Abstract

A method is described which permits specific hybridisation of RNA to complementary DNA in ultrathin sections of tissues prepared for electron microscopy after brief gluteraldehyde fixation and glycerol metacrylate embedding. After hybridisation with 3H-labelled 28-S RNA the intracellular location of ribosomal DNA complements was revealed by electron microscope autoradiography.
