Optimizing planting density enhances the multi-scale structural characteristics and in vitro digestibility of maize starch via modulating the size distribution of granules

The modification of starch through agricultural practices is becoming increasingly significant for producing healthy foodstuffs and raw materials for industrial applications, consequently gaining momentum in academic research. This study examined how three different planting densities influenced the distribution of granule sizes, multi-scale structural characteristics, and in vitro digestibility of maize starch. The results showed that planting density significantly enhanced grain yield and relative crystallinity, and significant increases were also observed in the contents of both rapidly and slowly digestible starch. The surface- and volume-weighted mean diameter of granules significantly increased under the medium level (6.75 × 104 plants ha−1), and then decreased under high planting density level. As planting density level increased, the amylose content, peak viscosity, and hardness varied from 23.3 to 26.4 %, from 1962 to 2659 mPa·s, and from 129.3 to 307.6 g, respectively. However, no change was found in crystalline structure of maize starch. These results indicated that optimizing planting density could effectively improve grain yield and starch characteristics of maize, with the best effect under the level of 6.75 × 104 plants ha−1.

Rice bran oil-in-water optimized emulsions for the development of plant-based foods and beverages

Aim: This work aimed to develop rice bran oil-in-water (O/W) emulsions, stabilized with different mixtures of pea protein concentrate (PPC), Arabic gum (AG), and maltodextrin (MD), as the basis for the formulation of plant-based food products. Methods: The effects of the aqueous phase formulation on the properties of the resulting O/W emulsions were evaluated by a mixture design approach. Volume-weighted mean diameter (D[4,3]) of the emulsion particles and polydispersity expressed as the difference of D[4,3] – D[3,2], apparent viscosity at a shear rate of 200 s–1 and backscattering at different times associated to the global stability of the emulsions were studied as response variables. A multi-response optimization was carried out and mathematical models were validated. Results: The ternary mixtures of the aqueous phase showed significant antagonism between the three components in all the response variables. The optimal formulation of the aqueous phase for the O/W emulsions obtained after three homogenization cycles was 78% PPC and 22% MD. The properties of the O/W optimal emulsion were according to the ones predicted by the model. Conclusions: The best-formulated emulsion is promising for developing plant-based foods and beverages.

Enhancing the Techno-Functional Properties of Lentil Protein Isolate Dispersions Using In-Line High-Shear Rotor-Stator Mixing.

In response to global challenges such as climate change and food insecurity, plant proteins have gained interest. Among these, lentils have emerged as a promising source of proteins due to their good nutritional profile and sustainability considerations. However, their widespread use in food products has been impeded by limited solubility. This study aimed to investigate the potential of high-shear mixing, a resource-efficient technique, to enhance lentil protein solubility and its functional properties. Red lentil protein isolate powders were rehydrated and subjected to a semi-continuous in-line high-shear treatment at 10,200 rpm for a timespan ranging from 0 to 15 min. The results highlighted a significant (p < 0.05) increase in solubility from 46.87 to 68.42% after 15 min of shearing and a reduction in particle size as a result of the intense shearing and disruption provided by the rotor and forced passage through the perforations of the stator. The volume-weighted mean diameter decreased from 5.13 to 1.72 µm after 15 min of shearing, also highlighted by the confocal micrographs which confirmed the breakdown of larger particles into smaller and more uniform particles. Rheological analysis indicated consistent Newtonian behaviour across all dispersions, with apparent viscosities ranging from 1.69 to 1.78 mPa.s. Surface hydrophobicity increased significantly (p < 0.05), from 830 to 1245, indicating exposure of otherwise buried hydrophobic groups. Furthermore, colloidal stability of the dispersion was improved, with separation rates decreasing from 71.23 to 24.16%·h-1. The significant enhancements in solubility, particle size reduction, and colloidal stability, highlight the potential of in-line high-shear mixing in improving the functional properties of lentil protein isolates for formulating sustainable food products with enhanced techno-functional properties.

Microplastic transport and deposition in a beach-dune system (Saunton Sands-Braunton Burrows, southwest England)

Although microplastics (MPs) are ubiquitous contaminants that have been extensively studied in the marine setting, there remain gaps in our understanding of their transport and fate in the coastal zone. In this study, MPs isolated from surface sediments sampled from a large beach-dune complex in southwest England have been quantified and characterised. Concentrations above a detectable size limit of 30 to 50 μm ranged from about 40 to 560 MP kg−1 dry weight but, despite local sources of plastics such as an estuary and seasonal tourism, there were no significant differences in median concentrations between different orthogonal foreshore transects and the dunes or according to zonal location on the beach. The majority of MPs were black and blue fibres of <1 mm in length that were constructed of polymers of density > 1 g cm−3 (e.g., rayon, polyester, acrylic). A significant correlation was found between MP concentration and the proportion of very fine sand (100 to 250 μm) but relationships with other granulometric or compositional markers of sediment (e.g., volume-weighted mean diameter, circularity, calcium content) were not evident. An association of MP concentration with very fine sand was attributed to similar particle depositional characteristics and the entrapment of fibres within small interstitial spaces. Overall, the observations reflect the wavelaid and windlaid deposition of MPs from a diffuse, offshore source, and, despite their role as accumulators of particles from the foreshore, dunes do not appear to act as a landward barrier of MPs.

The use of hydrodynamic cavitation for waste-to-energy approach to enhance methane production from waste activated sludge

Anaerobic digestion in wastewater treatment plants converts its unwanted end product – waste activated sludge into biogas. Even if the process is well established, pre-treatment of the sludge can further improve its efficiency. In this study, four treatment regimes for increasing methane production through prior sludge disintegration were investigated using lab-scale cavitation generator and real sludge samples. Three different cavitating (attached cavitation regime, developed cloud shedding cavitation regime and cavitation in a wake regime) and one non-cavitating regime at elevated static pressure were studied in detail for their effectiveness on physical and chemical properties of sludge samples. Volume-weighted mean diameter D[4,3] of sludge's particles decreased by up to 92%, specific surface area increased by up to 611%, while viscosity (at a shear rate of 3.0 s−1) increased by up to 39% in the non-cavitating and decreased by up to 24% in all three cavitating regimes. Chemical changes were more pronounced in cavitating regimes, where released soluble chemical oxygen demand (sCOD) and increase of dissolved organic matter (DOM) compounds by up to 175% and 122% were achieved, respectively. Methane production increased in all four cases, with the highest increase of 70% corresponding to 312 mL CH4 g−1 COD. However, this treatment was not particularly efficient in terms of energy consumption. The best energy balance was found for the regime with a biochemical methane potencial increase of 43%.

Ethanol, not water, should be used as the dispersant when measuring microplastic particle size distribution by laser diffraction

Size distribution is a crucial characteristic of microplastics (MPs). A typical method for measuring this property is wet laser diffraction. However, when measuring size distributions of MPs, despite it being a poor dispersant for many MPs, water is commonly selected, potentially limiting the reliability of reported measurements. To evaluate dispersant suitability, different aqueous concentrations of ethanol (0, 10, 20, 30, 40, 50, 75, 100 wt%) and aqueous solutions of 0.001 wt% Triton X-100 and a mixture comprising 10 wt% sodium pyrophosphate and 10 wt% methanol were used as dispersants in a laser granulometer (Mastersizer 2000) to determine particle size distributions (PSDs) of granular polyethylene MP35, MP125 and MP500 particles (nominally <35, <125 and, < 500 μm in size). The reliability of the PSDs depended on the dispersant used and size of primary MPs. With increasing ethanol concentrations, PSD curves of MP35 particles shifted from multi-modal to mono-modal distributions. The measured size distribution reduced from 1588.7 to 4.5 μm in water to 39.9 to 0.1 μm in 100 wt% ethanol. Generally, as ethanol concentration increased, uncertainty associated with the PSD parameters decreased. Although Triton X-100 and the mixed solution also showed better dispersion than water, measured particle sizes and coefficient of variation (COV, %) were notably larger than those for 100 wt% ethanol. Similar trends were observed for larger-sized MP125 and MP500 particles, but differences in PSD curves, PSD parameters, and COV (%) among dispersants were less pronounced. In all dispersants, the volume weighted mean diameters (VWMD) in 100 wt% ethanol (MP35: 14.1 μm, MP125: 102.5 μm, MP500: 300.0 μm) were smallest and close to diameters determined from microscope observations (MP35: 14.6 μm, MP125: 109.0 μm, MP500: 310.6 μm). Therefore, for accurate determinations of the PSDs of MP by wet laser diffraction, ethanol rather than water should be used as the dispersant.

Impact of adding xylooligosaccharides encapsulated in butter: Microstructural, optical, rheological and sensory aspects

This study investigated the microstructure, rheological properties, and sensory characteristics of butters produced with free and encapsulated xylooligosaccharides (XOS). Four formulations of butter were processed: BCONT: 0 % w/w XOS (control); BXOS: 20% w/w free XOS; BXOS-ALG: 20% w/w XOS microencapsulated with alginate (XOS-alginate ratio of 3:1 w/w); and BXOS-GEL: 20% w/w XOS microencapsulated with alginate-gelatin (XOS-alginate-gelatin ratio of 3:1:1.5 w/w). The microparticles showed a bimodal distribution, low size and low span values, demonstrating physical stability to be included in emulsions. The XOS-ALG presented surface weighted mean diameter (D3.2) of 90.24 µm, volume-weighted mean diameter (D4.3) of 131.8 µm, and Span of 2.14. In contrast, the XOS-GEL presented D3.2 of 82.80 µm, D4.3 of 141.0 µm, and a Span of 2.46. Products with XOS were characterized by higher creaminess, sweet taste, and lower salty taste than the control. However, the addition form significantly impacted the other evaluated parameters. The utilization of XOS in a free form (BXOS) resulted in smaller droplet sizes (1.26 μm) than encapsulated XOS and control (XOS-ALG = 1.32 µm / XOS-GEL = 1.58 µm, / BCONT = 1.59 µm), and changes in the rheological parameters (higher values of shear stress, viscosity, consistency index, rigidity (J0), and Newtonian viscosity (ηN) and lower elasticity (τ)). Furthermore, it changed the color parameters (more yellow and dark color, lower L* and higher b* values). On the other hand, the utilization of micropaticles of XOS (BXOS-ALG and BXOS-GEL) kept shear stress, viscosity, consistency index, rigidity (J0), and elasticity (τ) more similar to control. The products had a less intense yellow color (lower b* values) and was perceived with more consistency and butter taste. However, the presence of particles was perceived by consumers. The results suggest that consumers were more attentive to reporting flavor-related attributes than texture. In conclusion, adding microparticles of XOS could improve butter's rheological and sensory properties. Cymbopogon martinii essential oil nanoemulsions: Physico-chemical characterization, antibacterial and antibiofilm potential against Enterococcus faecalis

Cymbopogon martinii is an aromatic crop cultivated for the production of its essential oil (EO) which has different applications. Bearing in mind that C. martinii EO (CMEO) has already been proved for high antibacterial potential the aim of this study was to formulate stable CMEO nanoemulsions, being capable to combat Enterococcus faecalis biofilm, especially within infected tooth’s root canal. Ten emulsions with variable CMEO content (2.5–10 % v/v) and Tween 80 (T80, 2.5–15 % v/v) were formulated (designated as E CMEO:T80 ) and screened for volume-weighted mean diameter (D4,3) and stability, using dynamic light scattering measurements. Chemical composition of stable nanoemulsions was monitored by UV–VIS and FTIR, while their antibacterial activity was estimated in microdilution assay. Antibiofilm properties of selected nanoemulsions were tested in vitro (cristal violet assay) and ex vivo (within root canals of the extracted teeth). Results showed that formulations containing 10 % of T80 and CMEO volume ≤ 6 %, as well as those having constant 2.5 % of CMEO and variable T80 volume (≤ 10%), namely E 2.5:2.5 , E 2.5:5, E 2.5:7.5, E 2.5:10, E 4:10 , and E 6:10 , were stable and nanosized (D4,3 <100 nm). FTIR and UV–VIS analyses confirmed successful encapsulation of the CMEO by T80 and showed that nanoemulsification did not affect CMEO composition. Evaluation of antibacterial potential showed that the most efficient were E 6:10 , E 2.5:2.5 and E 4:10 (MIC values 0.37–1.97 mg mL −1 ). Analysis of dependency of determined MICs on mean diameter and CMEO volume showed that antibacterial potential increased with both parameters. E 6:10 , E 2.5:2.5 and E 4:10 also reduced in vitro biofilm (inhibitory range 17.6–58.4 %), as well as intracanal biofilm (reduction ~2Log CFU) . Presented results suggest E 6:10, E 4:10 and E 2.5:2.5 as the good candidates for further research. • Emulsions with different ratio of C. martini oil and Tween 80 were prepared. • Characterization of the nanoemulsions showed successful oil encapsulation. • Nanoemulsions E 6:10 , E 2.5:2.5 , E 4:10 proved high antibacterial effect. • Nanoemulsions E 6:10 , E 2.5:2.5 and E 4:10 reduced ex vivo intracanal biofilm. • Nanoemulsification improved antibiofilm potential of the C. martini oil.

Microphysical Characteristics of Precipitating Stratiform Clouds in North China Revealed by Joint Observations of an Aircraft and a Polarimetric Radar

Abstract Microphysical structures and processes in a case of precipitating stratiform clouds in North China on 21 May 2018 are investigated using joint observations from an aircraft and an X-band polarimetric radar. The results show that there are enhancements in differential reflectivity (ZDR) and specific differential phase (KDP) above the 7-km altitude, consistent with the existence of dendrites and platelike ice crystals. The horizontal reflectivity factor (ZH) increases and ZDR decreases downward above the melting layer (ML), due to the prevalent aggregation process, which is confirmed by the downward increasing volume-weighted mean diameter (Dm) and decreasing total number concentration (Nt) observed by the aircraft. Within the ML, the concentration of median-sized particles (2–5 mm) decreases rapidly downward due to the melting process. Within approximately the top 2/3 of the ML, the melting particles’ mean and maximum sizes increase, demonstrating the dominance of the aggregation process. This causes the enhancements of ZH and ZDR within the radar bright band together with the increase in the dielectric constant. Within the bottom 1/3 of the ML, the breakup process is responsible for the decreasing Dm and increasing Nt observed by the aircraft. Below the ML, the measurements by the polarimetric radar and the aircraft only show slight variance with altitude, indicating the near balance between microphysical processes favored by the nearly saturated air. The results of the microphysics in the stratiform case would help improve the microphysical parameterization of numerical modeling in the future.

Effects of growth temperature on multi-scale structure of root tuber starch in sweet potato

Sweet potato was planted at three soil and air temperatures (21, 25 and 28 °C) with the same humidity and light time/intensity. Root tuber starches were isolated, and their multi-scale structures were investigated to reveal the effects of growth temperature on starch properties. Growth temperature did not change the morphology and amylose content of starch, but markedly increased the size of starch from volume-weighted mean diameter 12.2 μm to 17.0 μm. Starch grown at high growth temperature exhibited less A branch-chains and lower branching degree of amylopectin and more B2 and B3+ branch-chains of amylopectin than at low growth temperature. With increasing growth temperature, starch changed from CC-type to CA-type, its relative crystallinity and lamellar peak intensity increased, and the thickness of crystalline and amorphous lamellae did not significantly change. Starch grown at high growth temperature exhibited significantly higher gelatinization temperature than at low growth temperature, but had similar gelatinization enthalpy.

Experimental and numerical investigations on the condensed combustion products of Mg-based solid fuels

The exploration of condensed combustion products (CCPs) is essential to reveal the ignition, combustion, and infrared radiation mechanisms of Mg-based (Magnesium/polytetrafluoroethylene/Viton) solid fuels that have been considerably employed in the field of aerospace. Systematic experiments and numerical simulations are performed with the physicochemical properties (including morphology, element distribution, phase composition and particle size distribution) and flow field distribution characteristics of CCPs, respectively. The effects of formulation and pressure on the combustion behaviors of pyrotechnic compositions are thoroughly investigated based on thermal analysis measurements, laser ignition, optical diagnosis and CCPs collection. The results demonstrated that the major solid products for all cases are MgF2 agglomerates and MgO smoke oxide particles (SOPs). Increasing the Mg content in the formulation is simultaneously beneficial for the reduction of large agglomerates and SOPs. As the pressure is decreased from 0.1 MPa to 0.02 MPa, the volume weighted mean diameter of CCPs increases significantly due to the decrease in burning rate. The numerical model established in this paper better approximates the realistic combustion process by coupling the heterogeneous condensation reactions, and is adopted to predict the distribution of CCPs in the combustion flow field under different experimental conditions. Numerical results reveal that the MgF2 is mainly distributed in the anaerobic combustion core zone immediately above the burning surface, while MgO is located in the peripheral aerobic combustion diffusion zone. The negative pressure environment has more profound influence on the oxidation reaction of Mg than fluorination reaction. The scientific findings are expected to facilitate the development of more complete combustion and infrared radiation models, while theoretically providing in-depth guidance for the increasing military and civilian application of Mg-based solid fuels.

Dual-Frequency Radar Retrievals of Snowfall Using Random Forest

The microphysical parameters of snowfall directly impact hydrological and atmospheric models. During the International Collaborative Experiment hosted at the Pyeongchang 2018 Olympic and Paralympic Winter Games (ICE-POP 2018), dual-frequency radar retrievals of particle size distribution (PSD) parameters were produced and assessed over complex terrain. The NASA Dual-frequency Dual-polarized Doppler Radar (D3R) and a collection of second-generation Particle Size and Velocity (PARSIVEL2) disdrometer observations were used to develop retrievals. The conventional look-up table method (LUT) and random forest method (RF) were applied to the disdrometer data to develop retrievals for the volume-weighted mean diameter (Dm), the shape factor (mu), the normalized intercept parameter (Nw), the ice water content (IWC), and the snowfall rate (S). Evaluations were performed between the D3R radar and disdrometer observations using these two methods. The mean errors of the retrievals based on the RF method were small compared with those of the LUT method. The results indicate that the RF method is a promising way of retrieving microphysical parameters, because this method does not require any assumptions about the PSD of snowfall.

Mind the Gap - Part 3: Doppler Velocity Measurements From Space

Convective motions and hydrometeor microphysical properties are highly sought-after parameters for evaluating atmospheric numerical models. With most of Earth’s surface covered by water, space-borne Doppler radars are ideal for acquiring such measurements at a global scale. While these systems have proven to be useful tools for retrieving cloud microphysical and dynamical properties from the ground, their adequacy and specific requirements for spaceborne operation still need to be evaluated. Comprehensive forward simulations enable us to assess the advantages and drawbacks of six different Doppler radar architectures currently planned or under consideration by space agencies for the study of cloud dynamics. Radar performance is examined against the state-of-the-art numerical model simulations of well-characterized shallow and deep, continental, and oceanic convective cases. Mean Doppler velocity (MDV) measurements collected at multiple frequencies (13, 35, and 94 GHz) provide complementary information in deep convective cloud systems. The high penetration capability of the 13 GHz radar enables to obtain a complete, albeit horizontally under-sampled, view of deep convective storms. The smaller instantaneous field of view (IFOV) of the 35 GHz radar captures more precise information about the location and size of convective updrafts above 5–8 km height of most systems which were determined in the portion of storms where the mass flux peak is typically located. Finally, the lower mean Doppler velocity uncertainty of displaced phase center antenna (DPCA) radars makes them an ideal system for studying microphysics in shallow convection and frontal systems, as well as ice and mixed-phase clouds. It is demonstrated that a 94 GHz DCPA system can achieve retrieval errors as low as 0.05–0.15 mm for raindrop volume-weighted mean diameter and 25% for rime fraction (for a −10 dBZ echo).

Experimental investigation in the permeability of methane hydrate-bearing fine quartz sands

The permeability is one of the intrinsic parameters that determines the fluid flow in the porous media. The permeability in hydrate-bearing sediments affects the gas recovery and production of hydrate reservoirs significantly. The irregular permeability characteristics are challenging for fine-grained hydrate-bearing sediments. In this study, a series of experiments was conducted using an one-dimensional pressure vessel to investigate the hydrate formation characteristics and the permeability in hydrate-bearing fine quartz sands (volume weighted mean diameter was 36.695 μm). Hydrate saturations (0–26% in volume) were controlled and calculated precisely based on the amount of injected water and gas, the system pressure and temperature. The results indicated that the hydrate nucleation induction period was completed during gas injection, and the average time of hydrate formation was within 500 min. The permeability of methane hydrate-bearing fine quartz sands was investigated by steady gas volume flow. For hydrate saturation lower than 13.94%, the hydrate mostly formed in grain-coating, the permeability reduction exponent calculated by Parallel Capillary, Kozeny Grain Coats and Simple Cubic Filling models were 2.00, 2.10 and 1.74 respectively, and Simple Cubic Filling model was in accordance with the experimental data best. However, for hydrate saturation ranged from 13.94% to 25.91%, the permeability increased due to the flocculation structure formation of fine quartz sands and hydrate, which caused the increase of effective porosity. A new relationship among hydrate saturations, effective porosity, the ratio of permeability in the presence and the absence of hydrate was developed. This study developed the mathematical models for predicting the permeability with hydrate saturation in fine quartz sands, which could be valuable for understanding the characteristics of hydrate-bearing fine-grained sediments.

Parameterized Forward Operators for Simulation and Assimilation of Polarimetric Radar Data with Numerical Weather Predictions

Many weather radar networks in the world have now provided polarimetric radar data (PRD) that have the potential to improve our understanding of cloud and precipitation microphysics, and numerical weather prediction (NWP). To realize this potential, an accurate and efficient set of polarimetric observation operators are needed to simulate and assimilate the PRD with an NWP model for an accurate analysis of the model state variables. For this purpose, a set of parameterized observation operators are developed to simulate and assimilate polarimetric radar data from NWP model-predicted hydrometeor mixing ratios and number concentrations of rain, snow, hail, and graupel. The polarimetric radar variables are calculated based on the T-matrix calculation of wave scattering and integrations of the scattering weighted by the particle size distribution. The calculated polarimetric variables are then fitted to simple functions of water content and volume-weighted mean diameter of the hydrometeor particle size distribution. The parameterized PRD operators are applied to an ideal case and a real case predicted by the Weather Research and Forecasting (WRF) model to have simulated PRD, which are compared with existing operators and real observations to show their validity and applicability. The new PRD operators use less than one percent of the computing time of the old operators to complete the same simulations, making it efficient in PRD simulation and assimilation usage.

Production of vitex ( Vitex agnus ‐ castus L.) extract in powder form using spray‐drying: Potential for the production of functional foods

Vitex (Vitex agnus-castus L.) have bioactive compounds with potential to reduce the symptoms of menopause and premenstrual syndrome, as proved by several studies. However, such compounds may have low stability, and the spicy sensation caused by vitex fruits is a challenge for application in foods. Thus, the aim of this study was to produce microparticles by spray-drying loaded with vitex extract, and evaluate the physical and chemical properties of the powders for further application in new functional foods. The powders presented good process yield (ca. 63.4%) and retention (ca. 77.7%), volume weighted mean diameter of approximately 20.3 µm, and high stability of the bioactive compounds during the storage. Moreover, they showed good storage stability in different relative humidity, and they were classified as very low hygroscopic powders. Overall, these results are very promising and they may promote the production of a potential functional ingredient for new functional foods. Practical applications Considering the good process yield, retention and stability of the bioactive compounds during the storage, and the powder produced and characterized in the present study represents a potential functional ingredient with good technological properties. Moreover, it may be an alternative to increase the consumers’ ingestion of vitex bioactive compounds by application of the spray-dried powder in new functional foods such as chocolate and cereal bars.

Relationship between chalkiness and the structural and thermal properties of rice starch after shading during grain-filling stage

Chalkiness is a major concern in rice production and its acceptance and is increased by shade stress. However, the relationship between rice chalkiness and the structural and thermal properties of starch is unclear. Here, we investigated the effect of shade stress on rice starch properties. The chalky grain rate and chalkiness degree significantly decreased with the amylose content, Mn, and ΔH and increased with surface area- and volume-weighted mean diameters, branching degree, ratio of 1022/995 cm−1, and molecular weight polydispersity. Shade stress significantly increased the volume- and surface area-weighted mean diameters and Mw and decreased the amylose content, A chain proportion of amylopectin, Mn, and regularity of starch. These effects led to an increase in the molecular weight polydispersity and branching degree and a decrease in the crystallinity degree and 1045/1022 cm−1 ratio, thereby reducing starch ΔH and uniformity. These factors contributed to increased chalkiness of rice under shade stress.

Healing activity of hydrogel containing nanoemulsified β-caryophyllene

β-caryophyllene is a sesquiterpene present in the oil of many plant species, such as Copaifera sp., which has been shown to possesses potent anti-inflammatory action; however, its healing activity remains under study. The objectives of the present study were to produce a nanoemulsion containing β-caryophyllene followed by a hydrogel containing nanoemulsified β-caryophyllene, to evaluate the permeation profile in vitro, and to assess the in vivo healing activity, which is so far unexplored in the literature for pure β-caryophyllene and in pharmaceutical formulation. The nanoemulsion was obtained through high-pressure homogenization and the hydrogel by direct dispersion with hydroxyethylcellulose. Both formulations were characterized according to droplet size, polydispersity index, volume-weighted mean diameters, particle distribution, droplets diameters tracking, zeta potential, viscosity and bioadhesion behavior. β-caryophyllene content was determined by gas chromatography coupled with mass spectrometry (GC/MS). Both formulations presented a nanometric droplet size, negative zeta potential, high β-caryophyllene content, and were stable for 60 days. In agreement with the viscosity results, the hydrogel containing the β-caryophyllene nanoemulsion showed superior bioadhesiveness than the nanoemulsion. The skin permeation study in Franz cells demonstrated that isolated β-caryophyllene was unable to cross the stratum corneum and that its nanoemulsification promoted its permeation. On the other hand, in the simulated deeply wounded skin (dermis), no significant differences were observed between the formulations and isolated β-caryophyllene with respect to the amount of marker retention in the dermis, suggesting saturation of this skin layer. For the study of healing activity, the dorsal wound model was performed with an evaluation of the lesion size, anti-inflammatory markers, and antioxidant activity. The initial closure of the wound was achieved sooner in the group treated with the hydrogel containing the β-caryophyllene nanoemulsion, indicating its anti-inflammatory effect. The histological analysis indicated that on day 12 day of the lesion, the hydrogel presented similar results to those of the positive control group (Dersani® oil), proving effectiveness in cutaneous tissue repair.

Gelatin Hydrolysate Hybrid Nanoparticles as Soft Edible Pickering Stabilizers for Oil-In-Water Emulsions.

The aim of this study was to fabricate edible gelatin enzymic digest (GED) based gel particles that can stabilize oil-in-water (O/W) microemulsions. The gel particles were generated by covalent crosslinking, with genipin, the individual protein molecules within tannic acid-induced gelatin hydrolysate (GED-TA) particles. The ability of the genipin-treated GED-TA (GP-GED-TA) to stabilize emulsions was evaluated by Turbiscan analysis and droplet-size changes. For comparison, gelatin hydrolysate (GE) and tannic acid-induced gelatin hydrolysate particles (GED-TA) were used as controls. The mean diameters of GED, GED-TA, and GP-GED-TA particles were 0.68 ± 0.1 nm, 66.2 ± 8.4 nm, and 66.9 ± 7.2 nm, respectively. Nanomechanic analysis using atomic force microscopy(AFM) indicated the average Young’s modulu of the GP-GED-TA particles was 760.8 ± 112.0 Mpa, indicating the GP-GED-TA were soft particles. The Turbiscan stability indexes (lower values indicate a more stable emulsion) of the emulsions stabilized with GED, GED-TA, and GP-GED-TA, after storage for three days, were 28.6 ± 1.5, 19.3 ± 4.8, and 4.4 ± 1.3, respectively. After one, or 60 days of storage, the volume-weighted mean diameters (D[4,3]) of oil droplets stabilized by GP-GED-TA were 1.19 ± 0.11 μm and 1.18 ± 0.1 µm, respectively. The D[4,3] of oil droplets stabilized by GED-TA, however, increased from 108.3 ± 5.1 μm to 164.3 ± 19.1 μm during the storage. Overall, the GP-GED-TA gel particles have considerable potential for stabilization of O/W emulsions in food products.

Lipid metabolic differences in cows producing small or large milk fat globules: Fatty acid origin and degree of saturation

This study compared cows that consistently produce milk with small (volume-weighted mean diameter of 2.92-3.83 µm, with an average diameter of 3.29 µm) or large (volume-weighted mean diameter of 4.58-5.67 µm, with an average diameter of 4.92 µm) milk fat globule (MFG) size distributions in terms of the fatty acid (FA) composition of the MFG core. Selected cows fell into the respective size group over at least 3 independent measurements, including an observation period before the experiment. Further selection criteria were similar milk production traits between cows (milk yield, fat yield, fat/protein ratio) and established lactation (>50 d in milk). However, the selected groups differed in parity (parity 1-3 and 3-5 in the small and large MFG groups, respectively), and the small MFG group was an average of 25 d in milk later in their lactation period. All cows were under the same nutritional management and environmental conditions. Here, we show that cows with the small or large MFG phenotype differed in their lipid metabolism in terms of the FA composition of the MFG core. Our results indicate that cows with the small MFG phenotype produced milk with higher concentrations of unsaturated FA despite being fed the same diet. We suggest that this characteristic of the small MFG phenotype is the result of increased uptake of long-chain FA from the blood circulation. A relationship between the degree of unsaturation and MFG size was also identified in preliminary studies across other species-namely, camels, sheep, and goats. These findings show the potential for on-farm selection of cows (and potentially other dairy species) based on MFG size to produce milk with improved nutrient composition. This could lead to purpose-specific separation of milk based on MFG size and FA profile, both known to alter the technological properties of milk.
