A Comprehensive Review on Anti-Cancer Properties of Oxalis corniculata

Oxalis corniculata L. belongs to the Family (Oxalidaceae), commonly known as “creeping woodsorrel, procumbent yellow sorrel, or sleeping beauty, " a common name. Oxalis corniculate contains several compounds like β-sitosterol, betulin, 4-hydroxybenzoic acid, ethyl gallate, methoxyflavones, apigenin, and 7-O-β-D-glucopyranoside were previously isolated from the whole plant of Oxalis corniculata. linn. The review reveals that wide ranges of phytochemical constituents have been isolated from the plant like flavonoids, tannins, phytosterols, phenol, glycosides, fatty acids, galactose-glycerolipid, and volatile oil. The leaves contain flavonoids, iso vitexin, and vitexin-2”- O- beta – D- glucopyranose. It is a rich source of essential fatty acids like palmitic, oleic, linoleic, linolenic, and stearic acids. It has been reported that the plant contains anti-inflammatory, anxiolytic, anticonvulsant, antifungal, antiulcer, antinociceptive, anticancer, antidiabetic, hepatoprotective, hypolipidemic, abortifacient, antimicrobial, and wound healing properties.

The Glucans Mushrooms: Molecules of Significant Biological and Medicinal Value

Mushroom polysaccharides, key components of fungal cell walls, exhibit various biological properties and hold significant medicinal and industrial value. These polysaccharides are known for their medicinal properties like antitumor, antioxidant, anticancer, immunomodulatory, and antiviral properties. Mushroom polysaccharides, particularly β-glucans, α-glucans, and chitin, have been associated with various health benefits. β-glucans are well studied for their bioactivities, while α-glucans and chitin have gained attention for their prebiotic, antimicrobial, and wound-healing properties. The therapeutic effects of these polysaccharides are closely linked to their chemical structures, including molecular weight, monosaccharide composition, and glycosidic bond types. This work aims to review the studies on mushroom polysaccharides, with a particular focus on their structural composition to deepen medicinal properties of mushroom polysaccharides. Also, the extraction methods and the pharmaceutical application of polysaccharides will be revised in this work.

GC-MS/MS Analysis and Wound Repair Potential of Urtica dioica Essential Oil: In silico Modeling and In vivo Study in Rats.

The study aimed to assess the antioxidant and wound healing properties of Urtica dioica essential oil (UDEO) through a comprehensive evaluation involving in silico, in vitro, and in vivo analyses. The phytochemistry of UDEO was also investigated to identify trace compounds crucial. Various injection methods of the multimode inlet (MMI) in chromatography were investigated to attain lower instrumental detection limits. Subsequently, in silico studies were employed to delve deeper into the potential biological activities of the identified compounds. Standard antioxidative tests, encompassing ABTS•+ and TAC, were performed. In vivo tests centered on wound healing were implemented using rat models. The rats were randomly allocated to four groups: saline solution, vaseline vehicle, cytol centella, and 5% UDEO ointment. Wound healing progress was evaluated through a chromatic study. Gas chromatography combined with triple quadrupole mass spectrometry (GC-MS/MS) analysis revealed the presence of 97 thermolabile compounds in UDEO. Subsequent in silico studies unveiled the potential of identified compounds to inhibit COX-2, TNF-α, and IL-6, suggesting a possible enhancement of anti-inflammatory responses and healing processes. In vitro tests elucidated the notable antioxidant capacity of UDEO, a finding reinforced by wound healing data, revealing a substantial closure rate of 89% following the topical application of UDEO. Notably, fibrinogen and C-reactive protein (CRP) levels were significantly reduced, indicating minimized oxidative stress damage compared to control. Additionally, UDEO exhibited an increase in antioxidant enzyme activities compared to control. The study concludes that UDEO possesses significant antioxidant and wound-healing properties, supported by its rich phytochemical composition. The findings suggest its potential application in therapeutic interventions for oxidative stress and inflammatory conditions.

Bergenin inhibits growth of human cervical cancer cells by decreasing Galectin-3 and MMP-9 expression

Cervical cancer is still the leading cause of cancer mortality worldwide even after introduction of vaccine against Human papillomavirus (HPV), due to low vaccine coverage, especially in the developing world. Cervical cancer is primarily treated by Chemo/Radiotherapy, depending on the disease stage, with Carboplatin/Cisplatin-based drug regime. These drugs being non-specific, target rapidly dividing cells, including normal cells, so safer options are needed for lower off-target toxicity. Natural products offer an attractive option compared to synthetic drugs due to their well-established safety profile and capacity to target multiple oncogenic hallmarks of cancer like inflammation, angiogenesis, etc. In the current study, we investigated the effect of Bergenin (C-glycoside of 4-O-methylgallic acid), a natural polyphenol compound that is isolated from medicinal plants such as Bergenia crassifolia, Caesalpinia digyna, and Flueggea leucopyrus. Bergenin has been shown to have anti-inflammatory, anti-ulcerogenic, and wound healing properties but its anticancer potential has been realized only recently. We performed a proteomic analysis of cervical carcinoma cells treated with bergenin and found it to influence multiple hallmarks of cancers, including apoptosis, angiogenesis, and tumor suppressor proteins. It was also involved in many different cellular processes unrelated to cancer, as shown by our proteomic analysis. Further analysis showed bergenin to be a potent-angiogenic agent by reducing key angiogenic proteins like Galectin 3 and MMP-9 (Matrix Metalloprotease 9) in cervical carcinoma cells. Further understanding of this interaction was carried out using molecular docking analysis, which indicated MMP-9 has more affinity for bergenin as compared to Galectin-3. Cumulatively, our data provide novel insight into the anti-angiogenic mechanism of bergenin in cervical carcinoma cells by modulation of multiple angiogenic proteins like Galectin-3 and MMP-9 which warrant its further development as an anticancer agent in cervical cancer.

Advanced stability-indicating gradient elution reverse phase-high performance liquid chromatography method development and validation for simultaneous estimation of rutin and curcumin using an analytical quality by design approach

This research delves into a systematic Analytical Quality by Design (AQbD) approach to develop and validate a stability-indicating robust RP-HPLC method for the simultaneous determination of Rutin and Curcumin, compounds known for their anti-inflammatory, antioxidant, and wound-healing properties. Utilizing QbD principles, critical method parameters (CMPs) were identified and screened using design of experiments (DoE) and optimized via response surface methodology (RSM) with Central Composite Design (CCD). The optimized chromatographic separation used a Phenomenex Luna C18 column with gradient elution, using methanol and 0.1% formic acid, a flow rate of 1 ml/min, and detection at 345 nm (isosbestic point). The retention times were 5.63 mins for Rutin and 17.01 mins for Curcumin. The stability indication of the developed HPLC method was performed, and validation parameters were meticulously conducted following the ICH Q2(R1) guidelines, demonstrating linearity over 2-10 μg/ml for Rutin and 5-25 μg/ml for Curcumin, with correlation coefficients of 0.9999 and 0.9998, respectively. Precision studies showed %RSD within acceptable limits, and accuracies ranged between 97-100%. This validated HPLC method offers a reliable, sensitive approach for simultaneous quantifying Rutin and Curcumin, suitable for routine analysis in pharmaceutical laboratories and quality control.

Investigating the Multi-Targeted Pharmacological profile of an Exopolysaccharide from Bacillus rugosus SYG20 via In Vitro Evaluation of its Antioxidant, Anti-inflammatory, Anti-diabetic, Wound Healing, and Antimicrobial Properties

IntroductionExopolysaccharides (EPSs) derived from marine microorganisms are a newly recognized reservoir of bioactive therapeutic compoundsMaterial and methodsWe isolated a high EPS-yielding bacterial strain from the Red Sea, identified as Bacillus rugosus SYG20. Its purified EPS (EPSR9) contains 45.33% uronic acid, 9.98% sulfate groups, and 5.40% N-acetyl glucosamine. The HPLC chromatogram revealed four monosaccharides - glucose, xylose, galacturonic acid, and arabinose, in a distinct molar ratio of 2:3:1:1. EPSR9 showed a wide array of bioactivities.ResultsIt displayed antioxidant activity with an IC50 of 25.6 μg/ml in the DPPH assay and a total antioxidant capacity (TAC) of 417.77μg/ml equivalent AAE and 62.67 μg/ml equivalent AAE in Ferric reducing antioxidant power (FRAP) assays. It exhibited substantial anti-inflammatory properties. The anticoagulant effect of the EPS was demonstrated by a dose-dependent increase in prothrombin time. The scratch assay resulted in a 72.66% increase in wound closure, promoting in vitro wound healing after 48 h. Anti-obesity activity was evidenced by 83.8% lipase inhibition at 1000 μg/ml with IC50 of 107.73μg/ml. EPSR9 demonstrated inhibitory effects on α-amylase with an IC50 value of 14.37μg/ml and α-glucosidase with an IC50 value of 26.73 μg/ml, highlighting its potential as an anti-diabetic agent. Then, EPS showed bactericidal properties with MBC/MIC≤2 against both G+ve and G-ve bacteria, Staphylococcus aureus , Enterococcus faecalis (MIC=3.9μg/ml), Salmonella typhi, and Helicobacter pylori.ConclusionsThe marine EPSR9 exhibited considerable potential for pharmaceutical applications as a multi-bioactive microbial metabolite. Its in vivo potency and mechanisms of action warrant further investigation.

Symphytum genus—from traditional medicine to modern uses: an update on phytochemistry, pharmacological activity, and safety

AbstractWith around 34 recognized species, Symphytum genus (comfrey) has a noteworthy position within the Boraginaceae family. Comfrey species have been empirically used since ancient times as wound-healing and skin-regenerating agents in ulcers, wounds, bone fractures, and rheumatic complaints. This review aims to provide a thorough examination of recent scientific advances and challenges within the Symphytum genus, covering data published between 2013 and 2023. It delivers an updated overview of the taxonomy, ethnopharmacological uses, chemical composition, and pharmacological activities of the genus. Special emphasis is put on molecular identification methods for species taxonomy, emerging extraction technologies for comfrey phytochemicals, metabolomics techniques for mapping chemical complexity, modern bioassay platforms revealing its poly-pharmacology, formulation strategies, and remediation approaches for removal of toxic pyrrolizidine alkaloids (PAs). For instance, recent metabolomic studies employing advanced spectro-chromatographic techniques have revealed a diverse chemical composition of comfrey plants, including polysaccharides, allantoin, benzoic and cinnamic acid derivatives, flavonoids, fatty acids, and unsaturated necine-structure-based PAs. The mechanisms underlying their anti-inflammatory, analgesic, wound-healing, anti-irritant, and osteo-regenerative properties were targeted in modern pharmacological setups. Thus, key compounds like allantoin, rosmarinic acid, globoidnans A and B, rabdosiin, and comfreyn A, have been identified as significant contributors to the anti-inflammatory and wound-healing effects of Symphytum-derived preparations. Despite their well-established clinical use, concerns about PAs-induced toxicity have prompted the development of novel PA remediation strategies, enabling the production of comfrey extracts with enhanced safety profiles that can meet the regulatory standards imposed by authorities.

Design and evaluation of a bilayered dermal/hypodermal 3D model using a biomimetic hydrogel formulation

Due to the limitations of the current skin wound treatments, it is highly valuable to have a wound healing formulation that mimics the extracellular matrix (ECM) and mechanical properties of natural skin tissue. Here, a novel biomimetic hydrogel formulation has been developed based on a mixture of Agarose-Collagen Type I (AC) combined with skin ECM-related components: Dermatan sulfate (DS), Hyaluronic acid (HA), and Elastin (EL) for its application in skin tissue engineering (TE). Different formulations were designed by combining AC hydrogels with DS, HA, and EL. Cell viability, hemocompatibility, physicochemical, mechanical, and wound healing properties were investigated. Finally, a bilayered hydrogel loaded with fibroblasts and mesenchymal stromal cells was developed using the Ag-Col I-DS-HA-EL (ACDHE) formulation. The ACDHE hydrogel displayed the best in vitro results and acceptable physicochemical properties. Also, it behaved mechanically close to human native skin and exhibited good cytocompatibility. Environmental scanning electron microscopy (ESEM) analysis revealed a porous microstructure that allows the maintenance of cell growth and ECM-like structure production. These findings demonstrate the potential of the ACDHE hydrogel formulation for applications such as an injectable hydrogel or a bioink to create cell-laden structures for skin TE.

Chitosan/rutin multifunctional hydrogel with tunable adhesion, anti-inflammatory and antibacterial properties for skin wound healing

Effective wound care remains a significant challenge due to the need for infection prevention, inflammation reduction, and minimal tissue damage during dressing changes. To tackle these issues, we have developed a multifunctional hydrogel (CHI/CPBA/RU), composed of chitosan (CHI) modified with 4-carboxyphenylboronic acid (CPBA) and the natural flavonoid, rutin (RU). This design endows the hydrogel with body temperature-responsive adhesion and low temperature-triggered detachment, thus enabling painless removal during dressing changes. The CHI/CPBA/RU hydrogels exhibit excellent biocompatibility, maintaining over 97 % viability of L929 cells. They also demonstrate potent intracellular free radical scavenging activity, with scavenging ratios ranging from 53 % to 70 %. Additionally, these hydrogels show anti-inflammatory effects by inhibiting pro-inflammatory cytokines (TNF-α, IL-6, and iNOS) and increasing anti-inflammatory markers (Arg1 and CD206) in RAW 264.7 macrophages. Notably, they possess robust antimicrobial properties, inhibiting over 99.9 % of the growth of Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus growth. In vivo testing on a murine full-thickness skin defect model shows that the hydrogel significantly accelerates wound healing by reducing inflammation, increasing collagen deposition, and promoting angiogenesis, achieving 98 % healing by day 10 compared to 78 % in the control group. These attributes make the polysaccharide-based hydrogel a promising material for advanced wound care.

A New Nanoplatform Under NIR Released ROS Enhanced Photodynamic Therapy and Low Temperature Photothermal Therapy for Antibacterial and Wound Repair.

Skin injury, often caused by physical or medical mishaps, presents a significant challenge as wound healing is critical to restore skin integrity and tissue function. However, external factors such as infection and inflammation can hinder wound healing, highlighting the importance of developing biomaterials with antibiotic and wound healing properties to treat infections and inflammation. In this study, a novel photothermal nanomaterial (MMPI) was synthesized for infected wound healing by loading indocyanine green (ICG) on magnesium-incorporated mesoporous bioactive glass (Mg-MBG) and coating its surface with polydopamine (PDA). In this study, Mg-MBG and MMPI was synthesized via the sol-gel method and characterized it using various techniques such as scanning electron microscopy (SEM), the energy dispersive X-ray spectrometry (EDS) system and X-ray diffraction (XRD). The cytocompatibility of MMPI was evaluated by confocal laser scanning microscopy (CLSM), CCK8 assay, live/dead staining and F-actin staining of the cytoskeleton. The antibacterial efficiency was assessed using bacterial dead-acting staining, spread plate method (SPM) and TEM. The impact of MMPI on macrophage polarization was initially evaluated through flow cytometry, qPCR and ELISA. Additionally, an in vivo experiment was performed on a mouse model with skin excision infected. Histological analysis and RNA-seq analysis were utilized to analyze the in vivo wound healing and immunomodulation effect. Collectively, the new photothermal and photodynamic nanomaterial (MMPI) can achieve low-temperature antibacterial activity while accelerating wound healing, holds broad application prospects.

Development of a Synergistic Nanomaterial Scaffold Combining Silver Nanoparticles, Collagen, and Doxycycline for Enhanced Scar-Free Skin Regeneration.

Introduction The efficacy of wound-healing treatments can be significantly enhanced through innovative combination therapies. This research investigates the wound-healing properties of a combination therapy involving silver nanoparticles (AgNPs) synthesized using Delphinium denudatum (Dd), bovine tendon collagen (BTC), and the antibiotic doxycycline (DOX) in Wistar albino rats. Each component has known therapeutic benefits: AgNPs possess antimicrobial properties, BTC aids in tissue regeneration, and DOX is an effective antibiotic. The synergy between these components is hypothesized to enhance wound closure, reduce inflammation, and promote scar-free healing. Methods The synthesis of DdAgNPs was carried out using Dd. The presence of AgNPs was confirmed by ultraviolet-visible (UV-Vis) spectroscopy and high-resolution transmission electron microscopy (HRTEM). The study was conducted on Wistar albino rats following ethical guidelines for animal research. The rats were divided into different groups to receive various treatments: DdAgNPs alone, BTC alone, DOX alone, combinations of two components, and the triple combination of DdAgNPs: BTC: DOX. Wound closure rates, epithelialization, and collagen deposition were monitored and recorded over time. Tissue samples from the wound sites were collected for histological analysis. Hematoxylin and eosin (H&E) staining was used to evaluate epithelialization and overall tissue architecture. Collagen deposition was assessed using Masson's trichrome staining. Additionally, the expression of cyclooxygenase-2 (COX-2) was measured as an indicator of inflammation. Results UV-Vis spectroscopy provided the characteristic surface plasmon resonance peak indicative of AgNPs, while HRTEM revealed the morphology and size of the nanoparticles, showing spherical particles with an average size of 35±10.42 nm. The combination therapy of DdAgNPs: BTC: DOX significantly enhanced wound closure compared to individual and dual-component treatments. This was evidenced by faster epithelialization and increased collagen deposition. The histological analysis showed that the triple combination treatment resulted in more organized tissue architecture and denser collagen fibers. Furthermore, the treatment led to a marked decrease in COX-2 expression, indicating reduced inflammation and potential for lower scar formation. Conclusion The synergistic application of DdAgNPs, BTC, and DOX presents a promising strategy for advanced wound healing and tissue regeneration. The combination therapy not only accelerates wound closure but also enhances the quality of healing by promoting epithelialization and collagen deposition while reducing inflammation. These findings offer a potential pathway for developing effective, scar-free healing solutions, highlighting the benefits of integrating multiple therapeutic agents in wound care.

Wound healing effect of polydeoxyribonucleotide derived from Hibiscus sabdariffa callus via Nrf2 signaling in human keratinocytes

There has been a growing interest in skin recovery in both the medical and cosmetics fields, leading to an increasing number of studies reporting diverse materials being utilized for this purpose. Among them, polydeoxyribonucleotide (PDRN) is known for its efficacy in skin repair processes, while Hibiscus sabdariffa (HS) is recognized for its antioxidant, hypolipidemic, and wound healing properties, including its positive impact on mammalian skin and cells. We hypothesized that these characteristics may have a germane relationship during the healing process. Consequently, we induced calli from HS and then extracted PDRN for use in treating human keratinocytes. PDRN (5 μg/mL) had considerable wound healing effects and wrinkle improvement effects. To confirm its function at the molecular level, we performed real-time polymerase chain reaction, western blotting, and immunocytochemistry. Furthermore, genes related to wound healing (MMP9, Nrf2, KGF, VEGF, SOD2, and AQP3) were significantly upregulated. Additionally, the protein expression of MMP9, AQP3, and CAT, which are closely related to wound healing and antioxidant cascades, was considerably enhanced. Based on cellular morphology and molecular-level evidence, we propose that PDRN from calli of HS can improve wound healing in human keratinocytes. Furthermore, its potential to serve as a novel material in cosmetic products is demonstrated.

Isolation and Characterization of Different Compounds Present in Pineapple -Ananas Comosus

It has a juicy taste and flavour and has been used from the historical ages and used for different purposes like cardiovascular effects, antianginal, wound healing, anticancer, osteoarthritis, antidiarrheal, and anti-aging properties. There are different important phytoconstituents available in different parts of pineapple like root, peel, core, and leaves like flavonoids, terpenoids, glycosides, carbohydrates, amino acids, and many more. Most important is Bromelain which is a very good enzyme having proteolytic activity, burn wound healing activity, and good digestive properties. Due to enormous uses in versatile pharmacological profiles, there are numerous formulations that have also been discovered. These formulations are overcoming the side effects of synthetic drugs and playing a crucial role in herbal drug technology. Bromelain is a complex enzyme and different amino acids are present in it. Many researchers are trying to isolate and are trying to establish unknown pharmacological activity. The pineapple is a magic fruit and many more are yet to be discovered.

Polydopamine-armored zeolitic imidazolate framework-8-incorporated zwitterionic hydrogel with multifunctional properties for infected wound healing

Diabetic skin wound healing is compromised by bacterial infections, oxidative stress, and vascular disruption, leading to delayed recovery and potential complications. This study developed an antibacterial, antioxidant, and adhesive hydrogel dressing that promotes rapid bacterial-infected diabetic wound healing using the biological macromolecule of polydopamine (PDA). This hydrogel comprised PDA-armored zeolitic imidazolate framework-8 nanoparticles (PDA@ZIF-8 NPs) combined with a second armor of zwitterionic polymer network (poly(acrylamide-co-sulfobetaine methacrylate); PAS), realizing low concentration Zn2+ release, good adhesion (14.7 kPa for porcine skin), and improved tensile strength (83.2 kPa). The hydrogel exhibited good antibacterial efficacy against both Staphylococcus aureus (S. aureus, 92.8 %), Escherichia coli (E. coli, 99.6 %) and methicillin-resistant S. aureus (MRSA, 99.2 %), which was attributed to the properties of the incorporated PDA@ZIF-8 NPs. Notably, in vitro, the PDA@ZIF-8 PAS hydrogel not only promoted fibroblast proliferation and migration but also facilitated endothelial cell angiogenesis. In vivo, the PDA@ZIF-8 PAS hydrogel retained its Zn2+-releasing function and effectively suppressed bacterial growth in infected wounds, thereby accelerating the regeneration of both normal and diabetic wounds. This multiarmored hydrogel is a promising sustained-release carrier for functional metal ions and drugs, making it applicable for not only skin healing, but potentially the regeneration of other complex tissues.

Engineering Titanium-Hydroxyapatite Nanocomposite Hydrogels for Enhanced Antibacterial and Wound Healing Efficacy.

External factors often lead to predictable damage, such as chemical injuries, burns, incisions, and wounds. Bacterial resistance to antibiotics at wound sites underscores the importance of developing hydrogel composite systems with inorganic nanoparticles possessing antibacterial properties to treat infected wounds and expedite the skin regeneration process. In this study, a promising TiO2-HAp@PF-127@CBM inorganic and organic integrated hydrogel system was designed to address challenges associated with bacterial resistance and wound healing. The synthesized TiO2-hydroxyapatite (HAp) nanocomposites were coated with an FDA-approved PluronicF-127 polymer and combined with a carbomer hydrogel (CBM) to accomplish the final product. The synthesized nanoparticles exhibit enhanced biocompatibility against L929 and HUVECs and cell proliferation effects. To mitigate oxidative stress caused by TiO2-induced reactive oxygen species in dark environments for effective antibacterial effects, HAp promotes cell proliferation, expediting wound skin layer formation. CBM binds to inorganic nanoparticles, facilitating their gradual release and promoting wound healing. The reduced inflammation and enhanced tissue regeneration observed in the TiO2-HAp@PF-127@CBM group suggest a favorable environment for wound repair. These results align with prior findings highlighting the biocompatibility and wound-healing properties of titanium-HAp-based materials. The ability of the TiO2-HAp@PF-127@CBM hydrogel dressing to promote granulation tissue formation and facilitate epidermal regeneration underscores its potential for promoting antibacterial effects and wound healing applications.

Anti-inflammatory, Antioxidant, and Wound-healing Properties of the Methanolic Extracts from Hedera helix Fruits and Leaves

Common Ivy (Hedera helix L.) is an ornamental plant that is known for its aesthetic qualities and ability to provide natural greenery. It is also associated with potential health benefits. When evaluating the phytochemical constituents of H. helix methanolic extracts, the fruit extract had the highest levels of total phenolic compounds (TPC) at 100 GAE mg/g extract, compared to 89.47 GAE mg/g in the leaves extract. In contrast, total flavonoid compounds and total tannin were higher in leaves extracts, 37.14 TE mg/g extract and 24.79 GAE mg/g extract respectively. fruit extracts showed the greatest level of antioxidant properties in the FRAP test 75.5 and 62.35 AscE mg/g extract also in DPPH tests, the IC50 were 3.49 and 8.79 mg/ml for fruit and leaves respectively, demonstrating their potent capacity to neutralize free radicals and high reducing power. However, when evaluated by the ABTS method, the leaves extracts indicated the strongest antioxidant activity, suggesting their potent capacity to neutralize free radicals, the IC50 were 4.54 and 8.69 mg/ml for fruit and leaves respectively, the extracts' inhibitory effects on albumin denaturation were also assessed. The findings demonstrated the potential of the extracts as anti-inflammatory, with the leaf extract having the lowest IC50 values in these tests 75.26± 3.87 µg/ml and 115.62± 56.47 µg/ml, for ripe fruit extract. Furthermore, the lowest contraction value was 81.12% for pure ointment alone, followed by drug ointment and fruit ointment at 86.43 %and 90.21%, respectively, and the extract had the highest contraction rate at 95.82%, which demonstrated the strongest wound healing activity.

Pharmacological Evaluation of Ayurvedic Nicotiana tobacum Formulation for Accelerated Wound-healing: An Experimental Study

Background: The need for pharmacognostic-based compounds with various medicinal actions is increasing to address issues such as drug resistance, new diseases, and the toxicity of already utilized chemical compounds. The plant kingdom provides numerous bioactive molecules that have great promise to help solve these challenges, resulting in the rise of innovative solutions in the pharmaceutical sector. Ayurvedic preparations, including tobacco (Nicotiana tabacum) are considered a viable method due to their long history in herbal therapy and several medical advantages. Methods: The study aims to investigate the wound-healing properties of the tobacco plant. Tobacco leaves were utilized in the production of a traditional ayurveda formulation, which included mashi formulations such as anterdhum padhati mashi, bahirdhum padhati mashi, and muffle furnace mashi, in accordance with traditional ayurvedic methods. Aqueous and alcoholic extracts of tobacco leaves were also produced. The effectiveness of these formulations in wound-healing was evaluated using an Excision model in mice. Results: The bahirdhum padhati mashi with methanolic discharge showed significantly greater wound-healing capacity compared to a standard medication formulation, as indicated by comparative study. Conclusion: Ultimately, the bahirdhum padhati mashi made from tobacco shows potential for healing wounds and could be a useful agent for wound-healing. The results of this experimental study suggest that ayurvedic formulations could be effective in treating wound-healing difficulties. Additional research and clinical studies are needed to confirm the safety, effectiveness, and possible use in wound treatment.

CeO2-loaded PVA/GelMA core-shell nanofiber membrane to promote wound healing

Chronic wounds are a significant health issue due to various pathogenic abnormalities that hinder the healing process. Nanofiber membranes, with their favorable conductivity for cell growth, proliferation, and adhesion, offer immense potential for effective chronic wound healing. In this study, we utilized the coaxial electrospinning method to synthesize core-shell structured nanofiber membranes for chronic wound healing. To obtain superior nanofiber membranes, we modified the shell part of the electrospun nanofiber by combining gelatin methacrylate (GelMA) with cerium oxide (CeO2). The results show that these nanofibers exhibit promising wound healing properties, including ideal hydrophilicity, swelling ratio, porosity, mechanical properties, antibacterial activity, and fibroblast cell viability. With the addition of 3 % w/v CeO2 nanoparticles (PGCe-3 CL sample), the nanofiber membrane had a 53.7° water contact angle, 384.47 % swelling ratio, 83.91 % porosity, and 8.49 MPa tensile strength, which are optimal for the wound healing process. The PGCe-3 CL sample also demonstrated increased antibacterial activity, reducing bacterial colonies by up to 88.46 % for S. aureus and 93.14 % for E. coli compared to PG-CL and PGCe-1.5 CL. Furthermore, the cell viability test showed that the nanofiber membranes are non-toxic to human dermal fibroblast cells and significantly enhance cell proliferation compared to other samples. Accordingly, the synthesized nanofiber membranes loaded with nano CeO2 in this study show promising performance as suitable dressings for chronic wounds. Moreover, the findings of this study provide valuable insights for developing more effective and innovative materials for chronic wound healing.

Hyaluron-Based Bionanocomposites of Silver Nanoparticles with Graphene Oxide as Effective Growth Inhibitors of Wound-Derived Bacteria.

Keeping wounds clean in small animals is a big challenge, which is why they often become infected, creating a risk of transmission to animal owners. Therefore, it is crucial to search for new biocompatible materials that have the potential to be used in smart wound dressings with both wound healing and bacteriostatic properties to prevent infection. In our previous work, we obtained innovative hyaluronate matrix-based bionanocomposites containing nanosilver and nanosilver/graphene oxide (Hyal/Ag and Hyal/Ag/GO). This study aimed to thoroughly examine the bacteriostatic properties of foils containing the previously developed bionanocomposites. The bacteriostatic activity was assessed in vitro on 88 Gram-positive (n = 51) and Gram-negative (n = 37) bacteria isolated from wounds of small animals and whose antimicrobial resistance patterns and resistance mechanisms were examined in an earlier study. Here, 69.32% of bacterial growth was inhibited by Hyal/Ag and 81.82% by Hyal/Ag/GO. The bionanocomposites appeared more effective against Gram-negative bacteria (growth inhibition of 75.68% and 89.19% by Hyal/Ag and Hyal/Ag/Go, respectively). The effectiveness of Hyal/Ag/GO against Gram-positive bacteria was also high (inhibition of 80.39% of strains), while Hyal/Ag inhibited the growth of 64.71% of Gram-positive bacteria. The effectiveness of Hyal/Ag and Hyal/Ag/Go varied depending on bacterial genus and species. Proteus (Gram-negative) and Enterococcus (Gram-positive) appeared to be the least susceptible to the bionanocomposites. Hyal/Ag most effectively inhibited the growth of non-pathogenic Gram-positive Sporosarcina luteola and Gram-negative Acinetobacter. Hyal/Ag/GO was most effective against Gram-positive Streptococcus and Gram-negative Moraxella osloensis. The Hyal/Ag/GO bionanocomposites proved to be very promising new antibacterial, biocompatible materials that could be used in the production of bioactive wound dressings.
