Pikeperch (Sander lucioperca) larval rearing optimization: utilization of lactic acid bacteria for improving microbiome diversity and digestive enzyme activity

Lactic acid bacteria (LAB) probiotics were evaluated for their impact on the microbiota and development of pikeperch (Sander lucioperca) larvae during their initial feeding stage (first 21 days). Pikeperch larvae were exposed to LAB probiotics in two ways: (1) via the live-feed only (Treatment 1, live-feed) or (2) via the live-feed and the larval culture water (Treatment 2, probiotic) in comparison to a control group without LAB supplementation. Total length (TL), myomere height (MH), and survival rate were significantly increased in the probiotic compared the Control group. The administration of probiotics significantly positively influenced the microbiome’s diversity. Specifically, the relative abundance of Cytophagales decreased and that of several other taxa increased in both probiotic treatments although differences between treatments became statistically insignificant by day 21. Furthermore, the different treatments had distinct and significant impacts on digestive enzyme development such as protease and lipase, with the most prominent differences occurring at seven days post-hatching (dph). Taken together, these results indicate that the use of LAB in both the live-feed and rearing water has a positive impact on the larvae microbiota and digestive enzyme development in turn positively impacting their development and viability under intensive rearing conditions.

Metagenomic analysis for exploring the potential of Lactobacillus yoelii FYL1 to mitigate bacterial diarrhea and changes in the gut microbiota of juvenile yaks

Diarrhea in calves is a common disease that results in poor nutrient absorption, poor growth and early death which leads to productivity and economic losses. Therefore, it is important to explore the methods to reduce diarrhea in yak's calves. Efficacy of lactic acid bacteria (LAB) for improvement of bacterial diarrhea is well recognized. For this purpose, two different doses (107 CFU, 1011 CFU) of Lactobacillus yoelii FYL1 isolated from yaks were fed to juvenile yaks exposed to E. coli O78. After a trial period of ten days fresh feces and intestinal contents of the experimental yaks were collected and metagenomics sequencing was performed. It was found that feeding a high dose of Lactobacillus yoelii FYL1 decreased abundance of phylum Firmicutes in the E. coli O78 infected group whereas, it was high in animals fed low dose of Lactobacillu yoelii FYL1. Results also revealed that counts of bacteria from the family Oscillospiraceae, genus Synergistes and Megasphaera were higher in control group whereas, order Bifidobacteriales and family Bifidobacteriaceae were higher in infected group. It was observed that bacterial counts for Pseudoruminococcus were significantly (P < 0.05) higher in animals of group that were given high dose of Lactobacillus yoelii FYL1 (HLAB). Compared to infected group multiple beneficial bacterial genera such as Deinococus and Clostridium were found higher in the animals that were given a low dose of Lactobacillus yoelii FYL1 (LLAB). The abundance of pathogenic bacterial genera that included Parascardovia, Bacteroides and Methanobrevibacter was decreased (P < 0.05) in the lower dose treated group. The results of functional analysis revealed that animals of LLAB had a higher metabolism of terpenoids and polyketides compared to animals of infected group. Virus annotation also presented a significant inhibitory effect of LLAB on some viruses (P < 0.05). It was concluded that L. yoelii FYL1 had an improved effect on gut microbiota of young yaks infected with E. coli O78. This experiment contributes to establish the positive effects of LAB supplementation while treating diarrhea.

Fermentation characteristics and microbial community composition of wet brewer's grains and corn stover mixed silage prepared with cellulase and lactic acid bacteria supplementation.

The objective of this study was to investigate how cellulase or/and lactic acid bacteria (LAB) affected the fermentation characteristic and microbial community in wet brewer's grains (WBG) and corn stover (CS) mixed silage. The WBG was mixed thoroughly with the CS at 7:3 (w/w). Four treatment groups were studied: i) CON, no additives; ii) CEL, added cellulase (120 U/g fresh matter [FM]), iii) LAB, added LAB (2×106 cfu/g FM), and iv) CLA, added cellulase (120 U/g FM) and LAB (2×106 cfu/g FM). All additive-treated groups showed higher fermentation quality over the 30 d ensiling period. As these groups exhibited higher (p<0.05) LAB counts and lactic acid (LA) content, along with lower pH value and ammonia-nitrogen (NH3-N) content than the control. Specifically, cellulase-treated groups (CEL and CLA) showed lower (p<0.05) neutral detergent fiber and acid detergent fiber contents than other groups. All additives increased the abundance of beneficial bacteria (Firmicutes, Lactiplantibacillus, and Limosilactobacillus) while they decreased abundance of Proteobacteria and microbial diversity as well. The combined application of cellulase and LAB could effectively improve the fermentation quality and microbial community of the WBG and CS mixed silage.

Influence of lactic-acid bacteria feed supplementation on free amino acid levels in serum and feces of rainbow trout (Oncorhynchus mykiss, Walbaum 1792)

IntroductionThe influence of intestinal microbiota, particularly lactic-acid bacteria, on digestion and nutrient metabolic activities has been extensively studied. In this research, we investigated the effects of supplementing the feed of rainbow trout (O. mykiss) with P. acidilactici or L. lactis probiotics. A Control group without probiotic supplementation was also included. The study aimed to evaluate growth, feed conversion indices, free amino acid levels in serum and feces, and the relative gene expression of amino acid solute carrier transporters (SLC).MethodsRainbow trout weighing 73.9±10 g were fed with a basal commercial diet supplemented with mono-strains of P. acidilactici or L. lactis at a dosage of 5 x 108 CFU per gram of feed for 60 days. The trout were reared in 100-liter tanks with independent closed recirculation systems, with a water replacement rate of 80% of the total volume daily and a controlled temperature of 15 ± 0.3 °C. The proximate composition of the basal diet consisted of crude protein (49.8%), fat (32.1%), fiber (1.31%), ash (13%), and moisture (8.3%) on a dry matter basis. The diet was provided at a rate of 2% of bodyweight per day.ResultsNo significant differences (p &amp;gt; 0.05) were observed in growth and feed conversion indices between the experimental groups. However, significant differences in free amino acid levels in feces and serum samples (p ≤ 0.05) were observed. The P. acidilactici group exhibited significantly higher levels of glutamate, lysine, proline, and tyrosine in feces samples, and higher levels of arginine, iso/leucine, phenylalanine, proline, serine, threonine, and valine in serum samples compared to the Control group. Additionally, the supplemented probiotic groups showed significant downregulation (p ≤ 0.05) of theslc6a19 and slc7a9 genes, which encode transporters for neutral and cationic amino acids.DiscussionThe autochthonous rainbow trout strain, P. acidilactici, demonstrated more pronounced effects on amino acid levels in feces and serum compared to the non-autochthonous L. lactis strain. However, the increased amino acid levels, particularly in the serum, resulting from P. acidilactici supplementation, did not lead to improved trout growth or feed conversion. Nonetheless, lactic-acid bacteria supplementation induced measurable metabolic effects, as evidenced by elevated levels of specific amino acids in the serum and feces.Future research should focus on evaluating these probiotic effects using specialized diets and considering the observed metabolic effects in this study.

Dietary Supplementation of Lactobacillus johnsonii RS-7 Improved Antioxidant and Immune Function of Weaned Piglets.

We investigated the effects of dietary supplementation of lactic acid bacteria on the immune and antioxidant performance of weaned pigs. A total of 128 Duroc × Landrace × Yorkshire piglets weaned on day 28 with an average body weight of 8.95 ± 1.15 kg were selected and randomly divided into four treatment groups according to body weight and sex for a 28-day study. The four dietary treatments were basal diet (CON), and CON with 0.05% (LJ0.05), 0.1% (LJ0.1), and 0.2% (LJ0.2) Lactobacillus johnsonii RS-7, respectively. The lowest feed-to-gain ratio (F:G) was found when LJ0.1 was added to the diet. The addition of compound lactic acid bacteria to the diet increased the concentrations of TP, ALB, IgA, and IgM on day 14 and IgG, IgA, and IgM on day 28 (p < 0.05) in the blood, with trait values greater for pigs fed LJ0.1 than CON pigs (p < 0.05). Concentrations of antioxidants (CAT, T-AOC, MDA, T-SOD, and GSH) in serum, intestinal mucosa, spleen, liver, and pancreas improved. In summary, dietary supplementation of Lactobacillus johnsonii RS-7 improved the antioxidant and immune function of weaned piglets.

High Antioxidant Capacity of Lacticaseibacillus paracasei TDM-2 and Pediococcus pentosaceus TCM-3 from Qinghai Tibetan Plateau and Their Function towards Gut Modulation.

Probiotic supplementation is a key therapeutic strategy for promoting gut health and maintaining gut homeostasis by modulating functional microbiota. In this study, we isolated two lactic acid bacteria (LAB) strains, Pediococcus pentosaceus TCM-3 and Lacticaseibacillus paracasei TDM-2, from Qinghai-Tibetan plateau, and evaluated their probiotic properties and antioxidant bioactivity. In which, TDM-2 had higher T-AOC activity than either TCM-3 or LGG (4.10 μmol/mL vs. 3.68 and 3.53 μmol/mL, respectively, p < 0.05). These strains have shown high antioxidant activity compared to the LAB strains and were found to be acid and bile salt tolerant, confronting the safety issues of antibiotic resistance and the capability of surviving in simulated gastric and intestinal juices. In vitro fermentation experiments with human gut microbiota revealed significant differences in microbial community composition between samples supplemented with TCM-3 and TDM-2 and those without. The addition of these two strains resulted in an enrichment of beneficial taxa, such as the Pediococcus, Lactobacillus, and Clostridium_sensu_strictos at the genus level, and Firmicutes and Proteobacteria at the phylum level. Notably, the TCM-3 group exhibited higher short-chain fatty acid production than the TDM-2 group and untreated controls (acetic acid at 12 h: 4.54 mmol L-1 vs. 4.06 mmol L-1 and 4.00 mmol L-1; acetic acid at 24 h: 4.99 mmol L-1 vs. 4.90 mmol L-1 and 4.82 mmol L-1, p < 0.05). These findings demonstrate that LAB supplementation with high antioxidant capacity and probiotic properties can promote gut health by modulating functional microbiota and is enriching for beneficial taxa. Our study provides guidance for therapeutic strategies that use novel LAB strains to maintain gut homeostasis and functional microbiota modulation.

Geroprotective Effect of Levilactobacillus brevis and Weizmannia coagulans in Caenorhabditis elegans.

The prophylactic use of lactic acid bacteria (LAB) to maintain human health is one of the most important research areas in recent times. LAB supplementation confers a wide range of health benefits to the host, but few studies have focused on their possible role in delaying the aging process. This study explored the health and life-promoting properties of two LAB, Levilactobacillus brevis and Weizmannia coagulans, using the Caenorhabditis elegans model. We found that L. brevis and W. coagulans enhanced the intestinal integrity and intestinal barrier functions without affecting the overall physiological functions of C. elegans. Wild-type worms preconditioned with LAB strains increased their survival under oxidative and thermal stress conditions by reducing intracellular reactive oxygen levels. Live L. brevis and W. coagulans significantly extended the lifespan of C. elegans under standard laboratory conditions independently of dietary restrictions. Genetic and reporter gene expression analysis revealed that L. brevis and W. coagulans extend lifespan via insulin/insulin-like growth factor-1 signaling and the p38 MAPK signaling axis. Furthermore, sirtuin, JNK MAPK, and mitochondrial respiratory complexes were found to be partially involved in W. coagulans-mediated lifespan extension and stress resilience. Preconditioning with LAB ameliorated age-related functional decline in C. elegans and reduced ectopic fat deposition in an NHR-49-dependent manner. Together, our findings indicated that L. brevis and W. coagulans are worth exploring further as "gerobiotic" candidates to delay aging and improve the healthspan of the host.

Supplementation of lactic acid bacteria has positive effects on the mucosal health of Atlantic salmon (Salmo salar) fed soybean meal

We investigated the ability of lactic acid bacteria, when added individually or in combination in feeds, to prevent soybean meal-induced enteritis in Atlantic salmon. A control diet, designed to induce enteritis, was formulated with marine ingredients and 20% soybean meal. Three more diets were produced by coating the control diet with two bacteria, either singly (Lactobacillus plantarum; L. fermentum) or in combination. The fish were fed the abovementioned diets for 38 days. We performed histological assessments and evaluated the expression of selected mucin and antimicrobial peptide genes in the dorsal skin, gills, and distal intestine. Digesta were also collected to study the short chain fatty acids. Feeding bacteria, individually or in combination, altered the short chain fatty acids−acetoacetic acid, lactic acid, succinic acid, propionic acid−and the total fatty acids in the digesta significantly. Of all the determined short chain fatty acids, the concentration of acetoacetic acid was the highest, and the fish fed the combination of the two bacteria had the significantly highest value. Succinic acid was also significantly higher in fish fed the combination compared to the control group and the L. fermentum group. Total fatty acids were significantly higher in fish fed the combination than those fed L. fermentum. Compared to the control and probiotic combination-fed fish, those fed L. plantarum had higher defensin1 expression in the skin. We also observed significantly higher number of gill mucous cells in the fish fed the blend compared to the control group. Lamina propria width was significantly reduced in fish fed the blend. Supra nuclear vacuoles were higher in fish fed the single species or the blend, compared to the control group. Thus, adding the probiotics to a soybean meal diet can elevate the digesta short chain fatty acids and intestine supranuclear vacuoles, and reduce the lamina propria width, which probably indicate prevention of enteritis.

Concomitant Administration of Red Ginseng Extract with Lactic Acid Bacteria Increases the Plasma Concentration of Deglycosylated Ginsenosides in Healthy Human Subjects.

With the increased frequency of red ginseng extract (RGE) and lactic acid bacteria (LAB) co-administration, we aimed to investigate the interactions between RGE and LAB with regard to in vitro and in vivo deglycosylation metabolism and the pharmacokinetics of ginsenosides. As a proof-of-concept study, five healthy humans were administered RGE (104.1 mg of total ginsenosides/day) with or without co-administration of LAB (2 g, 1 billion CFU/day) for 2 weeks, and the plasma concentrations of ginsenosides in human plasma were monitored. The plasma exposure to compound K (CK), ginsenoside Rh2 (GRh2), protopanaxadiol (PPD), and protopanaxatriol (PPT) in the concomitant administration RGE and LAB groups increased by 2.7-, 2.1-, 1.6-, and 3.5-fold, respectively, compared to those in the RGE administration group, without a significant change in Tmax. The plasma concentrations of GRb1, GRb2, and GRc remained unchanged, whereas the AUC values of GRd and GRg3 significantly decreased in the concomitant administration RGE and LAB groups. To understand the underlying mechanism, the in vitro metabolic activity of ginsenosides was measured during the fermentation of RGE or individual ginsenosides in the presence of LAB for 1 week. Consistent with the in vivo results, co-incubation with RGE and LAB significantly increased the formation rate of GRh2, CK, PPD, and PPT. These results may be attributed to the facilitated deglycosylation of GRd and GRg3 and the increased production of GRh2, CK, PPD, and PPT by the co-administration of LAB and RGE. In conclusion, LAB supplementation increased the plasma concentrations of deglycosylated ginsenosides, such as GRh2, CK, PPD, and PPT, through facilitated deglycosylation metabolism of ginsenosides in the intestine.

Fortification of cocoa semi-skimmed milk formulations with native lactic acid bacteria: Cell viability, physicochemical and functional properties for developing novel foods.

This study aimed to evaluate several cocoa semi-skimmed milk formulations (CSMFs) as potential carriers of native lactic acid bacteria (LAB) strains to obtain novel probiotic beverages (PBs) with improved technological and functional characteristics, and satisfactorily organoleptic acceptance. The viability of two native LAB (Lactiplantibacillus plantarum UTNGt2 and Lactiplantibacillus pentosus UTNGt5) was assessed in comparison with two references (Lactococcus lactis subsp. lactis ATCC11474 and Limosilactobacillus reuteri DSM17938) strains in supplemented CSMFs throughout storage with refrigeration. The optimum conditions to produce novel beverages supplemented with native LAB were pH 6.6, 42°C, and 1 h of fermentation. Moreover, the effect of LAB strains fortification on pH, titratable acidity, total solids (°Brix), total polyphenolic compounds (TPC), antioxidant capacity (AOX), and ascorbic acid content (AAC), total proteins and fat, at initial and final storage was evaluated. The addition of two native LAB strains did alter the physicochemical quality of CSMFs to a lesser extent, where the bioactive molecules improved significantly (p < 0.05) with the increase of cocoa concentration and depending on the supplied strain. Although a statistically significant (p < 0.05) decrease in cell counts was recorded during storage, the LAB cells were found to be viable up to 21 days of storage at 4°C (>6 logCFU/ml), which is sufficient in number to prove their stability in vitro. Overall organoleptic results suggested that LAB supplementation had a significant impact on sensory attributes with satisfactory acceptability (>78%) of PBs containing the native strains and 1–2% cocoa, while CSMFs counterparts were less appreciated (40%) as perceived off-flavor. It appears that supplying bacteria to CSMF preserves flavor in the final product. Furthermore, the final beverages were free of harmful bacteria; thus, they comply with consumer safety regulations. This study concludes that CSMF can be used as a carrier of native LAB strains, maintaining cell viability, unaltered physicochemical properties, and improved functional and sensory characteristics, for which final beverages can be regarded as functional food. From the application standpoint, these formulations are an alternative to delivering native LAB strains and could help the cocoa and dairy industry to develop more attractive products for the growing regional market.

Impact of native spray‐dried lactic acid bacteria, packing density and wilting time on fermentation characteristics of experimental maize and lucerne silages

AbstractThis study aimed at investigating the effects of an autochthonous microbial inoculant microencapsulated by spray‐drying on fermentation parameters, microbiological composition, mycotoxin level and aerobic stability of maize and lucerne silages. Experiment 1, a completely randomized design with a 2 × 2 × 4 factorial arrangement of treatments, evaluated inoculation (control or Lactobacillus plantarum Hv75, Pediococcus acidilactici 3903 and Lactobacillus buchneri B463), packing density (148.5 kg dry matter (DM)/m3 or 181.5 kg DM/m3) and storage period (0, 1, 30 or 60 days) in maize silage. Lactic acid bacteria (LAB) supplementation produced higher DM and lower acetic acid and deoxynivalenol contents compared to the control (p &lt; 0.05). However, these differences were small in terms of practical applications. Experiment 2 evaluated effects of microbial inoculation (similar as in experiment 1), wilting time (3 or 21 h) and storage period (0, 3, 30 or 60 days) in lucerne silage. LAB supplementation increased DM and lactic acid content, reduced pH and concentrations of acetic acid, ethanol and ammonia nitrogen/total nitrogen compared to the control (p &lt; 0.05). In addition, inoculation increased total LAB population and decreased yeast and mould counts (p &lt; 0.05). Although the strains displayed potential to be used as a bio‐inoculant at different lucerne moisture levels, prolonged wilting positively influenced DM, crude protein and fibre degradation (p &lt; 0.05). Therefore, the addition of this spray‐dried inoculant to lucerne silages with longer wilting could be recommended for the attainment of silage quality.

Efficacy of lactic acid bacteria supplementation against Fusarium graminearum growth in vitro and inhibition of Zearalenone causing inflammation and oxidative stress in vivo

Fusarium graminearum invasion and Zearalenone (ZEN)-mycotoxin contamination are considered the most global threat to food and feed. This study investigates the effect Lactobacillus plantarum MON03 viable cells (LPVC) and LP free cells supernatant (LPFCS) against Fusarium graminearum growth and ZEN production in vitro and evaluates if treatment with LP viable cells can counteract the negative effect of ZEN on inflammation and oxidative stress in mesenteric lymph nodes and serum biochemical parameters in mice.For the in vitro study, 7 days of LPVC, LPFCS and F. graminearum co-incubation at different concentrations was done in order to determine the antifungal activity and ZEN- production inhibition. Regarding the in vivo study, Balb/c mice were treated as following: Control, ZEN group, LP group and ZEN + LP group for 30 days.In vitro, LPVC showed an excellent antifungal activity after 7 days of co-incubation (103 CFU/ml). LPVC was succeeded also to inhibit ZEN production by the fungi. In vivo, ZEN has shown an important oxidative damage. As a result of the exposure to ZEN, an increase cytokines, as effectors of an inflammatory response, were observed in the mesenteric lymph nodes (MLN) of intoxicated mice. In parallel, a serum biochemical change was also observed. LPVC induced a reduction of ZEN-induced oxidative stress and counteracts also the biochemical parameters damage and the inflammatory markers increased by ZEN.LPVC can be valorized as an anti-cating agent in the vitro and in the gastro-intestinal tract to decrease ZEN-toxic effects.

Effect of Lactic Acid Bacteria on the Pharmacokinetics and Metabolism of Ginsenosides in Mice.

This study aims to investigate the effect of lactic acid bacteria (LAB) on in vitro and in vivo metabolism and the pharmacokinetics of ginsenosides in mice. When the in vitro fermentation test of RGE with LAB was carried out, protopanaxadiol (PPD) and protopanaxadiol (PPD), which are final metabolites of ginsenosides but not contained in RGE, were greatly increased. Compound K (CK), ginsenoside Rh1 (GRh1), and GRg3 also increased by about 30%. Other ginsenosides with a sugar number of more than 2 showed a gradual decrease by fermentation with LAB for 7 days, suggesting the involvement of LAB in the deglycosylation of ginsenosides. Incubation of single ginsenoside with LAB produced GRg3, CK, and PPD with the highest formation rate and GRd, GRh2, and GF with the lower rate among PPD-type ginsenosides. Among PPT-type ginsenosides, GRh1 and PPT had the highest formation rate. The amoxicillin pretreatment (20 mg/kg/day, twice a day for 3 days) resulted in a significant decrease in the fecal recovery of CK, PPD, and PPT through the blockade of deglycosylation of ginsenosides after single oral administrations of RGE (2 g/kg) in mice. The plasma concentrations of CK, PPD, and PPT were not detectable without change in GRb1, GRb2, and GRc in this group. LAB supplementation (1 billion CFU/2 g/kg/day for 1 week) after the amoxicillin treatment in mice restored the ginsenoside metabolism and the plasma concentrations of ginsenosides to the control level. In conclusion, the alterations in the gut microbiota environment could change the ginsenoside metabolism and plasma concentrations of ginsenosides. Therefore, the supplementation of LAB with oral administrations of RGE would help increase plasma concentrations of deglycosylated ginsenosides such as CK, PPD, and PPT.

Sequential Modulation of the Equine Fecal Microbiota and Fibrolytic Capacity Following Two Consecutive Abrupt Dietary Changes and Bacterial Supplementation.

Simple SummaryThe equine hindgut is colonized by microorganisms, some of which are involved in fiber digestion and are crucial for the horse’s nutrition and health. These key microorganisms are very sensitive to dietary changes, which have been identified as a risk factor for colics. This study assessed the stressful effect of two consecutive abrupt dietary changes on the diversity, the composition, and the activity of fecal microorganisms focusing on fibrolytic bacteria. Twelve horses were subjected to an abrupt change from forage to a concentrate-rich diet, followed by a second change from a concentrate to forage-rich diet 5 days later. Half of the horses were given a supplement of living bacteria as a probiotic. Two days after the sudden change from forage to concentrate diet, the proportions and types of microorganisms were altered drastically, as was their capacity to degrade fibers. After this dietary stress, it took 3–4 weeks of a high-fiber diet to recover the basal state. Supplementation with probiotics promoted an earlier recovery of fibrolytic bacteria after the dietary stress. Abrupt dietary changes should be limited in horse management to protect the hindgut microorganisms and their capacity to use forage fibers, and consequently to limit the development of colic.In horses, abrupt changes from high-fiber (HF) to high-starch (HS) diets can affect the cecal and colonic microbiota. This study investigated modifications and recovery of fecal microbiota after two consecutive abrupt dietary changes. Twelve horses fed HF for 2 weeks were changed to HS for 5 days then returned to HF for 7 weeks. Six received lactic acid bacteria supplementation. Bacterial population diversity, structure, and activity, especially fibrolysis, were assessed to obtain an overview of alteration in hindgut microbiota. Two days after the abrupt change from HF to HS, the findings in feces were consistent with those previously reported in the cecum and colon, with a decrease in fibrolytic activity and an increase in amylolytic activity. Fecal parameters stabilized at their basal level 3–4 weeks after the return to HF. A bloom of cellulolytic bacteria and lower pH were observed after 1.5 weeks, suggesting a higher level of fiber degradation. In supplemented horses the relative abundance of potentially fibrolytic genera was enhanced 2 days after HS and 2 days to 2–3 weeks after the return to HF. Fecal analysis could be a promising technique for monitoring hindgut microbial variations accompanying dietary changes.

Neuroprotective effects associated with immune modulation by selected lactic acid bacteria in a Parkinson's disease model.

Parkinson's disease (PD) is a neurodegenerative process that affects the motor function and involves an inflammatory response and B vitamin deficiencies. The aim of this study was to evaluate the effect of B-group vitamin-producing and immunomodulatory lactic acid bacteria (LAB) in a murine model of PD. The effect of Lactobacillus plantarum CRL 2130 (a riboflavin producer), Streptococcus thermophilus CRL 807 (an immunomodulatory strain), and Streptococcus thermophilus CRL 808 (a folate producer) were evaluated individually and as a mixture in mice injected with 1-methyl-4-fenil-1,2,3,6-tetrahidropiridina. Motor capacity, tyrosine hydrolase in the brain, and cytokine concentrations in serum and brain tissues were evaluated in 1-methyl-4-fenil-1,2,3,6-tetrahidropiridina-treated mice after bacterial supplementation. The mice receiving the selected LAB showed significantly improved motor skills compared with those that did not receive bacterial supplementation. When given the mixture of all 3 strains together, the animals had higher brain tyrosine hydrolase-positive cell counts, decreased inflammatory cytokines interleukin 6 and tumor necrosis factor alpha in serum, and increased antiinflammatory cytokine interleukin 10 in serum and brain tissues compared with animals that did not receive LAB supplementation. The results showed the potential of a selected LAB mixture to improve motor behavior and neuroinflammation in PD. This probiotic mixture could be used as an adjunct treatment in the control of PD.

Impact of yeast and lactic acid bacteria on mastitis and milk microbiota composition of dairy cows

This experiment was conducted to evaluate the impact of yeast and lactic acid bacteria (LAB) on mastitis and milk microbiota composition of dairy cows. Thirty lactating Holstein cows with similar parity, days in milk were randomly assigned to five treatments, including: (1) Health cows with milk SCC < 500,000 cells/mL, no clinical signs of mastitis were found, fed basal total mixed ration (TMR) without supplementation (H); (2) Mastitis cows with milk SCC > 500,000 cells/mL, fed basal TMR without supplementation (M); (3) Mastitis cows fed basal TMR supplemented with 8 g day−1 yeast (M + Y); (4) Mastitis cows fed basal TMR supplemented with 8 g day−1 LAB (M + L); (5) Mastitis cows (milk SCC > 500,000 cells/mL) fed basal TMR supplemented with 4 g day−1 yeast and 4 g day−1 LAB (M + Y + L). Blood and milk sample were collected at day 0, day 20 and day 40. The results showed efficacy of probiotic: On day 20 and day 40, milk SCC in H, M + Y, M + L, M + Y + L was significantly lower than that of M (P < 0.05). Milk concentration of TNF-α, IL-6 and IL-1β in M + Y + L were significantly reduced compared with that of M on day 40 (P < 0.05). Milk Myeloperoxidase (MPO) and N-Acetyl-β-d-Glucosaminidase (NAG) activity of M + Y, M + L, M + L + Y were lower than that of M on day 40 (P < 0.05). At genus level, Staphylococcus, Chryseobacterium and Lactococcus were dominant. Supplementation of LAB decreased abundance of Enterococcus and Streptococcus, identified as mastitis-causing pathogen. The results suggested the potential of LAB to prevent mastitis by relieving mammary gland inflammation and regulating milk microorganisms.

Effect of lactic acid bacteria on Listeria monocytogenes infection and innate immunity in rabbits

The present study aimed to investigate the effect of probiotic lactic acid bacteria (LAB) addition on Listeria monocytogenes translocation and its toxin listeriolysin O (LLO), proinflammatory factors, immune organ indexes and serum immunoglobulins in farmed rabbits. Five treatments included negative control (NC), positive control (PC) with L. monocytogenes infection and supplemental LAB at 3.0 × 10&lt;sup&gt;6 &lt;/sup&gt;(low-LAB, L-LAB), 3.0 × 10&lt;sup&gt;8&lt;/sup&gt; (medium-LAB, M-LAB) and 3.0 × 10&lt;sup&gt;10 &lt;/sup&gt;(high-LAB, H-LAB) CFU/kg of diet, respectively. The LAB was a mixture of equal amounts of Lactobacillus acidophilus (ACCC11073), Lactobacillus plantarum (CICC21863) and Enterococcus faecium (CICC20430). A total of 180 weaned rabbits (negative for L. monocytogenes) were randomly assigned to 5 groups with 6 replicates of 6 rabbits each in response to the 5 treatments. L. monocytogenes infection occurred on the first day of feeding trial and dietary LAB supplementation lasted for 14 days. The results showed that on days 7 and 14 post administration, L. monocytogenes in caecum, liver, spleen and lymph nodes was reduced in M-LAB and H-LAB compared to PC (P &amp;lt; 0.05), and linear and quadratic reducing trends were found in liver on day 7 (P ≤ 0.002). On day 14, mucosa LLO mRNA expression and serum TNFα, IL1β and IFNγ were reduced in the three LAB treatments (P &amp;lt; 0.05), and linear and quadratic trends were found on TNFα and IL1β (P ≤ 0.025); indexes of thymus and spleen, serum IgA and IgG were increased in the LAB treatments (P &amp;lt; 0.05). It is concluded that LAB can be used to alleviate L. monocytogenes infection and to improve the immune function of farmed animals.

Lactobacillus Dominate in the Intestine of Atlantic Salmon Fed Dietary Probiotics.

Probiotics, the live microbial strains incorporated as dietary supplements, are known to provide health benefits to the host. These live microbes manipulate the gut microbial community by suppressing the growth of certain intestinal microbes while enhancing the establishment of some others. Lactic acid bacteria (LAB) have been widely studied as probiotics; in this study we have elucidated the effects of two fish-derived LAB types (RII and RIII) on the distal intestinal microbial communities of Atlantic salmon (Salmo salar). We employed high-throughput 16S rRNA gene amplicon sequencing to investigate the bacterial communities in the distal intestinal content and mucus of Atlantic salmon fed diets coated with the LABs or that did not have microbes included in it. Our results show that the supplementation of the microbes shifts the intestinal microbial profile differentially. LAB supplementation did not cause any significant alterations in the alpha diversity of the intestinal content bacteria but RIII feeding increased the bacterial diversity in the intestinal mucus of the fish. Beta diversity analysis revealed significant differences between the bacterial compositions of the control and LAB-fed groups. Lactobacillus was the dominant genus in LAB-fed fish. A few members of the phyla Tenericutes, Proteobacteria, Actinobacteria, and Spirochaetes were also found to be abundant in the LAB-fed groups. Furthermore, the bacterial association network analysis showed that the co-occurrence pattern of bacteria of the three study groups were different. Dietary probiotics can modulate the composition and interaction of the intestinal microbiota of Atlantic salmon.

We investigated the serum cholesterol and visceral fat lowering effects of combined lactic acid bacteria (Lactobacillus casei WK3, Enterococcus faecium WK5, Bifidobacterium longum WK9, and Lactobacillus plantarum K-1) in high fat diet-fed mice. To investigate the hypocholesterolemic effects of lactic acid bacteria (LAB), six-week old male C57BL/6J high fat-diet fed mice were orally administered WK3 (2.0×109 CFU/mouse) alone or mixed with WK5, WK9 and K-1 (Combined LAB, 5.0×108 CFU/mouse) for 6 weeks. The normal diet control group and high fat-cholesterol diet control group (HFC) were given PBS instead of LAB. After 6 weeks, pathological and biochemical blood analyses were conducted. Upon blood lipid analysis, the levels of total cholesterol and low density lipoprotein cholesterol were significantly decreased in mice in the Combined LAB group (135.2±19.18 and 50.3±0.97 mg/dL, respectively) compared to the HFC group (154.3±15.60 and 64.1±0.73 mg/dL, respectively). However, treatments had no effects on the serum aspartate aminotransferase and alanine aminotransferase levels. In H and E stained liver sections, HFC mice showed accumulation of lipid droplets, but they were rare in livers of Combined LAB fed mice. The increased epididymal adipose tissue of high fat diet-fed mice was significantly reduced by Combined LAB supplementation. These results suggested that administration of Combined LAB had serum cholesterol and visceral fat lowering effects and could be useful probiotics for cholesterol and visceral fat lowering products.
