Research on the Theoretical Framework, Spatio-temporal Laws, and Driving Mechanism of Beautiful Human Settlements—A Case Study of the 14 Prefecture-Level Cities in Liaoning Province

The existing research on Beautiful China focuses on the major strategic research that is horizontal and comprehensive, and it is urgent to conduct a vertical and in-depth study from the important focus of human settlements. This paper proposes the theoretical framework of “five-sphere integrated plan”, “three-state entirety”, and “binary fusion” of beautiful human settlements, and integrates the entropy power method, kernel density estimation method, geographic probe, and spatial analysis to study the spatio-temporal Laws and the driving mechanism of beautiful human settlements in Liaoning Province. The results show that: (1) In terms of time course, from 2009 to 2018, the beautiful human settlement in Liaoning showed a fluctuating upward trend, and there were more obvious stage divergence characteristics and discrete characteristics; over time, they clustered from low level to high level. (2) In terms of spatial pattern, beautiful human settlements in general showed regional heterogeneous characteristics. The construction in western Liaoning is relatively stable, while northern Liaoning has changed greatly. There is an obvious inverted “U” spatial structure in the province, and it was both higher in the north and south and lower in the east and west, specifically decreasing from southeast to northwest. The distribution pattern is a “dicaryon” dominated by Shenyang and Dalian, and a “triad” trend of core area, development area, and starting area. (3) System properties, such as environmental, support, and social systems show fluctuating upward trends, while systems such as population show downward trends with different discrete characteristics. (4) Driving mechanisms, social systems, and support systems are the main driving systems, and it is also driven by a combination of urban economic strength, population quantity and quality, infra-structure development such as housing, and emerging technology development. The purpose of studying the beautiful human settlements is to promote the integration and development of the sciences of human settlements and other cross disciplines and to clarify the focus of Liaoning local government on building a Beautiful China.

Public awareness for “classic” childhood diseases and inflammatory syndromes in children during the COVID-19 pandemic

ObjectivesThe objective was to analyze in silico public search interest during the COVID-19 pandemic for some classic infectious childhood diseases, e.g., measles, mumps, chickenpox, scarlet fever, and inflammatory diseases like Kawasaki disease and the pediatric inflammatory multisystem syndrome (PIMS). Study designIn this study, a comparison of five childhood diseases in public search trends with the pediatric inflammatory multisystem syndrome was performed. MethodsGoogle Trends data for the period of five years for six childhood diseases were used. We used topics coverings all languages worldwide and all connected search queries. ResultsPublic search interest decreased during the COVID-19 pandemic for some classic infectious childhood diseases. Search interest for the pediatric inflammatory multisystem syndrome, despite strong indication of a connection with COVID-19, remained relatively low compared to Kawasaki disease. Practice implicationsBetter understanding of Google Trends can map public awareness of childhood diseases in terms of time course and search intensity. ConclusionsPublic interest during the pandemic was generated for diseases with suspected connection to COVID-19, presumably due to media triggers.

Differential uptake of three clinically relevant allergens by human plasmacytoid dendritic cells

BackgroundHuman plasmacytoid dendritic cells (pDC) have a dual role as interferon-producing and antigen-presenting cells. Their relevance for allergic diseases is controversial. and the impact of pDC on allergic immune responses is poorly understood.MethodsThis in vitro study on human pDC isolated from peripheral blood was designed to compare side by side the uptake of three clinically relevant representative allergens: fluorochrome-labeled house dust mite Der p 1, Bee venom extract from Apis mellifera (Api) and the food allergen OVA analyzed flow cytometry and confocal microscopy.ResultsWe found that the internalization and its regulation by TLR9 ligation was significantly different between allergens in terms of time course and strength of uptake. Api and OVA uptake in pDC of healthy subjects was faster and reached higher levels than Der p 1 uptake. CpG ODN 2006 suppressed OVA uptake and to a lesser extent Der p 1, while Api internalization was not affected. All allergens colocalized with LAMP1 and EEA1, with Api being internalized particularly fast and reaching highest intracellular levels in pDC. Of note, we could not determine any specific differences in antigen uptake in allergic compared with healthy subjects.ConclusionsTo our knowledge this is the first study that directly compares uptake regulation of clinically relevant inhalative, injective and food allergens in pDC. Our findings may help to explain differences in the onset and severity of allergic reactions as well as in the efficiency of AIT.

Generation and Characterization of Immortalized Mouse Cortical Astrocytes From Wildtype and Connexin43 Knockout Mice.

We transduced mouse cortical astrocytes cultured from four litters of embryonic wildtype (WT) and connexin43 (Cx43) null mouse pups with lentiviral vector encoding hTERT and measured expression of astrocyte-specific markers up to passage 10 (p10). The immortalized cell lines thus generated (designated IWCA and IKOCA, respectively) expressed biomarkers consistent with those of neonatal astrocytes, including Cx43 from wildtype but not from Cx43-null mice, lack of Cx30, and presence of Cx26. AQP4, the water channel that is found in high abundance in astrocyte end-feet, was expressed at moderately high levels in early passages, and its mRNA and protein declined to low but still detectable levels by p10. The mRNA levels of the astrocyte biomarkers aldehyde dehydrogenase 1L1 (ALDH1L1), glutamine synthetase (GS) and glial fibrillary acidic protein (GFAP) remained relatively constant during successive passages. GS protein expression was maintained while GFAP declined with cell passaging but was still detectable at p10. Both mRNA and protein levels of glutamate transporter 1 (GLT-1) declined with passage number. Immunostaining at corresponding times was consistent with the data from Western blots and provided evidence that these proteins were expressed at appropriate intracellular locations. Consistent with our goal of generating immortalized cell lines in which Cx43 was either functionally expressed or absent, IWCA cells were found to be well coupled with respect to intercellular dye transfer and similar to primary astrocyte cultures in terms of time course of junction formation, electrical coupling strength and voltage sensitivity. Moreover, barrier function was enhanced in co-culture of the IWCA cell line with bEnd.3 microvascular endothelial cells. In addition, immunostaining revealed oblate endogenous Cx43 gap junction plaques in IWCA that were similar in appearance to those plaques obtained following transfection of IKOCA cells with fluorescent protein tagged Cx43. Re-expression of Cx43 in IKOCA cells allows experimental manipulation of connexins and live imaging of interactions between connexins and other proteins. We conclude that properties of these cell lines resemble those of primary cultured astrocytes, and they may provide useful tools in functional studies by facilitating genetic and pharmacological manipulations in the context of an astrocyte-appropriate cellular environment.

Keeping up with the clinical advances: tardive dyskinesia.

Tardive dyskinesia (TD) was first described in 1964, but treatment for this sometimes poorly characterized condition lagged decades as it was labored by medico-legal implications. TD has often been lumped with other medication-induced disorders and incorrectly classified as extrapyramidal symptoms. TD is likely to be under-recognized for many of these reasons. Though diverse in its presentations, TD is distinct in terms of time course, pathophysiology, and phenomenology.

Describing Events: Changes in Eye Movements and Language Production Due to Visual and Conceptual Properties of Scenes.

How can a visual environment shape our utterances? A variety of visual and conceptual factors appear to affect sentence production, such as the visual cueing of patients or agents, their position relative to one another, and their animacy. These factors have previously been studied in isolation, leaving the question about their interplay open. The present study brings them together to examine systematic variations in eye movements, speech initiation and voice selection in descriptions of visual scenes. A sample of 44 native speakers of German were asked to describe depicted event scenes presented on a computer screen, while both their utterances and eye movements were recorded. Participants were instructed to produce one-sentence descriptions. The pictures depicted scenes with animate agents and either animate or inanimate patients who were situated to the right or to the left of agents. Half of the patients were preceded by a visual cue – a small circle appearing for 60 ms on a blank screen in the place of patients. The results show that scenes with left- rather than right-positioned patients lead to longer speech onset times, a higher probability of passive sentences and looks toward the patient. In addition, scenes with animate patients received more looks and elicited more passive utterances than scenes with inanimate patients. Visual cueing did not produce significant changes in speech, even though there were more looks to cued vs. non-cued referents, demonstrating that cueing only impacted initial scene scanning patterns but not speech. Our findings demonstrate that when examined together rather than separately, visual and conceptual factors of event scenes influence different aspects of behavior. In comparison to cueing that only affected eye movements, patient animacy also acted on the syntactic realization of utterances, whereas patient position in addition altered their onset. In terms of time course, visual influences are rather short-lived, while conceptual factors have long-lasting effects.

The posterior semantic asymmetry (PSA): specific to written not auditory semantic word processing

The present study replicates the finding of a posterior semantic asymmetry (PSA; Koppehele-Gossel et al., Brain Lang 157-158:35-43, 2016), a lateralized event-related potential (ERP) suggested to reflect semantic activation from visually presented single words. This ERP negativity, derived from the subtraction of right-side from left-side scalp activity, again peaked around 300ms at temporoparietal electrodes and was more pronounced in a semantic task, compared to both a silent naming task and a passive viewing task. With analogous tasks, no comparable negativity was found for auditorily presented words. This suggests that the PSA specifically reflects visual-verbal semantic activation. For auditory words, a later variation with the demands on semantic processing was observed for a left-lateralized late positive potential (500-800ms), which, however, showed a remarkably similar topography as the PSA. Thus, while semantic processing of visual and auditory words converges on left temporoparietal brain areas, the exact patterns of brain electrical activation in terms of time course and polarity are different.

Metabotropic Acetylcholine and Glutamate Receptors Mediate PI(4,5)P2 Depletion and Oscillations in Hippocampal CA1 Pyramidal Neurons in situ

The sensitivity of many ion channels to phosphatidylinositol-4,5-bisphosphate (PIP2) levels in the cell membrane suggests that PIP2 fluctuations are important and general signals modulating neuronal excitability. Yet the PIP2 dynamics of central neurons in their native environment remained largely unexplored. Here, we examined the behavior of PIP2 concentrations in response to activation of Gq-coupled neurotransmitter receptors in rat CA1 hippocampal neurons in situ in acute brain slices. Confocal microscopy of the PIP2-selective molecular sensors tubbyCT-GFP and PLCδ1-PH-GFP showed that pharmacological activation of muscarinic acetylcholine (mAChR) or group I metabotropic glutamate (mGluRI) receptors induces transient depletion of PIP2 in the soma as well as in the dendritic tree. The observed PIP2 dynamics were receptor-specific, with mAChR activation inducing stronger PIP2 depletion than mGluRI, whereas agonists of other Gαq-coupled receptors expressed in CA1 neurons did not induce measureable PIP2 depletion. Furthermore, the data show for the first time neuronal receptor-induced oscillations of membrane PIP2 concentrations. Oscillatory behavior indicated that neurons can rapidly restore PIP2 levels during persistent activation of Gq and PLC. Electrophysiological responses to receptor activation resembled PIP2 dynamics in terms of time course and receptor specificity. Our findings support a physiological function of PIP2 in regulating electrical activity.

Does the Processing of Sensory and Reward-Prediction Errors Involve Common Neural Resources? Evidence from a Frontocentral Negative Potential Modulated by Movement Execution Errors

In humans, electrophysiological correlates of error processing have been extensively investigated in relation to decision-making theories. In particular, error-related ERPs have been most often studied using response selection tasks. In these tasks, involving very simple motor responses (e.g., button press), errors concern inappropriate action-selection only. However, EEG activity in relation to inaccurate movement-execution in more complex motor tasks has been much less examined. In the present study, we recorded EEG while volunteers performed reaching movements in a force-field created by a robotic device. Hand-path deviations were induced by interspersing catch trials in which the force condition was unpredictably altered. Our goal was twofold. First, we wanted to determine whether a frontocentral ERP was elicited by sensory-prediction errors, whose amplitude reflected the size of kinematic errors. Then, we explored whether common neural processes could be involved in the generation of this ERP and the feedback-related negativity (FRN), often assumed to reflect reward-prediction errors. We identified a frontocentral negativity whose amplitude was modulated by the size of the hand-path deviations induced by the unpredictable mechanical perturbations. This kinematic error-related ERP presented great similarities in terms of time course, topography, and potential source-location with the FRN recorded in the same experiment. These findings suggest that the processing of sensory-prediction errors and the processing of reward-prediction errors could involve a shared neural network.

Neural Dynamics of Phonological Processing in the Dorsal Auditory Stream

Neuroanatomical models hypothesize a role for the dorsal auditory pathway in phonological processing as a feedforward efferent system (Davis and Johnsrude, 2007; Rauschecker and Scott, 2009; Hickok et al., 2011). But the functional organization of the pathway, in terms of time course of interactions between auditory, somatosensory, and motor regions, and the hemispheric lateralization pattern is largely unknown. Here, ambiguous duplex syllables, with elements presented dichotically at varying interaural asynchronies, were used to parametrically modulate phonological processing and associated neural activity in the human dorsal auditory stream. Subjects performed syllable and chirp identification tasks, while event-related potentials and functional magnetic resonance images were concurrently collected. Joint independent component analysis was applied to fuse the neuroimaging data and study the neural dynamics of brain regions involved in phonological processing with high spatiotemporal resolution. Results revealed a highly interactive neural network associated with phonological processing, composed of functional fields in posterior temporal gyrus (pSTG), inferior parietal lobule (IPL), and ventral central sulcus (vCS) that were engaged early and almost simultaneously (at 80-100 ms), consistent with a direct influence of articulatory somatomotor areas on phonemic perception. Left hemispheric lateralization was observed 250 ms earlier in IPL and vCS than pSTG, suggesting that functional specialization of somatomotor (and not auditory) areas determined lateralization in the dorsal auditory pathway. The temporal dynamics of the dorsal auditory pathway described here offer a new understanding of its functional organization and demonstrate that temporal information is essential to resolve neural circuits underlying complex behaviors.

An electrophysiological index of changes in risk decision-making strategies

Human decision-making is significantly modulated by previously experienced outcomes. Using event-related potentials (ERPs), we examined whether ERP components evoked by outcome feedbacks could serve as biomarkers to signal the influence of current outcome evaluation on subsequent decision-making. In this study, 18 adult volunteers participated in a simple monetary gambling task, in which they were asked to choose between two options that differed in risk. Their decisions were immediately followed by outcome presentation. Temporospatial principle component analysis (PCA) was applied to the outcome-onset locked ERPs in the 200–1000ms time window. The PCA factors that approximated classical ERP components (P2, feedback-related negativity, P3a, and P3b) in terms of time course and scalp distribution were tested for their association with subsequent decision-making strategies. Our results revealed that a fronto–central PCA factor approximating the classical P3a was related to changes of decision-making strategies on subsequent trials. The decision to switch between high- and low-risk options resulted in a larger P3a relative to the decision to retain the same choice. According to the results, we suggest that the amplitude of the fronto–central P3a is an electrophysiological index of the influence of current outcome on subsequent risk decision-making. Furthermore, the ERP source analysis indicated that the activations of the frontopolar cortex and sensorimotor cortex were involved in subsequent changes of strategies, which enriches our understanding of the neural mechanisms of adjusting decision-making strategies based on previous experience.

Acoustic and musical features of emotional response to orchestral gestures

Recent empirical research indicates the impact of prominent changes in instrumentation on the listening experience: several studies suggest that timbral changes evoke music-induced emotions. However, orchestration remains an underdeveloped area of music theory. A model of orchestral gestures defined by changes in instrumentation in terms of time course (gradual or sudden) and direction (addition or reduction) is presented. An exploratory behavioral study that tested the perceptual relevance of orchestral gestures on listeners’ continuous ratings of emotional intensity was conducted. We demonstrate a new type of visualization that illustrates the relative textural density of each instrument family over time combined with other time-varying parameters extracted from the signal (loudness, spectral centroid, tempo, and roughness) and calculated from the score (instrumental texture, melodic range, and attack density). Inhibition of return and schizophrenia: A meta-analysis

Inhibition of return (IOR) is a phenomenon that involves inhibited or delayed orienting to previously cued locations in favor of attending to novel locations. To date, research on IOR in patients with schizophrenia has generated mixed, and seemingly conflicting, results. Some researchers report patients with schizophrenia exhibit blunted or delayed IOR, while other researchers report normal IOR, in terms of time course and magnitude. This meta-analysis summarizes the literature that has employed an IOR task in patients with schizophrenia and with controls while focusing upon a procedural feature, the use of a cue back to fixation, between the cue and target that is known to be important when executive control has been hampered in non-clinical populations. Fifteen experiments were located yielding a total sample of 362 patients with schizophrenia or schizoaffective disorder and 285 controls. Using a meta-analytic approach, results of the present analyses show that patients with schizophrenia demonstrate delayed IOR in the single cue procedure. In the cue back to fixation procedure, the time course of IOR among patients is more consistent with that of controls. Differences in measured IOR between patients with schizophrenia and controls are largely related to a deficit in endogenous disengagement of attention.

Understanding hERG Channels Gating using Voltage-Clamp Fluorimetry

hERG channels encode the alpha-subunit that underlies the rapidly activating delayed rectifier K+ current (IKr) in the heart. These proteins play a central role in cardiac action potential repolarization. hERG channel is a tetramer of four subunits, each containing six segments, S1-S6, and a re-entrant P-loop with a pore helix. The importance of hERG channels resides in their unusual gating, that are slow activation and deactivation, and much faster inactivation/recovery from inactivation.One debated and undetermined issue is whether S4 movement in hERG channels is slow, or if its coupling to pore opening is. Previous studies have suggested that slow activation may be due to slow movement of the S4 segments, however, we recently showed that such movement is actually fast, but that a secondary conformational rearrangement, related to the pore, is directly connected to the slow kinetics of activation/deactivation of hERG channels1.To get an overall picture of the conformational changes that occur in the whole channel throughout its activation/deactivation mechanisms, we performed a fluorescence scan of the S4 segment, the S1-S2 and S3-S4 linkers using voltage-clamp fluorimetry technique. Results show different profiles of fluorescence depending on the location where the fluorescent dye is attached. But interestingly, two profiles seem to be highlighted: 1) minor fast fluorescence signals with voltage-dependence that correlates well with S4 segment movement, and 2) major slow fluorescence signals that consistently relate to pore opening/G-V in terms of time course and voltage-dependence, leading to the conclusion that a concerted channel rearrangement is responsible for or resultant of slow opening in hERG channels.Our data suggest that re-assessment of mechanisms underlying hERG activation gating is required, particularly with regard to the coupling mechanisms between voltage sensor movement and channel pore opening.1 Es-Salah-Lamoureux et al., 2010, PLoS One 28;5(5):e10876

Pain characteristics after laparoscopic inguinal hernia repair

Previous studies have shown different pain characteristics in different types of laparoscopic operations, but pain pattern has not been studied in detail after laparoscopic inguinal hernia repair. To optimise preoperative patient information and postoperative analgesic treatment the present study investigated postoperative pain in terms of time course, pain intensity and individual pain components during the first 4 days after transabdominal preperitoneal hernia repair (TAPP). The study was a single-centre prospective trial including 50 consecutive male patients with primary or recurrent inguinal and/or femoral hernia undergoing elective repair. Several pain components (visceral pain, incisional pain and shoulder pain) were studied as well as pain at the three different trocar incisions. Pain was recorded before operation and on a daily basis during the first four postoperative days using a visual analogue and a verbal rating scale. We used 1 × 12 and 2 × 5 mm trocars for TAPP. A total of 46 patients were available for analyses (age median 58 years, range 27-69 years). Visceral pain dominated significantly compared with incisional pain (P < 0.01), which again dominated over shoulder pain intensity and incidence (P < 0.01). Pain intensity did not significantly differ between different trocar incisions (5 and 12 mm) (P > 0.05). The overall pain intensity (a conglomerate of the different pain components) was most intense 3 h after TAPP and declined to preoperative levels on day 3 (P > 0.5). Pain was most intense 3 h after the operation and declined to low levels within the first 3 days. Visceral pain was by far the dominating pain component compared with incisional and shoulder pain.

Four Common Conceptual Fallacies in Mapping the Time Course of Recognition

Determining the moment at which a visual recognition process is completed, or the order in which various processes come into play, are fundamental steps in any attempt to understand human recognition abilities, or to replicate the corresponding hierarchy of neuronal mechanisms within artificial systems. Common experimental paradigms for addressing these questions involve the measurement and/or comparison of backward-masking (or rapid serial visual presentation) psychometric functions and of physiological EEG/MEG/LFP signals (peak latencies, differential activities, single-trial decoding techniques). I review and illustrate four common mistakes that scientists tend to make when using these paradigms, and explain the conceptual fallacies that motivate their reasoning. First, contrary to collective intuition, presentation times, or stimulus-onset asynchrony masking thresholds cannot be taken to reflect, directly or indirectly, the timing of relevant brain processes. Second, psychophysical or electrophysiological measurements should not be compared without assessing potential physical differences between experimental stimulus sets. Third, such comparisons should not be performed in any manner contingent on subjective responses, so as to avoid response biases. Last, the filtering of electrophysiological signals alters their temporal structure, and thus precludes their interpretation in terms of time course. Practical solutions are proposed to overcome these common mistakes.

Synthesis of controlled polymeric cross-linked coatings via iniferter polymerisation in the presence of tetraethyl thiuram disulphide chain terminator

A “grafting from” approach has been used for controlled deposition of cross-linked polymers by living radical polymerisation. Borosilicate glass was modified with N, N-diethylaminodithiocarbamoylpropyl(trimethoxy)silane, in order to confine the iniferter reactive groups solely at its surface, then placed in solution with monomers and cross-linker. The polymerisation was initiated by UV irradiation. Formation of the cross-linked polymers was studied in terms of time course of the reaction, type of monomers incorporated and influence of oxygen. Grafted surfaces were characterised by AFM, FT-IR, ellipsometry and contact angle measurements. The ability to control the grafted layer improved dramatically when the chain terminator agent, N, N- N′, N′-tetraethyl thiuram disulphide (TED) was added. Upon irradiation TED increases the concentration of passive capping radicals and decreases the possibility of recombination of active macro-radicals, thus prolonging their lifetime. In the absence of TED the thickness of produced coatings was below 10 nm. TED added at different concentrations assisted in the formation of grafted layers of 10–130 nm thickness. Iniferter chemistry in the presence of TED can be used for growing nanometre-scale polymer layers on solid supports. It constitutes a robust general platform for controlled grafting and offer a general solution to address the needs of surface derivatisation in sensors technology.

The temporal interaction of modality specific and process specific neural networks supporting simple working memory tasks

Several theories of brain function emphasize distinctions between sensory and cognitive systems. We hypothesized, instead, that sensory and cognitive systems interact to instantiate the task at the neural level. We tested whether input modality interacts with working memory operations in that, despite similar cognitive demands, differences in the anatomical locations or temporal dynamics of activations following auditory or visual input would not be limited to the sensory cortices. We recorded event-related brain potentials (ERPs) while participants performed simple short-term memory tasks involving visually or auditorily presented bandpass-filtered noise stimuli. Our analyses suggested that working memory operations in each modality had a very similar spatial distribution of current sources outside the sensory cortices, but differed in terms of time course. Specifically, information for visual processing was updated and held online in a manner that was different from auditory processing, which was done mostly after the offset of the final stimulus. Our results suggest that the neural networks that support working memory operations have different temporal dynamics for auditory and visual material, even when the stimuli are matched in term of discriminability, and are designed to undergo very similar transformations when they are encoded and retrieved from memory.

Chronic but not Acute Dopaminergic Transmission Interruption Promotes a Progressive Increase in Cortical Beta Frequency Synchronization: Relationships to Vigilance State and Akinesia

Dopaminergic (DA) denervation results in the appearance of an excessive cortical beta frequency synchronization in parkinsonian patients and animal models of the disease. The present study analyzed electrocorticogram signals in awake rats to further characterize this excessive synchronization in terms of time course, relation to motor activity and state of vigilance. Using substantia nigra pars compacta lesions and both acute and chronic pharmacological interruptions of DA transmission, the present data demonstrated that the appearance of excessive beta synchronization requires a prolonged interruption in DA transmission and builds up progressively. This synchronization was vigilance-state dependent and observed solely during awake-like activity. Furthermore, these data demonstrated for the first time that the appearance of akinesia preceded the excessive cortical beta synchronization. In addition, this synchronization was stronger in the motor than in the somato-sensory cortex and in unilaterally compared with bilaterally lesioned animals. Finally, excessive beta synchronization was accompanied by an increased coherence between motor and somato-sensory cortical activities.
