Does the peroral chromium administration in young Hariana calves reduce the risk of calf diarrhea by ameliorating insulin response, lactose intolerance, antioxidant status, and immune response?

BackgroundThe reduction in insulin sensitivity during rumen development may predispose dairy calves towards lactose intolerance, which could be the reason behind neonatal calf diarrhea (NCD). Chromium (Cr) results in a range of effects when fed to ruminants, but most studies have shown improved insulin sensitivity. The aim of this study was to determine the effect of Cr supplementation on insulin sensitivity, lactose intolerance, diarrhoea and antioxidant, and immune response in young Hariana calves. MethodsA total of 20 milk-fed Hariana calves were randomly assigned to 1 of 2 treatments, each consisted of 10 calves: (1) a control group without supplemental Cr and (2) a 0.15 mg Cr as Cr-picolinate (CrPic)/kg BW0.75 supplemented group (Cr0.15). ResultsA more rapid glucose disappearance with unaltered insulin kinetics during intravenous glucose tolerance test (IVGTT) and oral lactose tolerance test (OLTT) indicates greater insulin sensitivity in Cr supplemented calves. Better insulin sensitivity in Cr supplemented calves was further confirmed by higher values of the quantitative insulin sensitivity check index (QUICKI), revised quantitative insulin sensitivity check index (RQUICKI) and insulin receptor substrate-1 (IRS-1) and lower (P < 0.05) values of homeostasis model assessment-insulin resistance (HOMA-IR) and glucose-to-insulin ratio in Cr supplemented calves during IVGTT. Cr supplementation resulted in a lower (P < 0.05) serum cortisol concentration, whereas serum non-esterified fatty acid (NEFA) concentrations during IVGTT did not differ among the groups. The rise in serum glucose concentrations within 2 h post lactose infusion during OLTT peaked at more than twice the basal glucose concentration, therefore calves were not considered as lactose intolerant. Within monthly blood samples, concentrations of serum insulin were similar among treatments, whereas the Cr supplemented group had lower (P < 0.05) serum glucose concentration and glucose-to-insulin ratio compared with the control group. No treatment differences were detected in the biomarkers of antioxidant status and immunity. Serum Cr concentrations were higher (P < 0.05) in Cr supplemented calves while concentrations of other studied minerals were remained unaltered. The incidence, duration of diarrhea, and faecal score were better (P < 0.05) in calves fed on Cr supplemented diet whereas, no treatment effect was observed on average daily gain (ADG). During the study period, no calves died, and no calves were found to have pneumonia, navel or joint disease. ConclusionFeeding a Cr-supplemented diet improved insulin sensitivity and reduced the risk of diarrhoea in milk-fed young calves, but had no or minimal effects on lactose intolerance, antioxidant status, immune response, and growth performance.

Hepatoprotective effect of Hydro-alcohol extract of Mimusops elengi root against antitubercular drug-induced hepatotoxicity in rats

Introduction: Nowadays, different chemicals and drugs are causing liver injuries including first-line antitubercular drugs (ATDs). There is a need to find safe and potent moieties of hepatoprotective drugs against liver diseases from different natural resources including medicinal plants. So, the current study aimed to evaluate the hepatoprotective property of Mimusops elengi root. Methodology: Mimusops elengi root extract (200-400mg/kg) was evaluated in an induced hepatotoxicity model of oxidative stress in Wistar rats by ATDs orally for 14 days. Markers indicating oxidative stress and hepatic damage such as serum transaminase (SGOT / SGPT), and alkaline phosphatase (ALP) were measured. Biomarkers of antioxidant status, superoxide dismutase, catalase, glutathione reductase, and marker of lipid peroxidation, were evaluated using standard procedures. The hematological lipid profile, bilirubin, glucose, and histopathological examination were also assessed. Results: intoxication with ATDs markedly reduced the hematological indices and elevated the biochemical enzyme markers (SGOT, SGPT, ALP p&lt;0.001) and lipid profile (p&lt;0.001), decreased and glucose was elevated. However, pretreatment with Mimusops elengi root extract significantly (p&lt;0.001, p&lt;0.01) improved this alteration and sustained the antioxidant potential. The Histopathological and biochemical data support hepatoprotective action. Conclusion: The results of the current study reinforced the extract of Mimusops elengi possesses significant hepatoprotective and antioxidant activity against ATDs- induced hepatotoxicity.

Dietary Strawberries Improve Biomarkers of Antioxidant Status and Endothelial Function in Adults with Cardiometabolic Risks in a Randomized Controlled Crossover Trial.

Strawberries, a popularly consumed berry fruit, are rich in bioactive compounds with antioxidant effects. In this study, we examined the effects of two dietary achievable doses of strawberries on the antioxidant status and biomarkers of endothelial function in adults with features of metabolic syndrome and a confirmed low baseline of fruit and vegetable intake. In a 14-week randomized controlled crossover study, participants were assigned to one of three groups for four weeks separated by a one-week washout period: control powder, one serving (low dose: 13 g strawberry powder/day), or 2.5 servings (high dose: 32 g strawberry powder/day). Blood samples and health data were collected at baseline and at the end of each four-week phase of intervention. Thirty-three participants completed all three phases of the trial. Significant increases were observed in serum antioxidant capacity and superoxide dismutase activity as well as decreases in lipid peroxidation after both low and high dose strawberry phases when compared with the control phase. Significant decreases were also observed in soluble vascular cell adhesion molecule-1 and tumor necrosis factor-α with the high dose strawberry phase. These data confirm that consuming strawberries for four weeks significantly improves antioxidant status, endothelial function, and inflammation in adults with cardiometabolic risks.

Flaxseed Oil Supplementation Augments Antioxidant Capacity and Alleviates Oxidative Stress: A Systematic Review and Meta-Analysis of Randomized Controlled Trials.

Objective Studies have reported controversial findings regarding the flaxseed oil effect on antioxidant status biomarkers. The present meta-analysis aimed to determine the impact of flaxseed oil on the serum level of biomarkers of oxidative stress. Methods A systematic search was conducted up to November 2020 on PubMed, Embase, Web of Science, Scopus, and Cochrane Central Library. Random-effects model was employed to perform meta-analysis. Subgroup analysis was carried out to determine the effect across different ranges of dosages and durations. Results Eight trials were included with a total sample size of 429 individuals with a mean age range of 25 to 70 years. The results indicated that flaxseed oil supplementation led to a significant decrease in malondialdehyde (MDA) levels (SMD: −0.52 μmol/L; 95% CI: −0.89, −0.15; P=0.006, I2 = 71.3, P < 0.001) and increase in total antioxidant capacity (TAC) levels (WMD: 82.84 mmol/L; 95% CI: 19.80, 145.87; P=0.006, I2 = 92.7, P < 0.001). No significant effect was observed on glutathione (GSH). Conclusion Our findings revealed that flaxseed oil supplementation might play a beneficial role in the reinforcement of the antioxidant defense system and amelioration of oxidative stress in adults.

Spirulina supplementation as an adjuvant therapy in enhancement of antioxidant capacity: A systematic review and meta-analysis of controlled clinical trials.

Spirulina, a type of blue-green algae, is used as an adjuvant treatment of metabolic and inflammatory diseases. Evidence about the effects of spirulina on antioxidant system are conflicting. Thus, this quantitative review aimed to summarise the effects of spirulina administration on antioxidant status biomarkers. Systematic searches were conducted using the PubMed/Medline, Scopus, Web of Science and EMBASE, up to May 2021. Random effect analysis was applied to perform meta-analysis. Subgroup analyses and multivariate meta-regression were performed to find heterogeneity sources. Quality assessment was conducted using Cochrane Collaboration's tool. Trim and fill analysis were also carried out in case of the presence of publication bias. A total of nine articles that enrolled 415 subjects were included in the present meta-analysis. Obtained findings exhibited that spirulina supplementation had marginal significant effect on total antioxidant capacity (TAC) (SMD=0.49; 95% CI: -0.001, 0.98; P=.05) and superoxide dismutase (SOD) activity (SMD=0.72; 95% CI: -0.03, 1.46; P=.06), while did not affect glutathione peroxidase (GPx) activity (SMD=0.27; 95% CI: -0.23, 0.77; P=.29). Spirulina consumption may exert beneficial effects on enhancement of antioxidant system. A marginal significant increasing effect on TAC and SOD activity were found by spirulina administration. However, it did not affect GPx activity.

The association of vitamin D with survival in colorectal cancer patients depends on antioxidant capacity

Vitamin D plays a role in detoxifying free radicals, which might explain the previously reported lower mortality in colorectal cancer (CRC) patients with higher vitamin D concentrations. We aimed to assess whether the associations of 25-hydroxyvitamin D [25(OH)D] with prognosis in CRC patients differ by total thiol concentration (TTC), a biomarker of antioxidant capacity. CRC patients who were diagnosed from 2003 to 2010 and recruited into a population-based study in southern Germany (n=2,592) were followed over a period of 6 y. 25(OH)D and TTC were evaluated from blood samples collected shortly after CRC diagnosis. Associations of 25(OH)D with all-cause and CRC mortality according to TTC were estimated using multivariable Cox proportional hazards regression. There was a weak positive correlation between 25(OH)D and TTC (r=0.26, P<0.001). 25(OH)D was inversely associated with mortality among patients in the lowest and middle TTC tertiles, but no associations were found among patients in the highest TTC tertile (P-interaction=0.01). Among patients in the lowest/middle TTC tertiles, those in the middle and highest (compared with lowest) 25(OH)D tertiles had 31% and 44% lower all-cause mortality (P<0.001) and 25% and 45% lower CRC mortality (P<0.001), respectively. However, in the highest TTC tertile, 25(OH)D was not associated with all-cause (P=0.638) or CRC mortality (P = 0.395). The survival advantages in CRC patients with adequate vitamin D strongly depend on antioxidant capacity and are most pronounced in cases of low antioxidant capacity. These findings suggest that TTC and other biomarkers of antioxidant status may be useful as the basis for enhanced selection criteria of patients for vitamin D supplementation, in addition to the conventional judgment based on blood 25(OH)D concentrations, and also for refining selection of patients for clinical trials aiming to estimate the effect of vitamin D supplementation.

The Effects of 6-Week Supplementation with Multicomponent Herbal Extract on Exercise Performance, Antioxidant Status and Telomere Length, and Self-Reported Side Effects in Healthy Men: A Randomized Controlled Pilot Trial

The main aim of this study was to examine the effects of medium-term supplementation with an eight-herbs extract on running performance, biomarkers of antioxidant status and telomere length, and self-reported outcome measures of safety events in healthy men. Ten healthy young men (age 23.1±3.2 years, weight 73.7±9.9kg, and height 179.4±8.0cm) volunteered to participate in this randomized controlled trial. The participants were allocated in a double-blind cross-over design to receive either an eight-herbs extract or placebo during a 6-week intervention period. Two-way mixed analysis of variance (treatment vs. time interaction) revealed no significant differences for exercise performance outcomes and telomere length between groups (P&gt;0.05). Compared with placebo, P-DNA provoked a significant rise in serum total antioxidant capacity (316.0±183.4µmol/mL at baseline; 792.7±68.1µmol/mL at follow-up in the eight-herbs extract group vs. 298.1±90.7µmol/mL at follow-up in the placebo group; P&lt;0.001), and less reduction in serum superoxide dismutase levels at follow-up (150.4±5.1IU/mL at baseline; 145.5±3.0IU/mL at follow-up in the eight-herbs extract group vs. 139.3±3.3IU/mL at follow-up in the placebo group; P=0.01). No participant reported any side effects of either intervention. The eight-herbs extract can thus be recommended as a well tolerated dietary supplement that can enhance antioxidant capacity in healthy men. This trial was registered at ClinicalTrials.gov (NCT04263246).

Effect of synbiotic bread containing lactic acid on glycemic indicators, biomarkers of antioxidant status and inflammation in patients with type 2 diabetes: a randomized controlled trial

BackgroundThe aim of this randomized, double-blinded, controlled trial was to investigate the effect of daily consumption of a synbiotic bread containing lactic acid on glycemic status, antioxidant biomarkers and inflammation in patients with type 2 diabetes (T2D).MethodsT2D patients, aged 20 to 60 years, were randomly assigned to consume synbiotic + lactic acid (n = 30), synbiotic (n = 30), lactic acid (n = 30), or control (n = 30) bread for 8 weeks. Patients consumed bread 3 times a day in a 40 g package for a total of 120 g/day. Glycemic status, antioxidant capacity, and serum hs-CRP were assessed before and after the intervention.ResultsOf a total of 120 patients that were included in the study, 100 completed the trial. In the adjusted analysis, it was found that consumption of synbiotic + lactic acid bread caused a significant decrease in HbA1c compared to the control bread (− 0.41 ± 0.33 vs 0.004 ± 0.10%, respectively; P < 0.001), while it significantly increased serum superoxide dismutase (SOD) (0.87 ± 1.14 vs. 0.18 ± 0.85 mmol/L, P = 0.02). Also, changes in glutathione peroxidase (GSH-Px) were significantly higher following the consumption of synbiotic + lactic acid bread than lactic acid bread. However, it had no significant effect on fasting plasma glucose, serum insulin, and total antioxidant capacity.ConclusionOverall, daily consumption of a synbiotic bread containing lactic acid for 8 weeks had beneficial effects on HbA1c, SOD, and GSH-Px among T2D patients.Trial registration This study was registered in Iranian Registry of Clinical Trials with number: IRCT201505242709N33 (Registration date: 2015-11-23, http://www.irct.ir/trial/2544)

SOME BLOOD BIOCHEMICAL, ANTIOXIDANT BIOMARKERS, LIPID PEROXIDATION, PRODUCTIVE PERFORMANCE AND CARCASS TRAITS OF BROILER CHICKS SUPPLEMENTED WITH ALPINIA GALANGAL RHIZOMES EXTRACT DURING HEAT STRESS

One thousand one-day-old Cobb broilers were randomly allotted to four dietary groups to study the influences of various levels of nutritional supplementation of Alpinia galangal rhizomes extracts (AGRE) on blood biochemical components, antioxidant status biomarkers, lipid peroxidation, productive performance, and carcass traits of broilers beneath summer circumstances. The control set was fed a standard diet free of AGRE; the trial groups obtained the standard diet besides 250, 500, and 750 mg AGRE /kg. The trial time continued for six weeks. The obtain results showed that, in groups, 500 AGRE and 750 AGRE, plasma globulin, albumin, and total protein were significantly augmented. However, all levels of AGRE supplementary diets revealed significantly decreased in the plasma total cholesterol, triglycerides, low-density lipoproteins, and total lipids. Total antioxidant capacity, glutathione S -transferase, superoxide dismutase, catalase, and glutathione peroxidase were significantly augmented by supplementing nutritional AGRE, respectively. In contrast, malondialdehyde was lowered with rising AGRE levels. Chickens of group 750 AGRE had the lowest mortality and also, the most effective feed conversion ratio and body weight gain. Percentage of dressing, breast, and thigh significantly augmented for chicks consumed different levels of AGRE. Also, the group fed on AGRE recorded the highest abdominal fat compared with control group. Additional, liver, heart and gizzard weights were increased significantly in the group of chickens that received different levels of AGRE as compared with the control group. Ascending levels of AGRE supplementary in broiler diet affected the carcass meat, which was noticed an increase in crude protein, moisture and ether extract in breast meat compared to the control group. However, ascending levels of AGRE supplementary caused downward in crude protein contents of thigh meat compared to the control group. The present findings revealed that all tested levels of AGRE supplementations were useful against heat stress to downgrade the adverse effects of heat stress in broilers.

Effects of a hypoenergetic diet associated with açaí (Euterpe oleracea Mart.) pulp consumption on antioxidant status, oxidative stress and inflammatory biomarkers in overweight, dyslipidemic individuals

To evaluate the effects of a hypoenergetic diet (HD)associated with açaí pulp consumption on oxidative stress, antioxidant status and inflammatory biomarkers in overweight, dyslipidemic individuals. A randomized, double-blind, placebo-controlled clinical trial was conducted for 90 days. The study began with a 30-day run-in period, during which the intervention was exclusively a HD. Following this period, volunteers were randomized into 2 groups, and 200g of either açaí pulp or placebo were added to the HD for 60 days. Anthropometric measurements, arterial pressure, oxidative stress and antioxidant status biomarkers, inflammatory and biochemical biomarkers were evaluated. Sixty-nine volunteers completed the clinical trial, 30 of which were in the HD+açaí group and 39 in HD+placebo group. Plasma 8-isoprostane concentrations significantly reduced 60 days after the intervention in the açaí group (p=0.000), and there was a significant difference between the groups (açaí versus placebo; p=0.037). Regarding inflammatory status parameters, a significant reduction in IL-6 was observed in the HD+açaí group (p=0.042), and IFN-γ decreased significantly in both groups, HD+açaí (p=0.001) and HD+placebo (p=0.008); there were, however, no differences between the groups. Lipid profile parameters and blood glucose levels did not show change, regardless of nutritional intervention. The addition of açaí to a HD, for 60 days, reduced oxidative stress and improved inflammation in overweight, dyslipidemic individuals.

Effects of probiotics and synbiotic supplementation on antioxidant status: A meta-analysis of randomized clinical trials

Oxidative stress implicated in the pathogenesis of several diseases. Anti-oxidative characteristics of probiotics reported previously. Thus, we aimed to critically investigate the effectiveness of probiotics and synbiotics supplementation on antioxidant biomarkers. A comprehensive search of Scopus and Medline was performed up to November 2017. All randomized controlled trials (RCT) which evaluate the effect of probiotics or synbiotics on superoxide dismutase (SOD) activity, total antioxidant capacity (TAC) and glutathione (GSH) levels were included. Weighted mean difference (WMD) were pooled using random effect model. Sixteen eligible RCTs with 915 participants were included in present study. Findings showed that probiotics could significantly increase GSH level compared to the control groups ((WMD): 132.36, 95% CI: 27.76, 236.95, P=0.01). Because of considerable heterogeneity among included the studies, subgroup analyses were conducted. Subgroup analysis revealed that GSH level significantly increased in non-diabetic individuals; the effect size was not significant in diabetic patients. Furthermore, probiotics and synbiotics showed no significant effect on TAC level (WMD: 0.04, 95% CI:-0.07, 0.15, P: 0.50) and SOD activity (WMD: 0.04, 95% CI:-0.06, 0.13, P=0.43). Probiotics and synbiotics supplementation improve GSH as a biomarkers of antioxidant status in the body. However, additional studies needed for concluding about TAC and SOD activity.

Dynamic Thiol/Disulphide Homeostasis in Children with Nephrotic Syndrome

Background: There is a growing body of evidence showing that there is an association between oxidative damage and kidney injury in patients with nephrotic syndrome (NS). Dynamic thiol/disulphide homeostasis (DTDH) is an important antioxidant system. The aim of this study was to determine if DTDH could be used as a biomarker of antioxidant status in pediatric NS patients using a novel automated method. Methods: The study included 39 patients with NS and 40 healthy controls. The DTDH level was measured using a calorimetric and automated method developed by Erel and Neselioglu. In the NS group, DTDH was measured during first-attack episode, relapse, and remission. Results: The native thiol, total thiol, and disulphide levels were significantly lower, the native thiol/total thiol ratio was significantly lower, and the disulphide/native thiol and disulphide/total thiol ratios were significantly higher in the NS patients during relapse and first-attack episode than during remission and in the controls. In addition, the native thiol and total thiol levels, and the native thiol/total thiol ratio were significantly lower, and the disulphide/native thiol and disulphide/total thiol ratios were significantly higher in the NS group during remission than those in the control group. Conclusions: The present findings show that DTDH shifted toward disulphide formation in the NS patients, even during remission. A clearer understanding of the mechanism of this disruption to homeostasis in NS patients might lead to the development of novel therapeutic strategies.

Telfairia occidentalis (Cucurbitaceae) pulp extract mitigates rifampicin-isoniazid-induced hepatotoxicity in an in vivo rat model of oxidative stress

ObjectiveDrug-induced liver injury complicates antituberculosis drug treatment and is a leading cause of death worldwide. The aim of this study is to establish the ethnomedicinal claim of hepatoprotective effects of fruit pulp extract of Telfairia occidentalis against rifampicin (RIF) and isoniazid (INH)-induced oxidative stress in rats. MethodsT. occidentalis pulp extract (TOPE) (125–500 mg/kg) and silymarin (50 mg/kg) were evaluated in an induced hepatotoxicity model of oxidative stress in Wistar rats by intoxication with RIF and INH (100 mg/kg each) orally for 60 d. Markers indicating oxidative stress and hepatic damage such as alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were assessed. Biomarkers of antioxidant status, including catalase, glutathione reductase, glutathione peroxidase and superoxide dismutase, and marker of lipid peroxidation, malondialdehyde (MDA), were assayed using standard procedures. The hematological profile, lipid profile, serum markers for kidney function and histopathological examination were also assessed. ResultsIntoxication with RIF and INH markedly reduced the hematological indices and elevated the biochemical enzyme markers (AST, ALT and ALP, P < 0.001) and lipid profile (P < 0.001), while antioxidant biomarkers were significantly (P < 0.01) depressed and MDA was elevated. However, pretreatment with TOPE significantly (P < 0.001) alleviated this alteration and sustained the antioxidant potentials. The histopathological morphology supports the biochemical evidence of hepatoprotection. ConclusionCurrent study is indicative of potential antioxidant activity, hepatoprotective effects and plausible therapeutic alleviation of RIF-INH-induced hepatotoxicity of TOPE in laboratory animals.

&lt;p class="abstract"&gt;&lt;strong&gt;Background:&lt;/strong&gt; &lt;span lang="EN-IN"&gt;Free radicals have an important involvement in the underlying mechanism of lichen planus (LP) and also various skin diseases. The aim of the study was to evaluate serum uric acid (UA) levels as a measure of the antioxidant defense status in lichen planus patients.&lt;/span&gt;&lt;/p&gt;&lt;p class="abstract"&gt;&lt;strong&gt;Methods:&lt;/strong&gt; &lt;span lang="EN-IN"&gt;The duration of the present study was from September 2016 to October 2017 which includes 30 lichen planus patients and 30 other patients whose age and sex matched with the subjects served as controls&lt;/span&gt;.&lt;strong&gt;&lt;/strong&gt;&lt;/p&gt;&lt;p class="abstract"&gt;&lt;strong&gt;Results:&lt;/strong&gt; &lt;span lang="EN-IN"&gt;Most common type of lichen planus is classical lichen planus. In our study, serum UA levels were significantly decreased in patients with respect to controls 3.1±0.91 and 4.9±1.08 mg/dl respectively. The difference of means is 1.8 mg/dl and was found to be significant (p&amp;lt;0.001). &lt;/span&gt;&lt;/p&gt;&lt;p class="abstract"&gt;&lt;strong&gt;Conclusions:&lt;/strong&gt; &lt;span lang="EN-IN"&gt;UA may be a potential, useful biomarker of antioxidant status in LP and a rational strengthening of the antioxidant defenses should be part of an optimal treatment strategy.&lt;/span&gt;&lt;/p&gt;

Biomarkers of antioxidant status and lipid peroxidation in elderly patients with hypertension

ABSTRACTObjective: The objective of the present study was to examine selected parameters of the blood redox system in elderly patients with hypertension.examine selected parameters of the blood redox system in elderly patients with hypertension. Methods: We analyzed differences in redox-associated molecules and enzymes among elderly hypertensive subjects (age above 65 years, n = 49) and two groups of normotensive subjects (<65 years old – Control group I; n = 27, and >65 – Control group II; n = 30). Results: Decreased activity of antioxidant enzymes, increased lipid peroxidation and reduced production of nitric oxide were observed in hypertensive subjects, compared to healthy younger controls, or those of the same age. In healthy controls, an age-related decrease in the production of nitric oxide and the activities of SOD-1 and GPx-1 was also evident. The pathology of hypertension was characterised by further, significant decreases in the values of these parameters. When the subgroups of females and males were compared to their respective controls, a compromised redox balance was observed that was more evident in female hypertensives. Discussion: Hypertension in elderly patients is accompanied by changes in biomarkers of antioxidant status and lipid peroxidation status, which significantly differ from those observed in healthy ageing subjects. Our study also suggests that the relationship of gender and changes in redox balance with regard to hypertension should be further explored.

ObjectiveFree radical-mediated oxidative stress has been implicated in the etiopathogenesis of Hashimoto's thyroiditis (HT), which is the most common thyroid disorder in adolescents. HT requires lifelong thyroid surveillance, particularly in women of childbearing age to avoid adverse effects on reproductive function. The aims of this study were to investigate serum concentrations of anti-Müllerian hormone (AMH), a marker of ovarian reserve, in euthyroid adolescent girls with newly diagnosed HT and explore the relationships between AMH levels and biomarkers of antioxidant status. Study designWe recruited 57 non-obese (body mass index [BMI] Z-score<2) adolescent girls with newly diagnosed HT and 50 age- and BMI-matched healthy controls for this case–control study. All participants were euthyroid. Hormonal and metabolic parameters, serum levels of AMH, and antioxidant status [paraoxonase (PON) and arylesterase (ARE) activities] were assessed. ResultsSerum AMH levels were significantly higher and serum PON and ARE activities were significantly lower in adolescents with HT than in the controls (p<0.001 for all). No significant associations were detected between the AMH level and any of the clinical or biochemical parameters in the control group. Serum AMH levels were negatively correlated with PON (r=−0.435, p=0.001) and ARE (r=−0.422, p=0.001) activities in adolescents with HT. ConclusionThe AMH level was significantly higher while the PON and ARE activities were significantly lower in euthyroid adolescent girls with newly diagnosed HT.

Both genetic and dietary factors underlie individual differences in DNA damage levels and DNA repair capacity

The interplay between dietary habits and individual genetic make-up is assumed to influence risk of cancer, via modulation of DNA integrity. Our aim was to characterize internal and external factors that underlie inter-individual variability in DNA damage and repair and to identify dietary habits beneficial for maintaining DNA integrity. Habitual diet was estimated in 340 healthy individuals using a food frequency questionnaire and biomarkers of antioxidant status were quantified in fasting blood samples. Markers of DNA integrity were represented by DNA strand breaks, oxidized purines, oxidized pyrimidines and a sum of all three as total DNA damage. DNA repair was characterized by genetic variants and functional activities of base and nucleotide excision repair pathways. Sex, fruit-based food consumption and XPG genotype were factors significantly associated with the level of DNA damage. DNA damage was higher in women (p=0.035). Fruit consumption was negatively associated with the number of all measured DNA lesions, and this effect was mediated mostly by β-cryptoxanthin and β-tocopherol (p<0.05). XPG 1104His homozygotes appeared more vulnerable to DNA damage accumulation (p=0.001). Sex and individual antioxidants were also associated with DNA repair capacity; both the base and nucleotide excision repairs were lower in women and the latter increased with higher plasma levels of ascorbic acid and α-carotene (p<0.05). We have determined genetic and dietary factors that modulate DNA integrity. We propose that the positive health effect of fruit intake is partially mediated via DNA damage suppression and a simultaneous increase in DNA repair capacity.
