Induction of unique mRNAs by human interferons.

Treatment of human fibroblast cells with human interferon (INF-alpha, IFN-beta, or IFN-gamma) resulted in the accumulation of at least four newly synthesized mRNAs. The mRNAs code for proteins having molecular weights of 56,000, 57,000, 62,000, and 68,000 when characterized in a wheat germ cell-free translation system. A direct relationship was observed between the amount of IFN used and the degree of both the accumulation of the induced mRNAs and the development of an antiviral state. In the case of IFN-alpha or IFN-beta, time course studies indicated that the induced mRNAs appeared as early as 40 min, accumulated for 2 h, then remained ribosome bound for up to 16 h. The ability of fibroblast cells to develop an antiviral state always coincided directly with both the appearance and the level of accumulation of the induced mRNAs. Further mRNA synthesis beyond 2 h had a minimal effect on the development of an antiviral state. Human IFN-gamma also induced the synthesis of the same four mRNAs but required higher interferon titers and a longer incubation time. In addition, IFN-gamma induced a disproportionate amount of the mRNA coding for the 68,000 molecular weight protein and three new mRNAs not detected in cells treated with IFN-alpha or IFN-beta. Mouse interferon induces the original four mRNAs in human cells but to a far lesser extent. This correlated with the inability of these cells to develop much resistance to viral infection.

Translational properties of a non-polyadenylated oligo(U)-containing RNA fraction from wheat embryos

A non-polyadenylated oligo(U)-containing RNA (poly(A) −·oligo(U) + RNA) fraction was isolated from wheat embryo cytoplasm and its properties were compared with those of polyadenylated RNA (poly(A) + RNA) from the same source. Both RNA preparations were highly heterogeneous and effectively stimulated [ 14C]leucine incorporation in a wheat germ cell-free translation system. Electrophoretic patterns of the translation products appearing in the non-polyadenylated RNA- and polyadenylated RNA-supplemented translation assays, respectively, differed from each other. The non-polyadenylated RNA-specific translation products included, in particular, a series of high molecular weight polypeptides. It is concluded that a specific class of non-polyadenylated oligo(U)-containing mRNA species (other than histone mRNAs) occurs in the wheat embryo cells.

Wheat Storage Proteins

A total RNA extract was prepared from developing wheat seeds using guanidine-HCl to eliminate endogenous RNase activity. The RNA preparation, substantially free of protein, carbohydrate and DNA, was chromatographed on either a poly uridylic acid-agarose or poly guanylic acid-agarose column to yield a gliadin-enriched mRNA fraction. Only slight differences were observed for the products synthesized in a wheat germ cell-free translation system when either poly adenylic acid-enriched or cytosine-rich RNA was used as a template. These results are consistent with the high proline content of the gliadins and indicate that a large proportion of the mRNA activity in these RNA preparations is directed toward gliadin synthesis. After a second affinity chromatography step, the gliadin-enriched mRNA fraction was fractionated by two cycles on sucrose-density gradient centrifugation under denaturing conditions. The RNA sedimented as a broad band with a peak at 14S and a shoulder at the 11S region of the sucrose gradient. RNA from the peak 14S fraction translated predominantly the two major gliadin polypeptides which had molecular weights of 34,000 and 36,000. Analysis of the 14S RNA by methylmercury hydroxide-agarose gel electrophoresis revealed the presence of a predominant RNA species with a molecular size of 415,000 (1,200 nucleotides).

The importance of tRNA for the in vitro cell-free translation of messenger RNA isolated from the malaria parasite Plasmodium lophurae

Preliminary studies had indicated the inadequacy of the wheat germ and rabbit reticulocyte cell-free translation systems for the in vitro translation of mRNA isolated from Plasmodium lophurae. To identify the factors which are important for the efficient translation of parasite proteins, an homologous system was established using polysomes, the pH 5 fraction, and tRNA prepared from P. lophurae. For comparison, the same components were isolated from the host duck reticulocytes and tested. The effect of each of these factors was evaluated by analysis of the translation products and by comparison with products synthesized in vivo. The results indicated that P. lophurae tRNA had a marked stimulatory effect on the synthesis of parasite proteins while it inhibited the synthesis of host proteins. Duck reticulocyte tRNA could not be used as a substitute for the parasite tRNA. Based on these findings, a commercially available rabbit reticulocyte system was supplemented with P. lophurae tRNA, which markedly increased the efficiency of translation of P. lophurae proteins by this system.

In Vitro Synthesis and Processing of Wheat α-Amylase

Wheat (Triticum aestivum) RNA was used to program synthesis of the alpha-amylase protein by Xenopus laevis oocytes. A 41,500-dalton protein was made which was identified as alpha-amylase by immunoprecipitation with rabbit anti-alpha-amylase antiserum raised against the purified wheat protein and by its co-migration with authentic alpha-amylase on sodium dodecyl sulfate polyacrylamide gels. Synthesis of alpha-amylase was dependent upon injection of RNA extracted from gibberellic acid-induced aleurone layers from wheat. The amount of alpha-amylase produced was proportional to the amount of RNA injected and reached a plateau within 4 hours after injection. When the same RNA was translated in a wheat germ cell-free translation system, a 43,000-dalton protein was produced. Addition of dog pancreas microsomal membranes to the wheat germ translation system resulted in processing of the alpha-amylase protein to a form which co-migrated with authentic alpha-amylase purified from malted wheat and with the protein synthesized in oocytes.

Lactate dehydrogenase-C mRNA: Its isolation and [formula omitted] translation

Lactate dehydrogenase-C (LDH-C) mRNA was purified from DBA 2 mouse testes and translated in vitro . First, the LDH-C synthesizing polysomes were isolated by double immunoprecipitation using specific anti-LDH-C and anti-horse immunoglobulin antibodies. Extraction of mRNA was made from the isolated polysomes using hot sodium dodecyl sulfate-phenol method at alkaline pH. In a wheat germ cell-free translation system, the mRNA coded for a polypeptide chain that could be immunoprecipitated with specific anti LDH-C antibody and comigrated with authentic LDH-C in sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

Light-stimulated increase of translatable mRNA for phosphoenolpyruvate carboxylase in leaves of maize.

Polyadenylated RNA has been isolated from maize leaves at various times during greening of etiolated seedlings and used to prime the wheat germ cell-free translation system. Levels of translatable messenger RNA for phosphoenolpyruvate carboxylase increase with the length of the illumination period. The pattern of the increase in translatable mRNA for the enzyme is similar to that of the increase in phosphoenolpyruvate carboxylase protein previously observed in intact tissues. Phosphoenolpyruvate carboxylase synthesized in vivo migrates as a doublet band on gradient sodium dodecyl sulfate-polyacrylamide gels. A similar doublet has been seen occasionally with the products of cell-free translation.

A positive regulatory gene is required for accumulation of the functional messenger RNA for the glucose-repressible alcohol dehydrogenase from Saccharomyces cerevisiae

The amount of glucose-repressible alcohol dehydrogenase is regulated by the amount of its functional messenger RNA. ADHII † † Abbreviations used: ADHII, alcohol dehydrogenase, isoenzyme II: ADHI, alcohol dehydrogenase, isoerizyme 1: SDS, sodium dodecyl sulfate. protein was detected by a radioimmune assay and differentiated from ADHI, the classical ADH isozyme, by limited proteolysis with Staphylococcus aureus protease. When yeast containing the wild-type alleles for ADR2 (the ADH II structural locus) and for ADR1 (its positive regulatory gene) were pulse-labeled with [ 35S]methionine during derepression, radioactive label accumulated in the antibody-precipitated ADHII coterminously with the appearance of ADHII activity. The kinetics of functional ADHII mRNA appearance during derepression in this strain were shown to be the same as those for ADHII protein synthesis in vivo when RNA, extracted from derepressed cells, was translated in a wheat germ cell-free translation system. The role of the positive regulatory gene, ADR1, in ADHII expression was analyzed using two strains mutated at that locus. Yeast containing the adr1-1 allele are incapable of derepressing ADHII activity. When this strain was pulselabeled with [ 35S]methionine during derepression, approximately one-tenth to one-twentieth the level of ADHII protein synthesis was detected as in the wild-type strain. When RNA was extracted during derepression from cells containing the udr1-1 allele and translated in a wheat germ cell-free system, little functional ADHII mRNA was found to be present. The role of the ADR1 gene was further analyzed using a strain containing the ADR1-5 c allele, which allows constitutive synthesis of ADHII activity. In this strain during glucose repression. ADHII protein synthesis and amount of functional mRNA were at levels comparable to those found for the wild-type strain after complete derepression. Similar kinetics of ADHII protein synthesis and of mRNA accumulation during derepression were observed in the strain carrying the ADR1-5 c allele when compared to that carrying the ADR1 allele, but the absolute amounts were greater by three- to fourfold in cells containing the ADR1-5 c allele. These results indicate that the ADR1 gene acts to increase the level of functional ADHII mRNA during derepression.

Induction of rat liver metallothionein mRNA and its distribution between free and membrane-bound polyribosomes.

Total, membrane-bound and free polyribosomes were purified from livers of Zn2+-treated and control rats. Polyadenylated RNA was separated from the polyribosomal RNA extracts by oligo(dT)--cellulose chromatography and translated in a wheat-germ cell-free translation system. Newly synthesized 35S-labelled metallothionein was isolated from the other [35S]methionine-labelled translation products by activated-thiol--Sepharose 4B chromatography. The purity of the 35S-labelled metallothionein product was substantiated by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis. Zinc administration resulted in an elevation of metallothionein mRNA activity to 11% of the total polyribosomal mRNA activity. The vast majority of biologically active metallothionein mRNA was localized in the free polyribosomal pool, at least 94% and 97% in control and zinc-treated rats respectively. The increase in the percentage of polyribosomal mRNA coding for metallothionein after zinc administration was 3-fold, whether measured directly in total polyribosomal mRNA or as a combination derived from membrane-bound and free polyribosomal mRNA. These data indicate that the induction of metallothionein mRNA by zinc involves only free polyribosomes and suggest that the function of metallothionein is limited to intracellular processes.

Antheridiol-induced differentiation of Achlya in the absence of detectable synthesis of new proteins

Vegetatively growing mycelia of the oomycete Achlya ambisexualis (♂ strain E87) can be induced to form male sexual structures upon treatment with the steroid hormone, antheridiol. Proteins labeled with a 20-minute pulse of [ 35S]methionine at different developmental stages were examined by both one-dimensional and two-dimensional polyacrylamide gel electrophoresis. All of the proteins detectable during vegetative growth continue to be synthesized during differentiation. Increased synthesis of a 60,000-dalton protein is detectable between 30 and 90 minutes following addition of hormone. This protein has not been detected on two-dimensional gels and no other developmental changes were observed during induced differentiation. Poly(A) + mRNA isolated at specific times during differentiation was translated in a wheat germ cell-free translation system. Analysis of the products on polyacrylamide gels showed no detectable qualitative differences in the products of translation during differentiation. We conclude that antheridiol-induced differentiation is accompanied by minor transcriptional and translational changes (if any) and that post-translational regulatory events may be required for differentiation.

Partial amino acid sequence of the precursors of blocked heavy and light chains from C 3 H mouse myeloma 5563.

A messenger RNA fraction from the C 3 H mouse myeloma 5563 was used to direct the synthesis of heavy (gamma 2a) and light (chi) chain precursors. The synthetic products were radiolabeled by inclusion of [3H] or [35S] amino acids in wheat-germ cell-free translation system. Precursor peptides for both H and L chains were indicated by comparison by polyacrylamide gel analysis of apparent molecular weights of chains synthesized in vitro vs. in vivo. The nonglycosylated H chain synthesized in vivo in the presence of tunicamycin was used for comparison. This approach should be generally applicable for the demonstration of precursor forms of N-glycosylated polypeptides. Following immunoprecipitation and preparative polyacrylamide gel electrophoresis, the isolated chains were subjected to automated Edman degradation. The amino acid sequence data obtained for these precursors were significantly different from those reported for BALB/c immunoglobulins. The 5563 H chain precursor bears little homology to the previously reported H chain precursor from MOPC 315 (IgA) (Jilka, J. A. and Pestka, S., Proc. Nat. Acad. Sci. USA 1977. 74:5692). The 5563 H chain can be assigned to a heretofore undescribed H chain variable region subgroup on the basis of partial sequence data. The amino terminal sequence of the 5563 L chain precursor has maximum homology (25-50%) to the ssequence o the precursor peptide of MOPC 41 chi chain.

Isolation and characterization of calmodulin from spinach leaves and in vitro translation mixtures

Calmodulin, a multifunctional calcium-modulated protein, has been isolated from spinach leaf tissue and from spinach leaf messenger RNA translation products. The translation protein and the spinach leaf protein have been partially characterized and compared to vertebrate calmodulins. Spinach leaf calmodulin will quantitatively activate bovine brain phosphodiesterase and will undergo a calcium-dependent shift in electrophoretic mobility similar to that of bovine brain calmodulin. In the presence of Ca(2+) the spinach and brain proteins comigrate, but in the presence of chelators they do not. A polyadenylylated RNA fraction has been isolated from spinach leaf tissue and translated in a wheat germ cell-free translation system. The calmodulin synthesized in vitro has been isolated by using calcium-dependent affinity chromatography on phenothiazine-Sepharose conjugates. The translation protein comigrates with spinach calmodulin during polyacrylamide gel electrophoresis whether in the presence or the absence of Ca(2+). The translation protein also undergoes a calcium-dependent mobility shift identical to that of spinach calmodulin. Amino acid analysis of the translation calmodulin indicates that it does not contain N(epsilon)-trimethyllysine, an amino acid residue that is characteristic of all calmodulins previously examined. These studies suggest that N(epsilon)-trimethyllysine is not required for the calcium-dependent interaction of calmodulin with phenothiazines and indicate the potential utility of phenothiazine-Sepharose conjugates as affinity-based adsorbents in biological and biochemical investigations.

Synthesis of rat liver microsomal cytochrome b5 by free ribosomes.

Free and membrane-bound polyribosomes were separated from liver homogenates and characterized by electron microscopy. Using the wheat germ cell-free translation system, total translation products of poly A+RNA extracted from free polyribosomes (poly A+RNAf) showed some correlation to total liver cytosol proteins. In contrast, translation products of poly A+RNA from membrane-bound polyribosomes (poly A+RNAmb) showed some similarity to rat serum. Antibody to purified rat serum albumin immunoprecipitated from only the translation products of poly A+RNAmb a single polypeptide of mol wt 68,000. i.e., 3,000 greater than secreted serum albumin. In contrast, antibody to detergent-extracted cytochrome b5 immunoprecipitated from only the translation products of poly A+RNAf a single polypeptide of mol wt 17,500, identical to that of microsomal cytochrome b5. A consideration of the known properties of cytochrome b5 is consistent with an exclusive site of synthesis on free ribosomes.

Ribosome binding site analysis of ovalbumin messenger ribonucleic acid.

The region of the ovalbumin messenger ribonucleic acid (mRNAov) molecule bound to the 40S ribosomal subunit and its associated initiation factors in the wheat germ cell-free translation system were isolated and characterized. Two mRNAov fragments, 87 and 92 nucleotides in length, were protected from T1 ribonuclease digestion by binding of guanosine 5',beta,gamma-methylenetriphosphate and were shown by hybridization and fingerprint mapping to be derived from the 5' end of mRNAov. Both these mRNAov fragments were of sufficient length to contain both the cap structure and the AUG initiation codon. Four T1-resistant oligonucleotides, prepared by direct digestion of mRNAov with T1 ribonuclease were also found to bind to the wheat germ 40S ribosomal subunit. Nucleotide sequence analysis of these oligonucleotides revealed (1) that they were not a subset of the ribosome binding fragments described above, (2) that they were derived from within the mRNAov molecule (one from within the coding region and three from the noncoding region at the 3' end of the mRNAov molecule), and (3) that three of the four mRNAov nucleotides contained 3'-terminal AUG trinucleotides. These data suggested that features of the mRNAov molecule in addition to the nucleotide sequence might be important in specifying the correct ribosome binding site for the initiation of protein synthesis. The amount of mRNAov bound to the wheat germ 40S ribosomal subunit in a preinitiation complex was found to vary inversely with the potassium ion concentration. Lowering the potassium concentration to levels suboptimal for translation also resulted in the protection of larger fragments of the mRNAov molecule derived from the same 5'-end region as the ribosome binding fragments described above. The ability of the cap analogue 7-methylguanosine 5'-phosphate (m7G5'p) to reduce the amount of mRNAov bound to the wheat germ 40S ribosomal subunit was found to depend directly on thepotassium concentration. Interestingly, the effects of potassium on the amount of mRNAov bound in a preinitiation complex and the inhibition of this binding by m7G5'p could be observed by changing the potassium concentration after binding had occurred. These data suggested that the interaction between the wheat germ 40S ribosomal subunit and mRNAov was very sensitive to the ionic environment.

Developmental Changes in the Activity of Messenger RNA Isolated from Germinating Castor Bean Endosperm

The capacity of polyadenylated RNA from developing castor bean endosperm to program protein synthesis in a wheat germ cell-free translational system has been examined. Although the use of micrococcal nuclease-treated wheat germ extracts demonstrated a low but significant content of translatable mRNA in dry seeds, a large scale increase in total translational capacity was observed during germination. The cellular content of translatable mRNA peaked on the 4th day of germination and subsequently declined. It is concluded that protein synthesis in castor bean endosperm cells during germination is directed by newly transcribed mRNA.

Cell-free synthesis of rat liver zinc-thioneins.

The induction of rat liver zinc-thioneins mRNA was studied in a wheat germ cell-free translation system. Liver poly A rich polysomal RNA was isolated from rats which had been injected with zinc sulfate 5 h previously. These RNA preparations stimulated the incorporation of [35S]cystine into trichloroacetic acid insoluble proteins when assayed in the cell-free synthetic system. The translation products were characterized by Sephadex G-75 chromatography in 8 M urea--50 mM beta-mercaptoethanol, by disc gel electrophoresis in 4 M area--Tris-glycine buffer (pH 9.2), and by peptide fingerprinting with pepsin. These results were identical with authentic rat liver zinc-thioneins. The zinc-thioneins mRNA activity in the control rats, however, was minimal. The stimulation in zinc-thioneins synthesis observed in the cell-free synthesis was similar to the increased synthesis of these polypeptides in vivo.

In vitro translation of measles virus-induced RNA

RNA was extracted from Vero cells infected with measles virus and tested for its ability to direct protein synthesis in the wheat germ cell-free translation system. When in vitro products were subjected to immunoprecipitation with immunoglobulins directed against measles virus four major bands were found to correspond to the viral proteins.

Cloning of a double-stranded cDNA that codes for a portion of chicken preproalbumin. A general method for isolating a specific DNA sequence from partially purified mRNA.

A scheme is presented for cloning a double-stranded cDNA molecule that codes for a portion of chicken preproalbumin. This method, which does not require pure mRNA or cDNA, has widespread applicability. Chicken preproalbumin was identified as a Mr = 72,000 polypeptide by immunoprecipitation of proteins synehesized in a wheat germ cell-free translation system from total, guanidine.HCl-extracted, rooster liver RNA. After removal of the bulk of the ribosomal RNA by poly(U)-Sephadex G-10 chromatography, albumin mRNA was enriched approximately 2-fold by centrifugation through low salt, isokinetic sucrose gradients, until it represented about 30% of the mRNA sequences present. Double-stranded cDNA prepared from this mRNA was then inserted into the Pst 1 site of the plasmid PBR322 by the "G-C tailing" technique and the recombinant DNA was used to transform Echerichia coli stran X1776. Transformants containing putative albumin DNA sequences were identified by colony hybridization with a cDNA probe that was highly enriched for albumin cDNA sequences. This probe was isolated by hybridizing the partially purified RNA preparation to its cDNA, under conditions of RNA excess, to a R0t value such that only the most abundant cDNA sequences had hybridized. Unhybridized, less abundant, sequences were destroyed by subsequent S1 nuclease digestion. The identity of clones that hybridized to this abundant class cDNA was established by DNA-mRNA hybrid-arrested cell-free translation. Hybridization of nick-translated, albumin-containing, plasmid DNA to total liver poly(A)+ RNA, that had been separated on methyl mercury agarose gels and transferred to diazobenzyloxymethyl paper, established that avian albumin mRNA has a molecular weight of 850,000. This molecular weight corresponds to approximately 2,600 nucleotides, or 600 nucleotides longer than the size required to code for the preproalbumin polypeptide.

RNA Hybridization to DNA Coupled with Cyanogen‐Bromide‐Activated Sephadex

A method for purifying RNA by hybridization to complementary DNA immobilized on Sephadex is described and is illustrated by the purification of polyoma mRNA. Denatured DNA can be immobilized on cyanogen‐bromide‐activated Sephadex whilst remaining freely available for hybridization. However, after activation with cyanogen bromide, followed by the coupling of DNA and deactivation with ethanolamine, Sephadex acts as an ion‐exchange resin. It is therefore necessary to elute RNA from DNA‐Sephadex by a combination of formamide buffers containing NaCl and elevated temperatures, conditions which dissociate DNA · RNA hybrids and also prevent the binding of RNA to the resin by ion exchange.Polyoma DNA, or restriction fragments thereof, were bound to Sephadex and used to purify viral mRNA. RNA purified from polyadenylated RNA isolated from the cytoplasm of mouse cells 28–30 h after infection with polyoma virus sedimented at values between 19 and 16 S in sucrose/formamide gradients and, in a wheat germ cell‐free translation system, directed the synthesis of three polypeptides; these were identified by gel mobility, immunoprecipitation and fingerprinting as the three structural proteins of polyoma virus. Viral mRNA could also be purified by hybridization directly from total cytoplasmic infected‐cell RNA without prior selection of polyadenylate‐containing RNA.The advantages and limitations of DNA attached to cyanogen‐bromide‐activated Sephadex as an affinity matrix for RNA purification are discussed.

Synthesis of preprolactin and conversion to prolactin in intact cells and a cell-free system.

Monolayer cultures of pituitary cells were pulse-labeled with [3H]leucine for several minutes and the incorporated radioactivity was analyzed by immunoprecipitation and electrophoresis on sodium dodecyl sulfate containing polyacrylamide gels. Following a 3-min labeling period, a peak of radioactivity with a mobility similar to that of preprolactin was observed, as well as radioactivity co-migrating with prolactin. Competition with unlabeled prolactin demonstrated the specificity of the immunoprecipitation reaction. After 5 min of pulse-labeling followed by 5-min chase in medium with unlabeled leucine, only a product with the mobility of prolactin remained. Addition of a membrane fraction from dog pancreas to a wheat germ cell-free translation system containing pituitary mRNA resulted in the conversion of preprolactin to prolactin. Partial sequence analysis demonstrated that the processed product contained the correct NH2 terminus of prolactin. Thus, both intact pituitary cells and a cell-free heterologous system are able to synthesize preprolactin and cleave it to prolactin offering strong evidence that preprolactin is the biosynthetic precursor to prolactin.
