Spectrophotometric Method Development and Validation for the Estimation of Trelagliptin Succinate in Bulk and Pharmaceutical Dosage Form

A Simple, precise and sensitive ultraviolet spectrophotometric method was developed for the estimation of Trelagliptin succinate in bulk and pharmaceutical dosage form, method was run through LABINDIA UV-Visible spectrophotometer-3200 with solvent of acetonitrile was used. In this method working wavelength was selected at 276 nm. The developed method was validated as per the international conference on harmonization (ICH Q2(R1)) guidelines. The drug was obeying Beer-Lambert’s law revealed good correlation in the concentration range of 10 - 60 µg/ml with correlation coefficient (r2) of 0.9993. The validity of the proposed method was assessed by applying the standard addition technique where the percentage recovery of the added standard was found to be 99.97 % for Trelagliptin succinate. The absorbance was found to be 0.0365 - 0.0506 with %RSD for both inter-day precision & intraday precision. The limit of detection and quantification were calculated and was found to be 0.0563 µg/mL and 0.1689 µg/mL respectively. The developed method was successfully applied to the determination of Trelagliptin in commercially available dosage form with percentage assay of 98.52. The proposed method is recommended for routine analysis of Trelagliptin in bulk and dosage forms in quality control testing laboratories. Since it is rapid, simple, accurate and sensitive.

Development, Validation, Identification and Estimation of Quinoline Yellow in the Levodopa and Carbidopa Pharmaceutical Combined dosage form by Ultra violet Visible Spectrophotometer

A simple, cost-effective, précised, accurate and robust Ultra Violet spectrophotometric method [1, 2 ] has been developed for the Identification and estimation of Quinoline Yellow in the Levodopa and Carbidopa tablet dosage forms. It is used as a colourant in the pharmaceutical tablet’s dosage form. UV scan of Quinoline yellow was taken from the entire UV range of 200 to 800 nm. From the spectrum, maximum absorption was observed at 414nm (λmax) for the Quinoline Yellow. Method was found to be specific with no interference due to blank and placebo, The method was linear with a correlation coefficient of more than 0.999, and Accuracy was in the range of 98.3 to 101.3, In the robustness study employed for standard and sample preparation showed no impact on the results, by deliberate changes proves method is robust and can be utilized for regular analysis. Method validation was performed with reference to ICH guidelines Q2R1.

Physical and Chemical Scrutiny of Obizi River in Awka South Local Government Area of Anambra State, Nigeria for Domestic Consumption

The Obizi River in the Awka South Local Government Area of Anambra State was examined physically and chemically to evaluate its contamination level and suitability for home use. It is important for recreational, fishing, cooking, drinking, and other applications. Its typical applications for drinking, cooking, fishing, recreational activities and other uses are limited since it flows through a canal that might be contaminated by industrial, agricultural, and other human activities. Nitrate mg/l, nitrite mg/l, magnesium mg/l, zinc mg/l, total dissolved solid (TDS) mg/l, hardness mg/l, sulphate mg/l, phosphate mg/l, alkalinity mg/l, acidity mg/l, sodium mg/l, biological oxygen demand (BOD) mg/l, chemical oxygen demand (COD) mg/l, total suspended solid (TSS) mg/l, TS mg/l, oxygen demand (OD) mg/l, and potassium mg/l are among the parameters that were evaluated. Total dissolved solids (TSS) were calculated as the difference between total solids and total dissolved solids. The zinc and nitrate elements within the samples were measured using the ultraviolet (UV) VIS spectrophotometric method. Sulphate was determined turbidimetrically by the absorption spectrophotometry. Magnesium content, total hardness, and alkalinity were measured through titration. TDS (mg/L) was measured with a Multi-Meter (HI 991300, Hanna Equipments, Romania), potassium and sodium ions were assessed by flame photometric technique, chemical oxygen demand (COD) was identified photometrically using the SpectroQuant Nova 60 COD cell test (Merck) in the range of 10 – 150 mg/L. Biological oxygen demand (BOD) was determined using the OxiDirect BOD system and total suspended solids (TSS) was calculated as the difference between total solids and total dissolved solids. Phosphates were measured using the ascorbic acid method, total solids were estimated gravimetrically, and oxygen demand (OD) was measured on-site using a dissolved oxygen meter JENWAY-3405 (Manufacturer: Barloworld Scientific Ltd., England). To find out if the results were significant or not, the parameters were subjected to an ANOVA single factor analysis. The analysis's conclusion was that the data were statistically not significant. Since nitrate and OD were above the World Health Organization's (WHO) limit while most other physicochemical parameters were below it, the ANOVA result showed that there was no significant difference between the physicochemical characteristics of the water samples. With the exception of nitrite and OD, which are above the WHO limit for domestic drinking water and other purposes, it was discovered that the majority of the physicochemical parameters fell within the organization's water quality standards for these purposes. As a result, the water quality is not good and should not be drunk unless treated.

DETERMINATION AND ESTIMATION OF PRESERVATIVES IN PACKAGED FOOD

Objectives: Packaged food and beverages are consumed all over the world for their nutritional value, durability, their quenching properties, stimulating effect, and for their medicinal value. Food additives are substances that are added to food or animal feed during processing or storage. They include antioxidants, preservatives, coloring and flavoring agents, and anti-infective agents. This study focuses on the determination of the content levels of sodium benzoate in different samples of commercially available food products in the local market. Methods: Food products selected are Sting Energy Drink, Listerine Cool Mint Mouthwash, Kissan Fresh Tomato Ketchup, and Dabur Hommade Imli Sauce. Identification tests for the presence of benzoic and its salts in food products were conducted. A standard graph for benzoic acid at different concentrations is drawn. The concentrations of benzoic acid in selected food products were determined and evaluated by rapid and simple ultraviolet spectrophotometric methods. Results: The results obtained from this study indicated that the quantity of benzoic acid in analyzed food samples was in the range of 18.3– 160.5 μg/mL. Conclusion: The optimum benzoic acid concentration to be in marketed food preservatives is 20–700 μg/mL. As the benzoic concentration in selected food products determined were in range, we found that all the selected samples are safe to use.

Simultaneous Assessment of the Nitrates and Nitrites in the Ground Water Samples using Optical Spectrophotometry and Validating the Appropriate Wavelength of Measurement

Background: Nitrate is a renowned pollutant in water and soil. Nitrate is used mostly as a nitrogen fertilizer. When nitrate seeps into the ground it can get into drinking water. Traces of nitrite and nitrate in drinking water may lead to methemoglobinemia in infants and with long term exposure is a possible cancer risk. Concentration of nitrate above 45 mg/L as nitrate and 10 mg/L as nitrate-nitrogen is unsuitable for domestic use according to BIS (Bureau of Indian Standards). Methods: Nitrates are mainly determined by the spectrometric, colorimetric and ion chromatography methods. Majority of these methods are slow measurement, laborious and require the use of toxic and expensive reagents. In this study, ultraviolet spectrophotometric method is utilized for detection of nitrate optically with no chemical reagents. This method is simple and cost-effective. Nitrate and nitrite concentrations were analysed at various wavelengths (220 to 370 nm) and 240 nm was found to be efficient for detecting both nitrate and nitrite. Result: Various groundwater samples in and around Karunya Campus and nearby residential places, Coimbatore, Tamil Nadu were analysed and compared with one borewell water sample from West Bengal State which was prone to contaminated ground water. The results show that all the ground water samples have nitrate and nitrite concentration within the permissible limit but the sample from West Bengal has higher concentration compared to Coimbatore samples.

UV Spectrophotometric Quantification of Sudan-IV Dye in Palm Oil from Major Markets of Benin Metropolis

The deficit in the supply of palm oil in Nigeria over the years has been met in part through smuggling of the product from different sources into the Nigerian market by marketers of which some of the products have been reported to be adulterated with azo dyes. The aim of this study was to detect and quantify sudan dyes in adulterated palm oil from the open market using simple and inexpensive procedures such as analytical thin layer chromatography (TLC) and Ultra violet spectrophotometric methods respectively. Seven palm oil samples from the open markets within Benin metropolis labelled as (AUC, AUC2, IK1, IK5, OLK1, ADIT, and OB) were screened for the presence of Sudan-III and Sudan-IV dyes. Four of the samples (IK1, IK5, OLK1, ADIT) were confirmed to contain Sudan-IV dye with quantities of 22.5mg/L, 21.7mg/L, 29.5mg/L, 23.8mg/L respectively. This experiment shows that the use of easy and cheap methods such as TLC and UV-spectrophotometry can be used to detect and quantify Sudan dyes in adulterated palm oil.

Estimation of Paracetamol in various brands of Paracetamol Tablets and their Comparative Study

Paracetamol is a widely used non-prescription medication and is available in several brands in the market which makes it difficult to select the safe and effective one. Therefore, the study aims to establish the pharmaceutical equivalence of the different brands of paracetamol tablets and to justify the quality of different brands available in the market. The study involves the quantitative analysis of ten different brands of Paracetamol 500 mg tablets, using Ultra Violet Spectrophotometric methods, in which the samples were dissolved in 0.1M sodium hydroxide solution and distilled water and their various absorbances determined at the wavelength (λmax) of 257nm. Quantitative estimation of Paracetamol carried out by UV-Visible spectrophotometric methods, by using standard absorptivity value (assay method given in Indian Pharmacopoeia) and calibration curve method. The percentage content for each sample was calculated byboth methods using appropriate formulae and also determined whether or not they comply with standard specifications as per IP. The percentage content of paracetamol in different brands of paracetamol tablets was compared by IP Method. The results obtained by the IP method and Calibration curve method were also compared.

HSI combined with CNN model detection of heavy metal Cu stress levels in apple rootstocks

In recent years, the prolonged application of copper-based fungicides, ripening agents, and fertilizers in agricultural activities has resulted in copper (Cu) levels in the soil exceeding safe levels. Excess Cu has a negative impact on plant growth and crop yields. The practice of grafting horticultural crops onto appropriate rootstocks is frequently employed as a preventive measure against copper accumulation and its associated toxicity. Therefore, the selection of high-quality rootstocks that are not contaminated with heavy metal Cu is important for the growth of grafted crops and the breeding of grafted crops. This study proposed a technique based on convolutional neural network (CNN) and hyperspectral imaging (HSI) for the rapid, non-destructive, and accurate identification of copper contamination levels in apple rootstocks. The effect of different heavy metal Cu contamination levels on leaves was obtained by measuring chlorophyll content in leaves using the ultraviolet spectrophotometer method. A hybrid strategy for spectral variable selection was utilized, involving competitive adaptive reweighted sampling and successive projection algorithms. A traditional partial least squares discriminant analysis linear classification model, and least squares support vector machine, random forest, and extreme learning machine nonlinear models were developed. The results showed that the accuracy of the CNN model and Macro-F1 were 99.6% and 99.2%, respectively, which were better than the traditional linear and nonlinear models. Principal component analysis and t-distributed stochastic neighbor embedding were used for visual analysis to investigate the clustering patterns among sample classes. The results indicated that the combination of CNN and HSI techniques has great potential for classifying copper contamination levels in apple rootstocks.

Development and validation of a stability indicating green analytical method for the simultaneous estimation of l-glutathione, n-acetyl l-cysteine and Vitamin C in marketed formulation using UV–visible spectroscopy

BackgroundAn efficient ultraviolet spectrophotometric method has been devised for the simultaneous analysis of l-glutathione, n-acetyl l-cysteine and Vitamin C in combined tablet dosage forms. This method is characterized by its simplicity, rapidity, precision, accuracy and cost-effectiveness. It employs the simultaneous equation method to determine the concentrations of l-glutathione, n-acetyl l-cysteine and Vitamin C.ResultsThe concentration range of 50–250 μg/mL for l-glutathione, 15–75 μg/mL for n-acetyl l-cysteine and 2–10 μg/mL for Vitamin C demonstrated linearity in this study. The simultaneous equation method was employed to determine the drug concentrations. The average recovery rates were found to be 99.94 ± 0.61% for l-glutathione, 100.90 ± 0.96 for n-acetyl l-cysteine and 99.81 ± 0.58% for Vitamin C. Stability studies were performed under various stress conditions.ConclusionThe method employed in this study was observed to possess qualities of simplicity, accuracy and precision, making it suitable for the simultaneous determination of l-glutathione, n-acetyl l-cysteine and Vitamin C in pharmaceutical tablet dosage forms. The analysis results were statistically validated and supported by recovery studies and stability studies, further enhancing the credibility of the method.

Cadmium ion-imprinted polymers for adsorption and detection of cadmium ions

Cadmium is a typical heavy metal ion. Long-term exposure to cadmium can lead to diseases like renal tubular dysfunction and bone degeneration. Therefore, it is necessary to develop efficient separation methods and detection techniques for cadmium ions. In this paper, adsorption enrichment was combined with detection to rapidly detect cadmium ions in water. Cadmium ion imprinted polymers (Cd(II)-IIPs) were prepared by bulk polymerization using Cd(II) as template ion, itaconic acid (IA) as functional monomer, ethylene dimethacrylate (EGDMA) as cross-linker, 4-(2-pyridylazo)-resorcinol (PAR) as chromogenic ligand, and azobisisobutyronitrile (AIBN) as initiator. The preparation conditions were optimized and the prepared Cd(II)-IIPs were characterized by Fourier transform infrared spectroscopy (FT-IR), solid-state 13C nuclear magnetic resonance (13C NMR), scanning electron microscopy (SEM), Brunauer-Emmett-Teller (BET) and X-ray photoelectron spectroscopy (XPS). In the adsorption experiment, the adsorption equilibrium was reached within 10 min in a solution of pH 7.0, and the saturation adsorption capacity of IIPs and NIPs were 50.542 mg·g−1 and 33.762 mg·g−1, respectively. The Cd(II)–IIPs exhibited selectivity for Cd(II) ions in the presence of Ni(II), Fe(II), Co(II), Cu(II), and Pb(II) ions; and after eight adsorption–desorption cycles, the adsorption capacity of Cd(II)-IIPs retained > 90%. In addition, the detection performance of the Cd(II)-IIPs was investigated by digital image colorimetric (DIC) and ultraviolet spectrophotometric methods; the detection limits were 1.46 and 1.74 μg·L−1, respectively, and the quantitative detection range was 8–100 μg·L−1. The Cd(II)-IIPs exhibited excellent adsorption properties and an obvious color gradient variation, which could be used for quantitative colorimetric detection over a range of concentrations.

Quantitative analysis of favipiravir in bulk and pharmaceutical dosage forms using UV visible spectrophotometer

The main aim of this study is to develop a rapid and cheap Ultraviolet (UV) spectrophotometric method for the estimation of Favipiravir (400mg) in tablets and validate as per International Conference on Harmonization (ICH) guidelines. The developed method uses the solvent Ethanol: Water (30:70) for the assay of Favipiravir whose λmax was found to be 231nm. In this method, Favipiravirshows linearity in the range 2-10 μg/ml. The accuracy was carried out by using standard addition method and the precision is ascertained by calculating the relative standard deviation. A cheap and rapid UV spectrophotometric method was developed and validated for the estimation of Favipiravir and hence it can be used for the routine pharmaceutical analysis.

Development and validation of ultraviolet spectrophotometric method for Saquinavir estimation in bulk and dosage form

We developed and validated a simple, rapid, sensitive, precise, and reproducible specific UV spectrophotometric technique for determining Saquinavir in bulk medication and pharmaceutical dosage form. Methods: As per ICH recommendations, a simple double-beam UV spectrophotometric technique has been designed and validated with several criteria, such as linearity, precision, repeatability, the limit of detection (LOD), limit of quantification (LOQ), and accuracy. Results: UV-visible spectrophotometric approach, measurement of absorption at the maximum wavelength in 10 mL methanol, and volume made with water solvent system as reference Saquinavir was discovered at 239 nm. The medicine followed Beer's rule and showed a good correlation. Beer's law was followed for Saquinavir in the 1-5µg/ml concentration range, with a correlation coefficient of 0.999. Saquinavir's LOD and LOQ were determined to be 0.56 μg/ml and 2.78 μg/ml, respectively. The proposed method is simple, precise, accurate, and reproducible, and it may be utilized for routine analysis of Saquinavir in bulk and tablet dosage form.

Smart eco-friendly mathematically manipulated UV spectroscopic methods to resolve severely overlapped spectra of a binary mixture of dapagliflozin with sitagliptin and vildagliptin

The binary formulations consisting of dapagliflozin with sitagliptin and vildagliptin are prescribed for the management of diabetes. The current study presents novel eco-friendly ultraviolet spectrophotometric methods for concurrent quantification of dapagliflozin with sitagliptin or vildagliptin in their binary formulations. The ratio spectra of series of sitagliptin, vildagliptin, and dapagliflozin were generated using dapagliflozin 7.5 µg/mL, sitagliptin 10 µg/mL, and vildagliptin 10 µg/mL as divisor than the ratio difference and ratio first derivative techniques were established. Also, the parent spectra of analytes were extracted from the mixture spectrum by successive mathematical filtration for quantification of individual analytes from the binary mixture without prior separation. The procedure showed good correlation in the series of 5–100 µg/mL for sitagliptin, 7.5–75 µg/mL for vildagliptin, and 1–15 µg/mL for dapagliflozin with an excellent correlation coefficient and good levels of detection and quantification. For the quantification of dapagliflozin levels in combination with different analytes, a single calibration curve was constructed. To create repeatable methods, the wavelength selection for measuring the peak amplitude and analyte concentration of divisor spectra was carefully optimized. The explored methods showed a low relative error percentage of 0.44–1.52, 0.20–0.94, and 0.02–1.22 and relative standard deviation percentages of 0.47–1.44, 1.16–1.41, and 0.94–1.81 for SGT, VGT, and DAP respectively, which confirmed the trueness and precision of the established processes. The validity of the ultraviolet methods was confirmed by quantifying the laboratory mixtures with varying dapagliflozin, sitagliptin, and vildagliptin ratios in addition to the standard addition technique. Additionally, the method's greenness evaluated by hexagonal and AGREE models and whiteness by the red–greenblue model demonstrated greater environmental sustainability in terms of safety and waste generated. Thus, utilizing environmentally friendly UV spectrophotometric techniques, a binary formulation including dapagliflozin + sitagliptin and dapagliflozin + vildagliptin can be evaluated for routine quality control.

Antimycobacterial Compound of Cynoglossum lanceolatum Forsk.: Bioassay Guided Isolation, Molecular Docking, Synthesis of Analogs, and a Plausible Mechanism of Action.

Tuberculosis (TB) remains a major threat to human health. Due to the prevalence of drug-resistant Mycobacterium tuberculosis (Mtb), it is urgent to discover drugs with new mechanisms of action (MOA) to ensure effectiveness against strains that are resistant to existing TB drugs. Cynoglossum lanceolatum Forsk was used to treat TB in Traditional Chinese Medicine. In this article, shikonin, the anti-Mtb active component, was obtained from the whole herb extract of C. lanceolatum by bioassay-guided isolation. Using the microplate alamar blue assay (MABA), the minimum inhibitory concentration (MIC) of shikonin against Mtb was determined to be 128 μg/mL. In order to obtain a more efficient anti-Mtb molecule, (E)-1-(6-bromo-2,3-dihydrochromen-4-ylidene)thiosemicarbazide was synthesized based on the scaffold of shikonin, which exhibited potent activity against Mtb (MIC=4 μg/mL). These results highlight that both naphthalene-1,4-dione and chroman-4-one are pharmacophores with activities against Mtb. To investigate a plausible mechanism of action, the molecular docking was firstly performed against catalase-peroxidase enzyme (KatG) of Mtb using AutoDock 4 software. The results demonstrated that both shikonin and (E)-1-(6-bromo-2,3-dihydrochromen-4-ylidene)thiosemicarbazide could bind to the active site of Mtb KatG. KatG enzyme activity and intracellular reactive oxygen species (ROS) levels in Mtb cells were then measured by ultraviolet spectrophotometric method and fluorescence microplate reader assay, respectively. The experiments confirmed that above compounds could inhibit the catalytic activity of Mtb KatG, and cause the ROS accumulation in Mtb cells. Therefore, inhibition of KatG may be a novel mechanism of action for these two compounds to fight against Mtb.

Development of spectrophotometric method for simultaneous estimation of dexketoprofen and dicyclomine

Objectives: The objective of this work was to develop a simple, fast, and reliable ultraviolet (UV) spectrophotometric method for simultaneous estimation of dexketoprofen and dicyclomine in tablet dosage forms. Materials and Methods: Dexketoprofen and dicyclomine solutions were prepared separately in the range of 2 µg/mL–10 µg/mL. The solutions were scanned over a range of 200–400 nm. The isoabsorptive point for both drugs was determined. The developed method was evaluated for accuracy, precision, and linearity. In linearity correlation coefficient was found. Results: A simple, fast, and reliable UV spectrophotometric method was developed for quantitative estimation of dexketoprofen and dicyclomine. The precision study showed that the percent relative standard deviation was within the range of acceptable limits, and the percent recovery was found to be in the range of 0.3–1.5 (intraday) and 0.23–1.7 (interday). In the accuracy study, the percent recovery was found to be 97 (50%), 96 (100%), and 95 (150%). Conclusion: The developed spectrophotometric method in this study was simple, accurate, precise, specific, sensitive, and reproducible and can be directly and easily applied to pharmaceutical dosage forms.

Polystyrene is a widely used plastic for household purposes or in industrial packaging. However, polystyrene can be categorized as a potent carcinogenic due to its monomer unit. Polystyrene comprises several styrene units that easily leach as styrene microplastics on contact with hot or fatty material. A ultraviolet (UV) spectrophotometer method has been developed for the determination of polystyrene. Polystyrene in an aqueous solution with tetrahydrofuran was estimated at a wavelength ranging from 200 to 300 nm. The λmax of polystyrene was found to be 261.5 nm. The method was linear for the polystyrene concentration ranging from 20 to 120 µg/mL. The limit of detection was founded to be 1.65 µg/mL, and the limit of quantification was 5 µg/mL. DETERMINING THE TANNIN CONTENT IN PISANG AMBON (Musa × paradisiaca L.) WITH THE POTENTIAL AS ANTHELMINTIC

Tannins result from plants' secondary metabolism, closely associated with plant defense mechanisms against insects. Condensed tannins can disrupt the life cycle of parasitic nematodes starting from eggs, adult worms, and larvae. Currently, the antiparasitic properties of condensed tannins are being investigated as an alternative for controlling parasites. The people use the young Pisang Ambon (Musa × parasidiaca L.) as an anthelmintic. People generally use this part of the banana peel. Based on this, a study was conducted to determine the tannin content in the peel and fruit of Pisang Ambon (Musa × parasidiaca L.). The research was conducted by extracting the fruit and peel of young Pisang Ambon by maceration method using 96% ethanol as solvent. The Harborne method and Thin Layer Chromatography tested the presence of secondary metabolites. The tannin content was determined by the visible, ultraviolet spectrophotometric method. The qualitative results showed that the peel and fruit of Pisang Ambon contained condensed tannins. Meanwhile, the quantitative results showed that the tannin content of the fruit (54.98% w/w) was higher than the peel (14.32 %w/w).

Analysis of different brands of paracetamol tablet used in the market by assay method of UV spectroscopy and its evaluation

The study included analysis of five different brands of Paracetamol tablets used in market, using the ultra violet spectro-photometric method. The percent purity of the five brands of Paracetamol was determined by assay method on uv spectrophotometer. The result obtained was compared with those of standard. The Percent content of standard Paracetamol in IP contain no less than 95.0 percent and not more than 105.0 percent of the prescribed Paracetamol value. It was noted that samples A, B, D and E meet the I.P. specified limit in spectrophotometric method where as sample c was out of the specified limit. Also the evaluation test of paracetamol. Were carried out which complies the i.p. limit.

Fruits are a type of food that contains nutrients, vitamins and minerals which are generally very good for consumption every day. Vitamin C is one of the vitamins contained in fruits. The loss of vitamin C in fruit processing depends on the type and process used, as well as the water-soluble vitamins. The purpose of this study was to determine the effect of heating temperature and heating time on the vitamin C content in melon juice. The method used in this research is using the ultraviolet spectrophotometric method. Green melon juice was treated with heating temperature and heating time with variations in heating temperature (30 ° C, 60 ° C, and 90 ° C) and variations in heating time (15 minutes, 30 minutes, and 45 minutes). The results showed that the orange melon juice and green melon juice by heating decreased the Vitamin C content. Heating of fruit juices often occurs during the sterilization process, so if the vitamin content in the material is to be maintained, sterilization must be carried out in a very short time. Heating temperature and heating time have a significant effect on the content of vitamin C in green and orange melon juice.
