Physical Mechanism of One-Photon Absorption, Two-Photon Absorption, and Electron Circular Dichroism of 1,3,5 Triazine Derivatives Based on Molecular Planarity.

We provide a method to regulate intramolecular charge transfer (ICT) through distorting fragment dipole moments based on molecular planarity and intuitively investigate the physical mechanisms of one-photon absorption (OPA), two-photon absorption (TPA), and electron circular dichroism (ECD) properties of the multichain 1,3,5 triazine derivatives o-Br-TRZ, m-Br-TRZ, and p-Br-TRZ containing three bromobiphenyl units. As the position of the C-Br bond on the branch chain becomes farther away, the molecular planarity is weakened, with the position of charge transfer (CT) on the branch chain of bromobiphenyl changing. The excitation energy of the excited states decreases, which leads to the redshift of the OPA spectrum of 1,3,5-triazine derivatives. The decrease in molecular plane results in a change in the magnitude and direction of the molecular dipole moment on the bromobiphenyl branch chain, which weakens the intramolecular electrostatic interaction of bromobiphenyl branch chain 1,3,5-triazine derivatives and weakens the charge transfer excitation of the second step transition in TPA, leading to an increase in the enhanced absorption cross-section. Furthermore, molecular planarity can also induce and regulate chiral optical activity through changing the direction of the transition magnetic dipole moment. Our visualization method helps to reveal the physical mechanism of TPA cross-sections generated via third-order nonlinear optical materials in photoinduced CT, which is of great significance for the design of large TPA molecules.

Theoretical Studies and Design Strategies of Highly Efficient Two-Photon Excited Fluorescent Probes for Hydrogen Sulfide Detection through Simulation of Excited-State Dynamics.

Hydrogen sulfide (H2S) plays a critical role in numerous physiological and pathological processes, but an abnormal level of H2S in living systems can cause various diseases. To detect the level of endogenous H2S in a complicated biological system, the luminous mechanism of "turn-on" probe for H2S monitoring has been deeply explored through the simulation of excited-state dynamic processes, and the effect of different geometric modifications on optical properties has been minutely investigated based on molecular modeling. TD-DFT calculations demonstrate that line-type π-expanding in the molecular skeleton is beneficial for improving two-photon absorption (TPA) ability, but it can give rise to extremely large geometric relaxation, going against fluorescence emission. It is an effective way to suppress molecular skeleton scissoring vibration by introducing strong electron-withdrawing substituent groups (F, Cl, Br, CN) in benzopyran, and these compounds also have superior TPA properties in NIR. One of the potential materials in the application of biological imaging and H2S detection has been obtained, which simultaneously possesses easily distinguished spectra (with a Stokes shift as large as 77 nm), high luminous efficiency (with a quantum yield up to 20.07%), and large TPA cross section (952 GM at 950 nm).

Computational Investigations into Two-Photon Fibril Imaging Using the DANIR-2c Probe.

The design of novel fibril imaging molecules for medical diagnosis requires the simultaneous optimization of fibril-specific optical properties and binding specificity toward amyloid fibrils. Because of the possibility to monitor internal organs and deep tissues, the two-photon probes that can absorb in the infrared (IR) and near-IR (NIR) region with a significant two-photon absorption cross section are of immense interest. To contribute to this exploration of chemical compounds suitable for two-photon fibril imaging, we have computationally studied the one- and two-photon properties of a donor-acceptor-substituted DANIR-2c probe, which was used for in vivo detection of β-amyloid deposits using fluorescence spectroscopy. In particular, a multiscale computational approach was employed involving molecular docking, molecular dynamics, hybrid QM/MM molecular dynamics, and coupled-cluster/MM to study the binding of the studied probe to amyloid fibril and its one- and two-photon absorption properties in the fibrillar environment. Multiple binding sites are available for this probe in amyloid fibril, and the one corresponding to the largest binding affinity exhibits also the largest and experimentally meaningful two-photon absorption cross section, thus demonstrating the potential of the studied probe in two-photon microscopy.

Photophysical properties of Pt(II) and Pd(II) complexes for two-photon photodynamic therapy: A computational investigation

Two-photon photodynamic therapy (TP-PDT) plays a critical role of in cancer treatment due to its deep penetration into tissue and low photobleaching effect. In this paper, density functional theory (DFT) and time-dependent density functional theory (TDDFT) are used to calculate the one and two-photon absorption properties, spin-orbit coupling constants, triplet excited state lifetimes and type I/II mechanisms of Pt(II) complexes and two designed Pd(II) complexes. The results reveal that all complexes meet the standard of photosensitizer. More importantly, the designed Pd(II) complex 3 possesses both large two-photon absorption cross section up to 134.5 GM and long triplet excited state lifetime of 85.5 μs. It is expected that the inexpensive Pd(II) complex will replace the reported Pt(II) complexes as an effective two-photon absorption photosensitizer, which will provide some guidance for the design and synthesis of novel photosensitizers experimentally.

Theoretical Investigation of Ru(II) Complexes with Long Lifetime and a Large Two-Photon Absorption Cross-Section in Photodynamic Therapy.

Two-photon photodynamic therapy (TP-PDT), as a new method for cancer, has shown unique advantages in tumors. A low two-photon absorption cross-section (δ) in the biologic spectral window and a short triplet state lifetime are the important issues faced by the current photosensitizers (PSs) in TP-PDT. In this paper, the photophysical properties of a series of Ru(II) complexes were studied by density functional theory and time-dependent density functional theory methods. The electronic structure, one- and two-photon absorption properties, type I/II mechanisms, triplet state lifetime, and solvation free energy were calculated. The results showed that the substitution of methoxyls by pyrene groups greatly improved the lifetime of the complex. Furthermore, the addition of acetylenyl groups subtly enhanced δ. Overall, complex 3b possess a large δ(1376 GM), a long lifetime (136 μs), and better solvation free energy. It is hoped that it can provide valuable theoretical guidance for the design and synthesis of efficient two-photon PSs in the experiment.

Physical mechanism on the linear spectrum and nonlinear spectrum in a twist bilayer graphdiyne nanodisk.

The one-photon absorption properties (OPA), two-photon absorption properties (TPA), electronic circular dichroism (ECD) spectra and partial DOS (PDOS) of a twist bilayer graphdiyne nanodisk (TwBLGDY-ND) were investigated by using a variety of quantum chemistry and wave function analyses. The physical mechanism of the twist bilayer graphdiyne nanodisk (TwBLGDY) with optical properties regulated by twisting angles was revealed. The results show that the twist angle makes the TwBLGDY form a moiré superlattice structure, and electron excitation mainly occurs in the first ring of the moiré superlattice structure. The contribution of atomic orbitals in these fragments to transition dipole moments is greater and electronic transitions are more likely to occur. When the twist angle increases from 0° to 15°, the absorption spectrum of the system is red shifted, which is mainly due to the enhancement of electron excitation characteristics. When the twist angle increases from 15° to 27.5°, the absorption spectrum of the system is blue shifted, due to the enhanced charge transfer within the layer. On the other hand, the twist angle can regulate the TPA absorption cross section of the system to enhance the intensity of the absorption spectrum. The twist angle can also regulate chirality by adjusting the spatial distribution of electric dipole transition and magnetic dipole transition. This study can provide theoretical guidance for constructing chiral optical devices based on the TwBLGDY structure.

Strong fluorescence-detected two-photon circular dichroism of chiral gold nanoclusters.

Progress in syntheses and understanding of the intriguing properties of chiral noble metal nanoclusters sparks interest to extend investigations of their chiroptical response to the nonlinear optics regime. We present a quantitative determination of two-photon circular dichroism of chiral gold nanoclusters with ATT and L- or D-Arg ligands (ATT = 6-aza-2-thiotymine and Arg = arginine). Introduction of arginine ligands enables the formation of two enantiomers of the nanoclusters, with strong chiroptical effects in both linear and nonlinear regime. We present two-photon absorption and luminescent properties measured in a wide range of wavelengths, with the two-photon absorption cross section reaching 1743 GM and two-photon brightness ∼1102 GM at 825 nm. We report strong, 245-fold enhancement of the two-photon circular dichroism of nanoclusters with respect to the one-photon absorption counterpart - the dissymmetry factor. The presence of multiple advantages of nanoclusters: high fluorescence quantum yield, strong nonlinear optical properties and well-controlled chirality is a powerful combination for applications of such clusters in multiphoton microscopy.

Photophysical and anion sensing properties of a triphenylamine-dioxaborinine trimeric compound.

Herein, we report the synthesis and photophysical characterization of the novel tris(4-(2,2-difluoro-6-methyl-2H-1λ3,3,2λ4-dioxaborinin-4-yl)phenyl)amine trimeric probe (A2) via the reaction between triphenylamine (1), acetic anhydride, and BF3·OEt2 implying the twelve new bond formation in a one-pot manner. This highly fluorescent compound in solution (φ up to 0.91 at 572 nm) and solid state (φ = 0.24 at 571 nm) showed a better solvatofluorochromism than its analog monomeric A1 due to symmetry-broken charge transfer, which is consistent with high solvent dipolarity (SdP) response in Catalán's multiparametric regression. Notably, A2 had a high sensibility and selectivity for CN- or F- in solution (LODCN-/F- = 0.18/0.70 μM), and CN- can be discriminated from F- by the reaction of A2 with 3.0 equiv. of CN-. In addition, A2 was impregnated on filter paper to prepare test strips that were applied to naked-eye qualitative sensing of CN- or F-. Finally, the octupolar system in A2 allows for better action of two-photon excitation cross-section values when compared with that of the dipolar structure in A1. These findings provide further information for the design of new efficient two-photon absorption dyes.

Centripetal triazine chromophores: towards efficient two-photon absorbers and highly emissive polyimide films

Centripetal chromophores based on the s-triazine core with tuneable properties and excellent two-photon absorption properties were used for the construction of high-quality and well-transparent emissive polyimide films.

Unveiling the molecular structure and two-photon absorption properties relationship of branched oligofluorenes.

Organic molecules have been intensively studied during the last few decades because of their photonics and biological applications. In this material class, the fluorene molecules present outstanding optical features, for example, high values of two-photon absorption (2PA) cross-sections, visible transparency, and high fluorescence quantum yield. Also, it is possible to improve the nonlinear optical response by modifying the fluorene molecular structure. In this context, herein, we have synthesized V and Y-shaped branching oligofluorenes containing two and three fluorene moieties in each branch. Such a molecular strategy may exponentially enhance the nonlinear optical response due to the coherent coupling among the molecular arms. Thus, we combined the use of femtosecond Z-scan spectroscopy and white light transient absorption spectroscopy (TAS) to understand the molecular structure and 2PA property relationship of branching oligofluorenes. The results show that there is a universal relationship between the 2PA cross-section and the effective π-electron number (Neff) given by σ2PA(GM) = (079 ± 0.03)Neff2, which is independent of the molecular shape (linear, V or Y-shaped). Therefore, the intramolecular charge transfer responsible for the cooperative effect among the branches does not occur. This statement is corroborated by the results of the femtosecond TAS technique.

Research and Design of Aggregation-Induced Phosphorescent Materials for Time-Resolved Two-Photon Excited Luminescence Imaging.

Pure organic two-photon excited room temperature phosphorescent (RTP) materials have attracted great attention for time-resolved imaging due to their long emission lifetime and high resolution. The materials with an aromatic carbonyl group exhibit aggregation-induced emission (AIE) and RTP characteristics simultaneously. Here, we deeply explored the nature of aggregation-induced phosphorescence (AIP), especially the relationship between molecular configuration and optical properties. It was found that aggregation effect can suppress geometrical vibrations and regulate energy difference between S1 and T1. The aromatic carbonyl group plays significant roles in changing electronic configuration, resulting in large Stokes shift and spin-orbit coupling. It also leads to small transition dipole moment, decreasing two-photon absorption cross section and radiative decay rate. To improve two-photon absorption properties, we further designed a π-conjugated compound with large two-photon absorption cross section in the biological window (36.40 GM/656 nm) and AIP characteristics, which is a potential material in the application of time-resolved two-photon excited imaging.

Ultrafast Dynamics and Strong Two-Photon Absorption Properties of Nonplanar β-Functionalized “Push–Pull” Copper Corroles with a Mixed Substituent Pattern

A new series of nonplanar and unsymmetrically β-functionalized "push-pull" copper corroles, CuTPC(CHO)R7 [R = H, Br, Ph, Me, or 2-thienyl (Th)], were synthesized and characterized to elucidate the effect of β-functionalization and nonplanarity on the photophysical, redox, and nonlinear optical (NLO) properties on the corrole ring. The synthetic route to unsymmetrically β-octasubstituted copper corroles includes bromination of CuTPC(CHO) to get CuTPC(CHO)Br7 in 80% yield, which was further subjected to the Pd-catalyzed Suzuki reaction. CuTPC(CHO)Br7 exhibited a large red shift in the Soret band (Δλmax = 35-40 nm) and both the Q bands (Δλmax = 10-50 nm), as compared to CuTPC and CuTPC(CHO). CuTPC(CHO)Br7 was 510 and 290 mV anodically shifted in the first oxidation and the first reduction compared to CuTPC owing to the strong -I effect of CHO and Br groups. Density functional theory studies revealed that all the β-octasubstituted copper corroles exhibited highly nonplanar saddle-shape conformation of the corrole ring. Very high torsional saddling was observed for CuTPC(CHO)Th7 (79-83°) than that for CuTPC (49-53°), even larger than that for CuTPCBr8 (67-70°). Femtosecond laser-induced third-order NLO studies from these copper corroles showed strong two-photon absorption cross-sections (0.48-6.98 × 104 GM) and self-focusing-type positive nonlinear refraction behavior. The observed structure-dependent two-photon absorption coefficients (β) are in the range of ∼2.7-20.9 × 10-12 m/W, and the n2 values are in the range of ∼0.64-6.45 × 10-18 m2/W. The present results may facilitate a new window for these copper corroles in nonlinear optical devices, femtosecond optical limiters, and many other ultrafast photonic applications.

Fluorescence lifetime imaging of water-soluble porphyrin in human nasopharyngeal cells under two-photon excitation

A comparative study of the two-photon absorption and emission properties of two free-based porphyrins (H2TMPyP and H2TPPS) and their zinc(II) complexes (ZnTMPyP and ZnTPPS) in three polar solvents has been carried out. The presence of the central zinc(II) ion leads to smaller two-photon absorption cross sections (δTPA), while the introduction of methylpyridinium substituents (N•+–CH3) results in larger ones. The δTPA values of H2TMPyP and ZnTMPyP are affected by both the solvent polarity and the hydrogen bonding between porphyrin and solvent molecules. In this work, the strongest two-photon fluorescence is given by H2TMPyP, which may efficiently get into four kinds of nasopharyngeal cell lines (NP69, SUNE1, CNE2, and CNE1) and prefer to localize in the nucleus. Two-photon fluorescence lifetime images of all the cell lines treated with H2TMPyP consist of a short- (τ1) and a long-lived component (τ2). The τ2 distribution of normal cell line NP69 shows one peak lifetime (∼2.12 ns), while those of the cancerous cell lines (SUNE1, CNE2, and CNE1) have two peak lifetimes (∼2.14 and ∼3.77 ns). The τ1 distribution around 0.68 ns is higher for the well differentiated nasopharyngeal cancerous cell line CNE1 compared with the poorly differentiated ones (SUNE1 and CNE2).

Dual-function artificial molecular motors performing rotation and photoluminescence.

Molecular machines have caused one of the greatest paradigm shifts in chemistry, and by powering artificial mechanical molecular systems and enabling autonomous motion, they are expected to be at the heart of exciting new technologies. One of the biggest challenges that still needs to be addressed is designing the involved molecules to combine different orthogonally controllable functions. Here, we present a prototype of artificial molecular motors exhibiting the dual function of rotary motion and photoluminescence. Both properties are controlled by light of different wavelengths or by exploiting motors' outstanding two-photon absorption properties using low-intensity near-infrared light. This provides a noninvasive way to both locate and operate these motors in situ, essential for the application of molecular machines in complex (bio)environments.

Triphenylamine-based highly active two-photon absorbing chromophores with push-pull systems

Two triphenylamine-based star-type push-pull chromophores (T1, T2) were designed and synthesized. Triphenylamine serves as the central core and acts as an electron-donating group surrounded by electron-withdrawing pentafluorobenzene or N,N-dimethyl substituted tetrafluorobenzene, which are connected by ethylene bridges. Single-crystal X-ray diffraction confirmed the structures and molecular arrangement of two chromophores. The systematic photophysical research of T1 and T2 absorption characteristics was carried out to gain a better understanding of how structure-property relationships affect the observed nonlinear optical absorption phenomenon. Complementary calculations based on density functional theory (DFT) further confirmed the experimental results. Both chromophores exhibited excellent two-photon absorption (TPA) properties in CH2Cl2. Notably, T2 has more remarkable nonlinear optical absorption effects with the TPA cross-section up to 4.24 × 107 GM. By adjusting the electronic structures of the chromophores through introducing pentafluorobenzene or N,N-dimethyl as functional groups with different electron-donating or withdrawing behaviors, the TPA performance of the small organic molecule could be greatly enhanced. These molecular structures with push-pull systems were excellent candidates for different two-photon applications.

Theoretical study on a series of naphthalimide-contained two-photon fluorescent hypochlorite probe targeting endoplasmic reticulum: response mechanism and receptor effect.

The development of detecting hypochlorous acid (HClO) in living endoplasmic reticulum has attracted much attention in the fields of biology, medicine, and pharmacy. In the present work, the one-photon absorption (OPA), one-photon emission (OPE), and two-photon absorption (TPA) properties of a series newly synthesized chemosensors with naphthalimide as the fluorophore were systematically investigated using time-dependent density functional theory in combination with response theory. Special emphasis is placed on evolution of the probes' optical properties in the presence of HClO. These compounds show drastic changes in their photoabsorption and photoemission properties when they react with HClO, indicating them to be excellent candidates as fluorescent chemosensors. To further understand the mechanisms of the two probes, we have employed the hole and electron analysis to investigate the charge transfer process for the photoemission of the molecules. The receptor effect is found to play a dominant role in the sensing performance of these probes. Specifically, two-photon absorption properties of the molecules are calculated. We have found that all probes show significant two-photon responses in the near-infrared light region. And the maximum two-photon absorption cross section of probe 2 is greatly enhanced with the presence of HClO, indicating that probe 2 can act as a potential two-photon excited fluorescent HClO probe. The theoretical investigations would be helpful to build the structure-property relationships for the naphthalimide-contained probes, providing information on the design of efficient two-photon fluorescent sensors that can be used for biological imaging of HClO in endoplasmic reticulum.

Gold-Doping Effect on Two-Photon Absorption and Luminescence of Atomically Precise Silver Ligated Nanoclusters.

Noble metal nanoclusters allow for the atomically-precise control of their composition. However, to create nanoclusters with pre-defined optical properties, comprehensive description of their structure-property relation is required. Here, we report the gold atom doping impact on one-photon and two-photon absorption (TPA) and luminescence properties of ligated silver nanoclusters via combined experimental studies and time-dependent density functional theory simulations (TD-DFT). We synthesized a series of Ag25-x Aux (DMBT)18 nanoclusters where x=0, 1 and 5-10. For Ag24 Au1 (DMBT)18 we demonstrate that the presence of the central Au dopant strongly influences linear and non-linear optical properties, increasing photoluminescence quantum yield and two-photon brightness, with respect to undoped silver nanoclusters. With improved TPA and luminescence, atomically-precise AuAg alloys presented in our work can serve as robust luminescent probes e.g. for bioimaging in the second biological window.

One-Pot Synthesis of Pyreno[2,1-b]furan Molecules with Two-Photon Absorption Properties.

The development of large π-conjugated polycyclic heteroaromatic materials is of immense interest, both in the academic as well as the industrial community. Herein, we present the efficient one-pot synthesis of novel pyreno[2,1-b]furan molecules from a newly designed intermediate, which display intense green emission (505-516 nm) in solution and a large red shift emission (625-640 nm) in the solid state, because of strong π-π stacking. More interestingly, the compounds exhibit novel two-photon absorption (TPA) properties, and the TPA cross-section (δ) value was increased to 533 GM by regulating the electronic effects of the substituents of the pyreno[2,1-b]furan molecules. This study not only offers a facile strategy for constructing new pyrene-fused luminescence materials with two-photon absorption properties but also provides a new chemical intermediate that opens up a new pathway to advanced materials.

Effectiveness of collagen cross-linking induced by two-photon absorption properties of a femtosecond laser in ex vivo human corneal stroma.

This study aimed to investigate the effectiveness of two-photon induced collagen cross-linking (CXL) using femtosecond lasers in human corneal stroma. An 800-nm femtosecond laser optical path for CXL was established. Corneal samples that received two-photon induced CXL and ultraviolet-A (UVA) CXL underwent uniaxial stretching experiments, proteolytic resistance assays and observation of collagen fiber structure changes. Two-photon induced CXL can achieve corneal stiffening effects comparable to UVA CXL and showed better advantages at low strains. The cornea after two-photon induced CXL exhibited high enzymatic resistance and tight collagen fiber arrangement. Two-photon induced CXL promises to be a new option for keratoconus.
