Abstract

Owing to the excellent base resolution and good signal amplification of CRISPR-Cas-mediated systems, research on CRISPR-Cas has grown by leaps and bounds, opening up a novel epoch in the development of sensing. CRISPR-mediated sensing strategies allow rapid, high sensitivity, and specific determination of diverse targets, including DNA, RNA, enzymes, proteins, metal ions, small organic compounds, etc., in biological, environmental, or food samples. More importantly, the CRISPR-Cas-mediated systems have also demonstrated the versatility of integrating with various signaling modalities including fluorescence, colorimetric, and electrochemical. Therefore CRISPR-Cas-mediated systems have been becoming hopeful contenders for future-generation sensing strategies. This chapter emphasizes the latest research progress on the design and application of CRISPR-Cas-mediated systems, to highlight the functionality achievements of in vitro diagnostics. This chapter also discusses the problems and challenges facing the future application and development of CRISPR-Cas system.
