidative stress markers in endometriosis. Method: This case-control laboratory investigation was performed in the obstetrics and gynecology department of a tertiary center. Serum anti-carbonic anhydrase I and II antibodies and erythrocyte oxidative stress markers (superoxide dismutase, malondialdehyde, glutathione peroxidase and catalase) were compared with control group (n = 30) in the endometriosis group (n = 33). Correlation between carbonic anhydrase autoantibodies and oxidative stress markers were tested. Results: Serum levels of anti-carbonic anhydrase II antibodies were found to be significantly increased in the endometriosis group compared to controls. The erythrocyte antioxidant enzyme activities (superoxide dismutase, glutathione peroxidase and catalase) and malondialdehyde levels in erythrocytes were increased in endometriosis group; but only glutathione peroxidase activity and malondialdehyde levels were significantly higher in endometriosis group. No correlation was detected between anti-carbonic anhydrase antibodies and oxidative stress markers. Conclusions: Our results indicate that erythrocyte oxidative stress and anti-carbonic anhydrase antibodies may be involved in the pathophysiology of endometriosis.

The Level of Reactive Carbonyl Derivatives of Proteins, Methylglyoxal, and Malondialdehyde in Rats Experiencing Chronic Unpredictable Moderate Stress

BACKGROUND: Prolonged exposure to stress is known to change homeostasis and can lead to the development of psychopathological disorders, including depression. Exposure to stress factors contributes to the emergence of reactive oxygen species in the brain. The data characterizing changes of the oxidative metabolism under conditions of chronic stress are insufficient, which determine the relevance of the research in this direction. The determination of reactive carbonyl derivatives of proteins (RCDP), methylglyoxal (MG), and malondialdehyde (MDA) levels can be considered an informative method for studying the dynamics of oxidative metabolism.&#x0D; AIM: The aim of the study was to study the RCDP, MG, and MDA in red blood cells and blood plasma in rats experiencing chronic unpredictable moderate stress (CUMS).&#x0D; METHODS: A total of 20 male outbred rats weighing 450–500 g were used in the study. The animals were divided into two groups of ten rats each using randomized selection. The experimental group of animals was exposed to the impact of diverse stress factors, according to the CUMS model for 21 days. The control group of animals remained in standard conditions and did not undergo CUMS model. Blood sampling was performed twice for all rats the day before the experiment started and on 21st day to determine the level of RCDP, MG, and MDA in their erythrocytes and blood plasma.&#x0D; RESULTS: The study revealed a significant decrease of RCDP (p = 0.007) and MDA levels in erythrocytes (p = 0.013), and MDA level in blood plasma (p = 0.005) of rats of the main experimental group. The control group showed a significant increase of RCDP level in plasma (p = 0.008), a two-fold increase of MDA in erythrocytes (p = 0.05) with a decrease of MDA in plasma (p = 0.011). A tendency for MG increase was noted in erythrocytes of rats in both the main group and the control group.&#x0D; CONCLUSION: The early stages of CUMS were accompanied by the violation of oxidative metabolism with the emergence and accumulation of RCDP and MDA in the blood of animals. The decreased emergence of RCDP and MDA in the later stages can be associated with an excess of oxidized substrates in the blood of animals of the experimental group. The revealed trend toward MG growth in animals of both groups suggests its increased synthesis in erythrocytes, which, in our opinion, should be considered a negative factor affecting the deformability of red cells. In turn, a violation of the deformability of red blood cells leads to a violation of microcapillary blood flow, which contributes to the development of hypoxic damage in different tissues. Thus, the obtained data confirm the different directions of changes in RCDP and MDA levels in the blood of animals experiencing CUMS or acute stress. Our results prove the need for further research on changes in oxidative metabolism in chronic stress disorders.

Effect of turmeric and ginger supplementation on immunity, antioxidant, liver enzyme activity, gut bacterial load and histopathology of broilers

Day-old broiler chicks (182) were distributed randomly to 7 treatments with 2 replicates. Treatments were T1 (control), basal diet; T2, basal diet + turmeric powder (TP) (0.5% of basal diet); T3, basal diet + TP (1% of basal diet); T4, basal diet + ginger powder (GP) (0.5% of basal diet); T5, basal diet + GP (1% of basal diet); T6, basal diet + TP + GP (0.5% TP + 0.5% GP); T7, basal diet + TP + GP (1% TP + 1.0% GP). The experiment was continued for 35 days. Immunity, antioxidant, liver enzyme activity, gut bacterial load and histopathology of broilers were conducted at fifth week of age. Higher cellular response against PHA-P was recorded in T3 and T7. Higher antibody titre against SRBC was recorded in T3. The weight of lymphoid organs did not differ significantly. Significantly higher erythrocyte malondialdehyde (MDA) level was recorded in T1. Significantly higher alanine amino transferase (ALT) levels were found in T1 and T7. Significantly higher aspartate amino transferase (AST) level was found in T1. Higher total bacterial count and lower E. coli count were recorded in group T3 and lower total bacterial count was recorded in T7. In group T1, liver showed mild congestion to mild cellular swelling and varying degree of vacuolar degeneration. From this study, it may be concluded that supplementation of 1% turmeric in ration either alone or in combination with 1% ginger improved the immunity, antioxidant status and gut health of broilers.

Composición química por HPLC-ESI-QTOF-MS/MS: efectos estrogénicos y antioxidantes de extractos de Mangifera indica L. cv. “Kent” en ratas ovariectomizadas

The chemical composition of Mangifera indica L. cv. Kent leaves was determined by HPLC-ESI-QTOF-MS/MS. Polyphenolic compounds characterized as benzophenone derivatives were the main components found in extracts (1, maclurin 3-C-(2-O-galloyl)-D-glucoside isomer; 2, maclurin 3-C---D-glucoside; 3, iriflophenone 3-C---D-glucoside; 5, maclurin 3-C-(2,3-di-O-galloyl)---D-glucoside; 6, iriflophenone 3-C-(2-O-galloyl)---D-glucoside; 7, methyl-iriflophenone 3-C-(2,6-di-O-galloyl)---D-glucoside) and xanthones (4, mangiferin and 8, 6-O-galloyl-mangiferin). The estrogenic and antioxidant effects of aqueous extracts from Mangifera indica L. cv. Kent leaves on ovariectomized rats were determined by uterotrophic assay and malondialdehyde (MDA) levels in erythrocytes, bone, liver, and stomach. We conclude that the polyphenolic compounds from extracts act as exogenous antioxidant agents against oxidative damage in ovariectomized rats.

Effect of Orally Administered Glutathione Peroxidase Mimetic towards Glutathione and Malondialdehyde Blood Level and Otoacoustic Emissions Result in Soldiers with Acoustic Trauma Risk Caused by Howitzer 105 Artillery Weapon BLAST

Excessive noise exposure may cause mechanical and metabolic (oxidative stress) to the inner ear structure (cochlea), resulting in acoustic trauma (TA). Oxidative stress which caused by increasing of free radical ROS/RNS in the organs of Corti, causing apoptosis and necrosis of cochlear hair cell. Oxidative stress was characterized by increasing of malondialdehyde (MDA), decreasing of glutathione peroxidase (GPx) enzyme, and abnormal otoacoustic emission value (refer). Defense mechanism toward oxidative stress was mediated by endogenous antioxidant enyzme (superoxide dismutase/SOD, catalase/CAT, and GPx). Cellular mechanism of glutation peroxidase mimetic was similar as GPx.    Aim of this study was to determine the impact of glutathione peroxidase mimetic to glutathione peroxidase and malondialdehyde level in blood, also otoacoustic emission value on soldiers with risk of TA due to explosion of Howitzer 105 artillery weapon. The design of this study was clinical trial pre and post design, randomized, double blind and placebo controlled, on 34 new recruit soldiers in Artillery Academy of Indonesian Army, during the soldiers were trained to firing Howitzer 105 artillery weapon, from 7th-10th July 2014 in Cimahi and Batujajar. Subjects were divided into 2 groups, the exposure group (group given treatment) and control group. Exposure group was given glutathione peroxidase mimetic (Ebselen SPI 1005) 200mg, orally once daily during the training, meanwhile control group was given. Malondialdehyde and Glutathione Peroxidase level in blood, pure tone audiometry and otoacoustic emission value (DPOAEs) was measured from those two groups, before and after firing training. The data was analyzed using parametric and non parametric, with Number Needed to Treat (NNT, 95%CI) and significance value (p&lt;0.05).    The results of this study showed the number of acoustic trauma events in controle group based on pure tone audiometry test was 23.5%, exposure group was 0%, and based on DPOAEs test the controle group showed 47.1% abnormal/refer, exposure group showed 100% normally/pass, and this difference stastically significance (p&lt;0.051). The group which was given Ebselen SPI 1005 showed increasing 82.4% erythrocyte GPx level (NNT, 95%CI=1.889 [1.159-3.016]; p=0.004), showed increasing 88.2% plasma GPx level (NNT, 95%CI=1.417 [0.970-1.775]; p&lt;0.001), decreasing 100% erythrocyte MDA level (NNT, 95%CI = 2.125 [1.335-3.987]; p=0.01),  showed decreasing 94.1% plasma MDA level (NNT, 95%CI=2.125 [1.29-3.904]; p=0.01), and resulting 100% in normal/pass DPOAEs value (NNT, 95%CI = 2.125 [1.335-3.987]; p=0.01). This study concluded that giving Ebselen SPI 1005 to soldiers with risk of acoustic trauma during the firing Howitzer 105 artillery weapon, increasing GPx level in blood, decreasing MDA level in blood, and resulting normally DPOAEs value (pass)

Investigation of oxidative stress in patients with alopecia areata by measuring the levels of malondialdehyde and ceruloplasmin in the blood.

IntroductionAlopecia areata (AA) is a chronic, inflammatory and autoimmune disease, presenting with non-scarring hair loss. Although the precise etiopathogenesis of AA remains unknown, oxidative stress is thought to play a role.AimTo investigate the role of oxidative stress in AA by measuring the levels of plasma and erythrocyte malondialdehyde (MDA) and the ceruloplasmin (CER) in serum.Material and methodsThe study included 24 AA patients and a control group consisting of 24 age- and sex-matched healthy volunteers. The levels of MDA and CER were measured and compared between groups.ResultsPlasma MDA levels were significantly higher (p < 0.05) in patients with AA compared with controls. No significant difference was observed in MDA erythrocyte levels (p = 0.990) between the study group and the control group. Ceruloplasmin level was higher in the AA group, but this increase was not statistically significant (p = 0.156).ConclusionsPatients with AA displayed significant plasma MDA levels, which could lead to damage in erythrocytes exposed to high concentrations of free radicals. These results demonstrate the presence of an imbalance in the oxidant-antioxidant system and support the concept of a possible role of oxidative stress in AA etiopathogenesis.

Antiplasmodial and antioxidant activities of chloroform extract of Ganoderma lucidum fruit body in Plasmodium berghei-infected mice

From our previous study extract of the fruit bodies of Ganoderma lucidum possessed promising curative potential against Plasmodium berghei in mice. Thus, we hypothesized that infection with chloroquine-sensitive Plasmodium berghei together with crude chloroform extract (CCE) of G. lucidum, at 250 mg/kg body weight would enhance various aspects of antiplasmodial activities in mice. Mice were treated with CCE or chloroquine (CQ) for seventy-two hours prior to infection with P. berghei and monitored closely until signs of death were observed in untreated mice and then sacrificed. Parasitaemia, hepatic function, and oxidative stress markers analyses were performed at the end of the experiment. CCE-treated mice displayed lower parasitemia compared with untreated mice. Plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST) and gamma-glutamyltranspeptidase (γ-GT) activities; and the erythrocyte malondialdehyde (MDA) level were significantly (p<0.0001) lower in the extract and CQ-treated mice. Erythrocyte antioxidant enzymes (GPx, GST, SOD and G6PDH) activities were significantly (p<0.0001) higher in CCE-treated mice compared with CQ-treated and untreated control. Though chloroquine treatment reduced parasitemia and MDA considerably compared with extract, there was reduced production of the antioxidant defense enzymes. This study shows that CCE of G. lucidum reduces parasitemia and improves the attendant consequences of Plasmodium berghei-malarial infection in mice.

The Effect of Chitosan on the Erythrocyte Antioxidant Potential of Lead Toxicity-Induced Rats.

In the present study, the effects of chitosan on erythrocyte malondialdehyde (MDA) and glutathione (GSH) levels and glutathione peroxidase (GSH-Px), glutathione reductase (GR), and glucose-6-phosphate dehydrogenase (G6PDH) enzyme activities in lead toxicity-induced rats were investigated. Twenty-eight male Wistar albino rats were divided into four groups of control (C), lead group (Pb group), lead+chitosan group (Pb+CS group), and chitosan group (CS group). Lead groups were administered 50mg/kg lead acetate intraperitoneally (ip) for 5days and chitosan groups were administered 200mg/kg chitosan for 28days via gavage. At the end of the study, lead levels were measured in the blood; MDA and GSH levels and GPx, GR, and G6PDH activities were measured in the erythrocyte. It was determined that, in parallel with the increase of full blood lead levels in the Pb group, erythrocyte MDA levels increased significantly, while GSH levels and GSH-Px, GR, and G6PDH activities decreased when compared to those in the C and CS groups (p˂0.05). There was a statistically significant decrease in lead and MDA levels and GSH level and GSH-Px activity increased (p˂0.05) in the Pb+CS group, where chitosan was administered as a protective agent in addition to lead, when compared to the Pb group. There were no differences between the Pb+CS group and the other three groups based on GR and G6PDH activities (p˃0.05). No statistically significant difference was found between the C and CS groups based on the parameters of analysis (p˃0.05). The findings of the present study demonstrated that lead increased oxidative stress by increasing free radical production in erythrocytes, and chitosan was effective in removing the lead from the circulation and enforced the antioxidant defense system.

Cytoprotective Effects of the Red Marine Alga Chondrus canaliculatus Against Maneb-Induced Hematotoxicity and Bone Oxidative Damages in Adult Rats

The current study aimed at evaluating the ability of a mineral and antioxidant-rich extract from Chondrus canaliculatus to improve maneb (MB)-induced toxicity in adult rat. The animals were divided into four groups: group 1 used as a control group, group 2 received MB, group 3 received MB + C. canaliculatus extract, and group 4 received only the algal extract. MB, a Mn-containing ethylene-bis-dithiocarbamate fungicide, induced oxidative stress damages, mineral perturbations in the plasma, urine, and bone, and genotoxicity in rats. Hematological analysis revealed in the MB-treated group a disruption in the number of red blood cells, platelets, and white blood cells associated with a striking genotoxicity. Interestingly, a significant increase in malondialdehyde and advanced oxidation protein product levels in erythrocytes and bones were found. On the other hand, an impairment of the antioxidant status in both tissues was occurred. Along, our results revealed that MB injection caused a striking drop and disruption in bone's mineral rates, especially calcium and phosphorus. These biochemical results were in accordance with the histological and molecular changes. However, co-treatment with C. canaliculatus extract showed, for the first time, that this alga was effective against MB-induced hematotoxicity, genotoxicity, and oxidative stress in the blood and bone and maintained osteomineral metabolism and bone histo-architecture. Such observations might be explained by the strong in vitro antioxidant and antibacterial activities exhibited by the alga, as well as by its high levels in several minerals: calcium, phosphorus, sodium, potassium, magnesium, iron, and zinc.

Antioxidant and Protective Effects of Schinus terebinthifolius Raddi Against Doxorubicin-Induced Toxicity.

Doxorubicin is an anticancer drug whose toxic effects on non-cancer cells are associated with increased oxidative stress. This study investigated the chemical composition, antioxidant activity of the methanolic extract of Schinus terebinthifolius Raddi leaves (MESL) as well as effects against doxorubicin-induced toxicity in human erythrocytes, K562 human erythroleukemia cells, and mouse hearts. The chemical composition indicated the presence of phenolic compounds, flavonoids, tannins, and ascorbic acid. MESL showed antioxidant activity by scavenging free radicals and inhibiting hemolysis and lipid peroxidation in human erythrocytes incubated with an oxidizing agent, and was able to increase the enzymatic activity of superoxide dismutase and glutathione peroxidase in human erythrocytes, without influencing the activity of enzyme catalase. The increase of oxidative hemolysis and malondialdehyde levels in erythrocytes incubated with doxorubicin was reduced by treatment with MESL. The cytotoxic activity of doxorubicin in erythroleukemia cells treated with MESL was unmodified. Additionally, the extract protected mice against the doxorubicin-induced cardiotoxicity. In conclusion, the MESL exhibits antioxidant activity, reducing doxorubicin-induced oxidative stress without changing the anticancer action of the drug, and protects against doxorubicin-induced cardiotoxicity. Hence, these findings suggest that these effects are via anti-oxidative by inhibiting free radicals, decreased oxidative stress, and increased antioxidant enzyme activity.

Protective effects of Teucrium polium aqueous extract and ascorbic acid on hematological and some biochemical parameters against carbon tetrachloride (CCl4) induced toxicity in rats

Herbal drugs play a crucial function to protect organisms from the toxic effect of some compounds. The present study was designed to evaluate the protective effects of Teucrium Polium (TP) aqueous extract and vitamin C (Vit C) against carbon tetrachloride (CCl4) which induced toxicity in rats. Male albino Wistar rats were divided into six groups: group I was used as controls, group II received CCl4 in olive oil (0.5ml/kg) by gavage, and group III received CCl4 in olive oil (0.5ml/kg) by gavage after three days of receiving TP (5g/l), orally, for seven days; Group IV received TP (5g/l) alone, orally, for seven days, Group V received CCl4 in olive oil (0.5ml/kg) by gavage after three days of receiving Vit C (250mg/kg) by intramuscular injection and Group VI received Vit C (250mg/kg) alone by intramuscular injection, for day seven. Some biochemical and hematological parameters were investigated. Our results showed that the administration of CCl4 caused hepatotoxicity as monitored by the significant increase in the levels of hepatic markers enzymes (lactate dehydrogenase (LDH), Alkaline phosphatase (PAL), total bilirubin (TB) and Gamma glutamyl transferase (γGT) levels) and a decrease in hematological parameters such as white blood cell (WBC), red blood cell count (RBC), mean corpuscular hemoglobin concentration (MCHC) and blood platelet count (PLT). Treatment with TP or Vit C appeared to be effective against hematotoxic and the liver changes induced by CCl4, as evidenced by the improvement of the parameters cited above. The CCl4-treated group demonstrated a significant increase of malondialdehyde (MDA) levels in erythrocytes thus causing a reduction in antioxidant defense system. Pretreatment with TP or Vit C improved the biochemical analyses, hematological parameters, and antioxidant defense system.
