Leszl1, Rita Benko1, Zsombor Lacza1, Zoltan Jarai2, Peter Sandor1 and Zoltan Benyo1 1 Institute of Human Physiology and Clinical Experimental Research, Semmelweis University, Hungary 2 1st Department of Internal Medicine, Semmelweis University, Hungary The influence of endocannabinoids on the resting blood flow and on hypoxia/hypercapnia (H/H)-induced vasodilation was investigated in the cerebral cortex with laser-Doppler (LD) flowmetry in anesthetized adult male Wistar rats. Inhibition of endocannabinoid reuptake by AM-404 (10 mg/kg) resulted in a transient increase of the resting cerebrocortical blood flow (CBF), which had a peak (+65±16 %) at 5.2±0.6 minutes and disappeared within 10 minutes. The CBF changes were accompanied by reduction of the ventilation rate as well as by increased arterial CO2 tension (PaCO2) and decreased O2 tension (PaO2). Application of the CB1 receptor antagonist AM-251 (10 mg/kg iv) did not alter baseline CBF but significantly increased the H/H-induced hyperemia: by 30 % during mild H/H (PaO2=75-85, PaCO2=50-60 mmHg), by 22 % during moderate H/H (PaO2=55-65, PaCO2=80-90 mmHg), and by 14 % during severe H/H (PaO2=45-50, PaCO2=90-100 mmHg). In contrast, vehicle of AM-251 had no significant effect. H/H did not change mean arterial pressure in untreated or vehicle-treated animals, although following AM-251 treatment it induced slight hypertension (+11 ± 6 mmHg). Furthermore, AM-251 was able to prevent the effects of AM-404 while the VR1 receptor antagonist capsazepine (20 mg/kg) had no effect. In conclusion our results indicate that endocannabinoids may increase the resting CBF either by a CB1-mediated direct vasodilator mechanism or indirectly via respiratory depression and the consequent changes of the blood gas tensions. Interestingly, activation of CB1 receptors attenuates the H/H induced cerebrocortical hyperaemia by a yet undefined mechanism. Supported by OTKA K62375 and NKTH H07-BEL74286 grants.
