Influence of Foliar Application of Nanoparticles on Low Temperature Resistance of Rice Seedlings

Chilling stress, a common abiotic factor, adversely affects the growth and biomass of rice seedlings during the early stages, ultimately reducing the yield. Effective strategies to mitigate these negative impacts are essential for improving rice productivity. The application of nanotechnology in agriculture, particularly nanoparticles (NPs), has shown a promising effect in alleviating chilling stress in plants. This study evaluates the effects of various nanoparticles, ZnO (0, 50, 100, and 200 mg/L), Fe2O3 (0, 50, 75, and 100 mg/L), TiO2 (0, 50, 75, and 100 mg/L), and CeO2 (0, 50, 75, and 100 mg/L) on the chilling resistance with one control (a water spray) under a normal temperature. Four rice cultivars: LLY-7108 and XZX-6 (Low-temperature-tolerant), and LLY-32 and ZJZ-17 (Low-temperature-susceptible) were tested in this experiment. Rice seedlings were subjected to low temperature conditions (12 h light 14 °C/12 h dark, at 10 °C) for five days, followed by seven days of recovery. The results of this study demonstrate that NPs significantly enhanced seedling height fresh/dry weight and root length compared to untreated controls under chilling stress. NP treatment also reduced the reactive oxygen species (ROS), malondialdehyde (MDA), and proline content, while enhancing superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities, thereby mitigating oxidative damage. The four rice varieties exhibited clear signs of rapid growth recovery and positive physiological changes due to NPs’ application. Among the tested cultivars, LLY-7108 showed the most substantial recovery and physiological responses, while ZJZ-17 exhibited the least. The findings of this study indicate that the foliar application of ZnO (100 mg/L), Fe2O3 (50 mg/L), TiO2 (50 mg/L), and CeO2 (75 mg/L) NPs effectively mitigates chilling stress in rice seedlings, likely by enhancing the antioxidant enzymatic activity while reducing the oxidative damage. This study highlights the potential of NPs as effective agents in reducing the adverse effects of chilling stress on rice.

Intestinal Tissue, Digestive Enzyme, and Antioxidant Enzyme Activities in the Early Development Stage of Endangered Brachymystax tsinlingensis.

This work explores the digestive system characteristics of Brachymystax tsinlingensis during early developmental stages and aims to solve the problem of high lethality of fry during the transgression period, which is crucial for the artificial propagation and population conservation of endangered fishes. This study was carried out on intestinal tissue, digestive enzymes, and antioxidant enzyme activities in the early development stage of Brachymystax tsinlingensis. Ten random samples during endogenous nutrition (7, 10, and 11 days after hatching), mixed nutrition (13 and 19 DAH), and exogenous nutrition (31, 33, 39, 45, and 73 DAH) were collected by histological and biochemical analysis methods. The results showed that the intestine of Brachymystax tsinlingensis already has four layers initially at 7 DAH, and the intestinal gland tissue is evident at 73 DAH. The contents of total protein (TP) and the activities of lipase (LPS) and trypsin (TPS) were maximal at 39 DAH, and the activities were 3.20 ± 0.26 mg/mL, 2.52 ± 0.69 U/g, and 2717.45 ± 295.26 U/mg, respectively. Catalase (CAT) and glutathione peroxidase (GSH-PX) activities both showed the lowest values at 39 DAH, which were 0.57 ± 0.11 U/mg and 3.35 ± 0.94 U/mg, respectively. The activity of amylase (AMS) and the content of malonaldehyde (MDA) increased, and the highest values were reached at 45 DAH (1.32 ± 0.41 U/mg) and 73 DAH (1.29 ± 0.43 nmoL/mg), respectively. Superoxide dismutase (SOD) and GSH-PX activities both showed a peak value at 7 DAH (126.58 ± 20.13 U/mg and 6.47 ± 1.86 U/mg). Overall, the changes in intestinal tissue, digestive enzymes, and antioxidant enzyme activities at 39 DAH of Brachymystax tsinlingensis are inseparable from different vegetative stages during the developmental period, and these results can provide a reference for the proliferation and cultivation of Brachymystax tsinlingensis resources.

Synergy between bio-control agents to trigger the defensive responses against Rhizoctonia root rot and enhance the growth, yield, and physiological and anatomical traits of pea plants

AbstractRhizoctonia solani (Rs) is the fungus that causes the primary and deadly disease that attacks pea plants throughout the cool growing season. It causes seed rot, damping off, and pea root rot diseases. The current study used arbuscular mycorrhizal fungi; Rhizoglomus clarum, Gigaspora margarita, Rhizophagus irregularis and Funneliformis mosseae (AMF), Pseudomonas fluorescens HE21 (Pf), and Trichoderma harzianum HL9 (TH) singly or in combination to suppress Rhizoctonia root rot of pea in a greenhouse. Using the in vitro dual culture assay, TH and Pf inhibited the radial growth of the Rs by 73.3 and 60.0%, respectively. The results of greenhouse experiments showed that all treatments significantly decreased the percentages of pre- and post-emergence damping while significantly increasing the number of surviving plants, particularly in the dual and triple combination treatments. Furthermore, all treatments improved yield and seed quality in addition to plant growth, total phenol content and antioxidant enzyme activity. There were also noted modifications to the treated plants’ anatomical, physiological, and distinctive features. The synergistic triple treatment consisting of M, TH, and Pf achieved the maximum reduction of disease severity (78.5%) compared to the untreated control treatment. As a result of the synergistic triple treatment due to their effectiveness and eco-safety, we recommend using the synergistic triple by M + Pf + TH to manage the root rot disease in peas caused by Rs and to enhance the crop’s growth, yield, and seed quality.

Effect of Different Salinities on the Biochemical Properties and Meat Quality of Adult Freshwater Drum (Aplodinotus grunniens) During Temporary Rearing.

Salinity is a significant environmental component that affects the physiological state of aquatic species. This study aimed to investigate whether water salinity had an impact on the biochemical properties and meat quality of adult Aplodinotus grunniens during temporary rearing of 7 days. Salinity caused increased osmotic pressure and antioxidant enzyme activities of Aplodinotus grunniens, which were attributed to the increase in the content of alanine and glutamate. It raised the hardness and shear force with an increase in salinity, leading to an increase in water-holding capacity. Salinity enhanced the DHA ratio with a decrease in the atherosclerotic index and thrombosis index. Combined with the increase in flavor amino acids and nucleotides, salinity enhanced the umami taste of Aplodinotus grunniens. These findings suggest that temporary rearing in salinity might be a practical approach to improving the meat quality of adult Aplodinotus grunniens.

Cerium Oxide Nanoparticles (CeO2 NPs) Enhance Salt Tolerance in Spearmint (Mentha spicata L.) by Boosting the Antioxidant System and Increasing Essential Oil Composition

Salinity represents a considerable environmental risk, exerting deleterious effects on horticultural crops. Nanotechnology has recently emerged as a promising avenue for enhancing plant tolerance to abiotic stress. Among nanoparticles, cerium oxide nanoparticles (CeO2 NPs) have been demonstrated to mitigate certain stress effects, including salinity. In the present study, the impact of CeO2 NPs (0, 25, and 100 mg L−1) on various morphological traits, photosynthetic pigments, biochemical parameters, and the essential oil profile of spearmint plants under moderate (50 mM NaCl) and severe (100 mM NaCl) salinity stress conditions was examined. As expected, salinity reduced morphological parameters, including plant height, number of leaves, fresh and dry weight of leaves and shoots, as well as photosynthetic pigments, in comparison to control. Conversely, it led to an increase in the content of proline, total phenols, malondialdehyde (MDA), hydrogen peroxide (H2O2), and antioxidant enzyme activities. In terms of CeO2 NP applications, they improved the salinity tolerance of spearmint plants by increasing chlorophyll and carotenoid content, enhancing antioxidant enzyme activities, and lowering MDA and H2O2 levels. However, CeO2 NPs at 100 mg L−1 had adverse effects on certain physiological parameters, highlighting the need for careful consideration of the applied concentration of CeO2 NPs. Considering the response of essential oil compounds, combination of salinity stress and CeO2 treatments led to an increase in the concentrations of L-menthone, pulegone, and 1,8-cineole, which are the predominant compounds in spearmint essential oil. In summary, foliar application of CeO2 NPs strengthened the resilience of spearmint plants against salinity stress, offering new insights into the potential use of CeO2 NP treatments to enhance crop stress tolerance.

Nitric Oxide-Dependent Regulation of Oxygen-Related Processes in a Rat Model of Lead Neurotoxicity: Influence of the Hypoxia Resistance Factor

Background/Aims: Lead exposure is known to induce oxidative stress and neurotoxicity. Nitric oxide (NO) plays an important role in modulating oxidative stress, with L-arginine as a precursor of NO and Nω-nitro-L-arginine (L-NNA) as an inhibitor of NO synthase, an enzyme that catalyses the production of nitric oxide (NO) from L-arginine. Methods: This study investigated the differential effects of L-arginine and L-NNA on markers of oxidative stress and biochemical changes in brain tissue from rats with different levels of resistance to hypoxia exposed to lead nitrate. Rats with either low or high resistance to hypoxia were exposed to lead nitrate (oral 3.6 mg lead nitrate/kg b.w. per day for 30 days) and treated with L-arginine (600 mg/kg b.w., i.p., 30 min before and after exposure to lead nitrate) or L-NNA (35 mg/kg b.w., i.p., 30 min before and after exposure to lead nitrate). Brain tissue samples were analysed for lipid peroxidation, oxidative modification of proteins, and activity of antioxidant enzymes, including superoxide dismutase, catalase, glutathione reductase, and peroxidase, and total antioxidant status (TAS). We also examined the biomarkers of biochemical pathways involving the activity of alanine and aspartate aminotransferases, succinate dehydrogenase (SDH), and α-ketoglutarate dehydrogenase (KGDH). In addition, the trend observed was supported by assessments of the acetylcholine levels and acetylcholinesterase activity (ACh-AChE system) in brain tissue. Results: In rats with low resistance to hypoxia, the L-arginine treatment significantly reduced lipid peroxidation and oxidative protein modification but increased antioxidant enzyme activity, suggesting a protective effect against lead-induced oxidative stress. Conversely, in rats with high resistance to hypoxia, L-NNA had a protective effect, reducing lead-induced oxidative damage and decreasing lipid peroxidation, whereas L-arginine exacerbated oxidative stress and impaired antioxidant defences. These findings were supported by corresponding changes in the acetylcholine-acetylcholinesterase system, reflecting the observed patterns of lead-induced oxidative stress and neurotoxicity. The study shows that L-arginine exerts a protective effect by reducing lead-induced oxidative damage via an improvement in TAS. Our study shows that lead nitrate exposure significantly increases ala-nine and aspartate aminotransferase activity in brain tissue, with L-arginine exacerbating and L-NNA reversing this effect. The lead nitrate exposure also affected the activities of SDH and KGDH, which are important for cellular energy production and hypoxia resistance, with L-arginine altering SDH activity depending on the level of resistance and L-NNA enhancing both SDH and KGDH activities. These trends were further validated by alterations in the ACh-AChE system, highlighting the differential role of NO-dependent mechanisms in modulating lead-induced neurotoxicity based on hypoxia resistance. Conclusion: These findings suggest potential targeted therapeutic strategies based on the oxidative stress profile and highlight the potential of nitric oxide system modulators in counteracting lead-induced biochemical alterations and the dynamics of the ACh-AChE system depending on the individual physiological reactivity of organisms.

Assessment of Paraoxonase 1 and Arylesterase Activities and Lipid Profile in Bodybuilders: A Comparative Study of Physical Activity and Anthropometry on Atherosclerosis.

Background and Objectives: Atherosclerosis, driven by dyslipidaemia and oxidative stress, is a leading cause of cardiovascular morbidity and mortality. This study evaluates the effects of vigorous-intensity bodybuilding exercise (VIBBE) on atherosclerosis biomarkers-including paraoxonase-1 (PON1) and arylesterase (ARE) activities-and lipid profiles in male bodybuilders who do not use anabolic-androgenic steroids. Comparisons were made with individuals engaged in moderate-intensity aerobic exercise (MIAE), as well as overweight/obese sedentary (OOS) and normal-weight sedentary (NWS) individuals. Materials and Methods: A cross-sectional study was conducted involving 122 healthy male participants aged 18-45 years, divided into four groups: VIBBE (n = 31), OOS (n = 30), MIAE (n = 32), and NWS (n = 29). Anthropometric assessments were performed, and fasting blood samples were collected for biochemical analyses, including lipid profiles and PON1 and ARE activities. Statistical analyses compared the groups and evaluated correlations between adiposity measures and atherosclerosis biomarkers. Results: The VIBBE group exhibited significantly lower levels of low-density lipoprotein cholesterol (LDL-C), triglycerides (TG), and logarithm of the TG to high-density lipoprotein cholesterol (HDL-C) ratio [log(TG/HDL-C)] compared to the OOS group (p < 0.05 for all), indicating improved lipid profiles. However, these improvements were not significant when compared to the NWS group (p > 0.05), suggesting that VIBBE may not provide additional lipid profile benefits beyond those associated with normal weight status. PON1 and ARE activities were significantly lower in the VIBBE group compared to the MIAE group (p < 0.05 for both), suggesting that VIBBE may not effectively enhance antioxidant defences. Correlation analyses revealed significant inverse relationships between PON1 and ARE activities and adiposity measures, including body mass index (BMI), waist-to-hip ratio (WHR), waist-to-height ratio (WHtR), body fat percentage (BFP), fat mass index (FMI), and obesity degree (OD) (p < 0.05 for all). Positive correlations were observed between oxLDL and log(TG/HDL-C) and adiposity measures (p < 0.05 for all). Conclusions: Vigorous-intensity bodybuilding exercise improves certain lipid parameters compared to sedentary obese individuals but does not significantly enhance antioxidant enzyme activities or further improve lipid profiles beyond those observed in normal-weight sedentary men. Conversely, moderate-intensity aerobic exercise significantly enhances PON1 and ARE activities and improves lipid profiles, offering superior cardiovascular benefits. These findings underscore the importance of incorporating moderate-intensity aerobic exercise into physical activity guidelines to optimize cardiovascular health by balancing improvements in lipid metabolism with enhanced antioxidant defences.

Exogenous 5-aminolevulinic acid enhanced saline-alkali tolerance in pepper seedlings by regulating photosynthesis, oxidative damage, and glutathione metabolism.

A plant growth regulator, 5-aminolevulinic acid, enhanced the saline-alkali tolerance via photosynthetic, oxidative-reduction, and glutathione metabolism pathways in pepper seedlings. Saline-alkali stress is a prominent environmental problem, hindering growth and development of pepper. 5-Aminolevulinic acid (ALA) application effectively improves plant growth status under various abiotic stresses. Here, we evaluated morphological, physiological, and transcriptomic differences in saline-alkali-stressed pepper seedlings after ALA application to explore the impact of ALA on saline-alkali stress. The results indicated that saline-alkali stress inhibited plant growth, decreased biomass and photosynthesis, altered the osmolyte content and antioxidant system, and increased reactive oxygen species (ROS) accumulation and proline content in pepper seedlings. Conversely, the application of exogenous ALA alleviated this damage by increasing the photosynthetic rate, osmolyte content, antioxidant enzyme activity, and antioxidants, including superoxide dismutase, catalase, glutathione reductase, and glutathione peroxidase, and reducing glutathione to reduce ROS accumulation and malonaldehyde content. Moreover, the transcriptomic analysis revealed the differentially expressed genes were mainly associated with photosynthesis, oxidation-reduction process, and glutathione metabolism in saline-alkali stress + ALA treatment compared to saline-alkali treatment. Among them, the change in expression level in CaGST, CaGR, and CaGPX was close to the variation of corresponding enzyme activity. Collectively, our findings revealed the alleviating effect of ALA on saline-alkali stress in pepper seedlings, broadening the application of ALA and providing a feasible strategy for utilize saline-alkali soil.

Impact of Sulfur Deficiency and Excess on the Growth and Development of Soybean Seedlings.

Sulfur is a critical element for plant growth and development, serving as a component of amino acids (cysteine and methionine), iron-sulfur clusters, proteins, glutathione, coenzymes, and auxin precursors. Deficiency or low concentrations of sulfur in the soil can lead to significant growth retardation in plants. The objective of our study was to examine the effects of sulfur (S) deficiency and excess on morphological symptoms, sulfur and nitrogen (N) metabolism, as well as antioxidant activity in soybean. We found that S starvation decreased the fine root length, biomass, and activity, and the chlorophyll content was reduced, while excess sulfur promotes lateral root growth. In contrast to sulfur excess, sulfur deficiency inhibits N and S metabolism levels in both subsurface and above-ground parts, and induced the expression of some sulfur transporters (SULTRs). In this study, we created soybean hairy root lines overexpressing the SULTR gene (GmSULTR2;1a) to observe metabolic changes following sulfur deficiency treatment. The results showed that GmSULTR2;1a saved the sulfur-deficient phenotype, and the antioxidant enzyme activity was much higher than that of the wildtype in the absence of sulfur. Our study revealed the important role of sulfur element in soybean growth and development and the regulation of sulfur deficiency by GmSULTR2;1a.

Nano-enhanced storage of American cotton using metal-oxide nanoparticles for improving seed quality traits.

Cotton seeds have poorer germination than other crops because of their high sensitivity towards insect pests and other biotic and abiotic stresses during the germination process. In the present study, inorganic bulk and nano nutrients of zinc oxide (ZnO) and titanium oxide (TiO2) nanoparticles were synthesized using the chemical reduction method and invigourated with cotton seeds. The characterization of nanoparticles was done by FESEM, HRTEM, UV/Vis analysis and FTIR. The delinted and fuzzy seeds of two American cotton varieties (H 1300 and H 1098-i) were nano-primed for 10h with zinc oxide nanoparticles (ZnONPs) @ 400ppm and titanium dioxide nanoparticles (TiO2NPs) @ 100ppm. After nanoparticle invigouration, the seeds were analyzed for various parameters at different intervals (0months, 3months, 6months, 9months and 12months) such as germination percentage, seedling length, seedling dry weight, electrical conductivity, dehydrogenase activity, antioxidant enzyme activity. The results indicated that that different storage periods and nanopriming treatments had significant effects on all seed quality parameters except the effect of nanopriming treatments on germination percentage (excluding delinted seeds of H 1098-i). It is also revealed that the interaction effect of nanopriming treatment and storage period was non-significant on all parameters except EC. Maximum reduction in seed quality parameters was observed in control treatment and minimum was found when seeds were nanoprimed with ZnONPs @ 400ppm. The differences in the response for both NPs can be attributed to their surface charge, and concentration used. Overall, ZnONPs and TiO2NPs could hold seed quality and vigour during the storage of cotton seeds of American varieties (H 1300 and H 1098-i).

Genome-wide identification of Aux/IAA gene family members in grape and functional analysis of VaIAA3 in response to cold stress.

Twenty-five VvIAA genes and eighteen VaIAA genes were identified from Pinot Noir and Shanputao, respectively. The overexpression of VaIAA3 in transgenic Arabidopsis increased cold tolerance by regulating auxin, ABA and ethylene signaling. Aux/IAA genes are key genes involved in regulating auxin signal transduction in plants. Although IAA genes have been characterized in various plant species, the role of IAA genes in grape cold resistance is unclear. To further explore the members of the Aux/IAA gene family in grape and their functions, in this study, using genomic data for Pinot Noir (Vitis vinifera cv. 'Pinot Noir') and Shanputao (Vitis amurensis), 25 VvIAA genes and 18 VaIAA genes were identified. The VaIAA genes presented different expression patterns at five different temperatures (28 ± 1 °C, 5 ± 1 °C, 0 ± 1 °C, -5 ± 1 °C, and -10 ± 1 °C) according to qRT‑PCR results. VaIAA3 was selected as a candidate gene for further functional analysis because of its high expression level under low-temperature stress. Subcellular localization experiments revealed that VaIAA3 was localized in the nucleus. Additionally, under 4 °C treatment for 24 h, relative expression level of VaIAA3, antioxidant enzyme activity, survival rate, and cold-responsive gene expression in three transgenic lines (OE-1, OE-2, OE-3) were greater, whereas relative electrolytic conductivity (REC), malondialdehyde (MDA) content and hydrogen peroxide (H2O2) content were lower than those of the wild type (WT). Transcriptome sequencing analysis revealed that VaIAA3 regulated cold stress resistance in Arabidopsis thaliana (Arabidopsis) through pathways involving auxin, ABA, JA, or ethylene. Importantly, heterologous overexpression of VaIAA3 increased the resistance of Arabidopsis to cold stress, which provides a theoretical basis for the further use of VaIAA3 to improve cold resistance in grape.

Population Dynamic of Aphis gossypii, and Their Parasitoids, and Effect of Some Insecticides on Aphids, Chlorophyll Content, Non-Enzymatic and Antioxidant Enzymes Activity in Cantaloupe Leaves in Greenhouses

Under green house, the Population density of Aphis gossypii on cantaloupe plants recorded two peaks on May 9th and 23th with 1409 and 1561 individuals/ 10 leaves in season 2022, while one highest peak, on 16th May with 1650 individuals/ 10 leaves in season 2023. Parasitism status of Aphidius colemani and Lysiphlebus fabarum varied from 1.49% on 25 April to 35.79% on 9th of May with seasonal parasitism 2023 in 2022 and ranged between 0.73% on 25th of April and 30.06% on 16th of May with seasonal parasitism 16.57% in 2023. Acetamiprid, imidacloprid, imidacloprid + abamectin, jojoba oil, orange oil and thiamethoxam + abamectin compounds were tested to control aphids during two seasons. Imidacloprid recorded the highest reduction with all average 97.88 ±1.15 and 97.39 ±1.15 in the first and second season respectively. the highest mean reduction of emergence of the two-parasites caused by thiamethoxam + abamectin by 16.92% followed by Imidacloprid with 16.87%. While the least toxicity to emergence of the parasitoids recorded with jojoba oil 0.0% followed by orange oil 1.93% in season 2022, while in season 2023, the higher parasitoid reduction caused by acetamiprid 18.05% followed by Imidacloprid 17.69% and thiamethoxam + abamectin 17.53%. Chlorophyll content increased with treatment jojoba oil followed by Imidacloprid by 47.91 and 44.73 unit in the first season and Thiamethoxam with abamectin lead to the highest content by 44.05 unit in the leaves followed by jojoba oil with 43.65 unit. The least treatment was Imidacloprid + abamectin with 39.55 unit compared with control treatment 40.43 unit. Treatment Thiamethoxam + abamectin recorded the highest content of carbohydrates (48.43) compared with control (47.71). Treatments of insecticides led to increase of protein, fats and total lipid compared with control. Also, Antioxidant enzymes activity in cantaloupe leaves were found to be increased in the treatment of insecticides compared with control.

Exogenous Melatonin as Pre- and Postharvest Application on Quality Attributes, Antioxidant Capacity, and Extension of Shelf Life of Papaya

Papaya is widely grown in tropical and subtropical climates due to its high yield potential and high returns. The vital hormone melatonin, regulating various biological processes in plants, is eco-friendly and less harmful to humans than other chemicals. This study aims to enhance the quality and antioxidant enzyme activities and lessen postharvest senescence in papaya cv. CO 8 fruits during both ambient (32 ± 2 °C and 55 ± 5% RH) and cold storage (10 ± 2 °C and 90–95% RH) as exogenous melatonin (EMT) is applied in varying concentrations. An optimum melatonin dose of 1.5 mM was applied as a pre-harvest spray 15 days before harvest and a postharvest dip proved effective in prolonging shelf life (under ambient it prolonged to day 9 and under cold storage up to 28 days) and delaying ripening and softening. Exogenous melatonin application enhanced antioxidant activity, reduced weight loss, maintained firmness, delayed ripening enzymes, and lowered ethylene and CO2 levels. For instance, control fruits had weight losses between 7.42% and 10.09%, while fruits treated with 1.5 mM melatonin showed 5.74% and 9.06% weight loss under ambient and cold storage, respectively. In conclusion, applying EMT (1.5 mM) could be an economically viable and environmentally benign way to lessen senescence after harvest and preserve the qualities of the papaya fruit during ambient and cold storage.

Advanced chelate technology-based trace minerals reduce inflammation and oxidative stress in Eimeria-infected broilers by modulating NF-kB and Nrf2 pathways

This study investigated the effects of substituting inorganic trace minerals (ITM) with advanced chelate technology-based TM (ACTM) in broiler chicken feed on productive performance, metabolic profile, humoral immunity, antioxidant status, and modulation of NF-kB and Nrf2 signaling pathways in mixed Eimeria species exposure. The study involved 480 newly hatched male broiler chickens, which were divided into 5 treatment groups, each with 6 replicate cages and 16 chickens per replicate. The experimental treatments included an uninfected negative control group fed a basal diet with recommended inorganic TM levels (NC), an infected positive control group fed the same diet (PC), a PC group supplemented with salinomycin (SAL), and two PC groups in which the basal diet was replaced with 50% and 100% ACTM instead of inorganic TM (ACTM50 and ACTM100, respectively). All groups, except for the NC group, were orally challenged with mixed Eimeria species oocysts on day 14. According to the results, the PC group showed lower feed intake, breast yield, low-density lipoprotein-cholesterol concentration, lactobacillus spp. counts, and serum IgG levels, but higher jejunal TGF-β expression versus the NC group. The broilers in the NC, SAL, and ACTM100 groups showed higher body weight gain, carcass yield, and TGF-β expression, but lower serum alkaline phosphatase activity, ileal E. coli count, and jejunal expression levels of IL-1β, IL-6, IFN-γ, Nrf2, and SOD1 compared to the PC group, with the NC group having the highest body weight gain and lowest IL-1β and Nrf2 expression levels. Furthermore, the administration of ACTM100 treatment improved feed efficiency, increased serum iron, zinc, manganese, and copper levels, enhanced total antioxidant capacity and different antioxidant enzyme activities, and reduced malondialdehyde concentration. In conclusion, complete replacement of ITM with ACTM effectively protects broilers from Eimeria infection, with similar positive effects to SAL treatment in terms of productive performance and anti-inflammatory responses and better antioxidant responses and mineral availability.

Dynamic responses of germination characteristics and antioxidant systems to alfalfa (Medicago sativa) seed aging based on transcriptome

Seed aging poses a significant challenge to agronomic production and germplasm conservation. Reactive oxygen species (ROS) are highly involved in the aging process. However, dynamic response of germination characteristics and antioxidant system to seed aging are not yet very clear. This study explored the potential physiological mechanisms responsible for the reduced and rapid loss of seed vigor in alfalfa, and identified key genes regulating seed vigor. The germination percentage exhibited a decreased trend with the prolongation of aging duration. From 16 to 32 days of aging, the antioxidant enzyme activities of SOD, POD, CAT, DHAR and MDHAR declined significantly, which lead to the disruption of ROS balance and a significant increase in ROS levels, exacerbating seed aging. Based on transcriptome, 29 differentially expressed genes (DEGs) including SOD1, APX-2 and GST-7 within the ROS scavenging system showed a significantly down-regulated expression trend at aging of 16 and 24 days, indicating the abnormal function of antioxidant metabolism. Furthermore, some related genes including ATPF1B, ATPeF0C-3, NDUFS1, NDUFS3 and ND2 in the mitochondrial ETC exhibited a downturn following seed aging, which would result in the losing of seed vigor. This study has uncovered a significant array of potential target genes within the seed antioxidant system and mitochondrial ETC. These discoveries offer a wider lens for delving into the molecular regulatory mechanisms of seed aging. Further research is crucial to comprehensively elucidate the precise pathways through which these pivotal genes regulate seed vigor.

Ficus carica Puree Exerts a Higher Antioxidative Profile in Hypoxic Lungs after Intermittently Inducing Hypoxia in Sprague–Dawley Rats

Abstract Background: The body produces more hypoxia-inducible factor and reactive oxygen species in response to hypoxic situations because they cause unbalanced oxygen levels. Ficus carica provides numerous benefits because of its high antioxidant and mineral content. Nevertheless, the mechanism by which F. carica consumption confers this protective benefit remains incompletely understood. Aim: This study aimed to evaluate how F. carica puree (FCP) can affect the antioxidant enzyme activity and malondialdehyde (MDA) levels in the lung by mitigating the effects of intermittent hypoxia (IH). Methods: Thirty Sprague–Dawley rats were divided into five groups negative control (NC), untreated; positive control (PC), treated with aquadest; FCP1; FCP2; and FCP3, which received FCP at doses of 1.25, 2.5, and 5 mL/200 g body weight. The treatment was administered for 4 weeks before inducing IH (10% O2 and 90% N2) into all groups (except NC) for 4 h for 7 days. Furthermore, hemoglobin (Hb) level, lung MDA level, and lung superoxide dismutase (SOD) enzyme activity were assessed. Results: The Hb level did not exhibit a significant increase under IH conditions. Conversely, the PC group exhibited the least activity of lung antioxidant enzymes and the highest lung MDA levels. In addition, the FCP intervention group exhibited lower MDA levels than the PC group and ameliorated relative lung weight loss. Conclusion: All FCP intervention groups showed lower MDA levels and higher SOD levels compared to the PC group, suggesting that FCP could mitigate the effects of hypoxia in rat lungs.

Effects of nutritional stress on soil fertility and antioxidant enzymes of rice in different growth periods.

Stress in plants denotes the detrimental impact of alterations in external environmental conditions on regular plant growth and development. Plants employ diverse mechanisms to mitigate or evade nutritional stress-induced damage. In order to investigate the physiological response mechanism of plants to nutritional stress and assess its impact on soil nutrient content and antioxidant enzyme activity in rice, a field experiment was conducted applying five treatments: control, nitrogen (N) deficiency, phosphorus (P) deficiency, potassium (K) deficiency, and full fertilization. Rice leaf and soil samples were concurrently gathered during both the vegetative and reproductive growth stages of rice. Analysis was conducted on soil N, P, and K levels, as well as leaf antioxidant enzyme activities, to investigate the impact of nutrient stress on rice antioxidant enzymes and soil fertility. The research findings indicate that full fertilization treatment enhanced the agronomic properties of the soil compared to the control treatment. In the N-deficiency treatment, reactive oxygen species (ROS) levels increased by 16.53-33.89% during the reproductive growth period compared to the vegetative growth period. The peroxidase (POD) activity decreased by 41.39% and superoxide dismutase (SOD) activity increased by 36.22% under K-deficiency treatment during the reproductive growth period compared to the vegetative growth period. Consequently, applying N and P fertilizer during the vegetative growth period can decrease membrane lipid peroxidation levels by 7.34-72.53%. The full fertilization treatment markedly enhanced rice yield compared to other treatments and increased the Nitrogen activation coefficient (NAC) and Phosphorus activation coefficient (PAC) in the soil, while decreasing the PAC. Elevating NAC levels can stimulate the activity or content of PRO, MDA, and RPS during the vegetative growth stage, whereas in the reproductive growth stage, it will decrease the content of ROS, PRO, and MDA. This data offers valuable insights and theoretical support for nutritional stress research.

Frequency-Dependent Antioxidant Responses in HT-1080 Human Fibrosarcoma Cells Exposed to Weak Radio Frequency Fields.

This study explores the complex relationship between radio frequency (RF) exposure and cancer cells, focusing on the HT-1080 human fibrosarcoma cell line. We investigated the modulation of reactive oxygen species (ROS) and key antioxidant enzymes, including superoxide dismutase (SOD), peroxidase, and glutathione (GSH), as well as mitochondrial superoxide levels and cell viability. Exposure to RF fields in the 2-5 MHz range at very weak intensities (20 nT) over 4 days resulted in distinct, frequency-specific cellular effects. Significant increases in SOD and GSH levels were observed at 4 and 4.5 MHz, accompanied by reduced mitochondrial superoxide levels and enhanced cell viability, suggesting improved mitochondrial function. In contrast, lower frequencies like 2.5 MHz induced oxidative stress, evidenced by GSH depletion and increased mitochondrial superoxide levels. The findings demonstrate that cancer cells exhibit frequency-specific sensitivity to RF fields even at intensities significantly below current safety standards, highlighting the need to reassess exposure limits. Additionally, our analysis of the radical pair mechanism (RPM) offers deeper insight into RF-induced cellular responses. The modulation of ROS and antioxidant enzyme activities is significant for cancer treatment and has broader implications for age-related diseases, where oxidative stress is a central factor in cellular degeneration. The findings propose that RF fields may serve as a therapeutic tool to selectively modulate oxidative stress and mitochondrial function in cancer cells, with antioxidants playing a key role in mitigating potential adverse effects.

Exploring the synergistic benefits of biochar and gibberellic acid in alleviating cadmium toxicity

Cadmium (Cd) toxicity significantly threatens agricultural productivity and food safety. Developing effective strategies to enhance plant tolerance to Cd stress is essential. This study investigates the synergistic effects of biochar (BC) and gibberellic acid (GA3) on mitigating Cd toxicity in maize (Zea mays), focusing on their impact on oxidative stress markers and antioxidant enzyme activities. Soil samples were collected from the Cholistan Institute of Desert Studies (CIDS) and analyzed for trace metal ions and other properties. Biochar was produced from fruit and vegetable waste, washed, washed, deashed, and mixed with 10 ppm GA3. FH-1036 hybrid maize seeds were sterilized and planted in pots containing soil with varying Cd levels (0, 8, and 16 mg Cd/kg soil). Twelve treatments were established, including control, GA3, BC, and their combinations under different Cd stress levels. Plants were irrigated to maintain 60% field capacity and harvested at the V10 growth stage. Hydrogen peroxide (H2O2) contents and activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) were measured in roots, stems, and leaves. Statistical analysis was performed using OriginPro 2021, with ANOVA and Fisher’s LSD test used to determine significant differences. GA3 and BC treatments significantly reduced H2O2 levels in maize roots, stems and leaves under Cd stress. The combined treatment of GA3 + BC showed the most significant reduction in H2O2 levels across all plant parts, reducing root H2O2 by 50%, stem H2O2 by 55%, and leaf H2O2 by 53% under severe Cd stress (16 mg Cd/kg). SOD activity increased under non-stress conditions but decreased under Cd stress, with the highest activity observed in the combined treatment. POD activity followed a similar pattern, with GA3 + BC treatment resulting in the most significant increases under non-stress conditions and the least reductions under Cd stress. CAT activity showed substantial increases with GA3 + BC treatment, particularly under severe Cd stress, with a notable rise over the control. APX activity also exhibited enhancements with GA3 and BC treatments, especially in the combined treatment under various Cd stress levels. This study highlights the potential of combined BC and GA3 treatments in improving Cd stress tolerance in maize. Future research should focus on field trials and the long-term impacts of these treatments on crop productivity and soil health.
