Abstract
Polypharmacy leads to increased incidences of drug–drug interactions (DDIs), which results in severe and debilitating drug-induced side effects. The potential DDI becomes significantly important with the gradual increase in age and the associated surge in multimorbidity and polypharmacy. Furthermore, an accurate method to predict the new DDI is necessary to circumvent the unknown adverse effects of the drug combinations. Hence, this chapter aims to discuss the impact of DDI on drug pharmacokinetics, and we further briefly discussed the critical role of microbiota in DDI. Furthermore, we reviewed the data on strategies to prevent the DDI and systematically presented it in the chapter. Finally, novel DDI prediction techniques and current clinical trial updates on DDI were discussed to predict the DDI in the least possible time with high accuracy.
