Abstract

Agarose gel electrophoresis is a gel electrophoresis technique used in biochemistry, molecular biology, genetics, and clinical chemistry to separate a mixed population of macromolecules in an agarose matrix. Agarose gel electrophoresis is the most efficient method for isolating DNA fragments ranging in size from 100 bp to 25kb. An electric field is used to drive charged molecules across an agarose matrix, and the biomolecules are segregated by size in the agarose gel matrix. This chapter explains the principle, reagent mixture, and step-by-step protocol advantages and disadvantages of agarose gel electrophoresis.
