Insensitivity of cultured rat cortical neurons to mitochondrial DNA synthesis inhibitors: Evidence for a slow turnover of mitochondrial DNA

Mitochondrial dysfunction is a major contributor to aging and neurodegeneration. Defects in mitochondrial DNA (mtDNA) have been identified in several neuromuscular diseases. Even though there is a high rate of phenotypic expression of mtDNA mutations in the central nervous system and replication of DNA introduces errors, little is known about the replicative activity of mtDNA in the brain. In this study, we investigated the sensitivity of cultured rat cortical neurons to mtDNA synthesis inhibitors as a means to assess the turnover rate of mtDNA. Four-day treatment with dideoxycytidine (ddC) (0.2 μM) or ethidium bromide (EtB) (0.25 μg/mL) reduced the mtDNA content approximately 80% in the human lymphoblastoid cell line, CEM. Concentrations of ddC ranging from 0.2 to 10 μM did not reduce mtDNA content in primary cultures of rat cortical neurons. Similarly, treatment with EtB (0.1, 0.25, and 0.5 μg/mL) did not affect significantly neuronal mtDNA. EtB (0.25 μg/mL) was effective in reducing mtDNA content in the undifferentiated embryonic carcinoma cell line, P 19. However, once P 19 cells were differentiated into a neuronal phenotype, they became insensitive to inhibition of mtDNA synthesis by EtB. Thus, cultured rat cortical neurons were less sensitive to mtDNA synthesis inhibitors than cell lines, suggesting that the turnover of mtDNA in central neurons is very slow. This may protect central neurons from accumulating mutations during the replication of mtDNA.

A survey of genes expressed in mouse embryonal carcinoma F9 cells: characterization of expressed sequence tags matching no known genes.

We prepared 2,132 expressed sequence tags (ESTs) from undifferentiated mouse embryonal carcinoma F9 cells and found that 1,416 match known gene and/or protein sequences [Nishiguchi et al. (1996) J. Biochem. 119, 749-767]. To obtain information on the functions of the remaining 716 unidentified ESTs and to develop a system for characterizing ESTs matching no known genes, we analyzed their sequences by (i) repeated database searches, using the BLASTN, BLASTX, TBLASTX, and FASTA programs, (ii) using computer programs developed or modified for this work, such as the WFASTA, ORFTRNS, and MFASTA programs, together with the DBPROSITE and GRAIL programs, and (iii) examining the expression patterns of the corresponding mRNAs in F9 cells and several organs of adult mice, using the digoxigenin-labeled dot-blot method. We found that 216 of the 716 ESTs match known gene and/or protein sequences, and 307 show significant similarities to these sequences, with a Poisson p-value < 0.01. The strategy and usefulness of such analysis for characterizing unidentified ESTs are discussed.

In vitro propagation and inducible differentiation of multipotential progenitor cells from human fetal brain

Central nervous system neurons and glia arise from undifferentiated embryonic neuroepithelial cells. Such progenitor cells from the human fetal forebrain can be propagated in vitro for extended periods, when grown on non-adhesive substrates in medium containing epidermal growth factor and insulin-like growth factor-I. These actively-dividing cells can be induced to differentiate into a variety of histochemically-characterized neurons and glia consistent with their forebrain origin. Electrophysiological recording indicates that differentiated neurons derived from these progenitors mature slowly, and display a range of glutamate- and GABA-mediated conductances characteristic of normal mammalian forebrain neurons.Our observations support a role for these trophic factors in normal development of the human brain. The methods described here may provide abundant normal, untransformed human forebrain neurons and glia for research and therapeutic applications.

Primordial germ cell-derived embryonic germ cells of the mouse—in vitro model for cytotoxicity studies with chemical mutagens

Different screening methods to detect the toxic effects of xenobiotics using cells from vertebrates and invertebrates in cytotoxicity and viability assays have been developed, but up to now appropriate in vitro methods with mammalian germ cells have not been available. In the present study the primordial germ (PG) cell-derived permanent embryonic germ (EG) cell line EG-1 was used as in vitro model in toxicity studies with chemical mutagens. EG-1 cells and embryonic stem cells of line D3 were comparatively investigated for their cell survival in response to N-ethyl-N-nitrosourea (ENU), N-methyl-N-nitro-N-nitrosoguanidine (MNNG) and mitomycin C (MMC) and the results compared with those obtained for undifferentiated embryonic carcinoma cells of line P19 and differentiated epithelioid EPI-7 cells. As a prerequisite for in vitro toxicity and viability studies the cultivation conditions for EG-1 and D3 cells in the absence of a feeder layer were improved by a conditioned medium, increasing the plating efficiency from 0.08% to 17.5% and from 21.1% to 25.1% for EG-1 and D3 cells, respectively. The resulting mean generation time (MGT) of 16.9 hr for EG-1 cells was identical to the generation time of PG cells in vivo, and was not significantly different from the MGT of D3 (15.6 hr) and EPI-7 (13.7 hr) cells, but significantly longer than the MGT of P19 cells (9.3 hr). Calculations of the concentrations resulting in vitro in a 50% decrease in cell survival demonstrated that EG-1 cells were more sensitive to the toxic effects of ENU, MNNG and MMC than D3 and P19 cells and, with the exception of MNNG, also more sensitive than EPI-7 cells. It is proposed that EG cells are used as a model system to screen for toxic effects of teratogenic and embryotoxic chemical agents in vitro.

Pluripotent cell lines derived from common marmoset (Callithrix jacchus) blastocysts.

We report the derivation of eight pluripotent cell lines from common marmoset (Callithrix jacchus) blastocysts. These cell lines are positive for a series of markers (alkaline phosphatase, SSEA-3, SSEA-4, TRA-1-60, and TRA-1-81) that characterize undifferentiated human embryonal carcinoma cells and rhesus embryonic stem cells. All eight cell lines had a modal chromosome number of 46; seven cell lines were XX and one was XY. Two cell lines (Cj11 and Cj62) were cultured continuously for over a year and remained undifferentiated and euploid. In the absence of fibroblast feeder layers, these cell lines differentiated to multiple cell types, even in the presence of leukemia inhibiting factor. Differentiated cells secreted bioactive CG into the culture medium and expressed alpha-CG, beta-CG, and alpha-fetoprotein mRNA, indicating trophoblast and endoderm differentiation. Bioactive CG secretion in differentiating cells was increased substantially in the presence of GnRH agonist D-Trp6-Pro9-NHEt. When grown at high densities, these cells formed embryoid bodies with a close resemblance to early postimplantation embryos, including the formation of a yolk sac, amnion, and an embryonic disc with an early primitive streak. These results make these pluripotent cells strong candidates for marmoset embryonic stem cells.

Low mutagenic effects of mitomycin C in undifferentiated embryonic P19 cells are correlated with efficient cell cycle control

Pluripotent undifferentiated embryonic carcinoma cells of line P19 and their differentiated progeny, epithelioid ectoderm-like EPI-7 cells, showed different responses to mitomycin C (MMC) with respect to induction of micronuclei, mutations at the HPRT-locus and cell cycle control. Cytotoxic effects of MMC after a 5-h treatment were lower in undifferentiated P19 cells than in differentiated EPI-7 cells with IC 50 values of 1.3 and 0.25 μM for P19 and EPI-7 cells, respectively. MMC did not induce 6-thioguanine-resistant mutants in P19 cells but significantly increased the mutation frequency in EPI-7 cells with concentrations of 0.25, 0.5 and 1.0 μM MMC. Micronuclei determined by flow-cytometry were induced by MMC in both cell lines at equitoxic concentrations of 4.5 (P19) and 0.75 (EPI-7) μM, reducing the viability in both cell lines to 10%. Whereas the induction of micronuclei in P19 cells was maximal 28 h after treatment and declined thereafter, micronucleus induction peaked 48 h post treatment in EPI-7 cells and remained significantly increased even 67 h after the treatment. Flow-cytometric determination of the distribution of MMC-treated P19 and EPI-7 within the cell cycle revealed a distinct G 2 M -block in P19 cells, whereas EPI-7 cells showed normal progression through S-phase and a negligible G 2 M -block . Therefore, we conclude that the lower effectivity of MMC to induce gene mutations and micronuclei in P19 cells seemed to be correlated with a more efficient cell cycle control in undifferentiated compared to differentiated EPI-7 cells.

Transcriptional activation by Oct-3: evidence for a specific role of the POU-specific domain in mediating functional interaction with Oct-1

Oct-3, a member of the POU family of transcription factors, is expressed in pluripotent cells of early mammalian embryos and in undifferentiated embryonal carcinoma cell lines. Using a variety of Oct-3 mutants, we have identified two different domains of Oct-3 which activate transcription in transfected mammalian cells. One of these domains, located in the C-terminal part of the protein, plays a major role in transcriptional activation when Oct-3 is bound to its cognate site, the octamer motif. An Oct-3 mutant containing a single amino acid substitution in the POU homeodomain is unable to bind the octamer target in vitro, yet is still able to activate transcription in an octamer-dependent manner. We provide evidence that transactivation by this mutant involves protein-protein interactions with the ubiquitous octamer binding factor Oct-1. This interaction requires the POU-specific domain of Oct-3 and allows recruitment of Oct-3 to the target promoter even in the absence of Oct-3 DNA binding.

An upstream region of the H2AZ gene promoter modulates promoter activity in different cell types

Human H2AZ gene promoter fragments that included sequences upstream from the core promoter resulted in decreased activity of reporter constructs transfected into several human cell lines, but increased activity in the undifferentiated human embryonal carcinoma cell line Tera-2. Differentiation of Tera-2 cells in media containing retinoic acid restored the ability of the upstream region to downregulate H2AZ gene promoter activity. Levels of endogenous H2AZ mRNA were also found to be 2.5-fold higher in undifferentiated Tera-2 cells than in differentiated Tera-2 cells. A 128 bp region located 234 to 361 bp upstream from the transcription start site of the H2AZ gene was found to be responsible for the modulation of reporter activity. The upstream region also functioned similarly when removed from the H2AZ gene promoter and inserted upstream of the SV40 promoter in reporter constructs. Gel mobility shift studies of fragments of this region revealed two sequence elements, CTCCTCC and CACGTG, that bound nuclear factors in vitro.

Human JC Virus cAMP Response Elements Functional for Enhanced Glial Cell Expression in Differentiating Embryonal Carcinoma Cells

Human JC virus (JCV) is glial cell-specific for growth and expression. This specificity is attributed to thecis-acting sequences of the two 98-bp tandem repeats in the JCV regulatory region. JCV causes brain lesions, especially in AIDS patients. To study the expression of JCV in glial cells, the role of both repeat region TGAGCTCA sequences, which are homologous to the classical TGAGCTCA cAMP response element (CRE), was examined. The effect of the CRE on expression of the JCV early promoter (JCVE) in response to cAMP was studied with undifferentiated, glial and muscle P19 embryonal carcinoma cells. The results showed a threefold increase in response to cAMP only in the glial cells in which JCV is efficiently expressed. The directin vivorole of the JCV CRE was confirmed by site-directed mutagenesis. Additionally, a CRE oligonucleotide was induced by cAMPin vivo,and inin vitrotranscription assays with glial cell extracts. The early promoter of human BK virus containing nonhomologous CRE sequences was previously shown not to be glial cell-specific and failed to respond to cAMP in glial P19 cells in this study. Mobility shift assays showed the cAMP-inducedin vitrointeraction of glial cell protein(s) with the CRE oligonucleotide. Southwestern blot and uv crosslinking experiments identified an approximately 43-kDa protein interacting with the JCV CRE oligonucleotide. The results indicate that thein vivoexpression of JCVEis specifically increased in response to cAMP only in glial cells and JCV CREin vitroprotein complexes are only detected in response to cAMP for glial cell extracts.

Genetic Mapping of a Gene Encoding an Atypical Protein Kinase C, Protein Kinase C Lambda, to the Proximal Region of Mouse Chromosome 3

Protein kinase C are a class of proteins that mediate the control of cellular responses to external stimuli through the phosphorylation of target proteins. These proteins show a molecular heterogeneity by which it is possible to subdivide them into three subclasses (conventional, novel, and atypical) on the basis of their distinct structural and biochemical properties. We have recently described the isolation of a human gene encoding a new member of the atypical class of protein kinase C, protein kinase C{sub l} (PKC{sub l}), which we showed mapped to Xq21.3, close to the BTK gene. The atypical class of protein kinase C contains at least three members, protein kinase C{lambda}, protein kinase C{zeta}, and the previously mentioned protein kinase C{sub l}, although further members have been predicted to exist on the basis of genomic Southern blotting. To clone the mouse genes for new members of the atypical class of protein kinase C, we screened a mouse 11.5-day embryonic cDNA library with a 2.2-kb cDNA representing the entire human PKC{sub l} gene. Eight cDNA clones were isolated, of which six were either partially or fully sequenced. Their sizes ranged from 2.7 to 2.9 kb and appeared to be overlapping fragments of themore » same transcript. A FASTA search of the GenBank database using 1.5 kb of consensus cDNA sequence, containing mainly 3{prime} untranslated sequence, revealed 99% identity at the nucleotide level with mouse protein kinase C{lambda} (Accession No. D28577). Protein kinase C{lambda} (PKC{lambda}) has been previously characterized by Akimoto et al., but the gene encoding it had not been mapped in mouse or human. Mouse PKC{lambda} is expressed abundantly in undifferentiated P19 mouse embryonal carcinoma cells and shows a decrease in expression with differentiation. Gene expression in response to retinoic acid in novel human chromosome 21 monochromosomal cell hybrids

To access a wide a variety of expressed sequence from human chromosome 21 we have placed this chromosome into undifferentiated P19 mouse embryonic carcinoma cells. Cell lines resulting from these experiments have a range of morphologies and a wide variety of karyotypes. We have studied the retinoic acid response of five cell lines, compared to P19 cells, by observing three markers of retinoic acid induced P19 differentiation--cell morphology, RAR alpha and Wnt1 transcription. We see an 'early' retinoic acid response effect, however this response breaks down by the time the 'late' gene. Wnt1 would be transcribed in P19 cells. A highly responsive cell line will be useful for cloning expressed sequences from human chromosome 21 which are produced by early genes in retinoic acid inducible pathways, such as those involved in neurogenesis.

Effects of taxol on the polymerization and posttranslational modification of class III beta-tubulin in P19 embryonal carcinoma cells.

Undifferentiated P19 embryonal carcinoma cells and P19 cells induced to differentiate along a neuronal pathway by 10(-6) M retinoic acid were treated with taxol to examine the effects of this microtubule-stabilizing drug on the subcellular sorting of class III beta-tubulin and on neurite outgrowth. P19 cells were grown on cover slips and then treated with taxol at concentrations of 10(-6) to 10(-9) M for 24 h. The microtubule cytoskeleton was examined after double-immunofluorescence labelling with a monoclonal antibody to alpha-tubulin (YOL 1/34) and a monoclonal neuron-specific class III beta-tubulin antibody (TuJ1). Treatment of undifferentiated P19 cells with concentrations of taxol greater than 4 x 10(-8) M caused microtubule bundling and multiple aster formation and promoted polymerization of the low levels of class III beta-tubulin found in these cells. In neurons, at 2 x 10(-8) M taxol, bundling of microtubules at the base of the neurite was apparent. At taxol concentrations greater than 1 x 10(-7) M, enhanced assembly of class III beta-tubulin was apparent, although long neurites were not observed. Using isoelectric focusing followed by western blotting, we detected an additional isoform of class III beta-tubulin after treatment with 10(-6) M taxol. The results indicate taxol treatment alters the normal subcellular sorting of tubulin isotypes, promotes the polymerization and posttranslational modification of class III beta-tubulin, and interferes with neurite outgrowth.

Negative regulation of a special, double AP-1 consensus element in the vimentin promoter: interference by the retinoic acid receptor.

The growth-regulated vimentin gene contains a functional double AP-1 binding site formed by two nearly perfect inverted repeats. We present evidence for down-regulation of vimentin expression by the retinoic acid receptor (RAR) in two mesodermally derived cell types. By mutation analysis we prove that the double consensus element is responsible for this negative regulation. From in vitro protein-DNA interaction studies we conclude that AP-1 binding is inhibited at RAR amounts required for occupation of the cognate RAR binding site in nuclear extracts from 3T3 cells and differentiated embryonal carcinoma cells. Furthermore, we show that, unlike in other cases, trans-activation of the vimentin AP-1 enhancer element can occur in undifferentiated embryonal carcinoma cells, despite the low amount of Jun and Fos proteins present in these cells. Here, however, down-regulation by retinoic acid cannot be detected.

IPEB transcription factor regulating the intracisternal a particle gene during F9 cell differentiation is expressed at sites of lymphoid development

The murine intracisternal A particle (IAP) proviral elements are expressed at low levels in undifferentiated F9 embryonal carcinoma cells but are highly expressed when F9 cells are induced to differentiate into parietal endoderm-like cells. IAP elements are also expressed in parietal endoderm-like PYS-2 cells. We previously identified an IAP proximal enhancer (IPE) element that mediates a F9 differentiation-specific enhancer activity. We also identified a 60 kDa IPE binding (IPEB) protein whose activity is high in PYS-2 cells, where IAP is expressed, but very low in F9 cells. Transcription of IAP elements has also been shown in the adult mouse thymus and in activated splenic B cells. We have now shown by DNA affinity chromatography, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and band-shift analysis that the 60 kDa IPEB is expressed in adult T lymphocytes and in resting as well as lipopolysaccharide activated splenic B cells but not in adult liver cells, suggesting an important role for IPEB in IAP transcription in vivo. In addition, we find IPEB expressed in the fetal mouse at sites of lymphoid development, such as the liver, spleen, and thymus, suggesting it may play an important role in gene expression during lymphoid development. In support of this, we find IPEB in the human T cell tumor lines, Jurkat and Molt 13, as well as the Daudi B cell line and in the normal calf thymus and in the thymus and spleen of the chicken and rat.

Interaction between a novel F9-specific factor and octamer-binding proteins is required for cell-type-restricted activity of the fibroblast growth factor 4 enhancer.

Understanding how diverse transcription patterns are achieved through common factor binding elements is a fundamental question that underlies much of developmental and cellular biology. One example is provided by the fibroblast growth factor 4 (FGF-4) gene, whose expression is restricted to specific embryonic tissues during development and to undifferentiated embryonal carcinoma cells in tissue culture. Analysis of the cis- and trans-acting elements required for the activity of the previously identified FGF-4 enhancer in F9 embryonal carcinoma cells showed that enhancer function depends on sequences that bind Sp1 and ubiquitous as well as F9-specific octamer-binding proteins. However, sequences immediately upstream of the octamer motif, which conform to a binding site for the high-mobility group (HMG) domain factor family, were also critical to enhancer function. We have identified a novel F9-specific factor, Fx, which specifically recognizes this motif. Fx formed complexes with either Oct-1 or Oct-3 in a template-dependent manner. The ability of different enhancer variants to form the Oct-Fx complexes correlated with enhancer activity, indicating that these complexes play an essential role in transcriptional activation of the FGF-4 gene. Thus, while FGF-4 enhancer function is octamer site dependent, its developmentally restricted activity is determined by the interaction of octamer-binding proteins with the tissue-specific factor Fx.

Replication of a Moloney Murine Leukemia Virus Mutant Lacking the ELP Binding Site

A mutant Moloney murine leukemia virus was created in which the binding site for the embryonal long terminal repeat binding protein had been replaced with a synthetic linker sequence. The virus was found to replicate normally in murine 3T3 fibroblasts, and the mutation was stably transmitted throughout repeated passages. The binding site is therefore dispensable with respect to viral replication. Titers of the mutant virus were comparable to those of wild-type preparations, even when supernatants were prepared from undifferentiated P19 embryonal carcinoma cells Elimination of the repressor binding site therefore had little effect on viral replication in embryohal cells, an unexpected result in light of the reported activity of the binding protein as a transcriptional repressor.

A survey of genes expressed in undifferentiated mouse embryonal carcinoma F9 cells: characterization of low-abundance mRNAs.

As a first step to catalogue mRNAs present in mouse embryonal carcinoma F9 cells, 879 clones corresponding to low-abundance mRNAs were selected from among 2,896 randomly picked up clones of undifferentiated F9 cDNA libraries, using DNA probes complementary to poly(A)+RNAs prepared from undifferentiated F9 cells and to ones prepared from mouse fibroblast L cells. Five-hundred and eighty-two of the 879 clones were partially sequenced, and the subsequent homology search revealed that 201 corresponded to 180 known genes or known DNA sequences, which include not only housekeeping genes but also various tissue-specific genes. Interestingly, at least 24 of the 180 genes are development-related genes in mammals. Among these 24, those for midkine (growth and/or differentiation factor) and interferon-beta are reportedly up-regulated, and those for ECA39 (target for c-Myc regulation), REX-1 (zinc finger protein), and OCT-3 (POU-domain transcription factor) are down-regulated during the development of mouse embryonal carcinoma cells. Thirty-seven of the 582 clones matched the 36 previously reported unidentified ESTs (expressed sequence tags) and the remaining 344 corresponded to 329 novel ESTs. Therefore, partial sequencing of F9 cDNA clones corresponding to low-abundance mRNAs in F9 cells not only provides valuable information concerning development-related genes in mammals, but also many novel ESTs useful for studying mammalian genomes.

Lower mutation frequencies are induced by ENU in undifferentiated embryonic cells than in differentiated cells of the mouse in vitro

The pluripotent embryonic carcinoma cells of line P19 established from undifferentiated cells of the early mouse embryo and their differentiated progeny, the epithelioid ectoderm-like EPI-7 cells, were investigated for the induction of mutations at the HPRT locus by the alkylating agent N-ethyl- N-nitrosourea (ENU). We showed that the cytotoxic effects of ENU after a 5-h treatment were lower in undifferentiated P19 cells than in differentiated EPI-7 cells. The IC 50 values of ENU in the two cell lines amounted to 0.6 mg/ml and 0.09 mg/ml for P19 and EPI-7 cells, respectively. The induction of 6-thioguanine-resistant mutants by ENU (1.0 mg/ml) determined after an expression time of 8 days for both cell lines resulted in similar mutation frequencies. Using expression times of 8 days for P19 and 11.75 days for EPI-7 cells, taking into account the longer generation time of differentiated EPI-7 cells (13.7 ± 3.6 h) in comparison to undifferentiated P19 cells (9.3 ± 0.9h), ENU induced significantly higher mutant frequencies in EPI-7 cells (4865 mutants/10 6 cells) than in P19 cells (282 mutants/10 6 cells). Our results and data from the literature on UV irradiation-induced repair support the idea that the induction of lower mutation frequencies in embryonic cells may correlate with different proliferation capacities, cell cycle parameters and/or different mechanisms of DNA repair in embryonic stem cells and differentiated cells, respectively.

Identification and Cloning of an mRNA Coding for a Germ Cell-Specific A-Type Lamin in Mice

The nuclear lamins are karyoskeletal proteins which have important functions, such as maintaining nuclear envelope integrity and organizing high order nuclear structure during mitosis in higher eukaryotes. In somatic mammalian cells, the A-type and B-type lamins, composed of lamins A and C and lamins B1 and B2, are major components of the nuclear lamina. However, A-type lamins have as yet not been identified in germ cells and undifferentiated embryonic cells. Here we report the cloning of a new 52-kDa A-type lamin from mouse pachytene spermatocytes, termed lamin C2 because of its similarities with lamin C. It has a sequence identical to that of lamin C except that the N -terminal segment, containing the head and the α-helical coil 1A domains, is replaced with a short non-α-helical stretch of amino acids. In mice, lamin C2 was found to be specifically expressed in germ cells. This specific expression and unique structure suggests a role for lamin C2 in determining the organization of nuclear and chromosomal structures during spermatogenesis.
