Chemical composition and nutritional characteristics of popular wild edible mushroom species collected from North-Western Romania

Wild edible mushrooms contain substances of nutritional and biological importance for humans, but they can also accumulate potentially toxic elements. The study aimed to determine the proximate composition, fatty acids profile and content of 18 elements (As, B, Ca, Cd, Co, Cr, Cu, Fe, Hg, K, Li, Mg, Mn, Na, Ni, P, Pb, S, Zn) in four species of popular wild edible mushrooms (Agaricus campestris, Boletus edulis, Lepiota Procera, and Russula cyanoxantha) collected from NW Romania. The analysed mushrooms contained 22 – 28 % proteins and 2 – 5 % lipids dry weight (dw) basis, in which the total unsaturated fatty acids contents prevailed over saturated fatty acids. The average contents (mg kg−1dw) of the essential macroelements were: 22900 (K), 5500 (P), 1920 (S), 1250 (Na), 701 (Ca), and 565 (Mg). The contents of potentially toxic elements were lower and decreased in the order of Zn>Fe>Cu>Mn>Cr>Hg>Ni>Pb>As>Cd. The toxicity of these elements is related to the ingested amounts, and thus the health risk was assessed considering their contents. The obtained scores indicated non-carcinogenic health risks, primarily because of the high Hg contents, and children’s vulnerability compared to adults. Fortunately, no carcinogenic risk was indicated for the ingestion of the studied mushrooms.

Effect of Meal-Timing on the Association of Unsaturated Fatty Acids with All-Cause and Cardiovascular Mortality among Adults: A Prospective Cohort Study with 10-Year Follow-Up.

Conflicting results have been reported on the association of dietary unsaturated fatty acids (UFAs) with longevity and cardiovascular health. Most previous studies have focused only on the amount of UFAs consumed, not the timing of intake. This prospective cohort study used data from 30,136 adults aged 18 years and older. Intakes of UFAs by meal time and types were assessed by a 24-h dietary recall for two days. The covariate-adjusted survey-weighted Cox proportional hazards models were performed to evaluate the associations of dietary total unsaturated fatty acid (TUFA), polyunsaturated fatty acid (PUFA), and monounsaturated fatty acid (MUFA) intakes throughout the day and three meals with mortality. During a median of 10.0 years of follow-up, 4510 total deaths occurred. All-cause mortality decreased with increasing intakes at dinner of TUFA (HR: 0.87 [0.77-0.98]), PUFA (HR: 0.81 [0.73-0.91]), and MUFA (HR: 0.88 [0.77-0.99]). With an increased intake of PUFA at dinner, CVD mortality showed a decreasing trend. However, the inverted L-shaped non-linear trend in all-cause mortality was found with increasing intake at breakfast of TUFA (HR: 1.35 [1.17-1.57], Q3 vs. Q1), PUFA (HR: 1.30 [1.13-1.50]), and MUFA (HR: 1.28 [1.13-1.45]). Meanwhile, increased breakfast intake of UFAs was associated with increased CVD and heart disease mortality. Meal timing influences the association of UFAs with all-cause and CVD-related mortality.

Effect of Ginger (Zingiber officinale) and Cinnamon (Cinnamon zeylanicum) on Production, Fatty Acid Profile and Meat Quality of Broiler Chickens

Phytogenic feed additives play an important role in broilers' nutrition, contributing to the improvement of the performance and quality of meat. The study aimed to evaluate the effect of Ginger (Zingiber officinale) and Cinnamon (Cinnamon zeylanicum) on broiler chicken production, fatty acid profile, and meat quality. In the present study, 140-day-old Vencob-400 broiler chicks were divided into 7 groups, including the control group (with no additives, T0), and T1 to T6 groups receiving varying concentrations of cinnamon and ginger. Accordingly, the chickens’ diet in T1 was supplemented with 1.0% cinnamon, T2 with 2.0% cinnamon, T3 with 3.0% cinnamon, T4 with 1.0% ginger, T5 with 2.0% ginger, and T6 with 3.0% ginger, all calculated based on dry matter. The carcass traits, weight of immune organs, organoleptic tests, and fatty acid profile of meat (breast and thigh) were recorded after the age of 42 days. The findings indicated that the breast and thigh had the highest organ weights in group T4 compared to other groups, however, the neck, back, drumstick, wing, and heart were not affected. The inclusion of 2% cinnamon (T2) and 1% ginger (T4) in the diet, significantly enhanced the color, texture, flavor, juiciness, and overall acceptability of the meat, compared to the diet of the control group. Adding a supplement of 2% cinnamon or 1% ginger powder to the diet of broiler chickens significantly decreased the percentage of total saturated fatty acid and increased the total unsaturated fatty acid (breast and thigh). The improvement in fatty acid composition is beneficial for the quality of the broiler meat. Based on these findings, it is recommended to supplement the diet of the broiler with either 2% cinnamon or 1% ginger powder to improve the carcass parameters and quality of the meat.

Epoxygenase Cyp2c44 Regulates Hepatic Lipid Metabolism and Insulin Signaling by Controlling FATP2 Localization and Activation of the DAG/PKCδ Axis.

Cytochrome P450 epoxygenase Cyp2c44, a murine epoxyeicosatrienoic acid (EET) producing enzyme, promotes insulin sensitivity and Cyp2c44(-/-) mice show hepatic insulin resistance. Because insulin resistance leads to hepatic lipid accumulation and hyperlipidemia, we hypothesized that Cyp2c44 regulates hepatic lipid metabolism. Standard chow diet (SD) fed male Cyp2c44(-/-) mice had significantly decreased EET levels and increased hepatic and plasma lipid levels compared to wild-type mice. We showed increased hepatic plasma membrane localization of the FA transporter 2 (FATP2) and total unsaturated fatty acids and diacylglycerol levels. Cyp2c44(-/-) mice had impaired glucose tolerance and increased hepatic plasma membraneassociated PKCδ and phosphorylated IRS-1, two negative regulators of insulin signaling. Surprisingly, SD and high fat diet fed (HFD) Cyp2c44(-/-) mice had similar glucose tolerance and hepatic plasma membrane PKCδ levels, suggesting that SD-fed Cyp2c44(-/-) mice have reached their maximal glucose intolerance. Inhibition of PKCδ resulted in decreased IRS-1 serine phosphorylation and improved insulin-mediated signaling in Cyp2c44(-/-) hepatocytes. Finally, Cyp2c44(-/-) HFD-fed mice treated with the analog EET-A showed decreased hepatic plasma membrane FATP2 and PCKDδ levels with improved glucose tolerance and insulin signaling. In conclusion, loss of Cyp2c44 with concomitant decreased EET levels leads to increased hepatic FATP2 plasma membrane localization, diacylglycerol accumulation, and PKCδ-mediated attenuation of insulin signaling. Thus, Cyp2c44 acts as a regulator of lipid metabolism by linking it to insulin signaling.

Systematic characterization of CsbZIP transcription factors in Camelina sativa and functional analysis of CsbZIP-A12 mediating regulation of unsaturated fatty acid-enriched oil biosynthesis

The basic leucine zipper (bZIP) transcription factors (TFs) function importantly in numerous life processes in plants. However, bZIP members and their biological roles remain unknown in Camelina sativa, a worldwide promising oil crop. Here, 220 CsbZIP proteins were identified in camelina and classified into thirteen groups. Two and 347 pairs of tandem and segmental duplication genes were detected to be underwent purification selection, with segmental duplication as the main driven-force of CsbZIP gene family expansion. Most CsbZIP genes displayed a tissue-specific expression pattern. Particularly, CsbZIP-A12 significantly positively correlated with many FA/oil biosynthesis-related genes, indicating CsbZIP-A12 may regulate lipid biosynthesis. Notably, yeast one-hybrid (Y1H), β-Glucuronidase (GUS), dual-luciferase (LUC) and EMSA assays evidenced that CsbZIP-A12 located in nucleus interacted with the promoters of CsSAD2–3 and CsFAD3–3 genes responsible for unsaturated fatty acid (UFA) synthesis, thus activating their transcriptions. Overexpression of CsbZIP-A12 led to an increase of total lipid by 3.275 % compared to the control, followed with oleic and α-linolenic acid levels enhanced by 3.4 % and 5.195 %, and up-regulated the expressions of CsSAD2–3, CsFAD3–3 and CsPDAT2–3 in camelina seeds. Furthermore, heterogeneous expression of CsbZIP-A12 significantly up-regulated the expressions of NtSAD2, NtFAD3 and NtPDAT genes in tobacco plants, thereby improving the levels of total lipids and UFAs in both leaves and seeds without negative effects on other agronomic traits. Together, our findings suggest that CsbZIP-A12 upregulates FA/oil biosynthesis by activating CsSAD2–3 and CsFAD3–3 as well as possible other related genes. These data lay a foundation for further functional analyses of CsbZIPs, providing new insights into the TF-based lipid metabolic engineering to increase vegetable oil yield and health-beneficial quality in oilseeds.

Feeding pomegranate pulp to Ghezel lambs for enhanced productivity and meat quality

Agrifood by-products contain nutrients and bioactive compounds that can be used in the diets of livestock – thereby value-adding to an otherwise waste product of environmental and economic significance. This study investigated the effect of dietary pomegranate pulp in the total mixed ration of Ghezel lambs, evaluating its effect on growth performance, blood parameters, carcass traits, as well as meat quality and shelf life. 3-month-old Ghezel lambs (individually housed, n = 8) were randomly assigned to be either non-supplemented (control) or supplemented with 100 g/kg DM of sun-dried pomegranate pulp for 28 days, post-adjustment. Results showed that supplementation of lamb diets with pomegranate pulp significantly increased liveweight and average daily gains, while not significantly affecting dry matter intake. Lamb serum urea and alkaline phosphatase concentrations and hot carcass weight were increased with pomegranate pulp supplementation. Compared to control lambs, the meat from lambs fed the supplemented diet had higher concentrations of intramuscular fat, mono- and polyunsaturated fatty acid, total unsaturated fatty acid, and meat phenolic compounds. Pomegranate pulp supplemented lambs also had a higher ratio of polyunsaturated to saturated fatty acids; and produced liver tissue with less fat and ash contents. Meat oxidative status (thiobarbituric acid reactive substance) and quality (water holding capacity, colour, and pH) were improved when lambs were supplemented with pomegranate pulp. These findings demonstrate that using pomegranate pulp as a feed for Ghezel lambs has advantageous effects on animal performance and meat quality, offering valorisation of an agrifood by-product.

Utilization of hazelnut oil oleogels for the preparation of milk couverture type compound chocolates: Composition, properties, and sensory evaluations

AbstractThe aim of this study was to use oleogels in compound chocolates. A control sample with cocoa butter (CNT) and two compound chocolates with hazelnut oil‐sunflower wax oleogel (SWO‐CC), and polyglycerol stearate oleogel (PGSO‐CC) were prepared with the same recipe. The main properties and compositions of the samples were quite similar. While CNT sample included only 37.97% of total unsaturated fatty acids, it was 74.19 and 63.08% in the SWO‐CC and PGSO‐CC, respectively. The melting peak temperatures were 32.1, 25.4, and 23.8°C for CNT, SWO‐CC and PGSO‐CC. The samples had 11.75%, 74.25%, and 74.25% shape retention index values at 60°C. Clearly compound chocolates melted at lower temperatures, but retained their shape at higher temperatures. After 15‐day temperature fluctuation storage, no fat bloom was developed. Rheological data showed that the PGSO‐CC sample was stiffer among all, and the compound chocolates melted slowly up to 40°C, but CNT melted sharply at 38°C. Further, the PGSO‐CC sample showed a lubricating behavior. Although CNT sample included 7 volatile aromatics, SWO‐CC and PGSO‐CC samples had 17 and 14 compounds, respectively. Trained panel described the samples with 13 attributes, and most profoundly the compound chocolates had lower shape, surface gloss, coffee, bitter, cooling, hardness scores, and higher coating scores. Consumer test revealed that compound chocolates had lower appearance scores, and equal aroma scores. The flavor score and acceptability were highest for the PGSO‐CC sample. Overall, this study proved that heat stable and sensorially acceptable compound chocolates could be prepared from the oleogels.

Multivariate Analysis of Biochemical Properties Reveals Diversity among Yardlong Beans of Different Origins.

This study analyzed the nutrient levels, secondary metabolite contents, and antioxidant activities of 35 yardlong bean accessions from China, Korea, Myanmar, and Thailand, along with their key agronomic traits. Significant variations were found in all the parameters analyzed (p < 0.05). The crude fiber (CFC), dietary fiber (DFC), total protein, and total fat contents varied from 4.10 to 6.51%, 16.71 to 23.49%, 22.45 to 28.11%, and 0.59 to 2.00%, respectively. HPLC analysis showed more than a 10-fold difference in vitamin C level (0.23 to 3.04 mg/g), whereas GC-FID analysis revealed the dominance of palmitic acid and linoleic acid. All accessions had high levels of total unsaturated fatty acids, which could help in preventing cardiovascular disease. Furthermore, total phenolic, tannin, and saponin contents ranged between 3.78 and 9.13 mg GAE/g, 31.20 and 778.34 mg CE/g, and 25.79 and 82.55 mg DE/g, respectively. Antioxidant activities like DPPH• scavenging, ABTS•+ scavenging, and reducing power (RP) ranged between 1.63 and 9.95 mg AAE/g, 6.51 and 21.21 mg TE/g, and 2.02, and 15.58 mg AAE/g, respectively. Days to flowering, total fat, palmitic acid, oleic acid, and TPC were significantly influenced by origin and genotype differences, while seeds per pod, one-hundred seeds weight, CFC, DFC, vitamin C, RP, and TSC were not affected by these factors. Multivariate analysis categorized the accessions into four clusters showing significant variations in most of the analyzed parameters. Correlation analysis also revealed significant relationships between several noteworthy parameters. Overall, this comprehensive analysis of biochemical factors revealed diversity among the different yardlong bean varieties. These findings could have practical applications in industries, breeding programs, and conservation efforts.

Possibilities of Utilising Biomass Collected from Road Verges to Produce Biogas and Biodiesel

Grass collected as part of roadside maintenance is conventionally subjected to composting, which has the disadvantage of generating significant CO2 emissions. Thus, it is crucial to find an alternative method for the utilisation of grass waste. The aim of this study was to determine the specific biogas yield (SBY) from the anaerobic mono-digestion of grass from road verges and to assess the content of Fatty Acid Methyl Esters (FAMEs) in grass in relation to the time of cutting and the preservation method of the studied material. The biochemical biogas potential (BBP) test and the FAMEs content were performed on fresh and ensiled grass collected in spring, summer, and autumn. The highest biogas production was obtained from fresh grass cut in spring (715.05 ± 26.43 NL kgVS−1), while the minimum SBY was observed for fresh grass cut in summer (540.19 ± 24.32 NL kgVS−1). The methane (CH4) content in the biogas ranged between 55.0 ± 2.0% and 60.0 ± 1.0%. The contents of ammonia (NH3) and hydrogen sulphide (H2S) in biogas remained below the threshold values for these inhibitors. The highest level of total FAMEs was determined in fresh grass cut in autumn (98.08 ± 19.25 mg gDM−1), while the lowest level was detected in fresh grass cut in spring (56.37 ± 7.03 mg gDM−1). C16:0 and C18:0, which are ideal for biofuel production, were present in the largest amount (66.87 ± 15.56 mg gDM−1) in fresh grass cut in autumn. The ensiling process significantly impacted the content of total FAMEs in spring grass, leading to a reduction in total saturated fatty acids (SFAs) and an increase in total unsaturated fatty acids (USFAs). We conclude that grass biomass collected during the maintenance of road verges is a valuable feedstock for the production of both liquid and gaseous biofuels; however, generating energy from biogas appears to be more efficient than producing biodiesel.

Possibility of using fatty acid profiles for the authentication of beef adulterated with pork, donkey, and dog meat

Detection of meat adulteration is a critical issue in food labeling procedures and a serious concern related to food fraud, authenticity, and religious beliefs. The current work detected and quantified adulteration of raw ground beef with pork, donkey, and dog meat based on fatty acid profiles using GC-MS/MS. The study design incorporated pork, donkey, or dog meat with beef meat; negative and positive controls were used for the different meat species. Results demonstrated several significant differences (p &lt; 0.05) in fatty acid contents between mixed/adulterated meat and pure beef. In addition, higher total saturated fatty acid levels in ground beef (57.91%) compared to dog fat (46.44%), donkey (38.71%), and pork fat (lard) (40.23%). High total unsaturated fatty acids content was observed in donkey (61.92%), pork (59.77%), and dog (53.56%) fats compared to beef fat (42.09%). On the other hand, total unsaturated and monounsaturated fatty acids in beef meat were lower than in pork, donkey, and dog meat. Moreover, the highest trans-fatty acid content was found in pork compared to the other meat types. All incorporated samples correlated positively with pure ground beef concerning the fatty acid profiles. Therefore, alteration of the mixed beef fatty acid profiles was a potential indicator for adulteration since a marked decrease in total saturated fatty acid content and an increase in unsaturated fatty acids was observed in the substituted samples. We concluded that GC-MS/MS-based fatty acid profiling is a promising technique that can be used to detect meat adulteration.

Improving the soil K+/Na+ ratio under moderate salt stress synergistically increases the yield and quality of cotton fiber and cottonseed

Cotton production areas have been shifted to saline-alkaline soils with high Na+ content, resulting in low soil K+/Na+ ratios, leading to K+ deficiency in cotton, which is detrimental to the yield and quality of the fiber and cottonseed. It was hypothesized that improving the soil K+/Na+ ratio by applying potassium under moderate salt stress could affect the contents of K+ and Na+ in various organs of cotton bolls. This could affect the growth and biomass distribution of the cotton bolls, ultimately impacting the yield and quality of both the fiber and the cottonseed. To test this hypothesis, a pot experiment using the cotton cultivar CCRI-79 was conducted in 2021 and 2022, and the results showed that improving the soil K+/Na+ ratio under moderate salt stress (1) increased the cotton boll K+/Na+ ratio, accelerated boll growth, and increased the proportion of boll biomass distributed to the fiber and the cottonseed kernel. (2) increased fiber and cottonseed yield. Specifically, the boll retention rate and the number of fruit nodes increased, thereby increasing the boll number. The increase in boll weight can be explained by an increase in the components of fiber and cottonseed yield per boll. Compared with those in the control soil without added salts (C), the fiber and cottonseed yield at a K+/Na+ ratio of 1:9 (SK2) were only 7.1%–21.0% and 4.9%–17.0% lower, respectively. (3) increased fiber and cottonseed quality. SK2 can completely alleviate the effects of moderate salt stress on fiber uniformity, length, and micronaire. And the fiber strength of SK2 is only 5.0%–7.2% lower than C. The oil content and yield of cottonseed kernels increased, and the total unsaturated fatty acid (UFA) and unsaturated index (UI) increased. Although the protein content decreased, the protein yield increased, and the total amino acid (TAA) content increased. SK2 and C did not differ significantly in oil and protein contents. Taken together, these findings indicate that the optimal alleviation of moderate salt stress on the yield and quality of fiber and cottonseed was achieved at a K+/Na+ ratio of 1:9. These results will help to synergistically increase the yield and quality of cotton fiber and cottonseed in saline-alkaline soil and provide new clues for elucidating the mechanism through which the K+/Na+ ratio regulates salt tolerance in cotton.

Gluten‐free pasta fortified with moringa leaves: effect of recipe ratios on quality and structural properties

SummaryPasta has a big publicity worldwide and gone through lots of research to improve its quality. It is a good source of carbohydrates and recently new types of pasta are being good sources of fibres and proteins, too. The objective in this study is to produce highly nutritious pasta with good quality trying to reduce some of Kuwaiti population diseases such as obesity and diabetes. I optimal RSM design was used to make combinations of three ingredients with different ratios (oats, corn, and moringa) and check their effect on proximate composition, textural properties, colour, cooking loss percentage, microstructural properties, and fatty acid profile. Results found that run #2 (10:70:20) (Moringa: Corn: Oats) is the optimum run with 0.63 desirability through having high protein and fibre contents 11.5% and 5.1%, respectively, with low fat and cooking loss percentages (10.3%). The same run also had the highest total unsaturated fatty acids (78%). Both microstructure and the texture affected cooking loss results where more cavities in the microstructure increased cooking loss and harder samples reduced cooking loss.

Quality Properties and Flavor-Related Components of Beef Longissimus Lumborum Muscle from Four Korean Native Cattle Breeds.

This study was carried out to assess the quality properties, components associated with taste and aroma of beef as a function of breed. For this purpose, steers from four Korean native cattle breeds: Hanwoo (n=10), Chikso (n=10), black Hanwoo (n=12, BHW) and Jeju black cattle (n=12, JBC) were used. The steers all were raised under identical conditions and finished at a similar age of around 30-months old. Following 24 h of slaughter, all longissimus lumborum muscles were collected and used for analysis of meat quality, fatty acids, and flavor-related components (metabolic compounds, free amino acids, and aroma volatiles). The Hanwoo presented a significantly higher intramuscular fat content (IMF, 22.85%) than the BHW (11.78%), Chikso (9.25%), and JBC (9.14%; p<0.05). The meat of Hanwoo breed showed lighter and redder color, and lower shear force value (p<0.05). The JBC presented a "healthier" fatty acid profiles as it had a higher total unsaturated fatty acids content (p<0.05). With regard to flavor-related components, Hanwoo also had higher total contents of free amino acids and metabolites associated with umami and sweet tastes, and fat-derived volatile compounds (aldehydes, alcohols, and ketones) associated with fatty aroma. It may be concluded that there was a considerable difference in the meat quality properties among breeds. The variations of IMF content and flavor-related components may be the main factors contributing to the typical flavors of beef among the four Korean native cattle breeds.

Saturated, monounsaturated and polyunsaturated fatty acid profiles of commercially important traditional and new improved red and white rice varieties of Sri Lanka

The present study evaluated saturated, monounsaturated, and polyunsaturated fatty acid (FA) profiles of seven new improved and three traditional red and white rice varieties (RV) of Sri Lanka. Oil was extracted from whole grain rice flour, derivatized to methyl esters, and analyzed by gas chromatography (n = 3 each). Results showed significant differences (p < 0.05) in FA profiles among the RV, red vs white RV, and traditional vs new improved RV. The oil % of RV varied from 1.94–3.19%. Total FAs, saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs), polyunsaturated fatty acids (PUFAs), total unsaturated FAs, and PUFAs to SFAs ratios of studied RV varied from 19.40–31.90, 4.40–7.04, 8.35–13.60, 6.65–11.26 and 15.00–24.86 mg/g of rice and 1.44–1.83 respectively. The tested RV had four omega-6 FAs while omega-3 FAs were absent. The major FAs found in all RV were palmitic, oleic, and linoleic acids and these three FAs together contributed to 88–95% of the total FAs present in rice. Among these FAs, 70–76% were unsaturated FAs. Considering all, RV consisting of healthy fatty acid profiles could be identified by studying their total fatty acid profile rather than studying their oil % and the total fatty acid contents.

Ploidy and fruit trait variation in oil-tea Camellia: Implications for ploidy breeding

Plant polyploidy often occurs in conjunction with higher yield and superior quality. Therefore, obtaining polyploid germplasms is a significant part of breeding. The oil-tea Camellia tree is an important native woody plant that produces high-quality edible oil and includes many species of Camellia with different ploidies. However, whether higher ploidy levels in oil-tea Camellia trees are related to better traits remains unclear. In this study, the ploidy levels of 30 different oil-tea Camellia strains in three different species in the Sect. Paracamellia were determined by flow cytometry and chromosome preparation, and the phenotypic characteristics and fatty acid compositions of the fruits were examined by field observations and laboratory analyses. The correlations between the ploidy level of oil-tea Camellia and the main traits of the fruit were investigated. Our results showed that 10 Camellia lanceoleosa strains were diploid, 10 Camellia meiocarpa strains were tetraploid and 10 Camellia oleifera strains were hexaploid. Hexaploid C. oleifera had larger fruit size and weight, more seeds per fruit, greater seed weight per fruit, higher oil content and greater yield per crown width than tetraploid C. meiocarpa and diploid C. lanceoleosa, but their fruit peel thickness and fresh seed rate were significantly lower, and these traits were significantly correlated with ploidy level. In addition, in terms of fatty acid composition, hexaploid C. oleifera had a higher oleic acid content than tetraploid C. meiocarpa and diploid C. lanceoleosa, but their linoleic acid, linolenic acid and arachidonic acid contents were lower. The contents of palmitic acid, stearic acid and total unsaturated fatty acids were not significantly correlated with ploidy level. In conclusion, certain correlations exist between the main characteristics of oil-tea Camellia fruit and the ploidy level, and increasing the ploidy level led to an increase in fruit yield with no effect on oil composition. The discovery of variations in the main characteristics of oil-tea Camellia fruit with different ploidies will facilitate germplasm innovation and lay a foundation for ploidy breeding and mechanistic research on fruit traits.

Altered myocardial lipid regulation in junctophilin-2-associated familial cardiomyopathies.

Myocardial lipid metabolism is critical to normal heart function, whereas altered lipid regulation has been linked to cardiac diseases including cardiomyopathies. Genetic variants in the JPH2 gene can cause hypertrophic cardiomyopathy (HCM) and, in some cases, dilated cardiomyopathy (DCM). In this study, we tested the hypothesis that JPH2 variants identified in patients with HCM and DCM, respectively, cause distinct alterations in myocardial lipid profiles. Echocardiography revealed clinically significant cardiac dysfunction in both knock-in mouse models of cardiomyopathy. Unbiased myocardial lipidomic analysis demonstrated significantly reduced levels of total unsaturated fatty acids, ceramides, and various phospholipids in both mice with HCM and DCM, suggesting a common metabolic alteration in both models. On the contrary, significantly increased di- and triglycerides, and decreased co-enzyme were only found in mice with HCM. Moreover, mice with DCM uniquely exhibited elevated levels of cholesterol ester. Further in-depth analysis revealed significantly altered metabolites from all the lipid classes with either similar or opposing trends in JPH2 mutant mice with HCM or DCM. Together, these studies revealed, for the first time, unique alterations in the cardiac lipid composition-including distinct increases in neutral lipids and decreases in polar membrane lipids-in mice with HCM and DCM were caused by distinct JPH2 variants. These studies may aid the development of novel biomarkers or therapeutics for these inherited disorders.

Application of benzothiadiazole to Cabernet Gernischt grapes (Vitis vinifera L.) for quality improvement: Effects on aroma metabolism precursors and related genes expression

Pre-harvest spraying of benzothiadiazole (BTH) can improve the winemaking properties of grapes, especially their aroma compounds and phenolics. Limited research has explored the molecular mechanisms by which BTH influences the accumulation of grape aroma precursors during early grape development. This study investigated the effects and putative molecular mechanisms of applying 0.37 mM BTH through whole-plant spraying on the accumulation of aroma metabolism precursors and gene expression in Cabernet Gernischt grapes during ripening. The results showed that BTH treatment increased the levels of fructose, alanine, aspartate, threonine, myristic acid, myristoleic acid, palmitic acid, β-cryptoxanthin, norisoprenoids and methoxypyrazines. Contrarily, it decreased the levels of glucose, sucrose, phenylalanine, tyrosine, leucine, valine, glycine, arginine, histidine, total unsaturated fatty acids (particularly linoleic acid), zeaxanthin, lutein, and organic acids. Additionally, BTH upregulated the expression of genes associated with the production and degradation of amino acids, fatty acids, and carotenoids while decreasing the expression of genes involved in the synthesis and degradation of soluble sugars and organic acids. Ten different metabolites, including fumaric acid, were identified as potential biological markers for distinguishing BTH-treated grapes from control grapes. The study demonstrates that BTH treatment had a substantial impact on the concentration and developmental patterns of aroma metabolism precursors. Furthermore, it altered the winemaking characteristics of Cabernet Gernischt grapes by modulating genes associated with the production and breakdown of metabolites.

Effect of cinnamon powder on quality attributes and off-flavor in fried chicken drumsticks made from long-term thawed Korean native chicken

The effect of cinnamon powder on the quality and mitigation of off-flavor in fried chicken drumsticks made from long-term thawed Korean native chicken (Woorimatdag No. 1, WRMD1) was investigated. The WRMD1 drumsticks were categorized into 5 groups: conventional thawing (16 h, CT), long-term thawing (48 h, LT), cinnamon powder added into 'LT' as marinade (0.03%, CM) or incorporated into the batter (1.35%, CB), and long-term thawing with cinnamon powder incorporated both in the marinade and batter (0.03% + 1.35%, CMB). The crude fat content was significantly higher in the CT and CMB than that of the CB. The CM, CB, and CMB showed significantly lower levels of 2-thiobarbituric acid reactive substance compared with the CT and LT. The predominant fatty acids in all treatments were C18:1n9, C18:2n6, and C16:0. The LT displayed lower total unsaturated fatty acid content than the CT (P < 0.05). The CM effectively decreased lipid oxidative volatiles, such as 1-octanol, 1-octen-3-ol, and 2-octen-1-ol, (E), in the LT (P < 0.05). Both the CM and CB showed an inclination to increase specific pyrazines associated with pleasant notes compared with the LT, and showed higher levels of pyrazines, such as pyrazine, 2-ethyl-6-methyl-, and pyrazine, 3-ethyl-2,5-dimethyl-, than those of the CMB (P < 0.05). The CM contained higher levels of 2,3-butanedione when compared with the other groups (P < 0.05). Multivariate analysis demonstrated that cinnamon had an effect in discriminating the treatment groups with cinnamon addition from both the CT and LT, whereas the CM, CB, and CMB formed distinct clusters. The CM and CMB received significantly higher aroma scores from panelists in comparison to the other groups. These findings suggest that the CM (0.03% cinnamon powder) can be used to enhance the aroma in fried WRMD1 drumsticks by reducing or masking the off-flavor volatiles associated with long-term thawing.

Phytochemical profile of secondary metabolites in the phloem of mature Pinus pinaster trees attacked by the pine wood nematode

The present study highlights, for the first time, the chemical composition of secondary metabolites (lipophilic and hydrophilic compounds) present in the phloem tissues of infected mature Pinus pinaster Ait. trees (on average 74 years old), which may be potentially determinant in the development of the pine wood nematode (PWN) Bursaphelenchus xylophilus. The levels of secondary metabolites in the phloem of infected trees were fairly constant along the height of the trees (an average of 30% dry phloem), but slightly lower compared with total extractives content in the phloem of mature healthy P. pinaster trees (30.0 vs. 39.9%). The hydrophilic fraction represented 91% of the total soluble compounds (27.3% of phloem mass) and it was mainly composed of phenolic compounds and reducing sugars (44.8% and 58.8% of the ethanol-water extracts). The lipophilic fraction (2.7% of phloem mass), was dominated by tricyclic diterpenic compounds (54.2–63.2% of the total lipophilic extracts) and fatty acids. Differences in the relative concentrations of individual compounds were observed compared to healthy pines, suggesting a response to the biotic stress imposed by nematode damage by an increased content of diterpenic resin acids, specifically dehydroabietic and abietic acids, and of total unsaturated fatty acids with a predominance of 9-octadecenoic acid.

Effect of Different Edible Trichosanthes Germplasm on Its Seed Oil to Enhance Antioxidant and Anti-Aging Activity in Caenorhabditis elegans

The seeds of various Trichosanthes L. plants have been frequently used as snacks instead of for traditional medicinal purposes in China. However, there is still a need to identify the species based on seeds from Trichosanthes germplasm for the potential biological activities of their seed oil. In this study, 18 edible Trichosanthes germplasm from three species were identified and distinguished at a species level using a combination of seed morphological and microscopic characteristics and nrDNA-ITS sequences. Seed oil from the edible Trichosanthes germplasm significantly enhanced oxidative stress tolerance, extended lifespan, delayed aging, and improved healthspan in Caenorhabditis elegans. The antioxidant activity of the seed oil exhibits a significant positive correlation with its total unsaturated fatty acid content among the 18 edible Trichosanthes germplasm, suggesting a genetic basis for this trait. The biological activities of seed oil varied among species, with T. kirilowii Maxim. and T. rosthornii Harms showing stronger effects than T. laceribractea Hayata.
