3D-printing of alginate/gelatin scaffold loading tannic acid@ZIF-8 for wound healing: In vitro and in vivo studies

In the present study, ZIF-8 metal-organic framework (MOF) modified with Tannic acid (TA@ZIF-8) was synthesized and impregnated in alginate-gelatin (Alg-Gel) hydrogel. The Alg-Gel scaffolds containing 0, 5, and 10 % of TA@ZIF-8 were fabricated through the 3D printing method specifically denoted as Alg-Gel 0 %, Alg-Gel 5 %, and Alg-Gel 10 %. XRD, FTIR, FESEM, and EDX physically and chemically characterized the synthesized ZIF-8 and TA@ZIF-8 MOFs. Besides, Alg-Gel containing TA@ZIF-8 prepared scaffolds and their biological activity were also evaluated. SEM images verified the nano-size formation of MOFs. Improved swelling and decreased degradation rates after adding TA@ZIF-8 were also reported. Increased compression strength from 0.628 to 1.63 MPa in Alg-Gel 0 % and Alg-Gel 10 %, respectively, and a 2.19 increase in elastic modulus in Alg-Gel 10 % scaffolds were exhibited. Biological activity of scaffolds, including Live-dead and Cell adhesion, antibacterial, in-vivo, and immunohistochemistry assays, demonstrated desirable fibroblast cell proliferation and adhesion, increased bacterial growth inhibition zone, accelerated wound closure and improved expression of anti-inflammatory cytokines in Alg-Gel 10 % scaffolds. The findings of this study confirm that Alg-Gel 10 % scaffolds promote full-thickness wound healing and could be considered a potential candidate for full-thickness wound treatment purposes.

Cationized gelatin-sodium alginate polyelectrolyte nanoparticles encapsulating moxifloxacin as an eye drop to treat bacterial keratitis

A mucoadhesive polyelectrolyte complex (PEC) nanoparticles were developed for ocular moxifloxacin (Mox) delivery in Bacterial Keratitis (BK). Moxifloxacin-loaded G/CG-Alg NPs were prepared by an amalgamation of cationic polymers (gelatin (G)/cationized gelatin (CG)), and anionic polymer (sodium alginate (Alg)) along with Mox respectively. Mox@CG-Alg NPs were characterized for physicochemical parameters such as particle size (DLS technique), morphology (SEM analysis), DSC, XRD, encapsulation efficiency, drug loading, mucoadhesive study (by texture analyzer), mucin turbidity, and viscosity assessment. The NPs uptake and toxicity of the formulation were analyzed in the Human Corneal Epithelial (HCE) cell line and an ocular irritation study was performed on the HET-CAM. The results indicated that the CG-Alg NPs, with optimal size (217.2 ± 4 nm) and polydispersity (0.22 ± 0.05), have shown high cellular uptake in monolayer and spheroids of HCE. The drug-loaded formulation displayed mucoadhesiveness, trans-corneal permeation, and sustained the release of the Mox. The anti-bacterial efficacy studied on planktonic bacteria/biofilms of P. aeruginosa and S. aureus (in vitro) indicated that the Mox@CG-Alg NPs displayed low MIC, higher zone of bacterial growth inhibition, and cell death compared to free Mox. A significant reduction of bacterial load was observed in the BK-induced mouse model.

Utilization of nanotechnology and experimental design in the development and optimization of a posaconazole‒calendula oil nanoemulgel for the treatment of mouth disorders.

Introduction: Essential oil‒based nanoemulsions (NEs) are the subjects of extensive investigation due to their potential to address a variety of oral health issues. NEs are delivery systems that improve lipid medicine solubility and distribution to intended sites. The goal of the current study was to create and enhance a self-nanoemulsifying drug delivery paradigm based on calendula oil (CO) and decorated with chitosan (CS) that could deliver posaconazole (PSZ) for the treatment of gingivitis. Method: Employing a response-surface Box‒Behnken design, PSZ-CO-CS NEs were created with varying amounts of PSZ (10, 15, and 20mg), percentages of CO (6%, 12%, and 18%), and percentages of CS (0.5%, 1.5%, and 2.5%). Results and conclusion: The optimized formulation resulted in a 22-mm bacterial growth suppression zone, 25-mm fungal growth inhibition zone, droplet sizes of 110nm, and a viscosity of 750 centipoise (cP). Using the appropriate design, the ideal formulation was produced; it contained 20mg of PSZ, 18% of CO, and 1.35% of CS. Furthermore, the optimal formulation had a more controlled drug release, larger inhibition zones of bacterial and fungal growth, and desirable rheologic properties. Additionally, the optimized formulation substantially lowered the ulcer index in rats when tested against other formulations. Thus, this investigation showed that PSZ-CO-CS NEs could provide efficient protection against microbially induced gingivitis.

Impact of Plant Extracts and Chemicals against Bacterial Blight of Pomegranate

Bacterial blight of pomegranate caused by Xanthomonas axanopodis pv. puniceae, is devastating disease causing huge losses to the production especially in pakistan last few years. It is caused by a bacterium Xanthomonas axanopodis pv. puniceae which damage the crop severely in favorable environment conditions. Current research was conducted to evaluate four chemicals; Streptomycin, Copper oxy-chloride, kasugmicine, Oxy-tetracycline and four plant extracts; Citrullus colocynths, Calotropis gigantea, Accica nelotica, and Moringa oleifera against Xanthomonas axanopodis pv. puniceae.. In Laboratory conditions result showed that maximum inhibition zone of bacterial growth was expressed by streptomycin sulphate (38mm) followed by copper oxycholoride (38mm), kasugmicine (34mmm), and oxy-tetracycline (24mm) respectively at 3% concentration after 72 hours' time interval. In plant extracts, maximum inhibition was expressed by Moringa olifera (27.6mm) followed by Accica nelotica (26.3mm), Citrullus colocynths (16.6mm), and Calotropis gigantea (19.3mm) at 25%, 35% and 45% concentration. In laboratory conditions significant results shown by chemical (copper oxycholoride) and plant extract (Moringa olifera) Copper oxycholoride were applied in greenhouse. The copper oxychloride shows lowest disease severity (23%) while Moringa olifera 36% as compared to positive and negative control.

ANTIBACTERIAL PROPERTIES OF COMMERCIAL PEPPERMINT ESSENTIAL OIL AGAINST SOME GRAM-POSITIVE AND GRAM-NEGATIVE BACTERIA

The authors of this article conducted research and studied the antibacterial properties of commercial peppermint essential oil (PEO) against several gram-positive and gram-negative bacteria provided by Polish manufacturers of essential oils (Naturalne Aromaty sp. z o.o., Klaj, Poland). Therefore, to conduct research with the aim of to study the antibacterial properties of commercial peppermint essential oil (PEO), an antimicrobial susceptibility test (Kirby-Bauer diffusion test) was used to measure diameters of bacterial growth inhibition zones). In the current study, Gram-negative strains such as Escherichia coli (Migula) Castellani and Chalmers (ATCC® 25922™ ), Escherichia coli (Migula) Castellani and Chalmers (ATCC® 35218™ ), Pseudomonas aeruginosa (Schroeter) Migula (ATCC® 27853™ ) and Gram-positive strains such as Staphylococcus aureus subsp. aureus Rosenbach (ATCC® 29213™ ), methicillin-resistant (MRSA), mecA positive Staphylococcus aureus (NCTC® 12493), Enterococcus faecalis (Andrewes and Horder) Schleifer and Kilpper-Balz (ATCC® 51299™ ) (resistant to vancomycin; sensitive to teicoplanin) and Enterococcus faecalis (Andrewes and Horder) Schleifer and Kilpper-Balz (ATCC® 29212™ ) were used. Results of the current study revealed that resistant to the PEO were Gram-negative bacterial strains, such as E. coli (Migula) Castellani and Chalmers (ATCC® 35218™ ) and P. aeruginosa (Schroeter) Migula (ATCC® 27853™ ) strains. The authors found that the diameters of the inhibition zones after application of PEO were similar to the control samples (96% ethanol). It was also found that after the application of REO, the increase in the diameters of the inhibition zones was 60.3% (p < 0.05) for the Escherichia coli strain (Migula) Castellani and Chalmers (ATCC® 25922™ ) compared to control samples (96% ethanol ). Accordingly, Gram-positive strains such as S. aureus subsp. aureus Rosenbach (ATCC® 29213™ ) and methicillin-resistant S. aureus (NCTC® 12493) were equally resistant to PEO, similarly. On the other hand, Enterococcus faecalis (Andrewes and Horder) Schleifer and Kilpper-Balz (ATCC® 29212™ ) and Enterococcus faecalis (Andrewes and Horder) Schleifer and Kilpper-Balz (ATCC® 51299™ ) were sensitive to PEO. After the application of PEO, the largest diameters of inhibition zones were observed for the E. faecalis strain. The results suggest that commercial peppermint essential oil provided by Polish essential oil manufacturers (Naturalne Aromaty sp. z o.o., Kłaj, Poland) possesses some noteworthy antimicrobial properties. In vivo studies are necessary to calculate the effective dose of EOs and determine their possible side effects and toxicity.

Potencijal esencijalnog ulja ambrozije (Ambrosia artemisiifolia L.) u supresiji bakterijskog rasta

Plant essential oils have antimicrobial properties that are widely used in food preservation and aromatherapy and also have various medicinal uses. The way essential oil will affect the microorganisms widely depends on the type of microorganism and the type of oil used. Gram-positive bacteria are more sensitive to essential oil antimicrobial traits than gram-negative bacteria. Ragweed (Ambrosia artemisiifolia L.) essential oil has a wide range of antimicrobial applications; this oil is recommended for potential health benefits because of its antioxidant activity. The aim of this study was to examine the antibacterial potential of the ragweed essential oils against Escherichia coli, Bacillus subtilis and Salmonella spp. The antimicrobial effect of the oil was examined using a diffusion method test. Before the experiment, the inhibition zone of bacterial growth was determined using the antibiotic gentamicin. The results of the study confirmed the antimicrobial effect of ragweed oil on the growth of Salmonella spp. and Bacillus subtilis growth, while this oil had negligible effect on Escherichia coli growth. Moderate susceptibility of Salmonella spp. and Bacillus subtilis and resistance of E. coli was observed after ragweed treatment compared to the standard inhibition zone values proposed by EUCAST. This research confirms the potential application of ragweed essential oil in the inhibition of Salmonella spp. and Bacillus subtilis growth.

Ag-Containing Carbon Nanocomposites: Physico-Chemical Properties and Antimicrobial Activity

The subject of the present work is the synthesis and analysis of the structural and morphological properties of Ag-containing carbon composites and the investigation of their practical application in water purification and disinfection. A series of composites were synthesized by carbonization of resorcinol–formaldehyde polymers filled with Ag-containing fumed silica under an inert atmosphere at 800 °C. The as-synthesized micro- and mesoporous carbon composites were characterized by their specific surface area of 466–529 m2/g. The suitability of the composites for flow-through filters was evaluated by kinetic studies on the adsorption of 4-chlorophenol. The composite with the highest amount of metallic nanophase showed the most effective kinetics with a rate constant (log k) and half-life (t0.5) of −2.07 and 81 min, respectively. The antimicrobial susceptibility was determined against Gram-positive (Staphylococcus aureus ATCC 25923) and Gram-negative strains (Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 700603, Pseudomonas aeruginosa ATCC 27853, and Acinetobacter baumannii ATCC 19606). The zones of bacterial growth inhibition correlated with the silver nanoparticle content and were the lowest for RFC-02 (10–12 mm) and the highest for the RFC-1 composite (15–16 mm), resulting from the increase in number of evenly distributed small Ag nanoparticles (3–5 nm) in the samples.

Psidium guajava, Phalaenopsis sp., Syzygium aromaticum e Cinnamomum verum como inibidores naturais do crescimento de Streptococcus mutans

Currently, an increase on research on plant extracts seeking therapeutic action in the control of diseases of the oral cavity are observed, especially dental caries. Several plant extracts are already incorporated into toothpastes and mouthwashes. The objective of this study was to evaluate in vitro the antibacterial action of Psidium guajava (guava), Phalaenopsis sp. (orchid), Syzygium aromaticum (clove) and Cinnamomum verum (Ceylon cinnamon) extracts. The tests were carried out with the oral cavity bacteria Streptococcus mutans. The antibacterial activity of plant extracts was evaluated by the disk diffusion method in agar and by minimal inhibitory concentration (MIC). The tests showed the effectiveness of the extracts through inhibition zones of bacterial growth (from 5 to 20 mm in diameter) and MIC of 100 mg. mL-1 Phalaenopsis sp. extract and 50 mg. mL-1 P. guajava extract. Complementary studies will be needed to evaluate other pharmacological actions in order to use these plant extracts in different dental specialties and mainly in the treatment and prophylaxis of dental caries.

Undecylenic acid and N,N-dibutylundecenamide as effective antibacterials against antibiotic-resistant strains

Evaluation of undecylenic acid (UA) and its tertiary amide N,N-dibutylundecenamide (DBUA) activity­ in vitro against the standard and antibiotic-resistant Escherichia coli and Staphylococcus aureus strains was carried out. The antibacterial potential of the acid and its amide at 2.5 and 5.0 μM concentration both against gram-positive bacteria (S. aureus) and gram-negative (E. coli) cultures was confirmed by monitoring the diameter of the bacterial growth inhibition zones. The docking study identified methionine aminopeptidase (MAP) as the most energy-favorable potential biotarget associated with the drug resistance of E. coli and S. aureus with a binding energy in the range from -8.0 to -8.5 kcal/mol. The ligands complexation was due to the formation of hydrogen bonds with ASP108, HIS171, HIS178, GLU204, GLU235, HIS76, ASP104, GLU233, ASP93 and metal-acceptor interactions with Co2+. Overall, the results indicated that UA and DBUA activity against antibiotic-resistant strains creates prospects for the development of new antibacterial formulations. Moxifloxacin HCl-Incorporated Aqueous-Induced Nitrocellulose-Based In Situ Gel for Periodontal Pocket Delivery.

A drug delivery system based on an aqueous-induced in situ forming gel (ISG) consists of solubilizing the drug within an organic solution of a polymer using a biocompatible organic solvent. Upon contact with an aqueous medium, the solvent diffuses out and the polymer, designed to be insoluble in water, solidifies and transforms into gel. Nitrocellulose (Nc), an aqueous insoluble nitrated ester of cellulose, should be a promising polymer for an ISG using water induction of its solution to gel state via phase inversion. The aim of this investigation was to develop and evaluate a moxifloxacin HCl (Mx)-incorporated aqueous-induced Nc-based ISG for periodontitis treatment. The effects of different solvents (N-methyl pyrrolidone (NMP), DMSO, 2-pyrrolidone (Py), and glycerol formal (Gf)) on the physicochemical and bioactivity properties of the ISGs were investigated. The viscosity and injection force of the ISGs varied depending on the solvent used, with Gf resulting in higher values of 4631.41 ± 52.81 cPs and 4.34 ± 0.42 N, respectively. All ISGs exhibited Newtonian flow and transformed into a gel state upon exposure to the aqueous phase. The Nc formulations in DMSO showed lower water tolerance (12.50 ± 0.72%). The developed ISGs were easily injectable and demonstrated water sensitivity of less than 15.44 ± 0.89%, forming a gel upon contact with aqueous phase. The transformed Nc gel effectively prolonged Mx release over two weeks via Fickian diffusion, with reduced initial burst release. Different solvent types influenced the sponge-like 3D structure of the dried Nc ISGs and affected mass loss during drug release. Incorporating Nc reduced both solvent and drug diffusion, resulting in a significantly narrower zone of bacterial growth inhibition (p < 0.05). The Mx-incorporated Nc-based ISGs exhibited efficient antibacterial activity against four strains of Staphylococcus aureu and against periodontitis pathogens including Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis. This study suggests that the developed Mx-incorporated Nc-based ISGs using DMSO and NMP as the solvents are the most promising formulations. They exhibited a low viscosity, ease of injection, and rapid transformation into a gel upon aqueous induction, and they enabled localized and prolonged drug release with effective antibacterial properties. Additionally, this study represents the first reported instance of utilizing Nc as the polymer for ISG. Further clinical experiments are necessary to evaluate the safety of this ISG formulation.

To Evaluate the Antimicrobial Activity of PRP and PRF with and without Nanosilver.

To evaluate the antimicrobial activity of PRP and PRF with and without nanosilver. The materials were tested in powdered form is nanosilver. The nanosilver particles was mixed to form with PRP and PRF so as to placed in a wells followed the groups are experimental groups; Group I: PRP + nanosilver particles, Group II: PRF + nanosilver and control group: PRP and PRF and normal saline. Silver nanoparticles was tested at concentrations of 50 μ gram per mL. The powder was prepared for each group with identical amount of the powder (milligram/mg) and then mixed with 1 milliliter liquid. The plates are then incubated at 37°C under appropriate atmospheric conditions (80% N2, 10% CO2, 10% H2) for 24 hours, 48 hours, and 72 hours under anaerobic conditions in a CO2 incubator. The diameters of the zones of bacterial and fungal growth inhibition around the wells containing the test substances are then recorded after the period of incubation. The inhibitory zone determined in millimeter by measuring scale the shortest distance between the outer margin of the well and initial microbial as well as fungal growth. The experiments were performed 20 times and the mean and standard deviations of the inhibitory zones were calculated. Platelet rich fibrin is mixed with nanosilver particles showed higher antimicrobial efficacy than platelet rich plasma with nanosilver and simple platelet rich plasma and platelet rich fibrin are equivalent when it is placed against the anaerobic bacteria E.faecalis and yeast like fungi Candida albicans, respectively. Groups presented with antimicrobial efficacy in this order- Group IV > Group II > Group III > Group I.

Antimicrobial peptide-loaded decellularized placental sponge as an excellent antibacterial skin substitute against XDR clinical isolates.

Post-wound infections have remained a serious threat to society and healthcare worldwide. Attempts are still being made to develop an ideal antibacterial wound dressing with high wound-healing potential and strong antibacterial activity against extensively drug-resistant bacteria (XDR). In this study, a biological-based sponge was made from decellularized human placenta (DPS) and then loaded with different concentrations (0, 16µg/mL, 32µg/mL, 64µg/mL) of an antimicrobial peptide (AMP, CM11) to optimize an ideal antibacterial wound dressing. The decellularization of DPS was confirmed by histological evaluations and DNA content assay. The DPS loaded with different contents of antimicrobial peptides (AMPs) showed uniform morphology under a scanning electron microscope (SEM) and cytobiocompatibility for human adipose tissue-derived mesenchymal stem cells. Antibacterial assays indicated that the DPS/AMPs had antibacterial behavior against both standard strain and XDR Acinetobacter baumannii in a dose-dependent manner, as DPS loaded with 64µg/mL showed the highest bacterial growth inhibition zone and elimination of bacteria under SEM than DPS alone and DPS loaded with 16µg/mL and 32µg/mL AMP concentrations. The subcutaneous implantation of all constructs in the animal model demonstrated no sign of acute immune system reaction and graft rejection, indicating in vivo biocompatibility of the scaffolds. Our findings suggest the DPS loaded with 64µg/mL as an excellent antibacterial skin substitute, and now promises to proceed with pre-clinical and clinical investigations.

Utilization of experimental design in the formulation and optimization of hyaluronic acid–based nanoemulgel loaded with a turmeric–curry leaf oil nanoemulsion for gingivitis

Numerous problems affect oral health, and intensive research is focused on essential oil–based nanoemulsions that might treat prevent or these problems. Nanoemulsions are delivery systems that enhance the distribution and solubility of lipid medications to targeted locations. Turmeric (Tur)- and curry leaf oil (CrO)–based nanoemulsions (CrO-Tur-self-nanoemulsifying drug delivery systems [SNEDDS]) were developed with the goal of improving oral health and preventing or treating gingivitis. They could be valuable because of their antibacterial and anti-inflammatory capabilities. CrO-Tur-SNEDDS formulations were produced using the response surface Box-Behnken design with different concentrations of CrO (120, 180, and 250 mg), Tur (20, 35, and 50 mg), and Smix 2:1 (400, 500, and 600 mg). The optimized formulation had a bacterial growth inhibition zone of up to 20 mm, droplet size of less than 140 nm, drug-loading efficiency of 93%, and IL-6 serum levels of between 950 ± 10 and 3000 ± 25 U/ml. The optimal formulation, which contained 240 mg of CrO, 42.5 mg of Tur, and 600 mg of Smix 2:1, was created using the acceptable design. Additionally, the best CrO-Tur-SNEDDS formulation was incorporated into a hyaluronic acid gel, and thereafter it had improved ex-vivo transbuccal permeability, sustained in-vitro release of Tur, and large bacterial growth suppression zones. The optimal formulation loaded into an emulgel had lower levels of IL-6 in the serum than the other formulations evaluated in rats. Therefore, this investigation showed that a CrO-Tur-SNEDDS could provide strong protection against gingivitis caused by microbial infections.

Screening assessment of antibacterial activity of Thymus serpyllum L., Thymus marsсhallianus Willd. and Pimpinella anisum L. essential oils against uropathogens, isolated from pregnant women

Plant essential oils of certain genera and species are important sources of antimicrobial substances.The goal of this study was a screening assessment of antibacterial activity of Thymus serpyllum L., Thymus marsсhallianus Willd. and Pimpinella anisum L. essential oils against opportunistic gram-negative uropathogens. The essential oils were obtained by steam hydrodistillation, and the screening assessment of their antibacterial activity was performed by the disk diffusion method. Two standard species (Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 25922) and 6 clinical strains (1 — P. aeruginosa, 5 — E. coli), isolated from the pregnant women with infections of the urinary passages, were used as the test bacterial cultures. The study determined that T. marsсhallianus Willd. and T. serpyllum L. essential oils possess strong inhibitory action on all test cultures until the absence of the bacterial lawn. P. anisum L. essential oil did not show antibacterial activity against all test strains, no zones of visible bacterial growth inhibition were formed. T. serpyllum L. and T. marsсhallianus Willd. essential oils are promising antibacterial agents against uropathogens of the species P. aeruginosa and E. coli.

SOCIALIZATION OF ANTIBACTERIAL ACTIVITY OF KERSEN LEAF EXTRACT (Muntingia calabura L) AGAINST BACTERIA Staphylococcus aureus

Kersen (Muntingia calabura) is a shady plant that is usually used as a shade. Cherry leaves contain several secondary metabolites such as flavonoid, tannin and terpenoid. Flavonoid can inhibit bacterial growth by denaturing proteins and damaging cell membranes and dissolving fats contained in cell walls. According to the World Health Organization as many as 25 million deaths worldwide in 2011, one third were caused by infectious diseases. One of the most common infectious diseases caused by the bacterium Staphyloccus aureus. This socialization aims to provide information to the public about the antibacterial ability of kersen leaves against staphylococus aureus. The concentration of the extract used was 15%, 30%, 45% and 60% and erythromycin as a positive control. From the test results, it can be concluded that the largest inhibition zone diameter is at a concentration of 60% with an inhibition zone diameter of 6.78 mm and the smallest at a concentration of 30%, which is 0.42 mm. Concentration of 45% has (weak) activity inhibiting bacterial growth with an inhibition zone of 2.13 mm, while at a concentration of 15% it has no activity in the inhibition zone of bacterial growth

Synthesis of antibacterial hydroxypropyl methylcellulose and silver nanoparticle biocomposites via solution plasma using silver electrodes

The biocomposites of hydroxypropyl methylcellulose (HPMC)/silver nanoparticles (AgNPs) were synthesized using the solution plasma process (SPP). HPMC/AgNPs were synthesized in 1–5 % HPMC solutions using silver electrodes. UV–Vis spectroscopy showed a peak near 400 nm and the peak increased as the concentration of HPMC and discharge time increased. FTIR analysis indicated no change in the chemical structure of the HPMC based biocomposites. Spherical shaped AgNPs with size ranges about 2–18 nm and well dispersed in the porous HPMC matrices with fringed edges were observed by TEM and SEM/EDS analyses. The synthesized biocomposites were found to be thermo-stable by TGA analysis. The inhibition zones of bacterial growth formed by the HPMC/AgNPs biocomposites were in the range of 8–14.3 mm; minimal inhibition concentrations, in the range of 10–15 μg·mL−1 for Gram-negative bacteria; 25–30 μg·mL−1 for Gram-positive bacteria. The biocomposites were non-toxic to the HEK293 cells up to 125 μg·mL−1. The results indicated that the synthesis of antibacterial agents in the HPMC matrix using silver electrodes via SPP would be an efficient and safe way for the development of biopolymer based antimicrobials and wound healing biomaterials.

FORMULASI DAN UJI AKTIVITAS ANTIBAKTERI SEDIAAN GEL HAND SANITIZER EKSTRAK ETANOL DAUN BAYAM MERAH (Amaranthus Tricolor L.) TERHADAP BAKTERI Staphylococcus aureus &amp; Eschericia coli

Red spinach has function as skin antiseptic. It has antibacterial activity because it contains compounds such as tannins, flavonoids, saponins, and steroids. The purpose of formulation and test of antibacterial activity of hand sanitizer gel preparation red spinach extract and evaluation of physical quality of hand sanitizer. Antibacterial activity test was measured by diffusion method using paper disk to measuring the diameter of the bacterial growth inhibition zone of Escherichia coli and Staphylococcus aureus. Concentration extract ethanol of leaf red spinach (and formula of hand sanitizer gel used (F1) blanco; (F2) 5%; (F3) 10%; (F4) 20% and (F5) control positive/detol. Evaluation of physical properties of hand sanitizer gel include of: homogeneity test, stability test and pH test. The results has found showed that antibacterial activity test of gel extract leaves red spinach (on Escherichia coli inhibited at without extract/blanco was 0 mm, concentration of 5% with diameter of inhibit was 14,99 mm, concentration of 10% with diameter of inhibit was 17,26 mm, concentration of 20% with diameter of inhibit was 18,26 mm and using control positive was 19.33. And then for bacteri Staphylococcus aures at concentration of without extract/blanco was 0 mm, 5% with diameter of inhibit was 15,5 mm, concentration of 10% with diameter of inhibit was 15,96 mm and concentration of 20% with diameter of inhibit was 17,43 mm and using control positive was 18.33 mm. So we can conclude that hand sanitizer gel of leaf red spinach have less inhibited growth against Escherichia coli and Staphylococcus aureus.

Biosynthesis of ZnO Nanoparticles from Aqueous Extract of Moringa Oleifera L.: Its Application as Antibacterial and Photocatalyst

ZnO nanoparticles were successfully synthesized by the biosynthesis method using water extract of Moringa leaf (Moringa oleifera L.) as a reducing agent and stabilizer as well as a capping agent. This study aimed to characterize ZnO nanoparticles and their application as antibacterial and photocatalyst. Characterization was carried out using Fourier Transform Infrared (FT-IR) Spectroscopy and X-Ray Diffraction (XRD). The biosynthesis ZnO nanoparticles have a hexagonal wurtzite phase with an average crystal size of 16.97 nm and a crystallinity level of 78.49%. Antibacterial activity of ZnO nanoparticles with concentrations of 3%, 6%, 9% resulted in an average inhibition zone of bacterial growth in S. aureus was 19.6 0.16; 21.8 0.33; 24.3 0.14 and in E.coli it is 9.87 0.2; 11.3 0.04; 11.57 0.06. The greater the concentration of ZnO nanoparticles, the larger the diameter of the inhibition zone. The result of photodegradation of methylene blue is greatest with variations in the mass of ZnO nanoparticles and the concentration of methylene blue is 95% with a nanoparticle mass of 120 mg, methylene blue concentration of 10 ppm, and an irradiation time of 150 minutes.

Evaluation of antibacterial and mechanical features of dental adhesives containing colloidal gold nanoparticles

In this study, colloidal gold nanoparticles (NPs) with sizes less than 20 nm were added to the dentin adhesive in various concentrations to achieve favorable antibacterial properties. The effect of gold NPs on mechanical, antibacterial, and cytotoxicity features of adhesive was evaluated systematically. It was shown that the addition of gold NPs at the concentration of 5X (i) increased flexural strength of dentin tooth adhesive by 75%, (ii) increased the micro-shear bond strength by 60%, and (iii) increased the tensile diameter strength of adhesive by 65% compared to the based adhesive. Multiscale modeling was used to simulate and calculate stress and strain distribution around the nanoparticles and within the adhesive matrix. Antibacterial properties of adhesives containing gold NPs were examined using disk diffusion and the pour plate methods. The results showed that adhesive with the 5X concentration of gold NPs had a bacterial growth inhibition zone with a diameter of 2 mm is in the disk diffusion method which indicates the antibacterial characteristics of adhesive at this concentration. Also, the antibacterial effect of adhesive-containing NPs was observed in the pour plate method and the reduction of bacterial growth is increased by the addition of nanoparticles. The cytotoxicity of NPs is examined in pure form and combination with dental adhesive using the MTT assay. The results showed that pure gold NPs have shown no toxicity for the growth of cells and the incorporation of gold NPs has increased the cell viability in the base adhesive.

Antagonistic and Antibacterial Activity of Stapylocossus aureus and Isolates of Oral Bacteria from the Endogenous Fungus Apis dorsata Binghami Nest

Apis Dorsata Binghami honey bee is a honey bee endemic to Indonesia, living naturally in the forests of Sulawesi. This study aimed to obtain isolates and characteristics of endogenous fungi antibiotic activity from Apis dorsata Binghami nest. The research consisted of isolation of fungi from beehives using potato dextro agar, pure culture of fungi, antagonist test and antibiotic test using disc diffusion method. Antibiotic test was performed on oral bacteria and Staphylococcus aureus. The results obtained six isolates of fungi from the nest of Apis dorsata Binghami, namely isolates FAB1, FAB2, FAB3, FAB4, FAB5 and FAB6. The results of the antagonist test showed that the isolates FAB2 and FAB3 had the best antagonist properties, while the FAB6 isolates had the weakest antagonist properties. The FAB2 isolate showed the best bacterial growth inhibition zone average for the isolates of oral bacteria and Staphylococcus aureus. The inhibition zone lasts up to 3 x 24 hours so that the activity of the bacteria is bactericidal. From the results of this study, it can be concluded that the endogenous fungus Apis dorsata Binghami is a potential source of antibacterial bioactives.
