Abstract

A simple, precise, and accurate high-performance thin-layer chromatography (HPTLC) method for the simultaneous determination of pioglitazone, metformin, and glimepiride in multicomponent pharmaceutical preparations has been developed and validated. Pioglitazone, metformin, and glimepiride from the formulations were separated on silica gel 60 F254 HPTLC plates with acetonitrile, methanol, propyl alcohol, and ammonium acetate solutions in the proportion of 7:2:1:1 (ν/ν) as mobile phase. Densitometric quantification was performed at 240 nm. Well-resolved bands were obtained with RF values 0.83, 0.21, and 0.89 for pioglitazone, metformin, and glimepiride, respectively. The method was validated for precision, accuracy, specificity, and robustness. The calibration curve was found to be linear in the concentration range of 0.3–1.2, 10–40, and 0.04–0.16 μg per band by area with correlation coefficients of 0.995, 0.996, and 0.998 for pioglitazone, metformin, and glimepiride, respectively.
