Abstract

Objectives To assess the placental oxidative stress factors in various types of hypertensive disorders of pregnancy and to correlate with the pregnancy outcome. Methods Placental samples from Hypertensive disorders of pregnancy (HDP) patients and the control pregnant women (CPW) were collected. The levels of oxidative stress parameters MDA, Total antioxidant status (TAS) and protein carbonylation (PCO) were assessed in the placental lysates. Also the placental expression of antioxidant enzymes superoxide dismutase (SOD), Glutathione peroxidase (GPX1), Glutathione Reductase (GSH), catalase and stress sensitive serine kinase proteins ERK, p38-MAPK, JNK, IKBα and NOX-4 were found out by western blotting in HDP (gestational hypertension, Preeclampsia- eclampsia) and CPW. The Pregnancy outcomes like the gestational age at the time of delivery (GA) and birthweight (BW) and APGAR score of the baby also were measured. Results The GA, BW and APGAR score of the baby were found to be significantly lower in Preeclampsia and Eclampsia compared to CPW. The levels oxidative stress parameters MDA and PCO were elevated and the TAS were decreased in HDP compared to CPW. The placental expression of pERK, pNrf-2/Nrf-2, NOX-4 and IkBα were found to be elevated whereas the expression of CAT, JNK, pP38MAPK were found to be decreased in HDP when compared to CPW. Also a negative correlation was observed between the o0xidative stress markers and the pregnancy outcome in HDP. Conclusions An increased placental oxidative stress and a poor birth outcome were observed in hypertensive disorders of pregnancy.
