In-Vivo Antidiarrhoea Evaluations of Methanol Leaf Extract and Fractions of Chrysophyllum albidum

Background and Objectives: In Nigeria, traditional use of medicinal plants, like Chrysophyllum albidum, for diarrhoea persists. This study assessed its effectiveness in treating diarrhoea in rats. Methods: Extract and Fractions from the plant were tested for antidiarrhoea effect. The process involved methanol extraction and fractionation using hexane, ethyl acetate, and butanol. Rat models were used to evaluate the effects on castor oil-induced diarrhoea, enteropooling, and gastrointestinal motility. Results: Defecation caused by castor oil-induced diarrhoea was inhibited significantly by 53.55, 78.48 and 74.69 % at 500 mg/kg of methanol extract, ethyl acetate, and n-hexane fraction respectively, and by 50.99 and 63.69 % at 250 and 500 mg/kg of butanol fraction respectively. The volume of intestinal content and weight of intestinal content were reduced by 76.74 and 80 % at 500 mg/kg of ethyl acetate fraction. Also, the volume of intestinal content and weight of intestinal content were reduced by 70.93 and 70.83 % at 500 mg/kg of n-hexane fraction. At 250 and 500 mg/kg, the ethyl acetate fraction inhibited intestinal motility by 58.65% and 76.50% respectively. The n-hexane fraction at 500 mg/kg showed a 66.35% inhibition. Conclusion: These fractions from Chrysophyllum albidum leaves exhibit potential antidiarrhoea compounds, as evidenced across all three study models. Ongoing research aims to isolate these compounds for further investigation.

Evaluation of Antidiarrheal, Antispasmodic, and Antisecretory Activities of Extract and Fractions of Castanopsis costata Leaves in Animal Models

Background: The use of traditional medicinal plants Castanopsis costata in the management of diarrhea is an ancient practice in North Sumatra, Indonesia, but its safety and efficacy have not been examined scientifically in animal models. Therefore, this study aims to determine the antidiarrheal effect of C. costata leaf extract and fractions in vivo. Methods: The antidiarrheal activity was evaluated against animal models of diarrhea induced by castor oil, charcoal meal, and entero-pooling test. The extract group received doses of 25, 50, 100, and 200 mg/kg, while the fraction groups was treated with WFCC, EAFCC, and nHFCC, in doses of 50 and 100 mg/kg, respectively. The negative control received 10 mL/kg of the dissolving vehicle, while the positive was treated with loperamide 3 mg/kg. Results: The results showed that EECC doses of 200 mg/kg, WFCC, and EAFCC at 50 and 100 mg/kg, respectively significantly delayed the onset of diarrhea, while WFCC and EAFCC at 100 mg/kg significantly reduced the amount of wet feces output and total feces output. In addition, only WFCC and EAFCC (at 50 and 100 mg/kg, respectively) significantly reduced the mean weight of wet feces and total feces. The percentage of maximum diarrhea inhibition was observed at the EAFCC dose of 100 mg/kg. Based on the charcoal meal test results, only EAFCC significantly inhibited the distance traveled by charcoal meal and reduced the peristaltic index at a dose of 100 mg/kg, while the percentage decrease in gastrointestinal motility was 46.87%. Similarly, in the entero-pooling test, WFCC and EAFCC significantly reduced the volume and weight of intestinal contents at a dose of 100 mg/kg, respectively. Conclusion: The results confirm that the extract and fractions of C. costata have antidiarrheal activity. Therefore, this study provides scientific support for the traditional use of C. costata in treating diarrhea.

Evaluation of Antidiarrheal Activity of 80% Methanol Extract and Solvent Fractions of the Leaves of Withania somnifera (L.) Dunal in Swiss Albino Mice.

Background Withaniasomnifera is an important medicinal plant for the treatment of diarrhea in Ethiopian folklore medicine. The aim of this study was to evaluate the antidiarrheal activity of Withania somnifera leaves in Swiss albino mice. Materials and Methods Hydromethanolic crude extraction and solvent fractionation were done using cold maceration technique. 80% methanol was used as a solvent in crude extraction, while distilled water, n-butanol, and chloroform were employed during fractionation. Castor oil-induced diarrhea, enteropooling, and gastrointestinal motility models were employed to evaluate antidiarrheal activity. Mice were randomly divided into five groups (six mice per group): negative control, which received 2% Tween 80 in distilled water; positive control, which received 3 mg/kg loperamide; and three test groups (III, IV, and V), which were treated with 100 mg/kg, 200 mg/kg, and 400 mg/kg of crude extract and solvent fractions, respectively. Results The crude extract, aqueous, and n-butanol fractions significantly delayed the onset of diarrhea at 200 mg/kg and 400 mg/kg dose. There was a significant reduction in the number and weight of stools at all tested doses of the crude extract and aqueous fraction, and at 200 mg/kg and 400 mg/kg of n-butanol fraction. Significant reduction in volume and weight of intestinal contents was observed at all tested doses of the crude extract, and at 200 mg/kg and 400 mg/kg of aqueous and n-butanol fractions. All tested doses of the crude extract and 200 mg/kg and 400 mg/kg of the aqueous and n-butanol fractions significantly reduced the motility of charcoal meal. Conclusion This study demonstrated that the crude extract and solvent fractions of the Withania somnifera leaves have antidiarrheal activity and supported the folklore use of the plant.

Antidiarrheal Effect of 80% Methanol Extract and Fractions of the Roasted Seed of Coffea arabica Linn (Rubiaceae) in Swiss Albino Mice

Background Globally in 2019, diarrhea was the second leading cause of mortality in children, accounting for more than half a million under-five deaths yearly. Several societies use Coffea arabica Linn for the treatment of diarrhea. However, its use is not scientifically validated. Objective The study was conducted to evaluate antidiarrheal activity of 80% methanol extract and solvent fractions of roasted seed of Coffea arabica Linn in mice. Methods Coffea arabica Linn seed was roasted, milled, extracted, and fractionated using hexane, ethyl acetate, and distilled water. Castor oil-induced diarrhea, enteropooling, and motility tests were conducted. Effects on onset, number of feces, weight of feces, fluid content, volume and weight of intestinal content, and motility were evaluated by administering 100 mg/kg, 200 mg/kg, and 400 mg/kg of each extract. Negative controls received 10 ml/kg of the vehicle, and positive controls received either loperamide (3 mg/kg) or atropine (1 mg/kg). Data were analyzed using one-way ANOVA followed by Tukey's post hoc test. Results Ethyl acetate fraction at all tested doses significantly prolonged (p < 0.05) onset of diarrhea. The number and weight of feces were also reduced significantly by crude extract and ethyl acetate fraction. Reduction in fluid content was observed at 200 mg/kg and 400 mg/kg of the crude extract (p < 0.01) and aqueous fraction (p < 0.001) as well as all tested doses of ethyl acetate fraction (p < 0.001). Similarly, the crude extract, ethyl acetate fraction, and aqueous fraction showed a significant reduction in the volume and weight of intestinal content. At 400 mg/kg, the crude extract, hexane fraction, aqueous fraction, and all doses of ethyl acetate fraction showed significant antimotility activity. Conclusion The results of this study revealed that the roasted seed of Coffea arabica Linn has antidiarrheal activity.

Limonitum Ameliorates Castor Oil-Induced Diarrhoea in Mice by Modulating Gut Microbiota

Diarrhoea is a common clinical condition; its pathogenesis is strongly associated with gut microbiota dysbiosis. Limonitum is a well-known traditional Chinese medicine that exerts appreciable benefits regarding the amelioration of diarrhoea. However, the mechanism through which Limonitum ameliorates diarrhoea remains unclear. Here, the efficacy and underlying mechanism of Limonitum decoction (LD) regarding diarrhoea were explored from the aspect of gut microbiota. Castor oil (CO) was used to induce diarrhoea in mice, which were then used to evaluate the effects of LD regarding the timing of the first defecation, diarrhoea stool rate, degree of diarrhoea, diarrhoea score, intestinal propulsive rate, and weight of intestinal contents. The concentrations of short-chain fatty acids (SCFAs), including acetic, propionic, isobutyric, butyric and valeric acids, were analysed by gas chromatography-mass spectrometry (GC-MS). The 16S rRNA high-throughput sequencing technology was applied to evaluate changes in the gut microbiota under exposure to LD. LD was found to effectively ameliorate the symptoms of diarrhoea, and the diversity and relative abundance of gut microbiota were restored to normal levels following LD treatment. Additionally, LD significantly restored the observed reductions in SCFAs. These results provide strong evidence that LD can sufficiently ameliorate diarrhoea in mice by regulating their gut microbiota. The findings presented here highlight that Limonitum may constitute a prospective remedy for diarrhoea.

Antidiarrheal Effect of 80% Methanol Extract and Fractions of Clerodendrum myricoides (Hochst.) Vatke (Lamiaceae) Leaf in Swiss Albino Mice.

Background Diarrhea is one of the tempting symptoms of diseases in the world. In Ethiopian traditional medicine practices, Clerodendrum myricoides is utilized for the treatment of diarrhea without scientific evidence. Objective This study was aimed to evaluate the antidiarrheal activity of 80% methanol extract and fractions of the leaf of Clerodendrum myricoides in mice. Methods The crude extract was prepared by maceration in 80% methanol and then fractionated using hexane, chloroform, and distilled water. Antidiarrheal activity was assessed by castor oil-induced diarrhea, enteropooling, and gastrointestinal motility models using onset of diarrhea, number and weight of feces, volume and weight of intestinal contents, and distance travelled by charcoal meal as main parameters. Negative controls received either distilled water or 2% Tween 80 (10 ml/kg), positive controls received 3 mg/kg loperamide or 1 mg/kg atropine, and the test groups received 100, 200, and 400 mg/kg doses of the extract. Results The crude extract and chloroform fraction significantly prolonged the onset of diarrhea at 200 and 400 mg/kg and decreased the number of wet, total, and weight of fresh feces at all tested doses. Hexane fraction has a significant antidiarrheal effect on the onset, number, and weight of feces at 400 mg/kg. The crude extract and chloroform fraction at all tested doses, as well as aqueous fraction at 200 mg/kg and 100 mg/kg, produced significant reduction in volume and weight of intestinal contents. Additionally, hexane fraction showed significant reduction of volume and weight of the intestinal content at 400 mg/kg. In the gastrointestinal motility test model, both chloroform fraction and crude extract at all tested doses and aqueous fraction at 200 mg/kg and 400 mg/kg showed a significant antidiarrheal effect as compared to the negative control. Conclusion The leaf of Clerodendrum myricoides showed antidiarrheal activity which supports the traditional use.

Methanolic Crude Extract of Hagenia abyssinica Possesses Significant Antidiarrheal Effect: Evidence for In Vivo Antidiarrheal Activity.

Background Hagenia abyssinica is one of the most commonly used medicinal plants for the treatment of diarrhea in Ethiopia. Therefore, this study aimed to evaluate the antidiarrheal effect of methanol crude extract of H. abyssinica leaves in mice. Methods Acute toxicity testing was conducted using Organization for Economic Cooperation and Development guidelines. The antidiarrheal activity of the crude extract of H. abyssinica was investigated using three animal models such as small intestine transit, enteropooling, and castor oil-induced diarrhea models. The extract was administered at three different doses (100, 200, and 400 mg/kg) to the test groups, while the positive control group received 3 mg/kg of loperamide and the negative control group received 10 ml/kg of vehicle (distilled water). Results The crude extract of H. abyssinica did not exhibit death at the limit dose (2 g/kg) throughout the observation period. In the castor oil-induced model, the crude extract at a dose of 200 and 400 mg/kg exhibited a significant (P < 0.05) antimotility effect as compared to the negative control. The crude extract revealed a significant reduction in the volume and weight of intestinal contents at 200 and 400 mg/kg doses of the extract. Moreover, the highest antidiarrheal index (ADI) was obtained with the dose of 400 mg/kg of crude extract, which was comparable to the standard drug. Conclusion The crude extract of Hagenia abyssinica possesses antidiarrheal activity and supports the traditional use of H. abyssinica for the management of diarrhea.

Comparative performance of extreme learning machine and Hammerstein-Weiner models for modelling the intestinal hyper-motility and secretory inhibitory effects of methanolic leaf extract of Combretumhypopilinum Diels (Combretaceae).

In this article, three data-driven approaches were explored, including two artificial intelligence (AI) based models namely; Extreme Learning Machine (ELM) and Hammerstein-Weiner (HW) models and a trivial linear model namely; multilinear regression (MLR). In this context, the models were developed using the onset of diarrhoea, the total number of wet faeces, total number of faeces, weight of intestinal content (g) and length of the small intestine (cm) as the independent variables. In contrast, distance travelled by charcoal meal (C) and volume of intestinal content (I) were considered as the dependent variables for the prediction of the intestinal hypermotility and secretory inhibitory effects of the methanol leaf extract of Combretumhypopilinum (MECH). Three different performance indicators including; mean absolute percentage error (MAPE), Nash-Sutcliffe efficiency (NSE) and Root mean square error (RMSE) were employed in this research to calculate and determine the performance skills of the models. The obtained results indicated the reliable capability of ELM and HW over MLR model having NSE-values higher than 0.90 in both the calibration and verification stages. The results further demonstrated that, in terms of MAPE and RMSE, ELM and HW models showed higher performance efficiency than the MLR model. Even though HW outperformed the ELM and MLR models in the prediction of I. Whereas, ELM outperformed HW and MLR models in the prediction of C. Overall; the results proved the satisfactory ability of the AI-based models (HW and ELM) for modelling the Intestinal hypermotility and secretory inhibitory effects of MECH.

Evaluation of Anti-Diarrheal Activity of 80% Methanol Extracts of Vernonia amygdalina Delile (Asteraceae) Leaves in Mice.

IntroductionDiarrhea is a cause of morbidity and mortality, especially in children. Patients with diarrhea have been suffering from limited treatment options due to poor drug tolerance, side effect, and multi-drug resistance to almost all the current drug treatments. Therefore, it is important to search for a new therapeutic medicine that can effectively prevent the disease and safe. Vernonia amygdalina is an indigenous medicinal plant used traditionally for the treatment of diarrhea in Ethiopia. The aim of this study was therefore to confirm the antidiarrheal activities of 80% methanol leaves extract using mice models.Materials and MethodsThe antidiarrheal activities of the 80% methanol leaves extract were investigated using castor oil-induced diarrhea, intestinal motility, and enteropooling models in mice. The negative control received distilled water orally, the test groups received three dose levels (100, 200, and 400 mg/kg) of the plant materials, and the positive control is given 3 mg/kg Loperamide orally.ResultsIn the castor oil-induced diarrheal model, the extract delayed onset of diarrhea and reduced fecal parameters at all tested doses significantly as compared with the negative control. Results from the charcoal meal test revealed that the extract produced a significant anti-motility effect at all tested doses as compared with the negative control. In the enteropooling test, the extract produced a significant decline in the volume and weight of intestinal contents. The observed antidiarrheal activity could be associated with the phytochemicals present in this plant extract. It was also observed that the extracts have shown no acute toxicity at a dose of 2 g/kg.ConclusionThis study provides the antidiarrheal activity of the crude extract in all three models. Hence, the findings provide scientific support for the traditional use of Vernonia amygdalina leaves as treatment of diarrhea.

Evaluation of Various Solvent Extracts of Tetrastigma leucostaphylum (Dennst.) Alston Leaves, a Bangladeshi Traditional Medicine Used for the Treatment of Diarrhea.

Tetrastigma leucostaphylum (TL) is an important ethnic medicine of Bangladesh used to treat diarrhea and dysentery. Hence, current study has been designed to characterize the antidiarrheal (in vivo) and cytotoxic (in vitro) effects of T. leucostaphylum. A crude extract was prepared with methanol (MTL) and further partitioned into n-hexane (NTL), dichloromethane (DTL), and n-butanol (BTL) fractions. Antidiarrheal activity was investigated using castor oil induced diarrhea, enteropooling, and gastrointestinal transit models, while cytotoxicity was evaluated using the brine shrimp lethality bioassay. In antidiarrheal experiments, all doses (100, 200, and 400 mg/kg) of the DTL extract significantly reduced diarrheal stool frequency, volume and weight of intestinal contents, and gastrointestinal motility in mice. Similarly, in the cytotoxicity assay, all extracts exhibited activity, with the DTL extract the most potent (LC50 67.23 μg/mL). GC-MS analysis of the DTL extract identified 10 compounds, which showed good binding affinity toward M3 muscarinic acetylcholine, 5-HT3, Gut inhibitory phosphodiesterase, DNA polymerase III subunit alpha, and UDP-N-acetylglucosamine-1 carboxyvinyltransferase enzyme targets upon molecular docking analysis. Although ADME/T analyses predicted the drug-likeness and likely safety upon consumption of these bioactive compounds, significant toxicity concerns are evident due to the presence of the known phytotoxin, 2,4-di-tert-butylphenol. In summary, T. leucostaphylum showed promising activity, helping to rationalize the ethnomedicinal use and importance of this plant, its safety profile following both acute and chronic exposure warrants further investigation.

Curculigo recurvata W.T.Aiton exhibits anti-nociceptive and anti-diarrheal effects in Albino mice and an in silico model.

Background Curculigo recurvata (C. recurvata) is an enthnomedicinally important herb reported to have significant medicinal values. The present study aimed to explore the in vivo and in silico anti‐nociceptive and anti‐diarrheal effects of a C. recurvate rhizome methanol extract (Me‐RCR).MethodsThe analgesic effects of Me‐RCR were assessed using acetic acid‐induced writhing and the formalin‐induced flicking test. The drugs were administered intraperitoneally (IP) at doses of 200 and 400 mg/kg body weight (bw). Anti‐diarrheal activity was evaluated by assessing intestinal motility, hypersecretion, and fecal score in mice at oral doses of 200 and 400 mg/kg·bw. Computer facilitated analyses for anti‐nociceptive and anti‐diarrheal activities of three isolated compounds from C. recurvata were undertaken to identify the best‐fit phytoconstituents.ResultsThe Me‐RCR showed significant (P < .05) peripheral anti‐nociception at the highest dose. The extract inhibited both early and late phases of nociception in the formalin‐induced writhing test. In the castor oil‐induced diarrhoea model, the extract significantly (P < .05) prolonged the onset time of diarrhoea, inhibited percentage of diarrhoea, and decreased both the volume and weight of intestinal contents. Rates of intestinal fluid accumulation inhibition were (33.61 ± 1.00)% and (46.44 ± 0.89)% at Me‐RCR doses of 200 and 400 mg/kg·bw, respectively. Moreover, a significant (P < .05) reduction in gastrointestinal motility was observed. An absorption, distribution, metabolism, excretion and/or toxicity (ADME/T) test showed that the selected compounds yielded promising results, satisfying Lipinski's rule of five for predicting drug‐like potential. Notably, of the three phytoconstituents curculigine and isocurculigine possessed the highest affinity for the COX‐1 and COX‐2. Isocurculigine was also identified as the most effective anti‐diarrheal compound in the computer‐facilitated model.ConclusionAn extract of the plant C. recurvata showed potential analgesic and anti‐diarrheal activity due to the presence of one or more active secondary metabolite(s).

Regression analysis to predict the impact of dietary neutral detergent fiber on carcass yield in swine.

Research has shown that carcass yield in swine is reduced when ingredients with high neutral detergent fiber (NDF) content. Carcass yield reduction from feeding high-fiber ingredients results from an increase in the weight of intestinal contents. NDF has been shown to result in the digestive contents to swell in the large intestine by absorbing water thus increasing the fecal volume in the large intestine. Considering the financial implications of changing carcass yield, the objective of this project was to develop a regression equation to estimate carcass yield from dietary NDF and strategies where high-NDF ingredients are taken out of the diet in the last dietary phases before slaughter (withdrawal period; WP). Data from 8 experiments (43 observations) originated from 6 journal articles and 1 technical memo were used to develop the regression equation. The WP of high NDF ingredients was either none or ranged from 5 to 63 d in the experiments. Treatment diets of each trial were reformulated to obtain dietary nutrient content using the NRC ingredient library (NRC, Nutrient requirements of swine. 11th ed, 2012). Composition of experimental diets was used to calculate dietary net energy, crude protein, crude fiber, NDF, and acid detergent fiber in the last two dietary phases. These dietary compositions along with the number of days of WP were used to develop regression equations. The model was determined using a step-wise selection procedure starting with guided forward selection through individual predictor variables, with a statistical significance at P < 0.05 used to determine inclusion of terms in the final model. The regression analysis showed that WP, NDF level in the dietary phase prior to the final phase (NDF1), NDF level in the last dietary phase before marketing (NDF2), and the interaction between NDF2 and WP were the most important variables in the dataset to predict carcass yield. The resulting regression equation was as follows: carcass yield, % = 0.03492 ± 0.02633 × WP (d) - 0.05092 ± 0.02862 × NDF1 (%) - 0.06897 ± 0.02931 × NDF2 (%) - 0.00289 ± 0.00216 × (NDF2 [%] × WP [d]) + 76.0769 ± 1.33730. In conclusion, high levels of NDF up to slaughter had a negative impact on carcass yield. Increasing the length of the WP improved carcass yield; however, the effect of WP was dependent on the level of NDF2. The equation herein provides a tool to estimate of the impact of dietary NDF on carcass yield.

Evaluation of the Antispasmodic and Antisecretory Activities of the 80% Methanol Extracts of Verbena officinalis L: Evidence From In Vivo Antidiarrheal Study.

Verbena officinalis L. has a folkloric repute for the management of digestive disorders, including diarrhea. However, the safety and efficacy of the plant material has not been scientifically validated yet. This study was, therefore, aimed to evaluate the overall antidiarrheal activity of the 80% methanol extracts of V officinalis in mice. The antidiarrheal activity of the 80% methanol extracts of the roots (R-80ME) and the leaves (L-80ME) of V officinalis was tested in castor oil–induced diarrhea in mice. R-80ME was further evaluated using charcoal meal and entero-pooling. In each test, group I and group II (controls) received 10 mL/kg distilled water and standard drug (5 mg/kg loperamide), respectively, whereas groups III, IV, and V (test groups) received 100, 200, and 400 mg/kg of the 80ME, respectively. The R-80ME at 200 mg/kg (P < .01) and 400 mg/kg (P < .001) significantly delayed the onset of diarrhea compared with negative control. Both R-80ME and L-80ME at 200 and 400 mg/kg significantly decreased the frequency of wet fecal outputs (P < .01). Generally, 70.24% inhibition of the number of wet fecal output was recorded at R-80ME 400 mg/kg. Results from the charcoal meal test revealed that the R-80ME at 200 (P < .01) and 400 mg/kg (P < .001) produced a significant antimotility effect. In entero-pooling test, the R-80ME, at 200 and 400 mg/kg doses (P < .01), showed a significant decline in both the volume and weight of intestinal contents. The maximum in vivo antidiarrheal index was determined to be 95.25 at dose of 400 mg/kg R-80ME. This study demonstrated that the 80ME, mainly the root extract, produced promising antidiarrheal activity and hence provides a scientific support for acclaimed traditional use of the plant material for treatment of diarrheal diseases.

The immune-regulating effect of Xiao\u2019er Qixingcha in constipated mice induced by high-heat and high-protein diet

BackgroundXiao’er Qixingcha (EXQ) has been extensively applied to relieve dyspepsia and constipation in children for hundreds of years in China. However, the therapeutic mechanism underlying its efficacy remained to be defined. The present study aimed to clarify the possible laxative and immune-regulating effects of EXQ on two models of experimental constipation in mice, which mimicked the pediatric constipation caused by high-heat and high-protein diet (HHPD).MethodsThe two models of constipated mice were induced by HHPD or HHPD + atropine respectively. To investigate the laxative and immune-regulating activities of EXQ, animals were treated with three doses of EXQ (0.75, 1.5 and 3 g/kg) for 7 consecutive days. The fecal output parameters (number and weight), weight of intestinal content and, the thymus and spleen indexes were measured. The levels of sIgA, IL-10, TNF-α and LPS in colon and serum were determined by ELISA. Furthermore, the pathological changes of colon tissue were examined after routine H&E staining.ResultsBoth HHPD and HHPD + atropine treatments obviously inhibited the fecal output and reduced the colonic sIgA, prominently increased the levels of IL-10 and TNF-α in colonic tissue and elevated the contents of LPS in serum and colonic tissues. In contrast, oral administration of EXQ significantly improved the feces characters and dose-dependently decreased the intestinal changes in both models. In HHPD model test, EXQ efficaciously boosted the sIgA level in a dose-dependent manner, significantly elicited decreases in TNF-α and IL-10 levels, and evidently decreased the spleen and thymus indexes. In HHPD + atropine model test, EXQ treatment reversed the pathological changes by not only dramatically decreasing the spleen index and the levels of LPS and IL-10, but also markedly elevating the thymus index. Furthermore, microscopic observation revealed that EXQ treatment maintained the integrity of colonic mucosa, and protected the colonic tissues from inflammation in the both models.ConclusionsEXQ exhibited prominent laxative activity and effectively protected the colonic mucosal barrier in two models of constipated mice, of which the mechanism might be closely associated with its propulsive and immune-regulating properties. The current results not only validated the rationale for the clinical application of EXQ in pediatric constipation related symptoms, but also threw new light on the immune-inflammatory responses accompanied with chronic constipation pathology.

Investigating Potential Modes of Actions of Mimusops kummel Fruit Extract and Solvent Fractions for Their Antidiarrheal Activities in Mice.

Background Fruits of Mimusops kummel A. DC. (Sapotaceae) are traditionally used for the treatment of diarrhea. The present study aimed at investigating modes of actions of this fruits for antidiarrheal action to guide future drug development process. Methods Fractions of chloroform, n-butanol, and water were obtained from 80% methanol extract, which was prepared by maceration. Antidiarrheal activities and the modes of actions were investigated in mice. Results In castor oil induced diarrheal model, the extract delayed onset of diarrhea and reduced number and weight of feces at all tested doses significantly. In this model all fractions significantly delayed onset of diarrhea at all tested doses. Charcoal meal test showed that the extract and all the fractions produced a significant antimotility effect at all tested doses. Enteropooling test showed that the extract as well as n-butanol and aqueous fractions at all tested doses produced a significant decline in volume and weight of intestinal contents, whereas chloroform fraction had substantial effect only at high dose. Conclusion This study demonstrated that the extract and solvent fractions produced antidiarrheal activities due to dual inhibitory effect, intestinal motility, and fluid secretion, with the aqueous fraction being the most active among fractions in three models.

Anti-diarrhoeagenic Properties of Aqueous Extract of Phragmanthera capitata S. Balle in Albino Rats

Aim: To investigate diarrhoeagenic activities of aqueous extract of Phragmanthera capitata (AEPC) in albino rats. Place and Duration of Study: The study was carried out at the Department of Pharmacology, Faculty of Basic Medical Sciences, University of Calabar, Calabar, Nigeria, between October 2013 and January 2014. Methodology: Diarrhoeagenic activities were assessed using three models; enteropooling, gastrointestinal transit and faecal discharge. In each of these models, all rats were overnight fasted with access to drinking water until the start of the experiment. The rats were randomized into 5 groups of 7 rats. Group I (control) received saline (10 ml/kg), Group II (standard) and Groups III-V (test) received AEPC (100, 200, 300 mg/kg respectively) by oral gavage. To assess enteropooling activity, castor oil (1 ml) and magnesium sulfate (10 ml/kg in saline) were used to induce enteropooling in two separate experiments. Group II in both experiments received loperamide (3 mg/kg). Intestinal contents were weighed before and after discharging the contents and recorded. To investigate gastrointestinal transit, 0.25 ml of charcoal meal was administered to the rats Original Research Article European Journal of Medicinal Plants, 4(6): 743-752, 2014 744 with standard group receiving atropine (5 mg/kg i.p.). Distance traveled by charcoal meal was measured and recorded. To assess the rate of faecal discharge, castor oil (1 ml) was used to induce diarrhoea and faeces were collected, weighed and recorded. One way Analysis of Variance (ANOVA) was performed followed by Tukey test as post hoc. Results: The results revealed significant (P =.05) reduction in faecal discharge, weight of intestinal contents and distance traveled by charcoal meal. Conclusion: AEPC possesses anti-secretory, anti-electrolyte permeability and hence anti-diarrhoeagenic properties.

Phytochemical constituents and antidiarrhoeal effects of the aqueous extract of Terminalia superba leaves on Wistar rats

The aqueous extract of Terminalia superba leaves was subjected to phytochemical screening. Antidiarrhoeal property of the extract was determined at 50, 100 and 150 mg/kg body weight in castor oil-induced diarrhoeal Wistar rats. Phytochemical screening revealed the presence of saponins, cardenolides, triterpenes, flavonoids, steroids, phenolics and tannins, whereas alkaloids, anthraquinones and phlobatanins were not detected. The time of induction of diarrhoea was significantly (P < 0.05) prolonged at all the doses of the extract. The frequency of stooling and feacal parameters (total number of feaces and number of wet feaces), intestinal fluid accumulation (enteropooling) and the weight of intestinal content were significantly reduced. The data in the present study indicate that the aqueous extract of T. superba leaves possessed antidiarrhoeal properties. Effect of Guar Gum, Lignin and Pectin on Proteolytic Enzyme Levels in the Gastrointestinal Tract of the Rat: A Time-Based Study

The effects of dietary pectin (P), guar gum (G) and lignin (L) on stomach emptying and potential levels of pepsin, trypsin and chymotrypsin during a 2-h period after force-feeding were investigated in growing rats. All of the fibers delayed stomach emptying by 21–26 min. Total potential pepsin activity over 2 h decreased for P (57%), G (44%) and L (20%). In the intestine, total potential trypsin activity over 2 h increased for L (16%) but decreased for P (21%). Total potential chymotrypsin activity over 2 h increased for L (54%) and G (39%). Sixteen to 21% of the variability in intestinal activity over time was statistically attributable to variation in the weight of intestinal contents. The results indicate that fiber components altered proteolytic enzyme levels in the gastrointestinal tract, but the decreased protein utilization previously observed with these fibers is probably not due to reduced levels of intestinal proteases.
