Histological Assessment for the effect of deodorant blocker on the blood cells of albino rat

Twenty Rats were used in the present study to evaluate the effect of the deodorant blocker on the blood cells of the rat including the white blood cells and red blood cells. The animals were distributed into Group A control Five Rats and Group B Five treated animals. 
 The animals were put in steel cages in the animal house of veterinary medicine college. water with food free putto animals the skin of whole animals was shaved at the dorsum and at the flank region, then deodorant blocker was applied daily by whipping those areas for 30 days continuously, otherwise the control group was applying the distill water on the shaved areas. After the end of the experiment, a drop blood from the tip of tail was obtained and spread on the slide for staining by Leishman stain. 
 The results of blood examination were indicated release of the lymphocytes and monocytes percentage in comparison of control, also the certain number of RBCs indicated macrocytic, microcytic, incomplete hemoglobin ring and target cells as deformities of RBCs. In conclusions; the deodorant blocker induced imitation of immune system via increase the number of lymphocytes and monocytes which consider as main factors for immune response in cases of extrinsic insults.

Molecular Survey and Microscopic Examination of Haemoparasites Infecting Lizards from Morocco.

Understanding the factors influencing parasite diversification, their distribution and infection patterns among hosts provides valuable information on the impact of parasitism on natural populations and the dynamics and evolution of disease outbreaks. In this study, we investigated prevalence and intensity of infection by blood parasites, the diversity, distribution, host specificity and infection patterns of apicomplexan parasites in lizards of Morocco. Samples from lizard specimens were collected between 2019 and 2021 from different prospected localities in Morocco. In total, 1500 lizards were collected, and their blood and tail tips were screened for blood parasites. Using a PCR approach, we detected two distinct apicomplexan parasite groups (haemogregarines and haemococcidians) and also Stramenopiles (Proteromonas). A significant difference was identified between hosts families, with no agamids or skinks infected and with particularly high infection rates in lacertids. Twenty-four samples were sequenced and included in a phylogenetic assessment. Two samples of Atlantolacerta andreanskyi were infected with unidentified species of Proteromonas. Three samples were infected with haemococcidians, including two samples with Schellackia and another sample with Lankesterella, more closely related to others isolated from bird hosts. These are the first reports of Lankesterella and Proteromonas in these lizards from Morocco. Nineteen different isolates of the complex of Hepatozoon/Karyolysus were identical or very similar to previously sequenced parasites from North Africa and Iberian Peninsula and included new host species. Our results highlight the need for future research on the vectors involved in transmission that may influence the ecology and distribution patterns of these parasites. Given that Morocco is a hot spot of endemicity and reptile richness, our results are relevant not only in the context of their taxonomy, but can also have implications for conservation.

Numerical study of an individual bubble rise in a shear-thinning viscoelastic liquid based on a combination of the Carreau and FENE-CR models

The rise behavior of a single bubble in shear-thinning viscoelastic liquids was investigated using a volume of fluid method based on a combination of the Carreau and FENE-CR models. The effects of Weissenberg number (Wi), polymer stretching parameter (L), Eötvös number (Eo) and rheological index (n) on bubble shape and typical characteristics of flow fields were investigated in detail. The results show that bubble tail becomes pointed under the influence of elastic stress, and the bubble becomes longer along the longitudinal direction with a decrease in Wi. When the magnitude of n is relatively large, the bubble appears as an ellipsoidal shape with a cusp; however, when the magnitude of n is relatively small, bubble shape is closer to a disc, and the tip of bubble tail disappears. Wi affects the magnitude of elastic tensile stress, and affects the action rang of the large tensile stress. With an increase in Wi, the action rang of elastic tensile stress develops from both sides of the bubble towards regions below bubble tail. The appearance of negative wake is not directly related to bubble shape, but to the relaxation of elastic stres.

An incidental finding of microfilaria on a urine cytological smear study

In developing nations, lymphatic filariasis is a significant public health concern. Wuchereria bancrofti species, which are morphologically recognised as sheathed parasites with a tail tip free of nuclei, are responsible for almost 90% of the cases in India. Microfilariae have been found in a wide range of other body fluids as well as urine samples from patients with chylous and achylous hematuria. We report a case of microfilariasis in urine cytology. This study aims to raise awareness of the possibility of filariasis in bodily fluids and aspirates, particularly in areas where it is endemic. Identification results in a comprehensive diagnostic and treatment plan for the patient.

Ficophagus annulatae n. sp. (Nematoda: Aphelenchoididae), an associate of Ficus annulata in China

Summary A new species of the genus Ficophagus was recovered from the syconia of Ficus annulata from Xishuangbanna, Yunnan province, China. It is described herein as Ficophagus annulatae n. sp. and is characterised by possessing the combined characters of a short PUS (9-18 μm or 0.3-0.6 VBD long), an excretory pore (EP) located at the level between stylet basal knobs and metacorpus, presence of crustaformeria, amoeboid sperm, three pairs of subventral papillae on the male tail (one pair just adcloacal (P2), one pair halfway between cloacal aperture and tail terminus (P3), and one pair near tail tip (P4)), rounded male tail tip without mucron, absence of gubernaculum and bow-shaped spicule with indistinct rostrum. Ficophagus annulatae n. sp. was differentiated from other sequenced species by the partial small subunit (SSU) rRNA gene and D2-D3 expansion segments of the large subunit (LSU) rRNA gene. Phylogenetic analysis with the LSU D2-D3 expansion segment sequence suggested that F. annulatae n. sp. is clustered in the same highly supported monophyletic clade with F. benjamina, F. curtipes and F. microcarpus. It differs morphologically from these species in EP position, spicule and uterus morphology, and some morphometric characters.

Further studies on the morphology of Baylisascaris transfuga (Rudolphi, 1819) (Nematoda: Ascaridomorpha: Ascarididae) from the polar bear Ursus maritimus Phipps (Carnivora: Ursidae).

Baylisascaris transfuga (Rudolphi, 1819) is a common parasitic nematode in the digestive tract of various species of bears worldwide, with great veterinary significance. However, our present knowledge on the morphology of B. transfuga remains insufficient. In the present study, the detailed morphology of B. transfuga was studied using light and scanning electron microscopy (SEM), based on specimens collected from the polar bear Ursus maritimus Phipps (Carnivora: Ursidae) in the Shijiazhuang Zoo, China. The results revealed some morphological and morphometric variation between the present specimens and some of those from previous studies, including oesophageal length of female, number and morphology of postcloacal papillae and morphology of tail of males. Present SEM observations clearly showed the detailed morphology of lips, cervical alae, cloacal ornamentation, precloacal medioventral papilla, phasmids and tail tip. These supplementary morphological and morphometric data enable us to identify this ascaridid nematode more accurately.

Structural basis of bacteriophage T5 infection trigger and E. coli cell wall perforation

Most bacteriophages present a tail allowing host recognition, cell wall perforation, and viral DNA channeling from the capsid to the infected bacterium cytoplasm. The majority of tailed phages bear a long flexible tail (Siphoviridae) at the tip of which receptor binding proteins (RBPs) specifically interact with their host, triggering infection. In siphophage T5, the unique RBP is located at the extremity of a central fiber. We present the structures of T5 tail tip, determined by cryo-electron microscopy before and after interaction with its E. coli receptor, FhuA, reconstituted into nanodisc. These structures bring out the important conformational changes undergone by T5 tail tip upon infection, which include bending of T5 central fiber on the side of the tail tip, tail anchoring to the membrane, tail tube opening, and formation of a transmembrane channel. The data allow to detail the first steps of an otherwise undescribed infection mechanism.

Deladenus Bonabensis n. Sp. (Tylenchomorpha: Neotylenchidae) from East Azarbaijan Province, Northwestern Iran: A Morphological and Molecular Phylogenetic Study.

A population of Deladenus, representing a new species, was recovered from Bonab's Ghara-Gheshlagh lagoon. It is mainly characterized by having a long body (1051-1185 μm), long distance of anterior end to pharyngeal glands end (270-312 μm), six lines in lateral fields, and a short mucro-like differentiation on the tail tip. Furthermore, it has a small-sized stylet (8.5-11.0 μm) with three knobs, no postvulval uterine sac, and males with 24- to 28-μm-long tylenchoid spicules and penial tube. With six lateral lines, the new species is comparable with seven species of the genus: D. apopkaetus, D. brevis, D. cocophilus, D. durus, D. obtusicaudatus, D. processus, and D. ulani. It was furthermore compared with D. oryzae with an unknown number of lateral lines and D. aridus, D. obesus, and D. parvus having a different number of lateral lines but similar morphology. In the molecular phylogenic analyses using small and large subunit ribosomal DNA (SSU and LSU D2-D3 rDNA) sequences, the relationships of the new species with other species and genera were not resolved in SSU phylogeny. However, it formed a clade with Deladenus sp. and D. brevis in LSU phylogeny.

Muscles of the sex organs of Panagrolaimus detritophagus (Chromodorea: Rhabditida: Panagrolaimidae)

Summary The sex organs are important for nematode systematics, but the associated muscles are rarely used as phylogenetic characters. This research aimed to study the musculature of Panagrolaimus detritophagus, using confocal microscopy. The ring-shaped anal sphincter is pierced by a biconical X-structure and connected by two pairs of processes to the subventral stomato-intestinal muscles and dorsal body wall. The female has two pairs of vaginal dilators that are connected to the proximally invaginated vaginal sphincter and subventral body wall. The anal depressor is a pair of dorsoventral muscles. The male has 7-8 pairs of caudal diagonal muscles located beneath the ventral body wall muscles, followed by five pairs of postcloacal oblique muscles. The paired posterior longitudinal muscles are connected to the cloacal opening and tail tip. The paired anterior retractors of the spicule connect the spicule capitula to the lateral body wall; a pair of spicule-gubernacular protractors runs from spicule capitula to the anterior part of gubernaculum, and paired ventral protractors run from spicule capitula to anterior cloacal lip. Two pairs of gubernaculum erectors are attached to the mid-gubernaculum, and the unpaired gubernaculum protractor extends ventrally to attach to the body wall at the posterior cloacal lip. The sex muscle pattern is of the Rhabditidae-Cephalobidae type with a 15-character code for phylogenetic reconstructions: 63313-22234-42243.

New Records of Ditylenchus Species from Southern Alberta, Canada.

The presence of plant-parasitic nematodes (PPNs) in cultivated areas is a limiting factor in achieving marketable crop yield. To control and alleviate the effects of these nematodes and determine appropriate management strategies, species-level identification is crucial. Therefore, we conducted a nematode diversity survey, which resulted in the detection of four Ditylenchus species in cultivated areas of southern Alberta, Canada. The recovered species had six lines in the lateral field, delicate stylets (>10 µm long), distinct postvulval uterine sacs, and pointed to rounded tail tips. The morphological and molecular characterization of these nematodes revealed their identity as D. anchilisposomus, D. clarus, D. tenuidens and D. valveus, all of which are members of the D. triformis group. All of the identified species were found to be new records in Canada except for D. valveus. Accurate Ditylenchus species identification is crucial because false-positive identification can result in the implementation of quarantine measures over the detected area. Our current study not only documented the presence of Ditylenchus species from southern Alberta, but also described their morpho-molecular characteristics and subsequent phylogenetic relationships with related species. The results of our study will aid in the decision on whether these species should become a part of nematode management programs since nontarget species can become pests due to changes in cropping patterns or climate.

Deciphering Bacteriophage T5 Host Recognition Mechanism and Infection Trigger.

Bacteriophages, viruses infecting bacteria, recognize their host with high specificity, binding to either saccharide motifs or proteins of the cell wall of their host. In the majority of bacteriophages, this host recognition is performed by receptor binding proteins (RBPs) located at the extremity of a tail. Interaction between the RBPs and the host is the trigger for bacteriophage infection, but the molecular details of the mechanisms are unknown for most bacteriophages. Here, we present the electron cryomicroscopy (cryo-EM) structure of bacteriophage T5 RBPpb5 in complex with its Escherichia coli receptor, the iron ferrichrome transporter FhuA. Monomeric RBPpb5 is located at the extremity of T5's long flexible tail, and its irreversible binding to FhuA commits T5 to infection. Analysis of the structure of RBPpb5 within the complex, comparison with its AlphaFold2-predicted structure, and its fit into a previously determined map of the T5 tail tip in complex with FhuA allow us to propose a mechanism of transmission of the RBPpb5 receptor binding to the straight fiber, initiating the cascade of events that commits T5 to DNA ejection. IMPORTANCE Tailed bacteriophages specifically recognize their bacterial host by interaction of their receptor binding protein(s) (RBPs) with saccharides and/or proteins located at the surface of their prey. This crucial interaction commits the virus to infection, but the molecular details of this mechanism are unknown for the majority of bacteriophages. We determined the structure of bacteriophage T5 RBPpb5 in complex with its E. coli receptor, FhuA, by cryo-EM. This first structure of an RBP bound to its protein receptor allowed us to propose a mechanism of transmission of host recognition to the rest of the phage, ultimately opening the capsid and perforating the cell wall and, thus, allowing safe channeling of the DNA into the host cytoplasm.

Spirocamallanus delirae n. sp. (Nematoda, Camallanidae), a parasite of Propimelodus eigenmanni (Siluriformes: Pimelodidae) from the Brazilian Amazon.

A new species of Spirocamallanus Olsen, 1952 (Camallanidae), is described based on specimens found parasitizing Propimelodus eigenmanni (Siluriformes), an Amazonian fish. The new species has buccal capsule with 15-19 spiral ridges, males with caudal alae supported by 3 pairs of precloacal and 6 pairs of postcloacal papillae, and two terminal spikes on tail end, and females with a digitiform projection ending in two small spikes on tail end. The new species shares characteristics with a group of Neotropical Spirocamallanus that are characterized by the presence of caudal alae, 3 pairs of precloacal papillae, unequal spicules and the presence of spines on the tail tip of males. Several species of this group have been reported from marine environments, in contrast with Spirocamallanus delirae n. sp., which occurs in Amazon freshwaters. Among the Brazilian congeners placed in the referred morphological group are S. rarus, S. freitasi, S. macaensis, and S. halithophus, the last two being parasites of marine fishes. The new species differs from S. rarus (3-4), S. macaensis (12-15), and S. halithophus (10-15) based on the number of spirals in the buccal capsule and from S. fretasi based on the morphology of the larger spicule that is bifid in the former and undivided in the new species. The present description of Spirocamallanus delirae n. sp. adds new data to the biodiversity of parasites from freshwater siluriform catfish in the Neotropical region.

Experimental Assessment of Aero-Propulsive Effects on a Large Turboprop Aircraft with Rear-Engine Installation

This paper deals with the estimation of propulsive effects for a three-lifting surface turboprop aircraft concept, with rear engine installation at the horizontal tail tips, conceived to carry up to 130 passengers. This work is focused on how the propulsive system affects the horizontal tailplane aerodynamics and, consequently, the aircraft’s static stability characteristics using wind tunnel tests. Both direct and indirect propulsive effects have been estimated. The former produces moments whose values depend on the distance from the aircraft’s centre of gravity to the thrust lines and propeller disks. The latter entails a change in the angle of attack and an increment of dynamic pressure on the tailplane. Several tests were also performed on the body-empennage configuration to investigate the propulsive effects on the aircraft’s static stability without the appearance of any aerodynamic interference phenomena, especially from the canard. The output of the experimental campaign reveals a beneficial effect of the propulsive effects on the aircraft’s longitudinal stability, with an increase in the stability margin of about 2.5% and a reduction in the directional stability derivative of about 4%, attributed to the different induced drag contributions of the two horizontal tail semi-planes. Moreover, the rolling moment coefficient experiences a greater variation due to the propulsion depending on the propeller rotation direction. The outcomes of this paper allow the enhancement of the technical readiness level for the considered aircraft, giving clear indications about the feasibility of the aircraft configuration.

The novel gene, mtre-1 , is expressed downstream of MAB-3 and DMD-3 in the male tail tip at the termination of male tail tip retraction.

The development of the adult C. elegans male tail involves an extensive remodeling during the last larval stage where the pointed tail of the L4 male is converted into the blunt-ended adult tail with its collection of mechano-sensitive rays. The first step in this remodeling is the retraction of the four hypodermal cells of the tail tip to generate the blunt-ended tail. Male tail tip retraction is an excellent model for characterizing how upstream regulatory networks interact with the downstream cell biological effectors that drive morphogenetic changes in all animals. Previously, we've shown that two DM-domain transcription factors, MAB-3 and DMD-3 , are central regulators of male tail tip retraction. Using a microarray-based approach we have identified ~400 genes that are more highly expressed in the L4 male tail tip relative to the hermaphrodite L4 tail tip. The uncharacterized gene T05H10.3 , which we've named mtre-1 , was highly over-represented in the male tail tip vs. the hermaphrodite tail tip and was under-represented in mab-3 ; dmd-3 mutant male tail tips vs. wild-type male tail tips. A transcriptional reporter for mtre-1 shows clear expression in the male tail tip cells for a short period (~3 hours) at the end of retraction. This expression is dependent on the activity of MAB-3 and DMD-3 , since expression is reduced in dmd-3 single mutant males and absent in mab-3 ; dmd-3 mutant males. Finally, males homozygous for a putative null allele of mtre-1 display a phenotypically wild-type adult male tail, indicating that mtre-1 is not essential for male tail morphogenesis.

Quantifying the Over-retraction (Ore) phenotype in C. elegans male tail tip morphogenesis.

Morphometrics, the quantitative analysis of biological structures, reduces subjectivity and increases reproducibility in characterizing morphological phenotypes. In C. elegans males, the rounded adult tail tip emerges from a stage-specific retraction of epidermal cells regulated by the heterochronic pathway via LIN-41/TRIM71. Precocious tail tip morphogenesis in lin-41 reduction-of-function conditions results in a blunted tail (Ore) phenotype, previously described qualitatively (Del Rio-Albrechtsen et al., 2006). We present a quantitative method to assess the Ore phenotype by measuring the tail tip position relative to the cloacal opening. This method can be used to study variation in Ore phenotypes and to validate lin-41 loss-of-function reagents.

Redescriptions of Spinitectus acipenseri and S. micracanthus (Nematoda, Cystidicolidae), with notes on the taxonomy of Spinitectus-like nematodes parasitising North American fishes.

Based on light microscopical and scanning electron microscopical (SEM) examinations, two North American species of Spinitectus Fourment, 1884, S. acipenseri Choudhury & Dick, 1992 and S. micracanthus Christian, 1972 (Nematoda, Cystidicolidae) are redescribed from museum voucher specimens (S. acipenseri) and those newly collected from centrarchid and some other fishes in the Upper San Marcos River in Texas and the Santee River in South Carolina, USA. The first use of SEM to study S. acipenseri, a parasite of lake sturgeon Acipenser fulvescens Rafinesque (Acipenseridae) in Canada, made it possible to describe dorsal and ventral lips, amphids and sublabia, and the presence of a dorsal barb on the right spicule, which was confirmed to be the most characteristic feature of this species. The SEM study of S. micracanthus, a parasite mainly of centrarchids, enabled us to correctly determine the location of the excretory pore in relation to rings of cuticular spines in the male, and to describe the exact structure of the tip of the male tail, sublabia, phasmids and the presence of a median ventral protuberance on the male tail. Some taxonomic problems of North American species of Spinitectus are discussed. Filaria serrata Linton, 1901 is considered a junior synonym of S. oviflagellis Fourment, 1884. To date, there are 13 valid species of Spinitectus parasitising fishes in North America. Keys to species of Spinitectus-like nematodes from fishes in North American waters are provided.

Description of Aphelenchoides persicus sp. n. (Nematoda: Aphelenchoididae) from Iran

Abstract During a survey on the soil nematodes, a population of the genus Aphelenchoides was collected around the rhizosphere of persimmon in Guilan Province, Iran. The morphological and molecular characters confirmed the new species, namely A. persicus sp. n. The new species is characterized by a female body length (699–1068 μm), lip region offset from the rest of the body by a slight constriction, lateral fields with six incisures, stylet 12–13.5 μm long, with a clear basal swelling, excretory pore ca 1.5 metacorpal length posterior to base of the metacorpus, post uterine sac elongate, about 4–7 times than the vulval body diameter; conical female tail with a single centrally located mucron with tiny projection close to the tail tip, male body length (663–908 μm), and spicule well developed with rounded condylus, blunt conical rostrum, and a hook-like tip of dorsal limb. The new species belongs to the Group 2 category of Aphelenchoides species and was similar to seven known species with six lateral field incisures, including A. allius, A. chinensis, A. meghalayensis, A. nechaleos, A. paranechaleos, A. parasexlineatus, and A. sexlineatus. The molecular phylogeny based on 28S rDNA revealed that the new species stands close to A. hamospiculatus (MN931591; MN931592) and two unidentified Aphelenchoides (KY769057; LC583315). The measurements, line illustrations, LM photographs, and phylogenetic analysis are given for the new species.

Structure and host specificity of Staphylococcus epidermidis bacteriophage Andhra.

Staphylococcus epidermidis is an opportunistic pathogen of the human skin, often associated with infections of implanted medical devices. Staphylococcal picoviruses are a group of strictly lytic, short-tailed bacteriophages with compact genomes that are attractive candidates for therapeutic use. Here, we report the structure of the complete virion of S. epidermidis-infecting phage Andhra, determined using high-resolution cryo-electron microscopy, allowing atomic modeling of 11 capsid and tail proteins. The capsid is a T = 4 icosahedron containing a unique stabilizing capsid lining protein. The tail includes 12 trimers of a unique receptor binding protein (RBP), a lytic protein that also serves to anchor the RBPs to the tail stem, and a hexameric tail knob that acts as a gatekeeper for DNA ejection. Using structure prediction with AlphaFold, we identified the two proteins that comprise the tail tip heterooctamer. Our findings elucidate critical features for virion assembly, host recognition, and penetration.

Determining epigenetic memory in kidney proximal tubule cell derived induced pluripotent stem cells using a quadruple transgenic reprogrammable mouse

The majority of nucleated somatic cells can be reprogrammed to induced pluripotent stem cells (iPSCs). The process of reprogramming involves epigenetic remodelling to turn on pluripotency-associated genes and turn off lineage-specific genes. Some evidence shows that iPSCs retain epigenetic marks of their cell of origin and this “epigenetic memory” influences their differentiation potential, with a preference towards their cell of origin. Here, we reprogrammed proximal tubule cells (PTC) and tail tip fibroblasts (TTF), from a reprogrammable mouse to iPSCs and differentiated the iPSCs to renal progenitors to understand if epigenetic memory plays a role in renal differentiation. This model allowed us to eliminate experimental variability due to donor genetic differences and transfection of the reprogramming factors such as copy number and integration site. In this study we demonstrated that early passage PTC iPSCs and TTF iPSCs expressed low levels of renal progenitor genes and high levels of pluripotency-associated genes, and the transcriptional levels of these genes were not significantly different between PTC iPSCs and TTF iPSCs. We used ChIP-seq of H3K4me3, H3K27me3, H3K36me3 and global DNA methylation profiles of PTC iPSCs and TTF iPSCs to demonstrate that global epigenetic marks were not different between the cells from the two different sets of tissue samples. There were also no epigenetic differences observed when kidney developmental genes and pluripotency-associated genes were closely examined. We did observe that during differentiation to renal progenitor cells the PTC iPSC-derived renal cells expressed higher levels of three renal progenitor genes compared to progenitors derived from TTF iPSCs but the underlying DNA methylation and histone methylation patterns did not suggest an epigenetic memory basis for this.

Description and molecular characterisation of Ditylenchus pedrami n. sp. (Rhabditida: Anguinidae) from Iran

Summary Ditylenchus pedrami n. sp., recovered from the rhizospheric soil of date palm in Khuzestan province, southwest Iran, is described and illustrated based upon morphological and molecular data. The new species is characterised by having six lines in the lateral field, lip region smooth and continuous with body contour, stylet 9-11 μm long, median pharyngeal bulb oval with small valve, pharyngeal bulb offset from the intestine, V = 83.9 (80.2-88.1), conoid tail with a pointed, dull or rounded tip and males with 20.1 (17-24) μm long spicules. Morphologically, the new species comes close to D. africanus, D. anchilisposomus, D. australiae, D. clarus, D. clavicaudatus and D. parcevivens, mainly by having shared features like six lines in the lateral fields, stylet length and somewhat similar tail tip. The new species was also compared with D. stenurus and D. sarvarae, two species with close phylogenetic affinities to it. The phylogenetic relationships of the new species with representatives of the family Anguinidae were reconstructed and discussed using partial sequences of the small subunit, D2-D3 expansion segments of the large subunit, and internal transcribed spacer regions of ribosomal DNA (SSU, LSU D2-D3 and ITS rDNA) based on Bayesian inference (BI).
