Publisher Summary This chapter focuses on karyology of cells in culture and the characteristics of plant cells. The assessment of the karyological state of plant cells in culture is a primary objective for investigating nuclear mechanisms and while using the in vitro method of cell propagation. This is also an important assessment to be made when plant cells in vitro are used for any other purpose. The procedures described in the chapter are concerned only with the preparation of squash preparations. For the most general purposes, with adequate precautions, this method is quite satisfactory as well as expedient. If large numbers of samples are to be analyzed, it is the only practicable approach, the alternative of embedding and sectioning being tedious, time consuming, and not suited for making quick assessments. The procedures call for passing the tissue or cells through several treatments. Coherent tissues can simply be transferred from one treatment to the next with forceps. The chapter also elaborates the procedures for pretreatment of cells, sampling of the cell population, and fixation of cells. The chapter discusses the hydrolysis and maceration.
