Abstract
目的 研究鸦胆子油乳诱导白血病细胞的凋亡及其机制.方法 MTT法检测鸦胆子油乳对K562细胞增殖的抑制率;细胞形态及流式细胞术(FCM)检测细胞凋亡;FCM法检测凋亡相关基因Fas和Fas-L表达水平的改变.结果 鸦胆子油乳可抑制K562细胞的增殖并呈时间-剂量-效应关系,IC50为187μg/ml;K562细胞表现为典型的凋亡形态;125、250、500μg/ml鸦胆子油乳作用24、48、72 h的细胞凋亡率均高于对照组(P＜0.01);500μg/ml鸦胆子油乳作用48 h后,Fas阳性细胞率高于对照组(P＜0.01),而Fas-L的表达无明显改变.结论 鸦胆子油乳在体外可诱导K562细胞凋亡,上调Fas表达可能是其诱导凋亡的机制之一。
