Abstract

Microorganisms are isolated and propagated as a pure culture. The pure culture is stored for a longer time period with the protection from contamination. During storage in laboratories, the survival of microorganisms is retained by transferring periodically to a fresh medium. Recovery microorganisms involve inoculation into an appropriate medium and incubating at the optimal growth conditions. Transfer is always done under aseptic conditions to avoid contamination. Storage conditions (such as temperature, oxygen, and humidity) affect the microbial viability. There are short-term, long-term, and novel emerging preservation technologies for microbial cultures. In this practice, periodic transfer of microbial culture, storage at freezing, freeze-drying, ultra-freezing, immersing in paraffin preservation techniques are purposed.
